Reaction of 3,4,6-tri-O-acetyl-2-deoxyglucopyranosyl bromide (1) with silylated 5-benzyluracil and subsequent ammonolysis afforded α- and β-anomers of 5-benzyl-1-(2-deoxy-D-glucopyranosyl)uracil (2 and 3). Under catalysis with tin tetrachloride, silylated 5-benzyluracil reacted with 1-O-acetyl-2,3,5-tri-O-benzoyl-D-ribofuranose to give 2',3',5'-tri-O-benzoyl-5-benzyluridine (10), which was converted into the 4-thio derivative 11 by reaction with Lawesson reagent. Debenzoylation of compound 11 afforded 5-benzyl-4-thiouridine (12), whereas its reaction with methyl iodide and deblocking gave 4-me

The reaction of triethyl orthoformate with 5-amino-6-ethoxy-2-methyl-4-pyrimidinecarbonitrile (5), available from DAMN via the ammonium salt 4, afforded iminoformate 6. The latter cyclised with aqueous ammonia or equimolar amounts of aryl/benzyl amines to give the corresponding pyrimido[5,4-d]pyrimidines 8, 9a–c. Reaction with excess of aqueous ethylamine, methylamine or benzylamine caused cyclisation with substitution of an ethoxy group to give 9d–f. Hydrolysis of compounds 8, 9a–b under acidic conditions gave the corresponding pyrimido[5,4-d]pyrimidones 10, 11a–b. Heating 4 to reflux in THF

Taladegib (LY-2940680), a small molecule Hedgehog signalling pathway inhibitor, was obtained from N-benzyl-4-piperidone via Borch reductive amination, acylation with 4-fluoro-2-(trifluoromethyl)benzoyl chloride, debenzylation, substitution with 1,4-dichlorophthalazine and Suzuki cross-coupling reaction with 1-methyl-1H-pyrazole-5-boronic acid. The advantages of this synthesis route were the elimination of Boc protection and deprotection and the inexpensive starting materials. Recently, much effort has been expended on the development of non-viral gene delivery systems based on polyplexes of nucleic acids with various cationic polymers. Natural polysaccharide derivatives are promising carriers due to their low toxicity. In this work, chitosan was chemically modified by a reaction with 4-formyl-n,n,n-trimethylanilinium iodide and pyridoxal hydrochloride and subsequent reduction of the imine bond with NaBH4. This reaction yielded three novel derivatives, n-[4-(n’,n’,n’-trimethylammonium)benzyl]chitosan chloride (TMAB-CS), n-[(3-hydroxy-5-(hydroxymethyl)-2-methyl-4-pyr

The compatibility of graphene oxide with its dispersion medium (polymer) plays a critical role in the formation of nanocomposite materials with significant property improvements. Environmentally friendly miniemulsion polymerization, which allows a formation of nanoencapsulation in an aqueous phase and high molecular weight polymer/composite production is one promising method. In this study, we screened a series of amphiphilic modifiers and found that the quaternary ammonium (ar-vinyl benzyl) trimethyl ammonium chloride (VBTAC) pending carbon double bonds could effectively modify the graphene o

Polymer nano-composites (PNC) are polymers which are reinforced with less than 5% by volume of nano-sized particles with high aspect ratios (L/h > 300). Compared to conventional composites, where the reinforcement is on the order of microns, the nano-composites are reinforced on the order of a few nanometers with advantages in processing and toughness. Nano-composites of epoxy clay have been studied where epoxy is mixed at high shear rates with clay. In our method of making nano-composites, an epoxy, Diglycidyl ether of bisphenol (DGEBA) A was mixed under high shear with organically modifie
