Abstract:
ABSTRACT The role of human leukocyte antigen (HLA) class I supertypes in controlling human immunodeficiency virus type 1 (HIV-1) infection in African Americans has not been established. We examined the effects of the HLA-A and HLA-B alleles and supertypes on the outcomes of HIV-1 clade B infection among 338 African American women and adolescents. HLA-B58 and -B62 supertypes (B58s and B62s) were associated with favorable HIV-1 disease control (proportional odds ratio [POR] of 0.33 and 95% confidence interval [95% CI] of 0.21 to 0.52 for the former and POR of 0.26 and 95% CI of 0.09 to 0.73 for the latter); B7s and B44s were associated with unfavorable disease control (POR of 2.39 and 95% CI of 1.54 to 3.73 for the former and POR of 1.63 and 95% CI of 1.08 to 2.47 for the latter). In general, individual alleles within specific B supertypes exerted relatively homogeneous effects. A notable exception was B27s, whose protective influence (POR, 0.58; 95% CI, 0.35 to 0.94) was masked by the opposing effect of its member allele B*1510. The associations of most B supertypes (e.g., B58s and B7s) were largely explained either by well-known effects of constituent B alleles or by effects of previously unimplicated B alleles aggregated into a particular supertype (e.g., B44s and B62s). A higher frequency of HLA-B genotypic supertypes correlated with a higher mean viral load (VL) and lower mean CD4 count (Pearson's r = 0.63 and 0.62, respectively; P = 0.03). Among the genotypic supertypes, B58s and its member allele B*57 contributed disproportionately to the explainable VL variation. The study demonstrated the dominant role of HLA-B supertypes in HIV-1 clade B-infected African Americans and further dissected the contributions of individual class I alleles and their population frequencies to the supertype effects.

Abstract:
The root is the main site of nitrogen (N) acquisition and aluminum (Al) toxicity. The objective of this study is to investigate whether liming and cultivation of an Al-tolerant rice (Oryza sativa L.) cultivar can improve root growth, thereby increasing N acquisition by rice plants in acid paddy soil. Two rice cultivars (‘B690’, Al-sensitive, and ‘Yugeng5’, Al-tolerant) were cultivated with 15N-labeled urea, and with or without lime in an acid paddy soil (pH 4.9) in pots. We examined root and shoot growth, soil pH, soil exchangeable Al, N uptake, 15N distribution in plant-soil system, and fertilizer N recovery efficiency. Results showed that liming improved the root growth of ‘B690’ by decreasing soil exchangeable Al concentrations, in both N-limited and N-fertilized soils. Liming enhanced the N uptake of ‘B690’ only in the absence of N fertilizer. The root weight of ‘Yugeng5’ was greater than that of ‘B690’ without lime, but the two cultivars showed similar N uptake. The fertilizer N recovery efficiency and N loss did not differ significantly between limed and non-limed conditions, or between the two rice cultivars. Thus, liming an Al-sensitive rice cultivar and cultivating an Al-tolerant one improves root growth, but does not enhance fertilizer N recovery efficiency in the present acid paddy soil.

Abstract:
e16551 Background: Chowell et al. showed that HLA (human leukocyte antigen) class I (HLA-I) heterozygosity may avail to survival benefit from immune checkpoint blockade (ICB) in advanced cancer patients. Among their cohorts of melanoma patients, patients with the HLA-B44 supertype had improved survival, while the individuals with HLA-B62 supertype had worse outcome. However, these observations were not supported by another study of NSCLC patients treated with immune checkpoint inhibitors (ICIs). Now whether HLA-I genotype can serve as predictive marker for ICB treatment of other malignancies such as gastrointestinal cancer remains unclear. Methods: Tumor tissue samples before immunotherapy as well as related clinical data were collected from 91 Chinese gastrointestinal cancer patients receiving anti-PD-1/PD-L1 inhibitors and/or anti-CTLA-4 antibodies. Genomic features of tumor specimens were characterized by whole exome sequencing (WES) and HLA-I genotyping of normal DNA from peripheral blood cells were performed using WES data. Tumor immune microenvironment signatures were examined by RNA-sequencing targeting 395 immune related genes. Results: We observed that the individuals with HLA-B62 supertype (n = 15) had a significantly shorter PFS (median PFS, 1.83 months versus 3.53 months; HR, 2.3; 95% CI, 1.03-5.0; p= 0.009) than that of those without HLA-B62 supertype (n = 35) in a subgroup of patients with microsatellite stability (MSS, n = 50) and it was not so in other patients with microsatellite instability-high (MSI-H, n = 17) or unknown MSI status (n = 24). Moreover, the patients with B62 supertype and TP53 mutations (n = 15) exhibited more markedly shortened PFS (mPFS, 1.8 months versus 5.57 months; HR, 3.9; 95% CI, 1.5-9.6; p< 0.0001) and OS (mOS, 4.2 months versus 17.3 months; HR, 4.5; 95% CI, 1.5-13.2; p< 0.0001) compared to those patients with non-B62 supertype and TP53 wildtype (n = 26). Furthermore, gene expression levels of certain immune activation related pathways or markers (IFN-γ signature, cytolytic marker, etc.) in the patients with B62 supertype and without B44 supertype (n = 23) were statistically lower ( p< 0.05) than in those patients without B62 supertype (n = 62). Chi-square test revealed no correlation between HLA-B62 supertype and TP53 mutation, MSI status, tumor mutation burden (TMB) or PD-L1 expression. Conclusions: HLA-B62 supertype in combination with TP53 mutations may define a subset of gastrointestinal cancer patients who probably fail to benefit from ICIs, especially in MSS patients.

Abstract:
Abstract Rejection of a graft after human leukocyte antigen (HLA)-identical stem cell transplantation (SCT) can be caused by recipient's immunocompetent T lymphocytes recognizing minor histocompatibility antigens on donor stem cells. During rejection of a male stem cell graft by a female recipient, 2 male (H-Y)–specific cytotoxic T lymphocyte (CTL) clones were isolated from peripheral blood. One CTL clone recognized an HLA-A2–restricted H-Y antigen, encoded by the SMCY gene. Another CTL clone recognized an HLA-B60–restricted H-Y antigen. In this study UTY was identified as the gene coding for the HLA-B60–restricted H-Y antigen. The UTY-derived H-Y antigen was characterized as a 10-amino acid residue peptide, RESEEESVSL. Although the epitope differed by 3 amino acids from its X-homologue, UTX, only 2 polymorphisms were essential for recognition by the CTL clone HLA-B60 HY. These results illustrate that CTLs against several H-Y antigens derived from different proteins can contribute simultaneously to graft rejection after HLA-identical, sex-mismatched SCT. Moreover, RESEEESVSL-specific T cells could be isolated from a female HLA-B60+ patient with myelodysplastic syndrome who has been treated with multiple blood transfusions, but not from control healthy HLA-B60+ female donors. This may indicate that RESEEESVSL-reactive T cells are more common in sensitized patients.

Abstract:
ABSTRACT Human immunodeficiency virus type 1 (HIV-1)-specific cytotoxic T-lymphocyte (CTL) responses play a major role in the antiviral immune response, but the relative contribution of CTL responses restricted by different HLA class I molecules is less well defined. HLA-B60 or the related allele B61 is expressed in 10 to 20% of Caucasoid populations and is even more highly prevalent in Asian populations, but yet no CTL epitopes restricted by these alleles have been defined. Here we report the definition of five novel HLA-B60-restricted HIV-1-specific CTL epitopes, using peripheral blood mononuclear cells in enzyme-linked immunospot (Elispot) assays and using CTL clones and lines in cytolytic assays. The dominant HLA-B60-restricted epitope, Nef peptide KEKGGLEGL, was targeted by all eight subjects with B60 and also by both subjects with B61 studied. This study additionally establishes the utility of the Elispot assay as a more rapid and efficient method of defining novel CTL epitopes. This approach will help to define new CTL epitopes that may play an important role in the immune control of HIV-1.

Abstract:
1. B31 and B32 are pattern-initiator neurons in the buccal ganglia of Aplysia. Along with the B61/B62 neurons, B31/B32 are also motor neurons that innervate the 12 buccal muscle via the I2 nerve. This research was aimed at determining the physiological functions of the B31/B32 and B61/B62 neurons, and of the I2 muscle. 2. Stimulating the I2 muscle in the radula rest position produces radula protraction. In addition, in behaving animals lesioning either the muscle or the I2 nerve greatly reduces radula protraction. 3. During buccal motor programs in reduced preparations, B31/B32 and B61/62 fire preceding activity in neuron B4, whose firing indicates the onset of radula retraction. In addition, during both ingestion-like and rejection-like patterns the activity in the I2 nerve is correlated with protraction. 4. B31/B32 fire at frequencies of 15-25 Hz. Neither B31/B32 nor B61/B62 elicit facilitating end-junction potentials (EJPs) and electromyograms (EMGs) in the I2 muscle. EMGs from B31/B32 are smaller than those from B61/B62. B31/B32 and B61/B62 innervate all areas of the muscle approximately uniformly. 5. In behaving animals, EMGs consistent with B31/B32 activity are seen in the I2 muscle during the protraction phase of biting, swallowing, and rejection movements. In addition, the I2 muscle receives inputs that cannot be attributed to either the B31/B32 or B61/B62 neurons, either because the potentials are too large, firing frequencies are too low, or a prominent facilitation is seen. Such potentials are associated with lip movements, and also with radula retraction. 6. EMGs were recorded from the I2 muscle during feeding behavior after a lesion of the I2 nerve. Animals that had severe deficits in protraction showed no activity consistent with B31/B32 or B61/B62, but did show activity during retraction. 7. Our data indicate that the I2 muscle and the B31/B32 motor neurons are essential constituents contributing to protraction movements. Activity in these neurons is associated with radula protraction, which occurs as a component of a number of different feeding movements. The I2 muscle may also contribute to retraction, via activation by other motor neurons.

Abstract:
This paper presents a comparative analysis of the diesel engine performance and emission characteristics, when operating on diesel fuel and various diesel-biodiesel (B10, B20, B40, B60) blends, at various loads and engine speeds. The experimental tests were performed on a four-stroke, four-cylinder, direct injection, naturally aspirated, 60 kW diesel engine D-243. The in-cylinder pressure data was analysed to determine the ignition delay, the Heat Release Rate (HRR), maximum in-cylinder pressure and maximum pressure gradients. The influence of diesel-biodiesel blends on the Brake Specific Fuel Consumption (bsfc) and exhaust emissions was also investigated. The bench test results showed that when the engine running on blends B60 at full engine load and rated speed, the autoignition delay was 13.5% longer, in comparison with mineral diesel. Maximum cylinder pressure decreased about 1–2% when the amount of Rapeseed Methyl Ester (RME) expanded in the diesel fuel when operating at full load and 1400 min–1 speed. At rated mode, the minimum bsfc increased, when operating on biofuel blends compared to mineral diesel. The maximum brake thermal efficiency sustained at the levels from 0.3% to 6.5% lower in comparison with mineral diesel operating at full (100%) load. When the engine was running at maximum torque mode using diesel – RME fuel blends B10, B20, B40 and B60 the total emissions of nitrogen oxides decreased. At full and moderate load, the emission of carbon monoxide significantly raised as the amount of RME in fuel increased.

Abstract:
e21512 Background: Anti-PD-1 therapy has improved the outcome of advanced melanoma patients with a 5-year survival rate of about 40-45%. However, biomarkers predictive of response to immune checkpoint blockade therapy are lacking. There is limited data on the utility of host germline human leucocyte antigen (HLA) genotype as a predictor of response to anti-PD-1 therapy in advanced melanoma. Here, we investigate the prognostic value of HLA in predicting survival outcomes of patients with unresectable locally advanced, metastatic melanoma on anti-PD-1 therapy. Methods: Blood was collected from 113 metastatic melanoma patients who were treated with anti-PD-1 therapy at two major oncology centres in Western Australia. High quality DNA was extracted from white blood cells and subsequently HLA-I and HLA-II typed using clinically validated assay. Univariate analyses were conducted using Cox regression model correlating homozygosity at HLA-I and HLA-II loci with overall survival (OS). HLA-A and HLA-B were classified into 12 supertypes and correlated with OS. Multivariate analyses were performed while controlling for age, gender, prior therapy, BRAF mutation status, ECOG performance status and presence of liver and brain metastases. Results: Homozygosity at HLA-I or HLA-II loci was not associated with OS. However, the absence of HLA-B62 supertype was associated with a trend towards improved OS (HR: 0.53 [95% CI:0.25-1.10]; P = 0.09) as reported previously. Notably, the absence of HLA-B27 supertype was associated with improved OS which was statistically significant (HR: 0.45 [95% CI:0.24-0.85]; P = 0.01). In multivariate analyses, the prognostic value of HLA-B27 supertype (HR: 0.38 [95% CI:0.19-0.76]; P = 0.006) was maintained, whereas the prognostic value of HLA-B62 supertype significantly improved (HR: 0.42 [95% CI:0.19-0.94]; P = 0.03). Conclusions: Our results suggest a limited role of HLA homozygosity in predicting survival of melanoma patients treated with anti-PD-1 therapy. However, we identified that the absence of HLA-B62 and HLA-B27 supertype is associated with improved survival benefit. Therefore, HLA-B27 and HLA-B62 supertype may be used as adjunct biomarkers of response to anti-PD-1 therapy in patients with melanoma in addition to PD-L1 status, pending validation in prospective randomised clinical trials.

Abstract:
Abstract Exopolysaccharides (EPS) is a type of polysaccharide produced by lactic acid bacteria (LAB) that can be directly used in foods to make the products more excellent. Therefore, batch studies were performed to explore the effect of different LAB on the production of EPS and antioxidant activity. Five strains with high EPS yield and antioxidant activity were screened out from 66 strains isolated from Tibetan dairy products. The results show that EPS produntion of the five strains (B55, B62, B30, 7830 and K2) were 110.66, 145.48, 132.78, 122.11 and 111.72 mg·L−1, respectively, and they have a higher DPPH free radical scavenging activity (56.29, 66.43, 62.94, 68.71, 61.87%). Five LAB strains were identified and classified based on screening, purification and 16S rDNA sequences. Molecular characterization based on partial sequence 16S rDNA homology confirmed the initial identification as Lactobacillus fermentum (B55, B62), Lactobacillus plantarum (7830), Pediococcus acidilactici (B30) and Lactobacillus helveticus (K2).

Abstract:
Endothelial dysfunction in ischemic acute renal failure (IARF) has been attributed to both direct endothelial injury and to altered endothelial nitric oxide synthase (eNOS) activity, with either maximal upregulation of eNOS or inhibition of eNOS by excess nitric oxide (NO) derived from iNOS. We investigated renal endothelial dysfunction in kidneys from Sprague-Dawley rats by assessing autoregulation and endothelium-dependent vasorelaxation 24 h after unilateral (U) or bilateral (B) renal artery occlusion for 30 (U30, B30) or 60 min (U60, B60) and in sham-operated controls. Although renal failure was induced in all degrees of ischemia, neither endothelial dysfunction nor altered facilitation of autoregulation by 75 pM angiotensin II was detected in U30, U60, or B30 kidneys. Baseline and angiotensin II-facilitated autoregulation were impaired, methacholine EC50 was increased, and endothelium-derived hyperpolarizing factor (EDHF) activity was preserved in B60 kidneys. Increasing angiotensin II concentration restored autoregulation and increased renal vascular resistance (RVR) in B60 kidneys; this facilitated autoregulation, and the increase in RVR was abolished by 100 μM furosemide. Autoregulation was enhanced by Nω-nitro-l-arginine methyl ester. Peri-ischemic inhibition of inducible NOS ameliorated renal failure but did not prevent endothelial dysfunction or impaired autoregulation. There was no significant structural injury to the afferent arterioles with ischemia. These results suggest that tubuloglomerular feedback is preserved in IARF but that excess NO and probably EDHF produce endothelial dysfunction and antagonize autoregulation. The threshold for injury-producing, detectable endothelial dysfunction was higher than for the loss of glomerular filtration rate. Arteriolar endothelial dysfunction after prolonged IARF is predominantly functional rather than structural.

Abstract:
ABSTRACT The formation of seedlings is one of the most important phases of lettuce cultivation. Therefore, any strategy that aims to obtain high-quality seedlings can increase productivity. One of these strategies is the prediction of morphophysiological attributes based on optical properties. The objective of this study was to quantitatively estimate the biometric variables of lettuce from parametric and non-parametric models based on the response of multispectral camera images. The experiment was conducted in a greenhouse in the municipality of Uberaba, Minas Gerais State, Brazil. Twenty days after sowing, multispectral images of the plants were captured using a MAPIR Survey 3 camera. To compose the estimation models, along with the original bands of the camera, the multispectral vegetation indices were calculated using the calibrated original camera bands. Bands B550, B660, and B850 and the near-infrared indices contributed significantly to estimating the physiological variable models, with B850 contributing the most to the biometric and nutritional variables. From the near-infrared band (B850) and derived indices, it was possible to estimate all the agronomic variables from the models generated by the M5 algorithm, with an accuracy of up to 1.6% for the maximum quantum yield. Thus, it is possible to quantify the biometric, physiological, and nutritional variables of lettuce using a multispectral camera. Among the Mapir camera bands, B660 exhibited the greatest variability, showing that the red range was the most sensitive.

Abstract:
AbstractGrinding is widely known for its low material removal rates and high surface quality. However, recent developments in production processes for cubic boron nitride (CBN) abrasive grains have led to commercially available grain sizes larger than 300 µm. These superabrasive CBN-grains allow higher material removal rates during grinding of hardened steel components. Currently, these components are pre-machined with turning processes before hardening and finishing the work piece by grinding. However, the turning process can be eliminated by grinding with coarse CBN-grains since higher depths of cut are achievable when machining hardened components. This paper explores the limits of grinding wheels using grains with a size of B602 during soft and hard machining in comparison to conventional B252 grains. It is shown that the use of coarser grains leads to lower process forces, higher (tensile) residual stress and higher surface roughness. Residual stress and surface roughness are of less importance as these grains are to be used mainly in roughing operations with ensuing finishing operations for the required surface properties. Over all investigations, especially in hard machining, neither grain nor tool wear was observed for the B602 grains, whereas the B252 tool was severely clogged during the experiments. Additionally, the grinding force ratio indicates that the coarse grain tools have not yet reached their productivity limit as it increases over all investigated feeds. This indicates improving tool performance with lower amounts of rubbing for increasing feed rate during hard grinding and shows the potential for the industrial use of higher feed rates with larger grains.

Abstract:
Photoluminescence (PL) and reflectivity spectroscopy in magnetic fields up to 60 Tesla are used to investigate the excitonic optical transitions within CdTe/(Cd,Mg)Te 2D electron gases. The samples are 100 A and 200 A wide quantum well (QW) wherein the electron density is varied smoothly from 1 × 1011 to 4.5 × 1011 cm-3 across the wafer. This allows us to follow the energy diagram of the neutral exciton ( X 0) and the charged exciton singlet and triplet spin states ( X - s , X - t ) versus magnetic field and the electron density. We observe a weak PL peak above 12 T (15 T) for the 200 A QW (100 A) at the low energy side of the spectra in addition to the X -0 and X - s PL. This peak could be the signature of a fundamental dark triplet state recently predicted (Phys. Rev. B62, 4630). Moreover, for high density samples a balance of PL intensity between the X -s and an other X -t occurs in the 25T-30T range. These results are interpreted on the basis of the calculations by Wojs et al. (Phys. Rev. B62 4630) taking account the Zeeman energy of the charged exciton.
