Abstract:
Although the function of Auto CAD is powerful, its professional pertinence is weak, and partuicularly it cannot satisfy the requirement for the efficient drawing and annotation of the common mechanical structure. The author of this paper has a discussion on the development principle and methods of a 2D General Mechanical CAD System (MACD). Faced up with all the requirements of the mechanical designing 2D drawing, and on the basis of the overall extracting the special features of the mechanical structure 2D primitives, the designing habits and the designing standards and specifications, all the function points of the system have been set up through the intersection and integration of the Auto CAD working mode and the functions of the system. A mechanical CAD system with a general drawing fucntion has been complished with a comprehensive use of the tools of the Auto CAD like function expansion, menu customization and programme development. Applied practice shows that this system has satisfied all the requirements for the mechanical desginers to make the drawings, annotations, enquiry for the standard data, the structure enquiry and quotations so that the designing efficiency has been greatly increased.

Abstract:
STIM1 (stromal interaction molecule 1) is one of the key elements that mediate store-operated Ca2+ entry via CRAC (Ca2+- release-activated Ca2+) channels in immune and non-excitable cells. Under physiological conditions, the intramolecular auto-inhibitions in STIM1 C- and STIM1 N-termini play essential roles in keeping STIM1 in an inactive state. However, the auto-inhibitory mechanism of the STIM1 C-terminus is still unclear. In the present study, we first predicted a short inhibitory domain (residues 310–317) in human STIM1 that might determine the different localizations of human STIM1 from Caenorhabditis elegans STIM1 in resting cells. Next, we confirmed the prediction and further identified an aromatic amino acid residue, Tyr316, that played a crucial role in maintaining STIM1 in a closed conformation in quiescent cells. Full-length STIM1-Y316A formed constitutive clusters near the plasma membrane and activated the CRAC channel in the resting state when co-expressed with Orai1. The introduction of a Y316A mutation caused the higher-order oligomerization of the in vitro purified STIM1 fragment containing both the auto-inhibitory domain and CAD (CRAC-activating domain). We propose that the Tyr316 residue may be involved in the auto-inhibitory mechanism of the STIM1 C-terminus in the quiescent state. This inhibition could be achieved either by interacting with the CAD using hydrogen and/or hydrophobic bonds, or by an intermolecular interaction using repulsive forces, which maintained a dimeric STIM1.

Collaborations abstract the interactions among a group of objects above the level of an individual object-oriented programming (OOP) message send for an integrated design/manufacture process, they can realize information share and system integration, and can be viewed as a type of systematic approach. This paper proposes the collaborative mode in the interior of an enterprise and among enterprises and a novel knowledge-based intensive CAD/CAM framework for collaborative environment development, which integrates product design, design for process planning, NC codes auto-generation and manufacturing simulation. A unified class of knowledge intensive data structures is defined using the blackboard and OO knowledge-based approach for handling both the design and the process planning problems. The detailed knowledge-intensive framework with integration of multi-cooperative knowledge sources and software is given. Thus, product design, process planning and NC codes auto-generation can be carried out simultaneously and collaboratively in an entirely computer-aided collaborative environment.

Abstract:
The detail and design of numerical control system for SLS are discussed. With the help of Visuall C++ and tool kit ObjectARX in AutoCAD, the data information of the 2D contours are extracted from model in AutoCAD by data processing, and these data are transferred to laser scanning data, finally NC codes for processing are generated. A new kind of adaptive slicing algorithm that determines the slicing thickness by comprehensively considering the surface normal and the cross-sectional area, is adopted in data processing. The preheat temperature, scanning speed, laser power and thickness of spreading layer are main factors to part quality in Selective Laser Sintering(SLS). Sintering parameters are optimized by orthogonal experimental design. And a matching rule between scanning velocity and laser power is established so as to realize the laser power matching in different scanning velocities. In order to maintain and transplanted the system, the modular structure is used in the procedure of system design.

Abstract:
Precise matching of measurement data with CAD model for hull block is the key to accurately analysis manufacturing errors and rationally evaluate manufacturing precision. A algorithm to automatically match measurement data with CAD model is proposed. The measurement data is measured by total station, which is currently used in the shipyard. The algorithm is divided into two steps. One is rough matching and another is refined matching. Rough matching that adopts PCA algorithm can narrow the dislocation between measurement data and CAD model. The pair-wise points are matched by searching minimum distance. Initialized by the former result based on rough matching, refined matching that adopts Euler rotation theory leads to perfect matching by translating and rotating the measured points. Experimental results show that the proposed algorithm can automatically match measurement data with CAD model without prior information on transformation, and evaluates manufacturing precision of hull blocks accurately, and provides an instructive basis for subsequent assembly.

Abstract:
Auto-antibodies to oxidized low-density lipoprotein (ox-LDL) are thought to play a pivotal role in the pathogenesis of atherosclerosis. This study investigates the value of auto-antibodies to ox-LDL as a predictive marker of atherosclerosis in patients with both homozygous and heterozygous familial hypercholesterolaemia (FH), who are known to suffer from severe premature atherosclerosis. The influences of well-established risk factors for atherosclerosis such as age, LDL-cholesterol levels and smoking on the results were also determined. Auto-antibody titres to ox-LDL and fasting lipid profiles were measured in 26 homozygous FH patients, 20 heterozygous FH patients without documented coronary artery disease (CAD), 24 heterozygotes with overt CAD and 10 healthy normocholesterolaemic controls. Carotid intima-media thickness, used as an in vivo assessment of atherosclerosis, was also measured in the homozygous FH patients. Ox-LDL titres did not differ between the groups. There was also no association between ox-LDL titres and the LDL-cholesterol level ( P=0·14), presence or absence of CAD ( P=0·69), age ( P=0·50), carotid intima-media thickness ( P=0·51) or smoking ( P=1·0). In conclusion, antibody titres against ox-LDL cannot be used as a predictive marker of the presence or severity of atherosclerosis in patients with FH.

To reduce design man-hours and human errors an expert system shell and a geometric modeling kernel are integrated to automate the design process. Existing algorithms for routing problems have been analyzed -most of them are to solve 2-D circuit routing problems. Design of the ship piping system, especially within the engine room, is a complicated, large-scale 3-D routing problem. Methods of expert systems have been implemented to find the routes of ship pipes on the main deck of a bulk carrier. A framework of the intelligent CAD system for pipe auto-routing is suggested. The making of this workpiece uses three main media, namely Auto CAD 2000 software, CNC Milling KELLER Q plus software, and TU 3A CNC milling machine. The process of making the workpiece starts from making the workpiece design in Auto CAD software which is then stored on the CDR via DFX data. The design of the workpiece contained in the CDR is then transferred to CNC Millling KELLER Q plus software via Geometry. in Geometry, the design of the workpiece is dimensioned. Completed from the Geometry the workpiece design is included in the Work plan to be processed so that it can be simulated to move to a CNC program to convert the graphical language into a numeric language so that it can be read by a CNC machine. The programming results from the CNC KELLER Q plus software are then recorded to be executed on the CNC TU 3A milling machine to obtain the workpiece in accordance with the design in AutoCAD. Programming results from CNC Milling KELLER Q plus software cannot be directly used on the TU 3A milling machine (EMCO), because the programming language used is different. Abstract:
Abstract Primary cold agglutinin disease (CAD) is a hemolytic anemia mediated by monoclonal anti-I autoantibodies. CAD is caused by an underlying low grade B-cell lymphoproliferative disease of the bone marrow with a typical histology that is different from lymphoplasmacytic lymphoma and, accordingly, does not display the MYD88 L265P mutation (Randen et al., Haematologica, 2014). Since CAD is a clonal lymphoproliferative disorder, we studied the mutational landscape to further characterize the disease and identify potential novel treatment approaches. We prospectively collected bone marrow samples of CAD patients, enrolled in a clinical trial (CAD5; www.clinicaltrials.gov, NCT02689986). Exome sequencing of six cases was performed and findings were confirmed in ten additional cases using targeted sequencing. For these analyses, clonal B cells and normal T cells, used as control, were purified from bone marrow samples using fluorescent activated cell sorting. All mutations were verified by Sanger sequencing. Whole-exome sequencing was performed at BGI Tech Solutions (Hongkong) using the Agilent SureSelect Human All Exon V4 Reagent Kit and Illumina HiSeq technology. The bioinformatics pipeline consistent of BWA alignment tool (aligned to hg19); Picard tools FixMateInformation and MarkDuplicates; the Genome Analysis Toolkit (GATK) IndelRealigner and BaseRecalibrator; somatic variant detection tools Strelka, MuTect and Pindel; the annotation tool SnpEff. Recurrent somatic mutations were found in KMT2D (11/16 cases, 69%) and CARD11 (5/16, 31%) (Table 1, Figure 1-2). 7/16 (44%) of KMT2D mutations were deemed high impact mutations by SnpEff and to result in inactive protein. 2/16 (12,5%) of KMT2D mutations are missense mutations and are predicted to impair SET domain function. 2/16 (12,5%) of KMT2D mutations are classified by SnpEff as low impact mutations of which functional tests are necessary to demonstrate potential consequences for protein function. Of interest, two additional patients showed rare germline KMT2D variants that have also been seen in Kabuki syndrome patients, although these patients do not have Kabuki syndrome. CARD11 was somatically mutated in 5/16 (31%) cases. Four of those patients had a concurrent KMT2D mutation. All CARD 11 mutations were classified as moderate impact mutations by SnpEff. Mutations were tightly clustered in a 20bp sequence of the coiled-coil domain sequence (Table 1, Figure 2). KMT2D is a histone lysine methyl transferase that represses B cell lymphoma development. Mono-allelic mutations of KMT2D seem to act in a dominant fashion and cause partial loss of protein expression with cell growth advantage. KMT2D is frequently mutated in follicular lymphoma, diffuse large B cell lymphoma and nodal marginal zone lymphoma. Mono-allelic constitutional mutations cause Kabuki syndrome, characterized by distinct facial characteristics and multiple organ malformations. Patients frequently develop immune-mediated thrombocytopenia as well as auto-immune hemolytic anemia. CARD11 coiled-coil domain mutations result in constitutive NF-kB activation and enhanced NF-kB activity upon antigen receptor stimulation. Mutations were previously detected in diffuse large B cell lymphomas of activated B cell origin. Mono-allelic CARD11 coiled coil mutations are not oncogenic per se in mice and humans, but result in B-cell proliferation and auto-antibody production. Since four CAD patients showed concurrent KMT2D and CARD11 mutations, it seems likely that the mutations act in concert with anti-I B-cell receptor stimulation to contribute to CAD-associated lymphoproliferative disease. In conclusion, we demonstrated a high frequency of KMT2D and CARD11 mutations in the bone marrow B cell lymphoproliferative disease of patients with CAD. These results confirm that CAD-associated B cell lymphoproliferative disease is a distinct disease different from other known B cell lymphoproliferative diseases of the bone marrow, most notably lymphoplasmacytic lymphoma. The identification of these recurrent mutations in CAD may allow the design of novel treatment modalities. Table 1 Mutations in KMT2D and CARD11 gene in CAD. Table 1. Abstract:
This study aims to explore the causal relationship between fiscal deficit (FD) and current account deficit (CAD) along with policy recommendations based on long-run and short-run dynamics and sensitivities. A panel data span from 1990 to 2019 is analyzed based on panel unit root tests, panel co-integration with auto-regressive distributed lag (ARDL), panel co-integration regression with fully modified ordinary least squares (FMOLS) and dynamic ordinary least squares (DOLS), and causal analysis with the Dumitrescu and Hurlin (DH) technique. The results disclosed that all tested variables are stationary at the first difference I(1) except the real interest rate (IR), which is stationary at level I(0). The ARDL estimates suggested that there is a long-run relationship between tested variables and 92% annual convergence is possible for long-run equilibrium. The FMOLS and DOLS estimates indicated that the CAD is sensitive towards the FD and the exchange rate. The DH causality test showed that the CAD is significantly affecting the FD, supporting the current account targeting hypothesis. Furthermore, it is observed that the interest rate is acting as a moderating factor between the FD and the CAD because it causes both the deficits. Thus, reverse causality is concluded from the CAD to the FD. These results have macroeconomic implications for fiscal policy in the Association of South-East Asian Nations (ASEAN-10).

Abstract:
Abstract The capability to adapt a manufacturing plant to changing requirements gains increasing importance in industrial production environments, e. g., triggered by Industrie 4.0 scenarios. A virtual as-built model of a manufacturing plant and its surrounding factory building provides important decision support and relevant information for digital twins, e. g., to trace assets and asset types across their whole lifetime, planning of renovations, plant and machine topology changes, or the simulation-based analysis of production processes. Based on point clouds obtained by terrestrial laser scanning or photogrammetric acquisition, reverse engineering can be applied to extract and to reconstruct relevant objects in a form suitable for CAD programs. In this article, we review approaches to capture a scene by point measurements and to reconstruct the geometry of its components given specific object models. This comprises the discussion of various representation schemes for objects and their relations, strategies for object recognition, and the explication of methods for model instantiation. Furthermore, depending on the requirements for specific tasks, we identify technology gaps and specify the degree of maturity of the related techniques.

Abstract:
ABSTRACT Introduction: Orthodontic treatment for adults is currently increasing, and therefore the need to bond brackets to restorations and temporary crowns. The use of CAD/CAM PMMA provisional restorations for orthodontic purposes have not yet been described, and there is currently insufficient information regarding the strength of bracket adhesion. Objective: This study aimed at evaluating the effects of thermocycling (TC) and surface treatment on shear bond strength (SBS) of brackets to different provisional materials. Methods: Forty specimens were made from each material [PMMA (Telio Lab), bis-acryl (Telio CS C&B), and PMMA CAD/CAM (Telio CAD)], sandpapered, and divided according to surface treatment (pumiced or sandblasted) and TC (half of the samples = 1,000 cycles, 5°C/55°C water baths) (n = 10/group). Stainless-steel brackets were bonded to the specimens (using Transbond XT), and SBS testing was performed. Data were analyzed by three-way ANOVA and LSD post-hoc tests (α = 0.05). Failure types were classified with adhesive remnant index (ARI) scores. Results: SBS values ranged from 1.5 to 14.9 MPa. Sandblasted bis-acryl and sandblasted auto-curing PMMA groups presented similar values (p> 0.05), higher than the CAD/CAM material (p< 0.05), with or without TC. When thermocycled, pumiced bis-acryl showed higher SBS than pumiced acrylic (p= 0.005) and CAD/CAM materials (p= 0.000), with statistical difference (p= 0.009). TC showed negative effect (p< 0.05) for sandblasted bis-acryl and pumiced acrylic groups. ARI predominant score was mostly zero (0) for CAD/CAM, 1 and 2 for bis-acryl, and 1 for acrylic groups. Conclusion: In general, bis-acryl material showed the highest SBS values, followed by acrylic and CAD/CAM materials, which showed SBS values lower than an optimum strength for bonding brackets.

Abstract:
A seamless analysis of complex geometry is one of greatly interesting topic. However, there are still gaps between the industrial applications and fundamental academic studies owing to time consuming modeling process. To resolve this problem, an auto mesh generation program based on grid-based approach has been developed for IT-product in the present study. At first, base mesh and skin mesh are generated using the information of entities which extracted from IGES file. Secondly the provisional core mesh with rugged boundary geometry is constructed by superimposing the skin mesh as well as the base mesh generated from the CAD model. Finally, the positions of boundary nodes are adjusted to make a qualified mesh by adapting node modification and smoothing techniques. Also, for the sake of verification of mesh quality, the hexahedral auto mesh constructed by the program is compared with the corresponding tetrahedral free mesh and hexahedral mapped mesh through static finite element analyses. Thereby, it is anticipated that the grid-based approach can be used as a promising pre-processor for integrity evaluation of various IT-products.

Abstract:
Patients with severe chronic coronary artery disease (CAD) exhibit a highly altered myocardial pattern of perfusion, metabolism, and mechanical performance. In this context, the diagnosis of stunning remains elusive not only because of methodological and logistic considerations, but also because of the pathophysiological characteristics of the myocardium of these patients. In addition, a number of alternative pathophysiological mechanisms may act by mimicking the functional manifestations usually attributed to stunning. The present review describes three mechanisms that could theoretically lead to reversible mechanical dysfunction in these patients: myocardial wall stress, the tethering effect, and myocardial expression and release of auto- and paracrine agents. Attention is focused on the role of these mechanisms in scintigraphically “normal” regions (i.e., regions usually showing normal perfusion, glucose metabolism, and cellular integrity as assessed by nuclear imaging techniques), in which stunning is usually considered, but these mechanisms could also operate throughout the viable myocardium. We hypothesize that reversion of these three mechanisms could partially explain the unexpected functional benefit after reperfusion recently highlighted by high-spatial-resolution imaging techniques.
