Abstract:
Neste trabalho, analisamos a obra Mulheres que mordem (2015), de Beatriz Leal com o objetivo de identificar como a escritora explora, a partir de metáforas literárias os diferentes tipos de violência. A abordagem teórica está voltada o texto literário e o papel do crítico como responsável por ler a obra de uma perspectiva diferente do leitor. Foucault (2014); (2021), Motta (2002), Frye (1957), Culler (1999) e Norman Friedman (2002) conferem, portanto, robustez a esse corpus teórico, que tem como perspectiva metodológica a pesquisa bibliográfica. Mulheres que mordem é narrada a partir de vários pontos de vista que coincidem com o narrador onisciente seletivo, além de ser escrita majoritariamente em modo sumário, com poucas cenas descritivas. O simbolismo das metáforas empregadas pela autora mostra quatro personagens que mordem para obter o controle de si mesmas e do mundo que as cerca. Morder, neste sentido, está tanto no campo do real quanto no universo simbólico.

En este trabajo analizamos el libro Mulheres que mordem (2015), de Beatriz Leal, con el fin de identificar cómo la escritora explora, a partir de metáforas literarias, los diferentes tipos de violencia que experimentan los personajes Rosa, Laura, Elena y Clara. El enfoque teórico se centra en el texto literario y el papel del crítico como responsable de leer la obra desde una perspectiva diferente a la del lector. Foucault (2014); (2021), Motta (2002), Frye (1957), Culler (1999) y Norman Friedman (2002), por lo tanto, dan solidez a este corpus teórico, que tiene como perspectiva metodológica la investigación bibliográfica. Bite Women está narrada desde varios puntos de vista que coinciden con el selectivo narrador omnisciente, además de estar escrita mayoritariamente en modo sumario, con pocas escenas descriptivas. El simbolismo de las metáforas utilizadas por la autora muestran los cuatro personajes que muerden para hacerse con el control de sí mismos y del mundo que les rodea. Morder, en este sentido, está tanto en el ámbito de lo real como en el universo simbólico.

Hablar de críticas musicales brasileñas durante la primera mitad del siglo XX, dada la preponderancia de Río de Janeiro, es hablar de críticas musicales cariocas. Mediante la escritura, estas “mujeres nuevas” desarticularon la norma hegemónica del hombre-redactor. En este artículo delineamos algunas directrices sobre la crítica musical femenina en Brasil, temática no abordada en los debates académicos. Además de resaltar la labor de cronistas decimonónicas y finiseculares de rotundos ideales feministas, como Joana Paula Manso de Noronha y Sol Menor, y de refutar la instalación de Oscar Guanabarino y Mário de Andrade como precursores de la crítica musical brasileña, presentamos a Lúcia Branco, Beatriz Leal Guimarães, Ondina Portella Ribeiro Dantas, Magdala da Gama Oliveira y Mariza Lira. Defensoras de la música de arte, polemizaron sobre quiénes encarnaban “lo brasileño”. La pugna entre lo docto y lo popular —lo inocuo versus lo nocivo— es una prueba del racismo y el clasismo adheridos y propagados por los medios de comunicación.

Abstract:
O presente artigo insere-se no campo de estudo das relações entre literatura, história e memórias coletiva e individual. propõe-se o estudo da representação feminina republicana e das relações entre mulheres nos romances Historia de una maestra (1990), Mujeres de negro (1994), La Fuerza del destino (1997), da escritora Josefina Aldecoa, e Mulheres que mordem (2015) de Beatriz Leal, obras da literatura espanhola e hispano-americana que tematizam a Guerra Civil Espanhola, a Segunda Guerra Mundial e o período ditatorial tanto na Espanha quanto na Argentina, a partir de um protagonismo feminino. O Corpus literário é abordado com base em reflexões teóricas que partem do conceito de memória; trata-se de pensar como as rememorações das personagens surgem na fratura do discurso histórico e como influenciam no transcorrer dos romances. Assim, pretende-se refletir de forma crítica e analítica sobre tais narrativas, junto de argumentações embasadas em autores que trabalham nos campos, em confluência, da memória e da história, tais como Le Goff (2003), Halbwachs (1990), dentre outros. O trabalho possui o intuito de contribuir para as discussões sobre a memória coletiva e individual e a resistência feminina em contextos de opressão social espanhóis e argentinos, em especial durante o contexto da guerra e pós-guerra.

Abstract:
Abstract Background: RET (rearranged during transfection) is a proto-oncogene encoding for a tyrosine kinase membrane receptor that promotes protein synthesis and cell proliferation. A small percentage of human tumors harbor RET fusions or mutations as oncogenic drivers and RET-TKIs have been developed. Two of them, selpercatinib and pralsetinib, have recently been approved. However, their effect on tumors overexpressing wild-type RET (wt-RET) has not been investigated. A large-scale mRNA analysis performed by our group revealed that ~2% of lung tumors present high expression of wt-RET in absence of fusions. Interestingly, ~60% of high wt-RET tumors presented neuroendocrine characteristics. Methods: Twenty large cell neuroendocrine (LCNEC) and 19 small cell (SCLC) lung carcinomas from two additional institutions were analyzed by nCounter® to validate the previous results. For in vitro experiments, a panel of cell lines derived from lung NETs (DMS-53, NCI-H82, NCI-H727) was used, together with a lung adenocarcinoma cell line (LC2/ad, a positive control harboring the CDCC6-RET translocation). RET fusions and mRNA expression were analyzed by nCounter®, RET protein expression by Western blotting and immunohistochemistry (IHC) and RET mutations by next generation sequencing. The inhibitory effects of selpercatinib and pralsetinib were analyzed in vitro by MTT in 2D and spheroid/3D cultures. Finally, in vivo experiments implanting tumor cells into athymic nude mice are ongoing. Results: Among the 20 LCNECs and 19 SCLCs analyzed, 3 (15%) and 3 (15%) presented very high levels of wt-RET mRNA by nCounter, respectively. Fusions were absent in all cases. The NCI-H82 and NCI-H727 NET cell lines showed very low RET mRNA and protein expression by nCounter, Western blotting and IHC. In contrast, DMS-53 cells presented levels of RET mRNA and protein higher than the observed in the RET-fusion positive LC2/ad. In 2D models, the three NET cell lines tested showed moderate resistance to pralsetinib and selpercatinib, with IC50s of 1.5 - 9.7 µM for pralsetinib and 4.6 - 6.1 µM for selpercatinib; while the RET-fusion positive LC2/ad cells showed an IC50 in the nM range, as expected. In 3D cultures, DMS-53 was found to be sensitive to RET TKIs, with IC50s of and 0.3 µM for praseltinib and 2.5 µM for selpercatinib. In contrast, spheroids of the two RET low cell lines showed complete resistance to both drugs (IC50>10 µM). Experiments in animal models are currently ongoing, final results will be presented at the meeting. Conclusions: Overexpression of RET is frequent in lung tumors with neuroendocrine features (LCNEC, SCLC). Selpercatinib and Pralsetinib show antitumor activity in lung NETs cells expressing high levels of wt-RET. Pesquisadores e jornalistas dedicam-se a compreender que tensionamentos abalam o sistema de produção de sentido que até então ostentava certa hegemonia como discurso que representa um presente social de referência (GOMIS, 1999). Este artigo reflete sobre o modo como a crise do jornalismo tem aparecido nos discursos e nas práticas da própria imprensa. A suspeita inicial é a de que a crise configura-se em acontecimento silenciado pela mídia hegemônica. Por outro lado, inevitavelmente ela transparece também nas práticas jornalísticas, uma vez que tem atingido de forma intensa a estrutura de funcionamento das redações. Além disso, tem provocado os jornalistas a reverem suas competências e o campo a transformar - de certo modo - seus pressupostos e modos de fazer.PALAVRAS-CHAVE: crise do jornalismo; práticas; hegemonia; futuro do jornalismo. ABSTRACTResearchers and journalists are dedicated to understand the tensions that shake the production system of journalism, which has had certain hegemony as social reference speech (GOMIS, 1999). Abstract:
O presente artigo tem a intenção de fazer um diálogo entre as origens e os fundamentos do racismo ambiental no território brasileiro e suas consequências psicossociais nas periferias urbanas e/ou rurais do país. Partindo do entendimento que o racismo é um potente fator de distribuição seletiva das pessoas no seu ambiente físico, trouxemos alguns dados de saúde e da recorrência de crimes ambientais em territórios que estão em situação de vulnerabilidade socioambiental. Nossa conversa almeja, sobretudo, localizar as contribuições e reforçar a importância do campo da psicossociologia de comunidades e da ecologia social no acolhimento sensível da população, recorrentemente, vitimada pelo racismo ambiental e pela injustiça climática e social. Ants are important agents of seed dispersal and they can play a fundamental role in the establishment and survival of plants (Beattie 1985). Leaf-cutting or fungus-growing ants (Myrmicinae: Attini) are dominant elements of the ant fauna throughout the neotropics. They collect different types of vegetative and reproductive plant material that serve as substrate for underground fungus gardens,which are the only food source for their larvae (Stradling 1991).

Abstract:
This article begins by examining James Beattie's conception of speculative ethics, which he regards as the study of the foundation and nature of virtue. This leads to a discussion of the moral sense, or conscience, which Beattie claims is part of the nature of every rational being and which is designed to lead us to a virtuous life. Given this, I ask why Beattie thought himself warranted, or even needed, to dispense practical ethical advice. Answering this involves looking at Beattie's views on the importance of proper education, as well as the role played by his acceptance of providential naturalism. Beattie's answer is not only consistent with his ‘lecturing others’ as to their practical duties, his understanding of the relation between the speculative and the practical also allows him to respond to what contemporary ethicists call the Application Problem. A comparison with Reid's ethical thought will help bring out this latter point.

Selma Castilho and Dra. Beatriz Guitton. The award was presented on July 29th during the 2005 Intensive Care Society of Rio de Janeiro Conference. Simone Pereira Machado's research was astounding because it question the validity of Braden and Cubbin and Jackson scales to prevent pressure ulcers for patients at the ICU. This is important because this kind of research often provides the basis for the development of new, knowledge-based nursing care and high-standards professional patterns.This award is so important for us because it confirms that high-quality scientific research for professional problems goes hand in hand with high-quality university education.This research is supported by the Hospital de Clínicas de Niterói (HCN).

Based on the problematization of the lack of recognition of black intellectual productions and artists as an episteme, and in particular, of Afro-referenced content in Brazilian musical literature, this article seeks to lead the reader to reflect on the processes of musical teaching and learning, from a decolonial and Afro-diasporic perspective. Seeking to weave a dialogue with the concept of black-body-territory, by the black historian and intellectual, Beatriz Nascimento, the text seeks to relate how the black body is inscribed in different environments based on the awareness and resumption of its ancestry, culture and origin. In the lines that are woven here, I seek to provoke the reader to the need to decentralize the hegemonic discourse of white-European tradition, which are undertaken in debates and recorded in books of musical theory and analysis, as well as in books of music history. Thus, the aim is to understand how the mechanisms of racial, class and gender oppression operate in the perpetuation of the symbolic erasure of the history and influence of the African continent on our musicality.

Abstract:
The tomato (Lycopersicon esculentum Mill.) is the most important crop under greenhouse conditions and covers 73% of the area under that system in Mexico. To increase fruit quality and yield, leaf removal is necessary to produce a balance between the vegetative and generative growth of tomato production. The objective of this experiment was to determine the effect of leaf removal on the tomato quality and production under greenhouse conditions. The evaluation was carried out in the Experimental Station INIFAP-CIRNO inside a polyethylene greenhouse. The date of seedling establishment was 28 Sept. 2003. In this trial, we used the Beatrice variety and a plant density of 3.78 plants/m2. We compared four treatments of leaf removal: 1) two leaves below the cluster; 2) two leaves below the cluster and later to remove those leaves when fruit reaches commercial size; 3) severe leaf removal; and 4) the control with no leaf removal. The results show significant differences between the intensively of leaf removal. The highest yield values corresponded to the treatment 2) with a yield of 281 t·ha-1 and fruit weight of 153 g vs. 202 t·ha-1 and 137.7 g of fruit weight for the control treatment.

empresas e a sua exigibilidade pelos stakeholders.O nono artigo intitulado “Investigação defensiva: a evolução do tema e os problemas de sua aplicabilidade”, de Marcelo Navarro Ribeiro Dantas e João Carlos Faria da Costa analisa as atividades de investigação defensiva, abordando a partir dos precursores do tema até a materialidade de disposições normativas a respeito, trazendo seus contornos doutrinários e práticos, buscando uma persecução penal que seja a mais justa possível.Por fim, no último artigo, “Atuações no mercado de capitais combinadas em redes sociais: apontamentos para a atribuição de responsabilidade administrativa e penal por manipulação de mercado”, Marcelo Costenaro Cavali e Natália Naomi Ikeda examinam os elementos que devem ser considerados pelas autoridades públicas, para efeito da imputação de responsabilidade, administrativa e penal, em casos de atuações coordenadas de investidores, combinadas em redes sociais, com o objetivo de provocar a elevação dos preços de valores mobiliários, bem como o crime de manipulação de mercado e a responsabilização administrativa e penal dos líderes dessa conduta.Por derradeiro registro agradecimentos especiais à After a close analysis of Fazi’s main contribution to a new understanding of computational aesthetics, we will briefly outline the potential implications of her work to understand the contemporary evolution of creative software, and especially the implementation of machine learning algorithms in these kinds of software. This will lead us to contextualise the contemporary anxieties regarding how machines could replace humans in the act of artistic creation.

Following initial spinal cord injury (SCI), a cascade of pathological events, including oxidative stress, leads to secondary injury. Glutathione (GSH) plays a critical role in oxidant scavenging. Maintenance of GSH concentrations after SCI lessens secondary injury and improves recovery. Since glutamine promotes GSH synthesis, this nonessential amino acid was examined for therapeutic potential. Denervation alters the expression of myosin heavy chain (MHC) isoforms within skeletal muscles. The hypotheses of this study were that glutamine administration to SCI rats would lead to improved functional recovery and more preserved MHC phenotypes in representative locomotor muscles. Male Wistar rats were divided into four groups: healthy, sham with laminectomy, laminectomized SCI untreated, and laminectomized SCI treated with glutamine. Functional performance was measured weekly for 6 wk using Basso-Beattie-Bresnahan scale and angle board methods. MHC composition of rat soleus and extensor digitorum longus muscles was determined using SDS-PAGE. Glutamine-treated rats had significantly higher angle board scores ( P < 0.001) and Basso-Beattie-Bresnahan scores ( P < 0.01) than untreated SCI rats. Soleus of healthy rats contained 94% type 1 myosin isoform. Treated rats maintained 68%, which was significantly ( P < 0.001) greater than 28% in untreated rats. The extensor digitorum longus of healthy rats contained 55% type 2b myosin. There was a significant ( P < 0.001) decrease in this isoform following SCI, but no significant difference between treated and untreated groups. There were strong correlations between higher functional scores and more preserved MHC phenotypes. Our findings suggest glutamine improves functional recovery and helps preserve myosin profile by reducing secondary SCI, thereby maintaining more nerves.

The paper examines the changing cultural perceptions of one of the most famous and mysterious geographic expeditions of the 19th century, John Franklin’s search for the Northwest Passage. Analyzed herein in closer detail are two novels: The Discovery of Slowliness by Sten Nadolny and Wanting by Richard Flanagan, as well as three texts by Margaret Atwood: the short story The Age of Lead from the collection Wilderness Tips, her Oxford lecture “Concerning Franklin and his Gallant Crew”, published in the volume Strange Things. The Malevolant North in Canadian Literature, and the foreword to the reissue of O. Beatie and J. Geiger’s book Frozen in Time. The Fate of the Franklin Expedition. All of these tales of Franklin’s expedition reflect the vicissitudes of human fantasies about exploring unknown and menacing spaces – not just the mythical frozen North. They are also narratives about our relationship with Nature and all the fears and hopes associated with that relationship.

This study examines how Chimamanda Ngozi Adichie's Purple Hibiscus interrogates the oppressions of women in the Nigerian patriarchal society and how women empower each other lead them to women empowerment. The study shows how the oppressions of women is represented through female characters which perpetually put women in disadvantaged positions as portrayed in Purple Hibiscus. One of the most despicable oppression among the so well-known cultural practices in Nigeria is the patriarchal oppression. However, as the events unfold, efforts will be made in order to reveal of how African women are rated based on the good and real women as represented by Beatrice and Ifeoma. I argue that Adichie's approach to subvert patriarchal oppression describes that despite the struggle and pain, women assert themselves in the world of patriarchy through education and sisterhood. Adichie’s novel suggests women empowerment through social transformation confronted by women. The title of the novel, "Purple Hibiscus" may refer to a particular type of flower, but it also emphasizes the triumph of the innovative suggesting that the unusual is not necessarily bad as it looks which aims to women empowerment.

The discourse on women is one which cannot be over- emphasized. The United Nations in its Declaration on the Elimination of Discrimination Against Women in 1980, made expansion of opportunities for women as a key demand. The declaration contends among others, greater the participation of women in decision making. Hence, the present study examines women and leadership in modern African literature to account for their (women) involvement and the impact of their participation cum non participation, focusing on Chinua Achebe’s Anthills of Savannah. The aim is to ascertain women leadership and its impact in the wider society, with reference to Chinua Achebe’s Anthills of Savannah. The study adopts Feminism (Post structural perspective) as its theoretical consideration. Finding from the study shows that women are not adequately represented in leadership positions. However, in cases where they lead, they functioned optimally for the betterment of their society. Beatrice, for instance, a Senior Assistant Secretary in the Ministry of Finance, is a noble female leader who by her role and conduct stood out. She affected those around her positively and was a source of inspiration to Ikem, a male character, in the novel.

In this article, Robert LeVine, Sarah LeVine, and Beatrice Schnell develop and test a theoretical model of how women's schooling might contribute to social and demographic change. Drawing upon research from many disciplines, the authors propose that schooling leads to social change by imparting skills and fostering other individual changes that alter women's patterns of social participation. They argue that, in schools, girls acquire aspirations, identities, skills, and models of learning that eventually affect their decisions regarding reproductive, child-rearing, and health behaviors. Among other things, girls learn an academic register that, the authors argue, is the official language of all bureaucracies, including health and family-planning clinics as well as schools. Proficiency in using this academic language is advantageous in oral communication with the health bureaucracy, and may lead to greater utilization of health services and, thus, improved reproductive and health outcomes. After explaining their theoretical model, LeVine, LeVine, and Schnell present new findings from their research on maternal literacy in two very different settings, Nepal and Venezuela. The results of their quantitative analyses are consistent with the hypothesis that the literacy and language skills that women acquire in school provide an educational pathway to better health care.

Abstract:
The purpose of research is to establish the patterns of formation of weediness in crops depending on agricultural technologies and varietal composition of barley in forest-steppe agricultural landscapes of the Omsk Region. The studies were carried out in a long-term (2006–2021) stationary grain-fallow crop rotation on the trailing crop – barley and short-term experiments. The spring barley varieties Omsky 90, Sasha, and 5 brewing varieties were sown. It has been established that a more weedy agrophytocenosis is formed using extensive agricultural technology with zero tillage (on average up to 620 g/m2 – 44.2 %). With complex chemicalization, the biomass of barley increases by 2 times (from 752 to 1534 g/m2) while the weed component decreases by 4.2 times (from 44.1 to 10.6 %), with a predominance of bluegrass – 81–83 %. The relationship between barley yield and the biomass and number of dicotyledonous weeds is negative (up to 67–81 %). With the integrated use of chemical agents, plant damage by root rot is reduced by 19.8 %, leaf diseases of the upper tier of barley leaves, a decrease in the development of reticulate film – from 20.6 to 11.4 %, helminthosporiosis – from 10.3 to 5.8 %, brown rust – from 5.9 to 1.6 %. With intensive agricultural technology, the winner is resource-saving combined tillage in crop rotation (alternating moldboard and flat-cut) with a yield of 3.40 t/ha, the Beatrice variety – 2.74 t/ha with a protein content of less than 12 %.

First of all it’s important to recognize the fact that Vancouver is a town where educated and aware people think it’s clever to say that Shakespeare’s a bore. Vancouver theatre is entertainment theatre, like it or lump it, and even the greatest of plays must be made to conform. Hence, Richard Ouzounian, doing his M.A. thesis production at U.B.C. in 1972, was able to launch a stellar career by marrying Sondheim and the Marx brothers to Beatrice and Benedict in a production of Much Ado About Nothing, to set a tone for directorial approach that Vancouver theatre has still not been able to get past. Of course, it’s not fair to blame Ouzounian for all outlandish directorial concepts for Shakespeare. The precedents had been set in the sixties by Peter Brook with his gymnastic Midsummer Night’s Dream and existential Lear. Ouzounian’s awesome feat was to make it imperative that the post-Brookian director’s concept be fun. He put a joie in jeu that West Coast directors have been struggling with ever since. In their attempts to cram Shakespeare with entertainment values, they have frequently failed not only to make his plays more accessible, but also to impress audiences with any sense of the greatness of his writing. It isn’t my contention that these directors should not have attempted their radical pop interpretations of Elizabethan texts. I would suggest, however, that perhaps they failed because they didn’t go far enough. No matter how high their flashy stagings flew, when it came time to talk the whole thing bogged down in charmless anachronism.

Abstract Agonism of death receptor 5 (DR5), a tumor necrosis factor receptor superfamily member, induces the killing of tumor cells via the extrinsic apoptotic pathway upon receptor multimerization. We have engineered a multivalent IgM DR5 agonist antibody, aplitabart (IGM-8444), that induces robust anti-tumor activity in both in vitro and in vivo preclinical models. We also demonstrated potent synergistic in vitro activity of aplitabart with different classes of chemotherapeutic agents including microtubule inhibitors, nucleoside analogs and topoisomerase inhibitors in a range of solid cancer cell lines. Preliminary results from an ongoing phase 1a/ab clinical study show that aplitabart has an encouraging safety profile and promising activity in combination with FOLFIRI ± bevacizumab in heavily pretreated metastatic colorectal cancer patients. To further explore the combination therapies for aplitabart in lung and breast cancers, we evaluated standard of care chemotherapies, such as paclitaxel, carboplatin, and antibody drug conjugates (ADCs) in vitro on a range of human lung and breast cancer cell lines. We observed dose-dependent synergistic cytotoxic effects of aplitabart on tumor cells when combined with these agents. Additionally, these agents increased the expression of DR5 on tumor cells and could explain their synergistic combinatorial activity with aplitabart. Representative cancer cell lines were selected to evaluate these combinations in vivo using xenograft mouse models where robust anti-tumor responses were demonstrated in the xenograft lung and breast cancer models, resulting in enhanced tumor growth inhibition and extended survival. Our findings demonstrate potent in vitro synergistic cytotoxicity and enhanced anti-tumor response in preclinical models in vivo by combining aplitabart with ADCs, or carboplatin and paclitaxel. These results support the combination of DR5 agonist IgM antibodies with agents known to upregulate DR5 expression on solid tumors. Aplitabart is currently under evaluation in a randomized clinical study in combination with FOLFIRI + bevacizumab in comparison with FOLFIRI + bevacizumab in patients with colorectal cancer (NCT04553692). Abstract:
The paper discusses heraldic, cultural and art-historical aspects of two relief stone plaques from the former castle (Ožegovićianum) in Senj. Both plaques contain pairs of putti supporting leaf wreaths that surround specific heraldic representations. On the left panel it is the crowned coat of arms of the Neapolitan Aragon dynasty, recognizable by the characteristic alternation of fields with horizontal and vertical beams, stylized fleur-de-lis and multiple Jerusalem crosses. The more complex composition on the right plaque represents the Croatian-Hungarian lands under the rule of king Matthias Corvinus (Cluj-Napoca / Koloszvár, 1443. – Vienna, 1490; ruling: 1458-1490): its central place is occupied by a shield with four horizontal beams, surmounted by the royal crown and surrounded by six smaller coats of arms of Hungary (double cross); Dalmatia and Croatia (three crowned leopard heads); Beszterce (Bistriţa) County or the Bohemia (lion); the Hunyadi family (raven); Bulgaria or Slavonia (dog); and Galicia or Bosnia (crown). The arrangement and heraldic content of the coats of arms is identical to that of the so-called Second privy seal of Matthias Corvinus, of which an imprint in red wax is still kept in the City Museum in Senj. Given the above, the first coat of arms can be associated with Corvinus’ second wife, the Neapolitan princess Beatrice of Aragon (Naples, 1457-1508), whom the Hungarian-Croatian ruler married in 1476. This event also provides a firm terminus post quem for the Senj plaques, while their upper time limit is being determined by the Corvinus’ death in 1490. A comparative analysis of the Senj plaques (especially links with contemporary examples found in illuminated manuscripts) reveals their place in the context of the Pannonian Renaissance, testifying to the importance of Senj in the political and artistic topography of the Hungarian-Croatian Kingdom in the late 15th century.

Abstract We have developed a web based interactive platform cProSite (The Cancer Proteogenomic Data Analysis Site). It provides on-line proteomics and phosphoproteomics analysis for the data in The National Cancer Institute’s Clinical Proteomic Tumor Analysis Consortium (CPTAC), with a focus on tumor and adjacent normal comparisons and correlations between different protein or phosphorylation levels or between protein abundances and mRNA expressions. The web platform is designed for biologists and clinicians wishing to make rapid searches in a user friendly environment. The platform analysis has several advantages compared with standard analytical methods: 1) Faster analysis on-line; 2) More user-friendly; 3) Less need for bioinformatics expertise to perform analyses. Current cProSite includes 11 tumor types*. The web platform includes the following features: 1) Can compare selected protein abundance between tumors and normal adjacent tissues and visualization of fold-changes between paired tumor and control tissue. 2) Can compare selected levels of individual phosphorylation sites or levels of phosphorylation site per protein abundance and visualization of fold-changes between tumors and normal adjacent tissues. 3) Can correlate protein abundance, phosphorylation site levels, or phosphorylation site level per protein abundance between two selected proteins. 4) Can correlate selected protein abundance with its mRNA expression level if the data are available. The cProSite export feature allows users to download the analyzed data. In addition, search outcomes from cProSite can be used for clinical data validation and novel target identification. For example, cProSite can be used to validate findings from other studies; the upregulation of the Hyaluronan Mediated Motility Receptor (Rhamm/HMMR) protein abundance in non-small cell lung adenocarcinoma detected by tissue microarrays was confirmed by cProSite analysis. cProSite can also be used for hypothesis-generation and lead to hypothesis testing. In one example, cProSite identified several upregulated phosphorylation sites in the ECT2 (G2/M phase protein Epithelia Cell Transforming 2) protein in several tumor types. Consistent with that observation, mass spectrometry results showed increased phosphorylation levels from those phosphorylation sites in synchronized lung cancer cell lines in the G2/M phase. * The 11 tumor types in cProSite are: breast cancer, colon cancer, head and neck cancer, liver cancer, kidney cancer, lung adenocarcinoma, lung squamous carcinoma, ovarian cancer, pancreatic ductal adenocarcinoma, stomach cancer, and uterine cancer. Abstract Melanomas are responsible for about 80% of all deaths related to skin cancers. This is due to its high potential of metastasizing and developing resistance to treatments. Novel therapeutic modalities and the identification of biomarkers able to predict the prognosis are necessary. Studies have revealed the central role of long non-coding RNAs (lncRNAs) in regulatory networks controlling cell behavior. The disruption of these regulatory networks as a consequence of altered expression of lncRNAs can contribute to cancer development and progression. Our laboratory has developed a linear cellular model of melanoma progression consisting of distinct cell lines: melan-a (melanocytes), 4C (pre-malignant melanocytes), 4C11- (undifferentiated, slow-growing and non-metastatic melanoma cells) and 4C11+ (differentiated, highly proliferative and metastatic melanoma cells), which were analyzed for their lncRNA expression profile. Differentially expressed lncRNAs identified by RNAseq were selected by in silico analyses based on their proximity to differentially expressed coding-genes in the same melanoma model. Correlation analyses were performed between the expression of differentially expressed lncRNA neighboring genes and their melanoma patient survival, using two independent melanoma cohorts (TCGA and Leeds). Among those lncRNA located near genes presenting prognostic value is the lncRNA Gm20619, here named Slamon, neighbor to Slc25a13 gene (mitochondrial aspartate/glutamate carrier gene), which high expression correlates with poor prognosis. Both Slamon lncRNA and Slc25a13 are highly expressed in the metastatic melanoma cells 4C11+ compared to melan-a, 4C and 4C11- cell lines. The knocking down of Slamon or Slc25a13 were performed in 4C11+ melanoma cells, followed by analyses of collective migration, clonogenicity, proliferation, anoikis resistance, and dacarbazine and MEK inhibitor treatment. By knocking down Slamon lead 4C11+ cells significantly less migratory and less resistant to dacarbazine, and resulted in increased expression of Slc25a13 gene. 4C11+ melanoma cells knocked down for Slc25a13 also presented reduced migratory, clonogenicity, proliferation and anoikis resistance capability, diminished sensitivity to dacarbazine and MEK inhibitor treatment, and increased expression of Slamon lncRNA. The analysis of genes coexpressed with Slc25a13 revealed genes related to tumor progression, as Cdk15, Lsp1 and Pdgfrb. Our results suggest a crucial role of the lncRNA Slamon and Slc25a13 coding gene in melanoma progression which has never been described before. Slamon regulates tumor aggressiveness associated with Slc25a13, forming a regulatory network. These findings reveal the importance of lncRNAs in melanoma biology, potentially indicating transcripts that can serve as prognostic biomarkers and therapeutic targets in melanoma. This article aimed to evaluate the efficiency of public expenditure in the municipalities of the State of Amazonas with primary education in the years 2013 to 2017 by building a ranking of the efficiency of the municipalities relating the results obtained in IDEB by each city in the period, under the counterpart of the values of expenses paid for primary education, the average number of students enrolled in the period, average expenditure per student and GDP per capita of each municipality.

Magazine Luiza, founded in November 1957, a national retail icon, started in 2011 at B3, with exponential growth in the valuation of its shares. This case provides the following pedagogical objectives: a) to promote the problem analysis, to propose and inquire decisions regarding valuation; b) stimulate reflection between the theory that supports the evaluation purposes and the way it should be developed and applied; c) expand the ability to relate the valuation theoretical assumptions of valuation and their adequacy to reality.
 Article 2 (Section 4) - Budget Planning for education: overcoming a challenge of Jander Ângelo Diogo Ferreira, Naiany Gabrielle Pereira Lima, Juliana Saraiva Moreira, Odemir Vieira Baêta and Carla Beatriz Marques Rocha e Mucci. This teaching case was prepared from observations of issues experienced by the municipality of Cachoeiro de Santa Cruz, and adapted for didactic purposes, with the objective of assisting in the construction of an effective government budget planning. Its purpose is to foster reflection on the limitations and potentialitys of municipal budget management and also to propose solutions to funds for basic and fundamental education, in scenarios of material and financial restrictions.

Presentación En septiembre de 2017, la Asociación Colombiana de Alergia, Asma e Inmunología (ACAAI) y la Organización Mundial de Alergia (WAO) celebraron conjuntamente en la Ciudad de Cartagena, Colombia, dos eventos únicos: el XI Congreso Colombiano de Alergia, Asma e Inmunología y el Simposio WAO “Alergia a ácaros: de la ciencia básica a las aplicaciones clínicas”. Los organizadores se esmeraron a fondo en ofrecer un programa de contenido novedoso e interesante, pero también en lograr una convocatoria numerosa y de calidad que motivara la presentación de trabajos libres. Abstract Introduction: Intravesical Bacillus Calmette-Guerin (BCG) is an effective immunotherapy for non-muscle invasive bladder cancer (NMIBC). However, BCG treatment failure will lead to recurrence and tumor progression. In this study, urinary exosomes content (miRNA profile) was evaluated as a possible marker of BCG treatment response in NMIBC patients. Methods: Urine samples from patients with bladder cancer were collected at the time of surgery and during patient follow up to 1 year. Urine from healthy volunteers were also included. Clinical and pathological information such as tumor grade (High Grade (HG), Low Grade (LG)), depth of invasion (Ta, T1, CIS), and response to BCG treatment was also obtained. Exosome Isolation and total RNA extraction including microRNAs from cell-free urine after centrifugation were obtained. Library preparation for miRNA expression was done (QIAseq® miRNA Library) for Next-Generation Sequencing (NGS) analysis in a NextSeq 2000 single read platform, 75 bp with 15-20 million reads per sample. Reads were then queried against miRDeep2 software for identification. Only miRNAs having at least 20 counts considering all samples were included. After normalization, significantly and deferentially expressed microRNAs (>2-Fold) were selected for analysis. Bioinformatic analysis including sequence alignment was performed under the STAR-based approach. Identified microRNAs were then used to classify/predict the response to treatment and its relationship with other clinicopathologic variables. Results: A total of 56 urine samples from 13 patients were available/used for analysis including 10 High Grade Ta and 3 High Grade T1 patients. Urine from normal healthy donors (N=3) was also included. Clinicopathological features were patients with HGTa=10, HGT1=3 and 3 control samples. Regarding treatment response 9 patients were BCG responders and 4 BCG unresponsive. When compared to BCG unresponsive patients, BCG responders showed 45 differentially expressed miRNAs. Statistically significant differentially expressed miRNAs (Fold-change >2, p value <0.05) were 12 miRNAs, upregulated were miR132-3p (p=0.042); miR-187-3p (p=0.021); miR-409-3p (p=0.043) and miR1301-3p (p=0.048). Downregulated miRNAs were miR-let7-5p (p=0.007), miR-3605-3p (p=0.047), miR-140-5p (p=0.031), miR-500a-5p (p=0.051), miR-629-5p (p=0.039), miR-454-3p (p=0.05), miR-2110 (p=0.049) and miR-30c-5p (p=0.03). Interestingly, miRPathDABv2.0 analysis predicted those microRNAs be related to cancer, including bladder cancer. They showed targeting important pathways as PI3K.Akt, Wnt/B catenin and P53 signaling related with disease progression and treatment response. Conclusion: Our study supports the value of urinary exosomal microRNAs as non-invasive biomarkers to predict BCG treatment response in nonmuscle-invasive bladder cancer. Abstract INTRODUCTION: Breast MRI (bMRI) has become a standard of care (SOC) modality for both screening and diagnostic work-up of breast disease (1-2). Despite availability of larger bores, claustrophobia remains the most common reason for patients declining MRI (3). Anesthesia service can offer sedation during MRI improving patients’ acceptance. However, unlike routine MRI, bMRI is performed with patient positioned prone. Our anesthesiologists observed patients’ expectation of receiving general anesthesia, without understanding the risks associated with sedation/anesthesia in this particular setting and no prior trials of oral anxiolysis. OBJECTIVE: This project retrospectively reviewed bMRI exams with sedation for image quality and potential complication to identify areas for intervention such as provider education on anesthesiology risks in the bMRI environment and the implementation of an institution-approved oral anxiolysis program. METHODS: Patients receiving bMRI exams with sedation/anesthesia at a large academic cancer center were identified from the existing institutionally approved Report Imaging Quality Issue database after receiving institutional approval for this quality improvement project with waiver for consent obtained from Institution Review Board. Patient demographics, imaging data, and clinical notes were reviewed by two board certified breast radiologists with over 15 years of experience in MRI interpretation. Safety reports were reviewed in consensus between two radiologists and lead anesthesiologist for MRI department. RESULTS: Of 4844 bMRI exams, 33 were performed with sedation/anesthesia. Mean age was 60 years (range 37-77), with mean body mass index of 34 (range 21-45). Reason for anesthesia included claustrophobia in 13 (40%) patients, pain in 6 (18%), and 14 not clearly documented (42%). Eighteen of 33 (55%) exams were rated as poor quality, 7 (21%) as average, and 8 (24%) good or diagnostic. Most common reasons for poor quality included motion from snoring or pain and poor fat suppression due to body habitus. In 11/33 (33%), repeat imaging with still poor quality was documented in technologists’ comments. Length of exam appointment ranged from 52 – 155, average 90 minutes. Only one case documented facial bruising due to prone positioning and one case of positioning requiring up to 7 staff. DISCUSSION: Availability of on-site anesthesia has enabled some patients to receive bMRI. These patients present to MRI with expectation of sedation/anesthesia without understanding risks or prior trial of anxiolysis and most have a challenging body habitus contributing to difficulty in prone positioning and decreased image quality while under sedation/anesthesia. Development of an oral patient anxiolysis program and provider education may improve image quality and reduce utilization of anesthesia. Citation Format: H. T. The research is dedicated to the well-known visit of the Irish dramatist Bernard Shaw and the British MP Lady Nancy Astor to the USSR in 1931. However, it seeks to avoid the format of a clichéd observation of commonly known facts concerning their stay in the Land of the Soviets and aims to concentrate on previously unknown circumstances of the preparation of that trip organised by representatives of the People’s Commissariat for Foreign Affairs and diplomats of the Soviet Embassy in London, on the one hand, and the media struggle evoked by that visit within the Soviet and the British press. The novelty of the research is provided by the use of numerous archival documents (AVP RF, RGALI) as well as quoting Bernard Shaw’s private correspondence and his friend Beatrice Webb’s diaries that have not been translated into Russian. The methodological foundation of this article is built upon the biographical and cultural-historic approaches; it also employs narrative techniques in recreating the historical background of Shaw’s visit, and content analysis in commenting on the media publications, and methods of archival research per se. Having considered the abovementioned documents, one can conclude that despite Shaw’s avid interest in Russia, his visit there was to a large extent spontaneous, which, together with Nancy Astor’s unpredictable escapades, caused considerable difficulties to the Soviet officials. Nevertheless, the variety of experiences offered to the British guests as well as the satisfaction of both reasonable and whimsical requests made by the dramatist managed to produce a favourable impression on the company and lead to Shaw’s companions giving generally positive feedback of the trip to the USSR whereas Shaw himself exalted in rave reviews. As a result, the victory scored by the Soviet soft power instigated a deluge of publications in the British media that aimed to discredit Shaw and Lady Astor as central figures of the trip. However, it cannot be said that those angry and even harsh commentaries caused any serious damage to their reputation or influenced their opinions of the USSR at the time.

Since 2005, a disease of greenhouse tomatoes has been observed in the North Adelaide Plains of South Australia. Symptoms include chlorosis and necrotic lesions on the leaves, stunted plants, and leaves that are frequently distorted. Necrotic lesions typically had a light green or yellow margin and affected areas often fell out, leaving small holes. The onset of disease was generally associated with an increase in population of the greenhouse whitefly (Trialeurodes vaporariorum). Isometric virions (25 to 30 nm) were observed by electron microscopy in partially purified extracts, but no other virion types were observed. In 2008, reverse transcription (RT)-PCR products were obtained using primers specific for RNA-1 (TR1F/TR1R) and RNA-2 (TR2F/TR2R) of Tomato torrado virus (ToTV) (1) using a fresh diseased sample (isolate 2136) and a 2005 archived, lyophilized sample (isolate 1883). Cucumber mosaic virus was not detected in these samples by RT-PCR (data not shown). The RT-PCR products were cloned and sequenced. The partial 892-bp RNA-1 consensus nucleotide (nt) sequences of isolates 1883 (GenBank Accession No. GQ844869) and 2136 (GenBank No. GQ844871) were 100% identical. The partial 573-bp RNA-2 consensus nt sequences of isolates 1883 (GenBank No. GQ844870) and 2143 (GenBank No. GQ844872) were 99.8% identical. The partial RNA-1 and -2 nt sequences of isolate 1883 were 99.1% and 98.5% identical to the published ToTV sequences (RNA-1, GenBank No. DQ388879; RNA-2, GenBank No. DQ88880), respectively (2). For isolate 2136, the identities were 98.1 and 98.7%, respectively. The putative amino acid sequences were all 100% identical to the published sequences. The virus was graft transmitted to tomato cv. Grosse Lisse and Datura stramonium, in which typical disease symptoms and leaf chlorosis, respectively, were induced, and by mechanical inoculation to Nicotiana benthamiana, which displayed leaf chlorosis. All inoculated plant species indexed positive by RT-PCR for ToTV. Seeds were collected from known ToTV-infected plants and the seedlings were grown in isolation in an insect-proof glasshouse. None of the seedlings exhibited symptoms of virus infection, and ToTV was not detected in 97 seedlings from cv. Beatrice, 368 seedlings from cv. Ediez, or from 286 seedlings from cv. Loretto with the virus-specific RT-PCR assays. The pathway for entry of ToTV into Australia is unknown, and although seed transmission was suspected, no evidence for this could be found. Abstract INTRODUCTION: Since its introduction, the RENAL nephrometry score has proven to be an effective tool in medical decision-making and surgical planning for renal tumors and has been shown to be predictive of several important oncologic and operative outcomes. However, some studies have reported high interobserver variability, particularly within the endophycity or “E” portion of RENAL score. This variability can undermine the utility of the RENAL score in clinical management, as it may lead to inconsistent risk stratification and treatment recommendations. Here, we propose the use of center-based endophycity (CBE) scoring in place of the “E” score within RENAL as a means of facilitating interobserver agreement while maintaining utility. METHODS: Medical records for patients who underwent radical or partial nephrectomy for suspected renal malignancy at a single institution over a 9-year span were retrospectively collected. Exclusion criteria included unavailable imaging, multifocal disease, and incomplete pathology data. Preoperative imaging was evaluated with six independent observers manually assigning RENAL scores to each case, and three observers assigning CBE scores. CBE scores were determined by approximating the center of the tumor and assessing whether it was interior or exterior to the corticomedullary boundary of the kidney. Exophytic and endophytic tumors were given a score of 1 and 2 respectively. These scores were then integrated in place of the “E” score of RENAL for analysis. Kappa coefficient was used to analyze interobserver agreement, and logistic regression was used for predicting outcomes. RESULTS: A total of 288 cases met the inclusion criteria for this study. CBE scoring (kappa coefficient 0.77) had significantly higher interobserver agreement than RENAL “E” (kappa coefficient 0.17, p < 0.0001). Both traditional RENAL and RENAL with CBE were predictive of ISUP grade (p < 0.0001, p < 0.0001), tumor necrosis (p < 0.0001, p < 0.0001), and perioperative blood loss (p = 0.001, p < 0.0001). Traditional RENAL was predictive of malignancy (p = 0.001), while RENAL with CBE was not (p = 0.058). However, CBE independently predicted malignancy (p = 0.039), whereas RENAL “E” did not (p = 0.365). CONCLUSIONS: These results suggest that the RENAL “E” score is prone to particularly low interobserver agreement. When integrated in place of RENAL “E”, CBE scoring represents a promising modification to the RENAL nephrometry score, offering a more reproducible method for assessing endophycity in renal tumors while preserving much of the predictive power of the original model. These findings have important implications for improving the accuracy and consistency of preoperative renal tumor assessments. Further studies are needed to elucidate currently insignificant yet promising predictive data points from this study and to confirm our findings in other cohorts. The study was carried out in two curricular components, Spanish and English Literacies, of an Additional Languages undergraduate program, offered in a public university in Brazil. It aims to contrast the use of a holistic e-rubric, adopted to assess both the process and the outcome of the students' performances during an academic semester in which an interdisciplinary project was promoted, based on the theme of Happiness. Throughout the semester, a set of mentoring sessions aimed at qualifying the actors' assessment literacy. At the end of the academic semester, it was proposed that both professors and students used the same holistic rubric created for both subjects. The data showed that there were no statistical differences between the scores assigned by the professors and the self-rated scores by the students. Also, the final performances were higher with the adoption of the implemented assessment proposal, compared to the semesters in which there was no assessment through rubrics nor did the integrated work between both disciplines. The experience brought reliability to the professors about the students' sincere and adequate self-perception concerning their knowledge, and it proved to be favorable for the expansion, diffusion, and adoption of similar proposals in other contexts.ResumoEste texto está centrado na adoção de práticas avaliativas inovadoras no Ensino Superior. O estudo foi realizado em dois componentes curriculares, Letramentos em Espanhol e Letramento em Inglês, de um curso de Letras - Línguas Adicionais ofertado em uma universidade pública brasileira, tendo como objetivo contrastar a utilização de uma e-rubrica holística adotada para a avaliação tanto do processo quanto dos resultados de desempenhos discentes durante um semestre letivo em que se promoveu um projeto integrado a partir da temática da Felicidade. Ao longo do semestre, foram realizadas um conjunto de mentorias voltadas a qualificar o letramento em avaliação dos atores envolvidos. Ao final do semestre letivo, foi proposto que tanto as docentes quanto os discentes lançassem mão de uma mesma rubrica holística criada para ambas as disciplinas. Os dados evidenciaram que não ocorreram diferenças estatísticas entre as pontuações atribuídas pelas docentes e as pontuações autoatribuídas pelos discentes. Além disso, os desempenhos finais foram mais altos com a adoção da proposta avaliativa implementada, em comparação aos semestres em que não havia a avaliação através de rubricas e tampouco o trabalho integrado entre ambas as disciplinas. A experiência trouxe confiabilidade para as docentes sobre a autopercepção sincera e adequada dos discentes em relação ao seu conhecimento e se mostrou favorável para ampliação, difusão e adoção de propostas similares em outros contextos.ResumenEste texto se centra en la adopción de prácticas evaluativas innovadoras en la educación superior. El estudio se llevó a cabo en dos asignaturas, Literacidad en Español y Literacidad en Inglés, de un curso de Letras - Lenguas Adicionales que se ofrece en una universidad pública brasileña, con el objetivo de contrastar el uso de una e-rúbrica holística adoptada tanto para la evaluación de los procesos como de los resultados de las actuaciones de los estudiantes durante un semestre académico en el que se impulsó un proyecto integrado, con base en la temática de la Felicidad. A lo largo del semestre, se llevó a cabo un conjunto de sesiones de mentorías orientadas a calificar la literacidad en evaluación de los actores involucrados. Al final del semestre académico, se propuso que tanto las profesoras como los estudiantes utilizaran la misma rúbrica holística creada para ambas asignaturas. Los datos demostraron que no hubo diferencia estadística entre los puntajes asignados por las profesoras y los puntajes de las autocalificaciones asignadas por los estudiantes. Además, los desempeños finales fueron superiores con la adopción de la propuesta de evaluación implementada, en comparación a los semestres en que no hubo evaluación a través de rúbricas ni tampoco el trabajo integrado entre ambas asignaturas. Abstract Introduction: TV, a tissue factor (TF)-directed antibody-drug conjugate, is approved under accelerated approval in the US at a dose of 2.0 mg/kg IV administered every 3 weeks (Q3W) for adult patients (pts) with recurrent or metastatic (r/m) cervical cancer who have progressed on or after chemotherapy. TV is also being evaluated in several advanced solid tumors known to express TF, including SCCHN; antitumor activity has been observed at the approved dose level. Previous reports show that modifying the dosing schedule to optimize key PK parameters such as AUC, Cmax, and Ctrough can lead to further improvement in clinical efficacy. A population PK model based on 399 pts across clinical trials suggests that when TV is administered at 1.7 mg/kg IV on Days 1 and 15 of a 28-day cycle (Q2W), pts are predicted to achieve 24% higher AUC, a higher Ctrough level, and a lower Cmax, compared with the approved cervical cancer regimen (2.0 mg/kg Q3W). Here, we report the first analysis of TV 1.7mg/kg Q2W for r/m SCCHN that has progressed after prior platinum combination with or without immunotherapy. Methods: innovaTV 207 (NCT03485209) is an open label phase 2 multi-center study evaluating TV monotherapy or in combination for advanced tumors, including pts with r/m SCCHN. In Part C of the study, eligible pts could have received up to 3 lines of systemic therapy for r/m disease and must have received prior therapy with a platinum-based regimen and a checkpoint inhibitor (CPI), if eligible. TV was administered at 1.7 mg/kg IV Q2W. The primary endpoint was objective response rate (ORR), and secondary endpoints included safety and tolerability. Results: At data cutoff (28Nov2022), 15 pts with SCCHN were treated. The median number of prior lines for r/m disease was 2. All pts received prior platinum therapy and majority (93%) received a CPI. 67% of pts received cetuximab and 53% received taxanes, for r/m SCCHN. Confirmed ORR was 40% (95% CI: 16.3, 67.7), with 1 complete response and 5 partial responses. The safety profile was generally consistent with that observed across TV monotherapy clinical studies. 13 pts experienced a treatment-related adverse event (TRAE), most commonly asthenia (n=7), peripheral sensory neuropathy (PSN) (n=7), and vomiting (n=5). 4 pts experienced Grade ≥3 TRAEs. 2 pts discontinued treatment due to an AE (PSN and dry eye, n=1 each). 11 pts received 3 or more cycles of treatment (1 cycle = 28 days). Conclusions: For pts with r/m SCCHN who have progressed after prior platinum combination and immunotherapy, TV may be a promising treatment option. Preliminary data suggest encouraging antitumor activity at a higher exposure with Q2W administration while maintaining an acceptable safety profile. The innovaTV 207 study is still enrolling and different dosing regimens are currently being evaluated across advanced solid tumors. Abstract Introduction Insomnia and perceived cognitive impairments (PCI) affect 1 in 4 cancer survivors and negatively affects work productivity. This study explores the impact of cognitive behavioral therapy for insomnia (CBT-I) on work presenteeism and absenteeism in cancer survivors with comorbid insomnia and PCI. Methods A sub-sample of 42 Atlantic Canadian survivors with insomnia disorder and PCI who were currently working for pay were analyzed as part of a larger randomized clinical trial. Participants completed 7 virtual sessions of CBT-I. The Work Productivity and Activity Impairment Index Specific Health Problem Questionnaire (WPAI: SHP) was administered pre-and post-treatment. Absenteeism was defined as the percentage of time absent from work, while presenteeism was defined as the extent to which health problems affected productivity at work in the past 7 days. Costs were calculated by the human capital method. Significant changes in productivity loss were defined at ≥15% or 20% change in absenteeism and presenteeism, respectively. Descriptive statistics and paired samples t-tests were used to measure changes in productivity following treatment. Average net benefit was calculated by subtracting the average treatment cost from average treatment benefits after 1 year to reflect cost savings after CBT-I. The cost of treatment was estimated in terms of average cost per session and lost time at work. Results Participants (76% Female) were, on average, 50.4 years old with 16.6 years of education. Breast cancer was the most reported cancer type (43%). Overall rates of presenteeism were significantly lower at post-treatment (p=<.001, d=0.55), with 52.4% reporting significant reductions. Absenteeism rates from pre- to post-treatment were not significant (p=.11, d=0.20). Reduced productivity loss post-treatment was associated with an average savings of CAD$10,979/person/year. Average net benefits for the first-year post-treatment equaled CAD$9348/person. Conclusion CBT-I can improve work productivity in cancer survivors. Investments to increase and provide access to evidence-based treatment may lead to significant savings for individuals and businesses. Para isso, foi conduzido um experimento no Campus de Engenharia e Ciências Agrárias da Universidade Federal de Alagoas. O delineamento foi em blocos casualizados, com quatro tratamentos e cinco repetições (E1= 28/05/14, E2-11/06/14, E3-25/06/14 e E4- 23/07/14). A primeira época de plantio foi a que apresentou maior altura do dossel (2,25 m), índice de área foliar (4,0 m2 m-2) e rendimento agrícola (8,03 t ha-1). A E4 apresentou a menor altura do dossel (0,9 m) e a menor produtividade agrícola foi observada na E3 (5,9 t ha-1). Isso aconteceu porque com base no balanço hídrico da cultura, quando o plantio é realizado até o primeiro decêndio de junho há uma melhor distribuição da chuva o que maximiza o potencial da cultura. Abstract:
Compulsory notification provides a valuable means to monitor population health and indicate priorities for health policies and allocation of financial resources. Thus, it is also evident the need to stimulate adequate training, either at graduation or during professional practice. This study aimed to present the process of constructing an open educational resource on Compulsory Notification, created as a result of the lack of knowledge of the vast majority of students and health professionals and deficiency in teaching by higher education institutions on the subject. Materials and methods: this is an exploratory and descriptive study, which approach was qualitative, carried out in November 2018, from a discipline attended in a stricto sensu post - graduation course. Results: the construction of the resource occurred through six distinct steps presented throughout this article and which final product was the e-book about Compulsory Notification. Conclusion: The process of building the open educational resource presented in this article, along with the other information about Compulsory Notification, supports the learning of health professionals and students, who often need to overcome difficulties in their own knowledge and contribute to promotion and prevention strategies of the health of the Brazilian population.ResumoA Notificação Compulsória oferece um meio valioso para monitorar a saúde populacional e indicar prioridades para as políticas de saúde e alocação de recursos financeiros. Deste modo, evidencia-se também a necessidade de estimulação de uma formação adequada seja ainda na graduação ou durante o exercício profissional. Este estudo objetivou apresentar o processo de construção de um Recurso Educacional Aberto sobre Notificação Compulsória, criado em decorrência da falta de conhecimento da maioria dos estudantes e profissionais da saúde e da deficiência no ensino pelas Instituições de Ensino Superior sobre o tema. Materiais e métodos: Trata-se de um estudo exploratório, descritivo, cuja abordagem foi qualitativa, realizado no mês de novembro de 2018, a partir de uma disciplina cursada em uma pós-graduação stricto sensu. Resultados: A construção do recurso ocorreu através de seis etapas distintas apresentadas ao longo deste artigo e cujo produto final foi o e-book acerca da Notificação Compulsória. Abstract Background: SMDCs are gaining momentum with potential advantages over antibody-drug conjugates (ADCs), eg, better tumor penetration and lack of immunogenicity. We have developed a novel platform technology with tailored solutions for ADCs and SMDCs with intracellular and extracellular cleavage options. Key features of the SMDC technology comprise of a small molecule ligand such as an αvβ3 integrin binder showing efficient tumor homing and a linker extracellularly cleaved by neutrophil elastase (NE) in the tumor microenvironment allowing for seamless release of different payload classes.1 Our lead SMDC VIP2362 is currently in a Phase 1 clinical trial for the treatment of advanced solid tumors (NCT05712889). SMDCs often have a shorter half-life compared with ADCs, which may result in more frequent dosing schedules. Here we show significant reduction of clearance and drastic prolongation of half-life of SMDCs achieved by adding a second binding moiety. Methods: Payloads such as kinesin spindle protein inhibitors (KSPi), cyclin-dependent kinase 9 inhibitors (CDK9i), and monomethyl auristatin E (MMAE) have been linked via elastase-cleavable linkers to one or, via a branched linker, to two αvβ3 integrin binding moieties. Cytotoxicity of SMDCs was measured in cell lines with or without NE. Monovalent and bivalent SMDCs were investigated in DMPK studies in rats while monitoring the conjugate and the released payload. Results: Payloads were coupled to the NE-cleavable peptides via different linkages. MMAE was conjugated via sterically hindered ester bonds, CDK9i was conjugated via sulfoximide bonds, and KSPi payloads were coupled via amide bonds. Each of these SMDCs was highly stable in plasma and buffer. Without NE, in vitro cytotoxicity of SMDCs in each cell line was poor with IC50 in the micromolar range. In contrast, in the presence of NE, potency increased, and cytotoxicity (IC50) was in the low nanomolar to picomolar range depending on the cell line and payload. In each case, the SMDCs reached similar potency as the free payload indicating efficient cleavage of the SMDC by NE. In DMPK studies of SMDCs in rat, none or trace levels of free payload (<0.1% as compared to the respective conjugates) was measured. With each of the SMDCs and independent of the conjugated payloads, a remarkable impact on exposure was observed when attaching a second binding ligand. The SMDCs with two ligands showed a 37- to 81-fold reduced plasma clearance as compared with the respective monovalent SMDCs, which parallels a 12- to 57-fold increase in half-life (eg, 2.1 h and 28 h for mono and bivalent MMAE SMDCs respectively). Initial in vivo studies in a cell line-derived xenograft model showed efficacy of bivalent SMDCs with once weekly dosing. BackgroundSkin disease is one of the key domains of psoriatic disease (1) and most Psoriatic arthritis patients have skin psoriasis. However skin and joint disease are often managed separately by rheumatologists, dermatologists or primary care health care providers. This can lead to disjointed care, unmet need, and treatment decisions that don’t take into account all aspects of this heterogeneous condition.ObjectivesWe sought to evaluate how psoriatic skin disease is assessed and managed within our rheumatology services by identifying disease burden, approach to management and patient satisfaction.MethodsIn an existing psoriatic arthritis outpatient rheumatology clinic, patient swere asked to anonymously complete questionnaires on disease extent, management and satisfaction. Patients also completed dermatology quality of life index (DLQI) questionnaires. Case notes and clinic letters were reviewed to collect data relating to patient diagnosis, medications and extent of clinical assessment during consultations over a 4 month period.ResultsQuestionnaires were returned by 30 patients. 72% reported problems with psoriasis, with scalp was the most commonly affected body area. 77% participants reported receiving medication to manage joints only, 4% for skin only, 19% for both joints and skin. 13 participants were receiving topical medication for PsA, with the majority being managed by their GP.From the survey, 47% of respondents were asked about their skin in the rheumatology clinic, and 30% were given advice on its management. Of the patients that completed DLQI questionnaires mean score was 3.9 (SD 3.4). Average satisfaction with psoriasis management was 5/10 (SDEV 3.6).Of the 59 consultations from which data was extracted, 54% of consultations discussed joints only, and 37% discussed both joints and skin (remainder no discussion documented). At 45% of appointments joints and skin were examined, and at 35% of appointments joints only were examined (remainder no examination documented).ConclusionPsoriatic skin disease appears to be a significant burden in this population of patients, affecting 72% of those surveyed. Over half of these patients required topical therapy to manage their psoriasis, with some requiring systemic treatments. Of these patients, most of the management of their skin condition was done by their GP, with the topic being brought up at rheumatology clinic in only half of cases. Less than half had skin examined at rheumatology clinic and less than a third were advised on management of their skin disease by their rheumatologist.Satisfaction levels with psoriasis management were fairly neutral overall. Abstract Myeloid cells, unlike other immune cells such as T cells or NK cells, are known residents in the solid tumor microenvironment (TME). In the absence of checkpoints and in proinflammatory conditions, myeloid cells (particularly M1 macrophages) are known to be capable of direct phagocytosis of tumor cells and can present tumor-associated antigens to the host immune system. However, the immunosuppressive conditions within the TME restrict and limit the anti-tumor response of tumor associated macrophages (TAMs), including their ability to recruit and activate other immune cells against the tumor. Engineered CAR-Monocytes can serve a unique function in cell therapy by bridging a key gap in the treatment of solid tumors. We are developing an autologous engineered CAR-M cell therapy product targeting Glypican-3 (GPC3) to treat hepatocellular carcinoma (HCC). The CAR serves a dual purpose as a homing ‘GPS’ signal for trafficking directly to the tumor site, and for directing phagocytosis specifically at targeted tumor cells. We have designed a proprietary CAR molecule (M83 CAR) which contains a macrophage-specific costimulatory domain that significantly increases the phagocytosis function of the CAR-M cells. Furthermore, we have demonstrated that the M83 CAR-M cells are not inhibited by the prevalent CD47 checkpoint (‘do not eat me’ signal expressed on tumor cells), which is known to restrict myeloid function in the TME. Effective phagocytosis of engineered CAR-M cells is central to subsequent mechanisms of actions that can elicit robust anti-tumor immunity against patient-specific neoantigens. However, the first barrier to overcome is effective infiltration of engineered CAR-M into the tumor from the periphery. We have demonstrated that our engineered CAR-Monocyte drug product can successfully home to the targeted tumor specifically, from the periphery. Subsequent in situ differentiation of CAR-monocytes into CAR-macrophages (M1) enables robust tumor cell phagocytosis, the central mechanism that can result in the following series of events: 1) proinflammatory shift in the TME milieu, 2) recruitment of innate and adaptive immune cells, 3) activation of T-cells against tumor neo-antigens. This is a unique outcome of the CAR-M mechanism of action that is not capitulated by CAR-T or CAR-NK cells. Leveraging and further enhancing CAR-M function could lead to the rejection of the tumor and its metastases, particularly in combination with other immune-modulating therapies.
