Abstract:
The electrospinning method has been used successfully to make polyvinylpyrrolidone nanofiber containing Anredera cordifolia leaves (BLE). The research methods used were qualitative and pure experiment method. Polyvinilpirolidone nanofibers containing BLE were prepared with three mass variations of the polyvinylpyrrolidone (% w/w), namely 12%, 10%, and 8% w/w, respectively. The results of the macroscopic photo show that the fiber structure looks white for PVP nanofibers and yellow for PVP/BLE nanofibers. The fiber morphology was analyzed using SEM and the results showed that PVP and all PVP/BLE nanofibers were like a continuous strand of crossbars with a diameter of 590 – 1190 nm. The decrease in the concentration of the PVP polymer led to a reduction in the diameter of the resulting nanofibers. The coefficients of variance (ε), of the PVP, BLE1, BLE2, and BLE3 nanofibers were 0.06, 0.09, 0.11, and 1.22, respectively. The physicochemical structure of the nanofibers was evaluated using XRD and FTIR. The chemical analysis (FTIR) showed that there was a molecular interaction between Anredera cordifolia leaves extract and polyvinylpyrrolidone in the form of hydrogen bonds. The physics analysis (XRD) shows the effect of the electrospinning process, which is to change the structure of BLE crystals to semi crystals. The application of PVP/BLE nanofiber for wounds dressing

Abstract:
This study aims to determine the bioactivity of L.domesticum as cosmetic ingredient. The extraction was prepared by maceration with ethanol 96 % and ethyl acetate to obtain ethanolic extract of fruit flesh (BLE), ethanolic extract of fruit peel (KLE), ethyl acetate extract of fruit flesh (KLE) and fruit peel (KLEA). Then, determination of compounds was conducted by thin layer chromatography (TLC) and analysed of total phenolic and flavonoid content with colorimetric method. The antioxidant capacity was measured using radical scavenging assay of 1,2-diphenyl-2-picrylhydrazyl (DPPH), β-carotene bleaching assay (BCB) and ferric reducing antioxidant power assay (FRAP). The inhibitory effect of skin degradation enzymes (anti-aging) was carried out using elastase and collagenase assay. The tyrosinase inhibitory effect was determined using mushroom tyrosinase. Based on TLC result, extract of L.domesticum containing phenolic, flavonoids, terpenoids and steroids compounds. The largest of total flavonoid content was obtained in KLE (4,76±0,26 %b/b EQ) and phenolic total content in BLEA (6,4±0,15 %b/b EG). Sequence of antioxidant activity measured using DPPH method was BLEA> KLE> BLE> KLEA; while using BCB method and FRAP method were respectively KLEA>BLEA> KLE> BLE; and BLEA> KLE> BLE> KLEA. KLE and KLEA extracts showed the most potent to remaining activity of the enzyme elastase and collagenase. In addition, BLE extract has the higest tyrosinase inhibition than others. Based on these results it can be concluded that the extracts of the fruit flesh and fruit peel of L. domesticum can be used as a cosmetic active ingredient for anti-aging and anti-tyrosinase.

Abstract:

Abstract:
Historical surface irrigation of pastures in the Dutch province of Noord-Brabant: visible traces of a failed experiment In the second half of the 19th century, some major changes occurred in the water management of the eastern and southern provinces of the Netherlands. Unlike the low-lying western parts of the Netherlands which were characterised by polders and had a long history of formal water boards, the higher eastern and southern parts of the Netherlands were characterized by brook systems and sandy soils, and had no centralised water boards until 1850. From the 1850s onward, water boards were introduced in these higher regions as well, and agronomical scientists and organisations like the Nederlandse Heidemaatschappij endeavoured for the modernisation of agricultural water management. One of their priorities was the introduction of modern forms of surface irrigation of pastures, in order to increase crop yields. In various places modern irrigation systems were constructed. From the 1900s onward, these systems were abandoned due to the introduction of new chemical fertilizers, among others. This article describes the construction and abandonment of one of these modern irrigation systems that was located in Liempde, in the province of Noord-Brabant. The local farmers were not interested in the new technique, and within a few years the system was transformed in a poplar plantation. Nowadays, the area is part of a nature reserve. Nonetheless, the global layout of the irrigation system is still visible.

Abstract:
Abstract The ‘contemplative turn’ in today’s theology had already a predecessor in the theology of O. Noordmans. That theology, developed in the first half of the 20th century, was (as Noordmans himself put it) ‘embedded’ in ‘the church as mother church’, as a ‘womb of piety’. This article describes how that can be seen in Noordmans’s appreciation of the pastoral character of Reformed dogmatics and in his sharp distinction between ‘church’ and ‘school’. The church, according to him, is not a place for doing philosophy, rather for proclaiming the Gospel, and the dogma serves only to keep the preacher on the right path. In this context, special attention is paid to Noordmans’s doctrine of creation, as summarized in his statement: ‘Creating is not forming, but dividing’. In other words: creation cannot be a (philopsophical) theme in itself; rather it is the beginning of a story: the story of the procession towards the cross.

Abstract:
This article is about the dynamics of gender in workplaces that are undergoing changes in organization. More precisely it is about the instability in femininity and masculinity and the links between the construction of gender and the construction of the modern organizations. On a general level the traditional gender order is based on stability, that is to say that men and women are seen as very different, even as each other's antithesis, and with different values. This kind of gender stability works as a strong restorative mechanism in the organization of local workplaces since the resulting attempts to maintain segregation and hierarchy are directly contradictory to the modern organizational concepts that aim for integration and de-centralization. Gender order in such organizations does not only provide stability, at the same time we can also see dynamic processes at work whereby parts of the contents of the concepts masculinity and femininity are transformed. For example, social competence is often seen as a "female" characteristic or ability, but is more and more becoming a "male" competence, something that men should have as well. The construction of masculinity in local organizations is thus modified in accordance with what is important to modern organizational concepts. In other words, the dynamics of gender construction makes it possible to implement modern organization concepts without changing the gender order too much.

Abstract:
The success of trabeculectomy surgery depends on the longevity of the filtering bleb. Bleb failure can be categorized into two types: the scarred bleb with high intraocular pressure or the over-filtering and leaking bleb with low intraocular pressure. Bleb scarring is an insidious process over time as a consequence of excessive subconjunctival fibrosis. Timely recognition and early intervention utilizing a stepped-up approach are important for resuscitating the bleb and salvaging the trabeculectomy. Over-filtration and leaky bleb on the other end of the spectrum lead to failure of optimal bleb function and require a different management approach. This review discusses in detail various surgical techniques to revive dysfunctional blebs with a special focus on bleb needling. Timely identification and multifaceted management of bleb-related complications is the key to ultimately improving long-term success rates and patient outcomes.

Abstract:
Background: The intricate hemodynamic mechanisms of thrombosis in the bleb and/or dome of cerebral aneurysms remain unresolved. We encountered a unique case where the bleb of an unruptured internal carotid-posterior communicating artery (IC-PC) aneurysm underwent thrombosis over 7 years. Complete spontaneous thrombosis of a bleb in an unruptured cerebral aneurysm has not been previously reported. Therefore, a hemodynamic evaluation using computational fluid dynamics (CFD) analysis was conducted to examine the thrombotic development within the bleb of this aneurysm. Case Description: We observed a case in which thrombosis led to the disappearance of a bleb in the dome of an unruptured IC-PC aneurysm over 7 years. CFD analysis was employed to investigate the hemodynamics of bleb thrombosis and the thrombosed bleb-neck regions of the dome in this IC-PC aneurysm. The reduction and disappearance of the bleb were associated with a decreased flow rate within the bleb, reduced magnitude of wall shear stress (WSSm), a lower WSSm ratio between the bleb and dome, increased vector direction of wall shear stress (WSSv), and discrete streamlines entering the bleb-neck region, resulting in stasis and subsequent thrombosis within the bleb. Seven years later, the dome region corresponding to the thrombosed bleb-neck exhibited localized areas with low WSSm and high WSSv along the dome wall. Conclusion: Hemodynamically, spontaneously thrombosed bleb and thrombosed post-bleb-neck dome walls were characterized by low WSSm and high WSSv. These findings underscore the importance of CFD analysis in predicting thrombotic events in cerebral aneurysms, which can inform better clinical management strategies.

Abstract:
Aim When assessing bleeding risk in patients with atrial fibrillation (AF), risk stratification is often based on the baseline risks. We aimed to investigate changes in bleeding risk factors and alterations in the HAS-BLED score in AF patients. We hypothesized that a follow-up HAS-BLED score and the ‘delta HAS-BLED score’ (reflecting the change in score between baseline and follow-up) would be more predictive of major bleeding, when compared with baseline HAS-BLED score. Methods and Results A total of 19,566 AF patients receiving warfarin and baseline HAS-BLED score ≤2 were studied. After a follow-up of 93,783 person-years, 3,032 major bleeds were observed. The accuracies of baseline, follow-up, and delta HAS-BLED scores as well as cumulative numbers of baseline modifiable bleeding risk factors, in predicting subsequent major bleeding, were analysed and compared. The mean baseline HAS-BLED score was 1.43 which increased to 2.45 with a mean ‘delta HAS-BLED score’ of 1.03. The HAS-BLED score remained unchanged in 38.2% of patients. Of those patients experiencing major bleeding, 76.6% had a ‘delta HAS-BLED’ score ≥1, compared with only 59.0% in patients without major bleeding (p < 0.001). For prediction of major bleeding, AUC was significantly higher for the follow-up HAS-BLED (0.63) or delta HAS-BLED (0.62) scores, compared with baseline HAS-BLED score (0.54). The number of baseline modifiable risk factors was non-significantly predictive of major bleeding (AUC = 0.49). Conclusion In this ‘real-world’ nationwide AF cohort, follow-up HAS-BLED or ‘delta HAS-BLED score’ was more predictive of major bleeding compared with baseline HAS-BLED or the simple determination of ‘modifiable bleeding risk factors’. Bleeding risk in AF is a dynamic process and use of the HAS-BLED score should be to ‘flag up’ patients potentially at risk for more regular review and follow-up, and to address the modifiable bleeding risk factors during follow-up visits.

Abstract:
Purpose: To assess the ability of a noncontact optical coherence tomography to evaluate the morphological features of filtering blebs one year after glaucoma surgery.Design: Prospective study. Methods: Eighteen patients (18 eyes) with diagnosed primary open-angle glaucoma (POAG) assigned for trabeculectomy were included in the 12-month study carried out in the Eye clinic of the Lithuanian University of Health Sciences. All participants underwent trabeculectomy with 5-fluorouracil (5-FU). Bleb function was considered to be successful if the intraocular pressure (IOP) was ≤ 18 mmHg without glaucoma medications and a limited success if: 18 < IOP ≤ 21 mmHg with or without glaucoma medications at 12 months after surgery. The filtering blebs were imaged by anterior segment optical coherence tomography (AS-OCT) to evaluate the bleb wall reflectivity and measured bleb structures 12 months after trabeculectomy. Level of significance: p < 0.05 was considered significant. Results: The mean preoperative IOP was 25.7 (6.5) mmHg and the mean number of topical glaucoma medications was 3.0 (1.2). After surgery the mean IOP was 13.8 (3.4) mmHg and glaucoma medication was 0.3 (1.0) (Wilcoxon test, p < 0.001). Analyzing bleb morphology and bleb function it was found that with uniform wall reflectivity 0 out of 3 eyes (0%) had successful bleb function and with multiform wall reflectivity 14 out of 15 eyes (93.3%) had successful bleb function 12 months after surgery (p = 0.005).We found positive correlation between IOP changes and bleb wall thickness, height of internal fluid-filled cavity (bleb height) and total bleb height (r = 0.875, 0.897, 0.939, p < 0.001).Conclusion: AS-OCT is a useful device to assess the structure of the filtering bleb. Larger internal fluid-filled cavity, total bleb height, bigger bleb wall thickness and multiform bleb wall reflectivity were found to be good indicators of bleb function.

Abstract:
Membrane blebs are specialized cellular protrusions that play diverse roles in processes such as cell division and cell migration. Blebbing can be divided into three distinct phases: bleb nucleation, bleb growth, and bleb retraction. Following nucleation and bleb growth, the actin cortex, comprising actin, cross-linking proteins, and nonmuscle myosin II (MII), begins to reassemble on the membrane. MII then drives the final phase, bleb retraction, which results in reintegration of the bleb into the cellular cortex. There are three MII paralogues with distinct biophysical properties expressed in mammalian cells: MIIA, MIIB, and MIIC. Here we show that MIIA specifically drives bleb retraction during cytokinesis. The motor domain and regulation of the nonhelical tailpiece of MIIA both contribute to its ability to drive bleb retraction. These experiments have also revealed a relationship between faster turnover of MIIA at the cortex and its ability to drive bleb retraction.

Abstract:
Blebs are involved in various biological processes such as cell migration, cytokinesis, and apoptosis. While the expansion of blebs is largely an intracellular pressure-driven process, the retraction of blebs is believed to be driven by RhoA activation that leads to the reassembly of the actomyosin cortex at the bleb membrane. However, it is still poorly understood how RhoA is activated at the bleb membrane. Here, we provide evidence demonstrating that myosin II–interacting guanine nucleotide exchange factor (MYOGEF) is implicated in bleb retraction via stimulating RhoA activation and the reassembly of an actomyosin network at the bleb membrane during bleb retraction. Interaction of MYOGEF with ezrin, a well-known regulator of bleb retraction, is required for MYOGEF localization to retracting blebs. Notably, knockout of MYOGEF or ezrin not only disrupts RhoA activation at the bleb membrane, but also interferes with nonmuscle myosin II localization and activation, as well as actin polymerization in retracting blebs. Importantly, MYOGEF knockout slows down bleb retraction. We propose that ezrin interacts with MYOGEF and recruits it to retracting blebs, where MYOGEF activates RhoA and promotes the reassembly of the cortical actomyosin network at the bleb membrane, thus contributing to the regulation of bleb retraction.
