Abstract:
Les tissus vasculaires sont d’une grande importance pour la physiologie et le développement de la plante. Le xylème et le phloème composent ce tissu. En assurant le transport des sèves brutes et élaborées, ils permettent ainsi de redistribuer minéraux et métabolites entre les différents organes de la plante. La régulation de la mise en place de ces deux tissus est hautement contrôlée tant d’un point de vue génétique que spatio-temporel. Parmi les gènes impliqués dans la mise en place des tissus vasculaires, le gène OCTOPUS (OPS) intervient très précocement au cours du développement dans la mise en place du patron vasculaire ainsi que différenciation du phloème(Bauby et al., 2007; Truernit et al., 2012). OPS appartient à une famille de gènes spécifiques des plantes supérieures. Cependant la fonction moléculaire de la protéine OPS reste inconnue. Une combinaison d’approches physiologique, biochimique, moléculaire, cytologique et génétique a permis de montrer que le gène OPS est un nouveau régulateur positif de la voie signalisation des brassinostéroïdes (BR). Les BR sont des hormones végétales impliquées dans de nombreux processus développementaux incluant l’élongation cellulaire ou encore l’organisation des tissus vasculaires. Bien que la découverte des BR date seulement de 1970 (Mitchell et al., 1970; Mitchell and Gregory,1972; Grove et al., 1979), l’intérêt commun que les scientifiques lui ont porté fait que sa voie de signalisation est aujourd’hui très détaillée. Parmi les composants de la voie des BR, la protéine kinase BIN2 en est le régulateur clef (Li et al., 2001) réprimant la voie par son action sur les facteurs de transcription BES1 et BZR1(Yin et al., 2002; Wang et al., 2002). OPS interagit physiquement avec la protéine BIN2 au niveau de la membrane plasmique, pouvant créer une inhibition par délocalisation de la protéine BIN2 de son site d’activité. A ce titre, les défauts de phloème observés chez le mutant opssont restaurés lorsque l’activité de BIN2 est abolie ou lorsque la voie est induite en dessous de BIN2. Ainsi nous montrons que la voie des BR est directement impliquée dans la différenciation du phloème. Plus généralement, une étude de la redondance fonctionnelle d’autres membres de la famille OPS semble indiquer qu’ils pourraient exercer une fonction similaire de régulateurs positifs de la voie des BR au sein d’autres tissus de la plante
Vascular tissues play an important role in plant physiology and development. Vascular tissues consist on xylem and phloem that ensure sap transport and permit to redistribute mineral and metabolites between different organs. Regulation of vascular tissues establishment is highly controlled in space and time. Among genes involved in vascular tissues formation, OCTOPUS (OPS) gene operates early during development to control vascular patterning and to induce phloem differentiation(Bauby et al., 2007; Truernit et al., 2012). OPS belongs to a multigenic family conserved in high plant. However, molecular function of OPS protein remains unclear. A combination of physiological, biochemical, molecular, cytological and genetic approaches allowed to show that OPS gene is a new positive regulator of brassinosteroid (BR) signaling pathway. BR are phytohormones involved in many developmental processes such as cell elongation or vascular tissues organization. Although BR were discovered in 1970 (Mitchell et al., 1970; Mitchell and Gregory, 1972; Grove et al., 1979), interest of researchers for this hormone permitted its detailed description. Among the component of BR signaling, the BIN2 kinase is the key regulator (Li et al., 2001) which represses the pathway through its action on BES1 and BZR1 transcription factors(Yin et al., 2002; Wang et al., 2002). OPS interacts physically with BIN2 at the plasma membrane which could create an inhibition by delocalization of the BIN2 protein from its activity place. As such, opsphloem defects are restored when the activity of BIN2 is inhibited or when the pathway is induced downstream of BIN2. Thus, we show that BR pathway is directly involved in phloem differentiation. More generally, a study of the functional redundancy of other OPS family members suggests that they could have a similar function as positive regulators of BR pathway in other plant tissues

Abstract:
The purpose of bin packing is the effective apportionment of smaller elements in the bigger ones. The bin packing is being analysed in managment, computers, mathematics and researches of operations. The quality of the heuristic structural algorithm and time have been analysed in the work (the percent of the used material and the quality of the received waste). The results of the researches show that non-sorted bin packing algorithms cut better than bin packing sorted algorithms but the time of cutting grows longer when the number of finished products grow longer.

Abstract:
When a pile of scrap at Stena recycling has reached the maximum height, the loader operators need to drive around the facility to check whether or not the pile has to be emptied. This causes a lot of unnecessary driving, resolving in wear to the loaders and loss of time, but mostly the unnecessary use of diesel. In this paper different sensors is evaluated and tested against each other to find a system that can measure the volume of the pile. The sensors used is IFM O3D303 and the Kinect V1 sensor. These sensors are then used with two different volume calculation software, CloudCompare and 3DF Zephyr. The goal of the project is to get a system that can get a volume with an accuracy within 2.5 % of the actual volume. If a volume can be calculated from the scrap piles, then it is also possible to know when the pile should be cleared. Since the facility is a recycling plant, there is a lot of metal dust that is gathering in the indoor part of the facility. The sensors being used, should be able to work in this environment. To do that the sensor needs to have an IP certification higher than IP 55. The distance of measuring the pile can be more than 10 m, so the sensors should also be able to handle this requirement. From the sensors a point cloud and a mesh were captured, two data formats that contain information of where every point on the object is located in a three-dimensional space. Every system needs to be calibrated to suit the specific application. The results show that, even with a basic sensor such as the Kinect V1, an accurate volume of a scrap pile could, with calibration, be calculated. However, the cheap sensor does have limits, unlike the IFM which can cope with all the requirements. This report shows how the two sensors compare to each other and how the different software, combined with the sensors, can be used to calculate a volume of a pile of scrap.

Detta kandidatarbete har handlat om att ta fram en ergonomisk papperskorg som löser problemet med att en städare tvingas böja sig ned för att byta påse. Uppgiften kommer ur de önskemål som ett företag verksamt inom städbranschen har uttryckt. Efter att ha varit i kontakt med detta företag för att diskutera problemets bakgrund och vilka förväntningar som finns så utfördes en inledande marknadsundersökning vars syfte var att få en överblick på vilka produkter som fanns, vilken funktionalitet dessa hade och till vilket pris. Den lösningen som slutligen valdes var en höj och sänkbar papperskorg, där korgen installeras på två samverkande saxliftar som är parallellt monterade. En fotpedalen utsätts för nedåtriktad kraft vilket med hjälp av en vajerlösning får mekanismen att resa sig. Ett hölje är format för att täcka hela mekanismen och detta hölje har funktionaliteten av ett teleskoprör. Höljet består av tre segment som följer upp med korgen då den lyfts. Den fristående papperskorgen står på den platta som är en del av det översta segmentet. Styrning i form av plastglidlager är monterade på sidan av dessa segment för att stabilisera lyftet. Totalvikten för hela konstruktionen inklusive papperskorgen är 4,4 kg och de material som valdes var plast samt stål. Höljet och plattan tillverks av polypropen (PP), styrningar tillverkas av polyetylentereftalat (PET). CAD modeller användes för att illustrera underlaget för produkten, modellen visar och klargör mekaniskmens funktion. Målet för arbetet var att ta fram ett underlag för en produkt som är redo för produktion. Produkten ska underlätta arbetet för användaren, erbjudas till en rimlig kostnad samt kunna passa in i de flesta miljöer.
The purpose of this project has been to design an ergonomic litter bin that solves the problem of a cleaner always having to bend down when emptying the bag. This assignment was presented to us by a company in the cleaning industry. After contacting the company to discuss the background of the problem, as well as the expectations on this projects outcome, a market research was initiated to get an overview of what the market offers in terms of similar products, the functionality of these products and their price range. The chosen solution was a litter bin that has the ability to move vertically. A conventional plastic bin was installed on two scissor lifts mounted parallel to each other. By transferring a downward facing force on a foot pedal through a wire, the mechanism is lifted. The outer parts of the product are shaped in a way so that it covers the entire mechanism in a way similar in principle to a telescope. This housing is composed of three segments to have the ability to move alongside the basket when lifted. The basket stands on a platform which is a part of the innermost segment. Plastic rails are mounted on the side of the segment to stabilize the lift. Total weight of the structure including the basket is 4,4 kg, plastic and steel are the chosen materials. The housing is made out of polypropylene (PP) and the rails are made out of polyethylene terephthalate (PET). Our ambition was to develop a basis for a product that is ready for production. The product should ease the workload of the end user, it should be available to a reasonable cost and be able to fit into most environments.

Abstract:
In the past few years, there has been a strong and growing interest in evaluating the expected behavior of what we call combinatorial bin packing problems. A combinatorial bin packing problem consists of a number of items of various sizes and value ratios (value per unit of size) along with a collection of bins of fixed capacity into which the items are to be packed. The packing must be done in such a way that the sum of the sizes of the items into a given bin does not exceed the capacity of that bin. Moreover, an item must either be packed into a bin in its entirety or not at all: this "all or nothing" requirement is why these problems are characterized as being combinatorial. The objective of the packing is to optimize a given criterion Junction. Here optimize means either maximize or minimize, depending on the problem. We study two problems that fit into this framework: the Knapsack Problem and the Minimum Sum of Squares Problem. Both of these problems are known to be in the class of NP-hard problems and there is ample reason to suspect that these problems do not admit of efficient exact solution. We obtain results concerning the performance of heuristics under the assumption that the inputs are random samples from some distribution. For the Knapsack Problem, we develop four heuristics, two of which are on-line and two off-line. All four heuristics are shown to be asymptotically optimal in expectation when the item sizes and value ratios are assumed to be independent and uniform. One heuristic is shown to be asymptotically optimal in expectation when the item sizes are uniformly distributed and the value ratios are exponentially distributed. The amount of time required by these heuristics is no more than proportional to the amount of time required to sort the items in order of nonincreasing value ratios. For the Minimum Sum of Squares Problem, we develop two heuristics, both of which are off-line. Both of these heuristics are shown to be asymptotically optimal in expectation when the sizes of the items input are assumed uniformly distributed.

Abstract:
In this thesis we study online algorithms for dynamic bin packing. An online algorithm is presented with input throughout time and must make irrevocable decisions without knowledge of future input. The classical bin packing problem is a combinatorial optimization problem in which a set of items must be packed into a minimum number of uniform-sized bins without exceeding their capacities. The problem has been studied since the early 1970s and many variants continue to attract researchers’ attention today. The dynamic version of the bin packing problem was introduced by Coffman, Garey and Johnson in 1983. The problem is a generalization of the bin packing problem in which items may arrive and depart dynamically. In this setting, an online algorithm for bin packing is presented with one item at a time, without knowledge of its departure time, nor arrival and departure times of future items, and must decide in which bin the item should be packed. Migration of items between bins is not allowed, however rearrangement of items within a bin is permitted. The objective of problem is to minimize the maximum number of bins used over all time. In multi-dimensional generalizations of the problem, multi-dimensional items must be packed without overlap in multi-dimensional bins of uniform size in each dimension. In this work, we study the setting where items are oriented and cannot be rotated. We first consider online one-dimensional dynamic bin packing and present a lower bound of 8/3 ∼ 2.666 on the achievable competitive ratio of any deterministic online algorithm, improving the best known 2.5-lower bound. Since the introduction of the problem by Coffman, Garey and Johnson, the progress on the problem has focused on improving the original lower bound of 2.388 to 2.428, and to the best known 2.5-lower bound. Our improvement from 2.5 to 8/3 ∼ 2.666 makes a big step forward in closing the gap between the lower bound and the upper bound, which currently stands at 2.788. Secondly we study the online two- and three-dimensional dynamic bin packing problem by designing and analyzing algorithms for special types of input. Bar-Noy et al. initiated the study of the one-dimensional unit fraction bin packing problem, a restricted version where all sizes of items are of the form 1/k, for some integer k > 0. Another related problem is for power fraction items, where sizes are of the form 1/2k, for some integer k ≥ 0. We initiate the study of online multi-dimensional dynamic bin packing of unit fraction items and power fraction items, where items have lengths unit fraction and power fraction in each dimension, respectively. While algorithms for general input are suitable for unit fraction and power fraction items, their worst-case performance guarantees are the same for special types of input. For unit fraction and power fraction items, we design and analyze online algorithms that achieve better worst-case performance guarantees compared to their classical counterparts. Our algorithms give careful consideration to unit and power fraction items, which allows us to reduce the competitive ratios for these types of inputs. Lastly we focus on obtaining lower bounds on the performance of the family of Any- Fit algorithms (Any-Fit, Best-Fit, First-Fit, Worst-Fit) for online multi-dimensional dynamic bin packing. Any-Fit algorithms are classical online algorithms initially studied for the one-dimensional version of the bin packing problem. The common rule that the algorithms use is to never pack a new item to a new bin if the item can be packed in any of the existing bins. While the family of Any-Fit algorithms is always O(1)-competitive for one-dimensional dynamic bin packing, we show that this is no longer the case for multi-dimensional dynamic bin packing when using Best-Fit and Worst-Fit, even if the input consists of power fraction items or unit fraction items. For these restricted inputs, we prove that Best-Fit and Worst-Fit have unbounded competitive ratios, while for First-Fit we provide lower bounds that are higher than the lower bounds for any online algorithm. Furthermore, for general input we show that all classical Any-Fit algorithms are not competitive for online multi-dimensional dynamic bin packing.

Besides providing a biographical sketch of Bint al-Shati', this thesis analyzes her intellectual growth and provides the necessary information about her works relating to Qur'anic studies. The study also surveys al-Khuli's method of Qur'anic exegesis and Bint al-Shati' thinspace's attempts to apply it to her own works. Our analysis focuses on her works on Qur'anic studies such as al-Tafsir al-Bayani li al-Qur'an al-Karim, Maqal fi al-Insan and al-I'jaz al-Bayani li al-Qur'an wa Masa'il Ibn al-Azraq. The distinguishing features of her philological and thermatic approach are discussed and the main results obtained by the practical application of this method are shown to bring refreshing insights into the meaning of the Qur'an.

Abstract:
The purpose of this experiment was to empirically study and document the double bind. Twenty outpatient clinical and twenty matched non-clinical adolescents between the ages of 13 and 19 were studied. Three research questions were investigated. The first was the nature of the bodily responses in terms of cognitive complexity and emotional stress to inconsistent and consistent messages in the context of a close relationship. Cognitive complexity was measured by Reaction-time, and emotional stress by heart-rate and frequency of non-verbal movement (arm, leg and foot). The consistent and inconsistent messages were presented by the instrument developed by Reilly (in press) consisting of photographs and accompanying taped messages in a mother/child context. The second question studied was the nature of responses (double bound/non-double bound) made by the two groups to inconsistent and consistent messages. Finally, the nature of the attribution of meaning to messages, and the verbal/non-verbal focus used to do so by both groups, was studied. It was found that the inconsistent messages were more cognitively complex for both groups than consistent ones, and that inconsistent positive messages were the most complex of all. Perhaps this finding about inconsistent positive messages may be explained by their less frequent documentation in the double bind literature, and their not being part of the accepted social norm. Inconsistent messages were also found to be more stressful than consistent messages to both groups. This confirms the speculations in the theory of such communication being associated with subjective distress. In addition, the clinical group was found to make more double bound responses than the non-clinical group, especially in response to inconsistent messages. This is very supportive of the hypotheses of the double bind theory. Finally, it was found that meaning was correctly and unhesitatingly attributed to consistent messages by both groups. However there was less clarity and consensus in the attribution of meaning to the inconsistent messages by both groups. The non-clinical group tended to focus on non-verbal information in interpreting the messages, which is developmentally age-appropriate. The clinical group tended to focus at times on the non-verbal information, and at other times on the verbal or literal information, indicating a transitional stage between a developmentally younger stage and the age-appropriate stage.

Social media affects our daily lives. It is one of the first sources for finding breaking news. In particular, Twitter is one of the popular social media platforms, with around 330 million monthly users. From local events such as Fake Patty's Day to across the world happenings - Twitter gets there first. During a disaster, tweets can be used to post warnings, status of available medical and food supply, emergency personnel, and updates. Users were practically tweeting about the Hurricane Sandy, despite lack of network during the storm. Analysis of these tweets can help monitor the disaster, plan and manage the crisis, and aid in research. In this research, we use the publicly available tweets posted during several disasters and identify the relevant tweets. As the languages in the datasets are different, Bing translation API has been used to detect and translate the tweets. The translations are then, used as training datasets for supervised machine learning algorithms. Supervised learning is the process of learning from a labeled training dataset. This learned classifier can then be used to predict the correct output for any valid input. When trained to more observations, the algorithm improves its predictive performance.

This thesis is devoted to the study of Bint al-Shat&dotbelow;i''s method of interpreting the Qur'an. The problems of consistency in terms of her hermeneutical theory and of the application of this method represent the focus of the study. It furthermore discusses her attitude towards tendentious and i'jaz-misoriented interpretations of the Qur'an, of which both classical and modern exegetes are guilty in her eyes. Secondly, it studies how she applies her method in specific situations. The cross-referential method, and the concept of irtibat&dotbelow; (interrelation between verses) are two major points to be analyzed. Finally, it also discusses her theory concerning the asbab al-nuzul (occasions of revelation), and its application. Using phenomenological, comparative and analytical methods of analysis, and by means of primary and secondary sources, the thesis concludes that Bint al-Shat&dotbelow;i' in many cases is not consistent in applying the method she established.

Twenty years after the collapse of the Soviet Union, the Russian Federation has struggled to construct a new foreign policy paradigm in a world that is no longer bipolar. Instead of the Cold War era arms stockpiles, Moscow has signed multiple Strategic Arms Reduction Treaties (START), and instead of physically taking over neighboring territories, increasing diplomacy and economic incentives have become Moscow’s primary tool to garner and maintain influence in its near abroad. Soft power initiatives, that can all roughly fit into a foreign aid model, from the Russian Federation to the near abroad come in different forms: oil subsidies, aid in kind, and direct financial investment. The Russian Federation has used all three of these strategies in Ukraine after the fall of the Soviet Union in an attempt to keep the country in its sphere of influence. The use of foreign aid, be it oil subsides, aid in kind, or direct financial aid, all work towards the same goal of promoting Russian policies and interests in the near abroad. Ukraine, with its unique political, cultural and geographical importance, demonstrates the new struggle between Russia and the West. The struggle’s main actors are the Russian Federation against the eastern half of NATO and the EU, and the conclusion is unclear. Foreign aid and soft power are now playing a critical role in the outcome.

A significant percentage of breast and prostate tumours have a missing or aberrantly expressed Bin1 protein. However, preliminary work in skeletal muscle has shown that Bin1 may also be involved in regulating differentiation. Based on the variable roles attributed to Bin1, we proposed a hypothesis in which distinct domains of Bin1 regulate different functions of the protein. This study examines one particular domain of Bin1, the SH3 domain, through a dominant negative approach in a mouse model. Overexpression of the SH3 domain using a broadly expressed promoter resulted in a skeletal muscle phenotype that included large myofibre diameter in vivo and an inability to complete the differentiation process in vitro. Importantly, these deficits were not accompanied by disruptions in apoptosis, increased cell proliferation or tumorigenesis. These results suggest that Bin1 has distinct functions that are mediated by the modular SH3 domain and separate from its role in cell cycle regulation.

In this study, we consider two bi-objective bin packing problems that assign a number of weighted items to bins having identical capacities. Firstly, we aim to minimize total deviation over bin capacity and minimize number of bins. We show that these two objectives are conflicting. Secondly, we study the problem of minimizing maximum overdeviation and minimizing the number of bins. We show the similarities of these two problems to parallel machine scheduling problems and benefit from the results while developing our solution approaches. For both problems, we propose exact procedures that generate efficient solutions relative to two objectives. To increase the efficiency of the solutions, we propose some lower and upper bounding procedures. The results of our experiments show that total overdeviation problem is easier to solve compared to maximum overdeviation problem and the bin capacity, the weight of items and the number of items are important factors that effect the solution time and quality. Our procedures can solve the problems with up to 100 items in reasonable solution times.

O bin-picking começou a ter uma maior notoriedade principalmente a partir dos anos 80. As indústrias e laboratórios fizeram tentativas para desenvolver sistemas bin-picking eficazes, mas sem sucesso, principalmente devido aos recursos computacionais e tecnológicos da época. Hoje em dia, já existem alguns sistemas implementados, mas na maioria dos casos as peças estão semi-orientadas. São poucos os sistemas capazes de apanhar peças que estejam orientadas em qualquer posição, não havendo solução ainda para problemas mais complexos, como é o caso de molas e outras peças com geometrias muito irregulares. Este trabalho tem como objectivo principal desenvolver um sistema de binpicking capaz de apanhar objectos toroidais de diferentes dimensões, dentro de uma gama e empilhados de forma aleatória. O sistema deve conseguir através de dados sensoriais, determinar a localização e orientação dos objectos num procedimento dinâmico e de forma que sejam apanhados um a um. Neste caso em concreto, os objectos toroidais escolhidos para fazer os testes em laboratórios são pequenas réplicas de pneus. Depois de identificados os objectos potenciais, o manipulador deverá, autonomamente, fazer o reposicionamento da garra e sensores associados, para localizar e validar o objecto candidato, recorrendo à aproximação ao objecto ou à mudança do ângulo de visão se necessário. O sistema de visão baseia-se numa configuração de câmara monocular móvel juntamente com a iluminação localizada. A imagem é captada continuamente e é feito o tratamento com Sherlock. Por outro lado, como a câmara não permite obter informação sobre a profundidade, é usado um sensor de distância que complementa a informação da câmara. O programa de controlo recebe informação dos dispositivos de percepção e, com base nessa informação, toma as decisões, fazendo depois o controlo do manipulador e a actuação sobre a garra. O sistema mostrou-se capaz de identificar a localização e orientação dos pneus na pilha e de resolver problemas de oclusão e sobreposição dos mesmos. Além disso, o sistema permite posicionar o manipulador de forma a apanhar os pneus nas mais diversas situações, com a ajuda da garra desenvolvida para a aplicação em causa.
The bin-picking problem began to have great notoriety mainly from the 80’s.Companies and laboratories have made attempts to develop effective binpicking systems, but without success mainly due to the computing resources and technology of the time. Currently, there are some systems already implemented, but in most cases the pieces are semi-radom. There are few systems able to pick up pieces that are randomly oriented, still locking a solution for more complex items, such as springs and other parts with very irregular geometry. The main objective of this work is to develop a bin-picking system able to take toroidal parts of different sizes within a range and stacked randomly. The system uses sensories data to determine the localization and orientation of parts in a dynamic procedure, and catches them one by one. In this particular case, the toroidal objects chosen for the tests in labs are small replicas of tires. After having identified the potential objects, the robot must autonomously make the repositioning of the grip and associated sensors to locate and validate the candidate object using the approximation or to change the viewing angle if necessary. The vision system is based on a monocular camera mobile configuration with local illumination. The image is captured continuously and is analyzed with Sherlock software. However, as the camera doesn’t give information about the depth, a distance sensor is used to complement the information from the camera. The control program receives information from the sensing devices, and based on this information it makes decisions and then making the control of the robot and the activation of the grip. The system was able to identify the location and orientation of the tires on the stack and resolve problems of occlusion and overlap them. It is still able to position the robot according the orientation of the objects, as well as, pick them using the developed grip.

Detta har vi besvarat med följande frågeställning: Vilka relationer utifrån den didaktiska triangeln framkommer då pedagoger resonerar kring undervisning med bin som utgångspunkt? För att kunna formulera vår undersökningsfråga har vi genomfört systematiska sökningar i olika databaser. De källor vi ansåg var av relevans för vår frågeställning har därefter studerats, analyserats och sammanställts i förhållande till vårt syfte. I studien intervjuas fyra pedagoger som alla använder bin och bisamhället som en del i sin undervisning. Resultatet av intervjuerna analyseras sedan med hjälp av den didaktiska triangeln med fokus på relationerna lärare-elev-innehåll. Under analysprocessen utkristalliseras tre beskrivande kategorier för lärsituationer där bin utgör utgångspunkten för undervisningen: Natur, Engagemang och Ämnesintegrering. Utifrån kategorierna gjordes sedan en didaktisk analys av dessa kategorier. Intervjuerna som används i studien är av en semistrukturerad karaktär. Resultatet presenteras utifrån tre frågeställningar: Hur uttrycks framställning, Hur uttrycks erfarenhet? samt Hur uttrycks interaktion? Resultatet visar att naturen som undervisningsmiljö är en viktig del i framställningen när det kommer till undervisning med bin. Genom ett innehåll med bin kan eleverna erfara en medvetenhet om miljöfrågor och hållbar utveckling. Slutligen visar resultaten att lärarna genom interaktion med eleverna, i ett innehåll med bin kan utgå från elevernas behov av lärande och ge det en pedagogisk kontext.

Les travaux présentés dans ce mémoire traitent des propriétés optiques non-linéaires des cristaux photoréfractifs d'oxyde de bismuth et de silicium (Bi12 Sio20)) et d'oxyde de bismuth et de germanium(Bi12 Geo20)) ainsi que de la réalisation d'une expérience d'interconnexion dynamique. Nous nous interessons en premier lieu, a la diffraction d'une onde plane par un réseau épais photo induit dans des milieux présentant simultanément de l'activité optique et de la birefringence linéaire tels que les cristaux de Bi12 Sio20. Grâce a cette étude nous déterminons les caractéristiques (amplitudes phases et polarisations) des ondes transmises et diffractées. Nous exposons ensuite une méthode entièrement optique de caractérisation de ces matériaux, ce qui nous a permis pour différents échantillons, de mesurer tous les paramètres physiques dont dépend l'effet photoréfractif: - la constante de temps de relaxation diélectrique - la longueur de diffusion des électrons dans la bande de conduction - la densité de centres accepteurs d'électrons. La mise en évidence de variations importances de ces paramètres avec les conditions de cristallogenèse ouvre la voie a une optimisation possible de ces matériaux. Enfin, dans une dernière partie, une application a la déflexion optique est présentée. Le faisceau a défléchir est incident sur un réseau dynamique induit par la figure d'interférence de deux ondes planes dans un cristal photoréfractif épais. Les modifications simultanées, du pas du réseau d'une part, et de l'orientation de ses strates d'autre part, permettent d'adresser le faisceau tout en conservant une efficacité de diffraction maximale

Understanding the molecular biology of cells is a central field of research that is essential to all of the biological sciences. Messenger RNAs are the central link between DNA information contained in the nucleus and proteins that are synthesized in the cell cytoplasm. These mRNAs are subject to a substantial amount of modification and control, which are handled or under the supervision of RNA binding proteins. RNA binding proteins are trans-acting factors that interact with cis-acting elements present in the RNA sequence. They play major roles in mRNA processing including alternative splicing, translation, trafficking, localization and non-sense mediated decay. The STAR (signal tranduction activator of RNA metabolism) protein family are RNA binding proteins that can play multiple roles in RNA processing. This family of protein includes among others Quaking (QKI), SRC associated in mitosis (SAM68), SAM68-like mammalian protein 1 (SLM-1) and 2 (SLM-2), splicing factor 1 (SF1), germ-line development (GLD-1). All these family members contain a STAR/GSG domain that embeds a hnRNP K homology domain responsible for conferring the RNA binding properties. Because of the lack of genetic data providing insight about their physiological RNA targets, the function of these RNA binding proteins has been difficult to elucidate. Only a few mRNA targets are known across the whole family of STAR proteins. The hypothesis is that STAR proteins bind to a subset of determined mRNAs, and that identifying these RNAs and understanding how STAR proteins act on them would provide important evidence toward understanding the function of this familly of RNA binding proteins. Using systematic evolution of ligands by exponential enrichment (SELEX), we were able to define the QKI response element (QRE) as an hexanucleotide sequence (ACUAAY) with an additional half-site (UAAY). Bien comprendre la biologie moléculaire de la cellule est un champ de recherche central et essentiel à toutes les sciences biologiques. L'ARN messager est le lien central entre l'information de l'ADN contenu dans le noyau cellulaire et les protéines traduits dans le cytoplasme des cellules. Ces ARN messagers sont sujets à une quantité importante de modifications et de contrôle par des protéines liant l'ARN. Ces protéines jouent plusieurs rôles majeurs dans la maturation des ARN messagers incluant l'épissage alternatif, la traduction, le transport, la localisation ainsi que la dégénérescence d'ARN non-sens. La famille de protéine STAR « Signal Transduction and Activatior of RNA metabolism » sont des protéines liant l'ARN capable de jouer plusieurs rôles dans la maturation des ARNs. This research project explores the rural-urban bind in four novels by Thomas Hardy, Far from the Madding Crowd (1874), The Mayor of Casterbridge (1886), Tess of the d'Urbervilles (1891), and Jude the Obscure (1895). In each of these novels Hardy contrives to create an imagined nostalgia for a rural community losing its traditions, memory, and history due to social change brought about by the urban elements of modernisation, migration, and industrialisation. The purpose of this thesis is to expose the methods that Hardy employs to construct and conserve this rural-urban bind. Firstly, Hardy utilises the regional novel genre, which, as this essay shows, offers a felicitous framework for considering the problematic repercussions of the rural-urban bind, drawing attention to the importance of locality within the landscape. Secondly, Hardy draws on pastoral landscapes as a setting for representations of how social change, industrialisation and urbanisation affect characters, communities, environments and a nostalgic, rural way of life. Finally, this thesis explores Hardy's antagonistic portrayal of science, religion, industry, technology and travel to argue that the rural-urban bind is used negatively to convey encroaching modernity and portray a growing urban industrial landscape, which consumes rural traditions, erases memories and severs the individual's connection to place.

Many scholars celebrate the emancipatory potential of alter-globalisation networks. This thesis tests this claim, using a case study of the Global Call to Action against Poverty (GCAP), and analysing what the powers which constitute this network reveal about the powers 0/ it. GCAP is one of the largest networks of its type, mobilising nearly 175 million people on a single day in 2009 via national coalitions of civil society organisations in 115 countries. The PhD research focuses on two of these national coalitions in India and Malawi, as well as GCAP's broad governance structures, and utilises semi-structured and ethnographic interviews, participant observation and documentary analysis. The data was analysed through a methodological frame of governmentality and post-governmentality literatures, to analyse the full range of discourses and agencies which construct GCAP. The thesis interrogates the agency of GCAP through an exploration of three power-related themes, namely: the relationships GCAP enacts with processes of statist and neo-liberal hegemony; how GCAP develops relations of solidarity across distance; and the manner in which GCAP constructs subjects of legitimation. The thesis finds that GCAP embodies a monitored subjectivity vis-A-vis statist and neo-Iiberal hegemonic power, yet also retains a monitory agency on those powers. It furthermore finds that relations of solidarity developed in GCAP between areas of structural advantage and disadvantage are imbued with both colonial and postcolonial discourses, which simuhaneously buttress and contest neoliberal discourses of managerialism, resource-dependency and the fetisbisation of 'the poor'. These different sets of relations construct GCAP with a contingent, contradictory, yet at times emancipatory and transcendent subjectivity. By creating a snapshot of an alterglobalisation network in diverse social contexts, this thesis reveals the ways in which the power of such networks is uneven and immanent, dependent upon confluences of the various internal and external powers which constitute them

This thesis reports the syntheses and DNA binding properties of a number of novel DNA binding complexes. A library of ruthenium(II) and rhenium(I) complexes incorporating extended terpyridine type ligands has been synthesised (1). The library consists of a series of structurally related complexes in which we have varied the number of hydrogen bonding sites, charge, chirality and steric demand on the complexes. Experimental data offers clear evidence that these complexes interact with DNA, possibly in an intercalative mode, and that varying the characteristics above has significant effects on the nature of the interaction with DNA. Furthermore a series of achiral mono (2) and bimetallic complexes of ruthenium(II) (3) and rhenium(I) (4) incorporating the well-characterised intercalating domain dppz have been assembled. The novel achiral ruthenium system shows good affinity for CT-DNA and there is a 13-fold increase in binding affinity for the di-cationic complex over the mono-cationic complex. The binding of the di-rhenium and bis-ruthenium complexes show increased affinity for CT-DNA however bis-intercalation is not seen and the major contribution to the interaction comes from electrostatic and groove binding interactions.

Resumo: Neste trabalho descrevemos métodos holográficos para a caracterização de cristais fotorrefrativos do tipo Bi12SiO20. Foram aplicadas as técnicas de modulação de fase do feixe laser e de detecção sincrônica para medir a diferença de fase entre o padrão de interferência luminoso e a rede holográfica gerada por efeito fotorrefrativo. Essas técnicas permitiram operar uma montagem experimental estabilizada, verificar a existência dos hologramas "caminhantes" e medir sua velocidade de deslocamento nesses cristais. A fase e a velocidade do holograma são informações relevantes em experimentos envolvendo a mistura de duas ondas, e foram usados para avaliar parâmetros de transporte desses cristais
Abstract: We describe holographic methods for the characterization of Bi12SiO20 - type photorefractive crystals. Laser beam phase modulation and synchronous detection techniques were used to measure the phase-shift between the interference patern of light and the generated photorefractive holographic grating. These techniques allowed to operate a stabilized holographic setup, to verify the presence of running holograms and to measure their velocity in these crystals. Phase-shift and running hologram velocity are relevant informations for many aplications involving two-wave mixing experiments, and were used for computing transport parameters of these materials

Abstract:
Using survey data from 219 divorced custodial mothers randomly selected from public court records, this research investigated variables which may contribute to increased attachment to a former spouse, postdivorce. A path analysis was conducted to determine the effects of the selected variables on attachment. The findings indicated that attachment was greater for women whose husbands had wanted the divorce, for those who had been married for a longer period of time, for those who were experiencing a difficult time with parenting, and for those who had better coparental relationships with their former spouses. The degree of hostility surrounding the divorce, current conflict between the former spouses, the custody arrangement, and the mother's satisfaction with financial support all had an indirect effect on attachment through their effects on conflict and the quality of the coparental relationship. Possible explanations for these findings as well as implications for therapy are discussed.

Demência é uma síndrome clínica caracterizada pelo declínio progressivo das capacidades cognitivas, estando a tornar-se cada vez mais comum, devido ao envelhecimento da população mundial. Prevê-se que o número de doentes com demência aumente em cerca de 30 milhões nos próximos 15 anos, representando grandes gastos para os sistemas de saúde e sociais. Existem vários tipos de demência, sendo que a Doença de Alzheimer (DA) é a mais comum, afetando entre 20 a 30 milhões de pessoas em todo o mundo, das quais 90.000 são portuguesas. A compreensão das características genéticas e moleculares associadas a esta doença pode constituir um meio para descobrir novos métodos de diagnóstico e tratamento. A maior parte dos casos de Alzheimer tem início tardio, afetando indivíduos com 65 ou mais anos de idade. Até recentemente apenas o gene que codifica a Apolipoproteína E (APOE) foi associado com esta forma de DA. No entanto, estudos recentes de associação genómica identificaram o gene BIN1 como sendo o loci de risco associado ao Alzheimer tardio mais significativo depois do APOE. Além disso, vários SNPs do BIN1 foram associados a este tipo de Alzheimer, sendo que o polimorfismo rs744373 foi proposto como um dos mais relevantes para a DA. Dado que os SNPs mais significativos podem variar de população para população, o objetivo principal deste trabalho foi avaliar se o polimorfismo rs744373 do gene BIN1 pode ser associado a um aumento do risco de desenvolver DA, numa população portuguesa do distrito de Aveiro, que pertence a um estudo transversal baseado em populações, realizado na Universidade de Aveiro. Analisámos 63 indivíduos Portugueses, sendo 32 doentes e 31 controlos. Neste estudo conseguimos observar que, de uma forma geral, o alelo A é o mais frequente e que o alelo G (alelo de risco) foi o menos frequente, numa razão de 3:1. Não conseguimos encontrar uma forte evidência de associação entre o rs744373 e o risco de desenvolver DA (Razão de probabilidade [RP] = 0.733 , valor p = 0.464), o que está de acordo com estudos previamente publicados. Não foi detetada significância estatística entre o rs744373 e portadores do alelo APOE-Ԑ4 (valor p = 0.467) ou indivíduos com demência (CDR≥1) (valor p = 0.269). Foi detetada uma associação entre o alelo de risco do polimorfismo de estudo e a presença de Diabetes Mellitus (RP = 6.60, valor-p = 0.035). No entanto, como a nossa amostra era pequena, deve ser feito um novo estudo para avaliar se este resultado pode ser generalizado para a população Portuguesa.
Dementia is a clinical syndrome characterized by a progressive decline in cognitive skills, and is becoming increasingly common, due to the aging of the world’s population. It is expected that the number of patients with dementia will increase by 30 million in the next 15 years, representing a major factor of costs in health care and social systems. There are several types of dementia, and Alzheimer’s Disease (AD) is the most common, affecting 20 to 30 million people worldwide, of which 90.000 are Portuguese. Understanding the genetic and molecular characteristics associated with the disease may constitute a way to discover new diagnostic methods and treatments. Most cases of AD are late-onset (LOAD), affecting individuals with 65 or more years of age. Until recently only the Apolipoprotein E gene (APOE) had been associated with this form of AD. However, recent genome-wide association studies have identified Bridging Integrator 1 (BIN1) as the most significant LOAD-associated risk loci after APOE. Furthermore, several SNPs of BIN1 have been associated to this type of AD and rs744373 was proposed to be one of the most relevant for AD. Since the most significant SNP may vary from population to population, the main aim of this work was to evaluate if BIN1 polymorphism rs744373 can be associated with the risk of AD in a Portuguese population from the Aveiro district, belonging to a cross-sectional population-based study performed in Aveiro University. We analysed 63 Portuguese individuals comprising 32 cases and 31 controls. In this study we could observe that, overall, allele A was the most frequent and allele G (risk allele) was the least frequent, in a ratio of 3:1. We didn’t find strong evidence of association for rs744373 with the AD risk (odds ratio [OR] = 0.733 , p-value = 0.464), which is in agreement with some previous published studies. No statistical significance was detected between rs744373 and APOE-Ԑ4 carriers (p-value = 0.467) or individuals with dementia (CDR≥1) (p value= 0.269). We have detected an association between the risk allele of the study polymorphism and the presence of Diabetes Mellitus (odds ratio [OR] = 6.60, p-p-value = 0.035). Nevertheless, due to our small sample size, a follow-up study is required in order to evaluate if this result can be generalized to the Portuguese population.

Autonomy has become the villain in many debates about government bureaucracies, and reformers have increasingly urged public agencies to build stronger ties with the outside world. These relationships can make organizations more effective and improve their legitimacy, but they also create new challenges for practice that research has not yet fully explored. In this study, based on a comparative analysis of eleven diverse case studies in policing, I describe the problems and dilemmas that working in partnership entails, and how effective practitioners overcome them. I argue that the central problems of building and sustaining partnerships involve conflict over values: Different social institutions advance different social values, and when the partners who cling to them try to collaborate, conflict flares up at the point of contact. In policing, a small number of conflicts appear recurrently in four different types of partnerships: In community ties, these conflicts involve the relative importance of soft crime, the proper level of police authority, and the tension between equity and the interests of individual neighborhoods; in business ties, they center on the private sector's responsibility for social impacts; in political ties, they involve fights about money, mistakes, and responsiveness; and in inter agency ties, they concern the relative importance of crime control compared with other goals of public policy, and the proper ordering of organizational domains. In some views, conflicts like these are unavoidable, and calls for "partnerships" among organizations are futile. But these cases suggest that many effective police practitioners have been able to overcome them in two ways. First, they employ strategies of institutional change (like appealing to an existing value to institutionalize a new one) to shift their priorities or those of their partners. They thereby close the normative gap that divides them, putting their partnerships on less-contentious ground. Second, some police departments have developed a capacity to attend to competing values in the manner of Isaiah Berlin's fox: Instead of subordinating all but one value in a dilemma, their practitioners have a moral humility and a penchant for "incompletely-theorized agreements" that makes them effective in a world of value pluralism.

Abstract:
The obligate anaerobe Porphyromonas gingivalis is the etiological agent responsible for periodontal disease. It must withstand high levels of reactive nitrogen species in the oral cavity generated by the host and other oral flora. The mechanisms allowing for protection against such stress remain poorly understand. HcpR is an FNR-CRP family regulator that has been implicated in regulation of the nitrosative stress response. In this study we characterize the biochemical properties of HcpR. It is a homo-dimer that is composed of 3 domains – a heme-binding domain, dimerization helix, and a DNA-binding domain. Our studies show that HcpR binds the heme cofactor. UV-Vis and Raman spectroscopy reveal that the bound heme is capable of binding the diatomic gas molecule Nitric Oxide (NO)-a source of nitrosative stress. Binding of NO causes a change in the oxidation state of the iron. SAXS reveals the protein bears a structural resemblance to homology models generated from an ortholog. Promoterr studies reveal that mechanisms P. gingivalis-HcpR uses to modulate expression are novel and different than those found in E. coli and P. aeruginosa.
