This paper argues the flexibility of focus group in Design research as a controlled-experiment setting to study Designerly Intuition at concept-stage decision making. This paper outlines the focus group design in terms of the types and structure considerations and the moderator factor. Finally, the focus-group result gathers dual-mode input from novice Industrial Designers (1) 'as a group' and the intuitive attributes as (2) ‘individual participants’. Therefore, the focus group design for the data collection setting is established through the research layout and space design that accommodates both modes of inquiry. This case addresses two frequently heard questions in real-world business settings: Why are sales going down? and What decisions must we make to reverse the sales decline? These questions were directed to Dr. Felix Wilder, a professor of Marketing, by the senior management of Big Red, Ltd., headquartered in Waco, Texas, after sales reports indicated a significant decline in year-over-year sales in the Louisville, Kentucky, market region. This region represented one of Big Reds most important market areas for soft drinks outside of its home base in Texas. Members of Big Reds senior management team were unable to identify the specific factors that had contributed to the sales decline. As a result, they commissioned Dr. Wilder to conduct an extensive marketing research project to determine the factors contributing to the erosion in sales. Dr. Wilder proposed a multifaceted exploratory research project that involved an extensive review of industry and trade literature, in-depth interviews with grocery store and super center store managers, an in-store consumer survey of Big Red customers, and a consumer focus group. The research design focused on the marketing mix variables, environmental factors, the target market, and changing consumer purchase behavior and lifestyle issues as possible sources contributing to the sales erosion. The results of Dr. Wilders investigation conflicted with Big Red managements preliminary hypotheses and revealed unexpected factors that may have contributed to the downward drift in sales. Based on the research, Dr. Wilder presented management with various alternative courses of action to correct the sales decline.

A study was conducted on the performance of FPOs and the factors contributing to performance of FPOs in Medak district of Telangana State, India. Three FPOs were selected randomly from 3 different promoting institutes viz., Suraksha Farmers Producer Company Ltd (SFPCL) promoted by independent research organization Centre for Sustainable Agriculture (CSA), Marpalli Kisan Kranthi Producer Company Ltd (MKKPCL) promoted by Vrutti NGO and Siddipet Kisan Agro Farmers Producer Company Ltd (SKAFPCL) promoted by International Crops Research Institute for the Semi-arid Tropics (ICRISAT). The Ex-post facto research design was adopted for the study with a sample of 90 producer members, covering three FPOs in erstwhile Medak district of Telangana State. From the analysis, it was found that overall performance of FPOs in Medak district was average to poor. The factors viz., education, group leadership, group communication, adherence to rules, group participation, and team spirit had showed positive and significant relationship with performance. The results of regression analysis inferred that all the selected twelve factors put together explained the variation to the extent of 72.47% in the performance of the FPO with leadership of officials and participation of members contributing significantly to the performance of FPOs.

Background: In the daily practice, clinical effectiveness of pegfilgrastim compared to that of daily filgrastim in patients with malignant lymphoma is still unclear. This study aimed to clarify the effectiveness of pegfilgrastim versus daily filgrastim, using a national inpatient database in Japan. Study Design and Methods: We retrospectively reviewed 18095 patients with newly diagnosed diffuse large B-cell lymphoma (DLBCL) who received the first R-CHOP treatment between July 2007 and March 2017. Patients who received primary G-CSF prophylaxis were divided into pegfilgrastim group and filgrastim group. Outcomes included incidence of febrile neutropenia, all-cause in-hospital death, length of hospital stay, and total costs. To account for measured confounding, patients' characteristics were adjusted using two different propensity score utilizing methods: propensity score matching (PSM) and stabilized inverse probability of treatment weighting (IPTW). Instrumental variable (IV) analysis was also performed to account for unmeasured confounding. Results: We identified 1299 patients in the pegfilgrastim group and 2203 patients in the filgrastim group. FN occurred in 380 of the 3502 (11%) total patients with a median of 9 days (interquartile range [IQR] 7-10) after the initiation of the chemotherapy. All-cause in-hospital death occurred in 94 (2.7%) of 3502 patients. Median of hospital stay was 17 days (IQR 13-22), and total medical costs during hospitalization were 7300 (IQR 5700-9500) in USD. By one-to-one PSM, 1294 patients for both the pegfilgrastim and filgrastim groups were selected. Pegfilgrastim was administered 2 days (median, IQR 1-3) after the chemotherapy. Daily filgrastim administration was initiated 5 days (median, IQR 3-6) after the chemotherapy, and continued for 6 days (median, IQR 2-7). Compared with the filgrastim group, the pegfilgrastim group showed significant lower risk of the FN incidence (risk difference 6.1%, 95% confidence interval [CI] 4.1-8.1) and all-cause in-hospital mortality (risk difference 0.9%, 95% CI 0.05-1.8). In similar, stabilized IPTW showed that pegfilgrastim group was significantly associated with the lower risk of FN (risk difference 7.0%, 95% CI 5.1-8.9) and in-hospital mortality (risk difference 1.8%, 95% CI 0.9-2.7). In PSM, the length of hospital stay and total costs during hospitalization were also significantly decreased in the pegfilgrastim group compared to the filgrastim group (percent reduction 34% [95% CI 31-37], percent reduction 12% [95% CI 9-15], respectively). Stabilized IPTW also showed the significant percent reduction of the pegfilgrastim group to the filgrastim group for length of hospital stay and total costs (31% [95% CI 25-35], 19% [95% CI 13-24], respectively). In the sensitivity analyses, IV methods showed significantly lower FN susceptibility (odds ratio 0.14, 95% CI 0.07-0.26). However, pegfilgrastim was not associated with the reduction in susceptibility of in-hospital mortality (odds ratio 0.93, 95% CI 0.23-3.70). The ratio of the pegfilgrastim group to the filgrastim group for length of hospital stay and total costs were 55% (95% CI 51-60) and 78% (95% CI 73-84), respectively. Conclusion: Pegfilgrastim contributed to lower susceptibility of FN. Total length of hospital stay and medical costs were also decreased in the pegfilgrastim group. ABSTRACT Introduction Ceramics are widely applied in dentistry owing to their excellent mechanical and physical attributes. The most popular ceramics are Lava™, KaVo Everest, and Cercon. However, it is unclear whether or not a different surface treatment along with low-temperature aging and mechanical loading (ML) affects the physical properties of computer-aided design (CAD)/computer-aided manufacturing (CAM)-machined yttria-stabilized tetragonal zirconia polycrystal (Y-TZP) ceramic. Aim The objective of this research was to assess the impact of various surface treatments as air-particle abrasion, ML, low-temperature degradation (LTD), and their cumulative effects on biaxial flexural properties of Y-TZP. Materials and methods Totally, 50 specimens were fabricated by CAD–CAM machining from Cercon® and divided into five groups following different surface treatments as control (C), air-particle abrasion (Si), ML, LTD, and cumulative treatment (CT) group. Results were investigated by two-way analysis of variance (ANOVA) and Tukey honest significant difference (HSD) test. Results The highest biaxial flexural strength was observed in the Si group (950.2 ± 126.7 MPa), followed by the LTD group (861.3 ± 166.8 MPa), CT group (851.2 ± 126.5 MPa), and the least with ML (820 ± 110 MPa). A significant difference was observed in the two-way ANOVA test. X-ray diffraction (XRD) analysis showed that the control group consists of 100% tetragonal zirconia and the maximum amount of monoclinic phase was obtained after LTD. Conclusion No negative effect on biaxial flexural strength was observed; indeed, it increases the biaxial strength. Hence, these surface treatments can be done in routine clinical practice to improve the performance of ceramic restoration. Purpose One way to increase the availability and to add more value to exotic Brazilian fruits is to develop new products. However, prior to product development, there is a need to know consumer’s preferences and target audience. The purpose of this paper is to investigate the influence of different characteristics of Cerrado fruit preserves on the intention to purchase using conjoint analysis. Design/methodology/approach Conjoint analysis evaluated three factors with three levels each, defined by focus group technique. The attributes and levels evaluated were: flavor (marolo, marolo and soursoup, marolo, soursoup and sweet passion fruit), nutritional information (light, diet and absent) and health claims (aids reduction of cardiovascular disease risk; aids reduction of blood cholesterol; aids activation and regulation of the gastrointestinal system). Findings The results of conjoint analysis led to three consumers’ groups. Group 1 (majority) were more influenced by flavor (IR=26.5 percent) and nutritional information (IR=59.3 percent). Groups 2 and 3 were greatly influenced by flavor and nutritional information, respectively. Health claims influenced weakly on the intention to purchase regardless the groups. Thus, the concept of Cerrado fruit preserves for each group was different. Originality/value This research contributes to future studies, supporting the development of products based on Cerrado fruits and the variety of products on the market. There are few research works using conjoint analysis to evaluate consumer preferences in the early stages of new product development, which makes this paper even more relevant.

Purpose The purpose of this paper is to understand consumers’ behaviour in fast-food restaurants in Spain. To this end, the authors conducted a survey that combined a classification of food values, as proposed in the relevant literature, with a related model that links personal values to behaviour. Design/methodology/approach A sample of 400 consumers was gathered from two different, leading fast-food chains operating in Spain. With these data, respondents were grouped through hierarchical cluster analysis and K-measures, and in accordance with Lusk and Briggeman’s (2009) food values and the food-related lifestyle model. The authors validated these clusters by means of ANOVA and discriminant analysis, which led to useful observations about inter-group differences in consumers’ habits, as well as their satisfaction, trust and loyalty. Findings The results indicate that consumers can be clustered into three groups based their food values assessments: the “mainly utilitarian” group, the “mainly hedonic” group and the “ethical values” group. These groups not only demonstrate diverse habits, but also differ on key variables such as satisfaction, trust and loyalty. Practical implications The authors offer several managerial recommendations for designing and developing segmentation strategies in the fast-food industry. Any such strategies should acknowledge that all consumer groups appear to value restaurants’ efforts to provide them with both hedonic and utilitarian benefits, although the extent varies across groups. Originality/value Among the relevant literature, this research is the only one that examines the existence of distinct consumer groups based on their food values assessments. In addition, this paper analyses inter-group differences in terms of both diverse consumptions habits (frequency of visits, expenditure, etc.) and key marketing variables (satisfaction, trust and loyalty).

The result of this study was brought to identify the promotion of pedestrian safety zone. In the participatory process, the four groups were started up for collaboration including teacher and student group, local community group, academics group, and private organization group. Brainstorming is a series of procedures (rules) designed to maximize the productivity of groups engaged in idea generation by reducing production loss, popularized by Osborn, an advertising executive. The main concern in this tool is increasing creativity in an organization. This study examines two main factors, production loss and ownership of the topic that influence the performance in group brainstorming. Production loss includes activities in production blocking, social loafing and evaluation apprehension among individuals, while ownership of the topic relates to the interest of individuals in engaging the brainstorming activities especially in industrial design. Using a quasi-experimental research design, this study reports on the participation of115 groups of university students from 6 different universities. The data is analyzed at group-level. The hypotheses of this study are tested using Partial Least Squares (PLS) regression. Results reveal that out of the three dimensions of production loss, production blocking, social loafing, and evaluation apprehension, production blocking is negatively related to the group brainstorming performance whereas evaluation apprehension is positively related to the performance or group brainstorming. As expected, ownership or the topic is related to the performance too. However, social loafing is not related to the performance or group brainstorming. The results are of potential interest to educators, and researchers. This study responds 10 a call for further brainstorming research.

Taiyuan Boiler Group Co. Ltd. designed and developed an 850t/h CFB boiler with energy saving technology on the principle of “State Specification Design Theory”. By measuring the ash formation and attrition characteristic of the design coal, with the one dimension mass balance model developed by Tsinghua University, the bed quality of the bed inventory and the pressure drop were modified. Based on the particle density calculated, the heating transfer coefficients were chosen and the arrangements of the heating surfaces were modified. According to the special operation conditions of the user, the practical working conditions for the boiler are determined so as to assure the technical characteristics and structural properties in 850t/h CFB boiler.

Background Azacitidine (AZA) is one of the hypomethylating agents (HMAs) proven to significantly prolong overall survival (OS) for patients with higher-risk myelodysplastic syndromes (h-MDS) via a prospective phase 3 trial. However, the optimal treatment schedule of AZA for h-MDS has not yet been prospectively determined. As a 7-day treatment schedule includes weekends, a 5-day administration has also been applied with h-MDS, though solid evidence has not been established. Based on this background, we planned to compare AZA treatment on 7-day and 5-day schedules. Patients Patients diagnosed with de novo or treatment-related MDS (FAB-defined RAEB and RAEB-t), aged 16 years or older were eligible for this study, if they showed adequate performance status (ECOG PS 0-2) and no history of HMA treatment or chemotherapy. Candidates for allogeneic stem-cell transplantation were excluded. Study design This was a multicenter, randomized, open-label, phase 3 trial to compare the efficacy of AZA treatment for 7-days (AZA-7) to 5-days (AZA-5). The primary endpoint was 2-year OS rate (2y OS). AZA was given subcutaneously at 75 mg/m² every 28 days. To test the non-inferiority of AZA-5 to AZA-7, 2y OS of AZA-7 was estimated at 30%, and the delta was defined as 11%, for which 410 patients were needed to complete the trial. However, because of the poor recruitment, this study closed prematurely, and the protocol-planned interim analysis (when 200 patients were registered) was performed. This protocol was approved by the IRB at each trial center. All patients provided written informed consent, and the trial was conducted in accordance with the Declaration of Helsinki. The efficacy of AZA was evaluated using IWG2006 criteria, and treatment was continued until study completion, or relapse, unacceptable toxicity, or disease progression was observed. Efficacy was analyzed by intention-to-treat. Secondary endpoints were the hematological response rate, 2-year leukemia-free survival (2y LFS) rate, the cytogenetic response, and occurrence of adverse events. Results Between January 2013 and Jun 2018, 201 patients were randomly assigned to AZA-7 (n=99) or AZA-5 group (n=102). The median age of all patients at the enrollment was 73.5 years old (range, 48 to 91). Between the two groups, there was no significant difference in the baseline characteristics, such as sex, type of MDS (de novo or treatment-related), percent of BM blasts, FAB and WHO classification, and IPSS and IPSS-R risk. After excluding 5 and 6 patients from AZA-7 and AZA-5, respectively (poor data entry, change in diagnosis), at the time of last follow-up, 59 and 67 patients died in AZA-7 and AZA-5, respectively. The Kaplan-Meier estimates of the 2y OS and 2y LFS were 35.1% (95% CI, 23.8 to 46.7) and 27.3% (95% CI, 17.3 to 38.3) for AZA-7, and 22.4% (95% CI, 13.1 to 33.3) and 20.5% (95% CI, 11.6 to 31.2) for AZA-5, respectively. There was no significant difference in the frequency of erythroid, platelet or neutrophil improvement, or hematological response (complete or partial response, or any hematological improvement) between the two groups. The most common non-hematological adverse event of grade 3 to 4 (in ≥20% of the patients in either group) was febrile neutropenia (33.7% for AZA-7, and 30.5% for AZA-5). Conclusion and Discussion Because of the premature termination of this trial, statistical analysis for the primary endpoint (2y OS of 35.1% for AZA-7, and 22.4% for AZA-5) could not provide any solid evidence. However, considering the difference of more than 10% in 2y OS, and no major differences in the safety profiles, we favor AZA-7 to AZA-5 for MDS patients with RAEB and RAEB-t. Purpose The purpose of this paper is to report the results of a study that investigated the barriers and facilitators associated with the adoption of a hazard analysis critical control point based food safety management system (HACCP FSMS) in dairy processing companies of Armenia. Design/methodology/approach A cross-sectional qualitative research design was employed to examine managerial experience and perceptions of barriers and drivers to organizational adoption of HACCP FSMS procedures. The data were collected in 20 food processing facilities in the dairy industry through individual in-depth interviews with their representatives and in governmental and non-governmental organizations through focus group discussions with 23 policy makers and food safety consultants. Diffusion of innovations theory provided a framework for systematic exploration of the phenomenon under study. Findings Most frequently reported drivers of adoption included enhanced traceability, increased export opportunities, improved organizational image, and broader accountability. Major impeding factors, such as high investment costs, value incompatibility, excessive documentation, inadequate physical and technological infrastructures, and low observability led to less favorable attitudes toward the organizational change needed for HACCP FSMS adoption. The results indicate that larger organizations with well-developed infrastructures and skilled workforce have an advantage over smaller organizations in the process of adoption. Originality/value This study provides a comprehensive theory-based analysis of perceived determinants of organization-wide adoption of HACCP-based system in Armenia. The results of the study support and extend the findings of earlier research and provide new insights into HACCP FSMS adoption across different contexts.

Purpose Despite the perfect nutritional value of imperfect “ugly” produce, they are either never harvested or discarded, resulting in escalating farm-to-fork food waste problems in the USA. The purpose of this paper is to investigate US consumers’ perceptions of imperfect “ugly” produce and their willingness to purchase and consume these foods in their households. Design/methodology/approach Four focus group sessions with at least five voluntary participants per session were conducted. Each session used structured interview questions and the discussions were led by the same moderator. All sessions were recorded, transcribed and analyzed based on published focus group guidelines and recommendations. Findings The research study has revealed two categories of potential consumers, Influencers and Learners, when it comes to an initial understanding of “ugly” produce consumption. The study also identified nine themes (i.e. food safety, price sensitivity, location of sale, product display, name/label, purpose of use, fun, targeted education and food waste) that consumers hold regarding imperfect “ugly” produce. Research limitations/implications Insights from this study provide strategies for sustainable food management that could help stakeholders along all points of the food chain save money, reduce hunger and diminish food waste in our communities. Originality/value This study serves to provide preliminary insights in approaching the issue of consumers’ perception of imperfect “ugly” fruits and vegetables from a research perspective. Although a limited number of real consumers from the local community participated, their rich and in-depth qualitative responses defined the scope and the defining questions that need further exploration.

Abstract Background UCBT has proven effective in treating patients with hematologic malignancies (HM),but engraftment is slower and graft failure rates higher compared to other unrelated transplants. We have previously shown that incubation of UCB CD133+ cells with cytokines and the copper chelator TEPA (5µM), inhibits stem cell differentiation and results in a median of 89 and 30 fold expansion of CD34+ and CD34+CD38- cells, respectively (Cytotherapy 2004;6:344). Using this technology (StemEx®) we performed a prospective multicenter myeloablative UCBT trial in patients with HM. Methodology Patients were transplanted with a single CBU of which CD133+ cells from a segregated portion of the CBU (20-50%) were cultured for 21 days with hematopoietic cytokines and TEPA and transplanted along with a minimum of 107nucleated cells (NC)/kg from the un-manipulated (UM) portion of the same unit (NCT00469729). The primary endpoint of this study was 100 day overall survival. Using an intent to treat design, outcome was compared to a 2006-2010 double UCBT (dUCBT) control group (n = 295) collected from and by the CIBMTR and Eurocord registries using identical eligibility criteria to the StemEx® study: lack of a 5/6 or 6/6 matched sibling donor, age 12-55 years with high risk AML or ALL in 1stCR or subsequent, advanced CML or after failing TKI, MDS with Int-2 or high risk features or chemosensitive relapsed lymphoma. GvHD prophylaxis included a calcineurin inhibitor and mycophenolate mofetil. Comparison of the primary endpoint was based on a logistic regression model that adjusts the treatment comparison for imbalance between the groups in important prognostic factors found to impact mortality: age, sex, CMV status and disease risk. Results 25 centers in US, EU and Israel enrolled 101 eligible patients between Oct 2007-Feb 2012 with AML-43, ALL-30, MDS and CML-8 each, and lymphoma-12. Median age was 37 (12.6-55.8); median weight was 68 kg (42.5-128.5). The baseline NC and CD34 cell dose/kg were 3.06 (1.29-11.0) x 107 and 1.64 (0.24-9.23) x 105. 70% of the units were matched at 4/6 loci. Median NC and CD34 fold expansion were 400 (0-764) and 77 (6-280), respectively. StemEx yielded a median of 14-fold increase in the number of CD34+ infused, in comparison to the number of CD34+ cells the patients could have received from the entire UM CBU. In total, patients received a median dose of 2.2 x 107 NC/kg and 9.7 x 105CD34/kg. No significant acute toxicity was seen with the expanded cell infusions. The primary endpoint of this study has been met: 100 day survival was significantly higher in the StemEx® vs control group; 84.2 vs 74.6 % [95% CI 0.5 (0.26-0.95); p = 0.035]. Neutrophil and platelet engraftment rates were faster in the StemEx® vs control group: 21 vs 28 days (p< 0.0001) and 54 vs 105 days (p = 0.008), respectively. There was a trend in the reduction of engraftment failure from 14.4% in the control to 8.1% in the StemEx® group, p=0.086. Early engraftment (EE) (ANC ≤ day 20 and platelets ≤ day 60) was achieved more frequently in StemEx® (39.4%) than in the control arm (12.4%) , p<0.001. EE was associated with improved 100 day survival (p = 0.0028). Grade III/IV aGvHD and cGvHD rates did not differ between the study and control groups (19.4 vs 16.9%; p = 0.107) and 18.4 vs 16.0% (p = 0.731), respectively. Of the 101 patients, 16 patients died in the first 100 days. Causes of death were infection (5), multiorgan failure/ARDS (4), VOD (2) and relapse, acute GvHD, graft failure, hemorrhagic CVA and DIC (1 each). Importantly, the CD34+ cell dose from the StemEx expanded fraction was associated with time to ANC and platelet engraftment (p<0.001, 0.011, respectively) and inversely associated with grade III/IV infections during 100 day post-transplant (p = 0.023). Conclusions This multi-international study demonstrated the advantage of StemEx® over dUCBT historical controls,measured by a significant improvement in day 100 survival and faster engraftment of ANC and platelets. The robust associations obtained between CD34+ cell dose derived from the expanded portion and graft functionality provide additional support for the clinical benefit obtained over the dUCBT historical control arm. This technology holds the promise of increasing the number of UCBT being performed while potentially reducing its short term morbidity and mortality. In other words, cost, schedule adherence, and quality for very sophisticated ships are fully dependent on the performance of the electrical work. Ishikawajima-Harima Heavy Industries, Co., Ltd. (IHI) has been exploiting zone logic, also recognized as group technology, for construction of virtually all ship types. But, the extensive cable footage in sophisticated ships requires special considerations and techniques. This paper presents practical design and production processes for zone outfitting electric cable. Special focus is on:functional and detail design,conversion of system-oriented design data to zone-oriented work packages called "pallets," andwork methods currently employed in IHI shipyards.

Purpose The growing prevalence of single-person households in South Korea has started to affect the country’s restaurant business. The phenomenon has led to changes in how the interiors of food service establishments are designed. The purpose of this paper is to measure consumers’ emotions when they ate alone and when they saw someone else eating alone to investigate how these emotions can be used in marketing strategies. Design/methodology/approach The data were collected via self-administered surveys to individuals in Daejeon in South Korea. A total of 163 respondents were surveyed and divided into two groups on the basis of their frequency of eating alone: low frequency of eating alone and high frequency of eating alone (HEA). The HEA group was further sub-divided to identify significant differences in greater detail. An independent t-test and descriptive analyses were conducted on the data. Findings Significant differences were observed in the emotions of “be proud of oneself,” “sadness,” “extroversion,” and “loneliness.” A majority of the respondents ate alone because it saved them time. They also preferred to eat home meal replacements when they ate alone. Originality/value The results of this study help understand consumers’ emotions when eating alone in relation to their behaviors at food service establishments. They can inform decisions on market strategies developed by food businesses targeting single consumers.

PurposeThe purpose of this paper is to assess the relationship between nutrition knowledge and grocery store nutrition label use, with using nutrition information disclosure on menu selection in a group of hospitality management students, who shall be responsible for menu labelling in their future careers.Design/methodology/approachA between-subject design included 324 students, who were randomly assigned to choose from a menu labelled as follows: unlabelled; kcal label only; graphical label providing information on the per cent of the recommended daily intake of energy and four nutrients. Their nutrition knowledge and habit of reading grocery store nutrition labels were tested using an additional questionnaire.FindingsThe results showed that the provision of energy value information resulted in the selection of less energetic, less fat and less salted food, while a graphical label additionally led to the selection of food having a lower saturated fatty acid (SFA) and sugar content. Multiple regression analysis showed that the habit of packaged food nutrition label reading was a significant predictor of choosing food having a lower energy (p<0.001), fat (p<0.001), SFA (p<0.001), sugar (p<0.001) and salt (p=0.003) content, while the influence of nutrition knowledge on food selection was proven insignificant.Originality/valueGiven the established positive impact of menu labelling, these findings support the future European policy mandating energy and nutrient content disclosure on menus, but also point to the need for more-intense consumer education.

Abstract Treatment with DEX, rather than prednisone (PRED), improves outcome for children with standard risk ALL. However, DEX exposure is strongly associated with the development of therapy-related ON, particularly in adolescents. Previous COG HR-ALL studies have shown a lower ON risk for patients receiving 1 vs. 2 delayed intensification (DI) phases, and for DI using discontinuous DEX (days 1–7 & 15–21) vs. continuous DEX (days 1–21), suggesting a strategy for giving the drug with acceptable toxicity. The HRALL study COG AALL0232 utilizes a modified augmented BFM backbone that compares in a 2x2 randomized design: induction DEX (10 mg/M2/day x14 days) vs PRED (60 mg/M2/day x28 days), and interim maintenance (IM) escalating-dose “Capizzi” methotrexate vs. high-dose (HD) MTX. Induction rapid early responders (RER) receive single DI while slow responders receive double DI; all patients receive monthly 5-day DEX pulses during maintenance. To limit ON risk in adolescents, in the initial study design children ≥ 13y received discontinuous DEX during single or double DI; those <13y received continuous DEX. In 10/2006 the study was amended due to an unexpectedly high ON incidence in patients 10–12y receiving continuous DEX (28% @ 18 months) compared with historical controls given discontinuous DEX (3.4%). Subsequently all patients ≥ 10y have received discontinuous DEX; it is too early to assess the impact of this change. A comprehensive interim ON analysis was completed 4/2008 (reported as 24-month cumulative incidences). Overall ON incidence is 10.4% (110/1647), and is higher for those age ≥ 10 vs. <10y (15.2 vs 2.6%, p<0.0001, RHR=6.38); 99/110 cases of ON occurred in the older cohort. Among all patients, ON incidence is higher in DEX vs. PRED regimens (11.6 vs 8.7%, p=0.014, RHR 1.64); rates are similar for Capizzi MTX vs. HD-MTX regimens (10.4 vs 9.8%). Among patients ≥ 13y, incidence is higher in DEX vs. PRED regimens (18.9 vs 9.9%, p=0.02, RHR 1.97). There is no difference between regimens for children <10y. Among randomized RER patients ≥ 10y, incidence is higher in DEX vs. PRED regimens (17.2 vs 12.6%, p=0.006, RHR=1.79). For historical comparison, ON incidences by age cohort for RER patients on AALL0232 regimen PC (PRED + Capizzi MTX) vs. CCG-1961 regimen D (double DI with discontinuous DEX) were: <10y 4.1±3.4 vs. 2.0±1.4%, 10–12y 21.9±10.6 vs. 7.1±2.8%, and ≥ 13y 7.1±10.1 vs. 6.6±3.8%. To address these findings, the study was amended 6/2008. Patients ≥ 10y will be non-randomly assigned to induction PRED; the induction steroid randomization will continue for younger patients. Patients of all ages will receive discontinuous DEX during DI and PRED pulses during maintenance. Of note, compared with CCG-1961 the AALL0232 augmented BFM backbone was non-randomly modified in several ways that may affect ON incidence, including the use of pegaspargase during induction, a higher 15 mg intrathecal methotrexate dose for those age ≥ 9y, and monthly DEX instead of PRED maintenance pulses. Heightened awareness among caregivers may also have led to increased recognition and reporting of this toxicity. Using CTCAE v3.0 criteria, clinical ON severity among the 110 patients is: 3% grade 1, 60% grade 2, 35% grade 3, and 2% grade 4. Data regarding surgical intervention are being collected. The structure of half airtight cover is optimized by used of CFD, and the curve type is suitable shape to collect the dust.

The paper discusses the development of interview and focus group instruments to uncover the intuition occurrences in Designerly practices. The two data collecting method forms data source triangulation through in-depth interviews of eight experts and eight senior-level designers with four focus group sessions involving thirty-two novice-level designers. The instrument development took into consideration its reliability and validity for the triangulation analysis stage to follow. The result from four pilot interviews and one focus group discussion provides recommendation and refinement for the researcher’s guide as well as various data collection methods within the Design and Designerly research area. Over 100 years have elapsed since Sidney Ringer made the serendipitous discovery that calcium played a crucial role in amphibian cardiac contraction. Since then we have learned that this ion is an obligatory requirement for cardiac muscle of all species, and that the regulation of intracellular calcium levels is considerably more complex in the mammalian heart than previously thought. Part of this complexity is due to the involved design requirements of mammalian physiological processes. Another element of complexity is introduced by the quantitative differences in the involvement of various regulatory processes in different species. Finally, many significant technological advances in methods for investigating cardiac cellular functions have provided exciting experimental data. However, these data must be integrated into a unifying framework of knowledge of cardiac functions. Among the exciting recent developments are the use of a patch clamp technique to discover different kinds of calcium channels, a highly refined skinned fiber technique to study calcium-induced calcium release, and calcium indicator dyes and laser diffraction and scattering techniques to study the dynamics of calcium handling by the cell. Purpose Seaports are a signifier for the world economy and international trade. Notwithstanding the considerable role of Chinese ports in global trade, only few studies have explored the efficiency of Chinese container terminals. Furthermore, studies on Chinese port efficiency has typically centered on port-level analysis, not terminal level. Therefore, this study aims to examine the operation efficiency of Chinese container terminals. Design/methodology/approach This study uses super-efficiency data envelopment analysis (SE-DEA) approach. SE-DEA is superior than basic DEA model because it is feasible for categorizing and ranking the efficiency of container terminals more accurately and comprehensively. In the basic model, if the several decision-making units (DMUs) are efficient, the efficiency value of them is “1.” However, in the SE-DEA model, the most efficient DMU is over “1.” Based on the level of container throughput in 2018, the top 20 Chinese container terminal companies were selected. Various production quotas were selected as inputs, while the container throughput was considered output. Findings The findings show that Terminal Shanghai Mingdong Container Terminal Co., Ltd. was ranked 1, followed by Shanghai Shengdong International Container Terminal Co., Ltd., Shanghai International Port (Group) Co., Ltd. and Yidong Container Terminal Branch. Originality/value This study contributes to providing some insights into Chinese container terminal industry to augment the efficiency. This study also provides practical and policy implications (e.g. better terminal operations) for container terminals.

This study aims to present an adapted model of Rapoport’s environmental-behavioural study model for home interior spaces design according to family relations in Islam. Suggested model is designed in regards to Islamic content in the boundaries of Islamic-Iranian concept. The research results were confirmed by the use of Delphi Method. Proficiency examination of this model, introduced as an educational tool of home space design model, was carried out through two undergraduate student groups of analyse and control. The east wing of the third new mine of Fengfeng Group co., LTD has been designed to construct about 2490m with a long service time, bad geological conditions and slowly driving speed. Through improving rock drivage construction techniques, the mine has taken measures of driving by three teams, a scientific construction organization of digging while spraying and a kind of advancing crossover operation method of after four in the first four. In addition, the mine also strengthens the management and takes scientific and reasonable scoring model and acceptance criteria. Through all these measures the mine has improved the driving speed effectively and make on the single lane rock drivage footage of more than 120 m good grades. The effect of roadway supporting and forming is of good quality and it has never had safety accidents. Whats more, the economic benefits and social benefits are obvious. It provides an important reference significance for similar quick rock drivage project.

A cohousing community is created and managed by its residents. It is a new collaborative housing concept designed to foster meaningful relationships, closer social bonding and energy efficiency concepts. This paper aims to discuss the key principles, priorities and challenges of cohousing design process through interviews with four cohousing project architects and four members of the design group. Semi-structured interviews were carried out i) to understand residents’ thinking and behaviour change through living in the cohousing community, and ii) to establish the environmental and social sustainability in a cohousing setting. The study found that the development procedure of cohousing highly differs from the mainstream housing design. The quality of the design can potentially influence the residents’ daily life. The findings will benefit a range of groups. It will be an important reference for cohousing design standards. Case overview/synopsis The case study discusses the brand strategy, which Walmart Inc needs to adopt post its acquisition of Flipkart Pvt. Ltd (Flipkart) Group in India. Flipkart had acquired Myntra Designs Pvt. Ltd (Myntra) and Novarris Fashion Trading Private Limited (Jabong), but had kept their brand identity intact; Walmart Inc was faced with the decision on moving ahead with the brand strategy of keeping individual brand identities or merging all of these into a single brand identity. The study aims to provide valuable insights into the decision-making process adopted by Walmart Inc. It includes also the role of cause-related marketing in the positioning of Myntra as a socially responsible brand. The case study opted for an exploratory research design study using the qualitative research method of in-depth interviews. In total, 10 experts in the area of marketing, market research and marketing communication were interviewed. The qualitative data were analyzed using a template approach, which analyzes the text using a codebook or an analysis guide. The analysis guide already has clearly defined themes or categories. As the qualitative interviews progress, these themes get revised. These themes are analyzed qualitatively rather than statistically. The case study suggests to the management of Walmart Inc that they need to merge Myntra and Jabong based on the degree of similarity of consumer demographics, income/social class of buyers, brand identity and buying behavior. Myntra needs to retain as opposed to Jabong, as Myntra is perceived to be a socially responsible brand that creates a purchase disposition in the minds of the consumers. A more extensive quantitative study would offer better generalizability. It was not feasible to conduct a quantitative study due to time constraints. This research would have used advanced brand imagery assessment techniques such as multi-dimensional scaling to suggest if an overlap exists between consumer segments of Myntra and Jabong. The case study provides a decision-making framework to firms and individuals who are part of organizational teams to create a post-acquisition brand strategy in the e-commerce market. The case study fulfills a need for many academicians and practitioners to understand the decision-making process followed in devising a post-acquisition brand strategy in India. There are now more than 1,000 constructed wetland systems (CWs) in the UK. The first UK CW database was constructed by Water Research Centre (WRc) and Severn Trent Water Ltd to accompany a book on the design and performance of these systems. In that database, constructed by Gareth Job et al. (1996), only 154 beds were listed, most of which were tertiary sewage treatment sites in Severn Trent Water. The Constructed Wetland Association (CWA) was formed in 2000 as a UK water industry body in response to problems caused by unscrupulous constructors. A group of experienced, reputable designers and constructors formed the CWA to bring together best UK practice in order to counteract this problem. The group contains major water companies, designers, constructors, academics, plant growers and operators. They decided that one of the best ways of countering the problem was to assemble a database of design and performance from well-designed systems. After negotiation the CWA group took over responsibility for the database from WRc. The CWA has produced eight updates of the database which now contains information from more than 900 beds. It contains examples of the different variants of CWs in use in the UK. Most of these sites treat sewage/domestic wastewater but the database also includes examples of systems for the treatment of minewater, sludge, landfill leachate, industrial effluents, surface runoff and road runoff. Particular treatment applications are illustrated by case studies which are summary articles describing design, construction and performance.

Background: In electric-acoustic pitch matching experiments in patients with single-sided deafness and a cochlear implant, the observed “mismatch” between perceived pitch and predicted pitch, based on the amended Greenwood frequency map, ranges from –1 to –2 octaves. It is unknown if and how this mismatch differs for perimodiolar versus lateral wall electrode arrays. Objectives: We aimed to investigate if the type of electrode array design is of influence on the electric-acoustic pitch match. Method: Fourteen patients (n = 8 with CI422 + lateral wall electrode array, n = 6 with CI512 + perimodiolar electrode array; Cochlear Ltd.) compared the pitch of acoustic stimuli to the pitch of electric stimuli at two test sessions (average interval 4.3 months). We plotted these “pitch matches” per electrode contact against insertion angle, calculated from high-resolution computed tomography scans. The difference between these pitch matches and two references (the spiral ganglion map and the default frequency allocation by Cochlear Ltd.) was defined as “mismatch.” Results: We found average mismatches of –2.2 octaves for the CI422 group and –1.3 octaves for the CI512 group. For any given electrode contact, the mismatch was smaller for the CI512 electrode array than for the CI422 electrode array. For all electrode contacts together, there was a significant difference between the mismatches of the two groups (p < 0.05). Results remained stable over time, with no significant difference between the two test sessions considering all electrode contacts. Neither group showed a significant correlation between the mismatch and phoneme recognition scores. Conclusion: The pitch mismatch was smaller for the perimodiolar electrode array than for the lateral wall electrode array.

To save the labor cost, industry robot is applied in production line. This paper deals with the design of the beverage packaging line using high speed robots, which will be applied in Hangzhou Wahaha Group Co., Ltd. to satisfy their requirement. On the basis of investigation of the previous line, the packaging process is briefly addressed. Taking into account the circumstance of the workshop area, the layout of the packaging line is proposed. With Petri net technology, the model of the beverage packaging line is built and by analyzing the model, it is proved that the whole system is reachable, bounded and reversible. In addition, the Petri net model builds the theoretical basis for the network control of the whole packaging line.

Ltd. At present, the technology and service can be also provided by Henan Fang Yuan Group. However, it is still applied in submerged arc furnace for producing high-carbon ferromanganese. The application of condensing lining in large-scale submerged arc furnace for producing ferronickel is still in the exploration stage. In this article, based on communicating with technician worked in ferronickel corporation, enterprises devoted effort to the research & development of condensing lining and summarizing nickel iron production experiences, referenced advices for structure and design of condensing lining were put forward.

Background: Alvocidib is a potent cyclin-dependent kinase 9 inhibitor, which has previously been shown to downregulate the B-cell lymphoma-2 family member MCL-1 (myeloid cell leukemia-1) which, in turn, sensitizes tumor cells to apoptotic signals. Alvocidib has been studied globally in relapsed/refractory (R/R) AML in combination with cytarabine/mitoxantrone, as well as in newly diagnosed AML in combination with cytarabine/daunorubicin, and has demonstrated acceptable clinical activity. However, safety and tolerability in Japanese AML pts are unknown. Aims: To evaluate the safety and tolerability of alvocidib administered as ACM or A+7+3. Methods: This multicenter, open-label, uncontrolled phase 1 study (NCT03563560) included 2 regimens: ACM (in pts R/R to cytarabine/anthracycline based intensive chemotherapy and without a cumulative total exposure of anthracycline [daunorubicin-equivalent dose] exceeding 360 mg/m2 at entry) and A+7+3 (in newly diagnosed, treatment naive pts). Key eligibility criteria for both regimens were age 20-64 years, Eastern Cooperative Oncology Group performance status ≤ 2, a left ventricular ejection fraction ≥ 50%, and no major organ dysfunction. The ACM regimen (cohorts R1 and R2) comprised a fixed dose hybrid regimen of alvocidib (30 mg/m2/day as a 30-minute intravenous [IV] bolus followed by 60 mg/m2/day over 4 hours as a continuous IV infusion on days 1-3), and escalating doses of cytarabine (300 or 667 mg/m2/day by continuous IV infusion on days 6-8) and mitoxantrone (14 or 40 mg/m2/day IV infusion on day 9 or 10 starting 12 hours after completing cytarabine) in a standard 3+3 design. In the A+7+3 regimen (cohort F1) the dose of each component was fixed according to the recommended dose from the ongoing phase 1 study in the US (NCT03298984). Treatment consisted of the alvocidib hybrid regimen on days 1-3, cytarabine 100 mg/m2/day by continuous IV infusion on days 5-11, and daunorubicin 60 mg/m2/day IV on days 5-7. The primary study objective was to determine the safety and tolerability of alvocidib in combination with each type of chemotherapy, via the assessment of dose-limiting toxicities (DLTs) and treatment-emergent adverse events (TEAEs; graded according to the National Cancer Institute Common Terminology Criteria for Adverse Events v4.03). Secondary objectives were to evaluate the pharmacokinetics (including maximum plasma concentration [Cmax] and area under the concentration-time curve up to the last measurable concentration [AUC0-last]) and clinical activities of these regimens. Results: Between April 2018 to October 2019, 10 pts were enrolled, with a median (range) age of 45.0 (25-64) years. Of these, 6 pts (2 with relapsed disease and 4 with refractory disease) received ACM and 4 pts received A+7+3 (Table). Alvocidib was tolerated and DLTs were not observed in either of the two regimens. The most common TEAEs were hematologic events. The most common Grade ≥ 3 non-hematologic TEAEs occurring in ≥ 2 pts in the ACM regimen were diarrhea (50%), hepatic function abnormal (33%), sepsis (33%), cardiac failure (33%) and hypokalemia (33%). The most common Grade ≥ 3 non-hematologic TEAEs noted in ≥ 2 pts in the A+7+3 regimen were diarrhea (100%), tumor lysis syndrome (50%), and vomiting (50%). In the ACM regimen in R/R AML, all 6 pts were evaluable, of whom 5 (83%) achieved an objective response, including 4 complete remissions (CRs) and 1 morphologic leukemia-free state. The mean duration of CR was 13.6 months. In the A+7+3 regimen in newly diagnosed AML, 3 out of 4 pts were evaluable. All 3 evaluable pts achieved CR. One pt did not complete the A+7+3 regimen due to tumor lysis syndrome and was discontinued from the study prior to disease assessment. Overall, the mean Cmax of alvocidib in plasma was 1241 ng/mL on day 1 and 1217 ng/mL on day 3. The mean AUC0-last was 6555 h*ng/mL on day 1 and 7998 h*ng/mL on day 3. WSummary: Alvocidib in combination with cytarabine 667 mg/m2/day and mitoxantrone 40 mg/m2/day IV infusion in Japanese pts with R/R AML or cytarabine/daunorubicin (7+3) induction in Japanese pts with newly diagnosed AML showed an acceptable safety profile similar to previously investigated regimens in other studies. These data suggest that future studies of alvocidib in hematological malignancies are warranted, and planned directions for alvocidib development include phase 1/2 efficacy and safety studies in combination with other agents. This paper aims to investigate the usefulness of LEGO and other materials as prototyping tools in the co-creation process involving multiple stakeholders. The results demonstrate that using LEGO and other materials as prototypes in the co-creation process helps designers and stakeholders to explore, evaluate and modify ideas three-dimensionally to validate design solutions, idea generation and evaluation. This study believes that LEGO and other materials are efficient co-design prototyping tools to ensure its main purpose for team and strategy building in enhancing participant's ability to deliver more contribution during group discussions. The design of the public transport as to meet working women’s travelling needs is still lacking in Malaysian urban scenarios. Having that in mind, the increasing crime rate among these vulnerable group in urban area had further increase their level of fear while commuting to work. This is especially true for many working women who have no choice but to travel using public transport to their work place on a routine basis. To further investigate the factors of women travel pattern that affect the fear level among these vulnerable group of commuters, time and age were considered in this study.

Based on a new theoretical framework—the Social Relations Lens Model—this study examined the influence of personality on real–life attraction at zero acquaintance. A group of psychology freshmen ( N = 73) was investigated upon encountering one another for the first time. Personality traits, attraction ratings and metaperceptions were assessed using a large round–robin design (2628 dyads). In line with our model, personality differentially predicted who was a liker and who expected to be liked (perceiver effects), who was popular and who was seen as a liker (target effects), as well as who liked whom and who expected to be liked by whom (relationship effects). Moreover, the influence of personality on attraction was mediated by observable physical, nonverbal and audible cues. Results allowed a closer look at first sight and underline the importance of combining componential and process approaches in understanding the interplay of personality and social phenomena. Objectives/Hypothesis. Comparing long term stability of the Middle Ear Transducers (MET) of the 1st generation T1 (Otologics LLC) with the current generation T2 (Cochlear Ltd.) in all our clinical cases with standard incus coupling. Study Design. Retrospective chart review. Methods. 52 ears implanted with a MET device between 2008 and 2016 were analyzed retrospectively. All patients suffered from sensorineural hearing loss and the actuator was coupled to the body of the incus (standard coupling). 23 ears were implanted with the transducer T1 (Otologics LLC) between 2008 and 2011 and 29 ears were implanted with the current transducer T2 since 2011 (Otologics LLC/Cochlear Ltd.). Latest available in situ and bone conduction (BC) thresholds were exploited for a follow-up period of up to 7 years after first fitting. Long term stability of coupling and actuator performance was evaluated by tracking differences between in situ and BC thresholds. Results. In the T1 group, 9 out of 23 implants were still used by the patients at their last follow-up visit (average observation time 3.7 yrs.; min 1.0 yrs., max 7.4 yrs.). In 9 patients a technical failure identified by a decrease of in situ threshold of more than 15 dB compared to BC thresholds [Δ (in situ – BC)] lead to non-usage of the implant and 7 explantations. Five other explantations occurred due to medical reasons such as BC threshold decrease, infection, or insufficient speech intelligibility with the device. In the T2 group, 23 out of 29 implants were still used at the most current follow-up visit (average observation time 3.3 yrs.; min 1.0 yrs., max 4.8 yrs.). No technical failures were observed up to more than 4 years after implantation. Five T2 patients discontinued using the device due to insufficient benefit; two of these patients were explanted. One patient had to be explanted before the activation of the device due to disorders of wound healing. Nevertheless, a small but significant decrease of hearing loss corrected coupling efficiency [Δ (in situ – BC)] was seen in the T2 group. Conclusions. In contrast to the T1 transducers of the earlier generation of MET systems where technical failures occurred frequently, no technical failures were detected after 29 implantations with the current T2 transducers. However, a small but significant decline of transmission efficiency was observable even in the T2 implanted group.

Purpose Data envelopment analysis (DEA), a non-parametric technique is used to assess the efficiency of decision-making units which are producing identical set of outputs using identical set of inputs. The purpose of this paper is to find the technical efficiency (TE), pure technical efficiency and scale efficiency (SE) levels of Indian oil and gas sector companies and to provide benchmark targets to the inefficient companies in order to achieve efficiency level. Design/methodology/approach In the present study, a group of 22 oil and gas companies which are listed on the National Stock Exchange for which the data were available for the period 2013–2017 has been considered. DEA has been performed to compare the efficiency levels of all companies. To measure efficiency, three input variables, namely, combined materials consumed and manufacturing expenses, employee benefit expenses and capital investment and two output variables – operating revenues and profit after tax (PAT) have been considered. On the basis of performance for the financial year ending 2017, benchmark targets based on DEA–CCR (Charnes, Cooper and Rhodes) model have been provided to the inefficient companies that should be focused upon by them to attain the efficiency level. The performance of the companies for the past five years has been examined to check the fluctuations in the various efficiency scores of the companies considered in the study over the years. Findings From the results obtained, it is observed that 59 percent, i.e. 13 out of 22 companies are technically efficient. By considering DEA BCC (Banker, Charnes and Cooper) model, 16 companies are observed to be pure technically efficient. In terms of SE, there are 14 such companies. The inefficient units need to improve in terms of input and output variables and for this motive, specified targets are assigned to them. Some of these companies need to upgrade significantly and the managers must take the concern earnestly. The study has also thrown light on the performance of the companies over last five years which shows Oil India Ltd, Gujarat State Petronet Ltd, Petronet LNG Ltd, IGL Ltd, Mahanagar Gas, Chennai Petroleum Corporation Ltd and BPCL Ltd as consistently efficient companies. Research limitations/implications The present study has made an attempt to evaluate the efficiency of Indian oil and gas sector. The results of the study have significant inferences for the policy makers and managers of the companies operating in the sector. The results of the study provide benchmark target level to the companies of Oil and Gas sector which can help the managers of the relatively less efficient companies to focus on the ways to improve efficiency. The improvement in efficiency of a company would not only benefit the shareholders, but also the investors and other stakeholders of the company. Originality/value In the context of Indian economy, very limited number of studies have focused to measure the efficiency of oil and gas sector in the context of Indian economy. The present study aims to provide the latest insight to the efficiency of the companies especially operating in the Indian oil and gas sector. Further, as per our knowledge, this study is distinctive in terms of analyzing the efficiency of Indian oil and gas sector for a period of five years. The longitudinal study of the sector efficiency provides a bird eye view of the average efficiency level and changes in the efficiency levels of the companies over the years.

Background: Pelcitoclax (APG-1252), a novel dual inhibitor of Bcl-2/Bcl-xL, is active as monotherapy in patients with advanced solid tumors and well tolerated up to 240 mg twice weekly (NCT03387332). Preclinical data suggest that cells with Janus-associated kinase-2 (JAK2) mutations, including those associated with bone marrow fibrosis, are dependent on Bcl-2/Bcl-xL for survival and that addition of BH3 mimetics targeting Bcl-2/Bcl-xL induces apoptosis. Furthermore, in JAK2‒mutated cell models, apoptotic synergy is demonstrated when a JAK2 inhibitor and Bcl-2/Bcl-xL inhibitor are combined, as inhibition of Bcl-xL overcomes resistance to JAK2 inhibitors. Taken together, APG-1252 could overcome resistance to JAK2 inhibitors, and the combination could augment clinical benefit in patients with suboptimal responses to JAK2 inhibitor‒based therapy. Study Objectives: The primary objective of this open-label trial is to evaluate the safety and efficacy of APG-1252, as monotherapy and when combined with ruxolitinib, in adults with histologically or cytologically confirmed MF who require therapy and are ineligible for JAK2 inhibitors (and can receive single-agent APG-1252) or have had inadequate responses to ruxolitinib-based therapy (and can receive this treatment plus APG-1252). Secondary objectives include APG-1252 pharmacokinetics, time to response, and duration of response. Exploratory objectives include changes in cytogenetics and molecular mutations, bone marrow fibrosis, and cytokines on treatment. Study Design: The study is divided into Part 1 (APG-1252 monotherapy) and Part 2 (APG-1252 plus ruxolitinib). For Part 1, the key inclusion criterion is ineligibility for JAK2 inhibitors and for Part 2, inadequate responses to prior ruxolitinib-based therapy. A standard 3+3 dose-escalation design is being implemented to determine the maximum tolerated dose (MTD) of APG-1252 monotherapy in Part 1 and APG-1252 combined with ruxolitinib in Part 2. APG-1252 will initially be administered at 160 mg intravenously by 30-minute injection once weekly in a 28-day cycle. The dose can be escalated to a maximum of 240 mg or reduced to a minimum of 80 mg, depending on tolerability. Part 2 will begin once the MTD and recommended phase 2 dose (RP2D) of APG-1252 monotherapy have been determined. In Part 2, ruxolitinib will be administered orally twice daily per the package insert. After the MTD for APG-1252 monotherapy has been determined, no additional patients will be enrolled in Part 1; however, up to 15 to 30 additional patients can be enrolled in Part 2, to further evaluate the safety and anticancer activity of the combination at MTD or RP2D. Patients will continue treatment until disease progression or unacceptable toxicity. Clinical responses are being assessed every 12 weeks according to criteria from the International Working Group‒Myeloproliferative Neoplasms Research and Treatment and European LeukemiaNet panels, while optimal clinical benefit will be evaluated at 24 weeks. This research was aimed to describe the application of e-learning at the Department of Educational Technology, Faculty of Education Science, Jakarta State University. This research used case study research method with multiple data collection. Research findings revealed that (1) there are 20 courses conducted using e-learning by 7 lecturers; (2) blended learning was used; (3) e-learning itself developed through analysis, design and development phases; (4) analysis phase including students characteristics analysis, e-elarning environment analysis and content analysis; (5) design phase including online leaerning design (designing content, discussion for online discussion forum, assignment for online assignment, and test item for online evaluation) and face-to-face learning design including deep discussion, case study, and demonstration and practices; (6) student acceptance on blended was high; (7) there were several barriers in implementing e-learning due to lack of policy and ICT infrastructure support; and (8) supporting factors of implementation e-learning were the high commitment from lecturers and students, and the familiarity of students toward ICT. Based on research findings, Department of Educational Technology was recommended to have a minimum standard of development and implementation of e-learning and to enhance ICT infrastructure support, esp. access adequate internet connectivity.

Purpose. To compare the visual quality of patients with keratoconus who underwent penetrating keratoplasty (PKP) or deep anterior lamellar keratoplasty (DALK) with fluid dissection. Design. Cross-sectional, observational study. Methods. Twelve eyes that underwent PKP (PKP group) were compared to 24 eyes that underwent DALK (DALK group) after complete removal of sutures and stability of refraction. Visual, refractive, corneal topographic, corneal aberrometry, and ocular aberrometry parameters were compared for both groups. The χ2 and Mann–Whitney U tests were used for comparisons as appropriate. P<0.05 was considered statistically significant. Results. Uncorrected and best spectacle-corrected visual acuity (UCVA and BSCVA, resp.), mean refractive spherical equivalent and mean refractive cylinder (MRSE and MRC, resp.), root mean square of the 3 mm and 5 mm OPD Scan (NIDEK Co. Ltd., Gamagori, Japan), steep and flat meridians (SimK1 and SimK2, resp.), and the difference (corneal cylinder) were not statistically significantly different between groups (P>0.05, all comparisons). All aberrations, point spread functions (PSF), and the modulation transfer function (MTF) were not statistically different between groups (P>0.05). Conclusion. For our small study, the postoperative PKP and DALK with fluid dissection patient groups had vision/optical quality parameters that were not statistically different. This may indicate that DALK with fluid dissection can replace PKP for keratoconus without compromising vision quality.

Objectives Differences in peritoneal fluid handling in the acute setting can be expected if children are converted to pH-neutral dialysis solutions because conventional acidic solutions exert toxic effects on peritoneal mesothelial cells and microcirculation. Peritoneal fluid kinetics was therefore investigated with both types of solutions in a group of children. Design Peritoneal equilibration tests (PETs) were performed in 12 patients [mean age 70 months, mean time on peritoneal dialysis (PD) 18 months] using a pH-neutral PD fluid (Physioneal 3.86%; Baxter Ltd, Castlebar, Ireland) and dextran 70 as a volume marker. The results of these PETs were compared to those of a historic group of 12 children (mean age 75 months, mean time on PD 17 months). Setting Pediatric dialysis unit in a tertiary institute. Patients Stable pediatric PD patients. Main Outcome Measures Transcapillary ultrafiltration (TCUF) and marker clearance, dialysate-to-plasma (D/P) ratios for urea and creatinine, and Dt/D0 ratio for glucose. Results TCUF and lymphatic absorption were not different between the two groups. There was also no significant difference in small solute clearance measured by D/P ratio for urea and creatinine and Dt/D0 ratio for glucose. Conclusion Peritoneal fluid kinetics is not significantly altered if pH-neutral dialysis solutions are applied compared to acidic solutions. An altered TCUF, as is hypothetically possible using an acidic solution, was not established.

The pathological amyloid deposits of AD result in disruption of the balance between long-term potentiation (LTP) and long-term depression (LTD) of neuronal cells and synaptic plasticity. Brain stimulation in dementia research, especially with relatively safe tDCS, has been taken seriously recently. In theory, tDCS affects long-term synaptic plasticity through LTP and LTD, thereby improving cognitive ability. Recently, an increasing number of studies have been conducted to evaluate the efficacy of tDCS in AD and concluded a positive therapeutic effect. Currently, there are no studies of tDCS for AD in Taiwan. In this study, we investigate the effects of tDCS in AD.Method:Using a double-blind, randomized and sham- controlled trial design, Sixteen AD aged 55-90 years (8 active, mean age 73.88 and 8 sham, mean age 74.75) were included in the study. AD diagnostics is according to DSM-5 criteria. The CDR ratings of AD participants ranged from 0.5 to 2. All subjects completed ten consecutive daily sessions in which they received either an active or a sham tDCS over the left dorsal lateral prefrontal cortex (anodal) and a cathodal electrode on the right supraorbital area. In each session, we applied a current intensity of 2 mA and an electrode size of 35 cm2 for 30 min in the active group. All subjects received a series of neuropsychological tests, which included CDR, MMSE, CASI and WCST, before and after these treatment sessions on the first day and 4 weeks later. Chi- square test, Wilcoxon signed ranks test and Mann-Whitney U test were used to assess the differences in participant demographic characteristics and to compare the differences among groups.Results:The active group showed significant improvement in total correct item, Conceptual level Responses (reflecting insight into the correct sorting principles), Categories Completed (reflecting overall success), and Trials to complete first categories (reflecting initial conceptual ability) of WCST 4 weeks later after the final stimulation. There were no statistically significant differences between before and after the 10-session course for the sham group.Conclusion:tDCS stimulation improves cognitive operation and Conceptual Ability of AD.

BHP Petroleum Pty Ltd (BHPP) is subdivided, worldwide, into three 'regions'—Australia/Asia; Europe/Russia/Africa/Middle East; and Americas—each a self-contained, operating entity, each with its own system of management. The Australian/ Asian operations group within BHPP is implementing an integrated management system, of which the safety, occupational health and environmental elements will form crucial components. The management system has been designed to provide management control of safety, health and environment issues for all of the BHPP producing operations in Australasia. The Safety, Health and Environment Department of BHPP provides a region-wide function for Australasia, and the safety, health and environment portions of the management system, which the Department is helping to develop and implement, will document planned requirements at a number of levels: region-wide requirements for all departments (e.g. implementation of corporate policies, legislative compliance, emergency management, auditing, permit-to-work, health promotion, incident investigation, environmental approvals, etc), requirements specific to the Operations Department (e.g. safety/environmental auditing of plants and facilities, monitoring of oil spills/greenhouse gas emissions), and requirements specific to operational sites within the Operations Department. Responsibility for development, implementation, and maintenance of the management system, and compliance with its provisions, rests with line management—a logical extension of the accountability and responsibility for safety, health and environment matters that rests squarely on the shoulders of all line managers within BHPP. The Safety, Health and Environment Department provides expert advice, document control, training and auditing expertise, and offers a pool of experts available for participation in projects on a consultative basis. For contractor management, the BHPP system mandates intensive scrutiny of contractor safety, health and environmental performance and the systems that the contractor has in place to manage that performance, beginning at the stage of contract prequalification. Failure to meet the minimum standards set by BHPP will disqualify a contractor from contract tendering unless and until remedial action is taken. The ultimate aim of the BHPP system is a level of safety, health and environmental performance which leads the world—no injuries, no workplace health or hygiene problems and no adverse effect on the environment.

Introduction: Mosunetuzumab (M; RG7828) is a full-length, fully humanized immunoglobulin G1 (IgG1) bispecific antibody targeting both CD3 (on the surface of T cells) and CD20 (on the surface of B cells). It drives tumor B-cell killing independent of T-cell specificity and has been shown clinically to have single-agent activity in relapsed or refractory non-Hodgkin lymphoma, with durable complete responses (CR) at doses as low as 1.2mg (Budde et al. ASH 2018). We present biomarker data from the ongoing first-in-human multicenter Phase I/Ib study GO29781 (NCT02500407), highlighting acute pharmacodynamic effects that confirm mechanism of action (MOA), support Cycle 1 step-up dosing to mitigate cytokine-driven toxicities, and potentially predict response. Methods: Patients (pts) with biomarker data in Group A (0.05 to 2.8mg q3w dosing, n=30) and Group B (0.4/1/2.8 to 1/2/40.5mg Cycle 1 Day 1/8/15 step-up dosing, followed by q3w dosing) were included in the analyses. Peripheral biomarkers were evaluated using whole blood flow cytometry, plasma cytokine ELISA, and ex-vivo single-cell cytokine assay of cryopreserved peripheral blood mononuclear cells. Tumor biomarkers were assessed through CD20 immunohistochemistry and CD8/Ki67 immunofluorescence staining. Average CD20 receptor occupancy (RO%), which takes into account residual levels of rituximab that may compete with M for target binding, was used to characterize the exposure-pharmacodynamic response relationship (Li et al. ASH 2019). Results: Pharmacodynamic changes were observed in peripheral blood by 4 hours (hrs) after infusion of sub-efficacious doses of M in Group A. Transient reduction of circulating T cells was evident starting at the lowest doses administered of 0.05-0.2mg, with decreased cell numbers by 4 hrs after infusion and recovery by 72 hrs. At doses between 0.4-0.8mg, T-cell activation was observed in some pts, as evidenced by upregulation of CD69 on both CD8+ and CD4+ T cells and elevation of IFN-γ in plasma. At 1.2mg (minimal efficacious dose) to 2.8mg, all of these changes were detected in the majority of pts. M induced depletion of circulating B cells immediately after the first dose, which was sustained for the duration of treatment, even in pts refractory to prior anti-CD20 therapy. IL-6 levels peaked within 24 hrs of the Cycle 1 Day 1 dose and the kinetics of IL-6 increase were associated with the onset of cytokine release syndrome (CRS). Hence, step-up dosing at Cycle 1 was implemented in Group B to maintain the IL-6 peak at reasonably low levels and reduce the risk of toxicity. With step-up dosing, pharmacodynamic changes, including transient T-cell decrease, T-cell activation, and cytokine production, remained strongest within 4 hrs of the Cycle 1 Day 1 dose, despite higher subsequent and total cumulative doses. IL-6 peaks were similar in Group A and Group B and were in line with the low grades of CRS observed to date (Schuster et al. ASH 2019). In pts for whom the Cycle 1 Day 1 dose was fixed at 1mg, the acute pharmacodynamic changes were found to be associated with RO% at Day 1. In a subset of diffuse large B-cell lymphoma pts, functional assays showed that peripheral T cells taken at baseline from pts who achieved CR secreted multiple effector cytokines, such as IFN-γ, TNF-α, and Granzyme B, upon M stimulation ex vivo, whereas T cells from pts with progressive disease did not show this capacity. Analysis of tumor biopsies showed similar baseline levels of CD8+ tumor infiltrating T-cells (TILs) between responders and non-responders. M induced increases in CD8+ TILs, which were mostly Ki67-, as early as Cycle 1. The median on-treatment CD8+ TIL level was significantly higher in responders than non-responders. Conclusions: T-cell activation, cytokine elevation, and B-cell depletion in the periphery are among the earliest biomarkers of M activity and precede clinical responses. Acute pharmacodynamic changes confirm the MOA and inform dose/schedule selection. In particular, the 1mg Cycle 1 Day 1 dose, which was chosen to mitigate CRS and to dissociate toxicity and efficacy at higher subsequent doses, still induced potent pharmacodynamic changes. Finally, the data suggest a model whereby clinical efficacy may be critically dependent on activation and migration of functional T cells from the periphery into the tumor bed.
