Abstract:
Introduction Anopheles stephensi is a major urban malaria vector in Rajasthan, India, and is responsible for spreading persistent malaria throughout the year. In Rajasthan, An. stephensi is invariably distributed and has three bioforms discriminated based on the number of the ridge on the eggs viz; Type, Mysorensis, and Intermediate. The present study aimed to understand the distribution pattern of these bioforms in rural and urban setups as they also have differences in their malaria transmission capacity. Methods Gravid mosquitoes and the larvae were collected from different habitats of districts of Rajasthan. The gravid females An. stephensi were allowed to lay eggs. These eggs then were subjected to morphometric analysis and counted for the number of ridges for bioform identification. Results About 15000 ± 200 eggs were obtained from ~190 gravid An. stephensi collected from 45 localities (11 Urban & 34 Rural) of eight districts of Rajasthan. Out of which 3569 eggs were subjected to morphometric analysis. Mysorensis bioform (49.7%) was observed to have higher percent over Intermediate (25.5%) and Type (24.6%) bioforms. Mysorensis and Intermediate were found more in rural areas whereas the Type bioform dominated higher in urban areas. Discussion The Mysorensis bioform was found to be dominant throughout the year in all seasons in rural areas. Type bioform preferred indoor breeding places like underground tanks, cement tanks whereas other bioforms preferred outdoor breeding places. Egg size was found to be directly proportional to the number of ridges on the eggs (r = 0.55). No reproductive isolation was observed among these bioforms. Conclusions The Mysorensis bioform is more prominent than other bioforms. Subspecies level understanding helps to plan effective control measures according to the breeding site selection majorly by Type bioform, an efficient vector in this region.

Resumen. Se presenta una lista compilada de 763 especies de plantas acu&aacute;ticas no le&ntilde;osas de los humedales del Escudo de Guayana de Colombia y Venezuela, diferenciadas en cuatro clases estructurales y cuatro bioformas respecto a su posici&oacute;n en el agua. La mayor riqueza 749 especies (el 98% de la lista) ha sido registrada en la Guayana venezolana versus 412 especies (54%) en Colombia. La mitad habita en ambos pa&iacute;ses (52%, 394 especies) y el 15% son end&eacute;micas de Venezuela (113 especies) y de Colombia (12 especies). Predominan las hierbas arraigadas emergentes (91%) de las cuales un 16% ha sido observado al menos en dos ecofases. En t&eacute;rminos espec&iacute;ficos las familias mejor representadas son Poaceae (118 especies), Cyperaceae (75), Eriocaulaceae (57), Lentibulariaceae (48), Rapataceae (32) y Podostemaceae (30). The highest richness 749 species (98%&nbsp; of the list) has been recorded in Venezuelan Guyana versus 412 species (54%) recorded in Colombia. Half inhabit both countries (52% - 394 species) and 15% are endemic to Venezuela with 113 species and Colombia with 12. Emergent rooted herbs predominate (91%) of which almost 16% have been observed in at least two ecophases. The most species richest families are Poaceae (118 species), Cyperaceae (75), Eriocaulaceae (57), Lentibulariaceae (48), Rapataceae (32) and Podostemaceae (30). The genera with the most species are: <em>Utricularia</em> (41), Xyris (22), <em>Paepalanthus</em> (21), <em>Paspalum </em>(21), <em>Eleocharis </em>(20) and <em>Syngonanthus</em> (18). The list accounts for one of the richest aquatic plant floras in the Neotropics that can be related to the diversity of its wetlands, preliminarily classified here in 23 types according to their general morphology and altitudinal position along the gradient of highlands, midlands and lowlands.

Abstract:
Hand gesture recognition applications based on surface electromiographic (sEMG) signals can benefit from on-device execution to achieve faster and more predictable response times and higher energy efficiency. However, deploying state-of-the-art deep learning (DL) models for this task on memory-constrained and battery-operated edge devices, such as wearables, requires a careful optimization process, both at design time, with an appropriate tuning of the DL models’ architectures, and at execution time, where the execution of large and computationally complex models should be avoided unless strictly needed. In this work, we pursue both optimization targets, proposing a novel gesture recognition system that improves upon the state-of-the-art models both in terms of accuracy and efficiency. At the level of DL model architecture, we apply for the first time tiny transformer models (which we call bioformers) to sEMG-based gesture recognition. Through an extensive architecture exploration, we show that our most accurate bioformer achieves a higher classification accuracy on the popular Non-Invasive Adaptive hand Prosthetics Database 6 (Ninapro DB6) dataset compared to the state-of-the-art convolutional neural network (CNN) TEMPONet (+3.1%). When deployed on the RISC-V-based low-power system-on-chip (SoC) GAP8, bioformers that outperform TEMPONet in accuracy consume 7.8×–44.5× less energy per inference. At runtime, we propose a three-level dynamic inference approach that combines a shallow classifier, i.e., a random forest (RF) implementing a simple “rest detector” with two bioformers of different accuracy and complexity, which are sequentially applied to each new input, stopping the classification early for “easy” data. With this mechanism, we obtain a flexible inference system, capable of working in many different operating points in terms of accuracy and average energy consumption. On GAP8, we obtain a further 1.03×–1.35× energy reduction compared to static bioformers at iso-accuracy.

A pesquisa busca identificar como as formas existentes na natureza podem ser “captadas” através de dispositivos de alta tecnologia existentes hoje em dia em nosso aparato tecnológico e estudar possíveis aplicações em diferentes áreas da indústria e da educação. O processo e os resultados dessa pesquisa podem contribuir para a sociedade pois promove a ecoalfabetização, os princípios da Biomimética tanto no design de produtos quanto na estruturação de empresas e negócios e na produção de materiais didáticos. A equipe planeja a criação de uma plataforma de compartilhamento de um acervo digital visando a expansão das fronteiras do laboratório. The effect of two commercially produced biopreparations on the originators of chestnut ink disease in Slovakia &amp;ndash; soil fungi Phytophthora cambivora and Phytophthora cinnamomi was tested in laboratory conditions. Investigations of interrelations between Trichoderma harzianum (Pythium oligandrum) and isolates of Phytophthora sp. obtained from infected tissues of Castanea sativa Mill. proved more important inhibitive effects for Pythium oligandrum (biopreparation Polyversum) efficiency 25.3% (Ph. cambivora) and 26.2% (Ph. cinnamomi). Biopreparation Supresivit (Trichoderma harzianum) reached the efficiency 9.1% for Ph. cambivora isolates and 9.2% for Ph. cinnamomi isolates. Significant effect of the used bioformulas for inhibition of mycelial growth of both Phytophthora sp. and significant difference in the efficacy of two different bioformulas were confirmed statistically. &amp;nbsp;

El comportamiento térmico poblacional del manto de hojas jóvenes (MHJ) que rodea el ápice caulinar de Espeletia grandiflora Mutis ex Humb. &amp; Bonpl., E. incana Cuatrec. y E. hartwegiana Cuatrec., se aparta significativamente de la curva térmica del aire. Las diferencias térmicas aumentan con la altura de localidades de las cordilleras Oriental y Central de Colombia, situadas entre 3300 y 4300 msnm. Las temperaturas nocturnas medias del MHJ se relacionaron inversamente con la altitud y directamente con la humedad relativa. La bioforma característica de las Espeletiinae induce el sobrecalentamiento y el aislamiento térmico del MHJ a las bajas temperaturas nocturnas, debido a la configuración de un gradiente térmico interno altamente diferencial sostenido por el comportamiento térmico del suelo.

Traditionally, dental-shade-guide standards are designated in terms of Munsell hue (H), value (V), and chroma (C). However, ΔE color differences proposed as ADA tolerances for shade guides are in the CIE L*a*b* system. The purpose of this study was to evaluate a new color-difference equation, for estimation of small color differences by Munsell parameters. The published values of the Bioform shade-guide tooth colors determined with a Beckman spectrophotometer were used. Color differences among 276 combinations of the 24 Bioform shade-guide colors were calculated with Eq. 1, with use of the Munsell notation, and also with the CIE L*a*b* equation for ΔE. An estimate of the accuracy of Eq. 1 was 0.41 ΔE units when ΔE CIE was below 4.0. The Vita shade-guide colors were determined with a Beckman spectrophotometer. This data set contained 16 samples, and 120 combinations were used for calculation of color difference. An estimate of the accuracy for this set of data was 0.35 ΔE units when ΔE CIE was less than 4.0. The new color-difference equation provides a means for estimation of AE CIE L*a*b* color difference between dental shades with Munsell notation. This equation will be useful for estimation of small AE CIE L*a*b* values for shade-guide teeth that are designated in terms of Munsell notation.

A comprehensive analysis of alien flora from Dâmbovița county (Romania) was performed both on data from literature and personal observations in the field. The assessment of alien flora was focused on taxonomy, species origin, way of introduction, invasiveness status, introduction period, lifespan, bioforms, characteristic habitats and population size. There were identified 187 alien species of which 70 taxa are invasive and potentially invasive. A number of 138 species have been introduced intentionally in the area of Dâmbovița county, most of them being neophytes. There were identified some hotspots where 11 alien taxa have a high density, as follows: Ambrosia artemisiifolia, Sorghum halepense, Amaranthus retroflexus, Erigeron canadensis, Xanthium orientale subsp. italicum, Reynoutria x bohemica, Solidago canadensis, Reynoutria japonica, Erigeron annuus subsp. annuus, Robinia pseudoacacia, Eriochloa villosa. The largest populations of alien species were recorded in natural and anthropogenic habitats such as: railway embankments, roadsides, vacant lands, abandoned arable lands, wastelands, croplands, riparian habitats and degraded grasslands. The information regarding the habitats, size of populations and spreading of alien species are useful in order to take specific measures for their control.

The purpose of this research consists in the development of a floristic, phytocoenological, ecological, cytogenetic, syndamic, economic and environmentally-friendly study of the marshy meadows located in the subalpine peats of the Vlădeasa Mountains. A total of nine phytocoenological surveys were conducted in the most representative sample areas of the phytocenoses of the Eriophoro vaginati-Sphagnetum recurvi association, in order to find adequate answers to the five proposed objectives of the study. The species found as a result of the floristic inventory were included in an association synthetic table by affinity criteria for coenotaxa: alliance, order, class to which they are subordinate. In the results chapter, the phytocoenoses of the association's meadows were analysed statistically based on tables, histograms, diagrams, by weight in ecological categories of bioforms, phytogeographical elements, cytogenetic elements and the ecological behaviour of the species in relation to edaphic moisture, air temperature, and chemical reaction of the soil. The phytocoenoses of the association, the economic and scientific value and the measures of sustainable management and environmentally friendly management of the subalpine meadows were also studied syndamically. The results thus obtained were compared with two reference works belonging to authors who carried out similar research in different geographical regions. We formulate seven findings in which the results of the research are summarized.

Abstract:
The potential colonization by anchored plants (PCAP) and the potential areas for initial colonization of free floating plants were estimated during the early filling phase for the Yaciretá reservoir. In order to obtain the PCAP, the observed maximum depth of colonization of the anchored macrophytes before impoundment and the hypsographic curves were used. The species inhabiting the pre-impoundment area were classified according to the different bioforms before the inclussion in the analysis. The areal extent of PCAP (from depths between 0-4 m) could reach 275 km² at 76 m above sea level (current water level), whereas at 82 m above sea level (final filling level) the littoral zone will be increased by about 21.5%. The potential area for geophytes was estimated to be 99 km²; 131 km² for root-floating leaved plants and 120 km² for submerged plants, at 76 m above sea level. At 82 m above sea level, the geophytes could reach 271 km². The data for wind frequency, velocity and fetch, together with depth were used to calculate shallow and sheltered areas in which free floating plants could find favourable conditions to initial colonization. Physical and chemical features recorded at eight stations during the early filling phase are discussed in relation to potential plant development.

The main purpose of this work is the development of a phytocenologic, ecological, bioeconomic and eco-protective study of the natural meadows dominated by Festuca nigrescens and Scorzonera rosea in the Biharia Massif, Western Carpathians. In order to achieve the objectives pursued, a number of 11 phytocenologic surveys were performed on the most representative sample surfaces, homogeneous in terms of flora, with the same seasonal requirements and a similar physiognomy. The species from the floristic inventory were recorded in the association table subordinated to the corresponding coenotaxa, alliance, order, class, noted with the corresponding abundance and dominance coefficients. As a result, the phytocenoses of the association Scorzonero roseae Festucetum nigricantis were surveyed on the basis of tables, charts, diagrams, histograms on the distribution of species according to the ecological factors i.e. humidity, temperature, soil chemical reaction, ecological categories of bioforms, phytogeographic and cytogenetic elements. The succession dynamics, the importance, and the sustainable and eco-protective management of these meadows were dealt with in our study. The results thus obtained were compared with four reference papers of some authors who have carried out research in different geographical areas. We found the rare, endangered, endemic species for which suitable household and conservation measures have been proposed.

The aim of our research is to develop a floristic, phytocenological, ecological, cytogenetic, economic, syndynamic and ecoprotective study of the vegetation of the dominant grasslands of Festuca valesiaca Scleicher ex Gaudin and Agrostis capillaris L. in the North-West region of Romania. A numer of 20 phytocenological surveys were carried out in the most representative sample areas in order to find answers to the five objectives pursued. The species from the floristic inventory of the grasslands are included in the Association table according to the cenotic affinity criteria as characteristic and differential entities for the cenotaxons of the alliance, order and class. The phytocoenoses of the grasslands gathered in the association Agrostio-Festucetum valesiacae are statistically analysedin the results based on tables, histograms, dendrograms, diagrams on the distribution of species in ecological bioforms categories, phytogeographic elements, cytogenetic elements, ecological indices or factors: soil moisture, air temperature, soil chemical reaction. The succession dynamics of phytocoeneoses, the economic value and the productive potential, the sustainable and eco-protective management of the grasslands were all studied. The results thus obtained were compared with two reference papers belonging to authors who conducted more recent studies in different geographical regions. Eight conclusions were drafted in which both the results of the research and the original contribution of the authors are summarized.

Abstract:
The ability of Pluronic F127 (PF127) conjugated with tetrapeptide Gly-Arg-Gly-Asp (GRGD) as a sequence of Arg-Gly-Asp (RGD) peptide to form the investigated potential hydrogel (hereafter referred to as 3DG bioformer (3BE)) to produce spheroid, biocompatibility, and cell invasion ability, was assessed in this study. The fibroblast cell line (NIH 3T3), osteoblast cell line (MG-63), and human breast cancer cell line (MCF-7) were cultured in the 3BE hydrogel and commercial product (Matrigel) for comparison. The morphology of spheroid formation was evaluated via optical microscopy. The cell viability was observed through cell counting Kit-8 assay, and cell invasion was investigated via Boyden chamber assay. Analytical results indicated that 3BE exhibited lower spheroid formation than Matrigel. However, the 3BE appeared biocompatible to NIH 3T3, MG-63, and MCF-7 cells. Moreover, cell invasion ability and cell survival rate after invasion through the 3BE was displayed to be comparable to Matrigel. Thus, these findings demonstrate that the 3BE hydrogel has a great potential as an alternative to a three-dimensional cell culture for drug screening applications.

This study was carried out to investigate vascular host plant distribution in the hydrographical basin of the Otăsău River (Vâlcea County, in the Central Southern part of Romania). In order to determine the existing species in this area, fresh and dry material were collected. Specie identification was made based on a repetitive observation of the plants. A taxonomical aid was used to identify the species and classify them into different categories. In this research 368 taxa were investigated and registered as host vascular plants for pests from the studied area. The geoelement analysis shows that Eurasian, European, Central European, Cosmopolite and Circumpolar elements are predominant. Concerning the bioform spectrum the most representative components of the herbaceous layer are hemicryptophytes with 127 taxa. The study reveals that the studied area presents a high floristic diversity of the vascular plants (about 10% of the Romanian flora).

Abstract The aim of this paper is to point out plant species that are of benefit to pollinators, from the territory of the Băiceni (Botoșani county, NE region of Romania). The plant species were analyzed on the basis of the specialized literature as follows: bioform, flowering period, flower grouping, flower color, floral resources, melliferous potential. A number of 106 plant species belonging to 31 botanical families were identified; 41.50% are woody species and 58.50% are herbaceous species. The representative botanical families are: Rosaceae (19.81%), Fabaceae (14.15%) and Asteraceae (14.15%). The majority of the species recorded in the area of study have flowers grouped in inflorescences, blooming in spring and summer and are nectar-polleniferous. The color of the flowers varies from white, yellow, yellow-green to red, blue, purple. These species are an essential resource for pollinators (mainly for the honeybee) and thus contribute to keeping the ecological equilibrium of the ecosystems in the study area and to supporting local beekeeping.

Patterns in the temporal composition, abundance and diversity of the phytoplankton community of the Paraná river prior to and after the initial filling phase of the Yacyretá reservoir are analyzed. The study site is located 220 km downstream from the Yacyretá reservoir and 30 km downstream from the confluence with the Paraguay river. Because both rivers remain separate and unmixed at the study site, we compared the possible effects of the impoundment on both river banks (left and right banks) in hydrological periods with similar duration and magnitude of the low and high water phases. Physical and chemical conditions measured on the right bank (water from the Paraguay river) were similar at both periods (pre and post-impoundment) whereas conductivity, pH and orthophosphate concentration increased on the left bank (water from the High Paraná river and Yacyretá reservoir) after the impoundment. Changes in phytoplankton density and diversity were observed only in samples collected from water flowing from the reservoir (left bank). The density of Chlorophyceae (Chloromonas acidophila, Chlamydomonas leptobasis, Choricystis minor, Chlorella vulgaris, Scenedesmus ecornis, Monoraphidium minutum, M. contortum and M. pusillum) and Cryptophyceae (Rhodomonas minuta, Cryptomonas marssonii and C. ovata) increased while Cyanophyceae (Cylindrospermopsis raciborskii, Raphidiopsis mediterranea and Planktolyngbya subtilis) and Bacillariophyceae (Aulacoseira granulata and its bioforms) decreased compared to previous studies conducted on the left bank of the Paraná river. Phytoplankton collected from the right bank of the river did not differ in pre and post- impoundment samples because they originate from the Paraguay river, which remains relatively unaffected by human activities.

The purpose of this paper is to develop a phytosociological, ecological, cytogenetic, eco-protective and economic study of the vegetation of the wetland ecosystems in the Vlădeasa Massif built by the phytocoenoses of the association Carici echinatae-Sphagnetum (Balázs 1942) Soó 1955. In order to attain the aim and objectives put forward we carried out 10 phytocoenological surveys in the phytocoenoses during the optimal vegetation periods during 2020-2021. The taxonomically inventoried species were included in an association table by the criteria of their belonging to the basic coenotaxa of the association, alliance, order, vegetation class. The outcomes of the phytocoenosis research of the vegetation of the wetland ecosystems were processed, analysed, interpreted based on tables, histograms, diagrams, regarding the numerical and percentage weight of the species in the ecological categories of bioforms, phytogeographic elements (geoelements) and cytogenetic elements. The ecological behaviour of the species confined in ecosystems was also analysed by their relationship with ecological factors, soil moisture, air temperature and chemical reaction of the soil. The current state of ecosystems, potential threats, sustainable conservation of biodiversity, dynamics of phytocenoses, economic and scientific relevance were also subjected to research. The results obtained in the surveyed territory bring concrete answers to the questions formulated and were analysed, compared numerically and as percentage with the data provided by two reference scientific works belonging to authors who independently carried out research with a similar topic in two different geographical regions of the Western Carpathians. The conclusion provides us that the surveyed peatland in the Vlădeasa Massif has a predominantly mesohygrophilic (40%) to hygrophilic (28%), microthermic (46%) to micromesothermic (24%), ionic amphitolerant (38%) to acidophilic (22%) character, dominant species are hemicryptophytes (66.1%) and bryophytes (10.7%) and from the point of view of phytogeographic elements, the circumpolar plants are most representative (32.2%). Polyploid species (62%) dominate peat bogs from Vlădeasa Massif.

This article presents the results of research on pure European beech forests with Symphytum cordatum found in the northern part of the Semenic Mountains (southwestern Romania). The pure European beech forests, included in the association Symphyto cordati-Fagetum, Vida (1959) 1963, exhibit a characteristic forest formation area for the middle mountain layer, identified on districambosol and eutricambosol soil types, deep, moderately acidic, eutrophic, moist, with high trophic levels, rich in mull-type humus, while the river valley terrain has alluvial soils and humiferous deposits. The purpose of the research consists of conducting a floristic, phytocenological, ecological, eco-protective, and bioeconomic study of the Banat beech forests, composed of Fagus sylvatica and Symphytum cordatum, identified in the northern part of the Semenic Mountains-Romania. The phytocenoses of these forests have been analyzed in terms of physiognomy, floristic composition, bioform spectrum, floristic elements, influence of ecological indicators, soil moisture, temperature, chemical soil reaction, and karyological spectrum. The conclusions provide a synthesis of the research results regarding the ecological, eco protective, and bioeconomic aspects associated with the beech forests of the Semenic Mountains, which are mainly composed of the plant species Fagus sylvatica and Symphytum cordatum.

Tomato (Lycoperscion esculentum) is one of the most popular vegetables and a major source of nutrition and income for smallholders in Africa. Thrips-transmitted tospoviruses are among the economically important pathogens of tomatoes that cause significant crop losses worldwide (3). In surveys for Tomato spotted wilt virus (TSWV) in the major tomato production areas of Kenya between March 2010 and January 2012, tomato fruits with chlorotic ring spots on fruits with stem and leaf necrosis were observed frequently. The symptoms were more evident in the dry seasons and disease incidence ranged from 28 to 42%. The pathogen did not react with antiserum specific to TSWV (Agdia Biofords, Ervy, France) in double-antibody sandwich (DAS)-ELISA. Furthermore, the pathogen did not react with antiserum specific to Capsicum chlorosis virus (CaCV), Chrysanthemum stem necrosis virus (CSNV), Groundnut ring spot virus (GRSV), Impatiens necrotic spot virus (INSV), Iris yellow spot virus (IYSV), and Watermelon silver mottle virus (WSMoV) (Agdia Biofords and DSMZ, Germany) in DAS-ELISA, but reacted positively to antiserum specific to Tomato yellow fruit ring virus (TYFRV) (DSMZ, AS0526). The nucleocapsid (N) gene specific primers (TFfor: 5′-ACTCATTAAAATGCATCGTTCT-3′ and TFrev: 5′-CTAAGTAAACACCATGGCTACC-3′ as forward and reverse primers, respectively) were designed by choosing six conserved regions of the N gene sequences of known TYFRV and Tomato yellow ring virus (TYRV) sequences available from GenBank. Using these primers, TYRV infection of tomatoes collected from Loitokitok, Kenya (2.73°S, 37.51°E) was confirmed by reverse transcription (RT)-PCR. PCR products of approximately 912-bp were obtained from six out of 11 symptomatic tomato samples tested, but not from healthy and water controls. Amplicons were gel-purified using QuickClean II Gel Extraction Kit (GenScript, UK) and sequenced using TFfor and TFrev primers. A consensus sequence was generated using Geneious Pro 5.5.6 Software (Biomatters Ltd., Auckland, NZ). The BLAST revealed that the N-gene sequence of the Kenyan tomato isolate (GenBank Accession No. JQ955615) had sequence identity with the Cineraria isolate (98.5%) (Accession No. DQ788693.1) and the Anemone isolate (98.1%) (Accession No. DQ788694.1) of TYRV (4) from Fars Province, Iran; an Alstroemeria isolate (98.4%) (Accession No. HQ154130.1) and two tomato isolates (98.3%) (Accession Nos. HQ154131.1 and AY686718.1) of TYRV from northern Khorasan Province, Iran, and a tomato isolate (98.1%) (Accession No. AJ493270.1) of TYFRV from Varamin, Iran. The Kenyan tomato isolate differed from a TYFRV potato isolate (87.5%) from Iran (Accession No. EU126931.1) (1), a TYRV potato isolate (87.5%) from Iran (Accession No. JF836812.1); a soybean isolate of TYRV (87.4%) from Iran (Accession No. DQ462163.1) (2), and showed significant divergence from that of Polygonum ringspot virus from Italy (81%) (Accession No. EF445397.1). To our knowledge, this is the first report of TYRV infecting tomatoes in Kenya. Reporting biomarkers assessed by routine immunohistochemical (IHC) staining of tissue is broadly used in diagnostic pathology laboratories for patient care. To date, clinical reporting is predominantly qualitative or semi-quantitative. By creating a multitask deep learning framework referred to as DeepLIIF, we present a single-step solution to stain deconvolution/separation, cell segmentation, and quantitative single-cell IHC scoring. Leveraging a unique de novo dataset of co-registered IHC and multiplex immunofluorescence (mpIF) staining of the same slides, we segment and translate low-cost and prevalent IHC slides to more expensive-yet-informative mpIF images, while simultaneously providing the essential ground truth for the superimposed brightfield IHC channels. Moreover, a new nuclear-envelop stain, LAP2beta, with high (&gt;95%) cell coverage is introduced to improve cell delineation/segmentation and protein expression quantification on IHC slides. By simultaneously translating input IHC images to clean/separated mpIF channels and performing cell segmentation/classification, we show that our model trained on clean IHC Ki67 data can generalize to more noisy and artifact-ridden images as well as other nuclear and non-nuclear markers such as CD3, CD8, BCL2, BCL6, MYC, MUM1, CD10, and TP53. We thoroughly evaluate our method on publicly available benchmark datasets as well as against pathologists&#39; semi-quantitative scoring.&nbsp;Trained on IHC, DeepLIIF also generalizes well to H&amp;E images for out-of-the-box nuclear segmentation. DeepLIIF is deployed as a cloud-native platform at https://deepliif.org with Bioformats (150 input formats supported) and MLOps pipeline. Implementations for Torchserve/Dask+Torchscript deployment and auto-scaling via Pulumi available at our GitHub (https://nadeemlab.github.io/DeepLIIF/). ABSTRACT Salivary scavenger receptor cysteine-rich protein gp340 aggregates streptococci and other bacteria as part of the host innate defense system at mucosal surfaces. In this article, we have investigated the properties of fluid-phase gp340 and hydroxylapatite surface-adsorbed gp340 in aggregation and adherence, respectively, of viridans group streptococci (e.g., Streptococcus gordonii and Streptococcus mutans), non-viridans group streptococci (e.g., Streptococcus pyogenes and Streptococcus suis), and oral Actinomyces. Fluid-phase gp340 and surface-phase gp340 bioforms were differentially recognized by streptococci, which formed three phenotypic groupings according to their modes of interaction with gp340. Group I streptococci were aggregated by and adhered to gp340, and group II streptococci preferentially adhered to surface-bound gp340, while group III streptococci were preferentially aggregated by gp340. Each species of Streptococcus tested was found to contain strains representative of at least two of these gp340 interaction groupings. The gp340 interaction modes I to III and sugar specificities of gp340 binding strains coincided for several species. Many gp340 interactions were sialidase sensitive, and each of the interaction modes (I to III) for S. gordonii was correlated with a variant of sialic acid specificity. Adherence of S. gordonii DL1 (Challis) to surface-bound gp340 was dependent upon expression of the sialic acid binding adhesin Hsa. However, aggregation of cells by fluid-phase gp340 was independent of Hsa and involved SspA and SspB (antigen I/II family) polypeptides. Conversely, both gp340-mediated aggregation and adherence of S. mutans NG8 involved antigen I/II polypeptide. Deletion of the mga virulence regulator gene in S. pyogenes resulted in increased cell aggregation by gp340. These results suggest that salivary gp340 recognizes different bacterial receptors according to whether gp340 is present in the fluid phase or surface bound. This phase-associated differential recognition by gp340 of streptococcal species of different levels of virulence and diverse origins may mediate alternative host responses to commensal or pathogenic bacterial phenotypes.

The dendroflora of an urban arboretum (The Botanic Park of Timișoara, Romania), consisting of 193 species, was ecologically characterized as bioforms, phytogeographical elements, and preferences for moisture, temperature, and soil pH. The aim of the research was to determine whether the native ecological requirements of the woody species, along with certain biological and evolutionary traits of them, such as plant life-form and plant family, could serve as tools for explaining and understanding the strategies employed by the urban woody phytocoenoses to acclimate and adapt to an established environment, such as an urban arboretum. The inventoried species are grouped in 111 genera and 45 families. The native and non-native dendroflora share 16 common families. The most representative family both in the native and non-native dendroflora is Rosaceae. The monotypic families are largely present (22.22% in the native dendroflora, and 42.22% in the non-native dendroflora). The plant life-form spectrum is dominated by megaphanerophytes (49%), followed by mesophanerophytes (41%). The chorological spectrum of the native species comprises 16 chorological types and is dominated by Eurasians (32%) and Europeans (30%). The species characteristics of the Pontic-Carpathian space, to which Romania belongs, are rare in the analyzed urban botanical park (4%). The mesophyte, mesothermal and slightly acido-neutrophilous species dominate both the native and non-native dendroflora. In the acclimation process of the non-native dendroflora, 37% of species exceeded their native requirements for moisture, 41% for temperature, and 50% for soil pH. The species requirements for temperature are associated to those for moisture and soil pH. The results show the potential of the analysed woody species to exceed their native requirements within the acclimation and adaption process, and in this process, for the studied temperate site, the plant life-form is important, and also the plant family. The species of the analyzed urban arboretum, both native and non-native, are taxonomically and biogeographically diverse, with specific habitat requirements, suggesting their great ability in acclimating, adapting and resisting.

The present study established a maximum standard for printing quality and developed a preliminary ideal index to print three-dimensional (3D) construct using the Gly-Arg-Gly-Asp (GRGD) peptide modified Pluronic-F127 hydrogel (hereafter defined as 3DG bioformer (3BE)) as bioink. In addition, the biocompatibility of 3BE for 3D printing applications was carefully investigated. For biocompatibility study and ideal printing parameter, we used the formulation of 3BE in three different concentrations (3BE-1: 25%, 3BE-2: 30%, and 3BE-3: 35%). The 3BE hydrogels were printed layer by layer as a cube-like construct with all diameters of the needle head under the same feed (100 mm/s). The printing parameters were determined using combinations of 3BE-1, 3BE-2, and 3BE-3 with three different standard needle sizes (Φ 0.13 mm, Φ 0.33 mm, and Φ 0.9 mm). The printed constructs were photographed and observed using optical microscopy. The cell viability and proliferation were evaluated using Live/Dead assay and immunofluorescence staining. Results showed that a stable of printed line and construct could be generated from the 3BE-3 combinations. Cytotoxicity assay indicated that the 3BE hydrogels possessed well biocompatibility. Bioprinting results also demonstrated that significant cell proliferation in the 3BE-3 combinations was found within three days of printing. Therefore, the study discovered the potential printing parameters of 3BE as bioink to print a stable construct that may also have high biocompatibility for cell encapsulation. This finding could serve as valuable information in creating a functional scaffold for tissue engineering applications.

High-acuity αβT cell receptor (TCR) recognition of peptides bound to major histocompatibility complex molecules (pMHCs) requires mechanosensing, a process whereby piconewton (pN) bioforces exert physical load on αβTCR–pMHC bonds to dynamically alter their lifetimes and foster digital sensitivity cellular signaling. While mechanotransduction is operative for both αβTCRs and pre-TCRs within the αβT lineage, its role in γδT cells is unknown. Here, we show that the human DP10.7 γδTCR specific for the sulfoglycolipid sulfatide bound to CD1d only sustains a significant load and undergoes force-induced structural transitions when the binding interface-distal γδ constant domain (C) module is replaced with that of αβ. The chimeric γδ–αβTCR also signals more robustly than does the wild-type (WT) γδTCR, as revealed by RNA-sequencing (RNA-seq) analysis of TCR-transduced Rag2−/− thymocytes, consistent with structural, single-molecule, and molecular dynamics studies reflective of γδTCRs as mediating recognition via a more canonical immunoglobulin-like receptor interaction. Absence of robust, force-related catch bonds, as well as γδTCR structural transitions, implies that γδT cells do not use mechanosensing for ligand recognition. The αβ T-cell receptor (TCR) on each T lymphocyte mediates exquisite specificity for a particular foreign peptide bound to a major histocompatibility complex molecule (pMHC) displayed on the surface of altered cells. This recognition stimulates protection in the mammalian host against intracellular pathogens, including viruses, and involves piconewton forces that accompany pMHC ligation. Physical forces are generated by T-lymphocyte movement during immune surveillance as well as by cytoskeletal rearrangements at the immunological synapse following cessation of cell migration. The mechanistic explanation for how TCRs distinguish between foreign and self-peptides bound to a given MHC molecule is unclear: peptide residues themselves comprise few of the TCR contacts on the pMHC, and pathogen-derived peptides are scant among myriad self-peptides bound to the same MHC class arrayed on infected cells. Using optical tweezers and DNA tether spacer technology that permit piconewton force application and nanometer scale precision, we have determined how bioforces relate to self versus nonself discrimination. Single-molecule analyses involving isolated αβ-heterodimers as well as complete TCR complexes on T lymphocytes reveal that the FG loop in the β-subunit constant domain allosterically controls both the variable domain module’s catch bond lifetime and peptide discrimination via force-driven conformational transition. In contrast to integrins, the TCR interrogates its ligand via a strong force-loaded state with release through a weakened, extended state. Our work defines a key element of TCR mechanotransduction, explaining why the FG loop structure evolved for adaptive immunity in αβ but not γδTCRs or immunoglobulins.

OBJECTIVE: The aim of this study was to evaluate the microhardness in pressing layers of prefabricated modified acrylic resin teeth. METHODS: Lower first molar teeth with two (Biotone(r) IPN and Bioform(r)) or three pressing layers (Artiplus(r), Trilux(r) Eurovipi, and Natusdent(r)) were hemisected in a bucco-lingual plane and embedded in a self-curing acrylic resin (n =10). Specimens were then ground flat and polished using 400, 600 and 1200 aluminium oxide paper in a rotatory polisher. In each pressing layer, microhardness was measured using a Knoop indenter in three different locations spaced 300 µm apart under a 10-g load, applied for 5 sec. RESULTS: Analysis of variance and Tukey's tests demonstrated that there was no difference in microhardness in the first layer of the teeth analyzed (p = 0.355), whereas in the second layer, the brand Artiplus(r) showed higher values when compared to the brand Natusdent(r) (p = 0.018). For the third layer, the brands Artiplus(r) and Trilux(r) Eurovipi revealed higher microhardness when compared to Natusdent(r) teeth (p &lt; 0.001). CONCLUSION: For the outer most superficial layer of the artificial teeth, the microhardness of the different brands was similar, while differences were noted for the second and third layers among the artificial teeth, with Artiplus(r) teeth showing higher microhardness than Natusdent(r).

Age-related sarcopenia results in frailty and decreased mobility, which are associated with increased falls and long-term disability in the elderly. Given the global increase in lifespan, sarcopenia is a growing, unmet medical need. This report aims to systematically characterize muscle aging in preclinical models, which may facilitate the development of sarcopenia therapies. Naïve rats and mice were subjected to noninvasive micro X-ray computed tomography (micro-CT) imaging, terminal in situ muscle function characterizations, and ATPase-based myofiber analysis. We developed a Definiens (Parsippany, NJ)-based algorithm to automate micro-CT image analysis, which facilitates longitudinal in vivo muscle mass analysis. We report development and characterization of translational in situ skeletal muscle performance assay systems in rat and mouse. The systems incorporate a custom-designed animal assay stage, resulting in enhanced force measurement precision, and LabVIEW (National Instruments, Austin, TX)-based algorithms to support automated data acquisition and data analysis. We used ATPase-staining techniques for myofibers to characterize fiber subtypes and distribution. Major parameters contributing to muscle performance were identified using data mining and integration, enabled by Labmatrix (BioFortis, Columbia, MD). These technologies enabled the systemic and accurate monitoring of muscle aging from a large number of animals. The data indicated that longitudinal muscle cross-sectional area measurement effectively monitors change of muscle mass and function during aging. Furthermore, the data showed that muscle performance during aging is also modulated by myofiber remodeling factors, such as changes in myofiber distribution patterns and changes in fiber shape, which affect myofiber interaction. This in vivo muscle assay platform has been applied to support identification and validation of novel targets for the treatment of sarcopenia.

Nature is considered from the point of view of a formative source, the resource of which can be used in the design and construction of environmental objects in order to create new bioforms, eco-materials and structures to reduce energy consumption while maintaining comfort. The push for sustainable design has intensified with the launch of the Building Research Institute’s Environmental Assessment Method (BREEAM). It introduces the first green building rating system. In addition, the United States Green Building Council (USGBC) has formulated new criteria aimed at improving the environmental performance of buildings through its Leadership in Energy and Environmental Design (LEED). In Russia, the existing “Green” standards create practices for solving broader problems of sustainable development, such as zero-energy construction concepts, living, regenerative and restorative construction concepts that improve the natural environment or simulate natural processes. This will initiate the search for new design strategies using the tools of immersive design concepts for the development of socially-oriented projects aimed at supporting the global trend of total environmental design. The article analyzes the artificial and natural elements of the exposition complexes from the perspective of ecological design. In Russia, the existing “Green” standards create practices for solving broader problems of sustainable development, such as zero-energy construction concepts, living, regenerative and restorative construction concepts that improve the natural environment or simulate natural processes. Introduction: The cause–effect relationships between microbiota composition changes and type 2 diabetes (T2D) are complex, likely involving two-way interactions, and require further elucidation. Few studies have examined the interactions of antidiabetic drugs with the gut microbiota. This study’s goal was to evaluate the gut microbiota of patients with type 2 diabetes at first diagnosis and again after 12 weeks of taking oral antidiabetic drugs. Methods: We performed a fecal microbiota analysis of adult patients who recently received a T2D diagnosis and healthy adults. We compared the microbiota compositions between the T2D patients and healthy controls; we also evaluated changes from baseline to 12 weeks of treatment in the total group receiving oral antidiabetics, as well as in the subgroups receiving metformin and linagliptin. Results: The alpha diversity and beta diversity indices were different at baseline between patients with type 2 diabetes and healthy controls. The LEfSe analysis showed that, at the genus level, the Lactobacillus, Rothia, Collinsella, and Eubacterium genera increased in relative abundance in the T2D group while, at the species level, the Rothia mucilaginosa, Collinsella aerofaciens, and Eubacterium bioforme strains were found to be dominant in the T2D group. Faecalibacterium at the genus level and Faecalibacterium prausnitzii at the strain level increased in relative abundance in the T2D group after 12 weeks. After 12 weeks of intervention, the alpha diversity indices were significantly lower in the T2D group compared to the control group. At the end of the 12th week, the Gemmiger and Collinsella genera were dominant in the T2D group, with Gemmiger formicilis and Collinsella aerofaciens being dominant at the species level; in the control group, Bacteroides and Alistipes were dominant at the genus level, and Prevotella stercorea and Alistipes finegoldii were dominant. There was no difference in the LEfSe analysis results between baseline and 12 weeks of linagliptin treatment. At the strain level, Gemmiger formicilis, Ruminococcus bromii, Rothia mucilaginosa, and Lactobacillus ruminis were predominant at the start of metformin treatment; however, after 12 weeks, Collinsella aerofaciens became predominant. Conclusions: We report that there is a substantial change in the composition of the gut microbiota in patients with T2D. Oral antidiabetic treatments, especially metformin, have some beneficial effects on microbiota composition. Few studies have explored the interaction of antidiabetic drugs with the gut microbiota; further research will elucidate the clinical impact of these changes in gut microbiota composition in diabetes.

Adrian Tchaikovsky is renowned for exploring animal consciousness and non-human perspectives in his works. His 2017 novel Dogs of War exemplifies the dominance of anthropocentrism within the novel’s transhumanist context, wherein genetically engineered non-human bioforms are exploited by humans. This study argues the way humans perceive and treat non-human subjects in the novel, which are reduced to “disposable bodies” (Braidotti, 2013, p.15), extends the reach of human-centred ideology into transhumanism. Therefore, the termination of the bioforms once they are deemed obsolete demonstrates a utilitarian view of life within transhumanist thought. Building upon these arguments, this study examines whether genetically engineered non-human entities in the novel are entitled to the same rights as their human creators, who design and command them in the pursuit of technological and military advancements. By extension, this study proposes Tchaikovsky's portrayal of bioform animals resonating with Donna Haraway’s concept of the “cyborg,” which challenges the traditional concept of humans by attributing agency and sentience to these beings. The study seeks to illuminate the complex interplay between human and non-human agency, thereby questioning the established hierarchy of beings in the context of transhumanist thought.

The article is devoted to the actual problem in design projection of children’s choreographic costumes based on a bionic creative source. The analysis aimed to identify the scope of use of vegetables as a creative source for the production of fashion collections. It has been found that designers use vegetables for making clothes, for sketching the models of clothes, as well as a source of inspiration for form, decoration and colour choices. A figurative and stylistic analysis of bionic creative sources and the transformation of plant images into stage costumes for a children’s dance show was carried out in order to come up with models of stage costumes. During the process of costume design, the following principles were adopted: imitation of the bioform in separate elements of the costume, analogousness of silhouette solutions to natural bioforms, bionic design projection with the identification of functionality, and figurative and associative expressiveness of the costume. As a final step, the scenario of performance, as well as the requirements of the children and the creative source were considered when designing a collection of children’s stage costumes.

Abstract BioForms integrates sacrificial formworks, agent-based computational algorithms and biological growth in the generation of biodegradable internal wall panel systems. These wall panel systems are intended to minimize material waste, utilize local botany and generate a symbiosis between the artificially made and the naturally grown. This is achieved by utilizing local waste as a structural compressive core, mycelium as the binder, and recycled pellets as the architectural skin. Leveraging mycelium’s structural, acoustic and thermal properties, this exploration delves into unique methods of incorporating fungi and waste into architectural construction. The motivations for this research stem from the need to address the building industry’s contribution to climate change, by considering the lifecycle of our materials. BioForms aims to retrofit existing buildings by replacing foam insulation and MDF (medium-density fiberboard) wall panels with biodegradable and recyclable 3D-printed skins embedded with a mycelium core. Analysing mycelium’s reaction to BioForms I, the second iteration, BioForms II, evolves in design complexity and materiality. BioForms II explored robotically fabricated wood-based polylactic acid plastic (PLA) composite materials. Within the second iteration of this research stream, mycelia was both embedded within the compressed fabricated skins and on the external surface. Whilst BioForms explored the generation of biodegradable wall panel systems, the broader aims of this research is aimed at infiltrating biological matter into human-occupied spaces, completely omitting the use of synthetic building materials within the construction industry and advancing the architects relationship to nature in the generation of form.

The present study is focused on a volcanic mountain area of Maramures (Gutai Mountains) with specific vegetation that requires identification and analysis. The flora of these meadows has a natural character. The phytosociological study was performed and there was found vegetal associations belonging to classes Nardo - Callunetea Prsg. 1949, Molinio - Arrhenatheretea R. Tx. 1937 and Vaccinio-Piceetea Br.-Bl. in Br.-Bl. et al. 1939. The revealing species of the associations indicate a relatively poor floristic composition; great coverage has Nardus stricta, mainly due to acid substrate and to the bryophytes layer developed. Each vegetal association is described by analysis of the bioforms, of the floristic elements, geoelements, ecological indices and economic value. In all the vegetal associations hemicryptophytes exceeds other categories of bioforms, and considering the geoelements, the greatest rate is held by the Eurasian elements. From the point of view of the ecological character prevails mesophilic plants, micro-mesotherms and euritherms, euriionics and acidophilus. Meadows have a very low nutritive value and a low productivity, especially because of excessive grazing. Mosaic distribution of phytocoenosis is due to the uneven distribution of the climatic factors depending on the exhibition and on the degree of slope inclination.

AbstractSurficial sediments on the seafloor from passive continental margins can provide insight into recent Late Quaternary sedimentary dynamics acting over offshore sedimentary systems. This work focuses on the study of some particular ferruginous tubular structures resembling bioforms (FTB) located in the distal Galician Continental Margin (NW Iberian Margin) at water depths between ~ 1550 and ~ 2200 m. The characterisation of these structures made it possible to study in depth their formation environment and subsequent sedimentary evolution during the Late Pleistocene and Holocene. The FTB consist of goethite with a framboidal texture. They were interpreted as formed by an initial pyrite precipitation in reducing microenvironments conditioned by the activity of sediment-dwelling organisms during the early diagenesis. This is followed by the oxidation of pyrite by a combination of hydrothermal fluids and erosional processes, which triggers the formation of the framboidal oxyhydroxides. The data allowed obtaining a comprehensive understanding of the environmental context and the significance of these ferruginous tubules, as there are no previous studies in the scientific literature that describe these structures in a source-to-sink sedimentary system.
