Abstract:
One Thousand and One Nights is a composite, transnational work, consisting of popular stories originally transmitted orally within its embedded cultures and developed over several centuries. Ever since its translation into European languages in the nineteenth century, or perhaps even before, it has been adapted and appropriated into different forms and mediums and thus has reached different corners of the world. This project was inspired by the level of popularity of One Thousand and One Nights, often known as The Arabian Nights, in the world today. Although only a relatively small number of people might have read all the tales, we can safely assume that most people do have an idea of what the Nights are, whilst some could even name one or two films, series or cartoons that they think are based on the Nights. Indeed, only a very limited number of stories included in editions of the Nights have been adapted into films or TV series. There are two main characteristics of the Nights that help identify adaptations and adoptions in popular culture: embedded storytelling using a frame tale, and the ‘feminist’, emancipating heroine Scheherazade. The popular Turkish TV series Binbir Gece (One Thousand and One Nights) (2006‐09), which this article focuses on, not only makes use of these two popular features; it also offers a fresh and contemporary adaptation of the frame story of Shah Shahriyar and Scheherazade and elements from many other tales from the Nights, such as the emphasis on the importance of education for women, or the evil of cunning women. After analysing the degree of adaptation of the frame story in this series, this article sheds light on its global reach, reception and popularity.

Abstract:
A new trend in Greek TV is the screening of Turkish TV Serial, which seem to be very popular in many European countries. The effect of such Turkish Serial is so huge on the TV's audience that many times people prefer attending such soap-operas rather than other TV's very popular events, such as football games or Eurovision's competition.In this direction, as primary motivation for us, as academics and researchers was to explore the reasons for which the Greek audience likes attending Turkish TV Serial, focusing as an example on two very popular Turkish TV Serial which were shown at Greek TV, with sample University's students: these are "Binbir Gece" (Thousand and One Nights) and "Suleiman the Magnificent". The first TV Serial was shown by Greek TV in 2010 and the second since 2012. The protagonist in both TV Serial is the same actor, a very popular person for the Greek audience and both Serial were shown by Greece's big TV network, Ant1, at prime time, 9 p.m. Following, in this paper we present the methodology of the two studies and their results, i.e. frequency tables, we implemented at the University of Western Macedonia, a regional University in Northern Greece; an analysis of the results and conclusions follow. Birçok Avrupa ülkesinde popüler olan Türk TV dizileri Yunan televizyonunda yeni bir eğilimdir. Böyle Türk dizilerinin etkisi çok büyük olduğundan, TV seyircileri artık futbol maçları veya Eurovision yarışması gibi diğer çok popüler olan TV programları yerine çoğu zaman sabun köpüğü operaları seyretmeyi tercih etmektedirler. Bu doğrultuda, Yunan seyircilerin Türk dizilerini seyretme nedenini araştırmak için, üniversite öğrencileri örneği ile Yunan TV’sinde gösterilen iki popüler Türk TV dizisine odaklandık: bunlar: "Binbir Gece" ve Kanuni Sultan Süleyman’ın hayatını yansıtan “Muhteşem Yüzyıl” dizileridir. Yunan TV’sinde ilk Türk TV dizisi 2010 da ve ikincisi ise 2012’den itibaren gösterilmiştir. İki dizinin de kahramanı, Yunan seyirciler için çok popüler olan, aynı oyuncudur. İki dizi de Yunanistan’ın en büyük TV kanalı olan Ant1’de akşam saati’nde (Prime Time) 09:00 da yayınlanmıştır. Dahası, bu çalışmada iki araştırmanın metodolojisi ve sonuçları sunulmaktadır. Bunlar, Kuzey Yunanistan’da bir bölgesel Üniversite olan Batı Makedonya Üniversitesi ve uygulanan frekans tabloları ve sonuçların analizleridir.

Her halkın güzellik hakkında, yani gülün, doğanın, hayatın, insanın ve elbette kadının güzelliği hakkında kendi bakış açısı vardır. Bu görüşler edebiyat dairesinde daha da derinleşir. Dünya edebiyatına baktığımızda kadın karakterinin çok eskiden itibaren kült seviyesine yükseldiğine şahit oluruz. Bin bir gece masallarından Gümüş Bibi, Zeynep, Zebi, Zeberced gibi karakterlerin ele alındığı modern eserlere kadar edebiyatımızda ulusal kimliğimizi özünde taşıyan kadın karakterler bulunmaktadır. İnsan bilimi olarak bilinen edebiyat, her şeyden önce kadın bilimi, yani kadını araştıran, onu keşfeden bir bilimdir. Dikkat edilirse en eski eserlerin de kadına atfedildiğini anlaşılır. Söz konusu eserlerin çatışma merkezinde kadın ve ona ait unsurlar bulunur. Hatta edebiyatın gelişmesi kadın temasının edebî tetkiki, onun gittikçe derinlik kazanması ile sıkı sıkıya bağlıdır.  Nitekim, dünya edebiyatında kadın karakterin genel ve özel niteliklerinin araştırılması edebî sürecin genel kurallarını, özellikle milli edebiyatları karşılaştırmalı-tipolojik yönden araştırmaya yardımcı olur.  Kadın karakterin dünya edebiyatında ele alınışı ve toplumdaki yeri onun yaşadığı tarihî dönemle doğrudan ilintilidir. 18. Yüzyıl nesrinde kadınların romantik karakteri yaratılırken daha sonradan kadın haklarını savunan mücadeleci karakterler ortaya çıkmaya başladı. Dünya edebiyatına baktığımızda, İngiliz edebiyatında Elizabeth, Japon edebiyatında Kraliçe Otikuba, Rus edebiyatında Tatyana Larina ve Nataşa Rostova, Amerika edebiyatında Scarlett O’hara, Fransız edebiyatında Madame Bavary gibi o halkın kadınlarının sembolüne dönüşen karakterlere rastlarız. Bu tür karakterler XIX. Yüzyıl klâsik Rus edebiyatında daha geniş işlenmiştir. Dikkat edilirse en eski eserlerin de kadınlara atfedildiği anlaşılmaktadır. Söz konusu eserlerde kurgulanan çatışmanın merkezinde daima kadın ve ona dair unsurların yoğun olarak yer aldığını görmek mümkündür. Aslında edebiyatın teknik ve içerik olarak gelişimi, kadın temasının edebi olarak incelenmesi ve derinliğinin artmasıyla da yakından ilişkilidir. Nitekim dünya edebiyatında kadın karakterin genel ve özel niteliklerinin araştırılması, edebi sürecin genel kurallarının, özellikle de milli edebiyatların karşılaştırmalı-tipolojik açıdan araştırılmasına yardımcı olur. Dünya literatürüne bakıldığında kadın karakterinin eski çağlardan bu yana eserlerde süreç içerisinde kült düzeyine yükseldiğine tanık olunmaktadır. Kadın karakterinin dünya edebiyatında ele alınışı ve toplumdaki yeri, yaşadığı tarihsel dönemle doğrudan ilişkilidir. Dünya edebiyatında kadının bir kült düzeyine yükseldiğini gösteren örnekler arasında İngiliz edebiyatında Elizabeth, Japon edebiyatında Queen Otikuba, Rus edebiyatında Tatyana Larina ve Natasha Rostova, Amerikan edebiyatında Scarlett O'hara, Fransız edebiyatında Madame Bovary gibi ünlü isimleri saymak mümkün. Bu kahramanlar yaşadıkları dönemin kadınlarının sembolleri. Ayrıca Doğu medeniyetinde de Binbir gece masallarından Gümüş Bibi, Zeynep, Zebi ve Zeberced gibi karakterlerin işlendiği modern eserlere kadar, özünde milli kimliği taşıyan kadın karakterler var. Özbek edebiyatında kadın karakterleri ele alma ve farklı tiplerini tasvir etme deneyimi devam etmektedir. Bu makalede günümüz Özbek hikâyeciliğinde kadın karakteri ve kadının edebi yorumu konusu incelenmiştir.25 Temmuz 1908’de Meşrutiyet’in ilanıyla birlikte gazeteler sansürsüz yayımlanmaya başlamıştır. Gazete, dergi ve kitapların sayısında büyük bir artış yaşanmıştır. İkinci Meşrutiyetle birlikte başlayan hem İstanbul hem de taşra basınının özellikle de azınlık gazetelerinin son derece özgür bir ortamda yaptıkları gazetecilik, Birinci Dünya Savaşı ile birlikte sona ermiş ve yerini sıkı bir denetime bırakmıştır. Birinci Dünya Savaşı sonrasında başlatılan Millî Mücadele döneminde ise dönemin telgraftan sonra en etkili kitle iletişim aracı olarak görülen gazeteler; işgal kuvvetleri–İstanbul Hükümeti ve Ankara Hükümeti’nin kendi görüşlerini kitlelere duyurmasında birer araç ve ideolojik bir mücadele sahası olarak karşımıza çıkmaktadır.

Abstract:
Meru is a sacred structure in Balinese architecture, symbolizing Mount Sumeru in Hindu-Buddhist cosmology. Introduced by Mpu Kuturan in the 11th century, Meru is a key feature of Balinese temples, representing the connection between the divine and earthly realms. The tiered roof, which can range from one to eleven layers, reflects different levels of the universe, with higher tiers holding greater spiritual significance. Meru is not only a site of worship for deities and ancestors but also plays a crucial role in traditional ceremonies, such as the ngaben cremation rites. One of the remarkable aspects of Meru is its earthquake-resistant design. Built with natural materials like ijuk (sugar palm fibre) and ironwood, its proportional structure and construction techniques allow it to withstand seismic shocks effectively. However, while Meru can endure earthquakes, it remains vulnerable to fire, especially due to lightning strikes. The dry, flammable ijuk used for the roof makes Meru susceptible, as evidenced by recent fires at temples like Ratu Gede Bima Sakti and Dadia Bingin. Efforts to protect Meru from fire hazards are underway, including raising community awareness and improving fire safety protocols. Despite these challenges, Meru continues to stand as a powerful symbol of Bali’s spiritual and cultural identity, blending ancient traditions with modern efforts to preserve this sacred architecture.

The worship of local gods in the implementation of Saṃskāra in the Pedawa Village community is seen in the Sesapa spoken by Balian Desa in the implementation of the ritual. Not only local gods but worship of places and temples in Pedawa Village is also carried out. This study questions the worship of local gods that was carried out during the implementation of Saṃskāra in Pedawa Village. The method used in this research is descriptive qualitative. Data was collected by using interview and direct observation techniques. The results of this study show that the worship of local gods in the implementation of Saṃskāra in Pedawa Village can be divided into 4, namely (1) worship that is specifically shown to local gods, namely worship of gods who are believed by the Pedawa people to provide salvation as Ida Pengkan Prataksu , Ida Gede Kemulan, Rai Ida. (b) special worship for places in Pedawa Village such as Gunung Sari Pedawa, In Kamulan Bingin, Telaga Waja Pengampelan and other places in Pedawa Village (c) worship of holy places in Duurin Capah, namely worship of temples in the Tamblingan and Buyan Areas or known as Dalem Tamblingan Village Chess such as Ulun Danu Tamblingan Temple, Dalem Naga Loka, Sang Hyang Kangin, Dangin Capah taler ring Steaming. Abstract:
The useful life of a concrete structure is highly dependent upon its durability, which enables it to withstand the harsh environmental conditions. Resistance of a concrete specimen to rapid chloride ion penetration (RCP) is one of the tests to indirectly measure its durability. The central aim of this study was to investigate the influence of different variables, such as, age, amount of binder, fine aggregate, coarse aggregate, water to binder ratio, metakaolin content and the compressive strength of concrete on the RCP resistance using a genetic programming approach. The number of chromosomes (Nc), genes (Ng) and, the head size (Hs) of the gene expression programming (GEP) model were varied to study their influence on the predicted RCP values. The performance of all the GEP models was assessed using a variety of performance indices, i.e., R2, RMSE and comparison of regression slopes. The optimal GEP model (Model T3) was obtained when the Nc = 100, Hs = 8 and Ng = 3. This model exhibits an R2 of 0.89 and 0.92 in the training and testing phases, respectively. The regression slope analysis revealed that the predicted values are in good agreement with the experimental values, as evident from their higher R2 values. Similarly, parametric analysis was also conducted for the best performing Model T3. The analysis showed that the amount of binder, compressive strength and age of the sample enhanced the RCP resistance of the concrete specimens. Among the different input variables, the RCP resistance sharply increased during initial stages of curing (28-d), thus validating the model results.

Abstract G-quadruplex (G4) structures that can form at guanine-rich genomic sites, including telomeres and gene promoters, are actively involved in genome maintenance, replication, and transcription, through finely tuned interactions with protein networks. In the present study, we identified the intermediate filament protein Vimentin as a binder with nanomolar affinity for those G-rich sequences that give rise to at least two adjacent G4 units, named G4 repeats. This interaction is supported by the N-terminal domains of soluble Vimentin tetramers. The selectivity of Vimentin for G4 repeats versus individual G4s provides an unprecedented result. Based on GO enrichment analysis performed on genes having putative G4 repeats within their core promoters, we suggest that Vimentin recruitment at these sites may contribute to the regulation of gene expression during cell development and migration, possibly by reshaping the local higher-order genome topology, as already reported for lamin B.

Abstract:
The depletion of natural resources of river sand and its availability issues as a construction material compelled the researchers to use manufactured sand. This study investigates the compressive strength of concrete made of manufactured sand as a partial replacement of normal sand. The prediction model, i.e., gene expression programming (GEP), was used to estimate the compressive strength of manufactured sand concrete (MSC). A database comprising 275 experimental results based on 11 input variables and 1 target variable was used to train and validate the developed models. For this purpose, the compressive strength of cement, tensile strength of cement, curing age, Dmax of crushed stone, stone powder content, fineness modulus of the sand, water-to-binder ratio, water-to-cement ratio, water content, sand ratio, and slump were taken as input variables. The investigation of a varying number of genetic characteristics, such as chromosomal number, head size, and gene number, resulted in the creation of 11 alternative models (M1-M11). The M5 model outperformed other created models for the training and testing stages, with values of (4.538, 3.216, 0.919) and (4.953, 3.348, 0.906), respectively, according to the results of the accuracy evaluation parameters root mean square error (RMSE), mean absolute error (MAE), and coefficient of determination (R2). The R2 and error indices values revealed that the experimental and projected findings are in extremely close agreement. The best model has 200 chromosomes, 8 head sizes, and 3 genes. The mathematical expression achieved from the GEP model revealed that six parameters, namely the compressive and tensile strength of cement, curing period, water–binder ratio, water–cement ratio, and stone powder content contributed effectively among the 11 input variables. The sensitivity analysis showed that water–cement ratio (46.22%), curing period (25.43%), and stone powder content (13.55%) were revealed as the most influential variables, in descending order. The sensitivity of the remaining variables was recorded as w/b (11.37%) &gt; fce (2.35%) &gt; fct (1.35%).

One of the problems of optimization of concrete is to formulate a mathematical equation that shows the relationship between the various constituents of concrete and its properties. In this work, modelling of the compressive strength of concrete admixed with metakaolin was carried out using the Gene Expression Programming (GEP) algorithm. The dataset from laboratory experimentation was used for the analysis. The mixture proportions were made of three different water/binder ratios (0.4, 0.5, and 0.6), and the grades of concrete produced were grade M15 and M20. The compressive strength of the concrete was determined after 28 days of curing. The parameters used in the GEP algorithm are the input variables which include cement content, water, metakaolin content, and fine and coarse aggregate, while the response was designated as the compressive strength. The model was trained and tested using the parameters. The R-square value from the GEP algorithm was compared with the use of conventional stepwise regression analysis. With a coefficient of determination (R-square value) of 0.95, the GEP algorithm has shown to be a good alternative for modelling concrete compressive strength.

Abstract Introduction: Metastatic Lymph Node Gene 51 Protein (MLN51) also known as Cancer Susceptibility Candidate Gene 3 Protein (CASC3) or Barentsz (BTZ) is a molecule that was first identified in malignant breast tumours. The CASC3 gene encodes the protein that binds to mRNA and is a key component of the exon junction complex (EJC), playing an important role in the metabolism of mRNA and in cellular recovery after stress responses. It is not clear, however, if MLN51 has a clinical value in assessing the disease progression of breast cancer. Methods: We analysed the transcription expression levels of MLN51 in a cohort of human breast cancer tissues and correlated the expression pattern with clinical outcome, pathological type and hormonal receptor status. Results: High levels of MLN51 was found to be a favourable prognostic indicator for overall survival (p=0.008, Hazard Ratio [HR=0.262]), the prognostic value being independent of other clinical and pathological factors. Likewise, high levels of MLN51 were also a significant and independent predictive factor for disease free survival (DFS) (p=0.003, HR=0.262). It was found that the favourable prediction in both OS and DFS was associated with node positive tumours, with significance with OS (p=0.004), compared in node negative tumours (p=0.143) and to DFS (p=0.001 in node positive compared with p=0.079 in node negative tumours). Levels of MLN51 had a significant correlation with EGFR in both normal and tumour mammary tissues but correlated with Her-2 and Her-3 in tumour tissues only. The prognostic power of MLN51 for DFS was significant in both Her-2 positive and Her-2 negative (p=0.003 and p=0.012) tumours and in ER negative tumours (p=0.003). The prediction for OS by MLN51 was particularly strong for Her-2 positive and ER positive (p=0.007 for both) tumours. Conclusions: MLN51 (CASC3) is a favourable prognostic factor in patients with breast cancer. This is clearly linked to the nodal status and hormone receptor status, particularly ER and HER-2. IGFs play key roles in regulating vertebrate development, growth, reproduction, and aging. In extracellular fluids, IGFs are bound and regulated by a family of IGF-binding proteins (IGFBPs). Although all known IGFBPs are secreted proteins, some are also found in the nucleus and possess IGF-independent activities. When and how these distinct modes of biological actions have evolved is unknown. In this study, we identified and analyzed an IGFBP gene from amphioxus. Amphioxus shares a common ancestor with the modern vertebrate lineage that dates back to more than 520 million years ago. The amphioxus IGFBP shares all major structural characteristics of vertebrate IGFBPs. Phylogenetic analyses place it in a basal position in the IGFBP lineage. Ligand blot analysis reveals that amphioxus IGFBP does not bind to IGF-I or -II. Changing its Phe70 into Leu, however, is sufficient to convert it into a functional IGF binder. When tested in cultured cells, amphioxus IGFBP is localized in the nucleus, and this is attributed to 2 redundant nuclear localization sequences in its L domain. Furthermore, the amphioxus IGFBP N-terminal domain has strong transcriptional activation activity. Forced expression of amphioxus IGFBP in zebrafish embryos results in dorsalized phenotypes. This action requires nuclear localization. These results suggest that the nuclear localization and transcription activation activity of IGFBPs are ancient functions and the IGF-binding function may have been acquired by opportunistic gain-of-functional mutations later in evolution.

The study aims to develop a reliable model using gene-expression programming (GEP) technique for estimating the unconfined compressive strength (UCS) of soil stabilization by cement and fly ash. The model considered the effects of several parameters, including the fly ash characteristics such as calcium oxide (CaO) content, CaO/SiO2 ratio, and loss of ignition. The research results show that the proposed model demonstrates superior performance with a high correlation coefficient (R &gt; 0.955) and low errors. Therefore, the model could be confidently applied in practice for a variety of fly ash qualities. Besides, the parametric study was conducted to examine the effect of fly ash characteristics on the strength of soil stabilization. The study indicates that if the fly ash contains a high amount of calcium oxide, the strength of fly ash stabilized soil is significant. In addition, fly ash could be used in combination with cement to increase the strength of the mixture. A fly ash replacement ratio is suggested from 0.19 to 0.35, corresponding to the total binder used from 10% to 30%. The research findings could help engineers in optimizing the fly ash proportion and estimating the UCS of soil stabilization by cement and fly ash.

Feed safety is a necessity for animal health and welfare as well as prerequisite for food safety and human health. Wheat gluten (WG) is considered as a valuable protein source in fish feed due to its suitability as a feed binder, high digestibility, good amino acid profile, energy density and most importantly, due to its relatively low level of anti-nutritional factors (ANFs). The main aim of this study was to identify the impact of dietary WG on salmon health by analysing growth, feed efficiency and the hepatic and intestinal transcriptomes. The fish were fed either control diet with fishmeal (FM) as the only source of protein or diets, where 15% or 30% of the FM were replaced by WG. The fish had a mean initial weight of 223 g and approximately doubled their weight during the 9-week experiment. Salmon fed on 30% WG showed reduced feed intake compared to the 15% and FM fed groups. The liver was the less affected organ but fat content and activities of the liver health markers in plasma increased with the inclusion level of WG in the diet. Gene expression analysis showed significant changes in both, intestine and liver of fish fed with 30% WG. Especially noticeable were changes in the lipid metabolism, in particular in relation to the intestinal lipoprotein transport and sterol metabolism. Moreover, the intestinal transcriptome of WG-fed fish showed shifts in the expression of a large number of genes responsible for immunity and tissue structure and integrity. These observations implied that the fish receiving WG-containing diet were undergoing nutritional stress. Overall, the study provided evidence that a high dietary level of WG can have a negative impact on the intestinal and liver health of salmon with symptoms similar to gluten sensitivity in humans.

Chronic Lymphocytic Leukemia (CLL) is a heterogeneous disease characterized by variable clinical courses among different patients. This notion was supported by the possible coexistence of two or more independent CLL clones within the same patients, identified by the characterization of the B cell receptor immunoglobulin (BcR IG) idiotypic sequence. By using the antigen-binding site of the BcR IG as bait, the identification and isolation of aggressive and drug-resistance leukemic B-cell clones could allow a deeper biological and molecular investigation. Indeed, by the screening of phage display libraries, we previously selected a peptide binder of the idiotypic region of CLL BCR IGs expressing the unmutated rearrangement IGHV1-69 and used it as a probe to perform a peptide-based cell sorting by flow cytometry in peripheral blood samples from patients with CLL. Since the IGHV1-69 clones persisted during the follow-up time in both patients, we explored the possibility of these clones having acquired an evolutive advantage compared to the other coexisting clones in terms of a higher expression of genes involved in the survival and apoptosis escape processes. To this end, we studied the expression patterns of a panel of genes involved in apoptosis regulation and in NF-kB-dependent pro-survival signals by comparative qRT-PCR assays. According to the results, IGHV1-69 clones showed a higher expression of pro-survival and anti-apoptotic genes as compared to the other CLL clones with different immunogenetic characteristics. Moreover, these IGHV1-69 clones did not carry any characteristic genetic lesions, indicating the relevance of our approach in performing a comprehensive molecular characterization of single tumor clones, as well as for designing new personalized therapeutic approaches for the most aggressive and persistent tumor clones.

Confirmation of gene editing in livestock following CRISPR/Cas9 zygotic microinjection is often limited to either protein expression or gene sequencing analyses of unfixed embryos, but not both, due to the challenges of extracting DNA from fixed samples suitable for genotyping. Bovine embryos have been derived from gene-edited cell lines to screen for mutations followed by nuclear transfer, but these techniques limit embryo production efficiency, include technical challenges, and alter physiological relevance. Herein we report an integrative approach to evaluate both protein expression and the genotype of individual, fixed bovine embryos subjected to CRISPR/Cas9 microinjection. Bovine zygotes were derived from IVF and parthenogenetic activation of in vitro-matured oocytes followed by intracytoplasmic injection of CRISPR/Cas9 targeting embryonic POU5F1. Embryos were cultured for 7 days and fixed in 4% paraformaldehyde and stored at 5°C for up to 4 weeks. Fixed embryos were first subjected to immunohistochemistry to determine POU5F1 mutation success by protein expression. Binder reinforcement labels were affixed to glass slides as a substitute for adhesive spacers. Single embryos were added to 9µL of PBS with a coverslip for imaging. Individual embryos were then recovered and placed in 10µL of QuickExtract (Lucigen, Middleton, WI, USA) for DNA extraction by heating samples to 65°C for 6min and 95°C for 2min. Two rounds of PCR were applied to templates in 20-µL reactions consisting of 10µL of GoTaq Hot Start Green Master Mix (2×; Promega, Madison, WI, USA) with the addition of 0.4µL (10µM) of forward and reverse primer each and 9.2µL of template. The second PCR reaction contained 5µL of PCR product from the first reaction, nested primers, and 4.2µL of H2O. The PCR conditions were |95°C, 3 min| and 35 cycles of |95°C, 30s |56°C, 30 s|72°C, 30 s|72°C, 7 min|. The PCR product was run on an agarose gel to confirm DNA amplification of a single band, and the remainder was purified (QIAquick PCR Purification Kit, Qiagen, Hilden, Germany) and submitted (100ng) for Sanger sequencing. The CRISPR-injected embryos were aligned to wild-type embryo sequences using SnapGene (http://www.snapgene.com/) and TIDE software (https://tide.nki.nl/). Both DNA extraction (n=18/19) and Sanger sequencing (n=19/23) proved highly efficient and repeatable (83 and 95%, respectively). These methods provide an efficient, cost effective, and reproducible approach for confirmation of CRISPR gene editing in individual and fixed bovine embryos by both protein expression and gene sequencing. In addition, these techniques complement the production of gene editing in livestock after zygotic CRISPR microinjection by conclusively determining mutation efficiency through both proteomic and genomic analyses. In vivo embryos (n = 8) were derived by uterine flushing of artificially inseminated mares at 7 days after ovulation. For the production of the in vitro embryos (n = 8), oocytes from slaughtered mares were matured in DMEM-F12-based medium (Galli et al. 2007 Anim. Reprod. Sci. 98, 39-55) in 5% CO2 in air (maturation rate: 57%), fertilized by intracytoplasmic sperm injection, and cultured in DMEM-F12 with 10% fetal calf serum in 5% CO2, 5% O2, and 90% N2 for 9.5 days (cleavage rate: 74%; blastocyst rate: 7%). RNA was extracted from single early to expanded blastocysts and amplified and converted into cDNA with the WT-Ovation RNA Amplification System (NuGEN, San Carlos, CA, USA). Based on the presumed gene functions and differential gene expression as determined in a previously performed suppression subtractive hybridization (SSH; Smits et al. 2009 Reprod. Dom. Anim. 44, 75), 5 genes [brain expressed X-linked 2 (BEX2), Mps one binder kinase activator-like 3 (MOBKL3), fatty acid binding protein 3 (FABP3), minichromosome maintenance complex component 7 (MCM7), and ornithine decarboxylase (ODC)] were selected for quantification by RT-qPCR with the KAPA SYBR® FAST qPCR Kit (Kapa Biosystems, Belgium) on the iCycler iQ Real-Time PCR Detection System (Bio-Rad, Nazareth, Belgium). All data were normalized with previously determined stable reference genes (beta actin, ubiquitin C, ribosomal protein L32, and glyceraldehyde-3-phosphate dehydrogenase) and statistically analyzed by means of a Mann-Whitney test. The fact that all genes were expressed at greater levels in the in vivo-derived blastocysts than in the in vitro-produced blastocysts confirmed the results of the SSH. This difference was highly significant for MOBKL3, BEX2, and ODC (P &lt; 0.005), significant for FABP3 (P &lt; 0.05), and not significant for MCM7. These genes have already been shown to be important for embryonic cell survival (ODC), oocyte maturation and pre- implantation development (MOBKL3) in mice, regulation during embryonic development (BEX2) and fetal development (FABP3) in human, and genome replication in eukaryotes (MCM7) (Pendeville et al. 2001 Mol. Cell Biol. 21, 6549-6558; Han et al. 2005 Nucleic Acids Res. 33, 6555-6565). In conclusion, 4 genes (MOBKL3, BEX2, ODC, and FABP3) with greater expression levels in in vivo-derived equine blastocysts have been identified. Whether the up-regulation of these genes is important for normal embryonic differentiation in the horse embryo is currently under investigation.

265 Background: Treatment options for cholangiocarcinoma (CCA) after progression on first-line gemcitabine-based therapy are limited. Fibroblast growth factor receptor 2 ( FGFR2) gene fusions occur in 13–17% of intrahepatic CCA. A single-arm, phase II study (NCT02150967) evaluated infigratinib, an ATP-competitive FGFR1–3-selective oral tyrosine kinase inhibitor, in previously-treated advanced CCA with FGFR fusions/rearrangements. Methods: Adult patients with advanced/metastatic CCA with progression on ≥1 line of systemic therapy received infigratinib 125 mg orally for 21 days of each 28-day cycle until unacceptable toxicity or disease progression. All patients received prophylaxis with the oral phosphate binder sevelamer. Primary endpoint: objective response rate (ORR) by independent central review per RECIST v1.1, with duration of response (DOR). Secondary endpoints: progression-free survival (PFS), disease control rate, overall survival, safety, pharmacokinetics. Approximately 160 patients are planned (120/20/20 patients in Cohorts 1/2/3). This analysis focuses on Cohort 1 (patients with FGFR2 gene fusions or rearrangements without receiving a prior FGFR inhibitor). Results: As of 31 March 2020, 108 patients, including 83 (77%) with FGFR2 fusions, received infigratinib: median age 53 years (range 23–81 years); 54% had received ≥2 prior treatment lines. Median follow-up was 10.6 months (range 1.1–55.9 months). 96 patients (88.9%) discontinued treatment (12 ongoing). Centrally reviewed ORR was 23.1% (95% CI 15.6–32.2) including 1 CR and 24 PRs; median DOR was 5.0 months (range 0.9–19.1 months). Among responders, 8 (32.0%) patients had a DOR of ≥6 months. Median PFS was 7.3 months (95% CI 5.6–7.6 months). Prespecified subgroup analysis: ORR was 34% (17/50) in the second-line setting and 13.8% (8/58) in the third-/later-line setting (3–8 prior treatments). Most common treatment-emergent adverse events (TEAEs, any grade) were hyperphosphatemia (76.9%), eye disorders (67.6%, excluding central serous retinopathy/retinal pigment epithelium detachment [CSR/RPED]), stomatitis (54.6%), and fatigue (39.8%). CSR/RPED occurred in 16.7% of patients (including 1 G3 event; 0 G4). Other common grade 3/4 TEAEs were stomatitis (14.8%; all G3), hyponatremia (13.0%; all G3), and hypophosphatemia (13.0%; 13 G3, 1 G4). Conclusions: Infigratinib is associated with promising anticancer activity and a manageable AE profile in patients with advanced, refractory CCA with an FGFR2 gene fusion or rearrangement. Abstract Follicular lymphomas (FL) are associated with the chromosomal translocation t(14;18)(q32;q21). Most breakpoints of chromosome 18 (60%) occur in the major breakpoint region (MBR) of the BCL-2 gene. Further breakpoints have been detected in the minor cluster region (mcr), less frequent breakpoints are found in regions called 3′-MBR, 5′-mcr and icr. On chromosome 14 most breakpoints are located within one of the six JH-genes. Therefore, BCL-2 translocations with breakpoints within the MBR and mcr are generally detected by PCR using combinations of different BCL-2 primers with one JH-consensus primer. We have developed a multiplex quantitative real-time PCR strategy that that can be used to detect t(14;18) translocations with breakpoints located within all regions mentioned above. To minimize the costs for expensive probes we used the JH-consensus sequence as a target for one “consensus probe” (fluorescent labelled minor groove binder probe) for all assays in combination with 6 different JH intron primers. To reduce the size of amplified PCR fragments 12 BCL-2 primers were chosen in combination with 6 JH intron primers for the detection of all 5 breakpoint regions. It is very important to choose short DNA target sequences for amplification: (a) to establish a real-time PCR with a high amplification efficacy; (b) to be able to amplify target sequences also from partially degraded DNA isolated from formaldehyde-fixed paraffin-embedded tissue sections; (c) to achieve a high sensitivity to detect 1–3 copies per assay. Peripheral bood mononuclear cells (PBMNC) and formalin fixed, paraffin embedded lymph node tissue obtained from 139 FL patients at the time of diagnosis (LN and PBMNC, n = 54; LN only, n = 3; PBMNC only, n = 82) were tested by multiplex quantitative real-time PCR. 80 breakpoints were identified within the MBR (61%) region. For comparison, 78/80 breakpoints were also detected by our standard real-time PCR assay with one BCL-2-MBR- primer and one JH consensus primer in combination with a fluorescent probe located within the BCL-2 sequence [Doelken et al., BioTechniques, 1998]. Two additional translocations with breakpoints located 5′ of the target sequence of the standard PCR were found by using two additional MBR primers. In addition, five mcr breakpoints (5%), one breakpoint in the 3′MBR region and one breakpoint in the icr region were found. Based on these results the prevalence of breakpoints in various regions of the BCL-2 gene in FL patients is: MBR = 61% (80/139); mcr = 5% (5/139); 3′MBR = 1% (1/139); icr = 1% (1/139); 5′mcr = 0%). Furthermore, based on quantitative PCR results the t(14;18) translocations detected in this study were undoubtedly lymphoma associated and did not belong to t(14;18)-positive non-lymphoma B cell clones found in healthy persons. By applying this multiplex quantitative real-time PCR strategy t(14;18) translocations with breakpoints in five different breakpoint clusters can be detected in about 70% of patients with follicular lymphoma. The assays can be used for a fast and reliable quantitative detection of t(14;18) translocations on DNA isolated from fresh lymph nodes or pathological specimens as well as blood samples at the time of diagnosis. In almost all cases quantitative results will allow a distinction whether the translocation found is lymphoma associated or not, which will in turn allow a quantitative MRD analysis on follow-up samples during and after treatment.

Abstract Abstract 472 Background: Despite recent therapeutic advances, multiple myeloma (MM) remains primarily an incurable cancer. Patients experiencing rapid recovery of T cells post auto-SCT may have improved outcomes, and spontaneous cellular responses to tumor can occur, suggesting immune mediated control of tumor is possible. We and others have investigated therapeutic cancer vaccines that have shown promise in pilot studies. However, vaccine efficacy may be limited by thymic selection which restricts the repertoire of T cell receptors (TCRs) to low affinity TCRs that cannot recognize the reduced level of antigen present on most tumor cells. We hypothesized that incorporation of affinity-enhanced tumor antigen specific TCRs into autologous T cells would overcome this limitation. Methods: We report interim results of a Phase II clinical trial (NCT01352286) to evaluate the safety and activity of autologous T cells genetically engineered to express an HLA-A201 restricted, affinity-enhanced TCR targeting an epitope shared by the NY-ESO-1 and LAGE-1 tumor antigens (gene engineered cells, or GEC). Inclusion criteria include: 1) eligibility for auto-SCT, 2) PS of 0–2, 3) high risk MM or relapse after prior therapy (prior allo-SCT excluded), 4) HLA-A201 positive, and 5) NY-ESO-1 and/or LAGE-1 positive tumor. GEC are manufactured from monocyte and T regulatory cell depleted apheresis, followed by lentivector gene transfer and T cell expansion by bead-based artificial antigen presenting cells. GEC are administered four days after high dose melphalan and two days following auto-SCT, at a dose range of 1–10 billion cells. Patients are evaluated for MM responses in accordance with the IMWG criteria at 6 weeks, and 3 and 6 months post infusion. At 3 months, patients start lenalidomide maintenance. The initial 6 patient phase is complete and a 20 patient extension phase is ongoing. Results: 19 patients have been enrolled, 12 have been infused, 3 are pending infusion, and 4 were taken off study prior to infusion due to disease progression. The average TCR transfer efficiency was 31.9% (range 18.2%-45%). 6, 4, and 1 patients have reached the 6 month, 3 month, and 6 week post infusion MM assessment time points, respectively. All treated patients are alive. 3/11 (27%) and 7/11 (64%) patients have a stringent CR or best response of VGPR or better, respectively; 3/11 (27%) have stable or partial responses, and 1/11 (9%) had progressive disease. In patients with VGPR, oligoclonal banding was common, marrow showed no expansion of clonal plasma cells, and all had normalized light chain ratios, suggesting at most minimal residual disease. These results are encouraging, especially considering that 3/11 patients had prior ASCT and 8/11 had adverse cytogenetics. GEC infusions have been well tolerated. The majority of adverse events are related to the high dose melphalan. Several patients had a transient skin rash with lymphocytosis and others had a diarrheal syndrome that occurred later than expected for melphalan-induced mucositis. In all cases GI toxicity has been self-limited or treatable with a short dose of corticosteroids. Possibly related SAEs were cytopenias such as neutropenia and thrombocytopenia, and GI and metabolism disorders such as diarrhea, colitis, hyponatremia and hypomagnesemia. GEC have been detected for up to 1 year at 1% in the circulation and bone marrow. Detailed correlative analysis is reported elsewhere in this meeting (Kalos, M. et al). Summary: This is the first test of GEC in the MM setting. Infusion of GEC administered post auto-SCT is safe, well tolerated, and leads to high response rate in a high risk myeloma population. Yüzlerce hikâyeden oluşan bu masalların ana karakterleri, karısı tarafından ihanete uğrayan ve bundan dolayı bütün kadınlardan nefret eden Kral Şehriyâr ve vezirinin kızı Şehrâzât’tır. Tam olarak kaç hikâye olduğu hakkında tartışmaların bulunduğu Binbir Gece Masallarının birbirinden farklı birkaç baskısı bulunmaktadır. Arap edebiyat tarihinde Hikâyâtu/Kıssatu Elf Leyle ve Leyle olarak bilinen bu eserin barındırdığı hikâyeler, ait olduğu dönemlere ışık tutacak sosyo-kültürel konuları içermektedir. Kökeni efsanelere dayanan eser her ne kadar Şehrâzât tarafından söylendiği iddia edilirse de bu eserin barındırdığığı hikâyelerin çoğu Hint ve İran menşelidir. Firak ve visal olgusu insanlık tarihi kadar eskidir. Bu olgu, başta dini metinler olmak üzere destan, masal, hikâye ve diğer edebî türlerde yer almıştır. Edebî bir hazine olan Binbir Gece Masalları da bu edebî türlerden olup bir ansiklopedi hükmündedir. Arap dili ve edebiyatı alanına hitap edecek birçok bilgi ihtiva eden bu ansiklopedide firak ve visal/ayrılma ve kavuşma ile ilgi birçok anekdot bulunmaktadır. Öyle ki mezar ve mezarlıklar çevresinde gelişen algı merkezlerinin insana ölüm, öte dünyaya geçiş, kaçınılmaz son, ebedi istirahate çekilme, ebediyete intikal, sonsuzluk veya mutlak karanlık gibi fikirler aşılaması da bu bakımdan anlaşılabilir ve doğal karşılanmaktadır. Tarihin tüm devirlerinde ölüm gerçeği çokça düşünülmüş ve anlamlandırılmaya çalışılmıştır. Gerek semavi dinler gerekse yerel dinlerin ölüm ve ölümle bağlantılı olarak mezara bakışı konusunda birçok ortaklığın bulunması, nihai olarak her insanın ölüm denen sonu yaşayacak olması toplum nezdinde de ölüm ve mezar anlayışını, toplumun iç ve dış dünyasında gündemden düşmeyen bir yere taşımıştır. Ayrıca tarih boyunca birçok toplumun ölümden sonraki hayata inanmasına bağlı olarak, mezarlar ölümden sonraki aşamaya geçişte bir durak ve öte dünyaya açılan bir kapı olarak görülmüştür. Musa Abdi’nin 1429/30 yılları arasında kaleme aldığı Camasbnâme ışığında Çobanın Oğlu (Ignácz Kúnos) ve Mavi Gelin (Oğuz Tansel) isimli iki anlatıdan hareketle Cumhuriyet döneminde yayımlanan hikâye kitapları da mukayeseye dâhil edilmiştir.
