Abstract:

Abstract:
Abstract Abstract 3805 Background: Detection of aberrant antigen expression by multiparameter flow cytometry (MFC) is increasingly used in the diagnostic work-up of myelodysplastic syndromes (MDS). Myeloid nuclear differentiation antigen (MNDA) has been described to be strongly expressed in mature myeloid cells while myeloid progenitor cells show a weak expression only. MNDA has been suggested to be aberrantly expressed in MDS and therefore may improve the diagnostic capabilities of MFC. Aim: To analyze differences in the expression of MNDA between non-MDS cytopenias, MDS, and AML. Patients and methods: Bone marrow samples from a total of 269 patients with cytopenias and suspected MDS were analyzed by cytomorphology and MFC in parallel for diagnostic purposes. MNDA expression was determined by MFC in granulocytes, monocytes, and myeloid progenitor cells using the antibody clone 3C1 (provided by Trillium Diagnostics, Bangor, ME). MNDA expression was compared between patients grouped according to cytomorphology as: no MDS (n=103), MDS (n=85), CMML (n=9), AML (n=19), and possible MDS (n=53, based on cytomorphologic features of dysplasia but not sufficient to diagnose MDS). In addition, MNDA expression was compared between patients with aberrant and normal karyotype (cytogenetics was available in 238/269 patients, an aberrant karyotype was present in 59 patients). Results: A higher percentage of granulocytes (G) and monocytes (M) with weak expression of MNDA (%dimG and %dimM) was consistently found in MDS/AML samples as compared to samples without MDS. In detail, %dimG (mean±SD, 20±20% vs. 8±10%, p<0.001) and %dimM (31±24% vs. 16±11%, p<0.001) were significantly higher in cases with MDS as compared to those without. Furthermore, in patients with MDS, as compared to those without, the mean fluorescence intensity (MFI) of MNDA expression was found to be lower in monocytes (mean MFI, 71±36 vs. 85±27, p=0.004) as well as in myeloid progenitor cells with bright (67±24 vs. 86±66, p=0.007) or dim (2.5±0.9 vs. 2.8±1.2, p=0.037) expression of MNDA. In AML cases, as compared to cases with no MDS, similar figures were observed with higher values for %dimG (27±27% vs. 8±10%, p=0.007) and for %dimM (45±31% vs. 16±11%, p=0.001). In addition, in AML cases MFI of MNDA expression was lower in monocytes (55±39 vs. 85±27, p<0.001). Considering the diagnostically challenging cases with dysplastic features by cytomorphology but not sufficient to diagnose MDS similar although smaller differences were seen as compared to cases with no MDS with higher values for both %dimG (15±22% vs. 8±10%, p=0.026) and %dimM (25±19% vs. 16±11%, p=0.002) as well as a lower MFI of MNDA expression in monocytes (73±26 vs. 85±27, p=0.010). Interestingly, in CMML cases no reduced expression of MNDA could be found in any of the cell lines analyzed which might argue in favour of regarding CMML as myeloproliferative rather than myelodysplastic disease. From the cytogenetic point of view cases with an aberrant karyotype had higher values for both %dimG (22±20% vs. 12±18%, p=0.001) and %dimM (33±24% vs. 23±21%, p=0.004) as compared to those with a normal karyotype. Conclusions: These data suggests that MDS, as compared to non-MDS bone marrow, is associated with a reduced expression of MNDA in different cell lines which in part also applies to AML. Further analyses should define the potential of MNDA assessment to improve the MFC-based approach to diagnose MDS. Disclosures: Kern: MLL Munich Leukemia Laboratory: Employment, Equity Ownership. Machado, Wilson Pantoja. (2018). Superendividamento: a responsabilidade pré-contratual do credor.

Abstract:
He walks down the hall toward you. You are the only two in the hallway. You look at him, ready to smile and offer a cheery “Good morning!” With eyes downcast, he walks by you as if you are not there. What is your reaction? • Indignation. “Who does he think he is, not bothering to even look at me?” • Irritation. “Why can’t he make the effort to say a simple hello?” • Superiority. “With such poor social skills, he shouldn’t even be a teacher.” • Meanness. Silently you smirk at his indifference, eager to tell co- workers about this perceived slight. • Affirmation. “I knew he didn’t like me, and since no one is around, he doesn’t even pretend!” • Anger. “How dare he act so superior!” It may be a challenge to interpret this scene as anything but snobbery or poor manners. Yet the same curiosity and compassion that help us determine what our students need in order to learn can serve us well with other adults. The illusion in this scenario is our belief that we know the reason for another’s behaviors. The pitfall of this conclusion is the missed opportunity to wonder with good intentions. The risk to professional relationships is the narrow labeling of another’s behaviors and personalizing their intent. Asking ourselves “What else could it be?” implies that we have the power and presence of mind to press the pause button to offer a broader palette of possibilities. • Is he preoccupied with an upcoming meeting? • Is he concerned about a loved one? • Is he mentally preparing for his first class? • Is he an introvert who is much more comfortable keeping to himself than offering easy banter or automatic greetings? Why not be your best by offering a warm greeting to him with no expectation for receiving one in return? When faced with puzzling behaviors, we can avoid jumping to conclusions. We can take a moment, have a second thought. This opening of possibility offers a moment of grace to ourselves and our co- workers. We choose to suspend judgment for the good of the moment and for nourishing goodwill in schools.

Abstract:
“Don't pick the hard stories, sweetheart,” an editor told me long, long ago. “Those are the ones that will break your heart.” Nonsense, I thought. I was young and ambitious and eager to chase a story through multiple all-nighters. He was old and wily and appreciated those stories that would glide through the copy desk and get him home in time for a glass of scotch and dinner with the family. Now, more than 20 years after getting that good advice, I too appreciate the easy stories. But I'm still trying for the hard ones. Every few years, if I'm lucky, I manage to pull one off. When I do, the small, secret joy of having done so sustains me through months of too-short deadlines and too-tight space. In thinking about what elevates a story from okay to prizewinner, from another day at the office to the top of the clip file, I think again about that long-ago editor, a grizzled veteran of the Saturday Evening Post. Don't try to be different, he said. Write about what everyone else is writing about. Those are the big stories, the ones that matter. And he was right. In covering science and medicine, we're blessed with big stories galore. Cloning, cancer, Mars exploration, anthrax, the Big Bang, climate change, nanotechnology, heart disease—it's birth, death, creation, the meaning of life. If that can't get you on page Ai, what can? But that very abundance, and the flood of data that bears those stories along, make it all too tempting to settle for the easy get—to write off the journals, take your lead from the New York Times, and get by. A great story demands more. I like to think of journalism as bricklaying—a noble craft, but a craft all the same. To build a wall, I need bricks. To build a noble wall, I need the best bricks ever. Facts are the bricks of a story, and finding the right bricks requires serious reporting. I can't say that exhaustive research and reporting will guarantee a great story, but I've never been able to pull one off without it.

Abstract:
In this chapter, we will create a program that randomly generates pitches at a specified tempo. The program will have the ability to change a number of musical variables including timbre, velocity, and tempo. We will also write a program that allows your MIDI keyboard to function as a synthesizer. These two programs will be the basis of future projects related to composition and performance. Since you’ve already learned a number of objects in the previous chapter, let’s agree that when you’re asked to create an object that you already know, like button, for example, it will be sufficient for me to say “create a button” instead of repeating the process of creating a new object box and typing in the word button. Combining steps in this way will help us to get through the instructions with greater speed while reinforcing your understanding of how certain objects work. In this way, the instruction “create a message box containing the numbers 41 and 38” actually combines several smaller, and hopefully intuitive, instructions into a single step. I will slowly stop mentioning key commands and other shortcuts for objects and tasks that I’ve already introduced. Create a new patch and 1. Create a new object (press n) called random The random object takes a number as its only argument and randomly generates a number between 0 and one less than the argument when it receives a bang in its inlet. 2. Give this random object the argument 128 (Note: if you already clicked away from the object, double click it in order to, once again, enable typing within the object box) Be sure to put a space between the word random and the argument 128 or else Max will look for an object called random128 that does not exist. 3. Create a new button (press b) 4. Connect the outlet of button to the first inlet of random 128 5. Create a number box (press i) 6. Connect the outlet of random 128 to the inlet of the number box
