AbstractIntroduction. Sarcoidosis is a rare, multi-organ disease of unknown etiology, characterized by the formation of granulomas mainly in the respiratory system.Aim. The aim of the study is to present a plan of care for patients with sarcoidosis, including International Classification for Nursing Practice – ICNP® terminology.Material and methods. The study was conducted in April 2019 at the Internal Disease Unit of the 7th Polish Navy Hospital in Gdansk, and included a 54-year-old patient hospitalized for sarcoidosis. The research employed the case study method and the technique of the interview, observation, analysis of medical records and measurements. The C-HOBIC data set was used to assess the patient’s condition.Results and conclusions. The reasons for the patient’s admission to the hospital were a continued raised temperature, erythema, as well as joint pain. The assessment and analysis of a patient’s status allowed us to formulate seven nursing diagnoses characteristic for sarcoidosis, which included: arthritis pain, functional dyspnoea, fever, impaired active range of motion, fatigue, risk for medication side effect and lack of knowledge of disease and 2 diagnoses resulting from the lifestyle of the patient: tobacco abuse and overweight. The process of nursing care planning included interventions fundamental for the above-mentioned diagnoses. They concerned the monitoring of symptoms and health indices, the patient’s knowledge, as well as the role of the nurse in the therapeutic process. The ICNP dictionary includes a sufficient range of terms, which allows the creation of a satisfactory nursing care plan for a patient with sarcoidosis.

Abstract:
Extracellular nucleotides exert an important role in controlling cell physiology by activating intracellular signalling cascades. Osteoblast HOBIT cells express P2Y1 and P2Y2 G-protein-coupled receptors, and respond to extracellular ATP by increasing cytosolic calcium concentrations. Early growth response protein 1 (Egr-1) is a C2H2-zinc-finger-containing transcriptional regulator responsible for the activation of several genes involved in the control of cell proliferation and apoptosis, and is thought to have a central role in osteoblast biology. We show that ATP treatment of HOBIT cells increases Egr-1 protein levels and binding activity via a mechanism involving a Ca2+-independent protein kinase C isoform. Moreover, hypotonic stress and increased medium turbulence, by inducing ATP release, result in a similar effect on Egr-1. Increased levels of Egr-1 protein expression and activity are achieved at very early times after stimulation (5 min), possibly accounting for a rapid way for changing the osteoblast gene-expression profile. A target gene for Egr-1 that is fundamental in osteoblast physiology, COL1A2, is up-regulated by ATP stimulation of HOBIT cells in a timescale that is compatible with that of Egr-1 activation.

Dalam peningkatan produksi hortikultura, khususnya sayuran di Indonesia selama ini masih menggunakan sistem pertanian konvensional dengan masukan input luar. Semakin tinggi biaya produksi terhadap pertumbuhan tanaman budidaya, maka akan mengakibatkan semakin tinggi pula masukan input luar seperti pestisida dan pupuk yang diberikan terhadap tanaman.Banyak orang yang menanam kangkung hanya untuk sekedar dikonsumsi sendiri dan melepas hobi bercocok tanam. Namun dalam bercocok tanam sering kali yang menjadikan masalah adalah lahan yang dimiliki tidak dapat ditanami tanaman sesukanya. Jika ingin menanam tanaman, maka dibutuhkan lahan tanah yang cukup luas. Daerah perkotaan saat ini sudah jarang memiliki lahan yang luas yang dapat digunakan sebagai media bercocok tanam. Akan tetapi saat ini banyak berbagai cara yang dapat dilakukan untuk sekedar meluapkan hobi bertanam, salah satunya adalah dengan budidaya kangkung dalam polybag dengan tambahan sekam padi bekas ayam petelur. Polybag adalah menanam tanaman ke dalam kantong plastik hitam. Cara ini dapat digunakan untuk membudidayakan tanaman kangkung, selain tidak membutuhkan lahan yang luas cara ini juga sangat mudah dan hemat untuk dilakukan. Tujuannya adalah untuk meningkatkan hasil tanaman kangkung darat ( Ipomoea reptans) dilakukan untuk mengetahui pengaruh sekam padi yang berasal dari kotoran ayam terhadap pertumbuhan dan produksi kangkung darat (Ipomoea reptans), menggunakan bahan dalam berbudidayanya seperti benih kangkung ( Ipomoea reptans), dan sekam padi yang berasal dari kotoran ayam. Budidaya tanaman kangkung merupakan budidaya tanaman yang menggunakan tahapan-tahapan budidaya seperti langkah awal adalah sediakan media tanam dengan menggunakna polybag,masukan tanah dan campuran sekam padi bekas kotoran ayam ke dalam polybag, usahakan untuk memberikan pupuk organik sehingga tanaman yang akan dihasilkan akan terbebas dari bahan kimia yang berbahaya, pilihlah bibit kangkung yang unggul dengan kualitas yang baik dan tidak busuk atau cacat. Penyemaian bibit kangkung dilakukan ke dalam polybag. Setelah bibit kangkung mulai tumbuh hingga terdapat 3 sampai 5 helai daun lalu pindahkan ke dalam polybag yang sudah disiapkan. Dalam satu polybag dapat menampung sekitar 3 sampai 5 batang kangkung untuk mendapatkan nutrisi yang cukup dan juga dapat tumbuh dengan baik. Lakukan penyiraman dalam waktu 2 kali dalam sehari agar tanah tidak kering dan juga tidak terlalu lembab. Hasil analisis usaha kangkung darat memiliki nilai keuntungan yaitu berdasarkan perhitungan R/C ratio yaitu diperoleh 2,59 dengan menggunakan sekam padi bekas kotoran ayam petelur dan yang tidak menggunakan sekam padi ( control) diperoleh R/C ratio yaitu 1,71. Jadi usaha kangkung darat, setiap Rp. 1,- biaya yang dikeluarkan akan mendapatkan penerimaan sebesar Rp. 2,59 (perlakuan dengan menggunakan sekam padi bekas kotoran aya) dan Rp. 1,71 ( pada perlakuan control).

Abstract:
Perpanjangan Pembatasan Sosial Berskala Besar (PSBB) selama pandemi COVID-19 menyebabkan pembatasan kegiatan, bisnis, sekolah, serta mobilitas sosial. Hal ini menjadi faktor tidak langsung penyebab masalah dan komplikasi COVID-19 pada remaja di usianya yang masih rentan antara lain dikarenakan anxietas, stres dan imunitas tubuh. Pengabdian masyarakat ini dilakukan melalui webinar secara terbuka pada 41 remaja di Desa Cilangkap Kabupaten Lebak dan peserta lain yang tersebar di Provinsi Banten untuk memberdayakan remaja melalui pemberian informasi manajemen stres dan informasi vitamin C untuk mengurangi risiko stres dan meningkatkan imunitas tubuh. Metode pelaksanaan dilakukan dengan metode kaji tindak (action research), partisipatif (pemberian informasi, metode konsultasi, advokasi, pendampingan secara daring/Zoom Meeting), serta monitoring dan evaluasi pelaksanaan kegiatan. Hasil kegiatan diantaranya terlaksananya pengabdian masyarakat dengan pemberian informasi manajemen stres dengan sasaran remaja yang berpotensi mengalami stress, terciptanya kreasi materi video tentang manajemen stress dan vitamin C, serta HAKI. Hasil evaluasi didapatkan dengan dilakukan analisa selama proses dan kuesioner setelah webinar. Peserta menyatakan menjadi tahu informasi tentang manajemen stres, materi video sangat menarik, seru, relevan, mantap, informasi sangat jelas, sangat membantu, sangat bermanfaat, 100% peserta menyatakan baik, sangat baik, sangat memuaskan, sangat senang, dan pada narasumber (sekaligus pengabdi), dapat menambah informasi, pengetahuan, pengalaman dan relasi. Kesimpulannya, pengabmas ini sangat menarik dan bermanfaat karena topik relevan dengan yang dialami dan dirasakan peserta selama pandemi. Saran peserta, diharapkan adanya seminar lanjutan dan dilaksanakan secara luring, sehingga dapat langsung memfasilitasi pada peningkatan pemberdayaan remaja yang kreatif dan produktif atas hobi/bakat/dan minat remaja, serta munculnya inovasi baru yang menarik dan berprofit.

Abstract:
Previous work from our group (Ravasio A, Hobi N, Bertocchi C, Jesacher A, Dietl P, Haller T. Am J Physiol Cell Physiol 300: C1456–C1465, 2011.) showed that contact of alveolar epithelial type II cells with an air-liquid interface (IAL) leads to a paradoxical situation. It is a potential threat that can cause cell injury, but also a Ca2+-dependent stimulus for surfactant secretion. Both events can be explained by the impact of interfacial tensile forces on cellular structures. Here, the strength of this mechanical stimulus became also apparent in microarray studies by a rapid and significant change on the transcriptional level. Cells challenged with an IAL in two different ways showed activation/inactivation of cellular pathways involved in stress response and defense, and a detailed Pubmatrix search identified genes associated with several lung diseases and injuries. Altogether, they suggest a close relationship of interfacial stress sensation with current models in alveolar micromechanics. Further similarities between IAL and cell stretch were found with respect to the underlying signaling events. The source of Ca2+ was extracellular, and the transmembrane Ca2+ entry pathway suggests the involvement of a mechanosensitive channel. We conclude that alveolar type II cells, due to their location and morphology, are specific sensors of the IAL, but largely protected from interfacial stress by surfactant release.

Abstract:
The list of animal viruses has been frequently added of new members raising permanent concerns to virologists and veterinarians. The pathogenic potential and association with disease have been clearly demonstrated for some, but not for all of these emerging viruses. This review describes recent discoveries of animal viruses and their potential relevance for veterinary practice. Dogs were considered refractory to influenza viruses until 2004, when an influenza A virus subtype H3N8 was transmitted from horses and produced severe respiratory disease in racing greyhounds in Florida/USA. The novel virus, named canine influenza virus (CIV), is considered now a separate virus lineage and has spread among urban canine population in the USA. A new pestivirus (Flaviviridae), tentatively called HoBi-like pestivirus, was identified in 2004 in commercial fetal bovine serum from Brazil. Hobi-like viruses are genetically and antigenically related to bovine viral diarrhea virus (BVDV) and induce similar clinical manifestations. These novel viruses seem to be widespread in Brazilian herds and have also been detected in Southeast Asia and Europe. In 2011, a novel mosquito-borne orthobunyavirus, named Schmallenberg virus (SBV), was associated with fever, drop in milk production, abortion and newborn malformation in cattle and sheep in Germany. Subsequently, the virus disseminated over several European countries and currently represents a real treat for animal health. The origin of SBV is still a matter of debate but it may be a reassortant from previous known bunyaviruses Shamonda and Satuperi. Hepatitis E virus (HEV, family Hepeviridae) is a long known agent of human acute hepatitis and in 1997 was first identified in pigs. Current data indicates that swine HEV is spread worldwide, mainly associated with subclinical infection. Two of the four HEV genotypes are zoonotic and may be transmitted between swine and human by contaminated water and undercooked pork meat. The current distribution and impact of HEV infection in swine production are largely unknown. Avian gyrovirus type 2 (AGV2) is a newly described Gyrovirus, family Circoviridae, which was unexpectedly found in sera of poultry suspected to be infected with chicken anemia virus (CAV). AGV2 is closely related to CAV but displays sufficient genomic differences to be classified as a distinct species. AGV2 seems to be distributed in Brazil and also in other countries but its pathogenic role for chickens is still under investigation. Finally, the long time and intensive search for animal relatives of human hepatitis C virus (HCV) has led to the identification of novel hepaciviruses in dogs (canine hepacivirus [CHV]), horses (non-primate hepaciviruses [NPHV] or Theiler's disease associated virus [TDAV]) and rodents. For these, a clear and definitive association with disease is still lacking and only time and investigation will tell whether they are real disease agents or simple spectators.

11039 Background: To address the role of the local metastatic microenvironment in the antiestrogen (AE) resistance exhibited by disseminated breast cancer (BrCa), we assembled heterotypic in vitro three-dimensional (3D) tissue cultures comprised of estrogen receptor-positive (ER+) BrCa cells and non-malignant accessory cells from organs frequently targeted by metastatic disease, to model the composition and architecture of metastatic lesions. Methods: MCF7 cells expressing luciferase, cultured alone or with accessory cells, were exposed to dose-ranges of hydroxytamoxifen (4-OHT), fulvestrant or raloxifene. MCF7 cells were also injected s.c. in nude mice either alone or with HS-5 bone marrow stromal cells (BMSCs). Tumor spheroid viability or xenograft growth were measured by bioluminescence. Results: MCF7 response to AEs in 3D conditions (collagen type I and/or Matrigel) was marked by acinar differentiation of tumor spheroids that resembles normal breast epithelium. Co-culture of MCF7 and immortalized accessory cells from the bone (BMSCs; HOBIT and hFOB osteoblasts), brain (SVGp12 astrocytes), liver (THLE3 hepatocytes), and lung (MRC9 fibroblasts) in 3D (but not 2D) conditions, conferred resistance to 4-OHT, raloxifene, and fulvestrant at clinically relevant doses. Heterotypic xenografts (MCF7 + BMSCs) had reduced response to tamoxifen compared with monotypic xenografts (MCF7 alone). BMSCs induced in MCF7 cells downregulation of TGFβ2; upregulation of a transcriptional signature enriched for genes associated with high-grade tumors, including genes involved in interferon response, signaling through EGFR superfamily members, NFkB and other antiapoptotic pathways; and elevated c-Myc protein levels. Exogenous TGFβ2 or neutralizing antibodies against INFγ partially re-sensitized MCF7 cells to 4-OHT in co-cultures. Conclusions: Our results indicate that accessory cells representing the microenvironment of metastatic sites confer AE resistance to BrCa cells in 3D tissue cultures and xenograft models. The mechanisms of stroma-induced AE resistance may reveal new therapeutic targets for refractory BrCa patients with metastatic disease.

Abstract:
Objective: The researcher replicated 12 questions from a previous user satisfaction study for the C-HOBIC assessment tool at St. Boniface Hospital in Winnipeg Manitoba. The research questions were: (a) what is user satisfaction regarding the C-HOBIC assessment tool 20 months after implementation, (b) has user satisfaction changed since the first evaluation, and (c) do user demographics correlate with user satisfaction and the C-HOBIC assessment tool? Method: 20 months after the previous study (Canadian Nurses Association, 2015), a convenience sample of 71 participants from a pool of approximately 700 clinicians completed an online survey comprised of 12 questions taken from the previous study. Results: The data were analyzed using Shapiro-Wilk, descriptive statistics, chi-square test for independence, and Spearman’s correlation. The Likert-style survey produced discrete, ranked data that did not follow a normal distribution. Overall user satisfaction with the C-HOBIC assessment tool was rated higher in the previous group (n=59) as compared to user satisfaction in the current group (n=71). There was a significant but weak correlation with gender and C-HOBIC patient outcomes positively influencing patient care directions, and improving patient care planning. A significant but weak correlation existed between the years of a participant's clinical experience and the ease of integrating C-HOBIC into practice. Conclusions: There were more participants not satisfied with the use of the C-HOBIC assessment tool than were satisfied. Participants in this study had less user satisfaction with the C-HOBIC assessment tool and associated outcomes than participants from a similar study 20 months before. Gender and years of clinical experience are correlated with user satisfaction. The small sample size, the non-normally distributed data, and convenient sampling method do not support generalization of the results beyond the data set.
