Abstract:
Theodore Beza and the Quest for Peace in France examines the changing political strategies and religious attitudes of French Protestant leaders between the Saint Bartholomew's day massacres (1572) and the Edict of Nantes (1598). The hand-picked successor of John Calvin in 1564, Theodore Beza was an influential teacher, preacher, and power-broker in Geneva, as well as a prominent exiled leader of the French Reformed churches during the next four decades. Drawing on Beza's correspondence network, city archival materials and rare Huguenot pamphlets, I reconstruct the survival tactics of French Protestants in response to Catholic advances, document the decline in Huguenot expectations after 1572, and examine how social and political factors created widening ideological fissures within the Reformed movement by century's end. In highlighting the patterns of thought of the Huguenot leadership, my research contributes to an understanding of Protestant mentalities during the turbulent era of the French civil wars. In the aftermath of the massacres of 1572, Beza and other exiled leaders in Geneva were not only theorists of political resistance, but major players in Protestant agitation against the Valois monarchy. As the Reformed churches withered under royal persecution and Catholic missionary activities during the next decade, the reformer and his colleagues gradually aligned their political fortunes with Henri of Navarre. Beza tempered, but did not abandon his resistance theories when Navarre became presumptive heir to the French throne (1584). In return for a secret--hitherto unknown--annual stipend, Beza became Navarre's 'public relations agent' in Germany and Switzerland, raising money and mercenaries for Huguenot armies in the years prior to Henri's accession (1589). The bonds of friendship, patriotism and patronage made Beza a dedicated supporter of the person and program of Henri IV, even after the king converted to Catholicism in 1594. Thereafter, he urged the Reformed to trust the king's peace overtures, while attempting to silence 'moderates' who advocated doctrinal compromise in return for a political settlement. Though welcoming the Edict of Nantes, Beza and other Protestant leaders recognized that prospects for reform in France had been decisively curtained: 'the golden age has degenerated into a century of iron.'

Abstract:
Diabetes is a metabolic disease characterized by the loss of functional pancreatic beta cells. The incidence of diabetes has risen rapidly in recent decades, which has been attributed at least partially to alterations in host-associated microbial communities, or microbiota. It is hypothesized that the loss of important microbial functions from the microbiota of affected host populations plays a role in the mechanism of disease onset. Because the immune system also plays a causative role in diabetes progression, and it is well documented that immune cell development and function are regulated by the microbiota, most diabetes microbiota research has focused on the immune system. However, microbial regulation is also required for the development of many other important tissues, including stimulating differentiation and proliferation. We therefore explored the possibility that the microbiota plays a role in host beta cell development. Using the larval zebrafish as a model, we discovered that sterile or germ free (GF) larvae have a depleted beta cell mass compared to their siblings raised in the presence of bacteria and other microbes. This dissertation describes the discovery and characterization of a rare and novel bacterial gene, whose protein product is sufficient to rescue this beta cell developmental defect in the GF larvae. Importantly, these findings suggest a possible role for the microbiota in preventing or prolonging the eventual onset of diabetes through induction of robust beta cell development. Furthermore, the loss of rare bacterial products such as the one described herein could help to explain why low diversity microbial communities are correlated with diabetes.

Abstract:
Inspired by biological membrane shaping in the cell through means of curvature-inducing proteins, we investigate the interplay between membrane curvature and the distribution and movement of shape-inducing objects which are free to move as a consequence of the underlying shape. We initially study the self-assembly of a filament, taken as a proxy for the cross-section of a biomembrane, which is primarily driven by the chemical kinetics of attaching proteins and find that, under certain mechanical stiffness regimes of the attaching proteins, pattern formation occurs. Regions of high and low protein concentration form before spatially uniform filament shapes are obtained by means of protein adhesion and movement governed by diffusion and local curvature-seeking. However, noting that the curvature-mediated protein movement on membranes has been biologically observed to be long-range, we next study the self-assembly of embedded inclusions on a membrane as a result of the underlying geometry. We first derive an interaction law for the shape-mediated interaction of inclusions which break symmetry and find that there is a finite equilibrium distance to which the inclusions will aggregate. We derive corresponding equations of motion which describe this curvature-mediated aggregation mechanism and, using this framework, we investigate some of the properties of these self-assembled configurations, including their energy, stability, and their collective elastic behavior. Lastly, we consider the interaction energies of embedded inclusions on a periodic domain and determine that this mechanism may explain computational results of how proteins form rings to promote tubulation on cylindrical membranes.

Abstract:
Two dimensional (2D) crystals including graphene and monolayer transition metal dichalcogenides (TMD) such as molybdenum disulphide (MoS2) and tungsten disulphide (WS2) have attracted widespread attention for both basic scientific interest and potential in device applications. In this thesis, we investigate the electronic and optoelectronic devices based on 2D crystals grown using chemical vapour deposition (CVD). In our study on graphene transistors, we show that surface charge transfer between graphene and vertically stacked WS2 causes local modulation to doping levels of graphene, and can be further stimulated by photo-irradiation. We demonstrate the formation of an isotype junction in graphene, where two lateral sections of a graphene ribbon contain different dopant concentrations. Ultrathin lateral photodetectors of 2 nm thickness are made by incorporating graphene as electrodes and WS2 as active semiconductor. We show that graphene is different to conventional metal electrodes due to the finite density of states from the Dirac cones of the valence and conduction bands, which enables modulation of photoresponsivity through electrostatic gating and light input control. We demonstrate photodetectors with laterally spaced graphene electrodes and vertically stacked WS2/MoS2 as active light-adsorbing component, achieving photoresponsivity as high as 2340 A W-1. We observe in hybrid WS2/MoS2 photodetector massive enhancements in photogain compared to homobilayer and monolayer devices of MoS2 or WS2 due to efficient charge transfer between WS2 and MoS2 and long carrier lifetimes. Our results show that high performance photodetectors using all 2D materials can be realized using graphene electrodes and TMD heterostructures with type II band alignment.

Abstract:
Falciparum malaria remains one of the leading infectious causes of morbidity and mortality worldwide. Though the search for an effective vaccine has seen unprecedented advances in recent years and one of the leading vaccine candidates, RTS,S/AS01 is going to enter pilot deployments in Africa in 2018, there has been no vaccine that has demonstrated durable high level efficacy. This thesis describes four Phase I and Phase II clinical trials assessing novel pre-erythrocytic malaria vaccines in healthy adult UK volunteers. R21 has been developed at the Jenner Institute, University of Oxford. This is an improved RTS,S construct, an antigen derived from the pre-erythrocytic circumsporozoite protein, which is an abundant coat protein involved in sporozoite development and hepatocyte invasion. R21 comprises recombinant particles expressing the central repeat and the C-terminus of the circumsporozoite protein (CSP) fused to the Hepatitis B surface antigen (HBsAg), but without the excess of unfused HBsAg protein found in RTS,S. Both RTS,S and R21 predominantly function by inducing antibodies against the circumsporozoite protein. In addition to humoral immunity, it is widely recognised that T-cell mechanisms are likely to play an important role in malaria immunity and heterologous prime-boost regimes using viral-vectored vaccines have been shown to induce potent T-cell responses. ChAd63-MVA ME-TRAP is a regime that has been developed in the Jenner Institute and the first of the clinical trials described in Chapter 3 assesses the safety and immunogenicity of adjuvanting this regime with a novel saponin-based adjuvant, Matrix-MTM. This trial showed that this regime was safe, well tolerated and did not result in any immunological interference. R21 had never been administered to humans prior to the work described in this thesis. In Chapter 4, I describe two Phase I clinical trials assessing the safety and immunogenicity of R21 given at varying doses administered with two different adjuvants, Matrix-M (MM) and AS01B. Both these trials showed that R21 was safe and well tolerated when administered with either adjuvant and elicited comparable humoral immune responses to RTS,S/AS01B even at one-fifth of the dose (10μg). This formed the basis to test the efficacy of R21/MM, which had a favourable reactogenicity profile, using controlled human malaria infection in malaria-naïve UK volunteers. High level efficacy (> 80%) is reported in Chapter 5 with a three-dose schedule of 10/10/10μg R21/MM given 4 weeks apart. This provides the first evidence of proof-of-concept that this vaccine regime should be tested in malaria-endemic populations.

Abstract:
Gerald Heard (1889-1971) was an influential figure among the intelligentsia of the 1930s, once described by E.M. Forster as “one of the most penetrating minds in England.” However, he remains an ill-defined footnote, a marginal figure whose influence and reputation, although acknowledged, remains unexamined. This dissertation examines his life and work, and considers the role which Heard, as a generaliser and public intellectual, played in the intellectual landscape of the 1930s. Central to Heard’s philosophy was a belief that society was in need of a spiritual and psychological force which could allow isolated individuals to participate in community with one another. Heard’s solution to bring about this evolution of consciousness would prove to be partly psychological, partly mystical and partly down to the product of a particular way of living. The first chapter outlines Heard’s philosophy in detail. Subsequent chapters are structured so as to provide a loose biographical chronology, each focussing on a different phase of Heard’s career and examining the development of his thought. Running throughout the dissertation is a consideration of Heard’s role as a public intellectual. It was as a popular ‘generaliser’ of thought that Heard found his public, and the limited degree of success he found as a man of action could be seen to be a natural limitation of the role he had constructed for himself. Chapter II focuses on Heard’s time as personal secretary to Sir Horace Plunkett, father of the Irish Co-Operative Movement, and how the ideals of this movement can be seen to inform his developing ideas of human community. Chapter III looks at Heard’s role as a broadcaster with the B.B.C., where he became a noted populariser of science, firmly establishing himself as a public figure and cultural authority. It is arguably this increased public profile which provided Heard with a ‘public’ to whom he could address his ideas. Chapter IV, drawing on archival material from Dartington Hall, considers Heard’s role as a lecturer at Dartington School, and more importantly his first experiment to establish a small ‘group’ for meditation in an attempt to discover the mystical and psychological basis for a co-operative society. Chapter V examines his career as an outspoken pacifist, where he would advance his arguments for a radical reorganisation of society as a practical solution to the question of peace and further attempt to become a man of action.

Abstract:
This thesis is an early modern urban study that focuses both on national trends and events but also acknowledges the distinctive nature of the individual town. It takes a holistic approach to economic, administrative and socio-cultural changes and developments in a small but significant Berkshire town. The years after the dissolution of Abingdon abbey in 1538 were a critical time of losses, problems and opportunities for Abingdon. In the light of the town's successful development later in the century how serious were the challenges that it faced and what factors contributed to its survival? After a historiographical introduction and discussion of sources, two chapters investigate the town's medieval development under monastic lordship. The central chapters explore different aspects of change in Abingdon during the reformation period. In practical administrative terms the town's response was opportunistic and positive, due to a happy convergence of government policy and the continuity of local elite leadership. Economic and social strength and diversity gave stability, and a detailed rental survey demonstrates how the acquisition of public and private property benefited the town's elite. However, cultural and religious changes had some ill effects. Chapters 8 and 9 discuss Abingdon's successful administrative, economic and social development under the new institutions of the borough and Christ's Hospital established in the 1550s. Experienced leading men were supported by a thriving middling group of tradesmen. A brief discussion about the relationship between civic culture, civil discipline and puritanism later in the 1500s follows. The thesis concludes that Abingdon's resilience and survival was based on its diversified economic development and on social and cultural continuities. Abingdon's richly documented experience of reformation change demonstrates the need for continuing research into individual towns and offers an important contribution to our understanding of the age of reformation in English urban history.

Abstract:
Late-life depression (LLD) is common illness, frequently associated with neuropsychological impairment. Disruption of frontal-subcortical and limbic networks may play a key role in LLD and can be examined using magnetic resonance imaging (MRI). Grey matter (GM) can be examined using T1-weighted MRI, white matter (WM) using diffusion tensor imaging (DTI), and functional connectivity using resting-state functional MRI (fMRI). To clarify the roles of GM, WM and functional connectivity in LLD, systematic reviews and meta-analyses of T1-weighted MRI, DTI and resting-state fMRI studies of depression were performed. The literature provided evidence for GM and WM abnormalities within frontal-subcortical and limbic networks, and increased functional connectivity within the default-mode network, in depression. To examine whether results gained from different techniques are complementary, multi-modal MRI was used to compare GM, WM and functional connectivity between thirty-six participants with LLD and twenty-five control participants. WM integrity was widely reduced in LLD, without significant group differences in GM or functional connectivity. To investigate whether neuropsychological deficits represent independent processes with specific neural correlates, or whether they can be explained by a core deficit, the relationships between neuropsychological and MRI measures were explored. Executive function and processing speed were found to represent core deficits that contribute to impairment in other domains; and impaired performance was correlated with reduced frontal WM integrity. Episodic memory deficits were dependent on executive function and processing speed; and associated with reduced frontal and hippocampal WM integrity. The relationships between age at onset, severity and MRI measures of GM, WM and functional connectivity were also investigated. Later onset was associated with reduced WM integrity, in line with the vascular hypothesis. Earlier onset was associated with greater duration of illness and reduced hippocampal volume, consistent with the glucocorticoid cascade hypothesis. Severity was not associated with any MRI measure. This thesis strongly supports the hypothesis that WM abnormalities in frontal-subcortical and limbic networks play a key role in LLD, with abnormalities related to neuropsychological impairment and compatible with the vascular hypothesis.

Abstract:
Riboswitches are cis-acting gene regulatory RNAs, which function without involvement of proteins. They have been implicated as drug targets and are attractive systems for the study of RNA-ligand binding and RNA folding. The purine riboswitch was used as a model system for RNA-ligand docking. Published binding data was successfully reproduced in silico and compounds predicted to bind the riboswitch in a virtual screening were tested experimentally. Structural data confirming the predicted binding mode for several cases was obtained. The problems encountered were not specific to RNA-ligand docking but known from the far more explored field of protein-ligand docking.The SAM-I riboswitch was also subjected to virtual ligand screening. This receptor is a system of greater complexity than the purine riboswitch and consequently posed a harder challenge to the docking protocol. After initial validation of the docking setup based on previously published data, a set of compounds selected from the in-house database of commercially available compounds was screened. One compound identfied in silico was cofirmed to bind experimentally.The k-turn motif found in the SAM-I riboswitch was investigated with respect to its folding. The k-turn motif was found to be foldable in context of the SAMI riboswitch as well as in isolation as was expected. Furthermore, mutations disrupting key interactions within the k-turn motif were found to be prohibitive of k-turn folding in isolation as well as in context of the riboswitch, leading to a loss of ligand binding. Interestingly, two sequences were identfied which fold in context of the riboswitch but do not fold in isolation. This confirms the contribution of tertiary interactions to k-turn folding. This conclusion was backed up with structural data is a system of greater complexity than the purine riboswitch and consequently posed a harder challenge to the docking protocol. Abstract:
Bromodomains, protein modules found in 46 human chromatin-associated proteins, bind to acetylated lysine residues (KAc), and modulate the formation of complex protein scaffolds, which are often involved in transcription. The work in this dissertation focuses on the development of small molecule inhibitors for the BRD9 bromodomain, which is a component of nucleosome remodelling complexes, and has been linked to lung and cervical cancers. The lead molecule was based on the triazole KAc mimic, which is a promiscuous bromodomain ligand. Using a rational approach involving evaluation at the four positions shown on scaffold 1, this work led to compound 2, which has nanomolar affinity for the BRD9 bromodomain, and some selectivity over BRD4(1). Compound 2 was shown to have a high lipophilic ligand efficiency, high aqueous solubility, and moderate metabolic stability in an in vitro mouse model, making it suitable for further studies in a biological setting. Microscale Thermophoresis has been used as a new technique to measure interactions between small molecule ligands and their bromodomain binding partners. This work has provided useful structure-activity relationships for a subset of bromodomains, which will be useful in future ligand development programmes of this emerging therapeutic target class.

With the world's fourth-largest Muslim population, Bangladesh is an important case study in the exploration of what it means to be a 'secular' country with Islam as a state religion. One important mechanism through which to analyse the relationship between religion and the state is through the country's laws, and family laws are especially significant in that they represent the state's determination of which long-standing social and religious practices find their way into legislation as a representation of societal values. As with many other countries with significant Muslim populations, personal status legislation has remained relatively static in the years following independence, despite attempts at change. Inspired by studies of negotiations between state and civil society actors in bringing about changes in law, this study analyses the evolution of family laws for Muslims in Bangladesh, revealing a range of voices using such laws in their negotiations between competing notions of 'Islam' and 'secularism' and their role in governance. Using parliamentary and Supreme Court records, newspaper archives, expert interviews, and secondary literature, I show that there has been little change in personal status legislation beyond procedural simplification, and that the judiciary and policymakers have had a tendency to support freedom of religious practice except in family laws. This study explores why this is the case, and focuses on the discourse around the National Women Development Policy and its clause on property and inheritance as the greatest point of contention in enhancing women's rights in family laws.

Abstract:
This thesis focuses on the development of high harmonic generation (HHG) by using polarisation controlled quasi-phase matching QPM as well as related topics. A new class of QPM techniques called polarisation-controlled QPM is introduced where linear or circlar birefringence enables the modulation of the driving field's polarisation state called polarisation-beating QPM (PBQPM) for linear birefringence and optical rotation QPM (ORQPM) for circular birefringence respectively. PBQPM uses a linear birefringence to modulate periodically the driving pulse between linear and circular/elliptical polarisation states. Because elliptical or circular polarisation of the driving pulse suppresses harmonic generation, by appropriately matching the beat length of the driving field's polarisation state to the coherence length of the harmonic generation, QPM can be achieved. In the second technique, ORQPM, propagation of the driving radiation in a system exhibiting circular birefringence causes its plane of polarisation to rotate; by appropriately matching the period of rotation to the coherence length, it is possible to avoid destructive interference of the generated radiation. Not only does ORQPM have similar enhancements as true-phase matching, it is also the first proposed QPM source for circularly polarised high harmonics. The importance of phase modulation in QPM, especially relating to modebeating in hollow-core waveguides where harmonics is being generated are also explored theoretically. Based on this, a novel technique for analyzing random phase matching using a continuous phase-diffusion treatment has been developed; theoretical analytical models are shown to produce excellent agreement with simulations. It is further shown that random phase matching may be responsible for additional broadening of the high harmonic spectrum, especially at higher harmonic orders. Because mode and polarisation control is central to polarisation-controlled QPM, four waveguide mode decomposition techniques from single shot CCD data have been developed. The extraction of phase and coupling coefficients are demonstrated experimentally. A novel analytical general solution for the phase introduced by a phase-only spatial light modulator to generate a given far-field phase and amplitude was developed. The solution was demonstrated experimentally and shown to enable excellent control of the far-field amplitude and phase. Finally, circular and linear birefringent waveguides were explored. Analytic solutions to rectangular birefringent hollow-core waveguides were developed and some initial demonstration experiments were performed.

Abstract:
Cardiovascular disease is the leading cause of mortality in the developed world with up to fifty percent of cases being due to sudden cardiac death. Changes in sympatho-vagal balance underpin many cardiovascular conditions including heart failure and myocardial infarction. Neuraxial modulation of the autonomic nervous system is an emerging therapy to prevent ventricular arrhythmias, the main cause of sudden cardiac death. Chapter One reviews our current understanding of how the cardiac autonomic nervous system influences ventricular arrhythmogenesis. A particular focus was on the controversial role of cholinergic receptors and nitric oxide (NO) in parasympathetic protection from ventricular arrhythmias. Tetrahydrobiopterin (BH4), a critical cofactor for both tyrosine hydroxylase and NO synthases, and the co-transmitter neuropeptide-Y (NPY) may also influence sympathetic triggering of ventricular arrhythmias. This leads to the specific aims of the thesis which were to determine the mechanisms of the cholinergic antifibrillatory effect, investigate the role of cotransmission in arrhythmogenesis and, the mechanistic role of BH4 in autonomic cardiovascular control. Chapter Two detailed the experimental approach taken to investigate the hypotheses. A novel Langendorff heart preparation was developed with intact autonomic nerves to investigate how the stable analogue of acetylcholine, carbamylcholine (CCh) raises ventricular fibrillation threshold (VFT) and whether exogenous or endogenously released NPY lowers VFT. These actions are further investigated using optical mapping, dye free imaging of ventricular cell monolayers, immunohistochemistry, ELISA assays and measurements of NO metabolite production. To investigate the role of BH4 in the sympathetic control of the heart, an IRES-cre recombinase strategy was used to produce genomic deletion of GCH1 (the gene encoding BH4) in sympathetic neurons. Biopterins and plasma catecholamines were measured using HPLC, and blood pressure and heart rate via tail cuff plethysmography. Chapter 3 showed that CCh increased VFT, prolonged action potential duration and flattened the electrical restitution curve. This effect required stimulation of both muscarinic and nicotinic receptors and the generation of nNOS derived NO utilising a cGMP dependent pathway. These observations are in keeping with established evidence demonstrating the obligatory role of the muscarinic receptor and indicate that the role of NO is likely to be via modulation of cholinergic neurotransmission. Chapter 4 studied the role of the sympathetic co-transmitter NPY. NPY has been shown to increase ventricular myocyte calcium dynamics. Plasma levels are also increased post myocardial infarction and during heart failure, and correlate with outcomes. Perfusion of NPY decreased VFT via a Y1 receptor dependent mechanism and increased arrhythmic activity in myocyte monolayers. Direct sympathetic stimulation resulted in NPY release and remained pro-arrhythmic despite β-blockade, an effect that could be abolished by combined β-Y1 receptor blockade. These observations indicated that NPY may be a novel, pro-arrhythmic trigger amenable to therapeutic pharmacological modulation. Chapter 5 details the generation and phenotyping of two tissue specific Gch1 knockout mouse models. Whilst one model failed to produce significant lowering of BH4 in sympatho-adrenal tissue, the other did result in a marked neuro-motor phenotype. A biochemical rescue or alternative genomic modification approach would be required to study the cardiovascular phenotype of sympathetic Gch1 deletion in more detail. Chapter 6 is a concluding discussion summarising the main findings of the thesis, placing them in a clinical context and discussing avenues for further research.

Muitos estudos em diferentes áreas examinam como um conjunto de variáveis influencia algum tipo de percentagem, proporção ou frações. Modelos de regressão lineares não são satisfatórios para modelar tais dados. Uma classe de modelos de regressão beta que em muitos aspectos é semelhante aos modelos lineares generalizados foi proposto por Ferrari e Cribari--Neto~(2004). A resposta média é relacionada com um predictor linear por uma função de ligação e o predictor linear envolve covariáveis e parâmetros de regressão desconhecidos. O modelo também é indexado por um parâmetro de precisão. Smithson e Verkuilen,(2005), entre outros, consideram o modelo de regressão beta em que esse parâmetro varia ao longo das observações. Nesta tese foram desenvolvidas técnicas de diagnóstico para os modelos regressão beta com dispersão constante e com dispersão variável, sendo que o método e influência local (Cook,~1986) mostrou-se decisivo, inclusive no sentido de identificar dispersão variável nos dados. Adicionalmente, avaliamos através de estudos de simulação o desempenho de estimadores de máxima verossimilhança para o modelo de regressão beta com dispersão variável, as conseqüências de estimar o modelo supondo dispersão constante quando de fato ela é variável e de testes assintóticos para testar a hipótese de dispersão constante. Finalmente, utilizando um esquema de bootstrap (Davison e Hinkley,1997), desenvolvemos um procedimento de obtenção de limites de predição para o modelo de regressão com dispersão constante. Ilustramos a teoria desenvolvida com várias aplicações a dados reais.
Practitioners oftentimes wish to investigate how certain variables influence continuous variable that assumes values on the standard unit interval $(0,1)$, such as percentages, proportions, rates and fractions. Linear regression models are not suitable for modelling such data. A class of beta regression models which is in many aspects similar to that of generalised linear models was proposed by Ferrari and Cribari--Neto~(2004). The mean response is related to a linear predictor, which involves covariates and unknown regression parameters, through a link function. The model is also indexed by a precision parameter. Smithson e Verkuilen~(2005), among others, consider the beta regression model with variable dispersion, i.e., beta regression in which the precision parameter is not constant across observations. In this dissertation we develop diagnostic methods for beta regression models with both constant and variable dispersion. The method of local influence (Cook,~1986) proved to be particularly useful, since it is able to identify variable dispersion in the data. We have also used Monte Carlo simulation to evaluate the finite sample performance of maximum likelihood estimators in beta regression models with variable dispersion; we have also evaluated the consequences os misspecifying the model by incorrectly assuming constant dispersion when dispersion is variable and the finite sample behavior of heteroskedasticity tests based on first order asymptotics. of estimating the model supposing constant dispersion when Prediction bootstrap intervals (Davison e Hinkley,~1997) for the beta regression model with constant dispersion are also considered.Practical applications that employ real data are presented and discussed.

Nos últimos anos surgiu um tipo de bateria que tem gerado grande interesse para emprego em bancos de armazenamento estacionário. Tal tecnologia baseia-se na utilização de um eletrólito sólido β/β\'\'-Al2O3 que permite o transporte de íons de sódio entre um eletrodo positivo e um negativo. O eletrólito sólido β/β\'\'-Al2O3 apresenta uma estrutura cristalina diferenciada que permite grande mobilidade dos íons de sódio. O presente trabalho tem como objetivo principal obter eletrólito sólido β/β\'\'-Al2O3, utilizando pós preparados pelo processo mistura de óxidos e avaliar o desempenho elétrico dos eletrólitos com a deposição de filmes do mesmo substrato variando a concentração de sólidos presentes e correlacionar sua condutividade elétrica com sua porosidade. As amostras de β/β\'\'-Al2O3 foram sintetizadas, ocorrendo a formação da fase pretendida em temperaturas superiores a 1100°C. Os teores relativos das fases β/β\'\'-Al2O3 mostrou-se dependente da temperatura. Pós calcinados em temperaturas mais elevadas sofrem volatilização do sódio, sendo o teor máximo obtido da fase β\'\' durante o processo foi de 64%. Amostras sinterizadas em dois estágios, a 1600°C por 20 minutos seguida de tratamento térmico a 1475°C por 2 horas, apresentaram densificação maior em relação à amostras sinterizadas em um estágio. Sua microestrutura sofreu crescimento de grãos formando uma microestrutura duplex com grãos na forma de placas alongadas distribuídas em uma matriz de grãos finos. A partir da caracterização de suspensões aquosas de β/β\'\'-Al2O3 evidenciou a alteração do comportamento reológico exibido por suspensões dos pós cerâmicos em função da concentração em volume de sólidos de seu substrato em: 15%, 20%, 30% e 40%, e então os eletrólitos foram conformados por spin-coating. A técnica de espectroscopia de impedância foi importante na identificação das fases presentes na cerâmica β/β\'\'-Al2O3 e no mecanismo de condução elétrica, ocorrendo condução no volume (grãos), havendo uma condução maior na fase β-alumina.
In recent years has emerged a type of battery that has generated great interest for employment in stationary storage banks. Such technology is based on the use of a solid electrolyte β/β\'\'-Al2O3 that allows the transport of sodium ions between a positive and negative electrode. The solid electrolyte β/β\'\'-Al2O3 presents a differentiated crystalline structure that allows great mobility of the sodium ions. The main objective of this work is to obtain solid electrolyte β/β\'\'-Al2O3, using powders prepared by the process of mixing oxides and to evaluate the electric performance of the electrolytes with the deposition of films of the same substrate varying the concentration of solids present and to correlate their Conductivity with its porosity. The samples of β/β\'\'-Al2O3 were synthesized, with the formation of the desired phase occurring at temperatures above 1100°C. The relative levels of the β/β\'\'-Al2O3 phases were temperature dependent. Post calcined at higher temperatures undergoes sodium volatilization, the maximum obtained content of the phase during the process was 64%. Two-stage sintered samples at 1600°C for 20 minutes followed by heat treatment at 1475°C for 2 hours showed higher densification than sintered samples at one stage. Its microstructure underwent grain growth forming a duplex microstructure with grains in the form of elongated plates distributed in a fine grained matrix. From the formulation of aqueous suspensions of β/β\'\'-Al2O3 showed the alteration of the rheological behavior exhibited by suspensions of the ceramic powders as a function of the volume concentration of their substrate solids in: 15%, 20%, 30% and 40%, And then the electrolytes were formed by spin-coating. The impedance spectroscopy technique was important in the identification of the phases present in the ceramic β/β\'\'-Al2O3 and in the electrical conduction mechanism, occurring conduction in the volume (grains), with a higher conduction in the β-alumina phase.

Resumo: No presente trabalho desenvolvemos ferramentas de inferência bayesiana para modelos de regressão beta e beta inflacionados, em relação à estimação paramétrica e diagnóstico. Trabalhamos com modelos de regressão beta não inflacionados, inflacionados em zero ou um e inflacionados em zero e um. Devido à impossibilidade de obtenção analítica das posteriores de interesse, tais ferramentas foram desenvolvidas através de algoritmos MCMC. Para os parâmetros da estrutura de regressão e para o parâmetro de precisão exploramos a utilização de prioris comumente empregadas em modelos de regressão, bem como prioris de Jeffreys e de Jeffreys sob independência. Para os parâmetros das componentes discretas, consideramos prioris conjugadas. Realizamos diversos estudos de simulação considerando algumas situações de interesse prático com o intuito de comparar as estimativas bayesianas com as frequentistas e também de estudar a sensibilidade dos modelos _a escolha de prioris. Um conjunto de dados da área psicométrica foi analisado para ilustrar o potencial do ferramental desenvolvido. Os resultados indicaram que há ganho ao se considerar modelos que contemplam as observações inflacionadas ao invés de transformá-las a fim de utilizar modelos não inflacionados
Abstract: In the present work we developed Bayesian tools, concerning parameter estimation and diagnostics, for noninflated, zero inflated, one inflated and zero-one inflated beta regression models. Due to the impossibility of obtaining the posterior distributions of interest, analytically, all these tools were developed through MCMC algorithms. For the regression and precision parameters we exploited the using of prior distributions commonly considered in regression models as well as Jeffreys and independence Jeffreys priors. For the parameters related to the discrete components, we considered conjugate prior distributions. We performed simulation studies, considering some situations of practical interest, in order to compare the Bayesian and frequentist estimates as well as to evaluate the sensitivity of the models to the prior choice. A psychometric real data set was analyzed to illustrate the performance of the developed tools. The results indicated that there is an overall improvement in using models that consider the inflated observations compared to transforming these observations in order to use noninflated models

Como ponto de partida, este estudo propõe-se a uma revisão de paradigmas consagrados a respeito da HQ, também caracterizar HQ e HQ Moderna, estabelecer as diferenças entre os três grandes gêneros de HQ: a HQ na linha de montagem, a HQ de autor (nos seus dois casos) e o fenômeno intermediário, a HQ das grandes companhias, neste processo ressaltando a que alto nível estético a Nona-Arte pode ascender. Em seguida, demonstrar quão freqüente e rico é o diálogo mantido entre a Nona-Arte e as Belas- Artes e quão plástico-pictórica a HQ pode ser. Como conclusão, são lançadas as bases para a criação de um novo gênero de HQ, com a ênfase ultra-acentuada na plasticidade e de forte caráter alegórico, a Nona-Bela-Arte.

The partition of India in August 1947 remains a watershed in the subcontinent’s history, defining the post-independence relationship between the two countries. The event was marked by the greatest migration in the twentieth century and the death of an estimated one million persons. The causes of partition and reasons for the associated violence have been examined previously. However, existing accounts tend to focus in general terms or at best has a provincial angle with respect to patterns of violence, resettlement and rehabilitation. Research in the past has also tended to stop at August 1947 without looking beyond this period. While there has been move towards examining the “lived” experience of partition, there remains a tendency to avoid locality focused case studies. A comparative India-Pakistan dimension is also missing, even in the ‘new history’ of partition. This thesis seeks to adopt a comparative case study approach. In addition to providing new empirical data, it attempts to uncover the differential experiences of violence, migration and the resettlement of partition refugees within the Punjab region. The thesis argues, firstly that localized patterns of political authority and culture impacted on the differential experience of partition related violence; Secondly, that the experience of partition and dislocation was a process rather than an event confined to August 1947. Finally, the thesis considers the extent to which the input of refugee capital and labour were locally significant in the region’s post-partition urban economic development. The thesis adopts a comparative history methodology with the use of three case studies, namely Malerkotla, and Ludhiana in East Pubjab and Faisalabad, formerly Lyallpur in West Pubjab. The themes explored include the differential experience of partition violence through a comparison between the Muslim Princely State of Malerkotla and the neighbouring British administered districts of the Ludhiana district. Some comparative insights into the role of the state and communal violence are also drawn by means of a brief examination of the circumstances in the Sikh ruled Princely state of Patiala. Patterns of urban migration are also explored, shedding new light on the motives behind places of resettlement. Again, a comparative history methodology is used. Finally, the role of refugee capital and labour in post-independence Indian and Pakistan Pubjabs are examined through the study of Ludhiana and Lyallpur. This approach represents the most sustained comparative examination of partition and its aftermath to date based on locality case studies.

Abstract:
The identification of a novel human integrin $/beta$ subunit, $/beta/sb7$, has been reported in this dissertation. The full-length human $/beta/sb7$ cDNA contains 2798 nucleotides encoding a single long open reading frame of 798 amino acid residues of which 708 are extracellular and 52 are cytoplasmic. The human $/beta/sb7$ sequence displays strong homology with other integrin $/beta$ subunits, being most related to $/beta/sb2$. Cell distribution studies suggest that the expression of transcripts encoding the human $/beta/sb7$ subunit is restricted to leukocytes. This study was the first to show that the cysteine-rich repeats of the human integrin $/beta/sb7$ subunit and other integrin $/beta$ subunits are related to domain III of the laminin B chains. Furthermore, the integrin $/beta$ subunit and laminin cysteine-rich repeats are each related to the epidermal growth factor (EGF)-like repeats found in a growing number of adhesion molecules and regulatory proteins. The mouse homologue of the human $/beta/sb7$ subunit has also been cloned in this study, and it shares 87% amino acid identity with the human $/beta/sb7$ sequence. The mouse $/beta/sb7$ mRNA transcripts, like that of the human $/beta/sb7$, have a predominantly leukocyte-restricted expression, but in some haematopoietic lineage cell lines, in addition to the predominant 3 kb transcript, a less abundant 2 kb transcript is also detectable. Cultured mouse skin epithelial cells contain only the 2 kb transcript. The genes encoding the human and mouse $/beta/sb7$ subunits were mapped to human chromosome 12 (ITGB7) and mouse chromosome 15 (Itgb-7). The Itgb-7 gene was localised to the distal part of mouse chromosome 15, and exists in the gene order: Myc-Wnt-1-Itgb-1. The N-terminal 13 amino acids of the mature mouse $/beta/sb7$ subunit was shown to be identical to a 13 amino acid N-terminal sequence reported for the Mr 120,000 $/beta$ subunit of the M290 antigen, which is found highly expressed on mouse intestinal intraepithelial lymphocytes (IEL). This finding suggests that the integrin $/beta/sb7$ subunit may play important roles in IEL migration, and immunosurveillance in the gut mucosa.

ENGLISH ABSTRACT: The argument of whether to use food for biofuel (bioethanol) production prompted the search for an alternative non-food biomass, such as lignocellulose, as feedstock for bioethanol production. However, a hindrance in producing bioethanol from lignocellulose on an industrial scale is the cost associated with hydrolysing the lignocellulose to its respective sugar monomers. Improving enzyme production and enhancement of enzyme cocktails for efficient lignocellulose hydrolysis is, therefore, a necessary prerequisite. In this study, a yeast culture collection from the Wine and Fermentation Technology Division (ARC Infruitec- Nietvoorbij, Stellenbosch, South Africa), isolated from fruit from various regions in South Africa, was screened for β-glucosidase and β-xylosidase enzyme activities. β-glucosidases catalyse the hydrolysis of cellobiose and by doing so prevents end-product inhibition of cellobiohydrolases and endoglucanases during cellulose degradation. Similarly, β-xylosidases hydrolyse xylobiose and prevents end-product inhibition of endoxylanases during hemicellulose degradation. After initially screening 2180 non- Saccharomyces yeasts, two yeast isolates were selected that could potentially serve as enzyme source for lignocellulose hydrolysis; one as a producer of a β-glucosidase and another as a β-xylosidase producer. The yeasts were identified as a β-glucosidase producing Rhodotorula slooffiae-like yeast isolate 131B2 and a β-xylosidase producing Aureobasidium pullulans isolate 23B25, respectively. The production of β-glucosidase by Rhodotorula slooffiae-like yeast isolate 131B2 and of β-xylosidase by Aureobasidium pullulans isolate 23B25 was optimised using response surface methodology according to a central composite design. Subsequently, the crude and partially purified enzymes were characterised based on molecular mass, pH optima and stability, temperature optima and stability and inhibition by lignocellulose hydrolysis end-products, such as glucose, xylose and ethanol. The crude β-glucosidase from Rhodotorula slooffiae-like yeast isolate 131B2 was also compared to the commercial Aspergillus niger βglucosidase preparation (Novozyme 188) based on the characteristics mentioned above and as βglucosidase supplement during Avicel (microcrystalline cellulose) hydrolysis by the commercial cellulase preparation (Celluclast). The crude β-xylosidase by Aureobasidium pullulans isolate 23B25 could not be compared to a commercial β-xylosidase as none was available at the time of the study. During the study, the crude β-glucosidase 131B2 and β-xylosidase 23B25 showed potential as lignocellulose hydrolytic enzymes. Attempts were made to obtain the β-glucosidase and β-xylosidase genes from the respective yeast isolates using PCR-based approaches and by constructing cDNA libraries. However, cloning the β-glucosidase and β-xylosidase genes using these methods proved after several attempts to be unsuccessful, although, during this section of the study valuable information was obtained about the obstacles involved with using these approaches when the desired gene sequence is unknown and novel.

Abstract:
Bacterial diseases have an enormous impact on human health. The most widespread class of human antibacterials is the β-lactams that target the transpeptidase activity of penicillin-binding proteins, which are responsible for cross-linking the peptidoglycan cell-wall. However, bacteria have gained resistance to all major classes of β-lactams. To protect the clinical utility of the β-lactams it is essential to understand the structural basis for this resistance. This thesis aims to better understand the molecular details governing extended-spectrum β-lactamase mediated β-lactam resistance, and to gain insights into inhibition of these emerging resistance factors. Recently, a novel resistance factor known as the New Delhi Metallo-β-Lactamase-1 has been found to confer enteric pathogens such as Escherichia coli and Klebsiella pneumoniae with nearly complete resistance to all β-lactams. The 2.1Å resolution crystal structure of K. pneumoniae holo-NDM-1 revealed an expansive active site, which we propose leads to a broader β-lactam substrate specificity. Furthermore, NDM-1 localizes to the bacterial outer-membrane by sucrose density gradient centrifugation. The structural details underpinning the broad-spectrum resistance of NDM-1 was further investigated by analysis of the protein in complex with hydrolyzed β-lactams as well as bound to the inhibitor L-captopril. An analysis of the NDM-1 active site in these structures reveals key features important for the informed design of novel inhibitors of NDM-1 and other metallo-β-lactamases. The novel diazabicyclooctane (DBO) avibactam inhibits a wider range of serine β-lactamases than has been previously observed with clinical β-lactamase inhibitors. To understand the molecular basis and spectrum of inhibition by avibactam, we provide structural and mechanistic analysis of the compound in complex with important class A and D serine β-lactamases. A kinetic analysis of key active-site mutants for class A β-lactamase CTX-M-15 allows us to propose a validated mechanism for avibactam-mediated β-lactamase inhibition including a unique role for S130, which acts as a general base. We then show that avibactam derivatives retain β-lactamase inhibitory properties but also exhibit considerable antimicrobial activity against clinically relevant bacteria via targeting penicillin-binding proteins. Our results provide evidence that structure-activity relationship studies for the purposes of drug discovery must consider both β-lactamases and penicillin-binding proteins as targets.
