Abstract:
Plant glutathione peroxidases (GPXs) are the main enzymes in the antioxidant defense system that sustain H2O2 homeostasis and normalize plant reaction to abiotic stress conditions. To understand the major roles of the GPX gene family in rapeseed (Brassica napus L.), for the first time, a genome-wide study identified 25 BnGPX genes in the rapeseed genome. The phylogenetic analysis discovered that GPX genes were grouped into four major groups (Group I–Group IV) from rapeseed and three closely interrelated plant species. The universal investigation uncovered that the BnGPXs gene experienced segmental duplications and positive selection pressure. Gene structure and motifs examination recommended that most of the BnGPX genes demonstrated a comparatively well-maintained exon-intron and motifs arrangement within the identical group. Likewise, we recognized five hormones-, four stress-, and numerous light-reactive cis-elements in the promoters of BnGPXs. Five putative bna-miRNAs from two families were also prophesied, targeting six BnGPXs genes. Gene ontology annotation results proved the main role of BnGPXs in antioxidant defense systems, ROS, and response to stress stimulus. Several BnGPXs genes revealed boosted expression profiles in many developmental tissues/organs, i.e., root, seed, leaf, stem, flower, and silique. The qRT-PCR based expression profiling exhibited that two genes (BnGPX21 and BnGPX23) were suggestively up-regulated against different hormones (ABA, IAA, and MeJA) and abiotic stress (salinity, cold, waterlogging, and drought) treatments. In short, our discoveries provide a basis for additional functional studies on the BnGPX genes in future rapeseed breeding programs.

Abstract:
Plant height is an important agronomic trait that affects the establishment of the ideal plant type and yield formation of rapeseed. However, the genetic mechanism affecting plant height has not been fully elucidated. Building on previous genome-wide association studies (GWASs), we performed differential gene expression analysis using 14 shoot apex transcriptome datasets derived from materials exhibiting extreme seedling height phenotypes at the bud stage. The promoter and the first exon regions of the Glycosylphosphatidylinositol 2 gene (BnGPI2-A03) of rapeseed were identified as harboring two significant SNPs (Bn-A03-p717776 and Bn-A03-p7178917) associated with plant height. Co-expression network analysis shows that BnGPI2-A03 was associated with many hormone pathway genes, cell wall synthesis pathway genes, and transcription factor genes and formed a potential network regulating the plant height of rapeseed. To examine the function of BnGPI2-A03 on plant height in Brassica napus L., we generated transgenic plants in which BnGPI2-A03 was overexpressed using a 35 s promoter. Overexpression of BnGPI2-A03 elevated the height and main inflorescence length of the transgenic plant by an average of 17.95% and 25.92%, respectively, which showed a positive relationship with the level of BnGPI2-A03. Transcriptome sequencing analysis of the overexpressed transgenic rapeseed stem-tip tissue during the bud stage shows that BnGPI2-A03 is associated with plant cell wall synthesis and cell division pathways. The requirement of BnGPI2-A03 for plant height was further evidenced in natural germplasms by the higher expression levels of BnGPI2-A03 in the stem-tip tissue of extremely high genotypes (Hap1) relative to their ultra-short counterparts (Hap3). These discoveries highlight potential targets for the plant height genetic improvement of rapeseed in the future.

Abstract:
Germin-like proteins (GLPs) play important roles in the regulation of various plant development processes, such as seed vigor, root and leaf development and disease resistance, while the roles of GLPs on agronomic traits are rarely studied in Brassica napus. Here, we identified GLPs family genes in rapeseed and analyzed their potential functions. There are 77 GLPs family genes (BnGLPs) in the Zhongshuang11 rapeseed reference genome, divided into a, b, c, d, e, f six subfamilies. Tissue expression profile analysis of BnGLPs revealed the following: e subfamily genes were highly expressed in early stages of silique, cotyledon, vegetative rosette and leaf development; f subfamily genes were highly expressed in seed development; genes of a subfamily were mainly expressed in the root; and genes of b, c, d subfamily exhibited low-level or no expression in above mentioned tissues. RT-qPCR analysis confirmed that the transcripts of two f subfamily members decreased dramatically during seed germination, suggesting that f subfamily proteins may play vital roles in the early stage of seed germination. Transcriptome analysis of axillary buds in sequential developing stages revealed that the transcripts of eight e subfamily genes showed a rapid increase at the beginning of shoot branching, implying that the e subfamily members played vital roles in branch development. These results demonstrate that rapeseed BnGLPs likely play essential roles in seedling development, root development and plant architecture, indicating that harnessing certain BnGLPs may contribute to the improvement of rapeseed yield.

Abstract:
Plant glutamate receptor-like channels (GLRs) are homologs of animal ionotropic glutamate receptors. GLRs are critical in various plant biological functions, yet their genomic features and functions in disease resistance remain largely unknown in many crop species. Here, we report the results on a thorough genome-wide study of the GLR family in oilseed rape (Brassica napus) and their role in resistance to the fungal pathogen Sclerotinia sclerotiorum. A total of 61 GLRs were identified in oilseed rape. They comprised three groups, as in Arabidopsis thaliana. Detailed computational analyses, including prediction of domain and motifs, cellular localization, cis-acting elements, PTM sites, and amino acid ligands and their binding pockets in BnGLR proteins, unveiled a set of group-specific characteristics of the BnGLR family, which included chromosomal distribution, motif composition, intron number and size, and methylation sites. Functional dissection employing virus-induced gene silencing of BnGLRs in oilseed rape and Arabidopsis mutants of BnGLR homologs demonstrated that BnGLR35/AtGLR2.5 positively, while BnGLR12/AtGLR1.2 and BnGLR53/AtGLR3.2 negatively, regulated plant resistance to S. sclerotiorum, indicating that GLR genes were differentially involved in this resistance. Our findings reveal the complex involvement of GLRs in B. napus resistance to S. sclerotiorum and provide clues for further functional characterization of BnGLRs.

Abstract:
Sri Lanka is a nation in which multi-religious, multi-ethnic multi-language people live. Buddhists are the majority, while Muslims form the second minority group next to Tamils. Since historical times, the community relationship between Buddhists and Muslims has been prevailing. However, recently, a disturbing trend has been widely spreading among the Buddhists and Muslims. This situation has emerged during the aftermath of the anti-Muslim campaigns set by a number of Buddhist Nationalist Groups (BNGs), with their main goal being to propagate incorrect opinions about the Muslims to promote negative views about their socio-culture, and to distort the idea of a peaceful relationship between Buddhists and Muslims in the country. Accordingly, in the past several years, they have campaigned against halal certification on consumer goods, hijab and niqab of Muslim women, cattle slaughtering, places of worship and prayer services, among others. Moreover, they spread out the illusion that the above aspects of Muslim socio-culture are notable threats to the Buddhist people. So, these aspects are assumed by the Buddhists to be obstacles for maintaining a community relationship with Muslims. On the above background, analyzing the extent to which the above aspects influence the majority-Buddhists and minority-Muslims relationship, and determining as to whether an unfastened relationship will prevail between them, are the main objectives of this study. Based on the results, it is certainly affirmed that the above Muslim socio-cultural aspects, except slaughtering of cattle, have not pushed their influences to damage the Buddhist-Muslim relationship in Sri Lanka. In this respect, it was found that the aspect of ‘slaughtering of cattle’ is the only obstacle to the Buddhist-Muslim relationship. Furthermore, the recent campaigns have not changed the Buddhists’ mood in terms of maintaining a better relationship with Muslims. Moreover, the campaigns did not change their habits in keeping up the relationships with Muslims, without any break as how they behaved during the war (1984-2009) and pre-war periods. However, it is worthy to note that the BNGs have succeeded through their campaigns to create a negative Muslim stereotype among a small population of Buddhists in Sri Lanka.

Abstract:

Abstract:
碩士
國立勤益科技大學
工業工程與管理系
104
Taiwan has one of the fastest aging populations in the world. In 2015, citizens aged 100 years and older exceeded 3,000. As a niche market formerly neglected, the senior segment has been highly prioritized in recent years and become a global developmental topic. In accordance with current social trends, the capacity for families to care for their senior members has decreased, resulting in an increasing demand for senior citizens learning centers. Senior citizens learning centers offer a form of community daycare service by providing senior citizens with group leisure and entertainment activities during the day. Subsequently, at the end of each day, these centers return the senior citizens to their homes so that they can enjoy quality family time with their loved ones. This program permits offspring to focus on personal aspirations while also tending to senior citizens’ requirements. Based on the theory of planned behavior, this study created a behavioral intention model for senior citizens learning center participation. By using convenience sampling, this study distributed questionnaires to citizens aged 30 years and older in Taichung. The application of the following methodologies contributed to an insightful behavioral intention analysis about the critical factors influencing senior citizens learning center participation: structural equation modeling (SEM), independent-samples t-test, analysis of variance, and multiple responses. In this study, the allocated samples corresponding to a multivariate normal distribution and the SEM projected a high goodness of fit, reliability, and validity. The study results indicated that variables of attitude, subjective norm, and perceived behavior control caused a direct and positive impact on behavioral intention; among which, perceived behavioral control and subjective norm yielded considerable effects. Suggestions for the government and business investors seeking senior citizens learning center ventures are as follows: design marketing strategies tailored to specific segments and provide special services that satisfy psychological well-being and biological requirements. Such actions and measures would increase an overall participation in senior citizens learning centers, and offer a domestic senior care service.

Abstract:
Abstract The physical and chemical properties of secondary side steam generator deposits in the form of powder and flake obtained from Bruce Nuclear Generating Station A (BNGS A) Unit 3 were studied. The chemical phases present in both types of deposits, collected prior to the 1994 chemical cleaning during the pre-clean water lancing compaign, were magnetite (Fe3O4), metallic copper (Cu), hematite (Fe2O3) and cuprous oxide (Cu2O). The major difference between the chemical composition of the powder and the flake was the presence of zinc silicate (Zn2SiO4) and several unidentified silicate phases containing Ca, Al, Mn, and Mg in the flake. The flake deposit had high hardness values, high electrical resistivity, low porosity and a lower dissolution rate in the EPRI-SGOG (Electric Power Research Institute-Steam Generator Owner’s Group) chemical cleaning solvents compared to the powder deposit. Differences in the deposit properties after chemical cleaning of the Unit 3 steam generators and after laboratory cleaning were noted. The presence of silicates in the deposit inhibit magnetite dissolution.
