Abstract:
Activation of endothelial cells (EC) by inflammatory stimuli can result in heightened procoagulant properties towards coagulation proteins. These alterations can be readily studied in existing tissue culture models which lack a component of flow (shear). It is generally believed that thrombosis in the venous (low shear) regions of the vasculature are relatively dependent upon the coagulation proteins, whereas clots in the arterial system (high shear) are more dependent upon platelet plugs. Adhesion of the platelets to vessel wall elements under controlled conditions of flow have also been addressed in a number of models, such as the "Baumgartner chamber". In this model, blood or platelets are perfused over denuded, everted vessel segments, usually rabbit aortae. We have modified this model to study attachment of platelets to arterial segments of human origin which retain an intact layer of native EC. Arteries are aseptically removed from umbilical cords, trimmed, washed, and gently everted. The segments are placed in Medium 199 with 20% calf serum and standard growth supplements. After 4-10 days, these segments are placed on rods and perfused with whole citra-ted blood (800 sec−1). Such segments permit attachment of single platelets, but not platelet aggregates. Identical segments from which EC are mecnanically removed promote attachment of large (>5 cell) platelet thrombi. Endothelialized segments which are pre-treated (16 hours) with thrombin or phorbol myristate acetate show reduced attachment of platelets. In selected regions, clusters of leukocytes are attached to these segments, either upon or beneath the still- confluent layer of EC. Artery segments treated with bacterial endotoxin (1 mcg/ml) for 90 minutes, by contrast, show enhanced attachment of platelets, both as a monolayer of platelets and as small (2-5 cell) aggregates. This novel application of the Baumgartner assay demonstrates that platelet and leukocyte attachment to confluent EC may occur under conditions of moderate shear, and may be regulated in part by EC, which in turn may be modulated by exogenous mediators.

Platelet adhesion studies in perfusion chambers are generally performed with constant flow rate or with pulsatile flow generated by a roller pump. No information is available about the effect of flow pulsatility in the physiological range. For this purpose we devised a system consisting of a eccentric rotating disc which forces a driver toward a flexible tube. This movement causes a sinusoidal laminar flow component. The effect of this sinusoidal pulse on platelet adhesion was studied with the annulai^perfusion chamber of Baumgartner using umbilical arteries and In-labeled platelets. Conditions used are described below, where γ [1/s] is the constant flow component, γ0 [1/s] is the amplitude of the sinusoidal component and f [beats/min] is the frequency of the pulse (70 beats/min standard). Adhesion values are expressed as 105 III In platelets/cm2 (n=4).When osciallatory flow (abence of constant component) was generated, platelet adhesion incersed with increasing amplitude of the pulse.These data indicate that pulsatility as persent in the the aorta and large arteries has no effect on platelet adhesion.

Native Von Willebrand factor (NvWF) binds to platelets activated by thrombin, ADP or ristocetin, and also supports the adhesion of platelets to subendothelium at high shear rates. In contrast, asialo von Willebrand factor (AvWF) induces platelet aggregation in absence of platelet activators. We investigated the role of AvWF in supporting the adhesion of platelets to rabbit vessel subendothelium under flow conditions at a shear rate of 2000 sec-1 for 5 min using the Baumgartner perfusion system. We also studied the effects of blockage of platelet GPIb or GPIIb/IIIa on platelet adhesion using monoclonal antibodies (Mabs),and we measured the rate of binding of 111I-labeled NvWF and AvWF to subendothelium. Perfusates consisted of washed platelts and red cells resuspended in a 4% human albumin solution to which increasing concentrations of NvWF or AvWF had been added. Platelets interacting with the perfused vessels were evaluated morphometrically using a computerized system. At a concentration of 1.2 /ig/ml the percentage of total coverage surface was 21.3 ± 4.8% and 40.0±14.6%, for NvWF and AvWF, respectively (p<0.01). Addition of either Mab against GPIb (LJlbl) or against GPIIb/IIIa (CP8) to the perfusates, reduced platelet deposition (p <0.01). The rates of binding of 111I-labeled NvWF and AvWF to perfused vessel subendothelium were similar (0.83±0.1μg and 0.95±0.1 μg ,respectively).Our results indicate that AvWF enhances the interaction of washed platelets with the vessel subendothelium under flow conditions. Furthermore, they suggest that this effect is related to the interaction of AvWF with platelets and not to an increased affinity of AvWF for subendothelium.

Abstract:
Desmopressin (DDAVP) shortened the bleeding time and increased the platelet retention on glass beads and the platelet interaction on subendothelium measured by the Baumgartner perfusion system in 11 uremic patients in whom the three tests were abnormal previous to the treatment (1).Patients were chosen at random out of a group of 30 with prolonged bleeding time and decreased platelet retention on glass beads. All tests were performed on blood drawn before and one and six hours after a single dose of DDAVP (0.4 μg/kg body weight). Perfusion experiments were carried out at a shear rate of 800 sec-1.Levels of VIII:C, VIIIR:Ag and RiCof were at the upper limit of normality before DDAVP and significantly increased one hour after treatment. The multimeric structure of vWF in pretreatment plasma was normal ; one hour after DDAVP larger multimers appeared.After injection of DDAVP, the perfusion studies using reconstituted blood with uremic PPP in presence of isolated normal platelets and washed red cells, showed a statistically increased surface coverage and platelet aggregate formation on subendothelium (p < 0.05 respectively when compared to the pretreatment values). In the same perfusion assays, the in vitro addition to pretreatment plasmas of 1 u/ml of purified vWF with normal multimeric structure, or purified VIII:C and vWF (1 u/ml each) did not modify the decreased platelet interaction on subendothelium.These results confirm the shortening of bleeding time by DDAVP in uremic patients and reveal an increase of platelet interaction on vessel subendothelium mediated by a factor present in PPP. Abstract:
This paper presents and discusses the results obtained from a parametric study on the Baumgarte stabilization method for forward dynamics of constrained multibody systems. The main purpose of this work is to analyze the influence of the variables that affect the violation of constraints, chiefly the values of the Baumgarte parameters, the integration method, the time step and the quality of the initial conditions for the positions. In the sequel of this process the formulation of the rigid multibody systems is reviewed. The generalized Cartesian coordinates are selected as the variables to describe the bodies’ degrees of freedom. The formulation of the equations of motion uses the Newton-Euler approach that is augmented with the constraint equations that lead to a set of differential algebraic equations. Furthermore, the main issues related to the stabilization of the violation of constraints based on the Baumgarte approach are revised. Special attention is also given to some techniques that help in the selection process of the values of the Baumgarte parameters, namely those based on the Taylor’s series and Laplace transform technique. Finally, a slider crank mechanism with eccentricity is considered as an example of application in order to illustrated how the violation of constraints can be affected by different factors such as the Baumgarte parameters, integrator, time step and initial guesses.

Abstract A new automatic constraint violation stabilization algorithm for numerical integration of Euler-Lagrange equations of motion of multibody system dynamics based on Baumgarte constraint violation stabilization method and Taylor expansion of constraint functions is presented. The constraint equations may be holonomic or nonholonomic. The parameters α, β, σ used in Baumgarte’s method are determined automatically according to integration step. Some numerical examples compare the accuracy between the traditional method and the one suggested in this paper are presented finally.

Abstract:
This paper develops a discrete Hamiltonian system with holonomic constraints with Geometric Constraint Stabilization. It is first shown that constrained mechanical systems with nonconservative external forces can be formulated by using canonical symplectic structures in the context of Hamiltonian systems. Second, it is shown that discrete holonomic Hamiltonian systems can be developed via the discretization based on the Backward Differentiation Formula and also that geometric constraint stabilization can be incorporated into the discrete Hamiltonian systems. It is demonstrated that the proposed method enables one to stabilize constraint violations effectively in comparison with conventional methods such as Baumgarte Stabilization and Gear–Gupta–Leimkuhler Stabilization, together with an illustrative example of linkage mechanisms.

Abstract:
Abstract A relative coordinate formulation of the Hamiltonian equation of motion is derived for realtime vehicle simulation. The Baumgarte stabilization method [1] is adopted to solve the Differential-Algebraic Equations (DAE) of motion. The stability theory of multi-step integration methods is used to determine the stabilization constant. The equations of motion are first derived in Cartesian space and are reduced to relative coordinate space using the velocity transformation method [2]. Partial derivative of the kinetic energy with respect to the relative coordinate is obtained from equivalence of the Lagrangian and the Newton-Euler formulations. Parallel processing of the formulation is also discussed. Realtime simulation of a passenger vehicle is carried out to demonstrate the efficiency of the new formulation.

Abstract:
This paper presents an addendum to the Recursive Coordinate Reduction (RCR) algorithm for the efficient numerical analysis and simulation of modest to heavily constrained multi-rigid-body dynamic systems. The RCR algorithm can accommodate the spatial motion of broad categories of multi-rigid-body systems containing arbitrarily many closed loops in O(n + m) operations overall for systems containing n generalized coordinates, and m independent algebraic constraints. The presented approach does not suffer from the performance (speed) penalty encountered by most other of the so-called “(n)” state-space formulations, and does not require additional constraint violation stabilization procedures (e.g. Baumgartes method, coordinate partitioning, etc.). Due to these characteristics, the presented algorithm should offer superior computing performance relative to other methods in many situations involving both large n and m. This paper will specifically address an unpublished recursive step in the handling of “floating” loop base bodies, as well as present an extension to “spur” topologies.
