Full textSignifikant Svenska AB supplies an information system called Assert, developed to facilitate the aftermarket sales in the manufacturing and subcontractor domains. The information system offers companies and organizations the possibility to gather their product information in a joint database in order to increase the information availability and distribution.
The management of the documents is an important part of the customer need and can be improved in order to also support document maintenance directly in Assert. At the moment, users only have the possibility to add and view documents in the database. By also providing users the possibility to create documents, update document contents, effectively reuse document sections and ease the translation of documents within Assert, the document management will be better facilitated.
The purpose of this thesis is to develop a prototype which shows the concept and benefits when providing the possibility to share document contents between several documents. The prototype is developed in C#/WPF and provides a word processor with features to reuse document contents and translation management.

Full textUpon completion of a test where telemetry(TM) data was collected, the resulting TM file will usually contain millions of data points. Traditionally MatLab™, Mathlab™, or some third party software is used to plot the data. These methods may not always be desirable due to the expense of licensing, restrictions on the ability to create custom graphs and the inability to quickly plot large amounts of data. These problems were solved by using Windows Presentation Foundation (WPF) graphics capabilities in conjunction with C# to develop a unique set of algorithms to display custom graphs of unlimited size with quick response.

Full textTelemetry data from flight tests are normally plotted using MatLab™ or other third party software. For most of the trajectory and flight parameters, a static 2D or 3D line graph does not provide the proper data visualization that can be accomplished with 3D software. Commercial 3D software can be expensive and difficult to customize, and writing custom software using Direct3D or OpenGL can be complex and time consuming. These problems were overcome using C# and Windows Presentation Foundation (WPF) to quickly and easily create a 3D Telemetry Theater and Trajectory Visualization Tool to dynamically display actual and simulated flight tests.

This product is only available on an older mobile platform that has since been replaced. Saab is now in need of a rewrite of this product on a more modern platform. This dissertation describes the project Mobile Translator, which also became the products working title. The project's main aim was a new version of the previous product Paratus Pocket Translator for Windows with maintained functionality. The project has resulted in a Windows application based on modern technologies that satisfies Saab's needs and requests.

Full textThe mission was to create a prototype of a menu system for the company
System Andersson AB. The instructions were that it has to be as interesting and
modern now as in 20 years. We had to look at technology from the cutting edge
of development to make our prototype survive in the future. Windows
Presentation Foundation (WPF) was the technique we focused on. There are
only a few applications that take advantage of WPF today, but it is predicted
that WPF will be the next generation method for developing applications to the
Windows platform.
To make our menu visually appealing, easy to use and at the same time
functional, we followed the clear path and guidelines that are available in
interaction design. Those guidelines tell us how the design process should be
executed. Through usage of the six basic ideas of interaction design and taken
the principles of richer user experience further, we were able to build a menu
system that will be as modern in 20 years as it is now. We took the thoughts of
physical design into account to make the menu understandable and easy to use.
Examensarbetets uppdrag var att skapa en prototyp av ett menysystem åt
System Andersson AB, som enligt instruktioner ska vara intressant och
modernt, även om 20 år. För att lyckas med detta tittade vi på "cutting edge" av
vad som används idag för att inte systemet skulle riskera att bli föråldrat i
framtiden. Tekniken som vi undersökte och använde oss av var Windows
Presentation Foundation (WPF). I dagsläget är det endast ett fåtal applikationer
som använder WPF men utvecklingsmetoden spås vara morgondagens sätt att
programmera windowsapplikationer.
För att vår meny skulle bli estetiskt tilltalande, enkel att använda och samtidigt
funktionell så finns det inom interaktionsdesign klara riktlinjer om hur
designutvecklingen bör gå till väga. Genom att nyttja de sex grundtankarna: att
ett system ska vara effektivt, ha hög verkningsgrad, säkert att använda, vara
användbart, enkelt att förstå samt okomplicerat; i designen och att bygga vidare
med principerna så anser vi att vi har kunnat åstadkomma ett menysystem som
kommer att se lika intressant och modern ut om 20 år som det gör nu. För att
menyn skulle uppfylla de sex grundtankarna, bland annat att vara klart
förståelig och enkel att använda så använde vi oss av de tankar och teorier som
framgår i fysisk design.

Full textSistemas de Bilhética tem como foco as empresas de transportes públicos. As consolas de bordo são o componente principal destes sistemas, permitem a emissão de bilhetes, validação de passes, ou seja, o controlo de receita gerada a cada serviço prestado pelos motoristas. Estes sistemas são sistemas embebidos com algumas restrições de desenvolvimento, e no que diz respeito a emissão de faturas estes equipamentos não têm a possibilidade de integrar na própria maquina a emissão de faturas. Este projeto está focado em descobrir soluções para a emissão de faturas, incidindo sobre o processo de certificação de software de faturação, bem como o desenvolvimento de um posto de venda de passes que permitira criar e carregar passes emitindo a fatura na hora do pagamento. Sendo este um mundo de base de dados transacional existe o problema na parte de relatórios em que este projeto pretende dar resposta e ter uma solução mais rápida e eficiente, para ajudar na tomada de decisões operacionais.
Ticketing Systems focuses on public transport companies. The on-board consoles are the main component of these systems, they allow the issuing of tickets, validation of passes, the control of revenue generated for each service provided by the drivers. These systems are embedded systems with some development constraints, and with regard to the issuance of invoices these equipment’s do not have the possibility of integrating in the machine the issuance of invoices. This project is focused on finding invoice solutions, focusing on the billing software certification process, as well as developing a ticket sales point that would allow you to create and top-up pass by issuing the invoice at the time of payment. Being among a world of transactional database there is the problem in the part of reports in which this project intends to respond and have a solution, a fast and efficient decision support system.

Full textThis report considers a system where a database is used as the back-end storage for logging. A suitable method for extracting information from the logs was missing and the database design needed an improvement for log searching. An application for extracting and filtering the logs was created. An evaluation of how the database could be improved was also performed. Both parts were done in one project since they were heavily connected. The application would use the database. Since I couldn’t make arbitrary changes to the database only relatively limited changes were made in practice. Larger changes were evaluated theoretically. The application was made against the existing database, with one exception: a view was added. The report handles indexes and other methods for speeding up database searches. A method for fetching data inside an interval in a database was developed and is described in the report. The method searches for all data where the value of a column is inside an interval and the database is ordered, or almost ordered, on that column. The method gives inexact answers if the database is almost ordered on that column. It is faster than a corresponding exact search.

The platform Frama-C is a tool dedicated to analysis of source code of software written in C, with the possible types of analysis provided by plugins attached to the platform. One of its plugins is WP, used for deductive veri cation of C code with ACSL, a formal speci cation language. This dissertation describes the extension of this plugin, named WPTrans. This extension allows generated proof obligations from WP to be manipulated through inference rules and sent, at any stage of the proof, to automatic (mainly SMT solvers) and interactive theorem provers. Some proof obligations may be proved automatically, while others can be too complex to be solved by automatic theorem provers, requiring the users of Frama-C and WP to handle them manually. This approach usually requires a signi cant experience in proof strategies. Some interactive theorem provers provide communication with automatic provers. However, this connection can be complex and incomplete, leaving the user with the manual proof option only. The strength of WPTrans is to combine the features of automatic and interactive theorem provers in a precise and complete way, with a simple manipulation language. Thus, the user can simplify the proof obligations enough in order for its proof to be concluded with an SMT solver, letting the proof be partially manual and partially automatic. Nevertheless, the plugin is directly linked do WP, facilitating the installation of the extension in Frama-C. This tool is also a gateway to other possible features, which we discuss herein.

Full textThe messenger molecule c-di-GMP stimulates the formation biofilms in most bacteria species. The enzymatic activities of the diguanylate cyclases/DGC and the phosphodiesterases/PDE adjust the c-di-GMP content in response to diverse stress and suboptimal environmental conditions. Above all, Gram-negative bacteria have multiple GGDEF/EAL domain proteins. Escherichia coli K-12 possesses 29 of such proteins: 12 DGCs, 13 PDEs and 4 so called degenerate proteins without any enzymatical function. Mainly, this complex c-di-GMP control system regulates the production of the extracellular biofilm matrix, which in E. coli takes place during the switch into the stationary growth phase. The main compounds of the matrix are amyloid curli-fibers and exoplysaccaride cellulose. The goal of this work was to investigate, whether the 29 DGC/PDE from E. coli develop a specific interactome containing additional DGC/PDE pairs. In a comprehensive two-hybrid study, it could be demonstrated that there is indeed a specific interactome in the biofilm formation cascade. However, this interactome does not contain additional DGC/PDE pairs. Mainly, the core group of enzymes, which have multiple interactions among each other and with other DGC/PDE, controls biofilm formation. Under certain growth conditions the mode of action of the core enzymes might be adjusted through the interaction with other proteins. Presumably, the dynamics of the interaction network allows managing the resources in the different biofilm zones efficiently, which could conribute to the complex organisation of the matrix architecture. Therefore, the rapid adaptation of bacteria and the formed biofilm structures could be better organized. Altogether, this work provides a new model for the local regulation of the biofilm formation by the secondary messenger c-di-GMP guided from a systemical perspective. Hereby, the basis for further investigations on regulation mechanisms of individual DGC/PDE was set.
Переход от подвижного и планктонообразного образа жизни к формированию биоплёнок является важной и интересной особенностью различных микроорганизмов. Кишечная палочка (Escherichia coli) представляет собой удобный модельный организм для изучения подобных трансформаций. У этой грамотрицательной бактерии образование биоплёнки обусловлено внутриклеточной аккумуляцией циклического дигуанилата (цикло-диГМФ). Известно, что активность ферментов, синтезирующих (дигуанилатциклазы/ДГЦ) и разлагающих (диэстеразы/ДЭ) эти сигнальные молекулы, меняется в ответ на стрессовые и субоптимальные раздражители. Кишечная палочка имеет 12 ДГЦ, 13 ДЭ и четыре дегенерированных протеина. Цель данной работы – изучить специфический, важный при формировании биоплёнки интерактом и выяснить, способны ли другие ДГЦ/ДЭ образовывать дополнительные ДГЦ/ДЕ модули и вносить свой вклад в формирование биоплёнки. В работе были изучены молекулярные взаимодействия, ответственные за формирование биоплёнок у кишечной палочки. Так, было доказано отсутствие в интерактоме дополнительных локальных ДГЦ/ДЕ модулей, участвующих при каскадных процессах регуляции роста биоплёнки. Установлено, что процесс формирования биоплёнки в большей степени контролируется основной группой ферментов, которые имеют множественные взаимодействия между собой и с другими ДГЦ/ДЭ. Вероятнее всего, такие взаимодействия способны модулировать работу основных ферментов при определенных условиях культивирования. Динамика подобной сети взаимодействий позволяет микроорганизмам целесообразно использовать свои клеточные ресурсы при образовании биоплёнок и вносит свой вклад в её сложную архитектуру, повышая тем самым приспособляемость бактерий и созданных ими сложных биоплёночных структур к внешним условиям. В целом, в данной работе предложена новая модель локальной регуляции образования биоплёнки с помощью сигнальной молекулы цикло-диГМФ и заложен фундамент для дальнейших исследований механизмов действия отдельных ДГЦ/ДЭ.

Full textBiofilm formation of E. coli K12 requires the expression of the biofilm master regulator CsgD. This process depends on the signaling molecule c-di-GMP, which is synthesized by 12 diguanylate cyclases (DGCs with GGDEF domains) and degraded by 13 phosphodiesterases (PDEs with EAL domains). DgcE is the most complex DGC with a MASE1-containing transmembrane region (TM), three PAS, a GGDEF and a degenerate EAL domain, and it is essential for biofilm formation. This work shows that the regulation of the DgcE activity is highly complex. It was possible to assign activating and inhibitory roles to single domains of DgcE with regard to the expression of biofilm matrix components. C-di-GMP produced by DgcE is necessary for biofilm matrix production. The dimerization of DgcE is mainly mediated by the PAS region, whereas the EAL domain counteracts an active form of DgcE. DgcE-mediated matrix synthesis requires the activating signal input of the GTPase YjdA and its partner protein YjcZ. DgcE, YjdA and YjcZ form a protein complex in which YjdA directly interacts with YjcZ and the TM of DgcE. The interaction between DgcE and YjdA as well as the matrix expression depend on the GTPase activity of YjdA. Thus, it is proposed that GTP serves as an intracellular signal regulating the activation of DgcE by YjdA/YjcZ. The MASE1-containing TM proved to be a central hub for signal integration. It is both required for DgcE activity and for a massive degradation of DgcE. Furthermore, newly discovered curli-regulating genes (rbsK, rbsR, ydcI, yieP, puuR) have been analyzed. None of those gene products act on CsgD expression via the PdeR/DgcM/MlrA module. Flagellar entangling within the bottom layer of E. coli macrocolonies determines morphogenesis of macrocolonies. The data presented here suggest that the master PDE PdeH is somehow down-regulated by flagellar entangling, which probably results in a higher cellular c-di-GMP concentration, thereby promoting biofilm formation.

Full textCutaneous T cell lymphomas (CTCL) form a heterogeneous group of non-Hodgkin lymphomas. Its most frequent forms are Mycosis fungoides, Sézary syndrome and CD30-positive cutaneous anaplastic large-cell lymphoma. Death ligands critically contribute to lymphocyte homeostasis by induction of apoptosis and may further represent safeguard mechanisms to prevent lymphoma development and tumor growth. First, we analyzed induction of apoptosis by death ligands in CTCL cell lines and compared it to that in systemic T-cell lymphoma cells. This revealed for CTCL cells a pronounced resistance to death ligands. In particular, there was no caspase activation in non-responsive CTCL cells, indicating an early blockade of the apoptosis signal. Furthermore a loss of CD95, TNF-R1, caspase-10, as well as of Bid was found in several cell lines. Changes at the receptor expression level were largely ruled out, whereas, consistent and strong expression of c-FLIP was correlated with resistance. In addition, we investigated the CD30/CD95 crosstalk in CD95-sensitive cALCL cell lines. Experiments showed that CD30 ligation leads to NF-κB-mediated c-FLIP upregulation in cALCL cells, which in turn conferred resistance to CD95-induced apoptosis. Parallels with regard to expression of apoptosis regulators were seen in peripheral blood mononuclear cells and biopsies of CTCL patients. Finally, we evaluated non-steroidal anti-inflammatory drugs (NSAIDs) for their capacity to downregulate c-FLIP expression and apoptosis induction in CTCL cells as well as in tumor T cells from CTCL patients. NSAID treatment resulted in an activation of both apoptosis signaling pathways and, furthermore, Diclofenac pre-treatment sensitized for TRAIL-induced apoptosis. In conclusion, this study elucidates defects in apoptosis regulation, proved the significance of c-FLIP for the survival of CTCL cells and provides a rationale for the use of NSAIDs as a potentially new therapeutic option for CTCL patients.

Full textThe second messenger c-di-GMP promotes biofilm formation in many bacterial species. Phosphodiesterases (PDEs) and diguanylatcyclases (DGCs) - often controlled by various N-terminal sensor domains - degrade and synthesize c-di-GMP. The aim of this study was to characterize the mode of activation and physiological function of a new class of sensor domains, the CSS domain, which is coupled to an EAL domain with PDE activity and therefore potentially controlling c-di-GMP degradation. The CSS sensor domain contains two highly conserved cysteins in the periplasmic loop, of which one is arranged in the characteristic CSS motif. Integration into the inner membrane is ensured by two flanking transmembrane domains (TM1 and TM2), with TM2 providing a connection to the C-terminal EAL domain. Analysis of PdeC as one of 5 CSS-PDEs in Escherichia coli K-12 revealed a close linkage to the disulfide bond (DSB) system as well as important periplasmic proteases. Thus, formation of a DSB between the two conserved cysteins is promoted by the oxidizing DsbA/DsbB-system and reduces enzymatic activitiy of the EAL domain. In contrast, the free thiol form increases PDE activity and dimerizes via the TM2 domain. Moreover, reduction of the CSS domain results in degradation by the periplasmic HtrA proteases DegP and DegQ. These redundantly process PdeC to a shorter fragment containing the TM2 and EAL domain only, which shows dimerization as well and has high PDE activity. Degradation in the periplasm mediated by DegP and DegQ is very efficient. In contrast, further proteolysis in the cytoplasm by yet unidentified proteases is rather slow, allowing the accumulation of the highly active TM2+EAL form. Finally, the interplay of redox dependent activity control and proteolysis of the c-di-GMP specific PDE PdeC regulates production of curli and cellulose as major matrix components of the bacterial biofilm.

Full textNucleotide second messengers are used by all forms of life to transduce extra and intracellular signals and translate them into a physiological cell response. The cyclic dinucleotide c-di-AMP is a signaling molecule involved in diverse functions in bacterial physiology and is essential for many bacteria under certain growth conditions. However, this second messenger has to be tightly regulated since increased levels of c-di-AMP can be toxic. In many bacteria diadenylate cyclases with a conserved DAC domain synthesize c-di-AMP and phosphodiesterases (PDEs) with a DHH/DHHA1 or HD domain degrade it. Streptomyces spp. are soil-inhabiting gram-positive Actinobacteria characterized by a sophisticated developmental life cycle during which they produce various secondary metabolites, including antibiotics. The regulation and role of c-di-AMP is not well understood in Streptomyces biology. For c-di-AMP synthesis, streptomycetes utilize the DAC DisA but do not encode any canonical PDE and most of the known effector proteins for c-di-AMP signal transduction are absent. In this work, I demonstrated that DisA is the primary c-di-AMP synthetase in Streptomyces venezuelae and characterized AtaC as the founding member of a novel class of c-di-AMP-specific PDEs. In S. venezuelae, deletion of ataC interferes with development and growth, whereas disA inactivation affects bacterial survival under high ion osmotic stress conditions. Further, I identified CpeA and CpeD as the first c-di-AMP-binding proteins in Streptomyces. The respective genes are encoded in operons with the predicted cation/proton antiporters cpeB and cpeE, respectively, and the gene products interact in vivo in a c-di-AMP-dependent manner. Although neither cpe deletion nor overexpression of cpeABC produced a phenotype in S. venezuelae, expression of cpeABC in Escherichia coli improved growth in liquid media supplemented with potassium, suggesting that Cpe transporters are involved in potassium homeostasis.

Full textThe five CSS domain phosphodiesterases from Escherichia coli K12 (E. coli) belong to the widespread group of c-di-GMP PDEs. One representative, PdeC, has already been characterized by Herbst et al. (2018). DsbA/DsbB promoted disulfide bond (DSB) formation in the CSS domain of PdeC inhibits the PDE activity of the protein. On the contrary, the free thiol form is more enzymatically active, depending on the TM2 as the dimerization domain. This form is processed by the periplasmic proteases DegP and DegQ. An irreversibly activated TM2 + EAL fragment is generated that is slowly removed by further proteolysis. The reduction of the CSS domain of PdeC to the free thiol form stimulates the PDE activity of the EAL domain in vitro. Together with the data from Herbst et al. (2018) the CSS domain is characterized as a new sensory domain whose activity is regulated by a DSB / thiol switch mechanism. All five E. coli K12 CSS domain PDEs share a similar domain architecture, but redox biochemistry, proteolysis, and PDE activity differ within the protein family. On the basis of the PDE activity of non-DSB variants, PdeB, PdeC and PdeG were characterized as activatable (reduction increases PDE activity) and PdeD and PdeN as non-activatable (reduction inactivated PDE activity). Another representative of the CSS domain PDEs, PdeN, does not seem to be regulated and activated by forming a DSB in the CSS domain. After protein biosynthesis the protein concentration is rather regulated via the N-terminus, with acidic growth conditions significantly inducing the protein and increasing its activity. If the protein is successfully inserted in the membrane, it can develop its PDE activity due to the structural DSB and influence the biofilm matrix production significantly.

Full textStreptomycetes show a complex life cycle. The transition between the different developmental stages is regulated by the secondary messenger bis- (3´- 5´) -cyclic dimeric guanosine monophosphate (c-di-GMP) and the c-di-GMP effector proteins BldD and RsiG. c-di-GMP is synthesized by diguanylate cyclases (DGCs) with GGDEF domains, and its degradation is catalyzed by phosphodiesterases (PDE) with EAL or HD-GYP domains. In S. venezuelae, the Streptomyces strain which was used as a model organism in this work, there are ten potentially c-di-GMP metabolizing enzymes, of which two GGDEF-EAL tandem proteins, RmdA and RmdB, are the focus of this work. The deletion of the genes coding for RmdA and RmdB leads to a pronounced developmental delay in S. venezuelae. With the help of chromosomal mutations, the EAL motif was identified as essential for the in vivo function of RmdA and RmdB. Furthermore, both proteins were characterized in vitro as active PDEs and RmdA as a bifunctional enzyme, which also showed DGC activity. RmdB was identified as the master PDE in S. venezuelae by means of nucleotide extraction and is responsible for the hydrolysis of c-di-GMP over the course of development investigated. Also RmdA has an influence on the global cellular c-di-GMP concentration during the transition from the vegetative to the reproductive growth phase. A transcriptome analysis, qRT-PCR experiments and related follow-up experiments showed that the deletion of rmdA and rmdB leads to a differential expression of genes which code for important development-specific factors and regulators. This is presumably due to the increased c-di-GMP concentrations in the deletion mutants, with the c-di-GMP effector proteins BldD and RsiG delaying the transition to the next growth phase. Furthermore, it could be shown that RmdB can interact with the sigma factor of sporulation, WhiG.

Full textIn this work the affinity maturation of the murine anti c-myc-peptide antibody 9E10 was analysed. Therefore Fab fragments with reversed mutations directed towards germline genes were genetically produced and characterised for their binding to the human c-myc peptide. The epitope recognized by 9E10 consists of the amino acid sequence EQKLISEEDLLRKR of which the key positions LISEXXL are very selectively recognized. The maturation of 9E10 leads to a 3300-fold higher affinity, which is achieved by a faster association as well as by a slower dissociation of the complex. For the gain in affinity formation of additional contacts to the peptide is less important than conformational and/or flexibility changes of the CDRs which are involved in binding. The exceptionally long CDR-H3 contributes essentially to the affinity maturation. The variable light domain serves thereby with its long CDR-L1 and -L3 as a binding platform for the flexible CDR-H3. Changes in specificity of 9E10 are primarily due to maturation of the variable heavy domain. Selective recognition of the key positions in the peptide is already highly pronounced in the initial stage of affinity maturation of 9E10.

Full textLipopolysaccharide (LPS) binding protein (LBP) is an acute phase protein with the ability to bind and transfer LPS of Gram-negative bacteria. This soluble pattern recognition molecule represents an important defense principle of the host. Regulation of the hepatic acute phase response and its termination are important mechanisms for limiting systemic inflammatory activity of the host. Here were analyze the cooperation of Interleukin (IL)-1, IL-6, and Dexamethasone (Dex) at LBP expression in the hepatoma cell lines HuH-7 and Hep G2. The major inducer of LBP expression is IL-6. Within the LBP promoter numerously highly consensus binding sites such as AP-1, C/EBP-beta? and STAT3 are present, that confer transcriptional activity as shown by truncation and mutation experiments. Additionally, activate NF-kappaB sites activated by IL-1 were detected at the LBP promoter. By mutation experiments of the promoter furthermore were found differentially active glucocorticoid response elements (GREs). The promoter contains GREs enhancing the activity as well as inhibitory ones. The enhancing effect towards LBP expression by Dex was mediated by IL-6. Dex stimulated the expression of the IL-6 receptor and therefore upregulated the IL-6 pathway. Transforming Growth Factor (TGF)-beta is able to inhibit LBP expression in stimulated cells. An AP-1 binding site was identified mediating inhibitory TGF-beta effects towards LBP promoter activity. Furthermore it was shown that a growth factor independence (Gfi)-1 binding site localized near the AP-1 site is essential for mediating the TGF-beta inhibitory effect. The relevancy of the Gfi-1 site fore mediating TGF-beta effects indicates a novel mechanism for understanding inhibitory TGF-beta effects at the transcriptional level. In summary the complex regulation of LBP were elucidate which may help to eventually develop novel intervention strategies for acute phase, sepsis, and septic shock.

Full textThe present research considered the effects of agricultural management practice on the nematode community structure and biomass in three different depths and two successive vegetation periods at an arable field site. The resource quality was manipulated by crop plant and organic amendment, to investigate the major soil carbon pathways based on roots, bacteria and fungi. The nematode community analysis pointed to a highly enriched and disturbed ecosystem with low biodiversity. Along the depth gradient the nematodes formed distinct metacommunities, reflecting resource availability and abiotic environmental factors. Wheat supported significantly greater nematode biomass than maize. The litter amendment induced bottom-up effects, with greater biomass allocation at lower than at higher trophic levels. The biomass of nematode families as well as faunal indices (Channel Index, Enrichment Index) revealed a predominance of the bacterial carbon channel in the arable soil. A 13C pulse-labelling experiment investigating the flux of root- derived C into the soil food web revealed high turnover rates in the fungal carbon pathway. This was evident for soil fungi as well as for fungal-feeding nematodes and contradicts general assumptions of a more active bacterial pathway in arable soils. A laboratory experiment with 14C isotope was used to compile a complete budget for the root-derived carbon in the nematode food web. Here plant-feeding nematodes, which feed on living plant roots, thereby opening the root C cycling into the food web, showed highest amounts of 14C allocation within a few days.
