It is a complexation reaction and speciation of chromium(VI) and chromium(III) is possible because the reagent produced no colour reaction with chromium(III). The suitable acidity range for the green coloured chromium(VI) or blue green coloured vanadium added chromium(VI) complexes is 0.01&ndash;0.5 (N) HCl or 0.3 to 2.2 pH. Molar absoptivity of the blue green complex is 2.14 &times; 10<sup>4</sup> dm<sup>3 </sup>mole<sup>&ndash;1</sup> cm<sup>&ndash;1</sup> at 590 nm and Sandell sensitivity is 0.0025 &micro;g cm<sup>&ndash;2</sup>. A linear working range from the detection limit to 300 ng/ml of chromium(VI) was obtained. The lower detection limit (LOD) for the chromium-BPATCH complex and chromium-vanadium-BPATCH complex were 20 ppb and 5 ppb respectively at room temperature, when the absorbance of the colour reaction was 0.002 at 590 nm. The proposed spectrophotometric method was successfully applied to the determination of chromium(VI) in industrial effluent, USGS rock standard and speciation of chromium(VI) and chromium(III) in ground water sample.

Reactive oxygen species (ROS) signal vital physiological processes including cell growth, angiogenesis, contraction, and relaxation of vascular smooth muscle. Because cytochrome P-450 family 4 (CYP4)/20-hydroxyeicosatetraenoic acid (20-HETE) has been reported to enhance angiogenesis, pulmonary vascular tone, and endothelial nitric oxide synthase function, we explored the potential of this system to stimulate bovine pulmonary artery endothelial cell (BPAEC) ROS production. Our data are the first to demonstrate that 20-HETE increases ROS in BPAECs in a time- and concentration-dependent manner as detected by enhanced fluorescence of oxidation products of dihydroethidium (DHE) and dichlorofluorescein diacetate. An analog of 20-HETE elicits no increase in ROS and blocks 20-HETE-evoked increments in DHE fluorescence, supporting its function as an antagonist. Endothelial cells derived from bovine aortas exhibit enhanced ROS production to 20-HETE quantitatively similar to that of BPAECs. 20-HETE-induced ROS production in BPAECs is blunted by pretreatment with polyethylene-glycolated SOD, apocynin, inhibition of Rac1, and a peptide-based inhibitor of NADPH oxidase subunit p47phox association with gp91. These data support 20-HETE-stimulated, NADPH oxidase-derived, and Rac1/2-dependent ROS production in BPAECs. 20-HETE promotes translocation of p47phox and tyrosine phosphorylation of p47phox in a time-dependent manner as well as increased activated Rac1/2, providing at least three mechanisms through which 20-HETE activates NADPH oxidase. These observations suggest that 20-HETE stimulates ROS production in BPAECs at least in part through activation of NADPH oxidase within minutes of application of the lipid.

Abstract:
The present study evaluated the necessity of store-operated Ca2+ entry in mediating thrombin-induced 20-kDa myosin light chain (MLC20) phosphorylation and increased permeability in bovine pulmonary artery endothelial cells (BPAECs). Thrombin (7 U/ml) and thapsigargin (1 μM) activated Ca2+ entry through a common pathway in confluent BPAECs. Similar increases in MLC20 phosphorylation were observed 5 min after thrombin and thapsigargin challenge, although thrombin produced a sustained increase in MLC20phosphorylation that was not observed in response to thapsigargin. Neither agonist increased MLC20 phosphorylation when Ca2+ influx was inhibited. Thrombin and thapsigargin induced inter-endothelial cell gap formation and increased FITC-dextran (molecular radii 23 Å) transfer across confluent BPAEC monolayers. Activation of store-operated Ca2+ entry was required for thapsigargin and thrombin receptor-activating peptide to increase permeability, demonstrating that activation of store-operated Ca2+ entry is coupled with MLC20phosphorylation and is associated with intercellular gap formation and increased barrier transport of macromolecules. Unlike thrombin receptor-activating peptide, thrombin increased permeability without activation of store-operated Ca2+ entry, suggesting that it partly disrupts the endothelial barrier through a proteolytic mechanism independent of Ca2+ signaling.

Abstract:
In this study, N, N’-carbonic bis(piperonylic acid) dihydrazide (BPACH) was synthesized to broaden the category of piperonylic acid derivative and evaluate its influences on the thermal properties of poly(L-lactic acid) (PLLA). The geometry optimization of BPACH showed that the highest occupied molecular orbital mainly focused on the formed amide group and carbonic dihydrazide, whereas the lowest unoccupied molecular orbital mainly focused on the piperonylic acid, and the orbital energy gap was 0.10418 eV. The differences in melt-crystallization processes of the neat PLLA and PLLA/BPACH samples indicated that the BPACH could provide the effective nucleation site to accelerate the crystallization of PLLA, but the crystallization accelerating effect was still further improved compared to some reported nucleating agents. The melting behaviors of PLLA/BPACH samples after crystallization depended on the crystallization temperatures and heating rates; additionally, the melting processes could also effectively reflect the previous crystallization behaviors.

Peacekeeping is the important function of the Barangay Peacekeeping Action Team in their respective barangay, BPAT visibility is the one main focus of this peacekeeping activity. Hence, this study was conducted to determine the level of performance of the Barangay Peacekeeping Action Team (BPAT) in regards to their skills in patrolling, identifying violation in the community, basic self-defense skills and crowd control skills of selected Barangays in the City of Ozamiz during COVID-19 pandemic. The researchers employed a descriptive-correlational type of research characterized by gathering data from 100 respondents from the selected barangays in Ozamiz City. Moreover, survey questionnaire was used to gather the data. Furthermore, mean and standard deviation were the statistical tool used in the data analysis. Results revealed that the majority of Barangay Peacekeeping Action Team (BPAT) officers were very responsive in safeguarding and patrolling the neighborhood during the day and night while on the state/verge of pandemic. BPAT officers are highly knowledgeable and visible in securing their respective communities through ronda or patrol activity. BPAT officers also perform intelligence monitoring during the pandemic. The findings may provide additional information to the PNP and may give insights to the institution for the improvement of the performance of the Barangay Peacekeeping Action Team (BPAT) at the barangay level. Lastly this study recommends to maintain the BPAT’s excellent performance, and if possible to retrain them in upgrading the skills, and to have constant monitoring and evaluation of their performance and to establish a wider coverage of study or can be done to the entire city.

Abstract:
Community security involves the collective efforts of local authorities, law enforcement, and residents to ensure the safety and well-being of all members within a given area. This study explored the dynamics of collaboration between Barangay Peacekeeping Action Teams (BPATs) and police officers in ensuring the safety and security of the community in one of the barangays of Manolo Fortich Bukinon. This study utilized a phenomenological research design which is participated by total of twelve (12) participants chosen using purposive sampling. Moustakas’ method of data analysis was used in analyzing data. The study revealed five interrelated themes—ranging from collaborative synergy and trust-building to communication barriers and crime reduction—that underscore the critical role of sustained partnerships, clear communication, and coordinated efforts in enhancing public safety outcomes through BPAT-Police collaboration. The study concluded that strong collaboration among BPATs, police officers, and the community is essential in building trust, improving coordination, and achieving effective and sustainable public safety outcomes. It was recommended that local government units and law enforcement agencies institutionalize structured collaborative frameworks.

We (66) have previously described an NSAID-insensitive intramitochondrial biosynthetic pathway involving oxidation of the polyunsaturated mitochondrial phospholipid, cardiolipin (CL), followed by hydrolysis [by calcium-independent mitochondrial calcium-independent phospholipase A2-γ (iPLA2γ)] of oxidized CL (CLox), leading to the formation of lysoCL and oxygenated octadecadienoic metabolites. We now describe a model system utilizing oxidative lipidomics/mass spectrometry and bioassays on cultured bovine pulmonary artery endothelial cells (BPAECs) to assess the impact of CLox that we show, in vivo, can be released to the extracellular space and may be hydrolyzed by lipoprotein-associated PLA2 (Lp-PLA2). Chemically oxidized liposomes containing bovine heart CL produced multiple oxygenated species. Addition of Lp-PLA2 hydrolyzed CLox and produced (oxygenated) monolysoCL and dilysoCL and oxidized octadecadienoic metabolites including 9- and 13-hydroxyoctadecadienoic (HODE) acids. CLox caused BPAEC necrosis that was exacerbated by Lp-PLA2. Lower doses of nonlethal CLox increased permeability of BPAEC monolayers. This effect was exacerbated by Lp-PLA2 and partially mimicked by authentic monolysoCL or 9- or 13-HODE. Control mice plasma contained virtually no detectable CLox; in contrast, 4 h after Pseudomonas aeruginosa ( P. aeruginosa) infection, 34 ± 8 mol% ( n = 6; P &lt; 0.02) of circulating CL was oxidized. In addition, molar percentage of monolysoCL increased twofold after P. aeruginosa in a subgroup analyzed for these changes. Collectively, these studies suggest an important role for 1) oxidation of CL in proinflammatory environments and 2) possible hydrolysis of CLox in extracellular spaces producing lysoCL and oxidized octadecadienoic acid metabolites that may lead to impairment of pulmonary endothelial barrier function and necrosis.

Abstract:
Coronavirus disease (COVID-19) is a newly found coronavirus that causes an infectious disease. Infected patients may develop mild to moderate respiratory infections and recover without the need for specific treatment. Barangay Tanods is at the forefront of maintaining peace and order. They are made up of civilian volunteers who protect the community from irregular forces while also demonstrating their commitment to crime prevention by acting as deterrents to criminals, particularly in places where police are scarce. The BPAT is a national program of the PNP to encourage people empowerment from the community to address the real-time response in case a need arises, be it peace and order, security, or rescue related matters. The study will utilize a descriptive type of research. A descriptive type of research accurately and systematically describes the responses of the population considered in a particular study. The results revealed that the overall performance of barangay tanod was excellent, regardless of age, educational attainment, or gender. There were no significant differences in the performance of tanods divided into different groups. On the other hand, the delivery of BPAT services for crime prevention needs to be enhanced. The goal of this research is to assess the BPAT’s performance during the COVID-19 pandemic in the urban barangays in Ozamiz City, which includes the following: Barangay Aguada, Barangay Carmen Annex and Barangay Catadman in terms of patrolling, cooperation with other barangay officials, and health and safety protocol execution.

20-Hydroxyeicosatetraenoic acid (20-HETE) is an endogenous cytochrome P-450 product present in vascular smooth muscle and uniquely located in the vascular endothelium of pulmonary arteries (PAs). 20-HETE enhances reactive oxygen species (ROS) production of bovine PA endothelial cells (BPAECs) in an NADPH oxidase-dependent manner and is postulated to promote angiogenesis via activation of this pathway in systemic vascular beds. We tested the capacity of 20-HETE or a stable analog of this compound, 20-hydroxy-eicosa-5( Z),14( Z)-dienoic acid, to enhance survival and protect against apoptosis in BPAECs stressed with serum starvation. 20-HETE produced a concentration-dependent increase in numbers of starved BPAECs and increased 5-bromo-2′-deoxyuridine incorporation. Caspase-3 activity, nuclear fragmentation studies, and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays supported protection from apoptosis and enhanced survival of starved BPAECs treated with a single application of 20-HETE. Protection from apoptosis depended on intact NADPH oxidase, phosphatidylinositol 3 (PI3)-kinase, and ROS production. 20-HETE-stimulated ROS generation by BPAECs was blocked by inhibition of PI3-kinase or Akt activity. These data suggest 20-HETE-associated protection from apoptosis in BPAECs required activation of PI3-kinase and Akt and generation of ROS. 20-HETE also protected against apoptosis in BPAECs stressed by lipopolysaccharide, and in mouse PAs exposed to hypoxia reoxygenation ex vivo. In summary, 20-HETE may afford a survival advantage to BPAECs through activation of prosurvival PI3-kinase and Akt pathways, NADPH oxidase activation, and NADPH oxidase-derived superoxide.

Abstract:
I will analyze the transcript from the 2000 BPAC meeting on the reevaluation of the MSM deferral policy and will elucidate the role that scientific data, specifically the data associated with NAT, plays in the 2000 deliberations surrounding the MSM deferral. This examination reveals that while scientific data did play a significant role in the decision making process of the BPAC there were also a number of other factors that influenced their deliberations as well. Ultimately what I will argue is that there were two different platforms present in the meeting and that each platform performs and enacts the body and blood of the donor differently. One platform is all about the inclusionary principles of blood donation and is most concerned by the potential for discrimination toward the body of the donor. The other platform is all about risk regulation and economics and is most concerned about how the body and blood of the donor will affect the safety and integrity of the blood supply. These platforms are not perspectival or dependent on view; this is not an epistemological argument but rather, an ontological one that concerns the reality and materiality of the situation, not the perspective. Thinking about these two platforms gives a handle to the nature of this controversy and contextualizes the committee’s decision to continue with the MSM deferral.

Abstract:
Segregation of duties and least privilege are two business principles that protect an organisation’s valuable data from information leak. In this thesis we demonstrate how these business principles can be addressed through workflow-based access control. We present Business Process Access Control (BPAC), a workflow-based access control modelling environment that properly enacts the key business principles through constraints and we implement BPAC in the applied pi calculus. We ensure that constraints are correctly applied within our BPAC implementation by introducing the concept of stores. We propose a selection of security properties in respect of the business principles and we develop tests for these properties. The collusion metric is introduced as a simple indicator as to the resistance of a workflow-based access control policy to fraudulent collusion. We identify an anonymity property for workflows as the inability of an outside observer to correctly match agents to workflow tasks and we propose that anonymity provides protection against collusion. We introduce a lightweight version of labelled bisimilarity: the abstraction test and we apply this test to workflow security properties. We develop a test for anonymity using labelled bisimilarity and we demonstrate its application through simple examples.

Abstract:
In der Optogenetik werden genetisch codierbare Photorezeptorproteine in Zellen eingebracht, um spezifische Parameter durch Licht zu kontrollieren. Um den sekundären Botenstoff zyklisches Adenosinmonophosphat (cAMP) zugänglich zu machen, haben wir vier neu identifizierte photoaktivierte Adenylatzyklasen untersucht. Die vier Gene stammen aus einem Sulfid oxidierenden Bakterium Beggiatoa und aus einem Amoeboflagellaten Naegleria gruberi. Sie codieren für 350 bis 489 Aminosäuren lange Proteine und enthalten eine Blaulicht wahrnehmende BLUF- (blue light sensors using FAD) und eine Adenylatzyklasedomäne. Zellbasierte Funktionstests in E. coli, Xenopus Oozyten und CHO Zellen sowie die Bestimmung der Adenylatzyklase-Aktivität in vitro zeigen, dass alle vier PACs lichtaktiviert arbeiten. Dabei besitzt die bakterielle PAC (bPAC) das größte Potenzial für eine optogenetische Anwendung, da sie eine hohe Lichtaktivität und ein große Licht-zu-Dunkel-Aktivitätsverhältnis besitzt. Um den Vorteil bPACs gegenüber PACalpha aus Euglena zu demonstrieren, verglichen wir beide PACS in Xenopus Oozyten, in Ratten Neuronen und in Drosophila Fliegen. Gegenüber PACalpha besitzt bPAC eine geringere Aktivität im Dunkeln, eine höhere Lichtsensitivität und einen breiteren dynamischen Bereiche und ist damit für viele Anwendungen die bevorzugte PAC. In einem analytischen Ansatz untersuchten wir die Bedeutung der BLUF-Domänen für die Regulation von bPAC. Nach Entfernen der BLUF-Domänen verblieb nur die inaktive Zyklase. Mutationen innerhalb der BLUF-Domäne, die die Photochemie oder die Signaltransduktion beeinflussten, führten zur Ausbildung eines partiell aktiven Rezeptorgrundzustandes. Dies verdeutlicht die Bedeutung eines intakten Wasserstoffbrückennetzwerkes für die funktionelle Regulation von bPAC. Als Ergenbis einer anwendungsorientierten Optimierung wurden eine bPAC Variante mit reduzierter Dunkelaktivität sowie eine Variante erhalten, die ein stabiles, neutrales Flavinradikal ausbildet.<br>Optogenetics represents the use of genetically encoded photoreceptor proteins to control specific parameters of cells by light. To particularly access optic control of the second messenger cyclic adenosine monophosphate (cAMP) we tested the suitability of four newly identified photoactivated adenylyl cyclases (PAC). Their genes originate from a soil bacterium Beggiatoa sp. and from an amoeboflagellate Naegleria gruberi. They code for 350 to 489 aa small proteins consisting of one blue light sensing BLUF (blue light sensors using FAD) and one adenylyl cyclase domain. Cell based functional assays in E. coli, Xenopus oocytes and CHO cells as well as in vitro analysis of recombinant protein reveal that all four PACs are light activated adenylyl cyclases. Moreover the bacterial PAC (bPAC) has the highest potential for an optogenetic application due to its high activity in light and low activity in the dark. To demonstrate the advantage of bPAC over the established Euglena PACalpha we compared both proteins in Xenopus oocytes, rat neurons and Drosophila fruit flies. It appears that bPAC features a lower activity in the dark, a higher light sensitivity and a broader dynamic range. Therefore bPAC is the superior optogenetic tool for many applications. In an analytical approach we investigated the impact of the BLUF domains on regulation of the cyclase. Removal of the BLUF domains left the bPAC cyclase in an inactive state. Mutating BLUF residues, which are involved either in photochemistry or signal transduction, resulted in a partially active dark state and reduced light activation of bPAC. Thus the integrity of the BLUF domain hydrogen bond network is essential for functional regulation of bPAC. As a result from application oriented optimization a bPAC variant with reduced dark activity as well as a variant that forms a stable neutral flavin radical was obtained.

In fact, if you select Extras from the top menu, you will see an item marked EAMIR among the other extras. 1. Click on Extras&gt;EAMIR from the top menu to view the main menu of the EAMIR SDK 2. Abstract:
It is known that treatment of endothelial cells (ECs) with extracts of malignant cells results in an increased level of tPA release by such ECs. On the other hand, no data are available concerning an influence of ECs on the fibrinolytic activity (PA) of malignant cells. Therefore it was the aim of this investigation to study whether the tPA production of malignant cells is influenced by ECs. Eighteen different HMCLs were screened for their ability to produce tPA using a combined assay system for tPA activity and tPA antigen employing monoclonal anti tPA antibodies. All 18 HMCLs produced tPA antigen ranging from 0.2 to 15.5ng/105 cells/24 hours. Twelve of these cells produced also tPA activity from 0.2 to 5.0 IU/105 cells/24 hours. To determine the influence of ECs on the PA of HMCLs, BPAECs were selected because their PA consists mainly of urokinase type plasminogen activator (uPA). All 18 HMCLs were cultivated on monolayers of BPAECs and as a control on plastic. After 24 hours the average tPA antigen level in the supernatant for the HMCLs grown on monolayers of BPAECs was 4.2ng/105 cells as compared to 2.9ng/105 cells of the control. However, the tPA activity average level was &lt; 0.1 IU/105 cells for the HMCLs grown on BPAECs and 0.9 IU/105 cells for the HMCLs grown on plastic. To further investigate these effects two HMCLs were grown on plastic and incubated with conditioned media (CM) of BPAECs and compared to non conditioned medium (NCM). After 24 hours the levels of tPA antigen in the supernatant were 194 and 28.5ng/l05 cells after incubation with CM and compared to 14.0 and 14.9ng/105 cells for the control. In contrast to the first experiment the tPA activity level was also increased after incubation with CM. The respective values were 9.5 and 11.6 IU/105 cells and 3.8 and 4.8 IU/105 cells for the control. Our data suggest that BPAECs release a "factor" which is stimulating both the release of tPA activity and tPA antigen. This "factor" remains to be further characterized. On the other hand our results also suggest that BPAECs might release a plasminogen activator inhibitor (PAI) in its active form inhibiting the tPA simultanously released by cocultivated HMCLs.

Abstract This paper presents the results of ASME Boiler and Pressure Vessel Code (BPVC) Section VIII Division 2 analyses on a containment pressure vessel system designed to host small-scale shock physics experiments containing hazardous materials. These shock physics experiments are imaged using proton radiography at Los Alamos Neutron Science Center (LANSCE). The pressure vessel system examined is separated into two subsystems, the Outer Pressure Containment Vessel (OPCV) and the Beam Pipe and Auxiliary Hardware (BPAH). The OPCV mates with the BPAH to transport the proton beam and form the outer containment pressure boundary. An Inner Pressure Confinement Vessel (IPCV) is mounted inside the containment pressure boundary. The IPCV can be used to host a variety of physics experiments, which can include high explosive and hazardous materials. The IPCV is an impulsively loaded pressure vessel, while the OPCV and beam pipes are statically loaded. The OPCV and BPAH are designed to satisfy ASME BPVC Section VIII Division 2 Alternative Rules, Part 5 Design by Analysis Requirements for all load conditions expected during service. The BPAH is a series of stainless-steel tubes with low-profile flange assemblies, bellows, and aluminum windows to allow for proton imaging. The low-profile flanges are required for clearance of magnet bores located periodically along the length of the BPAH. The bellows are included along the beamline for ease of installation. The OPCV is a pressure vessel designed to interface with the BPAH, IPCV, and proton imaging alignment system. The OPCV and BPAH are both designed to support full vacuum loads. Major considerations for the BPAH are thermal elongation, collapse of bellows, and stresses due to clamp supports. Lifting loads and static equivalent loads transmitted from the IPCV are additional considerations for the OPCV. Under ASME BPVC Section VIII Division 2 Part 5.2.4, Elastic-Plastic Stress Analysis Method, Paragraph 5.2.4.3(a), Global Criteria, the requirements for analysis and acceptance are defined for the load cases described. A finite element analysis (FEA) model, performed in ANSYS 2020 R1, evaluates the design of the OPCV and BPAH under the aforementioned ASME guidance and criteria.

INTRODUCTION Rifamipcin-resistant tuberculosis (RR-TB) affects around 465,000 people each year globally. Current treatment is of 9-20 months’ duration; is toxic and poorly efficacious. TB-PRACTECAL is a multi-arm, 2-stage, randomised controlled, multi-country, non-inferiority trial comparing 24-week regimens to the locally approved standard of care (control). We report TB-PRACTECAL stage 1 and 2 outcomes as well as additional analyses from dropped arms. METHODS Participants 15 years and above with pulmonary RR-TB from Uzbekistan, South Africa, and Belarus were included regardless of HIV status or CD4 count. Patients were randomized in a 1:1:1:1 ratio in stage 1 and 1:1 in stage 2. Randomization lists were stratified according to trial site. The BPaL regimen was comprised of bedaquiline 400mg daily for 2 weeks then 200mg three times weekly for 22 weeks, pretomanid 200mg daily for 24 weeks, and linezolid 600mg daily for 16 weeks followed by 300mg daily for 8 weeks. BPaLM additionally contained moxifloxacin 400mg daily and BPaLC contained clofazimine 100mg daily. Treatment was administered daily under observation. Transition to stage 2 occurred after enrolment of 240 participants and BPaLM was found to be the most promising arm. Randomisation continued during transition and all participants continued their allocated regimen and were followed up to 108 weeks. A post-hoc analysis was conducted comparing the three investigational arms to control using the primary efficacy outcome: proportion of patients with unfavourable outcome (death, treatment discontinuation, treatment failure, recurrence, lost to follow-up) at 72 weeks. We also assessed the proportion of patients with grade =3 or serious adverse events (SAE) by 72 weeks and mean change in QT corrected using Fridericia’s formula (QTcF) at week 24. ETHICS This study was approved by the Ethics Review Board (ERB) of the London School of Hygiene and Tropical Medicine and the local ERBs in Uzbekistan, Belarus and South Africa; and by the MSF ERB. RESULTS In March 2021, TB-PRACTECAL was terminated for efficacy at which point, 552 patients were enrolled. In the modified intention-to-treat population (comprising all randomised patients dispensed study medication at least once, excluding patients who did not have microbiologically-proven RR-TB), 252 patients had reached 72 weeks of follow-up, 44.0% of whom were female and 22.6% were HIV positive. In the modified intention- to-treat population, the percentage of unfavourable outcomes was 48.5% (32/66) for control, 23.3% (14/60) for BPaL, 18.8% (12/64) for BPaLC, and 11.3% (7/62) for BPaLM. There were three recurrences in BPaL, one in BPaLC, and none in BPaLM. Percentage of Grade =3 or SAE were 19.4% (14/72; 16 events), 31.9% (23/72; 32 events) and 21.7% (15/69; 24 events) in BPaLM, BPaLC and BPaL respectively, compared with 58.9% (43/73; 69 events) in the control. Mean change in QTcF at week 24 was 27.0 milliseconds (ms), 40.2 ms, and 23.3 ms in BPaLM, BPaLC, and BPaL respectively; compared with 44.89 ms in the control. CONCLUSION 24-week all-oral regimens of bedaquiline, pretomanid and tapered-dose linezolid, with and without clofazamine or moxifloxacin are safe and efficacious in the treatment of RR-TB. Trial results show that treatment with BPaLM was more effective and had a better safety profile than the Control. BPaLC and BPaL were also highly effective. CONFLICTS OF INTEREST None declared.

Abstract Bovine pulmonary aortic endothelial cells (BPAEC) exposed to controlled levels of fluid shear stress in the absence of ATP exhibited transient intracellular free calcium (Ca2+i) responses that were sensitive to the onset rate of shear stress (ORSS). A coaxial parallel-disk device was used to control the application of a high (&amp;gt;900 dyn/cm s−1) or low (&amp;lt; 6 dyn/cm s−1) ORSS. The high rate elicited greater peak responses and final levels of intracellular free calcium concentrations, [Ca2+]i, compared to the low rate. The ORSS appears to be an important parameter characterizing the mechanical environment of endothelial cells and may play a role in their differential responses in disturbed versus undisturbed flow regions.

Abstract A system of nested pressure vessels is being developed to facilitate proton imaging of small-scale shock physics experiments at Los Alamos National Laboratory (LANL). Proton imaging provides valuable experimental data to characterize materials under extreme loading conditions. The main components of the nested system are the Inner Pressure Confinement Vessel (IPCV, which hosts the physics experiment), the Outer Pressure Containment Vessel (OPCV) and Beam Pipes and Auxiliary Hardware (BPAH). IPCV, OPCV, and BPAH together form a Pressure Vessel Containment System (PVCS). The detonation of high explosives (HE) inside the IPCV drives material inside the physics experiment package to extreme loading conditions, which are imaged using a proton beam and an imaging system. The IPCV is an explosively loaded high-pressure vessel which is rigidly mounted into the OPCV. The OPCV is a statically loaded vacuum vessel, where the OPCV and beam pipes form the containment barrier and are under vacuum for the purpose of proton imaging. The OPCV bellows are connected to a rotary platform allowing linear and torsional movements within a range of motion for alignment of the OPCV/IPCV assembly within a fixed beamline. The OPCV is supported by a novel alignment system which allows six degrees of freedom of motion to locate the experiment package relative to the proton beam for precise imaging. The IPCV is designed to the ASME Boiler and Pressure Vessel Code, Section VIII, Division 3, Code Case 2564. The aim is to make the confinement boundary Code-compliant while allowing for maximum resolution of proton radiography across a sufficiently large field of view. The nested vessels and beam pipes are connected to the Vacuum and Gas Handling System (VGHS). This paper provides an overview of the final design, analysis, and testing of the PVCS and its subassemblies.

Abstract:
INTRODUCTION The total duration of treatment for rifampicin-resistant tuberculosis (RR-TB) in Belarus prior to December 2022 was 18-20 months. The efficacy of treatment with such regimens is low, with the WHO’s Global TB Report suggesting that efficacy was around 73% in Belarus in 2018. The development of effective short regimens for RR-TB treatment is urgent. In Belarus, six-month long treatment with all-oral regimens is used in patients with RR-TB, under operational research conditions, following WHO recommendations. METHODS A preliminary assessment of the effectiveness of six-month all-oral regimens containing bedaquiline, pretomanid, linezolid, and moxifloxacin or clofazimine (BPaLM/BPaLC), was performed in a cohort of RR-TB patients. Treatment outcomes, time to culture conversion, and time to adverse event (AE) occurrence, AE types, frequency, and outcomes are described. ETHICS This study was approved by the MSF Ethics Review Board (ERB) and by the Belarus Ministry of Health ERB. RESULTS Of 177 patients who were enrolled from February 2022 to July 2022, one patient was excluded due to linezolid resistance; this patient continued treatment under an individualised regimen. Of the rest of the cohort (133 (76%) male, 43 female (24%); median age, 44 years (interquartile range, IQR, 25-29 years), 93.2% (164/176) had a favourable treatment outcome, 11 patients were lost to follow-up, and one died. 52 (30%) patients had a sputum smear positive result at treatment start, 59 (34%) a cavitary lesion on chest X-ray, and 42 (24%) patients had been previously treated. 12 patients (7%) were HIV-positive; 23 (13%) had had hepatitis C infection; 45 (26%) abused alcohol, and 6 (3%) of patients had diabetes mellitus. Median time to culture conversion was 27 days (IQR, 25-29). In 96.0% of patients, culture conversion was achieved within 2 months of treatment. 9% of patients had serious AE’s (SAE). Out of total 19 SAE’s, 12 resolved, two resolved with sequelae, three were resolving at the time of assessment, one did not resolve, and one was fatal. Median time from treatment start to the first SAE was 92.5 days (IQR, 12.5-143). The most frequent SAE’s were elevated liver function (6 (32%) cases), acute kidney injury (4 (21%) cases), and amylase increased/pancreatitis (3 (16%) cases). Two cases also revealed cancer or progression of cancer; one showed QTcF prolongation; one, anemia, one, thrombotic cerebral infarction, and one, Clostridium difficile infection. Two cases of cancer and thrombotic cerebral infarction (a patient with a long-standing history of multiple strokes) were assessed as unrelated to study drugs. The one death from cancer was assessed as not related to treatment. Permanent withdrawal of one study drug (linezolid or clofazimine) was done only in three instances (15.8%). CONCLUSION The effectiveness of six-month all-oral regimens in this cohort was very high (93.2%). BPaLM/BPaLC regimens were observed to be characterised by a good safety profile. Abstract:
Abstract A unique containment and confinement system is under development to conduct small explosively driven physics experiments containing hazardous materials at the Proton Radiography facility at Los Alamos National Laboratory (LANL). In these experiments, the detonation of high explosives (HE) is used to drive materials to extreme loading conditions, where some of the materials tested can be extremely hazardous (e.g. nuclear materials). The main components of the system are the Inner Pressure Confinement Vessel (IPCV, which hosts the physics experiment), the Outer Pressure Containment Vessel (OPCV) and Beam Pipes and Auxiliary Hardware (BPAH). This paper describes the design and preliminary analyses of the IPCV. The body of the IPCV, also referred to as the Inner Vessel, is being designed to the criteria of the ASME Boiler and Pressure Vessel Code, Section VIII, Division 3, Code Case 2564, with the exception of the materials of construction. The closure covers have different devices mounted on them, such as feedthrough devices for sending or receiving electrical and optical signals across the pressure boundary, and valves for venting the vessel interior. The unique feature in the vessel design are the radiographic windows, tentatively made of Beryllium, which need to be strong enough to maintain the pressure boundary during dynamic events, while being radiographically low-attenuating for the purpose of proton imaging.
