Abstract:
The Babi religion had, during its brief life, a tremendous impact on Iranian society. Its founder, Sayyid $ sp{ rm c}$Ali Muhammad, the Bab (1819-1850) wrote a great many works of several kinds. Of his major writings, the two earliest are partial commentaries on the Qur'an. The following study examines these two remarkably different commentaries in an attempt to appreciate the Bab's attitudes towards the Qur'an, Islam, and himself. The earliest work, the Tafsi r surat al-baqara, was written before the Bab had publicized a claim to messiahship. In the Tafsi r surat Yusuf, written only a short time later, this claim is made explicit. The radical difference in the style of the two commentaries, which may be seen as a reflection of a development in the Bab's perception of himself, is analyzed.

Abstract:
Depuis une dizaine d’années, l’auto-assemblage induit par polymérisation (PISA) s’est révélé être un outil efficace pour la préparation de nanoparticules à base de copolymères amphiphiles à des taux de solide élevés, aussi bien dans des solvants organiques que dans l’eau. Jusqu’à présent, la plupart des systèmes étudiés concernaient les copolymères diblocs AB (dans lesquels A = bloc solvophile stabilisant, et B = bloc solvophobe). Un autre type de copolymères à blocs intéressant est le tribloc associatif BAB. Peu d’études décrivent leur synthèse par PISA et les polymérisations sont réalisées dans des mélanges alcool/eau. Dans ce travail de thèse, et pour la première fois, la synthèse par voie radicalaire contrôlée de copolymères triblocs BAB par PISA a été réalisée dans l’eau pure et une étude approfondie du système a été menée. Un macro-agent RAFT hydrophile symétrique à base de poly(acrylamide de N,N-diméthyle) possédant une unité acide benzoïque au centre, et le diacétone acrylamide (blocs B) ont été sélectionnés pour étudier ce système. Des particules de différentes morphologies (sphères, fibres et vésicules) ont pu être obtenues avec de bons contrôles des polymérisations. Nous avons montré que le degré d’ionisation de l’unité centrale acide benzoïque du bloc stabilisant jouait un rôle crucial dans la stabilité du système et la morphologie obtenue. Le mécanisme de formation de ces copolymères triblocs au cours de la PISA a également été étudié. Enfin, nous avons montré qu’il était possible de former des réseaux dynamiques de copolymères BAB – via un procédé one-pot entièrement dans l’eau – menant à la formation de systèmes thermosensibles avec une viscosité modulable
Over the past decade, the polymerization-induced self-assembly (PISA) has become an efficient tool for the preparation of block-copolymer nanoparticles at high solid contents, in both organic solvents and water. Nonetheless, most of the studied systems are currently based on simple AB diblock copolymers (where A = solvophilic, stabilizer block and B = solvophobic block). Another interesting class of block copolymers are associative BAB triblocks. To the best of our knowledge, very few studies report their synthesis by PISA and the polymerizations are performed in mixtures of alcohol and water. In this work, for the first time, a straightforward strategy to synthesize, in pure water, BAB triblock copolymers through the use of reversible deactivation radical polymerization is developed and studied in details. To this end, an hydrophilic, bifunctional macroRAFT agent of poly(N,N-dimethylacrylamide) with a central benzoic acid group was used in the aqueous dispersion polymerization of diacetone acrylamide. Various morphologies (spheres, fibers and vesicles) could be obtained with good polymerization controls. Moreover, we showed that the particle morphology and colloidal stability strongly depend on the degree of ionization of the central charge in the stabilizer loop. Then, the mechanism of formation of these triblock copolymers during PISA was investigated. Finally, we showed the possibility to form dynamic networks of BAB copolymers – using a one-pot process entirely in water – leading to the formation of thermoresponsive copolymer networks with tunable viscosity

Abstract:
In this work new fluorinated and non-fluorinated mono- and bifunctional trithiocarbonates of the structure Z-C(=S)-S-R and Z-C(=S)-S-R-S-C(=S)-Z were synthesized for the use as chain transfer agents (CTAs) in the RAFT-process. All newly synthesized CTAs were tested for their efficiency to moderate the free radical polymerization process by polymerizing styrene (M3). Besides characterization of the homopolymers by GPC measurements, end- group analysis of the synthesized block copolymers via 1H-, 19F-NMR, and in some cases also UV-vis spectroscopy, were performed attaching suitable fluorinated moieties to the Z- and/or R-groups of the CTAs. Symmetric triblock copolymers of type BAB and non-symmetric fluorine end- capped polymers were accessible using the RAFT process in just two or one polymerization step. In particular, the RAFT-process enabled the controlled polymerization of hydrophilic monomers such as N-isopropylacrylamide (NIPAM) (M1) as well as N-acryloylpyrrolidine (NAP) (M2) for the A-blocks and of the hydrophobic monomers styrene (M3), 2-fluorostyrene (M4), 3-fluorostyrene (M5), 4-fluorostyrene (M6) and 2,3,4,5,6-pentafluorostyrene (M7) for the B-blocks. The properties of the BAB-triblock copolymers were investigated in dilute, concentrated and highly concentrated aqueous solutions using DLS, turbidimetry, 1H- and 19F-NMR, rheology, determination of the CMC, foam height- and surface tension measurements and microscopy. Furthermore, their ability to stabilize emulsions and microemulsions and the wetting behaviour of their aqueous solutions on different substrates was investigated. The behaviour of the fluorine end-functionalized polymers to form micelles was studied applying DLS measurements in diluted organic solution. All investigated BAB-triblock copolymers were able to form micelles and show surface activity at room temperature in dilute aqueous solution. The aqueous solutions displayed moderate foam formation. With different types and concentrations of oils, the formation of emulsions could be detected using a light microscope. A boosting effect in microemulsions could not be found adding BAB-triblock copolymers. At elevated polymer concentrations, the formation of hydrogels was proved applying rheology measurements.
Im Rahmen dieser Arbeit wurden neue fluorierte und unfluorierte mono- und bifunktionelle Trithiocarbonate der Typen Z-C(=S)-S-R und Z-C(=S)-S-R-S-C(=S)-Z zur Anwendung als CTAs (chain- transfer agents) im RAFT-Polymerisationsverfahren hergestellt. Alle CTAs wurden erfolgreich auf ihre Effizienz zur Steuerung des radikalischen Polymerisationsverfahrens hin durch Polymerisation von Styrol (M3) getestet. Neben GPC-Messungen wurden Endgruppenanalysen der synthetisierten Blockcopolymere mittels 1H-, 19F-NMR und in manchen Fällen auch UV-Vis Spektroskopie durchgeführt. Dazu wurden die Z- und/oder R-Gruppen der CTAs mit geeigneten fluorierten Gruppen versehen. Durch Anwendung des RAFT Verfahrens konnten symmetrische Triblockcopolymere vom Typ BAB bzw. mit einer Fluoralkylgruppe endgecappte unsymmetrische Polymere in nur zwei bzw. einem Polymerisationsschritt hergestellt werden. Das RAFT- Polymerisationsverfahren ermöglicht sowohl die Polymerisation hydrophiler Monomere wie N-Isopropylacrylamid (NIPAM) (M1) oder N-Acryloylpyrrolidin (NAP) (M2) für die A-Blöcke als auch der hydropoben Monomere Styrol (M3), 2-Fluorostyrol (M4), 3-Fluorostyrol (M5), 4- Fluorostyrol (M6) und 2,3,4,5,6- Pentafluorostyrol (M7) für die B-Blöcke. Die Eigenschaften der Blockcopolymere in verdünnten, konzentrierten und hochkonzentrierten wässrigen Lösungen wurden mittels DLS, Trübungsphotometrie, 1H- und 19F-NMR, Rheologie, CMC- sowie Schaumhöhen- und Oberflächenspannungsmessungen und Lichtmikroskopie untersucht. Weiterhin wurden ihre Eigenschaften als Emulgatoren und in Mikroemulsion untersucht. Das Micellbildungsverhalten der hydrophob endfunktionalisierten Polymere wurde mittels DLS Messungen in verdünnter organischer Lösung untersucht. Alle untersuchten BAB-Triblöcke bildeten Micellen und zeigten Oberflächenaktivität bei Raumtemperatur in verdünnter, wässriger Lösung. Weiterhin zeigen die wässrigen Lösungen der Polymere mäßige Schaumbildung. Mit verschiedenen Öltypen und Ölkonzentrationen wurden Emulsionen bzw. Mikroemulsionen gebildet. In Mikroemulsion wurde durch Zugabe von BAB-Triblockopolymeren kein Boosting-Effekt erzielt werden. Bei Untersuchung höherer Polymerkonzentrationen wurde die Bildung von Hydrogelen mittels rheologischer Messungen nachgewiesen. Verschiedene Substrate konnten benetzt werden. Die hydrophob endgecappten Polymere bilden in verdünnter organischer Lösung Micellen, die mittels DLS untersucht wurden, und zeigen somit Tensidverhalten in nichtwässriger Lösung.

Abstract:
This PhD research investigates the role of television in representing the past and constructing an idealized society using a case study of a phenomenal Ramadan drama series, Bab el-Hara. The television drama, a Syrian production, was funded by the pan-Arab satellite conglomerate, the MBC group, and it is set in a fictitious Damascus of the 1930s under the French Mandate. The series, airing its seventh season in Ramadan, 2015, succeeded in achieving pan-Arab fame and gave a boost to the “Damascene Milieu” drama genre. The study approaches this television phenomenon ethnographically, looking at the fiction's implicatedness in the everyday life of viewers and makers in Damascus and in Beirut, through a multi-sited approach investigating content, context and agency, engaging in questions on space, morality and patriotism. The objective is to investigate audiences, text and makers as distinct yet connected sites of meaning. This context based analysis of Bab al-Hara takes place against the backdrop of 2010/2011; the liminal state of a Levant entering deeper into a complex local, regional and international power struggle. The everyday life of Bab al-Hara’s viewers was characterized by a general sense of loss and mistrust, and an unclear and threatened future. Contrastingly, Bab al-Hara provided the nostalgic promise of ontological security, grounded as it was in the courtyard houses of Old Damascus. The Damascene courtyard house constituted the spatial anchor for an idealized moral past, an ahistorical Damascusfocused Arab cultural history, and an imagination of the domestic as sovereign. It thus promoted a view of the neighbourly, the city and the country as a system based on kin, or the family, as the frame in which to understand the collectivity. Bab al-Hara's cultural, moral and spatial telos, a fusion of religious and nationalist worldviews, amongst others, is negotiated by Bab al-Hara’s viewers. The older generation, with situated experience of the social relations during the 1930s, and the younger generation that is appreciative of the virility of the “real” Bab al-Hara man that they no longer encounter in their everyday life. The multiple generational readings in regard to the absent idealized strength and authority, became a dominant reading in relation to chastity and unity as two idealized values that are necessary to conserve, but that are facing serious challenges in the everyday. Bab al-Hara idealizes a moral domestic society that is set in the past and it aims to advance a discourse on unity and patriotism. In so doing, however, it only exposes the weakness of the national project. The Syrian social upheaval in 2011 shows how unity and patriotism as the binaries to sectarianism and treason, have not succeeded in protecting the inner domain of the house from external invasions or internal divisions. In fact, accusations of treason, instead of forcing the outsider to the outside and building solidarity within, accentuates mistrust between the insiders and reveals the power and the limits of t h e Bab al-Hara imaginary of a kin based collectivity, and the omnipresence of imperialism.

This study focuses on teacher s training in early childhood education, whose search problem lies in knowing the role of initial training in the reconstruction process of these professional identities, investigating the function of this training in the reconstruction of social representations of children education s future teachers about their profession. We developed a study based on qualitative research approach, whose theoretical and methodological framework was based on the Theory of Social Representations of Moscovici (2009). The data collection technique adopted was the application of questionnaires to 57 new students and 46 graduates of Pedagogy course of a private university in S?o Paulo state, among which we selected five graduating students to participate in semistructured interviews. The collected data were analyzed based on the identification of the following categories: teacher identity, conception of teacher and teacher training s challenges. The results indicated that this training course seems to have provided recognition to the early childhood s teacher as a professional one, with specific knowledge for its actions and, therefore, who needs to be professionally and socially recognized by the importance of its function. The investigation underscored how important is to rethink childhood s teacher identity in terms of professional recognition and its social, political and professional role. We value the contributions of this training course to the review of concepts and representations that interfere with this identity reconstruction and we realize that challenges are made to improve initial training in this course, such as internship, which needs to articulate the theory and promote more critical thoughts about the foundational concepts of the area; the need for deeper analysis of the curriculum to enable review of conceptions, allowing redraftings that deconstruct socially crystallized representations. Finally, by placing emphasis on these issues, we believe we are contributing to Educational research and, more specifically, to the identity formation of Childhood s Teachers, who belongs to a professional group entitled to exercise its teacher profession and the social function that historically characterizes it.
Este estudo aborda a forma??o de professores de educa??o infantil, cujo problema de pesquisa situa-se em conhecer o papel da forma??o inicial no processo de reconstru??o de sua identidade profissional, objetivando investigar o papel dessa forma??o na reconstru??o das representa??es sociais de futuras professoras de educa??o infantil sobre a profiss?o. Desenvolvemos um estudo baseado na abordagem qualitativa de pesquisa, cujo referencial te?rico-metodol?gico pautou-se na Teoria das Representa??es Sociais de Moscovici (2009). As t?cnicas de coleta de dados adotadas foram a aplica??o de 57 question?rios a alunos ingressantes e 46 concluintes de um curso de pedagogia de uma universidade particular do Estado de S?o Paulo, dentre os quais selecionamos cinco alunas concluintes para participarem da realiza??o de entrevistas semi-estruturadas. Os dados coletados foram analisados a partir da identifica??o das seguintes categorias de an?lise: identidade docente, concep??o de professor e desafios para a forma??o docente. Os resultados obtidos indicaram que esse curso de forma??o parece ter proporcionado o reconhecimento do professor da educa??o infantil enquanto pertencente ? categoria profissional professor, com saberes espec?ficos para sua atua??o e que, portanto, necessita ser reconhecido profissional e socialmente pela import?ncia de sua fun??o. A investiga??o ressaltou a necessidade de se repensar a identidade desse professor em termos de reconhecimento da profiss?o docente e do seu papel social, pol?tico e profissional. Destacamos as contribui??es desse curso de forma??o no que se refere ? revis?o de conceitos e representa??es que interferem nessa reconstru??o identit?ria e pudemos perceber que desafios est?o postos para aperfei?oar a forma??o inicial nesse curso, como a quest?o dos est?gios, que precisam articular a teoria e promover reflex?es mais cr?ticas sobre as concep??es fundantes da ?rea; a necessidade de uma an?lise mais aprofundada do curr?culo, a fim de permitir revis?es das concep??es, possibilitando reelabora??es que desconstruam representa??es cristalizadas socialmente. La zone d'etude est situee au front de la dorsale calcaire, a l'est du village de bab-taza. Elle est composee de formation calcaires, greseuses et marneuses, charriees les unes sur les autres. Cette zone connait de serieux problemes a cause des mouvements de terrain dus aux facteurs de declenchement et de site-soulevement orogenique, pluie, densite humaine, geologie (mineralogie, granulometrie, structure, etc. . . ) et morphologie (pente, concavite horizontale. . . ). Les types de ces mouvements sont differents selon les secteurs: translationnels a l'echelle de la montagne, rotationnels quand ils se declenchent a partir du sommet de celle-ci et rotationnels ou sous forme de coulees quand ils de declenchent dans les bassins et les cloisons. A partir des donnees ci-dessus, nous avons realise la carte de risque ; elle est une synthese des cartes de: localisation, susceptibilite des secteurs, intensite et d'alea. La carte de risque vise a attirer l'attention sur la fragilite du milieu
The studied zone is located on the calcareous-front-dorsal at east of the village bab-taza. The zone is cutted by the calcareous, sandstones and marls formation. This zone has serious problems due to earth movement. Which come from release factor and site-orogenic raising, rain, human density, geology (mineralogy, granulometric, structurs, and so on) morphology (slope, horizontal concavity)-, are differents according to the area, they are only translational or rotational at mountain scale, but in the basins and in the partitions rotational movements and also mudflow take place

Afin de mieux comprendre (i) les mécanismes moléculaires régissant le contrôle de l'expression des gènes impliqués dans le développement et (ii) les réseaux génétiques s'établissant en réponse à des processus de signalisation cellulaire, de plus en plus d'études s'intéressent aux éléments régulateurs en cis des gènes. C'est dans ce cadre que s'inscrit cette thèse intitulée "Contrôle de l'expression transcriptionnelle des gènes bric-à-brac lors de la morphogenèse des appendices chez le modèle drosophile". Le locus bric-à-brac (bab) chez Drosophila melanogaster se compose de deux gènes paralogues, bab1 et bab2, codant pour des protéines nucléaires à domaine BTB, et qui (i) sont exprimés dans les disques imaginaux de pattes et d'antenne selon des patrons très similaires et (ii) sont requis pour la segmentation normale des parties distales des pattes et des antennes, avec un rôle prédominant pour bab2. Au cours de ma thèse, je me suis intéressé aux mécanismes moléculaires contrôlant l'expression de bab2 dans les disques imaginaux de pattes et d'antenne, selon 4 ou 2 anneaux concentriques, respectivement. A partir (i) de constructions transgéniques exprimant un gène rapporteur GFP et insérées dans un site spécifique et (ii) d'un test de sauvetage phénotypique d'un mutant dépourvu de fonction bab, par un fragment d'ADN génomique (BAC) sauvage ou muté par recombineering, j'ai montré que l'expression de bab2 dans les disques de pattes et d'antenne dépend d'un module cis-régulateur (CRM) unique de 0,56 kilobases, que nous avons appelé LAE, pour "Leg and Antennal Enhancer". Par ailleurs, j'ai pu montrer que le LAE de D. Melanogaster est interchangeable avec la région équivalente de D. Virilis, révélant ainsi une conservation fonctionnelle chez ces deux espèces de drosophilidae séparées de 60 millions d'années. Moyennant des délétions et des mutations dirigées (en "Linker scanning") du LAE, j'ai identifié des motifs d'ADN agissant positivement ou négativement soit dans tout le domaine d'expression de bab2 ou seulement dans des sous-domaines (proximaux ou distaux), et ceci de manière similaire dans les disques de pattes et d'antenne. Enfin, par des expériences de perte et de gain de fonction in vivo, ainsi que des analyses biochimiques in vitro, j'ai montré (i) que la protéine à homéodomaine Distal-less (Dll) s'associe à une région conservée de l'enhancer LAE pour activer directement bab2 au niveau de tous les anneaux, (ii) tandis que la protéine à doigt de zinc Rotund lie une région non-strictement conservée chez les Drosophilidae, seulement nécessaire pour une activation maximale dans les anneaux proximaux
A proper spatio-temporal expression of bric-a-brab2 is required for the proximo-distal (PD) patterning of the Drosophila leg and antenna. Here, we deciphered the molecular mechanisms controlling bab2 expression in developing distal leg (4 rings) and antennal (2 rings) tissues. Using site-targeted GFP reporter assay and BAC recombineering, we show that restricted bab2 expression relies on a single 0. 56 kb cis-regulatory module (CRM), which is necessary and sufficient to drive normal Bab2 function in both developing leg and antenna. Through deletion and site-directed mutagenesis approaches, we identified discrete DNA motifs acting either positively or negatively. Finally, loss- and gain-of-function genetic analyses in combination with in-vitro electrophoretic mobility shift assays established that the Distal-less homeodomain protein directly activates bab2 in all rings, whereas the Rotund C2H2 zinc-finger protein is specifically required for direct activation in the most proximal rings. This work paves the way to decipher at the molecular level how to make concentric rings from an epithelial layer in order to organize the PD limb patterning

On this basis the claim by Ibn Mada'al-Qurtubi, in The Refutation of the Grammarians, that case is assigned solely by the language user on strictly semantic grounds, is rejected; also untenable, however, is the position of traditional Arab grammarians that case can, in some contexts, be assigned by "concealed" governors, as the Projection and Structure-Preserving Principles require that material having semantic content be visible at all levels of representation. Government and Binding principles are adequate to account for case-assignment in the issues raised by Ibn Mada'; they cannot, however, offer a clear solution to the apparent violation of Principle C of the Binding Theory found in some examples of bab al-tanazuᶜ, the category of "contention in government".

The word-of-mouth communications (WOM) has been considered a fundamental marketing element. The reason for this theme’s choice is the relevance that this uncontrolled communication has gained at the relationship marketing context, as costs free for companies and considered more reliable for costumers. Many researches point those relevant differences between behavior results of WOM showing that specific studies shall be amplified. So, this research applies the model of Brown et al. The use of the term structure of interest rates to price options is relatively new in the literature. It describes the relationship between interest rates and the maturities of bonds. The first model that described the interest rate process was the Vasicek (1977) model. There have been many studies on the formulation of theoretical pricing models. Yet limited empirical research has been done in the area of actually testing the models. In this thesis we report the results of a set of tests of the models indicated below. This paper involves analysis of the pricing errors of the Black model ( 1976), Asay model (1982), Extended-Vasicek model (1990) and Heath-Jarrow-Morton model (HJM) ( 1992) as applied to call options on 90-day Bank Accepted Bill (BAB) futures. Monthly yield curves are generated from cash, futures, swap and interest rate cap data. A number of different methods of analysis are used. These include the use of inferential statistics, non-parametric sign tests and Ordinary Least Square Regressions. The Wilcoxon non-parametric sign test assists the interpretation of whether the pricing errors are from the same distribution. Ordinary Least Square Regressions are used to assess the significance of factors affecting pricing errors. In addition, data are plotted against different variables in order to show any systematic patterns in how pricing errors are affected by the changes in the chosen variables. Monthly options data on BAB futures in the year 1996 suggest that the term structure models have significantly lower pricing errors than the Black and the Asay model. The Heath-Jarrow-Morton model (1992) is overall the better model to use. For the term structure models, pricing errors show a decreasing trend as moneyness increases. The Extended-Vasicek model and the HJM model have significantly lower errors for deep-in the-money and out-of-the-money options. Higher mean absolute errors are observed for at-the-money options for both term structure models. The HJM model overprices at-the money options but underprices in and out-of-the-money options while the Extended Vasicek model underprices deep-in-the-money options but overprices options of other categories. The mean and absolute errors for both the Black model and the Asay model rise as time to maturity and volatility increases. The two models overprice in, at and out-of-the money options and the mean pricing error is lowest for in-the-money options. The results suggest that the factor time to maturity is significant at the 0.05 level to the -mean pricing error for all four models. Moneyness is the only insignificant factor when the Asay model is used. It is also negatively correlated to mean pricing error for the Black model, the Asay model, the Extended-Vasicek model and the HJM model. The R-square for the Extended-Vasicek model was found to be the lowest. Overall, the HJM model gives the lowest pricing error when pricing options on 90-Day Bank Accepted Bill Futures.

Ce phénomène permet aux individus de s’adapter à des environnements fluctuants. Il peut également faciliter l’évolution en élargissant la gamme de phénotypes produits par un génotype. Comme modèle de plasticité phénotypique, nous étudions la pigmentation abdominale chez les femelles Drosophila melanogaster. En effet, ce caractère est sensible à la température : les femelles drosophiles sont plus pigmentées lorsqu’elles se développent à basse température, particulièrement dans les segments abdominaux postérieurs. Les études précédentes du laboratoire ont montré que le gène tan (t), codant une enzyme de pigmentation, est beaucoup plus fortement exprimé à 18°C qu'à 29°C. Par ailleurs, ce gène joue un rôle essentiel dans la plasticité phénotypique de la pigmentation abdominale des femelles Drosophila melanogaster. Au cours de ma thèse, je me suis intéressée à la caractérisation du réseau de gènes impliqué dans la régulation de l’expression de t dans l’épiderme abdominal des femelles Drosophila melanogaster. J'ai également cherché à identifier, dans ce réseau, les acteurs pouvant médier l'effet de la température sur l'expression de t. BBO. Os filmes finos ou espessos foram preparados através do método dos precursores poliméricos (Método Pechini) e através da técnica por evaporação por feixe de elétrons. Nas amostras preparadas através do método dos precursores poliméricos, a razão entre os compostos BaO e \'B IND.2\'\'O IND.3\' teve um papel fundamental no processo de estabilização fase BBO. Esta estabilização só foi possível quando a amostra foi preparada com óxido de boro em excesso, numa razão 70% \'B IND.2\'\'O IND.3\' e 30% BaO (% peso), muito distante da composição estequiométrica 32% \'B IND.2\'\'O IND.3\' - 68% BaO. A análise química desta amostra mostrou que a perda do elemento boro está ocorrendo durante a síntese das amostras. Devido à baixa viscosidade da solução estabilizada, foram obtidos filmes extremamente finos o que dificultou a análise por difração de raios-X. A análise por microscopia de força atômica destes filmes mostrou uma distribuição irregular de tamanhos e formas de grãos. A modificação do método Pechini pela adição de adição de poliidróxidos em substituição ao etilenoglicol mostrou ser eficiente na estabilização da fase BBO. A partir da substituição parcial ou total do etilenoglicol pelo sorbitol foi possível obter uma solução límpida, transparente e estável. Testes iniciais na deposição de filmes finos utilizando estas soluções mostraram que é possível obter filmes espessos (160 nm) por \"spin-coating\" após a deposição de apenas uma camada. No que tange aos filmes obtidos por evaporação de elétrons, diferentes fontes de evaporação foram utilizadas a fim de obter filmes da fase BBO. Os resultados de difração de raios-X mostraram que a fase obtida depende do material utilizado como fonte de evaporação, do tipo de substrato e da temperatura de tratamento térmico. O aquecimento do substrato \"in-situ\" promoveu uma cristalização parcial da amostra. A observação da formação da fase BBO só foi possível após um aquecimento \"ex-situ\" do filme. Comparando com a técnica dos precursores poliméricos, a técnica de evaporação por feixe de elétrons permitiu a obtenção de filmes mais espessos, cuja espessura variou entre 0.4 e 1.7m. A análise por microscopia de força atômica destes filmes mostrou em alguns casos a formação uma superfície uniforme e de baixa rugosidade
The main objective of this work was to prepare and characterize \'BETA\'-Ba\'B IND.2\'\'O IND.4\' (BBO) thin films. Thin and thick films were prepared using two different techniques, the polymeric precursor method (Pechini method) and the electron beam evaporation technique. In the samples obtained by the polymeric method, the BaO/\'B IND.2\'\'O IND.3\' ratio played an important role in the stabilization of the BBO phase. Stabilization, in this case, was only attained when the sample was prepared using a very high percentage of boron, i.e., a ratio of 70%\'B IND.2\'\'O IND.3\' and 30%BaO (weight percent). This composition is far from the stoichiometric one normally used to prepare the Ba\'B IND.2\'\'O IND.4\' compound. A chemical analysis of this sample revealed that there was a loss of boron during synthesis by evaporation. Due to the low viscosity of this solution, the films obtained by dip coating were very thin, making X-ray and microstructural analyses very difficult. The Pechini method was modified by replacing ethylene glycol with sorbitol, which stabilized the BBO phase in a stoichiometric composition. The solution obtained from this sample was transparent and stable, and thin films obtained from this solution presented a thickness of approximately 160 nm after only one deposition. For the films obtained by electron beam evaporation, different evaporation sources were used to produce the BBO phase. Heating the substrate in-situ was inefficient to obtain crystalline thin films, even when heated at 700°C. X-ray diffraction measurements showed that it was possible to produce the BBO phase when the sample was heated ex-situ at 650°C. The surface roughness and microstructure of the thin films was found to depend on the source of evaporation, the type of substrate and the ex-situ temperature to which the film was subjected. A comparison of the films obtained by the polymeric precursor method and those obtained by electron beam evaporation revealed that using the latter the films thickness can be varied from 0.4 to 1.7m

This thesis examines the ban on bar dancing in Mumbai and Maharashtra as an outcome of politics of gender and caste in a globalising India. By redeploying the historical erotic dancing in a globalising India, the dance bars emerged as the new market providing Bollywood-type entertainment with dance, music, and hospitality to the new consumer class. The Dance Bar market offered employment opportunities to poor women to earn a livelihood and proved to be an exceptional market where female labour was paid very well. By deploying their caste capital, i.e., the hereditary skills of dancing, drama and use of sexuality, the traditional dancing women occupied and ruled the dance bar market. To this extent, the bargirls may be viewed as a ‘performing caste’ continuing their hereditary caste occupations in a globalising capitalist market. However, while their relationship to the customer was defined through the market and they earned money, status and power through their occupation, the bargirls challenged, transformed and redefined the caste hierarchy by the use of ‘caste capital’. The demand for the ban on dancing in the bars came up through cultural discourses of protection of youth, family, Indian culture and the dignity of women, in which bargirls were projected as bad women earning too much money by cheating their customers and encouraging illegal activities in the dance bars. The ban diminished the freedom, earning potential and unique identity of the bar dancers, pushing them back into their original caste/class positionality. The legal ban can be studied as the effect of politics of caste and gender in Maharashtra intervening into the global market.

Resultados: Todas as proteínas examinadas foram observadas nos adenocarcinomas e mucosa não tumoral, porem com diferenças de expressão em relação à porcentagem de positividade e intensidade. Observamos: i) Houve associação entre 0 tamanho do tumor e a proteína p53. ii) Houve associação da proteína Bad no adenocarcinoma com a idade dos pacientes. iii) Associação das proteínas Bax, Bad e Ki-67 com 0 adenocarcinoma de tipo intestinal. iv) As proteínas Bcl-xl, Bak, Bad, p53 e Ki-67 apresentaram diferenças estatisticamente significantes entre a imuno-expressão no tumor e na mucosa não tumoral. v) Associação das proteínas Bax, Bak e Bad na mucosa não tumoral. vi) Não houve correlação da imunoexpressão das proteínas com a sobrevida dos pacientes. Conclusão: A expressão aumentada da proteína Bcl-xl nos adenocarcinomas, com evidente diferença de expressão entre 0 grupo teste e 0 grupo controle, esta relacionada com 0 efeito anti-apoptótico da proteína. A expressão reduzida das proteínas Bak e Bad e a expressão aumentada das proteínas p53 e Ki-67 nos adenocarcinomas demonstram 0 desequilíbrio entre morte e proliferação celular, permitindo 0 crescimento descontrolado das células neoplásicas.
Purpose: To study the immunoexpression of Bcl-2 family proteins (Bcl-2, Bcl-xl, Bax, Bak, Bad) and to evaluate the correlation between the immunoexpression of these proteins with the cleaved caspases 3, Ki-67 and p53 immuno-expression. Methods: A TMA paraffin block was constructed with gastric carcinoma tissue (test group) and normal gastric adjoining mucosa (control group) of 87 patients. The TMA block was submitted to immunohistochemistry for Bcl-2, Bcl-xl, Bax, Bak, Bad, p53 and-cleaved Caspase 3. Results: All studied proteins were present in tumor and normal gastric adjoining mucosa, but with different intensity and amount of positive cells. i) There was an association between tumor size and p53 expression. ii) association between Bad expression in the tumor and patient’s age. iii) Intestinal type adenocarcinoma was positively correlated with the expression of Bax, Bad and Ki-67. iv) The protein Bcl-xl, Bak, Bad, p53 and Ki-67 showed statistically significant differences between the immuno-expression in tumor and normal gastric adjoining mucosa. v) There was an association between the proteins Bax, Bak and Bad expression in the normal gastric adjoining mucosa. vi) No correlation between patient’s survival rates and the expression of the proteins was observed. Conclusions: The higher expression of Bcl-xl protein in adenocarcinoma, the difference of Bcl-xl expression between test group and control group, might be related with the anti-apoptotic effect of this protein. The lower expression of Bak and Bad and the increased expression of p53 protein and Ki-67 protein in adenocarcinomas demonstrate the imbalance between death and cellular proliferation, which allows the uncontrolled tumor cell proliferation.

Abstract:
Functional failure and loss of pancreatic β-cells are critical events in the pathogenesis of diabetes and there is mounting evidence that suggests chronic endoplasmic reticulum (ER) stress contributes significantly to β-cell dysfunction and apoptotic death. Two core pro-apoptosis proteins, Bax and Bak, mediate the execution of mitochondrial apoptosis and have also been suggested to regulate aspects of ER physiology and stress signalling. In this study we set out to determine the relative contributions of Bax and Bak in the execution of β-cell death and examine their putative roles in β-cell ER-stress signalling under diabetogenic conditions. We generated mice in which the single or combined knockout of Bax and Bak could be induced in the pancreatic β-cell. Physiological islet function assessed both in vivo and in vitro was not affected by the knockout of Bax and/or Bak. However, Bax and Bak knockout improved β-cell survival under stress conditions. Single knockout, double knockout, and wild-type cells were assayed for ER-stress and cell death following treatment with staurosporine, thapsigargin, and culture under glucolipotoxic conditions. Time-course kinetic cell death analysis demonstrated that single and double knockout cells were protected from staurosporine, and further revealed that Bax-Bak double knockout was required for significant protection from death under glucolipotoxic conditions. ER-stress signalling was evaluated by quantitative PCR for XBP1s and CHOP. Interestingly, spliced XBP1 expression was augmented in Bax-Bak double knockout islets in the early phase of ER-stress signalling compared to wild-type controls. Stress-induced CHOP expression increased in a time-dependent manner but was not significantly different between Bax-Bak double knockout and wild-type islets. These results suggest that Bax and Bak regulate the IRE1α arm of the ER-stress response upstream of apoptosis by suppressing maximal XBP1 splicing. Under glucolipotoxic conditions, pancreatic insulin content, insulin secretion, and insulin transcription were unaffected by Bax and Bak knockout, indicating Bax and Bak do not mediate their protective effects towards β-cell death by retaining islet function. Together these data demonstrate that Bax and Bak have both individual and combined contributions to β-cell death under various stress conditions and suggest novel non-apoptotic roles regulating early ER-stress signalling in the β-cell.

Abstract:
L'étude de l'apoptose est importante pour développer de nouvelles stratégies anti-cancéreuses. La voie mitochondriale de l'apoptose est finement régulée par le réseau d'interaction des protéines de la famille Bcl-2. D'autres acteurs moléculaires participent à la régulation de l'apoptose et notamment une famille de lipides bioactifs : les sphingolipides. De plus, des études récentes permettent d'envisager que les interactions entre le réticulum endoplasmique (RE) et la mitochondrie (via une région spécifique du RE appelée MAM) interviennent également dans la régulation de l'apoptose. Nous avons mis au point un protocole de fractionnement subcellulaire permettant de séparer une fraction mitochondriale, des mitochondries purifiées et les MAM, puis analysé les compositions protéiques et lipidiques de ces fractions (par WB et HPLC-MS). Nous avons ensuite comparé les compositions de ces fractions à l'état basal et en conditions apoptotiques. Nos résultats montrent que l'induction de l'apoptose induit une redistribution des protéines de la famille Bcl-2 avec notamment la transduction de Bax au niveau des mitochondries et des MAM ainsi qu'une forte augmentation de l'expression de la protéine Bcl-2 au niveau des MAM. En ce qui concerne les profils lipidiques, l'induction de l'apoptose entraine une augmentation du taux de céramide dans les mitochondries et une diminution du taux de sphingomyéline dans les MAM. L'ensemble de nos résultats suggère que les MAM pourraient jouer un rôle important dans la localisation subcellulaire des protéines de la famille Bcl-2 au cours de l'apoptose, potentiellement via la synthèse et/ou le trafic des sphingolipides
The study of apoptosis is important to develop new anti-cancer strategies. The mitochondrial apoptotic pathway is tightly regulated by the interaction network of the Bcl-2 family proteins but other molecular actors are involved in the régulation of apoptosis, in particular, a family of bioactive lipids : sphingolipids. The recent studies have shown that the interactions between endoplasmic reticulum (ER) and mitochondria (via a specific region of the ER called Mitochondria associated membrane, MAM) are also involved in the regulation of apoptosis. We have developed a subcellular fractionation protocol for the isolation of mitochondria fraction, purified mitochondria and MAM. The proteic and lipidic compositions of these fractions were analyzed by WB and HPLC-MS. We compared the compositions of subcellular fractions at normal and apoptotic conditions. Our results show that induction of apoptosis causes the redistribution of the Bcl-2 family proteins including Bax transduction to mitochondria and MAM and an important increase of Bcl-2 expression at MAM Regarding lipid profiles, inductions of apoptosis increases ceramide level at mitochondria and decreases sphyngomyelin in the MAM fraction. Taken together, these results may suggest that the MAM could play an important role in the subcellular localization of Bcl-2 family proteins during apoptosis, potentially through synthesis and/or trafficking of sphingolipids

Abstract:
The Y₁Ba₂Cu₃0x superconducting thin films were made by both sequential electron-beam evaporation and co-evaporation of Cu, BaF₂ and Y, followed with high temperature post-deposition annealing. The possibility of making Y₁Ba₂Cu₃0x superconducting film in situ with BaF₂ as the Ba source was investigated by studying the conditions of in situ conversion of BaF₂ to BaO. The BaO concentration in the film as function of the substrate temperature was estimated with an equilibrium thermodynamic model and found to agree with the experimental results. A brief experimental and theoretical exploration of BaCl₂ also showed that BaCl₂ does not have any advantage over BaF2 as the Ba source for in situ growth of Y-Ba-Cu-0 films from the growth temperature point of view.

Abstract:
Apoptosis, or programmed cell death, is an essential process in all multi-cellular organisms. It is indispensable to an organismâ s survival, preventing the malicious propagation of DNA damage and pathogenic alterations, through the clean disposal of afflicted cells. The BAD/tBID/BAK pathway is a portion of the apoptosis molecular pathway, albeit an important pathway since it is known to be deregulated and lead to pathological ailments such as cancer. Using chemical kinetics the BAD/tBID/BAK signaling pathway is modeled as a set of (nonlinear) ordinary differential equations. A first-cut numerical analysis reveals a mechanism where BAD sensitizes a switch from tBID activation to BAK activation. The phosphorylation of BAD is shown to inhibit this sensitizing effect. All behaviors are supported by experimental data, thereby validating the model of the BAD/tBID/BAK pathway. Moreover, modeling the phosphorylation of BAD as one of two modes, some conflicting experimental data about BADâ s phosphorylation can be disentangled. Parameter values (in this case the kinetic rate constants) are prone to error or missing altogether. Chemical reaction network theory, however, provides a theoretical approach to complement the initial numerical analysis without having to specify rate constant values. We extend the global asymptotic stability and robustness results in [92] to include any complex-balanced mass-action network. This enables us to study the BAD/tBID/BAK signaling network by breaking it into two sub-networks: one with BAD and tBID, and the other with tBID and BAK. The complex-balanced BAD/tBID sub-network is shown to possess a unique steady state which is globally asymptotically stable. This verifies the simple and dynamically well-behaved exchange of BAD for Bcl-2 proteins which guard against tBID activation. The second sub-network, tBID/BAK, is formulated as a complex-balanced network with a perturbation representing the reaction of tBID catalyzing the activation of BAK. Our theoretical results produce a non-conservative, though state-dependent, condition which can be used to prove global convergence to a neighborhood of the unperturbed steady state. We then illustrate the biological importance of the result for tBID/BAK sub-network with an example design for a drug delivery system.
