SummaryBackground: With the Act on the Reform of the Market for Medicinal Products (AMNOG) in Germany, pharmaceutical manufacturers are obliged to submit a dossier demonstrating added benefit of a new drug compared to an appropriate comparator. Underlying evidence was planned for registration purposes and therefore often does not meet the appropriate comparator as defined by the Federal Joint Committee (G-BA). For this reason AMNOG allows indirect comparisons to assess the extent of added benefit.Objectives: The aim of this study is to evaluate the characteristics and applicability of adjusted indirect comparison described by Bucher and Matching-Adjusted Indirect Comparison (MAIC) in various situations within the early benefit assessment according to §35a Social Code Book 5. In particular, we consider time-to-event endpoints.Methods: We conduct a simulation study where we consider three different scenarios: I) similar study populations, II) dissimilar study populations without interactions and III) dissimilar study populations with interactions between treatment effect and effect modifiers. We simulate data from a Cox model with Wei- bull distributed survival times. Desired are unbiased effect estimates. We compare the power and the proportion of type 1 errors of the methods.Results: I) Bucher and MAIC perform equiva- lently well and yield unbiased effect estimates as well as proportions of type 1 errors below the significance level of 5%. II) Both Bucher and MAIC yield unbiased effect estimates, but Bucher shows a higher power for detection of true added benefit than MAIC. III) Only MAIC, but not Bucher yields unbiased effect estimates. When using robust variance estimation MAIC yields a proportion of type 1 error close to 5%.In general, power of all methods for indirect comparisons is low. An increasing loss of power for the indirect comparisons can be observed as the true treatment effects decrease.Conclusion: Due to the great loss of power and the potential bias for indirect comparisons, head-to-head trials using the appropriate comparator as defined by the Federal Joint Committee should be conducted whenever possible. However, indirect comparisons are needed if no such direct evidence is available. To conduct indirect comparisons in case of a present common comparator and similar study populations in the trials to be compared, both Bucher and MAIC can be recommended. In case of using adjusted effect measures (such as Hazard Ratio), the violation of the similarity assumption has no relevant effect on the Bucher approach as long as interactions between treatment effect and effect modifiers are absent. Therefore Bucher can still be considered appropriate in this specific situation. In the authors’ opinion, MAIC can be considered as an option (at least as sensitivity analysis to Bucher) if such interactions are present or cannot be ruled out. Nevertheless, in practice MAIC is potentially biased and should always be considered with utmost care.

Abstract:
Among the different types of organic semiconducting materials, porphyrins, DPPs and BODIPYs based small molecules and conjugated polymers have attracted high interest as efficient semiconducting organic materials for dye sensitized solar cells (DSSC) and bulk heterojunction (BHJ) organic solar cells. Interestingly, they offer a synergistic effect when used together within a material, this coming from high complementarities of their absorption spectra, adapted prerequisites in order to be efficient energy transfer partners, and suitable frontier orbitals energy levels. We have recently designed different porphyrins, DPPs and BODIPYs based electron donor small molecules/polymers for bulk heterojunction organic solar cells (Figure), and now report their synthesis as well as the study of their electrochemical, photophysical and photovoltaic properties. ZusammenfassungDas Kapitel leitet in die Problemstellung des Buches ein: Foltersituationen als eine spezifische Form von sozialen Situationen, die durch extreme Gewalt und Asymmetrie geprägt sind. Als empirischer Fall dient die Folter durch den US-Geheimdienst CIA und Teile des US-Militärs im sogenannten War on Terror gegen Al-Qaida und die Taliban infolge der Terroranschläge vom 11. September 2001. Anhand des US-Folterkomplexes als ein Beispiel für moderne Folter durch liberal-demokratisch verfasste Staaten verfolgt das Buch folgende Leitfragen: Wie konstituieren verschiedene Elemente wie Menschen, Organisationen, Artefakte und räumlich-zeitliche Strukturen soziale Situationen als Foltersituationen? Welche Verflechtungen von (diskursivem) Wissen sind dabei relevant; Wissen etwa über ‚effektive‘ Verletzung, vermeintliche Eigenarten der zu folternden Feinde und rechtliche Normen? Inwieweit erleben sich Gefolterte als handlungsfähige Subjekte und inwieweit werden sie als solche adressiert?

Abstract:
Bucket Elimination (BE) is a universal inference scheme that can solve most tasks over probabilistic and deterministic graphical models exactly. However, it often requires exponentially high levels of memory (in the induced-width) preventing its execution. In the spirit of exploiting Deep Learning for inference tasks, in this paper, we will use neural networks to approximate BE. The resulting Deep Bucket Elimination (DBE) algorithm is developed for computing the partition function. We provide a proof-of-concept empirically using instances from several different benchmarks, showing that DBE can be a more accurate approximation than current state-of-the-art approaches for approximating BE (e.g. the mini-bucket schemes), especially when problems are sufficiently hard.

Abstract:
Two new tacrine/pyrazole conjugates were designed, synthesized, and pharmacologically evaluated for their inhibitory activity toward acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE). A scalable and cost-efficient synthetic route was developed, and key reaction steps for the synthesis of compounds 4a,b were nucleophilic substitution of α-aroylketene dithioacetals with tacrine intermediates, followed by cyclocondensation of respective N,S-acetals with hydrazine hydrate. The preliminary pharmacological evaluation revealed high inhibitory activities of 4a,b toward AChE (180 and 259 nM, respectively) and BuChE (51 and 95 nM, respectively) as compared with known inhibitor, tacrine. Overall, both compounds were more efficient BuChE inhibitors while 4a, with a shorter linker connecting tacrine and phenylpyrazole moieties, showed higher inhibitory activity toward both enzymes. Molecular docking analysis strongly corroborated pharmacological results since both compounds interacted favorably with target enzymes. Calculated pharmacokinetic properties (absorption, distribution, metabolism, and excretion (ADME) showed that 4a,b obey Lipinski’s rule of druglikeness and are promising lead compounds for the development of new drug candidates.

Abstract:
The composite bucket foundation of offshore wind power is a new type of foundation, which has been applied in practical engineering because of its advantages of large-scale prefabrication on land, integrated installation at sea and zero residual recovery. In order to overcome the shaft friction and tip resistance of the suction bucket, great passive suction is often applied during the penetration of the suction bucket. However, the large hydraulic gradient usually leads to seepage failure of the soil on beneath the top plate of the bucket foundation. As a result, the bearing capacity of the top plate of the bucket is often ignored. In this paper, the construction and construction process of composite bucket foundation are introduced, and series of model tests were carried out to reinforce the soil inside of the bucket foundation by the method of the vacuum electro-osmosis. The results show that the combined method of negative pressure and electro-osmosis can effectively improve the undrained shear strength of the soil and reduce the water content of the soil in the bucket foundation. Finally, the reinforcement effect of soil inside bucket foundation is analyzed according to reinforcement of the prototype construction.

Abstract:
Die Masterarbeit von Svenja Blumenrath mit dem Titel "Das Buch im Buch als Archiv" ist eine interdisziplinäre Zusammenführung von Literaturwissenschaft und Archivstudien. Anhand von sieben ausgewählten kinder- und jugendliterarischen Werken analysiert die Autorin die Funktion des Buches im Buch als Archiv für die erzählte Welt. Die Ergebnisse zeigen, dass das Buch im Buch nicht nur als narrative Struktur dient, sondern auch als Archiv fungiert, das Erinnerungen und Wissen bewahren, aber auch verwehren kann. Diese Erkenntnisse bieten neue Einblicke in die Verbindung von Literatur und Archivwesen.
