Abstract:
The current study was designed to evaluate the efficiency of broccoli plant in therapy of PCOS that induced for the first time by testosterone andriol (T.A). Forty-eight immature female rats (21 days old) were divided into 6 groups (8 rats each) as follows: G1, animals were injected with sesame oil for 39 cascade days (control). G2, animals were injected with 1mg/100g b.w of T.A for 39 cascade days. G3, animals were injected with 1mg/100g b.w of T.A gathered with gavaged broccoli juice (b.j) for 39 cascade days. G4, animals were injected with sesame oil for cascade 39 days at the end of last injection were gavaged with d.w for 30 cascade days. G5, animals were injected with 1 mg/100g b.w of T.A for 39 cascade days and at the end of last injection were gavaged with d.w for 30 cascade days. G6, animals were injected with 1mg/100g b.w of T.A for 39 days and at the end of the last injection were gavaged with b.j for 30 cascade days. T.A-induced PCOS rats (G2 and G5) in comparison with controls (G1 and G4) exhibited that had a significant (P≤0.01) increase in the b.w, ovarian, uterine and fat pads weights as well as the fat cells diameter. Additionally, these groups had acyclicity, high numbers of ovarian cysts, atretic and preantral follicles in the ovaries, lowering the number and the diameter of C.L., and uterine changes. The levels of T, E and LH, an insulin, glucose, LDL, cholesterol in the serum were increased whereas the levels of FSH, LDH were decreased. Therefore, T.A PCOS model could be appropriated for studied all the features of syndrome i.e, ovarian, metabolic and endocrinologiccal disturbances. Whereas all these measurements of G3 and G6 were ameliorated and recovered into their normalcy. Our findings have shown the benefit of consuming broccoli in the protection (G3) and prevention (G6) of PCOS.

Abstract:
The possible cardioprotective effects of translocator protein (TSPO) modulation with its ligand 4′-Chlorodiazepam (4′-ClDzp) in isoprenaline (ISO)-induced rat myocardial infarction (MI) were evaluated, alone or in the presence of L-NAME. Wistar albino male rats (b.w. 200–250 g, age 6–8 weeks) were divided into 4 groups (10 per group, total number N = 40), and certain substances were applied: 1. ISO 85 mg/kg b.w. (twice), 2. ISO 85 mg/kg b.w. (twice) + L-NAME 50 mg/kg b.w., 3. ISO 85 mg/kg b.w. (twice) + 4′-ClDzp 0.5 mg/kg b.w., 4. ISO 85 mg/kg b.w. (twice) + 4′-ClDzp 0.5 mg/kg b.w. + L-NAME 50 mg/kg b.w. Blood and cardiac tissue were sampled for myocardial injury and other biochemical markers, cardiac oxidative stress, and for histopathological evaluation. The reduction of serum levels of high-sensitive cardiac troponin T hs cTnT and tumor necrosis factor alpha (TNF-α), then significantly decreased levels of serum homocysteine Hcy, urea, and creatinine, and decreased levels of myocardial injury enzymes activities superoxide dismutase (SOD) and glutathione peroxidase (GPx) as well as lower grades of cardiac ischemic changes were demonstrated in ISO-induced MI treated with 4′-ClDzp. It has been detected that co-treatment with 4′-ClDzp + L-NAME changed the number of registered parameters in comparison to 4′-ClDzp group, indicating that NO (nitric oxide) should be important in the effects of 4′-ClDzp.

Abstract:
The present study explored the assessment of the antidiabetic potential of Tinospora cordifolia & Juglans regia composite extract in STZ induced diabetes in wistar rats. As streptozotocin -associated infiltrations of increase glucose level has been reported to be responsible for diabetes. We evaluated the glucose lowering potential of Tinospora cordifolia & Juglans regia on the basis of its anti-diabetic property. Rats were administered streptozotocin (55 mg/kg i.p., once) with nicotinamide (120mg/kg) to induce experimental toxicity. The development of diabetes was assessed biochemically as well as histologically 72 hours after induction of diabetes. Body weight and blood glucose levels were determined in (0, 7th, 14th, 21st, 28th) days. Serum lipid profile and enzyme estimated, (kidney, liver, pancreas) tissue was measured at the end of the experimental period. Treatment with composite extracts TCAE high dose (350 gm/kg b.w.) & JRAE high dose (800 mg/kg b.w.) and TCHE high dose (350 gm/kg b.w.) & JRHE high dose (800 mg/kg b.w.) were noted to be more effective against the streptozotocin- induced toxicity as compared to Glibenclamide (5 mg/kg b.w.). it may be concluded that streptozotocin-induced glucose may be accountable for the induction of diabetes toxicity in rats. Interestingly, improvement in body weight, glucose level, lipid profiles, biochemical parameters and histopathological changes in kidney, liver and pancreas was observed following herbal treatment in STZ induced diabetic rats. Furthermore, composite extract of TCAE (350mg/kg b.w.) & JRAE (800mg/kg b.w.) was found to be efficacious than the composite extract of TCHE (350mg/kg b.w.) & JRHE (800mg/kg b.w.).

Abstract:
BACKGROUND: Oxidative stress in type 2 diabetes mellitus (T2D) causes insulin resistance and disordered insulin secretion. Pathomechanisms of T2D consist of dysfunctional pancreatic β-cell and insulin resistance caused by free radical (reactive oxygen species and reactive nitrogen species) that produced from the glucose metabolism pathway. Insulin resistance can be measured using the homeostatic model assessment of insulin resistance (HOMA-IR). Oxidative stress can measure through the activities of malondialdehyde (MDA) and superoxide dismutase (SOD). AIM: This research aims to study the potential of chayote (Sechium edule Jacq. Swartz) to be used as antihyperglycemic in T2D. MATERIALS AND METHODS: This research was conducted with a post-test randomized controlled group design. Eleven groups with four male rats each were used. Normal untreated rats were treated under ad libitum feeding and drinking condition. Meanwhile, the rat models were induced with the combination of 45 mg/kg b.w. streptozotocin, 110 mg/kg b.w. nicotinamide, 40.5 mg/kg b.w. metformin, high-fat diet, and/or chayote extract. The chayote extract was orally administered to the rat in the form of ethanol extract and/or ethyl acetate fraction, with three dosages of 45 mg/kg b.w., 100 mg/kg b.w., and 150 mg/kg b.w. for each extract type. The body weight, glucose level, insulin level, MDA, and SOD activities were measured. The HOMA-IR was used. RESULTS: The lowest body weight of the rat model in week 0 was 145 ± 25.31, founded in Group H that was treated with ethyl acetate fraction of chayote extract (45 mg/kg b.w.). The lowest blood sugar level in the group with 2 h glucose load was 112.5 ± 27.00 on average, found in Group G that was treated with chayote ethanolic extract (150 mg/kg b.w.). The highest SOD in the group treated with chayote extract was 1.27 ± 0.20, founded in Group H treated with ethyl acetate 45 mg/kg b.w. The lowest level of MDA was 0.86 ± 0.70 in Group H treated with ethyl acetate 45 mg/kg b.w. The lowest fasting blood sugar spectrophotometer level was 150.54 ± 17.24 mg/dl in Group K with metformin treatment, followed by 155.16 ± 31.92 mg/dl in Group K treated 45 mg/kg b.w. ethanol treatment. The highest insulin level was 6.14 ± 0.71, founded in Group F that was treated with chayote ethanolic extract 100 mg/kg b.w. The lowest measurement of HOMA-IR was 0.16 ± 0.80 in Group E treated with ethanol extract of chayote 45 mg/kg b.w. CONCLUSION: Ethanol extract and fractionation of chayote work as an antioxidant and anti-insulin resistance.

The possible protective action of sodium-2-mercaptoethanesulfonate (mesna) on hemotoxicity induced by anticancer drugs was tested in normal mice. Mesna (100 mg/kg b.w.) was injected i.v. 1 h before the i.v. administration of cyclophosphamide (10, 120, and 200 mg/kg b.w.), doxorubicin (4.32, 7.2, and 12 mg/kg b.w.) and cis-diamminedichloroplatinum (5.4, 9, and 15 mg/kg b.w.). Results show that in this experimental model mesna does not seem to protect against toxicity of the tested anticancer drug on hemopoietic progenitor cells.

The present study was undertaken to evaluate the anti-inflammatory potential of Murraya koenigii root extracts petroleum ether, ethyl acetate and chloroform (MKPE, MKEA and MKCF, respectively) and Ficus lacor aerial root extracts petroleum ether, ethyl acetate, chloroform and ethanol (FLPE , FLET, FLCF and FLET, respectively) at doses of 50 and 100 mg/kg body weight (b.w.) using animal models of acute inflammation (carrageenan-, histamine- and serotonin-induced inflammation). The results of the Murraya koenigii roots chloroform extract caused 66.4% inhibition and the ethanol extract of Ficus lacor aerial roots caused 68.3% inhibition at the dose of 50 mg/kg b.w. At a higher dose of 100 mg/kg b.w., MKPE and MKCF showed 55.10% and 70.10% inhibition, respectively. FLPE and FLET showed 74.50% and 75.40% inhibition, respectively, in the carrageenan-induced inflammation model. In histamine-induced inflammation, the MKCF showed 60% inhibition, and 67.01% and 68.02% inhibition with the petroleum ether and ethanol extracts, respectively, in Ficus lacor aerial roots at the dose of 50 mg/kg b.w. At a higher dose (100 mg/kg b.w.), MKCF showed 64% inhibition. FLPE and FLET showed 70.13%and 74.01% inhibition, respectively; 62.15% and 66.10% inhibition was observed with the petroleum ether and ethanol extracts of Ficus lacor aerial roots at 50 mg/kg b.w. At higher dose (100 mg/kg b.w.), FLPE and FLET showed 69.10% and 68.72% inhibition in serotonin-induced inflammation.

Abstract:
In the present work, the antinociceptive action was assayed in several experiment models in mice, Hot plate, writhing and formalin test. The alcoholic extract of MP leaves at a dose of 150mg/kg B.W and 300 mg/kgB.W showed antinociceptive effects in different methods, where the dose of 300 mg/kg B.W showed significant reduction of the nociception by acetic acid. In the formalin test, the extract (300mg/kg B.W) also significantly reduced painful stimulus in both phases of the test. Treatment with extract (300mg/kg B.W) when given orally produced significant increase of the reaction time in hot plate test. These results showed that the leaves extract of MP contain active analgesic principles acting both centrally and peripherally.

Abstract:
Diabetes mellitus (DM) is a chronic metabolic disease with the highest rates of prevalence and mortality worldwide. Chloroxylonswietenia has been used extensively in folkloric medicine. The present study aims to determine the acute toxicity of Chloroxylonswietenia bark methanol (CSBMEt) and aqueous extracts (CSBAEt) (100, 150, 250, 500 and 1000 mg/kg body weight) and dose fixation of CSBMEt and CSBAEt in streptozotocin induced diabetic rats. Diabetes was induced in male albino wistar rats by single intraperitoneal injection of streptozotocin (50mg/kg b.w). The diabetic rats were administered with Chloroxylonswietenia bark extracts (CSBMEt and CSBAEt) (75,125 and 250mg/kg b.w) orally by intragastric intubation for 15 days. Acute toxicity studies revealed the non-toxic nature of the CSBMEt and CSBAEt. No lethality or toxic reactions found at any doses until the end of study, whereas 75 and 125 mg/kg b.w. doses of CSBMEt and CSBAEt produce no significant changes in the diabetic rats and 250mg/kg b.w. of CSBMEt and CSBAEt have significant change in the blood glucose. The results conclude that, there was no toxicity observed up to 1000mg/kg b.w. of both the extracts and 250mg/kg b.w. of CSBMEt and CSBAEt can be used as effective dose to treat diabetes.

Die doel met hierdie navorsing was om ‘n analise van die selfgereguleerde leervaardighede van leerders in die BEd Honneurs Oopafstandsleerprogram te doen en ‘n onderrigmodel daar te stel om genoemde vaardighede te verbeter. Om hierdie doel te bereik is ‘n literatuurstudie onderneem ten einde a) leer te omskryf volgens die behavioristiese, sosiaal–kognitiewe, inligtingverwerkings, konstruktiwistiese teorieë en volwasse leerteorieë; b) selfgereguleerde leer te omskryf vanuit die behaviouristiese, fenomenologiese, Vygotskiaanse, kognitief–konstruktivistiese en sosiaal–kognitiewe beskouinge, met die klem op laasgenoemde beskouing en c) afstandsleer en die vereistes wat afstandsleer en selfgereguleerde leer aan afstandsleerders stel, te beskryf. Vanuit literatuur is bepaal dat effektiewe leer gedefinieer word vanuit die navorser se oriëntasie tot leer sowel as die aard van leer in ‘n spesifieke konteks. Die verskillende leerteorieë beklemtoon verskillende aspekte van leer en hou verskillende implikasies in vir programontwerp, rolle van onderriggewers, leermetodes en assessering. Verder dui literatuur op ‘n positiewe verband tussen selfgereguleerde leer en akademiese prestasie. Terselfdertyd dui bestaande literatuur daarop dat suksesvolle afstandsleerders selfregulerend moet kan leer en dus oor sodanige vaardighede moet beskik. Die navorsingspopulasie het bestaan uit BEd Honneurs oopafstandsleerders van Noordwes–Universiteit asook fleksieleerders van die Noordwes–Universiteit, Potchefstroomkampus wat alreeds eksamen afgelê het in die Leerperspektiewe module. Die teikengroep het bestaan uit leerders wat reeds vroeër eksamen afgelê het in die module LEON 611 en tydens kontaksessies in 2010 by spesifieke kontaksentrums en vakansieskole bygewoon en die vraelyste voltooi het (n=264). ‘n Multi–metode ontwerp is geïmplementeer waar ‘n kombinasie van kwantitatiewe en kwalitatiewe navorsing aangewend is om die navorsingsprobleem beter te verstaan. Kwantitatiewe data is ingesamel aan die hand van vier vraelyste waarná aanvullende kwalitatiewe data–insameling met semi–gestruktureerde onderhoude gevolg het. Kwantitatiewe navorsing het geskied aan die hand van ‘n ex post facto–benadering en data is statisties ontleed deur middel van a) faktoranalise, b) meervoudige regressie–analise, c) stapsgewyse regressie–analise en d) effekgroottes. Kwalitatiewe data is met behulp van ATLAS.ti.6.0 - ‘n rekenaarondersteunde stelsel, ontleed. Resultate van die ondersoek dui daarop dat deelnemers nie voldoen aan al die vereistes vir effektiewe afstandsleer nie. Bereiking van leerdoelwitte is in ‘n groot mate afhanklik van die steun van onderriggewers en porture. Hul beskik nie oor inligtingstegnologiese vaardighede nie en kan nie effektief leer waar nie–moedertaalonderrig ter sprake is nie. In teenstelling met bestaande literatuur dui kwantitatiewe en kwalitatiewe resultate op ‘n lae verband bestaan tussen afstandsleerders se selfgereguleerde leervaardighed en hul akademiese prestasie. Deelnemers maak selde gebruik van selfgereguleerde leervaardighede soos ter sprake tydens die voorafdenkfase maar meer tydens die wilsbeheer– en selfreaksiefases. ‘n Model is ontwikkel vir die verbetering van selfgereguleerde leervaardighede van oopafstandsleerders, inbegrepe deurlopende ontwikkeling van akademiese taalvaardigheid en inligtingstegnologiese vaardighede. Aanpassings in studiemateriaal en verandering van onderrigstrategieë en terugvoer is belangrike aspekte van die model.

Le travail est basé sur l'édition scientifique de trois traités antihérétiques anonymes et inédits. L'étude menée à partir de cette édition a permis d'apporter un éclairage nouveau sur la compréhension de la construction du discours antihérétique. Il apparaît que ces traités ne présentent pas une hérésie cathare constituée comme une vraie religion. Il faut les comprendre comme un ensemble de chapitres qui sont autant de problèmes qui peuvent se poser à propos du dogme catholique, problèmes qui ne sont pas envisagés comme solidaires les uns des autres mais comme susceptibles de se poser de façon autonome. Ces traités ont été rédigés par un compilateur plus que par un auteur, puisqu'il s'agit de textes qui sont constitués par l'organisation d'autorités réparties par thèmes. Le but de ce compilateur n'était pas, malgré l'emploi du style direct dans le discours, de « polémiquer » avec d'éventuels hérétiques, mais plutôt d'informer les prêtres avec charge d'âme. Ces trois textes ne présentent, sur chaque thème, que des lieux communs, donnant aux traités une image atemporelle. Cela peut paraître étonnant de prime abord, mais cela s'éclaire assez bien si l'on considère que ces traités sont avant tout des ouvrages de vulgarisation qui ont été composés pour des prêtres considérés a priori comme ignorants par des hommes formés dans les écoles. Our work is based on the scientific study of three anonymous and previously unpublished antiheretic treaties. The study of this edition has allowed a new light to be thrown on the understanding and construction of the antiheretic discourse. These various treaties do not portray a Cathar heresy constituted as a true religion. They must be taken into consideration as a group of chapters that are as many problems as can be raised about the Catholic dogma – problems not to be looked upon as interdependent but liable to exist for themselves. The treaties have been written by a compiler rather than by an author as their constitution by the organization of authorities classified by themes makes obvious. In so doing, the goal of the compiler was not, though he uses free direct speech in his discourse, to become involved in a controversy with possible heretics but rather to instruct priests who had the cure of souls. The three texts only have commonplaces to offer on each of the various themes, thus conferring a timeless image to the treaties. This may seem surprising at first but becomes clear enough when one considers they were in the first place popularizing works written by scholars and intended for priests looked down upon as ignorant. A crucial theme, that of ecclesiology and sacraments, runs through the three treaties while that of dualism gains both in importance and clarity. At the same time other themes, such as those dealing with priesthood as well as oath and murder, are obviously loosing ground

Methodology: The Behavioral and Environmental Safety Program (B.E. Safe) for Early Childhood Professionals is being developed in response to Federal Safety regulations for childcare procedures and addresses social and emotional safety concerns. The curriculum includes Family Life Education content areas focusing on diversity, inclusion, social and emotional health, bullying prevention, supporting children with special needs, building relationships with parents, and understanding children’s behavior. Discussion of existing research: Research states that high quality childcare and education benefits all children (DHSS, 2015). Best practices indicate that Early Childhood Professionals need high quality training to be equipped to understand child development, strategies for working with families, and having the resources for supports when including children with disabilities in early childcare programs (DHSS, 2015). Not only should Early Childhood Professionals be equipped with the tools and resources to understand children’s behavior, educators need to demonstrate competency in state regulations, environmental safety for children, and playground safety (Sheridan, Edwards, Marvin & Knoche, 2009). B.E. Safe allows Early Childhood Professionals to engage in a research-based training that will help to ensure safety within their center. Connection to Theory: Behavior theories of child development incorporate environmental influences on behavior (Shute & Slee, 2015). A child’s early learning environment and the adults influence of children affects emotional, social, and physical development. B.E. Safe relates to Vygotsky’s Sociocultural Theory (1978) suggesting that children learn actively through hands on experiences. The Sociocultural Theory suggest that parents, caregivers, and a child’s environment from a young age is largely responsible for higher order functions (Shute & Slee, 2015). Proficiency in Adverse Childhood Experiences (ACEs) will allow participants to grasp how traumatic events can affect a child’s developmental process. Educating early childcare professionals on ACEs will allow for adults to be aware of what to look for, and how to keep children protected from adverse experiences (Sacks, Murphey & Moore, 2014). How will the study add to knowledge/innovative practice? Family Scientists will gain knowledge on best practices for providing Family Life Education to Early Childhood Professionals. Specifically, participants will gain knowledge on promoting inclusion, understanding behavior issues, developmental delays, and how to keep children safe from potential environmental hazards.

Architecture and mobility merge together to provide space for activities away from both home and office. The densification of people’s living space and the delocalisation of the workplace are two of the biggest emerging trends in the near future. Society is constantly changing with new needs and desires. The Internet of Things, new technologies and possibilities of autonomous driving enable new approaches to redesign the traditional car. The aim of this project is to rethink mobility and especially the car as we know it today. Since it has been shown that cars are no longer used exclusively as a means of transport, but also as mobile space, the image of the car as people know it could be redrawn Since BMW i is an innovative brand, which can break new ground, it can be ahead of its time and lead society instead of following it. After the first research and examination of relevant topics, the results were further developed and worked out. The system in which the concept takes place is defined in more detail based on further investigations such as scenario, marketplace and target group. The focus is mainly on the interior and exterior of the vehicle. In addition, an insight into the inspiration and the process of aesthetic design is described and illustrated in several sketches and renderings. Then the further development in 3D and VR (Virtual Reality) is shown up to the final digital model. iSpace is a system that brings attention to  the potential new role of the car in the coming years. iSpace is rather a mobile architecture than a conventional car, which means that it is used in a completely new and fresh way. The vehicle is designed to be used mainly in the static mode and offers the possibilities of a mobile space to support the flexible and delocalised lifestyle of people. In that sense it shows a completely new view how a vehicle could look like, while creating new kind of benefits to enrich and improve the quality of life. iSpace is part of a bigger system containing several vehicles to cover different types of needs and HUBs, which are the extension of the vehicles to various locations in urban areas.

Perpetuating African ancestral customs, Mardi Gras Indians in New Orleans avoid the African American identity crises illuminated by the poetry of the Harlem Renaissance. The poetry of Langston Hughes, Claude McKay and Waring Cuney incorporate W.E.B. DuBois’ double-consciousness theory to reveal the identity issues and ancestral alienation plaguing African Americans at the turn of the twentieth-century. In comparison, unique political and social circumstances in New Orleans allowed enslaved Africans to practice their ancestral customs weekly. The preservation of this heritage fostered a black community in New Orleans rich in traditions, pride and self-conviction. The development of Mardi Gras Indian culture and the allusions to Africa in Harlem poetry reveal the power of ancestry to establish identity.

The aim of this thesis project was to create something, that has never been there before in motorcycle design. A new, deep collaboration between the future generation of riders and their bikes. It should create a whole new riding experience. And this experience should be inspirational and new to several fi elds of transportation design besides motorcycling. The process started with the research and inspiration fi nding. It continued with form exploration through sketching, ergonomic tests and photoshop sketches. The whole process was based on an interdisciplinary working method. Being in touch with modelers, engineers, generative designers, color and trim designers was an essential part of the project. The interdisciplinary work continued digitally, with many different types of software. Creating the 3D visualisation of the project required the use of Alias, Cinema 4D, Rhino, Grasshopper, Fuse, Speedform, Maya and VRED. Frequently reviews on the current project status and taking part in technical meetings, along with responsible designers at BMW, in combination with weekly reviews with the university, structured the time plan throughout the whole project. As well as fl ying to the university for check-ups and presentations. BMW Spirit provides a revolutionary experience for future motorcycle riders. Focusing on the interaction between the rider and the bike, it creates the opportunity to build a deep relationship. Since AI becomes more and more important in our daily lives developing ways of human-machine interactions will be crucial for our future. The concept is focused on the opportunities it could bring to combine an Artifi cial Intelligence with motorcycling. BMW Spirit creates a human-machine symbiosis and is controlled like an extension of the human body, with intuitive movements. The built in Artifi cial Intelligence is the rider’s companion and develops its own character and consciousness over time. It learns and grows together with the rider and adapted to the rider. Growing trust enables more possibilities to push the limits and enjoying the power of this unique riding experience.

Abstract:
This historical commentary on the Metz Epitome, a late fragmentary account of Alexander's exploits, compares the work with the extant early Alexander historians. The sources of the anonymous author have much in common with the Cleitarchan historians, in particular, Diodorus Siculus and Quintus Curtius Rufus. Non-Cleitarchan elements in the text seem to reflect a certain affinity with a Hebraic tradition concerning Alexander. An examination of the author's methodology suggests that "epitome" is not an accurate description of the work in question. The anonymous author has achieved a unique portrait of Alexander and included information not found elsewhere. In view of its late authorship and the few new crumbs of historical fact it offers, the value of the Metz Epitome lies in its interpretation of Alexander's career rather than as a source for it.

Low molecular weight (LMW) heparlns have been developed with the aim of reducing anticoagulant activity thereby minimizing the bleeding complications of conventional heparin. Unexpectedly, bleeding events were reported during treatment with some LMW-heparins, in clinical and experimental studies. We studied the effect of four different LMW-heparlns on primary haemostasis In male rats (CD COBS, Charles River) after l.v. administration of 0.75 mg/kg b.w. of the drugs. LMW heparin A was devoid of any activity on an experimental model of “template” bleeding time in rats (110.6±5.9 sec versus 108.7±4.1 control values) whereas LMW-heparins B, C and D prolonged the bleeding time to a different extent (228.7±19.9, 161.5±6.4 and 161.7±8.6 respectively). Pretreatment of animals with aspirin (100 mg/kg b.w. per o.s). resulted In a significant potentiation of the “template” bleeding time. In vitro platelet aggregation Induced by collagen (20 μg/ml) or by collagen in combination with ADP (5-10 μM) was strongly inhibited by LMW-heparln B, while LMW-heparln A showed no effect. LMW-heparins C and D exerted an Intermediate level of Inhibition of platelet aggregation. The same pattern of aggregating response was found when LMW-heparins A and B were given i.v. to rats (0.75 mg/kg b.w.) and platelet aggregation was studied “ex vivo” 15 min after drug administration.These data may help explain the impairment of primary haemostasis associated with some LMW-heparin preparations.

An enzymatically degraded low molecular weight heparin (mean mol wt 4900 dalton, LOGIPARIN, NOVO In-dustri, Denmark) was given s.c. in a dose of 35 Xal u/kg b.w. once daily to 10 patients undergoing elective hip surgery in order to study the plasma levels of Xal and Hal activity and with special regard to safety. The Xal activity in plasma was ≤ 0.24 Xal u/ml and the Hal activity ≤ 0.043 Hal u/ml. No bleeding complications were observed, and no increase in per- and postoperative blood loss was seen in the present study. It is therefore postulated that plasma levels of the Xal and Hal activities within the range obtained are safe with regard to bleeding complications for the LMW heparin used and in the dose administered. No accumulation was seen, which may contribute to the safety. No phlebographically verified thrombi were registered but the antithrombotic efficacy of the dose regimen used remains to be demonstrated in a larger number of patients under controlled conditions, a study that is well under way.

The influence of low molecular weight heparin (Mw7600) in combination with dihydroergotamine (DHE) onthrombus formation and primary haemostasis was investigated in rabbit models. The jugular veins were used, thrombosis being induced by a combination of endothelial damage (ethoxysclerol) and flow reduction (standardized narrowing of the vein). Haemostatig plug formation was studied by transecting isolated microvessels in the mesentery. Conventional heparin in the dose of 60 units anti Xa activity/kg b.w. effectively prevented thrombus formation but the same dose of a low molecular weight fragment only gave a marginal decrease of the frequency of thrombosis (from79 to 57%). Doubling the dose of the heparin fragment totally abolished thrombus formation as did thecombination of 60 units with DHE.With DHE it was also possible to diminish the low molecular weight heparin dose to 30 units with a good prophylactic effect(7 ?6).There was a small but significant increaseof haemostatic plug formation time in all treatment groups except the one with low molecular weight heparinfragment 30 units anti Xa activity combined withDHE.Thus, by combining low molecular weight heparin withDHE it is possible to prevent thrombus formation without influencing primary haemostasis in rabbits.

Clinical use of heparin as an antithrombotic drug Is limited by the risk of excessive bleeding, generally ascribed to the anticoagulant activity of this mucopolysaccharide.Unexpectedly, some low molecular weight heparins with reduced anticoagulant activity are reported to cause bleeding in clinical and experimental studies. To approach this problem, a number of heparin preparations with various molecular weights were characterized by analysis of their 13C-NMR spectra. The bleeding potential of the same heparins was tested by measuring the "template" bleeding time (BT, a method exploring the mechanisms of primary haemostasis) in the rat tail, 15 min after i.v. administration of 0.75 mg/kg b.w. of the drug.Besides major NMR signals associated with known units of the regular and irregular regions of heparins, some of the preparations display 13C-NMR "extra-signals", which have a higher degree of relative intensity in the spectra of samples with higher haemorrhagic potential; indeed, all the samples exceeding in "extrasignals" a conventional intensity level of 2 had at least 70% prolongation of the BT over controls.These results suggest that specific contaminants, responsible for these13C-NMR "extra-signals", may also be responsible for the unusually high bleeding expressed by some heparin preparations. Work is in progress to identify these components and to evaluate which mechanism of primary haemostasis is involved in their haemorrhagic effect.

We have evaluated the pharmacological efficacy of a concentrated DDAVP preparation (40 μg/mL) (herein referred to as C-DDAVP) administered subcutaneously (s.c.) in mild (n=23) and moderate (n=2) hemophilia A patients. A comparison between the response to s.c. C-DDAVP and the "dilute" DDAVP (4 μg/ml) (administered s.c. in 16 patients with mild (n=13) and moderate (n=3) hemophilia A and i.v. in 18 patients with mild (n=16) and moderate (n=2) was also carried out. In all instances Desmopressin was given at a dose of 0.3 μg/kg b.w. in absence of bleeding. The increase of F. VIII:C (expressed as post/pre ratio) after s.c. C-DDAVP was 2,55 at 30', 3,50 at 60'and 3,21 at 120'. The comparison among the three schedules of DDAVP administration showed that s.c. C-DDAVP elicited an increase of F. VIII :C at least as high as that induced by the dilute DDAVP with differences not statistically significant.C-DDAVP was also administered s.c. in 4 patients with type I vWD (platelet normal subtype) with a normalization of bleeding time 60' after s.c. C-DDAVP, concomitant with a rise of activities related to F. VIII/vWF complex. Side effects were modest and transient. We can conclude that s.c. C-DDAVP is equally effective and safe in comparison with the dilute brand with the advantage of the minimal adminstration volume (<1 mL).

After prolonged stimulation venous endothelium becomes refractory to otherwise efficacious stimuli for tissue plasminogen activator (t-PA) release. When the stimulus is removed, the restitution of the response is observed. The distention of the veins distally from the occlusion site in deep venous thrombosis (DVT) could represent such a chronic stimulus. We studied t-PA release in patients with DVT during 20-min venous occlusion (VO) and DDAVP infusion (0,4 ug/kg b.w. in 10 min). t-PA release was estimated as the difference in euglobulin clot lysis time (ECLT in U) and fibrin plates (FP in mm2) before and after VO (fibrinolytic potential). Two groups of patients were studied: 15 recumbent patients with oneside iliofemoral DVT, and 15 patients with oneside postthrombotic syndrome.In both groups the response of legs was lower than that of arms. The comparison of the healthy and the affected leg in postthrombotic group showed no difference in t-PA release after VO. A higher release of t-PA was seen in the acute DVT group in all three tested limbs, explainable by a restitution of fibrinolytic potential due to the reduction of hydrostatic stimulus. However, the response was slightly lower in the diseased leg. In 5 patients from the acute DVT group DDAVP infusion induced higher t-PA release after VO in both legs compared to VO before DDAVP. We can conclude that hydrostatic pressure in upright position is such a strong stimulus that an additional decrease of t-PA release due to chronic venous stasis cannot be expressed. In recumbent patients DVT hinders the restitution of the response to V0 in the diseased leg. DDAVP seems to act independently of hydrostatic pressure and venous stasis.

In this work we investigated the possible relationship between plasmatic activity and antithrombotic activity of a Low Molecular Weight Heparin Fragment (CY222, Choay-Paris) compared with Heparin (Hep). A randomized administration of both test drugs was carried out on 198 rats receiving 4 different dosages (4,2,1 or 0.5 mg/Kg b.w.) or a placebo solution (normal saline). A ligature of the inferior vena cava was performed on rats at T = OH and test drugs were administered subcutaneously at T - 2H. Blood and thrombus samples were taken at T = 6H. These parameters may use in human Low Molecular Weight Heparin therapy.

Abstract:
The aim of the study was to investigate the effect of aspirin upon the development of occlusive arterial thrombosis in rats of either sex. For this purpose 3 months old male and female Wistar rats were pretreated with three different doses of aspirin: 10, 30 and 60 mg/kg b.w., orally by stomach tube, 3 hours before the thrombosis procedure. Occlusive arterial thrombosis was induced by the method of Hornstra and Vendel-mans-Starrenburg (1973). The dry thrombus weight (TW) and the obstruction time (OT) of the bypass-cannula served as criteria for the degree of the development of thrombosis.The results showed a dose-dependent statistically significant inhibition of arterial thrombosis in male rats by all doses of aspirin studied. TW was decreased 2, 2.5 and 9 times by 10, 30 and 60 mg/kg aspirin respectively. OT was significantly prolonged in male rats by all doses of aspirin studied. Inhibition of arterial thrombosis in female rats was achieved only by the largest dose of aspirin tested - 60 mg/kg: TW was decreased and OT was prolonged. Paradoxically, female rats treated with 10 mg/kg aspirin showed a tendency for augmentation of arterial thrombosis which contrasted to the significantly decreased TW and prolonged OT of male rats given 10 mg/kg aspirin. Female rats treated with 30 mg/kg aspirin showed only a tendency for inhibition of arterial thrombosis without any significant difference for both, TW and OT. The results were interpreted as suggestive the existance of sex differences in cyclooxygenase inhibition by aspirin and/or the prostaglandin system in rats. The data obtained were in agreement with epidemiological observation of uneffectiveness of aspirin treatment of female patients with stroke. They also showed the need for the most appropriate dose of aspirin for clinical purposes which might be effective in females too.

Abstract:
Administering PGI2 instead of heparin (H) during HD enables the risk of bleeding to be reduced. A serious disadvantage of this method of inhibiting coagulation by inhibiting thrombocyte function is however that, in contrast to anticoagu1 ation with H alone, there exists no quickly measurable monitoring parameter. Recently Kratzer and Born (Haemostasis 1985; 15: 357-52) presented a new method for measuring the primary haemostatis: the “in vitro BT”. Our pilot study was to determine whether this “in vitro BT” is concentration - d epe nd ent 1 y prolonged by administering a stable PGI2 analogue during HD. In 5 HD’s the stable P G I 2 analogue CG 4203 (dose gradually raised starting 45 rfnn before HD to 25 ng/kg b.w./min during HD; Gruenenthal GmbH, FRG) were given. Before, during and after the HD the “in vitro BT” (Thrombostat 4000, VDG, FRG) was measured under standardised conditions 15 min after taking 1 ml of citrate blood from the arterial branch of the extracorporeal system. The “in vitro BT” was concentration-dependent!y prolonged by CG 4203. The time interval between taking and measuring the blood sample affect the measured value. If the blood sample is taken directly before the dialyser, the “in vitro BT” is greater than if the sample is taken directly after the dialyser; from which it can be assumed that the dialyser activates the platelets. 1 h after ending HD and discontinuing the CG 4203, the “in vitro BT” is shorter than it was before starting HD; perhaps this is due to platelet activation during HD in the extracorporeal system and to removal of uraemic toxins. Furthermore, the “in vitro BT” was measured in a healthy volunteer 20 times consecutively. The coefficient of variability was 9.3 %. Further studies should be performed to see whether adjusting the “in vitro BT” to a given range allows the dosing of PGI2 or a stable PGI2 analogue to be so controlled as to prevent occlusion in the extracorporeal system.

Abstract:
Purified human FVIIa has been shown to induce haemostasis in pat. with haemoph. A compl. with antibodies against VIII:C (Hedner & Kisiel 1983). The in vitro effect of addition of pVIIa or rVIIa was studied by adding 0, 44, or 180 u pVIIa/rVIIa/ml to haemoph. A or haemoph. B plasma. The APTT shortened from 61 s (mean of 5 determin.) without any Vila added to 38 s after addition of 44 u pVIIa to haemoph. A plasma, and to 33 s after addition of 180 u/ml. In haemoph. B plasma the corresponding APTTs were 64 s, 35 s (44 u pVIIa/ml Vila) and 31 s (180 u pVIIa/ ml). Similar results were obtained when rVIIa was added to the same plasmas. In haemoph. A plasma APTT was shortened from 66 S to 52 s (50 u rVIIa/ml) and to41s(159u rVIIa/ml), and in haemoph. B plasma 71 s,44 s,and 37 s.Also in plasma from pat. with acquired antibodies against VIII:C a shortening of APTT was found both of pVIIa (60 s, 37 s, and 32 s) and rVIIs (65 s, 38 s, and 33 s). In normal plasma the APTT only shortened a few seconds after addition of the same amounts of pVIIa/rVIIa per ml (30 s, 27 s, 25 s). The doses required to normalize the APTT in vitro exceed substantially the doses used in vivo so far. The pVIIa and rVIIa were therefore also given i.v. to rabbits.Plasma samples were drawn before inj., 15 min, 2 hrs,4 hrs, 8 hrs, and 24 hrs after and platelets, APTT, fib.gen, ATIII, α2M, α2AP, ethanol gel. test, FVII,Hb, Hkr were followed. Doses of 500 u/kgb.w.,1000 or 5000 u/kg b.w. were given. No signs of a gen.activ.of the coag. were observed. The FVII level in plasm rose adequately. In conclusion higher doses of Vila than used clin. so far may be needed to achieve full haemost. effect in haemoph. pat. or pat. with acq. antibodies against VIII:C. Such doses also seem to be safe. rVIIa was as active as pVIIa and as safe in rabbits.
