Full textEnvironmental biotechnologies can bring both environmental and economical benefits, playing an especially important role in eliminating pollution and improving the environmental quality. The present thesis deals with the development and application of two environmental biotechnologies: microbial flocculant (MBF) and conductometric biosensor. A MBF named TJ-F1 of strong flocculating activity, produced by strain TJ-1 identified as Proteus mirabilis, was investigated with regards to its production, characterization and applications. The flocculating activity of TJ-F1 obtained under the optimized culture conditions was up to 93. 13%. About 1. 33 g of the purified solid MBF with a molecular weight of 1. 2×105 Da could be recovered from 1. 0 L of fermentation broth. Chemical analysis of TJ-F1 indicated that it contains proteins (30. 9%, w/w) and acid polysaccharide (63. 1%, w/w). Scanning electron microscopy image of the purified solid-state TJ-F1 showed that it has a crystal-linear structure. Spectroscopic analysis of TJ-F1 by Fourier-transform infrared spectrometry indicated the presence of hydrogen bonds, hydroxyl, amino and carboxyl groups which are the preferred ones for the flocculation process. The flocculation mechanism of TJ-F1 was discussed according to its physical & chemical characteristics and to the Zeta potential variation in the flocculation system. The successful application to the dewatering of activated sludges and the dye removal from water validated its excellent flocculating performance. A cost-effective culture medium using wastewaters as nutriments was also developed, which can not only cut down the production cost of TJ-F1, but also save the treatment cost of the wastewaters. Two conductometric biosensors respectively for phosphate and nitrite analysis were developed. The process parameters for the fabrication of the biosensors and various experimental variables were investigated with regards to their influence on the sensitivity, the detection limit, the dynamic range, the operation and storage stability. The biosensor for phosphate analysis showed a fast response (about 10 s) to phosphate with a detection limit of 1. 0 μM, and two linear ranges of 1. 0-20 μM and 20-400 μM. The biosensor for nitrite analysis showed a similar fast response to nitrite with a detection limit of 0. 05 μM and a linear range of 0. 2-120 μM. Both of them showed satisfactory reproducibility and good stability. For both biosensors, no obvious interference from other ionic species familiar in natural waters was detected. The application experiments showed that the biosensors are suitable for use in real water samples

This new more complex innovation, which requires environmental and competitiveness performance for its development and diffusion, occupies a more and more central place in the firm. We have in this work particularly analyzed the effectiveness of voluntary approaches (namely the European Ecolabel certification) on the innovation of products in the firm and, at the same time, identified the key determinants to adoption of this ecolabel. We focused our analysis on the detergents and maintenance-products sector in France. This study is a part of an evolutionary approach and considers the process of technical change (development and diffusion of European Ecolabel certified product) to be a complex and uncertain process incorporating different factors in co-evolution, beyond a limited conception of the role of environmental policies. The case study shows the role of the firm strategy in the process of technical change. The empirical analysis confirms the role of this determinant (strategy of suppliers of bimolecular substances) and presents, moreover, the importance of regulation (REACH…), the benefits of certification (brand image) and demand as key factors in adoption of this ecolabel certification. Indeed, although this voluntary policy drives some firms to innovation, the environmental performance and accession by most firms in this sector remains a challenge. In the current context, firms which have used biomolecular substances, under suppliers' strategic influence, have developed green products but not systematically European Ecolabel certified products

Full textDHs are a class of synthetic anionic clays whose structure is based on brucite-like layers Mg(OH)2 inwhich some of the divalent cations have been replaced by trivalent ions giving positively-charged sheets.This charge is balanced by intercalation of anions in the hydrated interlayer regions. The identities andratios of the di- and trivalent cations and the interlayer anion may be varied over a wide range, giving rise toa large class of isostructural materials. The parent material of this class is the naturally occurring mineralhydrotalcite and LDHs are consequently also known as hydrotalcite-like materials. Although the basicfeatures of the structure are well understood, detailed structural aspects have been the subject of somecontroversy in the literature. In this thesis, we have selected two types of LDH, MgAl and ZnAl, which havebeen widely introduced in various applications, such as sorption of molecules of biological interest (enzymeand drug) and the development of electrodes. The specificity of this work lies on the immobilization of amodel enzyme, lactate dehydrogenase in both matrices as well as an anti-bacterial drug, berberine, inorder to study the interactions between these two biomolecules and the introduced LDH phase and tobetter address their challenges of applications in the medical field. Second, we have tried to study the twophases mentioned above more and more accurately in terms of structure, morphology and electrochemicalprofile in order to use them as electrode materials for microbial fuel cell device

Full textA photosensitizer is a compound capable of capturing photons of light and transferring this energy to neighboring molecules. In particular, molecular oxygen can absorb this energy to lead to the formation of singlet oxygen, a very reactive oxygen species that can react with a large variety of organic compounds, cellular constituents and environmental pollutants. With a singlet oxygen quantum yield near unity in all of the most common solvents, phenalenone is an excellent photosensitizer which has the advantage of facile synthetic accessibility. However, chemical modification of this molecule drastically impacts the singlet oxygen quantum yield, thus limiting its use. Hence, during this work, we have proposed a series of phenalenone functionalizations with various functional groups like azide, propargyle, carboxylic acid, amine, alcohol and much more, enabling the expansion of its use without any loss of efficacy. In a second part, some of these derivatives were used for new applications of phenalenone. It was grafted on lignocellulosic materials (wood, paper) and siliceous materials (glass, sand). Antifungal properties of grafted wood are currently under study, while some preliminary tests of tebuconazole photodegradation of grafted sand in water gave promising results leaving open the possibility to use this material in water photodepollution. Phenalenone was also linked to one or two stearic acid chains and analysis of these derivatives highlighted their capacity to form supramolecular structures. Lastly, phenalenone was coupled to triazolium salts, and several derivatives have shown excellent photo-inhibition against microbial growth, in particular against Gram positive strains

Full textChlordecone is a synthetic organochlorine pesticide bearing a perchlorinated bis-homocubane structure. It was extensively used in banana plantations of the French West Indies (FWI) from 1972 to 1993. Even if its use has been prohibited in the FWI more than two decades ago, chlordecone is present in all environmental compartments thus contaminating local food chain. Chronic exposure increases risk of cancers and induces reproductive and developmental disorders. Due to its environmental persistence, chlordecone pollution in soils and water is estimated to last for numerous decades or even centuries. Previous works reported on chlordecone biodegradation research led to low efficient transformations: one or two chlorine atoms at most lost and bis-homocubane core structure untouched. Recently in Genoscope, bacterial consortia and isolated Citrobacter strains were shown to mediate complete transformation of chlordecone into three main metabolites families within several weeks of action.In the present work, this efficient microbiological transformation was investigated through complementary perspectives: structure elucidation of the main metabolites produced by bacteria-mediated transformations and study of their formation in order to approach the mechanistic pathways involved. Biomimetic transformation protocols were developed and optimized to produce several milligrams of each main metabolite. After successful isolation, the main metabolites were studied using mass spectrometry and nuclear magnetic resonance. The structures of three hydrochlordecones and two polychloroindenes (PCI) were elucidated. Even if previously postulated as transformation product of chlordecone, the chlorinated indene core has been unambiguously demonstrated for the first time. The originality of the work also lies in the novel structure identification of four chlordecone transformation products bearing an unexpected polychloroindenecarboxylic acid (PCICA) structure. These results shed new lights on numerous earlier unclear results. Degradation experiments were run on the purified metabolites to explore their role in the chemical and microbiological transformations as intermediate or dead-end products. A general transformation mechanistic pathway that accounts for the formation of all three main metabolite families was proposed. PCI and PCICA metabolites resulting from ring-opening of the chlordecone bishomocubane core show a peculiar indene structure. Electrochemical study demonstrated the necessity of strong reductive conditions and presence of a catalyst such as vitamin B12 in order to favour PCI and PCICA formation. Finally, to better understand the transformation dynamics, carbon isotope fractionation during biotic and abiotic transformations were measured.In this work, several chlordecone biological transformation products were produced, isolated and fully characterized. These results open the way towards new perspectives: evaluation of metabolites toxicity, development of reliable quantification methods for monitoring chlordecone transformations in all types of matrix, including environmental samples. Last but not least, chlordecone metabolites biodegradation will be investigated in order to pursue complete biomineralization of chlordecone, pre-requisite for any bioremediation attempts in the FWI
