Investigation and utilization of molecularly targeted agents has induced a number of drug adverse effects that are not typically associated with conventional chemotherapy. QT interval prolongation, a cardiac toxicity that increases the risk of fatal arrhythmia, is associated with several novel oncology therapies. Classes of molecularly targeted agents with described QT effects include histone deacetylase inhibitors, multitargeted tyrosine kinase inhibitors, vascular disruption agents, farnesyl protein transferase inhibitors, Src/Abl kinase inhibitors, and protein kinase C inhibitors. Concurrently, guidelines for monitoring the QT-prolonging effects of drugs under development have become increasingly rigorous. Although these guidelines apply to anticancer agents, they were not specifically designed for the oncology patient population. This article will review the pathophysiology of QT prolongation, methods of preclinical QT assessment, and current guidelines for QT evaluation in early phase trials. Additionally, molecularly targeted agents with QT effects will be summarized, and mechanisms of addressing this toxicity in the context of oncology drug development will be explored.

Abstract:
Background and objectives: Electrocardiograph abnormalities (i.e., QT interval prolongation) have been described in inflammatory bowel diseases (IBD). We aimed to measure the QT interval in a cohort of patients with IBD and to analyze its relationship with clinical and inflammatory activity. Materials and Methods: We performed a cross-sectional study that included 38 IBD outpatients and 38 “age- and sex-matched” healthy controls. Nine patients had active IBD, and 29 were in clinical remission. Among the latter, 10 patients had sustained (lasting >1 year) and 19 had short-term remission (≤1 year). Corrected QT (QTc) interval was measured on standard 12-lead electrocardiograph. A systematic review of the literature on studies investigating the QT interval in patients with IBD was also performed. Results: QTc interval values were similar between IBD patients and healthy controls (417.58 ± 22.05 ms vs. 409.13 ± 19.61 ms, respectively; p: 0.479). Patients with active IBD had significantly higher QTc values (435.11 ± 27.31 ms) than both controls (409.13 ± 19.61 ms) and patients in remission (412.14 ± 17.33 ms) (p: 0.031). Post hoc analysis showed that the difference in QTc values between active IBD and remission was attributable to the group of patients with sustained remission (p < 0.05). Lastly, a significant correlation between QTc interval and C-reactive protein (CRP) values was observed (Spearman test: r = 0.563; p: 0.0005). Conclusions: Our study demonstrates an association between QTc duration and both clinical and inflammatory activity in patients with IBD. The higher the CRP value, the longer is the QTc duration. For practical purposes, all patients with active IBD should undergo a standard ECG. Prescription of drugs able to modify the QT interval should be avoided in patients with active IBD. The systematic review of the literature indicated that this is the first published study demonstrating an association between the QTc duration and CRP values in patients with IBD.

AbstractAimMost long QT syndrome patients are associated with genetic mutations. We aimed to investigate the clinical and biochemical characteristics and look for genotype-based preventive implications in Chinese long QT syndrome patients.Methods and resultsWe identified two missense mutations of the KCNQ1 gene in two independent Chinese families, including a previously reported mutation R380S in the C-terminus and a novel mutation W305L in the P-loop domain of the Kv7.1 channel, respectively. The proband with R380S was an 11-year-old girl who suffered a prolonged corrected QT interval of 660 ms, recurrent syncope, and sudden cardiac death, whose father was an asymptomatic carrier. The mutation W305L was detected in a 36-year-old woman with long QT syndrome and her immediate family members including the proband’s younger sister with an unexplained syncope, her son, and her elder daughter without symptoms. Metoprolol appeared to be effective in preventing arrhythmias and syncope in long QT syndrome patients with mutation W305L. Both R380S and W305L mutations led to “loss-of-function” of the Kv7.1 channel accounting for the clinical phenotypes.ConclusionsWe first show two missense KCNQ1 mutations – R380S and W305L – in Chinese long QT syndrome patients, resulting in the loss of protein function. Mutation W305L in the P-loop domain of the Kv7.1 may derive a pronounced benefit from β-blocker therapy in symptomatic long QT syndrome patients, whereas mutation R380S located in the C-terminus may be associated with a high risk of sudden cardiac death.

Abstract:
Background: The World Health Organization (WHO) has approved the use of two new drugs, namely Bedaquiline (Bdq) and Delamanid (Dlm), for treatment of Drug Resistant Tuberculosis (DR-TB). One of the concerns raised with the use of these drugs was QT-interval prolongation. This condition could be serious and life threatening. Hence, knowing the magnitude and its management is very important. This case series identifies the incidence and discusses the management of clinically significant QT-interval prolongation amongst a cohort of patients who have been on these medicines. Materials and Methods: Patients with reports of high grade QT-Interval prolongation (i.e. Grade-3 and Grade-4) were identified from the cohort of 265 patients enrolled on bedaquiline and/or delamanid and discussion is made on the pattern, severity and management of each cases identified. Results: Only 4 (1.5%) out of all 265 patients enrolled on Bedaquiline and/or Delamanid have developed high grade QT-Interval prolongation. And all are managed without permanent discontinuation of both drugs. Conclusion: The Incidence of clinically significant QTcF-interval prolongation among DR-TB patients taking bedaquiline and /or delamanid in Lesotho is low. And almost all cases can be managed with more frequent Electrocardiogram (ECG) monitoring and management of other possible causes of QT-interval prolongation without the need to stop one or both drugs permanently.

The software development for embedded systems is getting faster and faster, which generally incurs an increase in the associated complexity. As a consequence, consumer electronics companies usually invest a lot of resources in fast and automatic verification mechanisms, in order to create robust systems and reduce product recall rates. In addition, further development-time reduction and system robustness can be achieved through cross-platform frameworks, such as Qt, which favor the reliable port of software stacks to different devices. Based on that, the present work proposes a simplified version of the Qt framework, which is integrated into a checker based on satisfiability modulo theories (SMT), name as the efficient SMT-based bounded model checker (ESBMC++), for verifying actual Qt-based applications, and presents a success rate of 89%, for the developed benchmark suite. We also evaluate our simplified version of the Qt framework using other state-of-the-art verifiers for C++ programs and an evaluation about their level of compliance. It is worth mentioning that the proposed methodology is the first one to formally verify Qt-based applications, which has the potential to devise new directions for software verification of portable code.
O desenvolvimento de software para sistemas embarcados tem crescido rapidamente, o que na maioria das vezes acarreta em um aumento da complexidade associada a esse tipo de projeto. Como consequência, as empresas de eletrônica de consumo costumam investir recursos em mecanismos de verificação rápida e automática, com o intuito de desenvolver sistemas robustos e assim reduzir as taxas de recall de produtos. Além disso, a redução no tempo de desenvolvimento e na robustez dos sistemas desenvolvidos podem ser alcançados através de frameworks multi-plataformas, tais como Qt, que oferece um conjunto de bibliotecas (gráficas) confiáveis para vários dispositivos embarcados. Desta forma, este trabalho propõe uma versão simplificada do framework Qt que integrado a um verificador baseado nas teorias do módulo da satisfatibilidade, denominado Efficient SMT-Based Bounded Model Checker (ESBMC++), verifica aplicações reais que ultilizam o Qt, apresentando uma taxa de sucesso de 89%, para os benchmarks desenvolvidos. Com a versão simplificada do framework Qt proposto, também foi feita uma avaliação ultilizando outros verificadores que se encontram no estado da arte para verificação de programas em C++ e uma avalição a cerca de seu nível de conformidade. Dessa maneira, a metodologia proposta se afirma como a primeira a verificar formalmente aplicações baseadas no framework Qt, além de possuir um potencial para desenvolver novas frentes para a verificação de código portátil.

Tolking och tillgodogörande av datastrukturer, organiserad information ochprogramkodsfiler förekommer frekvent i arbete med mjukvaruutveckling. Dennainformation är lagrad i textbaserad form och dess förståelse kräver stornoggrannhet och tidsinvestering från utvecklarens sida. I syfte att försöka förenklaprocessen beskriver detta examensarbete utvecklingen av en prototyp till ettverktygsprogram, vilket automatiserar tolkning av XML-data och källkodsfiler förprogrammeringsspråken C och C++. Programmet skapar och presenterar sedanen visuell graf av den undersökta strukturen. Algoritmen klarar av att presenteragodtyckligt stora XML-filer samt ett begränsat antal samtidigt inlästakällkodsfiler. Effekterna på tolkningens tidsåtgång och dess tillförlitlighet harutvärderats i en undersökning bland studenter inom mjukvaruutveckling.Resultatet visade på en viss mätbar ökning i antalet korrekta slutsatser somanvändaren drog efter att ha studerat datasammanhanget grafiskt jämfört meddess ursprungliga textform. Tidsåtgången mättes inte mer noggrant än subjektivthos användarna, av vilka en övervägande andel ansåg att tiden förkortades medden grafiska representationen till deras hjälp. Examensarbetet visar attanvändandet av detta eller motsvarande verktyg kan öka tillgodogörandet avdatastrukturer genom att både höja graden av tillförlitligheten hos dennainformation och samtidigt minska tidsåtgången. Däremot är den kvantifierbaravinsten av dessa resultat inte statistiskt säkerställd till en högre grad.
Interpretation and assimilation of data structures, organized information andsource code files are frequently occuring during software development. This kindof information is stored in text-based form and its understanding requires greatthoroughness and investment in time from the developer's part. This thesisdescribes the development of a utility program prototype, which automates theparsing of XML data and source code files from the programming languages Cand C++, in purpose of trying to simplify the interpretation process. The programcreates and presents a visual graph of the structure found, using an algorithmwhich can present arbitrary large XML files as well as a limited number ofconcurrent source code files. The effects on the interpretation time and itsreliablity has been evaluated in a survey among software development students.The result showed a certain increase in the number of correct conclusions fromthe participants' side after studying the visual representation compared to itsorignial text-based form. The amount of time used was not measured other thansubjectively by the users themselves, of which a predominant proportionconsidered a reduction in needed time when using the graphical representation.The thesis shows that the use of this or an equivalent utility can enhance theassimilation of data structures by increasing the rate of reliabilty whilesimultaneously decreasing the needed amount of time. Still, the quantifyable gainsof these results remains statistically largely uncertain.

Abstract:
This thesis presents a method to develop a visualization software for time-varying and geographic-based data. The machine learning team at Ericsson has collected data from their machine learning algorithms. The data set contains timestamped and geographic information. To have a better understanding of the result made by the machine learning algorithms, it is important to understand the pattern of the data. It is hard to see the pattern of the data by only looking at the raw data set, and data visualization software will help the users to have a more intuitive view of the data.  To choose a suitable GUI library, three common GUI libraries were compared. The Qt framework was chosen as the GUI library and development framework because of its wide-range support to user interface design. Animation is the main method to visualize the data set. The performance evaluation of the software shows that it handles the back-end data efficiently, renders fast in the front-end and has low memory and CPU usage. The usability testing indicates that the software is easy to use. In the end, the thesis compares its method to a previous method, developed in R. The comparison shows that even though the old method is easier to develop, it has worse performance.

Abstract:
The goal of this master's thesis is to provide an introduction to MSP430 microcontrollers and to design a simulator of these microcontrollers, focusing on easy implementation of extensions using peripherals. After a short introduction, the MSP430 microcontrollers are briefly described, including their internal peripherals and the formats used to store the binary executable code. The thesis continues with a  description of discrete event simulation using the DEVS formalism. Based on previous chapters, the new simulator (consisting of a simulation core, graphical user interface and library for MSP430 microcontroller simulation)  is designed and implemented. The implementation is tested by comparison with real microcontroller and in the end of the thesis, there is a summary and evaluation of the implemented simulator.

Detta projekt har utförts på Saab Dynamics. Projektets syfte var att utveckla en applikation, TBFO, för att rapportera information om hur missilen GLSDB ska slå till ett mål. TBFO är ämnat för att användas i närheten av målet och information skickas till planeringssystemet GLSDB MPS. Applikationen byggdes runt 3D-motorn Vricon och är anpassat för lättast möjliga användning med pekskärm. Huvuddelen av arbetet berörde utveckling av gränssnitt för pekskärm och utveckling av systemets applikationsprotokoll. Denna rapport redogör för framtagning av systemet samt de verktyg och metoder som användes. Rapporten fördjupar sig inom utveckling av applikationer anpassade för pekskärm. Slutsatsen som kan dras från resultatet av detta projekt är att idén om systemet som utvecklats är användbart för processen att planera angrepp med GLSDB MPS.
This project has been carried out at Saab Dynamics. The project's purpose was to develop an application, TBFO, for reporting information containing how the missile GLSDB would strike a target. TBFO is intended to be used in the proximity of the target and information is sent to the planning system GLSDB MPS. The application was built around the 3D engine from Vricon and is developed to fit for use of touch devices. The main part of the work concerns the development of user interface for touch input and the system’s application protocol. This report describes the processes of developing the system, including what tools and methods that have been used during development. The report also provides an in-depth look at processes used when developing applications for touch devices. The conclusion from the results of this project is that the idea of the described system is useful for the process of planning an assault with GLSDB MPS.

Diplomová práce se zabývá návrhem termální kamery pro biologické aplikace. V první části je popsáné infračervené záření, také známé jako thermální záření. Pokračuje popisem emisivity objektů, která hraje důležitou roli při stanovení teploty snímaného objektu. Dále je v první kapitole uveden přehled typů termálních detektorů a jejich základní vlastnosti. Druhá kapitola stručně seznamuje se zákládními hardwarovými komponenty vyvíjené kamery. V poslední kapitole je popsán celý proces vývoje softwaru. Ten zahrnuje vývoj IP jader, kernel modulů a C++ knihoven.

This thesis describes the design and implementation of an client-server application. This application is used to remote access to models of systems, which are saved on the server. Application also provides editation of the models and their simulation. In the thesis there is a design of Communication Protocol between the client and server too. For the implementation of the client and prototype of the server was used Qt library. Server is realized as a part of existing simulation core (SmallDEVS), which is implemented by Smalltalk.

The purpose of this thesis is to introduce a reader with existed commercial applications and control the FITkit using QDevKit. The main goal of the thesis is proposed and implemeted modul for application QDevKit which makes easy proposal and progress applications for platform FITkit.

Abstract:
Primary care clinicians commonly encounter patients with substance use disorders and can provide effective treatment for their problems. Many of the medical complications associated with drug use are due to the use of needles; these include transmission of HIV and hepatitis C, soft tissue infections, and endocarditis. Harm reduction strategies reduce the harms associated with drug use without targeting use itself; these include syringe distribution, safe consumption facilities, naloxone distribution, and pre-exposure HIV prophylaxis. Patients on opioid agonist treatment may develop a number of problems, including hypogonadism, constipation, and psychomotor impairment; those on methadone may develop prolonged QT syndrome. Other issues include drug interactions, treatment of acute pain, and perioperative care. Treating pain in patients with substance use disorder can be complicated; for many, especially those with opioid use disorder, treatment with buprenorphine or enrollment in methadone maintenance is the best option.

Abstract:
The design of RoboSim, a virtual environment for modular robots which controls simulated robots with code written for the hardware robots without modification, is described in detail in this paper along with its applications in educational environments. RoboSim integrates into the Ch programming environment, a C/C++ interpreter, that provides the ability to remotely control robots through interpreted C/C++ code allowing users to alternate between hardware and virtual robots without modifying the code. Open source software projects Open Dynamics Engine, OpenSceneGraph, and Qt are employed to produce the virtual environment and user interface which provide the capability of running on all major software platforms. The design of the software includes multiple library modules each specific to a particular task; therefore the simulation library and Graphical User Interface (GUI) can link against only the necessary libraries. The GUI links against the graphical library and XML library to give an interactive view of the RoboSim Scene as users are adding robots and obstacles into both the GUI and simulation. Execution of Ch code generates a new RoboSim Scene window which has the entire simulation that utilizes the simulation, graphical, xml, and callback libraries, in addition to the identical Scene from the GUI. It generates its own window for the user to view and interact with the progress of the simulation.

A software tool named “T4T” (Tools for Turbomachinery) has been developed for the parametric design of turbomachinery and wind turbine blades. The complete design procedure is object-oriented and parametric, providing the ability to the user to define various types of blades. It has been developed in QT C++, utilizing OpenCascade graphical and computational libraries. The software allows the user to design the outer surface either by specifying some physical parameters for each blade section, or by directly interpolating a surface through a cloud of points. The new/enhanced version of “T4T” software tool, introducing the definition of internal blade structure for wind turbines rotors, is fully parametric and customizable, allowing the user for defining the internal blade structure, including shear webs. The computational procedure finally produces compound solids, which can be further imported to mesh generation and analysis software through standard geometry exchange protocols, for cooperation with CFD and CSD solvers.

This paper describes an open source computer application developed at the University of Missouri - Rolla (UMR) for archiving product design information. The Repository Entry application is designed to work with the UMR design repository to record and upload product information. Written in C++, the application and user interface is compiled in Qt allowing for native Macintosh and Windows executables. The Repository Entry application can record all of the design information types allowed by the repository including: artifact-, function-, failure-, physical-, performance-, sensory- and media-related information types. By using XML, files can be seamlessly transferred between the Windows and Macintosh entry application versions as well as the online repository. Through an example product the procedure of using the entry application to record and upload design information is demonstrated. The result of this research is a fully functional, easy to use and multi-platform application to aid in the design information archival and reuse process.

Abstract:
Recent trends for linepipe projects reflect a sustained increasing complexity: Sour Service (SS), High Temperature & High Pressure (HT/HP) field conditions, deep and ultra-deep water oil and gas transportation, Artic and Artic “alike” areas. This reflects into stringent requirements for manufacturing and testing of the concerned products, which are not only the straight pipes but also a number of accessories, among which bends are the ones presenting the most complex combination of critical issues. As long as a whole chain reliability standpoint is assumed as the main concern, the design, production and supply of the bends have become a key stage within a linepipe project. Bends for linepipe projects are produced in general by hot induction bending (HIB). Two different fabrication routes can be clearly identified: HIB followed by Stress Relieving (SR) and HIB followed by off-line full quenching and tempering (Q&T). The first method is known as “Traditional route (TR)”, while the second one as “Quenching-Tank (QT)”. A large investigation program was carried out involving the most recognised benders in Europe. The matrix of industrial trials comprised a dimensional size range from 168 to 406 mm OD, 8 to 34 mm WT; X60 to X70 steel grades, different bending and post-bending heat treatments conditions and mother pipe chemistries. For each analysed item, the final bend, the corresponding mother pipe and samples taken in as-bent / as-quenched (TR / QT) conditions were fully characterised in terms of mechanical properties, hardness profiles and microstructure features. As a result, a much better performance was found for the bends produced by the off-line full Q&T method, principally due to the better quenching efficiency with respect to the in-line system. Production of bends through out the traditional method can be seen as a reliable option for bends which are not going to face hard conditions in the field, and therefore the corresponding specification requirements are no stringent as well (i.e. X52 grade or lower, Charpy tests required at 0°C or higher temperature, no corrosion tests required, etc.). If either stringent conditions are required or X60-X80 steel grades are involved, the off-line quenching tank route becomes the reliable option.

Abstract:
As shown previously, cleavage of surface glycoproteins on the platelet membrane shortens platelet survival. Since immunoglobulins have been implicated in the condition of idiopathic thrombocytopenic purpura in which patients have an elevated level of platelet-associated immunoglobulin G (I g G) and shortened platelet survival, we measured the binding of IgG to rabbit platelets that had been treated with neuraminidase or plasmin, to determine whether increased IgG binding to these platelets might be the signal that leads to their accelerated clearance. Rabbit platelets were washed and resuspended in calcium-free Tyrode-albumin solution (1.0 x 106/ μL), pH 6.5, and then incubated with either neuraminidase (0.1 U/mL at 22°C) or plasmin (activity: 1.6 ymol BAEE/min/mL platelet suspension at 37¼C) for 30 min. The platelets were then centrifuged and resuspended in heat-inactivated rabbit serum for 30 min to allow IgG to bind to the surface. After additional washing steps to remove non-specifically bound IgG, the amount Qt IgG that associated with the platelets was measured using an 125I-labeled goat anti-rabbit IgG binding assay. Binding data were analyzed by plotting the calculated fg of IgG bound/platelet versus the amount of 125-1 goat anti-rabbit IgG added. The binding curves for both neuraminidase and plasmin indicated significantly increased amounts of specifically bound IgG compared to control platelets. At a concentration of 30 μg/mL of labeled goat antirabbit IgG added, the amount of label associated with untreated rabbit platelets had plateaued at 0.36± 0.11 fg/platelet whereas the amount associated with neuraminidase-treated platelets was 5 times greater and with pi asmin-treated platelets, 1.5 times greater, and neither had plateaued. The results obtained with plasmin are of particular interest since this proteolytic enzyme shortens platelet survival and is present in vivo during thromboembolic events. These results indicate that IgG binds specifically to the surface of rabbit platelets following enzymatic alterations of the membrane glycoproteins and therefore may represent at least one mechanism that determines which platelets are cleared by the reticuloendothelial system.

Abstract:
It is very well-known that fracture toughness depends on loading rate. Higher strain rates can shift the ductile to brittle transition curve to higher temperatures, resulting in a more brittle structure at the same temperature. However, there is little effort to relate the testing rate to the loading rate within the offshore and maritime industry. For example, BS 7448-1 requires that the stress intensity factor loading rate be 0.5 MPa√m/s to 3.0 MPa√m/s. The loading rates of BS 7448-1 are very far away from the vibrational modes of the specimen, so these limitations are not necessary in order to assure a quasi-static test. In comparison, SSC 275 indicates that normal ship loading rates can be of the order of 220–440MPa√m/s. The results of SSC 275 are consistent with results obtained from a Dutch offshore equipment supplier, who indicates a time to maximum loading of 0.25–1.3 seconds. In general, a conservative loading scenario for the maritime and offshore industry is on the order of 200 times faster than the loading rate that is recommended by BS 7448-1. Testing at the standard rate has the consequence of artificially lowering the ductile to brittle transition temperature by 8–35°C in comparison to a real loading scenario, thus possibly giving a false impression of safety. This means that a CTOD measured as 0.2 mm for static testing conditions could be 0.08–0.15 mm for actual loading. The analysis is shown to be consistent with CTOD test data on a Quenched and Tempered (QT) and a Thermo-Mechanically Controlled Processed (TMCP) S690 grade steel.
