A disfunção erétil pode ser definida como a dificuldade parcial ou total de se obter e/ou manter a ereção peniana para um desempenho sexual satisfatório. Para que a ereção peniana ocorra, há necessidade de aumento do fluxo sanguíneo para o pênis por relaxamento de seu tecido erétil, o corpo cavernoso. Além disso, é preciso que o retorno venoso do pênis diminua para que a ereção se mantenha, processo que conta com a participação da veia dorsal peniana. O papel da serotonina (5-hidroxitriptamina, 5-HT) para a ereção peniana não está bem estabelecido, apesar de crescer o uso de fármacos que alteram sua homeostasia, como a fluoxetina. Além disso, estudos em nosso laboratório demonstraram a capacidade do corpo cavernoso (CC) em captar a 5-HT e metabolizá-la em 5-HIAA, seu principal metabólito. Outros trabalhos mostraram que, após ser captada pela célula vascular, a 5-HT é capaz de modular a contração do músculo liso por um mecanismo denominado serotonilação. Deste modo e sabendo preliminarmente da capacidade do CC de rato contrair na presença da 5-HT, o objetivo do presente trabalho foi o de caracterizar farmacologicamente essa resposta. Na análise de contrações neurogênicas do CC por estímulo elétrico de campo, observamos que o principal neurotransmissor liberado no CC de rato capaz de contraí-lo é a noradrenalina, a qual contrai o CC por ativação dos receptores a1-adrenérgicos. Esses dados sugerem pouca ou nenhuma liberação neuronal de 5-HT. Apesar disso, observamos que a 5-HT exógena é capaz de gerar uma resposta contrátil não reprodutível em CC de rato, sugerindo a existência de taquifilaxia. No entanto, não foi observada resposta taquifilática na veia dorsal peniana exposta à 5-HT 10 µM. Utilizando a metil-ß-ciclodextrina, um tensoativo capaz de sequestrar colesterol causando um desarranjo de estruturas de membrana como as cavéolas, verificamos que a resposta taquifilática não ocorreu através da internalização de receptores pelas caveolas. Além disso, verificamos que a inibição do metabolismo da 5-HT com o uso de pargilina, bem como a inibição da captação de 5-HT com o uso de fluoxetina, não alteraram o perfil da resposta contrátil induzida por 5-HT. Além disso, o metabólito da 5-HT, o 5-HIAA foi incapaz de gerar uma resposta contrátil. Esses dados sugerem ser improvável que a 5-HT intracelular seja importante no processo de contração do CC de rato. Com o uso de agonistas dostipos de receptores 5-HT1 e 5-HT2 e de antagonistas dos tipos de dos receptores 5-HT1, 5-HT2, 5-HT3 e 5-HT7, demonstramos que receptores 5-HT2 e, mais especificamente os do subtipo 5-HT2A, sejam os principais responsáveis por gerar a contração observada no CC de rato. Podemos concluir que a ativação de receptores 5-HT2A pela 5-HT em CC de ratos induz uma resposta contrátil que sofre dessensibilização com exposições repetidas à amina, fenômeno não observado na veia dorsal peniana. Desta forma, por não manter seu efeito contrátil no CC, e por contrair a veia dorsal peniana, a 5-HT pode contribuir para a função erétil em ratos.
Abstract : Erectile dysfunction is defined as the inability to obtain and/or maintain an erection for satisfactory sexual performance. Penile erection occurs by the increase in blood flow into the penis in parallel with the relaxation of the corpus cavernosum (CC), its erectile tissue. Furthermore, a decrease in venous return from the penis is also necessary to maintain erection, a process that involves the participation of the dorsal penile vein. The role of serotonin (5-hydroxytryptamine, 5-HT) in penile erection is not well established despite the increasing use of drugs that alter its homeostasis, such as fluoxetine. Moreover, data from our laboratory show the ability of the CC to take up 5-HT and metabolize it into its major metabolite, 5-HIAA. Other studies have shown that after 5-HT uptake by the vascular smooth muscle cell, it is able to modulate smooth muscle contraction intracellularly by a mechanism called serotonylation. In light of these aspects, and knowing preliminarily the contractile ability of 5-HT, the purpose of this study was to characterize pharmacologically the contractile response of the rat CC to 5-HT. By evaluating neurogenic contractions induced by electrical field stimulation, we observed that norepinephrine is the main contractile neurotransmitter released in the rat CC, acting via stimulation of a1-adrenergic receptors. These data suggest nerves in the CC release little or no 5-HT. Nevertheless, we observed that exogenous 5-HT is able to induce a non-reproducible contractile response in the rat CC, suggesting tachyphylaxis. However, this tachyphylactic response to 5-HT was not observed in the dorsal penile vein. Using methyl-ß-cyclodextrin, a drug that depletes cholesterol from the cell membrane and causes a disruption of caveolae structures, we found that the tachyphylactic response does not occur through 5-HT receptor internalization via caveolae. Furthermore, we found that the inhibition of 5-HT metabolism by pargyline, as well the inhibition of 5-HT uptake by fluoxetine did not alter the profile of the contractile response induced by 5-HT. Moreover, 5-HIAA was unable to produce a contractile response. These data suggest that intracellular 5-HT is unlikely important to contraction of the rat CC. Using agonists of the 5-HT1 and 5-HT2 families of 5-HT receptors and antagonists of the 5-HT1, 5-HT2, 5-HT3 and 5-HT7 families of 5-HT receptors, we observed that the 5-HT2 receptors, more specifically the 5-HT2A, are the main responsiblefor the contraction induced by 5-HT in the rat CC. We conclude that the activation of the 5-HT2A receptor by 5-HT induces a tachyphylactic contractile response in the CC, but not in the dorsal penile vein. This inability of 5-HT to maintain a sustained contraction of the CC, allied to its ability to contract the penile dorsal vein, suggest it may contribute to erectile function in rats.

Full textLocated between the city of Marseille and the Camargue region, the territory known as the "Etang-de-Berre" is centered on a 15. 500 hectares lagoon. This territory was created soon after the end of world war II in relation with the settlement of the industrial port of Fos-sur-Mer. Despite the positive and original qualities of this region, the global amount of industrial pollution greatly affected a potential outcome engaged ten years ago. The present analysis is conducted through the historical study and classification of social representations from the process of disqualification to today's effort of rehabilitation. It is based on the three anthropological scales of space : the planning, the landscape and the individual perception. Finally, the descrption of this modern space contributes to elaborate new ways of conceiving recent territories with the definition of an "inhabitant body" and its values, which could become essential figures for imagining a "modfern landscape"

Full textFor forty years, important transformations have been disrepting agriculture in this area. Traditional crops and methods are being abandoned in provence, extensive cereal farming, olive and wine-growing are declining while sheep and pig breeding are disappearing. Farmers from the marseilles area have revitalized agriculture in places. Since 1955, the abundant water resources combined with an exceptionally favourable microclimate have, with the development of irrigation and sheltering techniques in market-gardening for example, resulted in higher crop-yields. However, the production of olive oil in the lower valley of the arc, characteristic of the bouches-du-rhone, remains a traditional activity. Nevertheless, urban and industrial expansion together with new means of communication are gradually encroaching upon the rural world, causing the decline of an agriculture which would already have required a protectionist policy at a time when it was still dynamic and capable of modernizing itself. As early as 1970, official efforts were initiated for safeguareling the semi-rural milieu of the region, including one section withen the new town of vitrolles. The future of agriculture remains precarious : urban and industrial growth is still a threat. The only alternative is investing in advanced farming adapted to the new conditions of the e. C. C

Full textThe Berre Lagoon is an ecosystem historically disturbed by strong industrial discharges and significant freshwaters inputs from both natural and anthropogenic (EDF hydroelectric power station) origins. While rehabilitation policies initiated in 1994 are already showing some success, the introduction and proliferation of Mnemiopsis leidyi since 2005 could limit their effectiveness. The originality of this study is to associate oceanography and sociology in order to estimate the impact of M. leidyi on the functioning of this socio-ecosystem. We were able to show that the population of M. leidyi is maintained within a large range of temperatures (3 °C-28 °C) and salinities (10-30), with a quantity of carbon available ~ 3 mg C L-1 or more, temperature being a determining factor in population dynamics. The absence of this ctenophore on during cold events and its recurrence several months later suggests either the existence of an external source or the presence of a refuge zone that has been determined (Vaine lagoon) favourable to the maintenance of the ctenophores. The strong proliferation of M. leidyi in the Berre lagoon mainly affects professional fishermen. The clogging of nets, the mutilation of catches, the accelerated degradation of the material and the increase in the strenuousness induce an annual economic loss estimated at 50 %. Here in our interdisciplinary framework, the understanding of the biological and the anthropogenic interactions has shown that Mnemiopsis is tempering with the implementation of the rehabilitation efforts

Full textFor twenty years, cultural and creative activities (art,architecture, etc.) have been promoted by public authorities and scientists for their cultural, economic,social and urban roles. Their spatial agglomeration informer industrial districts implies multiple issues on these territories. Planned by political decision-makers or resulting from spontaneous dynamics, this agglomeration transforms industrial wasteland into creative quarters.Clusters that link creative workers in collaborative social networks are embedded in these quarters. These social networks are characterized by cooperation and a senseof togetherness.This thesis aims to question the functioning and the stakes of creative quarters by confirming the hypothesis of the establishment of a creative workers’ club within these territories. The club is defined as a voluntary grouping of individuals around the exclusive enjoymen tof goods and services. It carries both collective dynamics of sharing and logics of exclusion and segregation.Questioning its existence in creative quarters induces a reflection on social cohesion and fragmentation on the one hand, and creativity and innovation on the other one. Based on a qualitative analysis of three creative quarters in the cities of Nantes, Marseille and Grenoble, this thesis illustrates how clubs of creative workers are formed in these territories, how they work and what their social and economic impacts are

Full textThe industrialization of the Berre lagoon in the 20th century was accompanied by large releases trace metals, which were partially accumulated in sediments and are now likely to be remobilized to the water column or be integrated into the food chain and cause an ecotoxicological risk.The reconstruction of the temporal and spatial trends of sediment contamination shows that current levels of contamination of surface sediments have been the lowest for decades in agreement with the effectiveness of regulations on industrial releases set up in the years 1970. These levels are low to moderate in surface but very high contamination exist a few centimeters below the sediment surface.The role of Fe and Mn oxy-hydroxides and sulfides in controlling the mobility of ETM in the sediment and fluxes at the water/sediment interface has been demonstrated through the modeling of transport and reactions of chemical compounds and trace metals in the pore waters, their concentration profiles in the reactive fraction of the particulate phase and experiments under controlled laboratory conditions. The oxygenation of the water column is the main parameter influencing the mobility and fluxes and the influence of reoxygenation of bottom water column in the Grand Etang is discussed.Finally, the bioavailability of trace metals and adverse effects they may constitute for a target benthic organism, the polychaete Nereis succinea, were evaluated by estimating the potentially bioavailable fraction in sediments (chemical extractions and Diffusive Gradient in Thin-films), by measuring bioaccumulated concentrations and by the use of biomarkers (metallothioneins and genotoxicity assays). Some highly bioaccumulated trace metals pose a potential risk and might be involved in the degradation of the benthic macrofauna

Full textBerre lagoon was occupied by extensive meadows at the turn of the 20th century which regressed down under the impact of urban and industrial pollution and inflow of the EDF canal. Even though freshwater inputs and pollutions were drastically reduced respectively in the 1980s and 1990s, meadows have not significantly gained ground. This thesis aims at analysing the interactions between seagrass meadows of Berre lagoon, hydrodynamics and sedimentary processes, based on the postulate that these mechanisms are important in the maintenance of the meadows in their present dispersed form. In the lagoon, winds constitute the dominant influence on hydrodynamics in the lagoon by generating wind waves and currents. Wave attenuation is linked to wave height, which is, in turn, dependent on wind intensity and fetch length and modified by the bay morphology. Wave attenuation is also modulated by meadow biometry, and by water levels and currents.Whereas currents are strong and strongly influenced by wind and wind waves above the meadow, a transition canopy-water layer dissipates waves and currents. In the canopy, currents are thus attenuated.The meadow is not just a passive element in the overall sediment dynamics since it reduces energy and thus modifies substrate changes within and in the back of the meadow, thus protecting the shoreline. However, it is the recurrence of strong wind that seems to drive sedimentary changes. The strong interactions between the meadow and the hydrodynamic and sedimentary processes could limit the extension of the meadow in areas more exposed to waves

Full textThe Berre lagoon receives excess freshwater leading to strong changes in the ecosystem salinity and in the nutrient inputs. Regeneration processes and the N2 removal of nitrogen are of a particular interest as they can explain the planktonic development and control the eutrophication state. These processes were studied in 2 stations with different patterns of long term stresses. DNRA and anammox were negligible. Denitrification (particularly the one coupled to nitrification) would be the main N2 removal in the lagoon. The high primary production based on N-NH4+ can be explained by the regeneration processes, while the primary production based on N-NO3- might be sustained by external inputs. As salinity susceptible to influence directly or indirectly the nitrogen processes, a microcosm approach was used in order to assess their sensitivity (resistance, resilience, shifts) to short term salinity fluctuations. Overall, towards the same kind of perturbations, the two stations responded differently (denitrification and nitrate reduction exhibited showed higher resilience than nitrification.), suggesting that long-term saline stresses would influence short-term responses. We suggest that the diversity and the structure of the communities would be crucial in their functional redundancy and thereby in the maintenance of the rates. In this study, we focused on the total structure of the nitrifying community (AO). AO and their transcripts were site-specific and their abundances changed slightly between microcosms (higher resistance in the site subject to frequent salinity fluctuations compared to the other site which is not affected by changes in the in situ salinity)
