Full textCutin is a polymer of oxidized fatty acids and glycerol specific to plants. It forms the structural matrix of the waxy cuticle covering the epidermis of the aerial parts and plays a vital role in plants by preventing desiccation. Biosynthesis of cutin polymer was studied in the model plant Arabidopsis thaliana. Mutant loss-of-function monooxygenase type cytochrome P450 CYP77A4 were isolated. The analysis of transgenic lines expressing the GUS reporter gene under the control of the promoter of CYP77A4 showed that the gene was expressed mainly in floral organs and seeds. The analysis of various organs in the cutin demonstrated that the gene was essential for the synthesis of a C18 trihydroxy polyesters present in seed polyesters (9,10,18-trihydroxyoctadecenoic acid). A method for the separation of the embryo and seed coat allowing to analyze embryo polyesters was developed. The trihydroxy C18 fatty acid was found to be the major cutin embryo monomer. Profile of cutin monomers in mutant embryos showed that CYP77A4 is an epoxygenase in the biosynthetic pathway of C18 cutin monomers. The study of the physiology of the mutants also showed that the trihydroxy- fatty acids of the embryo cuticle play an important role in the germination of the seed under conditions of salt stress. In a second study, mutant loss-of-function and overexpressors for the Arabidopsis gene BODYGUARD encoding a protein of the α / β hydrolase fold superfamily have been characterized. Analysis of polyesters in these lines showed that this protein, whose role in the formation of the cuticle was not understood, plays in fact a role in cutin biosynthesis

Full textHost behavioral manipulation, a widespread strategy among parasitic organisms, has evolved to favor the transmission and/or survival of parasites. Understanding such a strategy is at the heart of applied aspects of parasitology, such as epidemiology and medicine, but is also important for evolutionary and ecological reasons. The aim of this study was to improve our knowledge on a category of manipulation that remains poorly documented: the bodyguard manipulation, initially described on parasitoid insects. Following their larval development, certain parasitic wasps (Braconidae) are able to usurp the behavior of their host and force them to serve as bodyguards to protect parasitoid cocoon against natural enemies. Studying the association between the parasitic wasp Dinocampus coccinellae and one of its host the spotted lady beetle Coleomegilla maculata, this work is structured in five axes: i) Efficiency of the bodyguard protection and associated costs, ii) Energetic constraints for manipulative parasites, iii) Diversity and evolution of bodyguard manipulation, iv) Host survival from parasitism and v) Host nutrition effects on a host–manipulative parasitoid association. Our results provide original findings about the ecology and evolution of the bodyguard manipulation, for instance the energetic costs associated with host manipulation or the role of host nutrition in this type of association. New perspectives are identified in order to stimulate future research in this area

This thesis is an interdisciplinary scholarly attempt to explore how individual traumas and colletive traumas are represented in Hualing Nieh’s Mulberry and Peach and Jade Y. Chen’s Mazu’s Bodyguards. In Chapter One, I juxtapose the two novels with Nieh’s and Chen’s accounts of their lived experiences, in order to investigate how Nieh and Chen transfer their traumatic memories into their literary works, and how the historical and social contexts they were in affected their artistic representation of traumas. As Nieh and Chen represent traumas in the novelistic mode, their deeds correspond to a crucial point held by many trauma studies scholars: since the overwhelming power of trauma leads to its impossibility of being fully perceived and accurately presented by people, in trauma narratives too definite positions of recounting the traumatic experiences should be avoided and recreating the traumatic events is needed. This recreation process corresponds to the basic philosophy of narrative therapy, a branch of postmodern psychotherapy approaches. Narrative therapy suggests that the story told by every client is constructed within certain context; during the treatment, the predominant problematic story should be deconstructed and replaced by “alternative stories.” Due to the fact that in both Mulberry and Peach and Mazu’s Bodyguards the two authors reveal their individual and family traumatic stories embedded in national traumatic events through the female perspective, I consider that there are two aspects in which the two texts constitute their authors’ “alternative stories.” First, by interweaving depictions of their lived experiences with fictional elements in the texts, Nieh and Chen tell the “alternative stories” as substitutes for the traumatic memories that have been haunting them in real life. Moreover, Nieh and Chen provide “alternative stories” to counter the male-dominated discourses in mainstream national narratives. In Chapter Two, I examine how Nieh and Chen, as witnesses to women’s sufferings during national unrest, eschew symbolically re-victimizing those female bearers of historical traumas in their “alternative stories.” Borrowing bell hooks’ theory about the margin as a strategic space of resistance, I consider that Nieh’s and Chen’s portrayals of those traumatized women constitute “the subversive voices from the margins” that can provide counter-perspectives against the predominant discourses.

Abstract At present most attention and discussion on 陳玉慧’s Mazu’s Bodyguards from the academic circle focus on the family’s story extending outward and overlapping the country’s fate (family to nation). In my point of view, this kind of discussion regretfully does not refer to the alienation and depressed relationships between people in modern world. I saw alienation between people in modern society in the whole alienated content. Why do tragic lives keep coping to the next generation and cannot be got rid of? Why must one leave? Why is every single character so lonely? Why cannot the thirst for being loved, identified and cared be satisfied throughout the time? Why cannot be accepted when the characters love other people with all their love? Therefore I focus on how a subject looks upon his/her own and family situations and deconstruct the alienation in Mazu’s Bodyguards from objective psychological points of view. This dissertation starts from the author’s semi-autobiography and discusses alienation between men and women’s love, generations and in-law relationship, followed by the author’s life history, compare the alienated topics in the author’s proses and novel, brings out the alienation and identification between the author and other people, and analyze where the root of 陳玉慧’s alienated homeless feeling and pessimistic life is. And then I analyze the relationship between alienation and subject’s self identity. In my point of view, in order to analyze the alienated situation, we have to start from constructing one’s self subject and search for the answer in one’s inside world. Only after recognizing self’s ‘position’, adjusting the interactive attitude between the subject and the outside environment, establishing closer relationships with other people starting from a health self, one subject can truly escape from the alienated relationships. He/she then can reach his/her own peace and comfort in life. Keywords:陳玉慧, alienation, identification, Mazu’s Bodyguards, attachment relationship

The purpose of this study is to explore the application of the traditional martial arts Bajiquan during the close combat (about 6 feet) of personal protection. To better understand the history and characteristics of Bajiquan, the job nature of personal protection and the significance of close compact as a personal defense technique, an extensive literature research was conducted. Through over 20 in-depth interviews with martial art masters, personal security experts and practitioners, relevant data were analyzed and summarized as follows: 1. the uniqueness of martial art: Bajiquan and the personal protection professions originated from the protection of the last emperor Pu Yi of the Qing Dynasty. Until today, it is still a must-have combat skill for Taiwan’s national security secret service personnel. The action characteristics of Bajiquan include “leaning shoulders and striking with elbows, pounding and moving by pelvic limb”, which wins the martial art a reputation of "striking with the elbow and the mountain is knocked down, stamping one’s feet and the entire continent is shaken up." Personal security personnel use close combat technique to ensure the employer’s safety and security. Bajiquan features its action in a way of “Constantly keeping one’s strength ready while holding the actions until the right moment”, which deals with any situation in a low-profile manner while prepares to launch required action at any moment. Such strategy takes into account the employer’s personal and professional image. 2. The relations between the martial art theory, its moves and techniques, and the application of the concept and the move in actions: the close-combat defense technique for personal protection uses the principle of “shielding with body, escorting to leave under cover” as the main tactics. Different scenarios in various situations are classified into three different natures and degrees of risks: low risk (disturbance), medium risk (nuisance) , and high risk (harmful). To deal with different scenarios, Bajiquan’s boxing theory states: "reserves the strength inner-body invisibly, rendering oneself without reservation, preparing to kill with one single blow", with which to exercise the tactics of "attack by strength of the elbow in close range" to eliminate threats and ensure the safety of the employer. 3. Establishing a training system for Bajiquan close combat personal defense skill which suits the need of personal protection requirements: the traditional training system should be used as the basis, combined with the actual work needs and the principles of business management, to establish a "modular" training system, which include real combat practice and the introduction of the training concept and methodology of sports science, meanwhile to facilitate professional teaching resources for inheritance of experience so as to enhance the overall training effectiveness. The research concludes in in attempt to contribute to the promotion of Chinese traditional martial arts culture, and to provide a reference for the use of Bajiquan as a close combat defense technique for the personal security service industry or the continuous development and advancing of government agencies in the coming future.
