This Ph.D. dissertation analyses the Nūberu Bāgu film trend based on the premise that its characteristics allow it to be considered an integral component of the Japanese New Left ideology, with both phenomena belonging to what is known as the “season of politics” (seiji no kisetsu) (1966-1971). From this approach, the understanding of a certain type of cinema is a way to gain a better insight into the ideology to which it belongs, just as the understanding of an ideology as a whole is a way to gain a better insight into the elements comprising it, cinema among them. On the methodological level, first I propose my own definition of the concept of “ideology” starting with some of the approaches by Plamenatz (1979), Eagleton (1991), Žižek (1994) and Kellner (1995). Based on this definition, I formulate a methodology for the “ideological analysis” of cinema starting with some of the conceptions by Thompson (1984, 1990), Zavarzadeh (1991) and Kellner (1993; 1995). The core of this methodology is a reading of films as texts whose internal elements serve the external interests of certain socio-political groups in a significant power relationship with other socio-political groups. In the case at hand, this means fundamentally decoding the continuous lines connecting the Nūberu Bāgu cinema with other ideological components of the Japanese New Left, as well as with this socio-political group’s ideology as a whole, opposed mainly to the Japanese Old Left. In order to conduct an “ideological analysis” of the Nūberu Bāgu, I select a sample of two paradigmatic films through which a set of characteristic elements of this film trend can be distinguished, which enables to configure a model to address this cinematographic phenomenon as an integral part of the Japanese New Left ideology; a model that can be used for future research. These films are Eros + Massacre (Erosu purasu Guakusatsu, 1969), by Yoshida Kijū, and Throw Away Your Books, Rally in the Streets (Sho o suteyo machi e deyō, 1971), by Terayama Shūji. The films were released at historical moments with significantly different implications, and filmed by directors with divergent sensitivities, but it is precisely these discontinuities that enable us to establish a coherent model (under the name of Nūberu Bāgu), belonging to a coherent ideology (under the name of New Left). The “ideological analysis” of the Nūberu Bāgu conducted in this dissertation is made up of three stages. In the first stage, I present the methodological approach for the “ideological analysis” of cinema as opposed to that of national cinema; by means of this contraposition, the convenience of “ideological analysis” when addressing cinema will be grasped. Secondly, I address the context of power relationships in Japan at the time when the Nūberu Bāgu emerged, and then I analyse Eros + Massacre by decoding the connections between the film text and this ideology as a whole (to which the film belongs as a doctrinal component). In the second stage, I investigate the relationship between the Nūberu Bāgu cinema, still using the illustrative case of Eros + Massacre, and the prevailing collective consciousness in the Japanese New Left of the importance of human subjectivity (shutaisei) conceived as the subject’s self-negation (jiko hitei) (and which is a subjective component of this ideology). To do so, I focus on two key aspects of Yoshida Kijū’s cinema through which he displays his view of subjectivity as self-negation: space and memory. In the third stage, I analytically relate the political theory of one of the most important thinkers in the shaping of the Japanese New Left, Yoshimoto Takaaki, and Terayama Shūji’s conception of cinema, through the illustrative case of his film Throw Away Your Books, Rally in the Streets. In this stage I focus on the way Yoshimoto, through his political theory, and Terayama, through his film practice, express a similar view of the relationship between the figure of the intellectual and the masses, taking into account that these expressions can be considered as part of the same doctrinal body of the same ideology: the Japanese New Left. Finally, I add an appendix in which I present the preliminary research that led to the approach of this Ph.D. dissertation. I have included it in this compendium as a reference for a general perspective of the starting point of the Nūberu Bāgu as a reaction to the humanist cinema hegemony in Japan until the end of the 1950s. The introductory understanding of the confrontation between both kinds of cinema enables us to gain a better understanding of the conflict between the New Left and the Old Left prevailing in the ideological dimension of the post-war Japanese society. This Ph.D. dissertation affords a better comprehension of the Nūberu Bāgu and the Japanese New Left ideology to which it belongs, and of their interrelationship. In the first place, the stages of the “ideological analysis” enable us to better understand the cinema, the political theory and the subjective consciousness that make up the Japanese New Left, by means of the analysis of their interrelationships and their relationship with this ideology as a whole. Secondly, these stages dialectically refine our understanding of the Japanese New Left ideology as a whole through the analysis of its indissoluble relationship with some of the components comprising it, and of the interrelationships between these components. Thirdly and on a more abstract level, they constitute, through the proposed approach to the relationship between the Nūberu Bāgu and the Japanese New Left, my critical rethinking of the very notion of ideology.

Full textThis study compares academic writing conventions in French and Chinese from the point of view of Contrastive Rhetoric in the field of French teaching and learning as a foreign language. We spot respectively in French and Chinese education systems a specific genre: the French dissertation and the Chinese yilun wen. They have a comparable status, a clear structure and strict rules to follow. The comparison between these two genres indicates that the rules are not shared between French and Chinese students and that certain criteria required in the Chinese yilun wen appear far removed from French dissertation. Thus Chinese students encounter intricate difficulties in writing French dissertations and negative transfers under the influence from Chinese writing conventions. This result is also confirmed by the analysis of dissertations written by the Chinese students of our corpus. The surveys conducted among French and Chinese learners show that they are not fully aware of the cultural differences of standards in writing. The analysis of comments by French teachers of dissertation written by Chinese students shows that they are not better armed than their students in face of the differences of textual organization (both the construction and the way of realization) in French and Chinese. We attend to hold endly how the organization of traditional Chinese Rhetoric (the bagu wen and the Qi-Cheng-Zhuan-He schemes), still proposed by textbooks and practiced by learners, exert more or less influence on French writing of the Chinese students

Full textThe present project of handbags for women students of universities and institutes and housewives has allowed us to validate the problem assumed based on information gathered in the analysis to the sector to which our product is directed, the NSE "C". In this way, we managed to detect that women belonging to this level did not have the necessary resources to adapt to the changing world of fashion. Currently, the fashion and beauty sector are constantly changing, because there are several designers and brands that year after year launch new products to the market, generating trend and attraction for women around the world. However, there are many categories of products in this sector that present very high prices for people who receive a minimum remuneration or who do not generate monthly income. From this, the different products offered in this sector were analyzed, with the bags being the product chosen for its preparation and sale, because it is very required by women and it is expected that its level of consumption will increase in the following years. Queen Bags will launch to the market a wide catalog of handbags elaborated based on the tastes and preferences of the clients. Likewise, it will stand out for the variety of designs offered, the launching of new catalogs for each station and the quality-price ratio of the final products. The business will start operations in 2019 and for its start-up the shareholders will make a total investment of S/12,719 formed by the purchase of assets and cash contribution.

Full textThis academic study aims to explain the reasons why the change in the consumption of plastic products for ecological in order to preserve natural resources is necessary. Excessive use of non-recyclable products is damaging to the environment, not only in our country but around the world. Therefore, we have developed a project in which we prove we can romove progressively the use of plastic bags, mostly made of polypropylene. Daily, see everything we carry tends to be inside a plastic container. These bags are used only for a few minutes to transport products from supermarkets, warehouses and shops to our homes or offices, but the remain on the planet by 150 years during the degradation process. According to the Ministry of the environment (2015), each year we use 791 tons of plastic bags in Peru. Only in supermarkets, one million six hundred thousand bags are used every day. The use of reusable bags could last for years and they are made of tocuyo and jute with recycled fabric strokes, thus managed to preserve the ecosystem. Our business model better alternatives in the market regarding the daily activities of mass consumption in major chains of stores and retail sales. Satisfactory results and showing signs of innovation, performing a project viable and beneficial for the company and the public in general.

Full textThe objective of this study is a reflection about visions and methodological approaches to analyse more specifically guinean traditional art. In fact, masks and sculptures formed, realised, used, transformed and or neglected by the nalu and the baga of guinea can permit a better acknowledgment of the history and the social context of these two tribe. Items of cult at the beginning, masks and sculptures were first known as simple curiosity before becoming an "objet d'art", wellknown outside. Many regards were glanced at them: colonial figure, origin of new discipline, theorical reflection in history of art, anthropology, philosophy. . . , research and methodological approaches adapted to every african tribe. Nalu and baga example of guinea offer a very good experimentation to day. After defining the geographical and historical context, 32 sculptures selected for the study are presented of their formal aspect, in the museum with conditions of collect or acquisition. On the field, theses sculptures have a basic place because there are for the searcher material source of information and for the users evidence of a lost civilisation and sociological indicators for the present and the future

Full textConstant change is closely associated with the fashion industry, as new trends and styles tent to change continuously. In the Western world, people consume considerably more fashion products than what the natural resources allow. The fashion consumers, however, gained an insight into the fashion industry´s negative environmental impact in 2017. A new law was founded to alert the fashion consumers of the negative effect of the shopping bags, especially made with plastic. In association with the new law, an organization, naming One Bag Habit was created. This led the fashion consumers to think extra before deciding to consume a shopping bag when they made a purchase of fashion item. Due to One Bag habit, fashion consumers now avoid from consuming shopping bags because of environmental aspects. The reluctance to consuming a shopping bag in addition to the fashion purchase has decreased yet impacts on the actual fashion consumption have not changed. Previous research has identified an attitude-behaviour gap between how fashion consumers value sustainable products and their actual behaviour in accordance with these values. This has led to this paper highlighting how this contrast is formed between consuming fashion products and refraining from the shopping bags because of environmental reasons. This study has a textile management approach, which means that the study is primarily aimed at companies in order to help them understand how fashion consumers value and act in different purchasing situations. This study will examine how fashion consumers consume shopping bags in relation to fashion products. The findings will contribute to creating an understanding of the created contrast between sustainable consumption regarding shopping bags and non-sustainable one regarding fashion consumption itself. Moreover, the findings will contribute to a deeper understanding of why the attitude-behaviour gap has created such a contrast among the fashion consumer. In order to understand and interpret the empirical material of this study, a theoretical model of attitude-behaviour gap was created. In addition, an understanding of how needs are created among consumers will be contributing to the formation of the theoretical framework. Furthermore, data was collected through qualitative interviews, three focus groups, and an observation. The data sample frame consisted of students from three different universities in Sweden (the Swedish School of Textiles in Borås, University of Gothenburg and Mälardalens University in Västerås). The findings of the study show that fashion companies need to be more specific when informing the fashion consumers on why and how they can act more sustainably when consuming fashion and shopping bags. In addition, the sustainable fashion products should be comparable in prices terms to non-sustainable fashion products in order for fashion consumers to act sustainable. Finally, it is suggested that the sustainable fashion products have an appealing designs in order for the fashion consumers to invest in sustainable fashion products.

Full textThis study examines consumers’ behavior in the purchase of grocery bags in food stores. Consumption of all types of bags is a problem in several ways, and this report will examine consumers’ knowledge about resource and energy use in manufacturing, pro-environmental behavior and consumers’ suggestions to behavioral change while purchasing grocery bags. The purpose is to investigate whether knowledge causes the consumer to act in a certain way. To achieve the purpose, a survey was conducted, both in a selected ICA store and on Facebook. The survey contained questions about consumption such as, what bag consumers usually buy and how often. Even how the consumer went to the store is of interest as well as the costumers’ knowledge previous to the study regarding resource utilization in manufacturing. Lastly, the consumers’ attitude towards a potential ban was asked for, as well as what could make the person reduce his or her own consumption. The study found that there is no difference in knowledge between those who buy bags and those who do not. This leads to the conclusion that there is a value-action gap, one is aware that the consumption is not sustainable, but one does not act according to his or her knowledge. Thus, the most effective way of reducing consumption is not considered to be a better knowledge of environmental impact. Instead, a ban on plastic bags based on oil is preferred. Hence, environmental bags and Swedish paper bags would be more promising options. Relatively few of those who participated in the survey are against a ban on disposable bags. Because even the consumption of environmental bags and Swedish paper bags has a negative impact on the environment, a reduction of consumption of all bags is therefore preferable. This can be achieved with an increase in price.

Full text"The present project “Fancy Rent Bag” aims to develop an innovative and scalable model, which allows contributing sustainably to society, so that it can face our national competitors through different marketing strategies focused on optimizing our operational and sales processes. Thus, the company will be able to position in the market and increase its market share to reach its target segment and potential customers effectively. Likewise, our target audience is divided in two segments. On the one hand, the first segment or also called ""consumers who want to rent luxury or designer bags"" focuses on modern peruvian women residing in Metropolitan Lima from 18 to 39 years of age from the socioeconomic status A, B and C. The proposal for segment 1 is to offer a digital space in which users can find different exclusive bags at affordable prices. On the other hand, the second segment or also called “owners” are those women who want to rent their luxury bags and earn extra money. This segment includes Peruvian women of sophisticated style residing in Metropolitan Lima from 20 to 40 years of age from the socioeconomic status A and B. Given this, during the course of the project it was possible to notice an incremental reception of the rent of luxury bags, which was with the impressions and interactions on social networks, Google Ads and Google Analytics.

Full textThis final thesis is written by Jelena Todorovic, studying Product Design at Malmö Högskola. The purpose with this project was to develop a sport bag for people living nearby a gym and who showers at home instead of the gym after training. The main focus of the project is User Centered Design, which means that the product development of the concept is based on the end users, or the customer’s preferences, needs and wishes. The user is included in every step of the project development, such as commenting and criticizing sketches and concepts, being interviewed and observed, and evaluating the different stages of the concept and product. The student saw a need of a different bag than the traditional sport bag. The pilot study resulted in a list of requirements based on the target group and their needs and preferences. A gym bag with focus on the functionality became the project. The student wanted the project and the final concept to be environmentally friendly, why a research in that field was necessary. We need to think about our planet, and for a Designer, it is a challenge to develop a product which is environmentally friendly and sustainable. It is also desirable for a product to fill its function and last for a long time. Finally, the project resulted in a functionality model of the new gym bag.

Full textThe province of Bagua, in the Amazon region, bases the operation of its economy on agricultural activity, with emphasis on the cultivation of coffee, cocoa and rice. For this reason, it’s proposed to repower the products with the different long-term objectives, ranging from the improvement in their productivity to the increase of their quality and, to increase the income of the producers to define the economy of the area and close the gaps in education and health of the province. All under a policy to respect the environment that translates into the goal of reducing the amount of wastewater discharged into rivers without treatment. The five strategies proposed were raised from a comprehensive assessment of the actual situation of Bagua, which includes a critical and systematic analysis of the determining factors of success, both internally and of its environment. The evaluation process has considered strategic aspects which include the analysis of opportunities, threats, strengths and weaknesses, as well as ethical and legal aspects. Thus, in the vision proposed to the year 2030, the human development index is expected to be at least 0.5, thanks to the strengthening of crops and raised with facilities such as the creation of a municipal agricultural nursery that ensures the safety of products and strengthening agricultural extension services to improve agricultural practices by increasing production and reducing the cost of crops

Full textAutophagy, literally meaning self-eating, is a highly evolutionary conserved process in eukaryotes where elements of the cytoplasm (organelles, macromolecules) are degraded by lysosomes. Autophagy can occur randomly in the cytoplasm or can be selective of a specific organelle. Among other, the specific degradation of mitochondria is called mitophagy. Autophagy and mitophagy have been implicated in several physiopathologies such as neurodegenerative diseases or cancer. Deregulations of autophagy/mitophagy may profoundly affect homeostasis. The aim of my thesis is to characterize the role of the co-chaperonne protein BAG6 in the regulation of mitophagy.BAG6 is a 150kDa protein, also known as BAT3 or Scythe, which functions in the quality control of the cytoplasm. Moreover BAG6 is also involved in immunity, apoptosis or autophagy. Our work showed that it is implicated in the regulation of mitochondrial morphology by inducing mitochondrial fission. Also, BAG6 induces mitophagy: in presence of BAG6, mitophagy markers such as PINK1 and PARKIN are more localized at the mitochondria whereas the expression of mitochondrial specific protein’s (TOM20, TFAM and TIM23) decreases. After its sequence analysis, we discovered that BAG6 is composed of many domains such as the UBL domains and two LIR domains (LC3- Interacting Region) and that BAG6 interacts with LC3 through its LIR2 domain. These features lead to identify BAG6 as a new potential receptor of mitophagy

Full textThe annular floating ring seals can significantly reduce the leakage in rotating machines operating with gas or cryogenic fluids. Their integration into the machine is not simple because the floating ring must be able to follow the vibrations of the rotor. The presented work has two parts. The first part consists of the analysis of static and dynamic behavior of the annular seal which forms the primary seal. The model developed is based on the theory of thin lubrication films dominated by inertia effects together with a total enthalpy form of the energy equation. This model can handle the phase change in cryogenic fluids. The dynamic response of the annular seal is expressed as the linear dynamic coefficients (derived for small perturbations). The dynamic coefficients vary with the operating regime of the seal ring (eccentricity, upstream / downstream pressure, rotation speed) but do not depend on the excitation frequency. The second part is dedicated to the development of a dynamic model. The floating ring is subject to forces generated by the fluid flow in the annular (primary) seal, the mixed friction forces between its nose and the stator (the secondary seal is similar to a mechanical face seal) and the inertia forces caused by its own weight. The rotor acts as an external excitation. A suitable model is developed for each of these forces. Two dynamic models are proposed. The first model is based on the assumption that the floating ring follows the rotor according to a periodic precession motion. This model allows a simple and fast quasi-analytical solution but is limited by the fact that the dynamic response of the ring may be more complex. The second model is fully nonlinear and takes into account the possible impacts between the rotor and the ring and between the ring and the anti-rotation pins. The model shows that under certain conditions the trajectory of the ring can be quasi-periodic or chaotic. Overall, the results of the second part underline design rules for properly integrating annular floating ring seals into a rotating machine

Full textThis strategic planning for the Province of Bagua has been developed aligned to the methodology proposed by D’Alessio (2016). Long-term objectives are proposed for the year 2030, as a development plan that seeks to improve the competitiveness of the province and reach national scope and international notoriety through its main current and future economic activities. According to the 2007 Census, the main economic activity of the Province of Bagua is agriculture and has not reached adequate technical development yet. The main trade derived from this activity is the commerce of products in the form of grains such as coffee, cocoa and rice. Another activity such as tourism is at an initial stage, which is very low due to the lack of infrastructure and very poor quality of service, moreover the lack of promotion and advertising. In regard to the long-term objectives of 2030, they have considered to increased the productivity of cocoa and coffee by more than double their current performance; boost Bagua within the top 10 of provinces with the largest number of tourists; improve their percentage of satisfactory level in reading comprehension from 10.5% up to 43.15%; and Bagua by 2030 will produce value-added cacao products from 0% nowadays up to 75% of the total cacao production, getting to be inside the top five of provinces leading the exportation of them. To achieve these long-term objectives, a set of short-term goals are proposed, and they are based on strategies that will impact and improve Bagua’s current situation, promoting the development of agriculture, tourism sector an education for the walfare of its citizens.

Full textOil-based fuels are notably recognized as unsustainable sources of energy, as their sources are impoverished, as well as their contribution to global climate change. In this scenario, biofuels appear as a renewable alternative to non-renewable ones. In Brazil, the insertion of renewable sources in the energy matrix has grown over the last decades, where biodiesel has played a prominent role. The production front line is centered on top vegetables and residues containing vegetable and animal oils, however, these are not the only sources of raw material for the production of biodiesel. Oil microalgae are a potential alternative that solves, in part, some of the problems associated with biodiesel from vegetable and animal oils. Microalgae can accumulate significant amounts of lipids (up to more than 50% of their dry weight) and generate large amounts of biomass, but are not available in nature at concentrations that support commercial exploitation, except for exceptions. Therefore, to be produced on a large scale, controlled cropping systems can be used. Within this context, the research was divided into 3 stages that aimed to achieve the following objectives: (1) Through axenization techniques, to purify two strains of Chlorella spp. So that the results obtained could be attributed exclusively to the algae; (2) To study the behavior of two strains in two different culture media (WC and TAP) using glucose as an organic carbon source and to investigate physical and chemical factors to optimize biomass and lipid productivity in relation to these factors. (3) With the chosen and optimized factors, to evaluate in a simplified way the cultivation in medium scale (10B photoboreaders). The Chlorella strain DAM5% 04 presented the best results regarding growth rates, and it was therefore chosen. To monitor growth, the absorbances previously calibrated by suspended solids were used. The Gompertz model was used for growth modeling and generated growth curves and parameters (amplitude, maximum growth speeds and generation time). The addition of glucose to the culture media increased the growth rates 4 fold compared to standard WC media (0.30 CFU mL-1 day-1 to 1.23 CFU mL-1 day-1). In the investigation of the chemical factors, the removal of sulfur from the medium was carried out in the same way as the biomass produced, which doubled its value in relation to WC and standard TAP (from 500 mgL-1 to 1090 mg L-1). Which is responsible for increasing the fluorescence value of neutral lipids by more than 7 times (from 1 ua to 7.8 ua), being the most efficient. In the combination of the factors involved, biomass production reached 2000 mg.L-1, Whereas the lipid fluorescence reached 14 ua. The optimal point for the production of biomass and fluorescence of neutral lipids found by the optimization model was 98 mg L-1 of nitrogen, 51 mg L-1 of phosphorus, 0 mg L-1 of sulfur and 200 μmol photons m-2 s-1 for the culture medium. Phase, culture in photobioreactors. It has been found that the airlift is more susceptible to contamination of the crop (microscopically proven) than bags. The highest specific growth rates were obtained in the airlift reactor (5.77 d-1) for CO2-enriched air (2%) cultivation, 4 to 5 times higher than those obtained on a small scale (test tubes). In relation to biomass, the airlift culture presented 2550 mg L-1. Neutral lipid fluorescence did not exceed 1 u.a. in all experiments performed on photobioreactors, well below the values found in the tubes.

Full textThe BAG3 gene was identified as a novel gene responsible for dilated cardiomyopathy (DCM), a major cause of heart failure (HF). The BAG3 protein is a co-chaperone that participates in the control of protein homeostasis via its role in autophagy, protecting cells against the proteotoxicity induced by degraded or misfolded proteins. The hypothesis that inactivation of the autophagic pathway controlled by BAG3 would induce cardiomyocyte proteotoxicity behind the CMD appears particularly attractive and is the central hypothesis of this work. Our results indicate that BAG3 mutations abolish the interaction with the chaperone HSP70, a central actor of the protein quality control. We observed cytotoxicity of BAG3 mutants, an impaired HSP70-dependent chaperone function and absence of autophagic response under stress conditions (starvation, heat shock, expression of a pro-aggregating protein). In vivo (zebrafish model) the extinction of BAG3 expression or mutants overexpression lead to the occurrence of a heart failure phenotype (pericardial edema) in injected embryos. Through genomic edition, we also develop a model of iPS-derived cardiomyocytes carrying or not the mutation in order to further explore the contractile function of these cells. Our results confirm the role of BAG3 in DCM and indicate that the alteration of the proteostasis function is the cause of the disease. This new pathophysiological pathway in DCM may prove to be more generally, a central line in the IC

Heart disease has been the leading cause of death in the United States for more than 90 years. The leading cause of death in individuals aged 65 and older has remained diseases of the heart from 1950 to the current time. According to the CDC, once diagnosed with heart disease, individuals have an approximately 50% chance of dying within 5 years, regardless of race. Mortality related to heart disease increased dramatically from the start of the 1900s to 1921, but subsequently experienced a steady decline from the mid-1960’s to 2000. However, when the decrease in heart disease is examined at the level of race it is clear that the decrease is not equally shared. While the leading cause of death among both Caucasian American men and women and African American men and women remains heart disease, the decrease in incidence of coronary heart disease among African American men was only half of the decrease in incidence among Caucasian American men. Genetic variants in BAG3 (Bcl-2 associated athanogene 3), a highly evolutionarily conserved gene that has recently emerged as a major dilated cardiomyopathy locus, are prevalent in isolated populations. This led us to hypothesize that variants in BAG3 might contribute to the increased prevalence of IDC in individuals of African ancestry. Expressed predominantly in the heart, the skeletal muscle and in many cancers, BAG3 has pleotropic effects in the heart. It inhibits apoptosis by binding to Bcl-2, facilitates protein quality control by binding to both large and small heat shock proteins, mediates adrenergic responsiveness by coupling the β-adrenergic receptor and the L-type Ca2+ channel, and maintains the integrity of the sarcomere by anchoring actin filaments to the Z disc. However, a paucity of subjects of African ancestry have been included in cohorts of probands with familial dilated cardiomyopathy whose exomes or genomes have been sequenced. Based on our previous observations and reports from other groups we postulated: 1) that mice with haplo-insufficiency of BAG3 will re-capitulate disease seen in humans and serve as a model for studying the pathogenesis of BAG3. 2) The prevalence or identification of specific BAG3 variants will differ by race and/or ethnicity. 3) SNVs of BAG3 may contribute to disease progression and thereby be pathogenic. Our study points out that we cannot understand population-based differences without enhancing the diversity of populations included in genomic studies. Similarly, in the era of big data, efforts must be undertaken to assess the genetic profile of both probands and their family members as without the ability to measure segregation, penetrance and plasticity we can only ascribe associations to functional genetic variants.
