Abstract:
ABSTRACT BRUCE is a highly conserved 528-kDa peripheral membrane protein of the trans-Golgi network. Owing to the presence of an N-terminal single baculovirus inhibitor repeat, BRUCE functions as an inhibitor of apoptosis protein and blocks apoptosis when overexpressed. In addition, due to the presence of a C-terminal ubiquitin-conjugating domain, BRUCE can covalently attach ubiquitin to substrates. Here we report the generation and characterization of BRUCE-deficient mice. Complete inactivation of the BRUCE gene resulted in perinatal lethality and growth retardation discernible after embryonic day 14. The growth defect is linked to impaired placental development and may be caused by insufficient oxygen and nutrient transfer across the placenta. Chorioallantoic placentation initiated normally, but the mutant placenta showed an impaired maturation of the labyrinth layer and a significant reduction of the spongiotrophoblast. No evidence for an elevated apoptosis rate was detectable in embryonic and extraembryonic tissues and in knockout fibroblasts. Thus, although BRUCE is broadly expressed in embryonic, extraembryonic, and adult mouse tissues, this bifunctional protein might play a unique role in normal trophoblast differentiation and embryonic survival.

AbstractLet (V,0) be the germ of an analytic variety in $\mathbb{C}^n$ and f an analytic function germ defined on V. For functions with isolated singularity on V, Bruce and Roberts introduced a generalization of the Milnor number of f, which we call Bruce–Roberts number, μBR(V,f). Like the Milnor number of f, this number shows some properties of f and V. In this paper we investigate algebraic and geometric characterizations of the constancy of the Bruce–Roberts number for families of functions with isolated singularities on V. We also discuss the topological invariance of the Bruce–Roberts number for families of quasihomogeneous functions defined on quasihomogeneous varieties. As application of the results, we prove a relative version of the Zariski multiplicity conjecture for quasihomogeneous varieties.

Abstract:
The DNA damage response (DDR) is crucial for genomic integrity. BRIT1 (breast cancer susceptibility gene C terminus-repeat inhibitor of human telomerase repeat transcriptase expression), a tumor suppressor and early DDR factor, is recruited to DNA double-strand breaks (DSBs) by phosphorylated H2A histone family, member X (γ-H2AX), where it promotes chromatin relaxation by recruiting the switch/sucrose nonfermentable (SWI–SNF) chromatin remodeler to facilitate DDR. However, regulation of BRIT1 recruitment is not fully understood. The baculovirus IAP repeat (BIR)-containing ubiquitin-conjugating enzyme (BRUCE) is an inhibitor of apoptosis protein (IAP). Here, we report a non-IAP function of BRUCE in the regulation of the BRIT1–SWI–SNF DSB-response pathway and genomic stability. We demonstrate that BRIT1 is K63 ubiquitinated in unstimulated cells and that deubiquitination of BRIT1 is a prerequisite for its recruitment to DSB sites by γ-H2AX. We show mechanistically that BRUCE acts as a scaffold, bridging the ubiquitin-specific peptidase 8 (USP8) and BRIT1 in a complex to coordinate USP8-catalyzed deubiquitination of BRIT1. Loss of BRUCE or USP8 impairs BRIT1 deubiquitination, BRIT1 binding with γ-H2AX, the formation of BRIT1 DNA damage foci, and chromatin relaxation. Moreover, BRUCE-depleted cells display reduced homologous recombination repair, and BRUCE-mutant mice exhibit repair defects and genomic instability. These findings identify BRUCE and USP8 as two hitherto uncharacterized critical DDR regulators and uncover a deubiquitination regulation of BRIT1 assembly at damaged chromatin for efficient DDR and genomic stability.

Abstract:
Over eighty polar-themed exhibitions were held in Britain between 1819 and the 1930s, a time of intense exploration of both the Arctic and Antarctic. These varied from panoramas and human exhibits to displays of ‘relics', equipment, photographs and artwork, waxworks and displays shown as part of a Great Exhibition. This period also saw the creation of the first dedicated polar museums. These displays were visited by thousands of people throughout the country, helping to mediate the subject of exploration for a public audience. Despite this, the role exhibitions played in forming popular views of the polar regions has not been fully assessed. This thesis addresses this gap. It is the first to consider all the polar exhibitions held during this period as a collective body, making it possible to study how they developed over time and in response to changing circumstances. The thesis uses a variety of archival sources to both reconstruct the displays and place them in their historical and museological contexts. The study shows that exhibitions evolved in response to changes both in the museum sector and in exploration culture. It demonstrates that, while they were originally identified with the shows of the entertainment industry, polar exhibitions began to take on more of the characteristics of museum displays. At the same time their dominant themes changed; the natural world was relegated in favour of ideas relating to the human experience of the regions such as heroism, adventure and everyday life in an exotic environment. While other media may have been more effective in disseminating ideas about exploration, visitors could find the experience of visiting an exhibition more compelling. This thesis contributes to our understanding of this distinct role that exhibitions played in presenting the polar regions to the British public.

Abstract:
One of Africa’s neglected tropical diseases, African Trypanosomiasis, is not only fatal but also has a crippling impact on economic development. Heat shock proteins play a wide range of roles in the cell and they are required to assist the parasite as it moves from a cold blooded insect vector to a warm blooded mammalian host. The expression of heat shock proteins increases during these heat shock conditions, and this is considered to play a role in differentiation of these vector-borne parasites. Heat shock protein 70 (Hsp70) is an important molecular chaperone that is involved in protein homeostasis, Hsp40 acts as a co-chaperone and stimulates its intrinsically weak ATPase activity. In silico analysis of the T. brucei genome has revealed the existence of 12 Hsp70 proteins and 65 Hsp40 proteins to date. A novel Hsp70, TbHsp70.c, was recently identified in T. brucei. Different from the prototypical Hsp70, TbHsp70.c contains an acidic substrate binding domain and lacks the C-terminal EEVD motif. By implication the substrate range and mechanism by which the substrates are recognized may be novel. The ability of a Type I Hsp40, Tbj2, to function as a co-chaperone of TbHsp70.c was investigated. The main objective of this study was to biochemically characterize TbHsp70.c and its partnership with Tbj2 to further enhance our knowledge of parasite biology. TbHsp70.c and Tbj2 were heterologously expressed and purified and both proteins displayed chaperone activities in their ability to suppress aggregation of thermolabile MDH. TbHsp70.c also suppressed aggregation of rhodanese. ATPase assays revealed that the ATPase activity of TbHsp70.c was stimulated by Tbj2. The targeted inhibition of the function of heat shock proteins is emerging as a tool to combat disease. The small molecule modulators quercetin and methylene blue are known to inhibit the ATPase activity of Hsp70. However, methylene blue did not significantly inhibit the ATPase activity of TbHsp70.c; while quercetin, did inhibit the ATPase activity. In vivo heat stress experiments indicated an up-regulation of the expression levels of TbHsp70.c. RNA interference studies showed partial knockdown of TbHsp70.c with no detrimental effect on the parasite. Fluorescence microscopy studies of TbHsp70.c showed a probable cytoplasmic subcellular localization. In this study both TbHsp70.c and Tbj2 demonstrated chaperone activity and Tbj2 possibly functions as a co-chaperone of TbHsp70.c.

Abstract:
Le trypanosome sud américain, Trypanosoma cruzi, transmis par un insecte hématophage de type triatome est le protozoaire connus pour causer la maladie de Chagas chez l’Homme. Le cycle de vie de ce parasite alterne à la fois sur le type d’hôte, insecte ou hôte vertébré, et sur la forme :trypomastigote pour la forme quiescente et amastigote et épimastigote pour les formes prolifératives. Concernant la forme, seuls les parasites épimastigotes évoluent et prolifèrent dans le tube digestif des triatomes et possèdent un système endocytaire actif nécessaire à leur besoin énergétique. Toutefois, cette endocytose est restreinte à deux sites membranaires, la poche flagellaire et le cytostome, à partir desquels se créent des cargos endocytaires. Ces cargos endocytaires fusionnent ensuite avec un réseau vésiculaire endosomal qui délivre son contenu dans des réservosomes, compartiments similaires aux lysosomes.Chez le trypanosome africain (Trypanosoma brucei brucei), l’endocytose ne se réalise qu’au niveau de la poche flagellaire. Certaines protéines appartenant à cette voie endocytaire sont modifiées par de longues chaines de résidus poly-N-acétyllactosamine (pNAL) de manière post-traductionnelle. Initialement, il a été proposé que ces résidus puissent agir en tant que signal de tri dans le processus d’endocytose chez ces parasites.En nous basant sur les travaux qui ont été réalisés chez le trypanosome africain, nous nous sommes proposés d’approfondir les connaissances sur la voie endocytaire du trypanosome sud américain (Trypanosoma cruzi) qui est beaucoup moins étudié. Pour ce faire, à l’aide de deux lectines, la tomatolectine et la lectine de Griffonia simplicifolia qui présentent respectivement une affinité pour les résidus pNAL et les résidus N-acétylglucosamine (GlcNAc) en fin de chaine, nous avons pu enrichir et caractériser par LC-MS², 173 glycoprotéines putatives dont plus de 13% sont localisées dans la voie endocytaire. Parmi les protéines identifiées, en plus des nombreuses hydrolases lysosomiales, nous avons pu identifier une lectine de type C localisée dans la partie antérieure des parasites, au niveau des principaux sites endocytaires. Cette dernière possédant de nombreux résidus en commun avec les récepteurs de type scavengers, elle pourrait donc jouer un rôle important dans la fixation et l’endocytose de certains nutriments.Nos travaux ont ainsi permis d’établir que similairement aux trypanosomes africains, Trypanosoma cruzi possèdent des glycoprotéines modifiées par des N-glycanes contenant des pNAL. Nos travaux ont également permis d’établir que ces résidus s’associent préférentiellement aux glycoprotéines de la voie endocytaire (au niveau du cytostome et du réservosome) de la forme épimastigote. L’ensemble des résultats obtenus durant cette thèse tendent à montrer que les résidus pNAL des glycoprotéines présentes dans la voie endocytaire ont été conservées entre les deux parasites étudiés (Trypanosoma cruzi et Trypanosoma brucei brucei).<br>Doctorat en Sciences<br>info:eu-repo/semantics/nonPublished

L’objet de ce travail est d’expliquer le comportement des dirigeants par rapport à la publication volontaire des flux de trésorerie dans le contexte français et de son impact sur la valeur de la firme. Tout d’abord, nous avons tenté d’expliquer ces comportements en se référant aux théories fondatrices de la firme, en l’occurrence la théorie d’agence, la théorie des coûts de transaction et la théorie des stakeholders. Cependant, ces théories supposent un rôle passif aux comportements des dirigeants, d’où l’intérêt d’analyser cette problématique dans le cadre de la théorie de l’enracinement et de la théorie du signal qui, en revanche, attribuent un rôle actif et stratégique aux comportements des dirigeants. De plus, nous avons réalisé une étude empirique sur le marché français afin d’expliquer les publications volontaires des flux de trésorerie par les dirigeants dans les comptes intermédiaires. Ainsi, nous avons supposé que les incitations des dirigeants à publier telle information peuvent être expliquées par des facteurs qui ont trait à la structure de la firme, son audit, ses performances et sa gouvernance. Nos résultats –portant sur 122 sociétés et couvrant la période [1996-2004]– montrent qu’effectivement les variables : taille, présence de pertes, présence d’auditeurs de renommée internationale et le pourcentage d’administrateurs indépendants peuvent expliquer les comportements des dirigeants vis-à-vis de la publication volontaire des flux de trésorerie. Par ailleurs, l’étude du comportement des dirigeants par rapport à la publication volontaire des flux de trésorerie ne peut être complètement appréhendée sans la prise en compte de l’impact de telle publication sur la valeur de la firme ; autrement dit, sans la considération des comportements du principal utilisateur de l’information comptable qui sont les investisseurs. Ainsi, nous avons tenté, dans le cadre de l’hypothèse d’efficience du marché financier, d’analyser le contenu informationnel des flux de trésorerie par rapport à d’autres mesures comptables, en l’occurrence les flux de fonds et le résultat net<br>This research aims to explain the behavior of firm managers as to the voluntary disclosure of cash flows in a French context as well as to evaluate the impact of such disclosures on the value of firms. We first attempted to account for managers' behaviors by referring to the firm's founding theories, i. E. Agency theory, transaction cost theory and stakeholders theory. However, these theories imply that firm managers have but a passive role; in this respect, it therefore seemed more appropriate to analyse the problem focusing on the entrenchment theory and signal theory, which, by contrast, cast managers in a more active and strategic role. We also carried out an empirical study in order to account for voluntary cash flow disclosure in interim reporting within the French market. Thus, we supposed that incentives to publish such information may be explained by factors related to firm structure, its audit, performance and governance. Our results –examining 122 companies over the [1996-2004] period– show that variables such as size, losses, internationally renowned audits and the ratio of independent directors do play a part in the voluntary disclosure of cash flow policy. In addition, the study of managers' behaviors as to the deliberate publication of cash flows cannot be fully grasped without acknowledging the impact of such a disclosure on the value of the firm, in other words, without taking into consideration the behaviors of the main users of the accounting information, to wit, investors. Thus, our purpose was to analyse the information content of cash flows compared to other accounting measurements, more specifically funds from operations and net earnings, within the efficient market hypothesis. We have studied accounting data published by companies listed on the SBF 120 index of Paris Euronext, and shown that cash flows do add incremental information to that provided about funds from operations and earnings. Additionally, they offer predictive capacity for future cash flows. Thus do cash flows constitute a reliable and relevant piece of information in the evaluation of firm performance, hence their interest for investors

Abstract:
When Argentina invaded the Falkland Islands in 1982, it was the beginning of a two month long war. The aim of this essay is to find alternative explanations to the Falklands war with the main question being whether there were hidden motives for Thatcher and her reactions in 1982.   The essay will focus on the period 1965-1982 in which different texts will be analyzed by applying theories. The diversionary war theory describes how state leaders can improve their political popularity by intentionally escalating a conflict. The geopolitical theory explains how a state, by investments, constructions and military, can create demographic and international perceptions regarding a territory while foreign political theory describes how decision making is affected by the bureaucracy, psychology and the international system.   The ministry of defense and the navy was heavily affected by the financial cuts during the conflict and may also have affected the manner in which they advised Thatcher in 1982. Because of the British unwillingness to invest geopolitically in the Falklands and their aggravation of the diplomatically efforts to find a peaceful solution, the conflict got worse. In addition, Thatchers intentions to implement economic reforms and her record low political popularity make it plausible to suspect her for having, intentionally, escalated the conflict to gain political popularity and to enable the economic reforms.

Abstract:
[EN] Small dwelling and urban form. German modern city bases. Small dwelling and urban form are the main concepts in which this research is based. The intention is to determine the characteristics of the German city where the modern architecture of Neues Bauen is developed. The small dwelling, as a new type lacking in an immediate historical precedent, arises from the need to house a new class of the industrial proletariat. The urban form as a different configuration which the city acquires. This is a result of the transformations that its growth demands in order to adapt to a new social reality. In Germany, as in the rest of Europe, the origin of the small dwelling came from the need to improve the living conditions of the worker. A reaction that was produced mainly from philanthropy but also as a guarantee in keeping some established social conditions. The movement for housing reform, set out by private initiatives, effectively solves new forms for a small, hygienic and functional dwelling that was accessible to a part of the working masses, establishing solid and functional criteria that would be maintained as the principles of a typology in the configuration of the modern dwelling at the beginning of the 20th century. The great German city, represented mainly by Berlin in this research, outlines its expansion through an imported urban idea that has, as its main interest, the monumental expression of the urban space. With a system originated in the interior reform processes of the French city, it outlines the exterior expansion of the German city as a continued imposing construction, in which the small dwelling only constitutes a dense internal filling. This "petrous sea", as Muthesius defines a city that extends concentrated to its limits, would be seen as incompatible with the small dwelling that was needed. Once this inconsistency was recognized and with a new scientific approach to the construction of the city, a transformation process was begun towards a new urban model that took into consideration the need for a small and healthy dwelling and returns to the citizen the contact with nature, a sense that is a deeply ingrained in German people. From the ideas and actions of the leading figures in this process, gathered in original documents, the research advances in the recognition of the typical and essential characteristics of a German city, that, at the start of the third decade of the 20th century, bases its modern quality in common criteria relative to the separation of urban functions, the insertion of green in the city and the value of public space. On this modern German city of first post-war Europe, the specific solution of the architecture of Neues Bauen will be built.<br>[ES] Vivienda pequeña y forma urbana. Fundamentos de la ciudad moderna alemana. Vivienda pequeña y forma urbana son los conceptos sobre los que se fundamenta esta investigación que quiere determinar las características de la ciudad alemana en la que se desarrolla la arquitectura moderna del Neues Bauen. La vivienda pequeña como un nuevo tipo carente de un antecedente histórico inmediato, que surge de la necesidad de alojar a la nueva clase del proletariado industrial. La forma urbana como la distinta configuración que adquiere la ciudad, consecuencia de las transformaciones que exige su crecimiento para adecuarse a una nueva realidad social. En Alemania, como en el resto de los países de Europa, la vivienda pequeña tiene su origen al reconocerse la necesidad de mejora de las condiciones de vida del trabajador. Una reacción que se produce desde la filantropía principalmente, pero también como garantía del mantenimiento de unas condiciones sociales establecidas. El movimiento para la reforma de la vivienda, planteado desde iniciativas particulares, resuelve de manera eficaz nuevas formas para una vivienda pequeña, higiénica, funcional y accesible a una parte de esta masa obrera, estableciendo sólidos criterios funcionales que se mantienen como principios de una tipología en la configuración de la vivienda moderna en los comienzos del siglo XX. La gran ciudad alemana, representada en esta investigación por Berlín principalmente, plantea su expansión sobre una idea urbana importada, que tiene en la expresión monumental del espacio urbano su mayor interés. Con un sistema originado en los procesos de reforma interior de la ciudad francesa, se traza la ampliación exterior de la ciudad alemana como un continuo edificado imponente, en el que la vivienda pequeña sólo constituye un relleno interior denso. Este "mar pétreo", como define Muthesius a una ciudad que se extiende concentrada hasta su límite, se mostrará incompatible con la vivienda pequeña que se necesita. Desde el reconocimiento de esta incoherencia y con una nueva aproximación científica a la construcción de la ciudad, se inicia el proceso de transformación hacia un nuevo modelo urbano que considere la necesidad de la vivienda pequeña saludable y retorne al ciudadano el contacto con la naturaleza, un sentimiento vernáculo de gran arraigo en el pueblo alemán. A partir del estudio de las ideas y actuaciones de los protagonistas de este proceso, recogidas en documentos originales, la investigación avanza en el reconocimiento de las características propias y fundamentales de una ciudad alemana que, en el comienzo de la tercera década del siglo XX, basa su cualidad de moderna en criterios comunes relativos a la separación de las funciones urbanas, la inserción del verde en la ciudad y el valor del espacio público. Sobre esta ciudad moderna alemana de la primera posguerra europea se construirá la específica solución de la arquitectura del Neues Bauen.<br>[CAT] Habitatge petit i forma urbana. Fonaments de la ciutat moderna alemanya. Habitatge petit i forma urbana són els conceptes sobre els quals es fonamenta aquest treball de recerca, que vol determinar les característiques de la ciutat alemanya en la qual es desenvolupa l'arquitectura moderna del Neues Bauen. L'habitatge petit com un nou tipus mancat d'un antecedent històric immediat, que sorgeix de la necessitat d'allotjar la nova classe del proletariat industrial. La forma urbana com la diferent configuració que adquireix la ciutat, conseqüència de les transformacions que exigeix el creixement d'aquesta per a adequar-se a una nova realitat social. A Alemanya, com a la resta dels països d'Europa, l'habitatge petit té l'origen en el reconeixement de la necessitat de millora de les condicions de vida dels treballadors. Una reacció que es produeix principalment des de la filantropia, però també com a garantia del manteniment d'unes condicions socials establides. El moviment per a la reforma de l'habitatge, plantejat des d'iniciatives particulars, resol de manera eficaç noves formes per a un habitatge petit, higiènic, funcional i accessible a una part d'aquesta massa obrera, i estableix uns criteris funcionals sòlids que es mantenen com a principis d'una tipologia en la configuració de l'habitatge modern en els inicis del segle xx. La gran ciutat alemanya, representada en aquest treball per Berlín principalment, es planteja l'expansió sobre una idea urbana importada que té el major interès en l'expressió monumental de l'espai urbà. Amb un sistema originat en els processos de reforma interior de la ciutat francesa, l'ampliació exterior de la ciutat alemanya es traça com un contínuum edificat imposant, en el qual l'habitatge petit només constitueix un farciment interior dens. Aquesta "mar pètria", com Muthesius defineix una ciutat que s'estén concentrada fins al límit, es mostrarà incompatible amb l'habitatge petit que es necessita. Des del reconeixement d'aquesta incoherència i amb una nova aproximació científica a la construcció de la ciutat, s'inicia el procés de transformació cap a un nou model urbà que considere la necessitat de l'habitatge petit saludable i retorne al ciutadà el contacte amb la natura, un sentiment vernacle de gran arrelament en el poble alemany. A partir de l'estudi de les idees i actuacions dels protagonistes d'aquest procés, recollides en documents originals, la recerca avança en el reconeixement de les característiques pròpies i fonamentals d'una ciutat alemanya que, al començament de la tercera dècada del segle xx, basa la seua qualitat de moderna en criteris comuns relatius a la separació de les funcions urbanes, la inserció del verd a la ciutat i el valor de l'espai públic. Sobre aquesta ciutat moderna alemanya de la primera postguerra europea es construirà la solució específica de l'arquitectura del Neues Bauen.<br>Bosch Abarca, J. (2016). Abstract:
The chapter examines the multiple representations, evolution and opposition of masculine constructions in John Barbour’s Brus (c.1375) which tells the story of Scotland’s First War of Independence. The two main heroes, Robert Bruce and James Douglas are shown to represent a fluid masculinity, adapting to the changing social and political circumstances of medieval Scotland. This is contrasted with the negative models of Edward I and Edward Bruce. The chapter discusses the results of the initial absence of proper leaders and male models, secondly the evolution of Bruce and Douglas into ideal king and knight, and finally the interactions of the male protagonists with women. Patriarchy is shown to operate not only in the power of men over women, but also in the authority that groups of men exercise over other groups according to social hierarchies.

After returning home from Europe in 1978, Bradbury was unable to come to agreement with the Smithsonian over “The Ghosts of Forever,” an animated film fantasy tour of the various Smithsonian museums. Chapter 12 goes on to document how the Voyager 1 and Voyager 2 journeys to the outer solar system prompted NASA’s Jet Propulsion Laboratory at Caltech to bring Bradbury back together with his “Mars and the Mind of Man” colleagues Arthur C. Clarke, Carl Sagan, and JPL director Bruce Murray to form the symposium “Jupiter and the Mind of Man.” The chapter also describes the uneven production and mixed reception of the NBC miniseries of The Martian Chronicles, and Bradbury’s Emmy-winning ABC collaboration with Malcolm Clarke on “Infinite Horizons: Space Beyond Apollo.”
