Abstract:
Botho Strauss a writer of prose, essays and plays, belongs to the most interesting and controversial authors of the last thirty years. Like a seismograph he picks up numerous discourses, the most important one being Memoria . His peculiar and eccentric way of representing and reflecting on memory, in the early texts, is innovative, critical, self reflective, but in the later texts becomes neo-conservative, elitist and patriarchal. This later discourse sparked off a discussion, involving writers such as Peter Handke, Martin Walser and Peter Sloterdijk, about the role of the intellectual in German society.<br>I begin by examining how cultural studies approaches the topic of Memoria. I then apply to the texts of Strauss a 'mnemonic reading,' meaning, to explore and discover topographies of memory. Literature and the arts in general are perceived as places of memory, or as archives where cultural memory is kept alive. Quoting, or rewriting selections from texts, as described in theories of intertextuality, changes and keeps alive a tradition of literature within literature. A text becomes a palimpsest in which fragments of foreign texts suddenly become visible and can enter the readers memory. In Strauss, to form a memory from the fragments is not without major problems. The pieces move in all directions to form a paradoxical body of text.<br>In his early texts the narrator laments the impossibility of creating an identity based on personal memories and so he concentrates on the problem of how to represent memory within the context of increasing amnesia. The seemingly dead subject moves about within the texts where he hopes to build himself a place for his identity to survive. In reaction to loss of memory and identity, a calculated game appears in which the representation and experience of the past plays only a minor role. The author turns towards the models of aesthetic memory, adapting, for example, the romantic mode of writing. The metamorphosis of texts, such as those from the romantic period, occurs as a backdrop against which the writer projects his erotically inspired allegorical "memory theater." Dramatization of the memory process leads him to conclude that memories only exist within the aesthetic world of literature, where they manifest themselves beyond a historical and social reality. At the end of his writing project, Strauss creates an aesthetic refuge of personal memories by turning himself into a "diarist." The male voice regresses to an aggressive and melancholic mode of limited perception, which the author knows cannot be more than an artificial construction.

Ce travail se propose de definir le concept de devenir scenique (formule par j. -p. Sarrazac), associe a la notion de forme-sens (selon la definition de h. Meschonnic) a travers l'analyse de deux elements essentiels, l'espace et le regard. L'analyse, qui porte sur trois auteurs contemporains, bernard-marie koltes, peter handke et botho strauss, comprend, sans les juxtaposer, d'abord une analyse textuelle et ensuite celle d'un corpus de mises en scene des oeuvres des auteurs examines, dans la conviction que le devenir scenique et la forme-sens constituent des instruments permettant une nouvelle comprehension du texte et de son rapport avec la scene. Apres une enquete sur le nouveau statut du texte et de la scene dans le theatre contemporain, l'analyse met en evidence le role fondamental de v espace dans ces ecritures: "espace structurant" chez koltes, "espace traversee" chez handke, "espace-temps" ou "espacemetaphore" chez strauss. Le chapitre "figures du regard" etudie le pouvoir structurant du regard dans l'ecriture des auteurs en question, tandis que le suivant, "mise en scene et devenir scenique", examine les relations que les metteurs en scene ont entretenues avec les textes. L'approche des textes a partir de leur devenir scenique, qui comporte le deplacement de l'attention du contenu a la forme, reorientation fondamentale pour la comprehension du texte theatral contemporain, permet de rappeler le double statut du texte - ferme et ouvert - ainsi que de reflechir sur l'influence, considerable dans le theatre contemporain, de l'ecriture sur la scene, qui va jusqu'a bouleverser les modes de jeu et l'emploi de l'espace. Enfin, par son caractere "polyvalent" et englobant, cette approche permet de mieux cerner la theatralite textuelle et d'entrevoir une "nouvelle alliance" entre le texte et la scene.

Abstract:
Botho Strauß, source for polemic and target of vitriol over three decades, proposes an unsettling understanding of the poetic in his prose works, from his earliest writing to most recent publications. The thesis contends that this understanding of the poetic is deeply indebted to the late thought of Martin Heidegger: it investigates the nature of the debt, highlighting Strauß’ adoption and adaptation of ideas central to the philosopher, including his thinking on the work of art, technology, language and poetry. The body of the thesis examines Strauß’ views through detailed exegeses of Beginnlosigkeit, Wohnen Dämmern Lügen and Fragmente der Undeutlichkeit, while drawing extensively on other works and writing. The readings identify and elucidate a number of key terms critical to Strauß’ proposed poetic. Underpinning these terms, the thesis contends, and bound to the understanding of the poetic, is an ontological concern for philosophical truth derived from Heidegger. The thesis concludes that far from a retreat by Strauß into obscurantist mysticism and resignation from a putative cultural, social and political collective, accusations repeatedly levelled at him and here grouped under the rubric of fatalism, Strauß offers in and through his works a dynamic engagement with this conception of truth, which the thesis hypothesises as a poetics of dwelling.

Streptomyces clavuligerus is a Gram-positive, filamentous bacterium which produces several important secondary metabolites, including isopenicillin N, cephamycin C and the &beta<br>-lactamase inhibitor clavulanic acid. Among these compounds, clavulanic acid is being used in combination with commonly used &beta<br>-lactam antibiotics in order to fight against bacterial infections that are resistant to such antibiotics. Among these combinations, Augmentin, composed of amoxicillin and clavulanic acid, is the most widely prescribed drug and has a market value of more than one billion dollars per year. There are two genes that act in regulation of clavulanic acid biosynthesis: ccaR located in cephamycin C gene cluster and claR located in clavulanic acid gene cluster. The goal of this study is to improve clavulanic acid production capacities of both wild type and industrial S. clavuligerus strains by integrating extra copies of claR gene into S.clavuligerus genome and its overexpression via a multicopy plasmid. Although previously has shown to be quite effective on wild type S. clavuligerus strains, claR overexpression in the industrial strain used in this study yielded only 1.4-fold increase in volumetric and 1.7-fold increase in specific CA production by the recombinant strains MA28 and MA16, respectively.

Abstract:
The famous contemporary German playwright Botho Strauß is also a prominent author of philosophical and aphoristic short prose, among others a volume named Paare, Passanten (Pairs, Passers by), which has been examined from the standpoints of formal, topical and ideological particularities. From the formal standpoint, the book is distinguished by occurrence of mixed literary forms dominated by short reflective, philosophical and personal notes, narrations and "portraits of moments", which bear both aphoristic and essayistic styles. The transitions between individual literary forms are often unclear and inconspicuous, even within an individual text. The texts are loosely connected to each other, yet they form a sophisticatedly composed series. The style of Strauß's texts is heavily enigmatic, fantastic and often allegorical. The texts are distinguished by a multilayered, at least dual structure (compare Welsch 26), which is typical for post-modernistic literature and offers to the reader a great variability in interpreting the allegorically encoded abstract phenomena, while the actual contents of these texts mainly depends on the contents of reader's mind. Abstract:
Activation induced Cytidine Deaminase (AID) contributes to the generation of antibody affinity by participating in two reactions, class switch recombination (CSR) and somatic hypermutation (SHM). Both reactions occur after VDJ recombination, subsequent to antigen exposure. During CSR, a deletion and recombination event occur to alter the effector function from IgM to either IgG, IgE, or IgA. SHM then occurs, which introduces point mutations at a high frequency into the variable regions of both the immunoglobulin heavy and light chains. These point mutations increase the antibody binding affinity for antigen, and antibodies with greatest affinity for antigen will be positively selected and further expanded during an immune response. The ability of AID to act as a mutator gene underscores the importance of understanding its regulation throughout the genome. Action of AID on genes outside of the Ig loci can lead to genomic instability. Hyperactivity of AID has been shown to cause chromosomal translocations and other oncogenic malignancies. Loss of AID can lead to immunodeficiencies. Therefore, it is imperative to understand how AID identifies and interacts with target sequences and mutates both strands of the DNA. Previous studies have identified DNA secondary structure such as R loops, transcription factors, miRNA, and phosphorylation as events important for determining AID's ability to access its substrate sequences. However, none of these studies demonstrated how AID mutates both strands of DNA, reminiscent to its in vivo mode of action.The focus of this thesis is to identify how AID mutates both strands of the DNA duplex, and how target genes are identified. To this end, we have discovered that AID functionally interacts with the cellular non-coding RNA degradation complex, RNA exosome. We observe that the RNA exosome stimulates AID activity on both strands of DNA in in vitro reconstituted reactions. The RNA exosome/AID complex binds to switch (S) sequences in a manner that is both transcription- and AID-dependent. Knockdown of exosome core component ExoSc3 results in defects in CSR. Additionally, this work focuses on the role of the neddylation (Nedd8) of AID in recruitment to its target sequences. Neddylation, a 10kDa modifier, is a small ubiquitin like modifier which functions in a variety of cellular processes. We have used a combination of proteomics, computational approaches, and candidate screening to identify and validate the role of E1, E2 and E3 in CSR. We have identified NEDD4 as the AID-specific E3 Neddylation ligase and demonstrated its requirement for CSR in mouse B cells. Using mass spectrometry, we have identified AID neddylation sites from in vitro neddylated AID proteins. We observe that mutation of these AID-neddylation sites affects AID/RNA exosome interaction and CSR efficiency in B cells. These observations point towards a role of NEDD4 in recruiting AID/RNA exosome complex to the immunoglobulin locus. Additionally, we confirm the role of NEDD4 as an E3 ubiquitin ligase of RNA polymerase. In both cell lines and primary cells, we observe an increase of germline transcripts and S region resident RNA polymerase in the absence of NEDD4. We propose NEDD4 ubiquitination can promote the degradation of stalled RNA polymerase complexes at the Ig S region, facilitating exosome access to germline transcripts and AID access to the template strand.

Under the globalization, the the trade in service is promoted to be more international and liberal. After Mainland China and Taiwan have become new members of WTO in 2001 and 2002 respectively, the Chinese trade in service industry faces huge competition from the enterprises of other developed countries. Consequently, it becomes more and more urgent for Mainland China and Taiwan to find out the method that promotes competence of both in the international trade in service markets while obeying the international regulations. Besides the introduction and conclusion, this thesis is mainly divided into three major parts. GATS is the main analysis construction. Under GATS, this thesis analyzes Mainland China and Taiwan’s trade in service industry by different viewpoints and observe opportunities and threats of Chinese trade in service industry during the huge fluctuation period in a pratical way. Further, an analysis of the new liberlization measures will be introduced, the impacts of service trade liberlization to China’s service trade and counter measures will also be discussed. Finally, the challenges and the approaches to the development of Mainland China and Taiwan’s trade in service, the comparative advantages and relative strategy, the difficulties and advices for solving the current problems will also be provided. The conclusion of this thesis are that the Mainland China’s trade service market is tremendous and the Taiwan government should adopt a more open strategy to enable enterprises to get in and become the center of international trade service market. In domestic, the Taiwan government should improve the tradet service industry to be more regulated and resolve the chaotic condition while expanding their business territory as fast as possible. In the future, especially in service industry, the competence trend will become more and more knowledge-based. In other words, the key factor of competition is talent persons, consequently, enterprises and individuals who have well negotiation skills and are familiar with the internationally investment and economic regulations will have more chance to be granted preferential treatment in most sectors when setting up operations in the Mainland China. Moreover, by citing related examples and views at home and abroad, the author hopes to help promote a better understanding of the agenda of the thesis and the development of the economic and trade co-operation between the Mainland and Taiwan, and the co-operation in bilateral relations could be consolidated and strengthened eventually. Taiwan will be able to take full advantage of Mainland China’s strengths and vice versa. This will not only enable each other to further accomplish its investmenal enviroment, but also to contribute to each other’s demsetic development.

Therefore, in 1999 OECD announced “Principles of Corporate Governance （revised in 2004）” which covers the rights of shareholders, the equitable treatment of shareholders, the role of stakeholders, disclosure and transparency of information and the responsibility of the board and provides references for its member countries and non-member countries. The era for globalization and standardization of corporate governance has come. The flaws of company governing have been faced by most of the countries in the world, and Taiwan and the PRC are no exception. To mend these flaws, Taiwan and the PRC try systematically to introduce the system of independent directors, as is adopted by some developed countries. It is hoped that independent directors will be able to resolve the difficulties created by the inability of the Supervisors（Supervisory Committee） to perform their duties properly. After the Independent Director’s system has been introduced and legislated, the situation that two kind of supervisions （Supervisory Committee）coexist has appeared in the structure of the company in Taiwan and the PRC. To be sure, there are some conflicts between these two systems. Hence, this article is try to understand in depth the flaw of internal corporate governance and then discuss how to improve the Supervisors（Supervisory Committee）system of Taiwan and the PRC in order to avoid or reduce enterprise’s financial crisis and take place once again . This article sets out the comparison between the basic mediation system at the local level in Taiwan and Mainland China and explores the relationship and differences between the township and county-administered city mediation of Taiwan and people’s mediation of Mainland China. Through such comparison and mutual learning, mutual aid and systematic exchange are hoped to be achieved. This research first explores the township and county-administered city mediation of Taiwan and people’s mediation of Mainland China from the history of Chinese mediation system and introduces the development and features of each country’s mediation system, as well as the laws of current alternative dispute resolutions. Second, this research describes the history and current implementation of the township and county-administered city mediation of Taiwan and people’s mediation of Mainland China in details and then analyses and compares the two systems. Establishing the relationships between parents and teachers should be valued as a key point to maintain connections between children and the kindergartens. Thus, this research would focus on the comparison of the interaction and contents between teachers and parents in private kindergartens in both China and Taiwan. The research method is individual interview. The interview subjects includes three kindergarten teachers from China, three parents from China, three kindergarten teachers from Taiwan, two parents from Taiwan, and a parent from Taiwan who were a kindergarten teacher in China before. The interview subjects were twelve in total. After analyzing based on the contents of interviews, the research would propose a conclusion, some suggestions, and elaborations of the differences between the interview result and references, in order to respond the question and purposes of this research. Under the affection of declining birth rates, governmental policies, and competition between the same trade, Taiwan has more multiple ways of interaction and deeper discussions between parents and teachers. On the other hand, the policies in China have made a stable birth rate, so the relationship between parents and teachers are comparatively passive. With the development of technology, the way of interaction between parents and teachers changes. And the content of discussions has gradually changes from larger, mutual aspects to more separative, individual ones. Positive interactions between parents and teachers will be an important factor for the development of both the kindergartens and the children in there. The establishment of interactions between parents and teachers depends on the way they choose and what their communication contents are based on. Thus, the appropriate ways of interactions and proper communication contents should be the two main targets for parents and teachers to pursue in order to enhance their interactive relationship.

Mainland China hopes to create employment opportunities through entrepreneurship. Taiwan government encourages entrepreneurship because the unemployment rate is too high. Most of the undergraduates are involuntarily unemployed. Government and businesses have also encouraged young people to create their own business; however, the number of self-employed graduate is less than the neighboring countries. Previous work on entrepreneurship issue mostly focuses on personality and demographic variables, which makes it difficult to measure properly the characteristics of entrepreneurs. Therefore, this study attempts to apply Entrepreneurial Attitudes Orientation (EAO) using EAO Scale and Big Five Model to evaluate the difference on four dimensions: innovation, achievement, personal control, and self-esteem among undergraduate students of general and vocational universities. This study quantified the questionnaire survey methods and explored whether there were significant differences in EAO four facets general and vocational universities different personality traits, and different population variables of students through an online questionnaire. 　　The results of the study show that cross-strait college students do not influence their entrepreneurial attitudes because of gender, college differences, and whether their family members have entrepreneurial experience. The personality traits of Taiwan’s college students show that there is a positive correlation between prudence and entrepreneurial attitudes. It also means that the higher the Taiwanese’s prudent score, the more stringent it will be in the entrepreneurial process; in mainland China, the top five dimensions all influence the orientation of entrepreneurial attitudes. After 1990s, they became active members of the society, received higher education, no longer focused on the constant advantages of low-cost manufacturing, but were more sophisticated in grasping market changes and focusing on new business models. 　　Finally, based on the conclusions of the study, recommendations are made to provide government, education units, entrepreneurs, and future researchers as reference.

Fire resistance design and analysis is an under-studied and under-codified area of bridge engineering. With the lessening of conservatism in bridge design, the aging or our bridge infrastructure, and the increase in the ground transport of highly-flammable and -combustible materials, it is essential that the bridge engineering community better understand and incorporate methods for modeling the effects of fire on bridges. Typical fire resistance analysis looks at the response of individual structural components. Analysis for the component of a bridge is nowhere more important than for that of the main cables of suspension bridges. As such, we will survey and introduce the necessary analysis techniques to provide the bridge engineering community with the knowledge and tools to understand fire modeling and both rapidly and accurately assess their effects on suspension bridge main cables. The work of this dissertation is twofold. In the first portion, we address proper fire modeling techniques for bridge conditions and apply them in a sequential thermal-mechanical analysis of a three-dimensional model main cable with thermally-dependent material and mechanical properties. Although fire modeling has been addressed in a variety of scenarios, including extensive studies for building design and analysis as well as tunnel design and analysis, the types of fires, fire geometries, and air conditions associated with bridge fires vary drastically. Our work identifies the time to failure for our particular main cable example and subsequently compares both the temperature and strain distributions for temperature-dependent and temperature-independent models. Although the three-dimensional analysis is complete, we hope to emulate and expand on the work done in the building fire engineering community and bring to the literature methods to produce significant two-dimensional temperature distributions for when a main cable component is either partially or fully-exposed to fire. As such, the main fire modeling analyses mentioned in the previous paragraph lay the groundwork for our pursuit of closed-form analytical solutions necessary to rapidly and accurately assess the time-dependent temperature distribution within a cable cross-section exposed to fire. These solutions are formed with different approaches depending on the fire scenario in question. They include a separation of variables (eigenfunction) approach, sinusoidal transforms, Laplace transforms, Green's function solutions, and a semi-analytical hybrid method. We validate each of the approaches numerically using three different fire models.

First, we focus our analysis on the post-war Taiwan advertising of background and effect, and on post-war advertising of features, national on advertising of management, and advertising with national of special relationship for discussion. Second, we emphasize the background and effect of post-war Chinese Communist Party advertising, and analyze with first part by the features, management, national relationship coordination. Third part is a comparison between both sides of the Taiwan Strait advertising development,using plane and space for dimensional of description, and research on both differences. After the Chinese Civil War, the different governing authorities of China and Taiwan led to divergent developments in their advertising industries. At the end of 1966, Taiwan’s advertising industry experienced a period of substantial growth, whereas, because the Chinese Communist Party (CCP) had declared the Cultural Revolution, advertising was strictly banned in China. During this study, we found that the Kuomintang (KMT) government could not fully eliminate the influence of Japan in Taiwan. Japanese medicine advertisements dominated Taiwan’s advertising industry, continuously influencing Taiwanese people’s lives. Because of the government’s weakening management of advertising and the chaotic post-war environment, Japanese drug manufacturers gradually gained control of advertising development, promoting the excellent quality of their medication through word of mouth, with minimal interference from the KMT government. Using medication advertisements, we analyzed the national relationship with advertising in post-war Taiwan. The Japanese government had a greater influence on Taiwan compared to that of the KMT, indicating that Japan, a previous colonizer of Taiwan, did not retreat from Taiwan after the end of World War II. Conversely, the CCP adopted a dual approach regarding advertising. Left-wing politics relentlessly attacked advertising, which was then horizontally connected with corruption, counterfeit medicine, imperialism, and extravagance. On the other hand, through the advertising and promotion of the Patriotic Health Campaign, dependence on domestic drugs in modern China was promoted by the chief of the Communist Party of China at every level in rural areas, substantially influencing the fundamental health issue in these areas. Promotion did not replace advertising until the Cultural Revolution. Third, although Taiwan exhibited an advantage in printed advertisements compared to China, Japan was the actual manipulator. However, employing wise use spatial management, the Communist Party of China created a new publicness of public health consciousness by the parade experience of the Chinese people, which may be paradigm of the current age. In this study, we explore how a national leader exploited advertising during a specific period to develop a new advertising function of modern society. Our unique results distinguish this study from conventional studies on advertising. Furthermore, we also considered the national relationship with advertising from a novel perspective.

Therefore, the writer specifically chose the “ Traditional Chinese Medicine ” (TCM) that maintains the lives of Chinese people on both sides of the Taiwan straits to be the subject of this study. Furthermore , the writer chose the “United Daily News”, which can best represent a spirit of “professional newspaper”, applying news theories of “frames and media representation” and “health and risk management”, to sieve and analyze great amount of news documents with content analysis and history comparison. The writer has achieved 571 valid samples. The writer has studied news on “ Chinese medicine on both sides of the Taiwan straits ” reported within 17 years (1996-2012) to discover the general condition of news report on TCM and , a step further, to probe into the difference made by Party Alternation in 2008 and ECFA on the report of such news. The writer also attempted to demonstrate through analysis the manner and framing of reports on this issue by news media. This study has found : (1) number of news of ” both sides page ” is the second in “ news page ” but is 29.77% of the total. 50.96% of “ news form ” belong to “ pure news report ” and is the highest. (2) “ Health and risk management on both sides of the Straits ” is the top issue of 9 issues of “ news theme ” and its number is 21.02% of the total. News related to “ traditional Chinese medicinal materials ” are the second with a percentage of 14.71%. (3) Number of “ news from unofficial sources ” is 296 and prevails by 52.88% of the total in “ information sources ”. (4) 71.10% of “ category of position ” belong to “ positive report ” and is the highest of 406. Comparing the two stages separated by the Party Alternation, this study has found: (1) In “ news page ”, the latter stage has more news in the “ major page ” by a double number. In both stages , numbers of news in “ both coasts page ” are nearly the same. And concerning “ news form ”, no significant difference is found in both stages. (2) Regarding “ news theme ”, the latter stage had four times of news on the issue of “ the institution and status quo of TCM on both coasts ”. Cross analysis and Chi-square test have found that “ news page ” and “ news position ” had been closely related to “ information sources ” in two stages of both the Party Alternation and ECFA. It has also been found that there are three times more of news in “ both coasts page ” in the former stage of ECFA than the latter stage.
