Spelutveckling är något som ständigt förändras, möjligheterna vi har idag är väldigt olika de möjligheter vi hade för tio år sedan och de möjligheter som vi kommer att ha om tio år. I denna rapport vill vi utforska några av dessa nya möjligheter och vi vill därför ta reda på hur väl lämpat C# och WPF är för implementationen av ett timingbaserat fightingspel till rörelsesensorn Microsoft Kinect genom att genomföra sagda implementation. Resultatet av detta examensarbete är en tidig version av spelet och en analys av de svårigheter som har uppstått och de begräsningar som vi har upptäckt med C#, WPF och Kinect.

System Andersson AB is a company located in Jönköping that works with developing computer systems for the mechanical engineering industry. As of today, when this report is written, the company is working on the development of a new Manufacturing Planning and Control (MPC) system. This new system aims to create not only a user friendly interface but also a modern, stylish interface. To achieve this, the company has decided to work with a, for them, new application framework; Windows Presentation Foundation (WPF).System Andersson is well known for developing systems for different kinds of touch screens, this project is no exception. For this new user interface the company wishes to implement different types of Drag&Drop functionality. They have not had enough time themselves to study and finish these parts and therefore they have chosen to hand out the task as a thesis project.The first step in the process was to get to know what WPF was and how it works, and especially how it handles Drag&Drop. The goal was to put together some sort of class library and a presentation that together were to give the company a quick guide on how they could implement the functionality into their system.As the framework was new for the company there were also a wish for alternative solutions that demonstrated how the WPF framework could be used to solve the task in as many ways as possible.This report contains a summary of the projects procedure and result. In the report the results are presented and motivated as well as demonstrated in the appendix.

Abstract:

Signifikant Svenska AB supplies an information system called Assert, developed to facilitate the aftermarket sales in the manufacturing and subcontractor domains. The information system offers companies and organizations the possibility to gather their product information in a joint database in order to increase the information availability and distribution.

The management of the documents is an important part of the customer need and can be improved in order to also support document maintenance directly in Assert. At the moment, users only have the possibility to add and view documents in the database. By also providing users the possibility to create documents, update document contents, effectively reuse document sections and ease the translation of documents within Assert, the document management will be better facilitated.

The purpose of this thesis is to develop a prototype which shows the concept and benefits when providing the possibility to share document contents between several documents. The prototype is developed in C#/WPF and provides a word processor with features to reuse document contents and translation management.

Upon completion of a test where telemetry(TM) data was collected, the resulting TM file will usually contain millions of data points. Traditionally MatLab™, Mathlab™, or some third party software is used to plot the data. These methods may not always be desirable due to the expense of licensing, restrictions on the ability to create custom graphs and the inability to quickly plot large amounts of data. These problems were solved by using Windows Presentation Foundation (WPF) graphics capabilities in conjunction with C# to develop a unique set of algorithms to display custom graphs of unlimited size with quick response.

Telemetry data from flight tests are normally plotted using MatLab™ or other third party software. For most of the trajectory and flight parameters, a static 2D or 3D line graph does not provide the proper data visualization that can be accomplished with 3D software. Commercial 3D software can be expensive and difficult to customize, and writing custom software using Direct3D or OpenGL can be complex and time consuming. These problems were overcome using C# and Windows Presentation Foundation (WPF) to quickly and easily create a 3D Telemetry Theater and Trajectory Visualization Tool to dynamically display actual and simulated flight tests.

Software development is of great significance in manufacturing and process industry. Software can, for example, make production more efficient and monitoring easier. In paperboard industry testing the product, that is, paperboard, is important. Testing is done to make sure that the product meets the requirements of the customers who transform the paperboard to packaging products. In the Skoghall paperboard Mill operators make use of a range of different test methods. Since the work pace is high and the product is being continuously tested improvements of the workflow at the laboratory are always of interest.   The aim of this project is to develop a program that connects test instruments at Skoghall Mill’s test laboratory to the Mill’s network. The motivation is to automate part of the work process by removing the step of manually reading measurement values from the test instruments. The scope of this project includes developing software that connects one of the instruments (the Short Span Compression tester [1] or SCT) at the laboratory to the Mill’s network. This program, named TestLink, will work as a proof of concept for connecting instruments using a common interface. The software is constructed using a modular approach forming a platform where an additional instrument can be connected by creating a new module. The software is developed in Visual Studio using C# as the programming language. The software architecture follows the MVVM [2] (Model View ViewModel) design pattern paradigm and WPF [3] (Windows Presentation Foundation) is used to construct the graphical user interface. The result shows that time savings can be made when automating part of the work with the introduction of new software.

Reliable authorization and authentication is now becoming necessary for many everyday activities such as boarding an aircraft, doing financial transactions etc. This work deals with the possibility of proposing an appropriate method of acquisition, processing and evaluation of data from the tablet itself, focusing on the possibility of obtaining data from the graphics tablet used in conjunction with a personal computer, for the purpose of verifying identity. It contains an overview of biometrics, as independent developing science over the last decades of the 20th century to the present, a summary of the development of a graphics tablet, the current situation, a brief overview of the most popular technologies used in their construction and distribution. Furthermore, it focuses on the design and implementation of appropriate procedures to obtain data provided by the Trust tablet TB - 7300, using available software development kit for the Microsoft Tablet PC, and a comparison of WinForms and WPF model (.NET Framework), with the declaration of suitability for the use of the second one. Last but not least, according to the response from the available set of users, this work focuses on the design of appropriate types of input data, test patterns, method and application for collecting and processing data from the graphic tablet, as well as their use for the verification of the identity of the individual. Ultimately, it is possible to discuss the accuracy of the method on a set of users, its suitability for use and shortcomings. Based on the analysis of the issue, the proposed method can be declared as under-optimal - it is safe, but inaccurate with low stability and repeatability. It is also important to accent that this is just the initial proposal, the use of applications in everyday life is for current phase impossible and the transition from laboratory conditions would require further development and improvement of the process.

Saab har utvecklat en produkt, Remote Tower, som tillåter flygledningen att fjärrstyra en flygplats, det vill säga att övervaka och kontrollera en flygplats på distans. Ett viktigt verktyg för flygledarna är VCS, Voice Communication System, som är Saabs egna röstkommunikationssystem, där både radio- och telefonkommunikation ingår. VCS används för att kommunicera med piloter och markpersonal på flygplatsen. Målet är att i framtiden kunna fjärrstyra fler flygplatser samtidigt, och då finns ett behov att visualisera de inkommande samtalen från de olika flygplatserna i realtid. Detta för att undvika ett scenario där flygledaren missar ett samtal. Ett system har utvecklats för just detta ändamål vars huvuduppgift är att visualisera de samtal som VCS producerar. Systemet kan hantera tre flygplatser samtidigt, och varje samtal kan spelas upp genom att klicka på de inkommande samtalen.

The goal of this thesis is to develop SCADA/HMI system for stations on leak test. The software will be able to communicate with the station via the OCP server, by which the control station and store measured data. For this purpose, it was used more fiber processing, as for the actual communication, as well as to run the program and work with databases to store the results. The resulting program will be placed on the personal computer, which is part of the station to check for leaks.

This master thesis describes development of Photography Geotagging Application from an introduction to the issues, through the analysis and design to the resulting implementation. The reader should be able understand to issues of time in geotagging using the GPS tracks records. He will also understand where and how metadata are stored. The big part of thesis is dedicated to the implementation of application which leads the reader through building the application architecture, the photography representation and loading their metadata with program ExifTool. Gradually will be explained the convertors of coordinates and GPS track parsers, which are used to synchronization. The final application allows the user synchronize photography with GPS track and display them on the map.

Företaget Saab AB i Karlstad har utvecklat produkten Paratus Pocket Translator för att hjälpa ambulans\-sjukvårdare kommunicera med patienter som inte talar samma språk. Eftersom denna produkt enbart finns på en utgående mobil plattform behöver Saab en omskrivning av denna produkt på en modern plattform. Denna uppsats beskriver projektet Mobile Translator vilket även blev produktens arbetsnamn. Projektets huvudsakliga syfte var en ny version av den tidigare produkten Paratus Pocket Translator för Windows med bibehållen funktionalitet. Projektet har resulterat i en Windows applikation baserad på moderna tekniker som tillfredställer Saabs behov och önskemål.
The company Saab AB in Karlstad has developed the product Paratus Pocket Translator to help paramedics communicate with patients who do not speak the same language. This product is only available on an older mobile platform that has since been replaced. Saab is now in need of a rewrite of this product on a more modern platform. This dissertation describes the project Mobile Translator, which also became the products working title. The project's main aim was a new version of the previous product Paratus Pocket Translator for Windows with maintained functionality. The project has resulted in a Windows application based on modern technologies that satisfies Saab's needs and requests.

Intrusion alarm systems (IAS) make inseparable part of our personal and work life which helps us with protection of important work data or personal property. This paper looks into the theory of IAS design and analysis which are two important factors influencing final system effectiveness. Furthermore, general procedure of IAS design, a number of analysis methods and IAS components are discussed. This theoretical knowledge is used to design and implement an aplication called IASPlanner, which allows users to draw a layout of IAS and subsequently analyse designed systems. The aplication can be used not only for the design itself, but also for educational purposes.

This diploma thesis deals with the study of leakage problems in industry and the use of Agilent weight detectors for its detection. Based on the studies conducted, a hardware and software solution has been proposed for connecting the measuring elements to a helium detector at a simple configurable station. Additionally, a C# application was created. This application allows the leak detector to be controlled and data collected from the detector and its surroundings. The created application was tested in a real environment. The conclusion of this work is devoted to the possibilities of using the created application.

The mission was to create a prototype of a menu system for the company

System Andersson AB. The instructions were that it has to be as interesting and

modern now as in 20 years. We had to look at technology from the cutting edge

of development to make our prototype survive in the future. Windows

Presentation Foundation (WPF) was the technique we focused on. There are

only a few applications that take advantage of WPF today, but it is predicted

that WPF will be the next generation method for developing applications to the

Windows platform.

To make our menu visually appealing, easy to use and at the same time

functional, we followed the clear path and guidelines that are available in

interaction design. Those guidelines tell us how the design process should be

executed. Through usage of the six basic ideas of interaction design and taken

the principles of richer user experience further, we were able to build a menu

system that will be as modern in 20 years as it is now. We took the thoughts of

physical design into account to make the menu understandable and easy to use.


Examensarbetets uppdrag var att skapa en prototyp av ett menysystem åt

System Andersson AB, som enligt instruktioner ska vara intressant och

modernt, även om 20 år. För att lyckas med detta tittade vi på "cutting edge" av

vad som används idag för att inte systemet skulle riskera att bli föråldrat i

framtiden. Tekniken som vi undersökte och använde oss av var Windows

Presentation Foundation (WPF). I dagsläget är det endast ett fåtal applikationer

som använder WPF men utvecklingsmetoden spås vara morgondagens sätt att

programmera windowsapplikationer.

För att vår meny skulle bli estetiskt tilltalande, enkel att använda och samtidigt

funktionell så finns det inom interaktionsdesign klara riktlinjer om hur

designutvecklingen bör gå till väga. Genom att nyttja de sex grundtankarna: att

ett system ska vara effektivt, ha hög verkningsgrad, säkert att använda, vara

användbart, enkelt att förstå samt okomplicerat; i designen och att bygga vidare

med principerna så anser vi att vi har kunnat åstadkomma ett menysystem som

kommer att se lika intressant och modern ut om 20 år som det gör nu. För att

menyn skulle uppfylla de sex grundtankarna, bland annat att vara klart

förståelig och enkel att använda så använde vi oss av de tankar och teorier som

framgår i fysisk design.

The aim of this project is to design an application for mobile platform which can insert virtual objects to camera shots without disturbing the scene. The application is implemented on Windows 8.1 and Windows Phone 8.1 platforms. The scene rendering is done in DirectX with C++ programming language and the user interface is created in XAML. This text describes the relevant fundamentals of augmented reality and the selected platform and it describes advanced programming techniques such as asynchronous resources loading or advanced post processing rendering techniques. The result of this project is a mobile application for inserting a virtual object with its shadow to camera shot. It is possible to set some aspects of the light and take a picture with the virtual object in high resolution.

The thesis is dealing with implementation of IMPRODEMO image processing application. Author has used existing image processing libraries and has implemented complex application with modern user interface. In order to develop the application, he used framework WPF, method for developing application using tests called Test Driven Development, design pattern MVVM and MEF tool for easy integrating of extensions. Chapter two deals with these technologies and methodologies and describes them. Third chapter describes design of application and presents class diagrams. Fourth chapter describes test driven development and particular pitfalls that emerged from using this methodology. Fifth chapter presents the mean of integrating code written in C++ language into .NET framework and into C# language. At the end, whole work is summarized, resuls are defined and possibilities of resuming and extending the work are proposed.

Sistemas de Bilhética tem como foco as empresas de transportes públicos. As consolas de bordo são o componente principal destes sistemas, permitem a emissão de bilhetes, validação de passes, ou seja, o controlo de receita gerada a cada serviço prestado pelos motoristas. Estes sistemas são sistemas embebidos com algumas restrições de desenvolvimento, e no que diz respeito a emissão de faturas estes equipamentos não têm a possibilidade de integrar na própria maquina a emissão de faturas. Este projeto está focado em descobrir soluções para a emissão de faturas, incidindo sobre o processo de certificação de software de faturação, bem como o desenvolvimento de um posto de venda de passes que permitira criar e carregar passes emitindo a fatura na hora do pagamento. Sendo este um mundo de base de dados transacional existe o problema na parte de relatórios em que este projeto pretende dar resposta e ter uma solução mais rápida e eficiente, para ajudar na tomada de decisões operacionais.
Ticketing Systems focuses on public transport companies. The on-board consoles are the main component of these systems, they allow the issuing of tickets, validation of passes, the control of revenue generated for each service provided by the drivers. These systems are embedded systems with some development constraints, and with regard to the issuance of invoices these equipment’s do not have the possibility of integrating in the machine the issuance of invoices. This project is focused on finding invoice solutions, focusing on the billing software certification process, as well as developing a ticket sales point that would allow you to create and top-up pass by issuing the invoice at the time of payment. Being among a world of transactional database there is the problem in the part of reports in which this project intends to respond and have a solution, a fast and efficient decision support system.

The work includes implementation of the server portal for real-time data evaluation. The next part of the thesis describes current software technologies used in the development of desktop and server applications. The design and implementation of both parts of the solution is a major part of the work. In the last part, the real-time functionality test of the application is presented.

This master’s thesis deals with the analysis, design and implementation of a system for organizing running races with the support of racers registration and the possibility of multiple online and offline checkpoints during the race. The system is composed of two web applications and a client desktop application. An administration web application is used to create and manage the race. A presentation web application with optional registration is responsible for publishing the race. It is possible to scan RFID chips of the racers on multiple track stations during the race using a client application. The RFID chips are scanned by an RFID reader connected to host device. If the client application is connected to the internet, the scanned times of racers are displayed on the presentation application in real time. Web applications are implemented with the ASP.NET MVC technology. Authorization and Authentication is provided by ASP.NET Identity. The client application is implemented using the WPF technology.

Den här rapporten behandlar ett system som använder en databas som lagringsplats för loggar. En bra metod för att hämta ut dessa loggar saknades och databasdesignen behövde förbättras för sökningar i loggarna. En applikation för att hämta och söka i loggposter från databasen skapades. En undersökning om hur databasdesignen kunde förbättras genomfördes också. Båda delarna gjordes i ett projekt för att de hörde ihop. Applikationen skulle använda databasen. Då jag inte kunde göra vilka ändringar jag ville i databasen gjordes relativt begränsade ändringar i den. Större ändringar utreddes teoretiskt. Applikationen gjordes mot den existerande databasdesignen, med ett undantag: en vy lades till. Rapporten undersöker index och andra metoder att göra sökningar i en databas snabbare. En metod för att hämta data inom ett intervall i en databas utvecklades och den beskrivs i rapporten. Metoden söker efter all data som har värden på en kolumn som faller inom ett intervall och där databasen är ordnad, eller nästan ordnad, på den kolumnen. Metoden ger oexakta svar om databasen är nästan ordnad på den kolumnen. Den är snabbare än en motsvarande exakt sökning.
This report considers a system where a database is used as the back-end storage for logging. A suitable method for extracting information from the logs was missing and the database design needed an improvement for log searching. An application for extracting and filtering the logs was created. An evaluation of how the database could be improved was also performed. Both parts were done in one project since they were heavily connected. The application would use the database. Since I couldn’t make arbitrary changes to the database only relatively limited changes were made in practice. Larger changes were evaluated theoretically. The application was made against the existing database, with one exception: a view was added. The report handles indexes and other methods for speeding up database searches. A method for fetching data inside an interval in a database was developed and is described in the report. The method searches for all data where the value of a column is inside an interval and the database is ordered, or almost ordered, on that column. The method gives inexact answers if the database is almost ordered on that column. It is faster than a corresponding exact search.

The platform Frama-C is a tool dedicated to analysis of source code of software written in C, with the possible types of analysis provided by plugins attached to the platform. One of its plugins is WP, used for deductive veri cation of C code with ACSL, a formal speci cation language. This dissertation describes the extension of this plugin, named WPTrans. This extension allows generated proof obligations from WP to be manipulated through inference rules and sent, at any stage of the proof, to automatic (mainly SMT solvers) and interactive theorem provers. Some proof obligations may be proved automatically, while others can be too complex to be solved by automatic theorem provers, requiring the users of Frama-C and WP to handle them manually. This approach usually requires a signi cant experience in proof strategies. Some interactive theorem provers provide communication with automatic provers. However, this connection can be complex and incomplete, leaving the user with the manual proof option only. The strength of WPTrans is to combine the features of automatic and interactive theorem provers in a precise and complete way, with a simple manipulation language. Thus, the user can simplify the proof obligations enough in order for its proof to be concluded with an SMT solver, letting the proof be partially manual and partially automatic. Nevertheless, the plugin is directly linked do WP, facilitating the installation of the extension in Frama-C. This tool is also a gateway to other possible features, which we discuss herein.

Bei den meisten Bakterien wird die Biofilmbildung durch das Botenmolekül c-di-GMP stimuliert. Durch die enzymatische Aktivität von c-di-GMP-synthetisierenden Diguanylatzyklasen/DGC und c-di-GMP-abbauenden Phosphodiesterasen/PDE wird der c-di-GMP-Gehalt als eine Antwort auf diverse Stress- und suboptimale Umweltbedingungen reguliert. Vor allem Gram-negative Bakterien haben multiple DGC/PDE. So besitzt Escherichia coli K-12 29 solche Proteine, darunter 12 DGC, 13 PDE sowie 4 degenerierte Proteine ohne enzymatische Funktion. Dieses komplexe c-di-GMP-Kontrollsystem reguliert die Produktion der extrazellulären Biofilmmatrix, die in E. coli während des Übergangs in die stationäre Wachstumsphase stattfindet. Das Ziel dieser Arbeit war es zu untersuchen, ob die 29 DGC/PDE von E. coli ein spezifisches Interaktom bilden, das DGC/PDE-Pärchen enthält. Durch umfangreiche Two-Hybrid-Untersuchungen konnte gezeigt werden, dass es ein solches Interaktom in der Biofilmregulationskaskade tatsächlich gibt, das allerdings nicht in Pärchen organisiert ist. Vielmehr wird die Biofilmbildung von einer Kerngruppe von Enzymen, welche multiple Interaktionen untereinander und mit anderen DGC/PDE aufweisen, kontrolliert. Die Funktionsweise der Kerngruppe von Enzymen könnte jedoch möglicherweise unter bestimmten Wachstumsbedingungen durch Interaktionen mit weiteren Proteinen moduliert werden. Die Dynamik des Interaktionsnetzwerks ermöglicht vermutlich eine rationelle Ressourcenverwaltung in den verschiedenen Zonen des Biofilms, was zum Aufbau der komplexen Matrixarchitektur beitragen könnte, und eine hohe Anpassungsfähigkeit der Bakterien und der von ihnen aufgebauten Biofilmstrukturen gewährleisten könnte. Insgesamt führt diese Arbeit aus einer systemischen Perspektive zu einem neuen Modell der lokalen Biofilmbildungsregulation durch den Botenstoff c-di-GMP und legt die Basis für weitere Untersuchungen der daran beteiligten Mechanismen einzelner GGDEF/EAL-Domäne-haltiger Proteine in E. coli.
The messenger molecule c-di-GMP stimulates the formation biofilms in most bacteria species. The enzymatic activities of the diguanylate cyclases/DGC and the phosphodiesterases/PDE adjust the c-di-GMP content in response to diverse stress and suboptimal environmental conditions. Above all, Gram-negative bacteria have multiple GGDEF/EAL domain proteins. Escherichia coli K-12 possesses 29 of such proteins: 12 DGCs, 13 PDEs and 4 so called degenerate proteins without any enzymatical function. Mainly, this complex c-di-GMP control system regulates the production of the extracellular biofilm matrix, which in E. coli takes place during the switch into the stationary growth phase. The main compounds of the matrix are amyloid curli-fibers and exoplysaccaride cellulose. The goal of this work was to investigate, whether the 29 DGC/PDE from E. coli develop a specific interactome containing additional DGC/PDE pairs. In a comprehensive two-hybrid study, it could be demonstrated that there is indeed a specific interactome in the biofilm formation cascade. However, this interactome does not contain additional DGC/PDE pairs. Mainly, the core group of enzymes, which have multiple interactions among each other and with other DGC/PDE, controls biofilm formation. Under certain growth conditions the mode of action of the core enzymes might be adjusted through the interaction with other proteins. Presumably, the dynamics of the interaction network allows managing the resources in the different biofilm zones efficiently, which could conribute to the complex organisation of the matrix architecture. Therefore, the rapid adaptation of bacteria and the formed biofilm structures could be better organized. Altogether, this work provides a new model for the local regulation of the biofilm formation by the secondary messenger c-di-GMP guided from a systemical perspective. Hereby, the basis for further investigations on regulation mechanisms of individual DGC/PDE was set.
Переход от подвижного и планктонообразного образа жизни к формированию биоплёнок является важной и интересной особенностью различных микроорганизмов. Кишечная палочка (Escherichia coli) представляет собой удобный модельный организм для изучения подобных трансформаций. У этой грамотрицательной бактерии образование биоплёнки обусловлено внутриклеточной аккумуляцией циклического дигуанилата (цикло-диГМФ). Известно, что активность ферментов, синтезирующих (дигуанилатциклазы/ДГЦ) и разлагающих (диэстеразы/ДЭ) эти сигнальные молекулы, меняется в ответ на стрессовые и субоптимальные раздражители. Кишечная палочка имеет 12 ДГЦ, 13 ДЭ и четыре дегенерированных протеина. Цель данной работы – изучить специфический, важный при формировании биоплёнки интерактом и выяснить, способны ли другие ДГЦ/ДЭ образовывать дополнительные ДГЦ/ДЕ модули и вносить свой вклад в формирование биоплёнки. В работе были изучены молекулярные взаимодействия, ответственные за формирование биоплёнок у кишечной палочки. Так, было доказано отсутствие в интерактоме дополнительных локальных ДГЦ/ДЕ модулей, участвующих при каскадных процессах регуляции роста биоплёнки. Установлено, что процесс формирования биоплёнки в большей степени контролируется основной группой ферментов, которые имеют множественные взаимодействия между собой и с другими ДГЦ/ДЭ. Вероятнее всего, такие взаимодействия способны модулировать работу основных ферментов при определенных условиях культивирования. Динамика подобной сети взаимодействий позволяет микроорганизмам целесообразно использовать свои клеточные ресурсы при образовании биоплёнок и вносит свой вклад в её сложную архитектуру, повышая тем самым приспособляемость бактерий и созданных ими сложных биоплёночных структур к внешним условиям. В целом, в данной работе предложена новая модель локальной регуляции образования биоплёнки с помощью сигнальной молекулы цикло-диГМФ и заложен фундамент для дальнейших исследований механизмов действия отдельных ДГЦ/ДЭ.

Für die E. coli K12 Biofilmbildung ist die Expression des Masterregulators CsgD essentiell. Dies erfordert das Signalmolekül c-di-GMP, dessen Auf- und Abbau durch 12 Diguanylatzyklasen (DGCs mit GGDEF-Domänen) und 13 Phosphodiesterasen (PDEs mit EAL-Domänen) erfolgt. DgcE ist mit einer MASE1-umfassenden Transmembranregion (TM), drei PAS-, einer GGDEF- und einer degenerierten EAL-Domäne die strukturell komplexeste DGC und notwendig für die Biofilmbildung. Diese Arbeit zeigt, dass die Aktivität von DgcE einer hoch komplexen Regulation unterliegt. Einzelnen DgcE-Domänen konnten aktivierende bzw. inhibierende Rollen hinsichtlich der Biofilmmatrixsynthese zugeordnet werden. Die Biofilmbildung hängt von DgcE-produziertem c-di-GMP ab, wobei die DgcE-Dimerisierung v.a. durch die PAS-Region vermittelt wird. Die EAL-Domäne wirkt einer aktiven DgcE-Form entgegen. Für die DgcE-vermittelte Matrixproduktion sind die GTPase YjdA und sein Partnerprotein YjcZ nötig. Über Interaktionen mit YjcZ und der TM von DgcE vermittelt YjdA eine Komplexbildung. Die Interaktion von YjdA und DgcE sowie die Matrixproduktion hängen von der GTPase-Aktivität von YjdA ab. GTP wird daher als intrazelluläres Signal vorgeschlagen, das die Aktivierung von DgcE durch YjdA/YjcZ reguliert. Die MASE1-umfassende TM agiert als Zentrale der Signalintegration. Einerseits ist sie nötig für die DgcE-Aktivität und andererseits ist sie an einem massiven Abbau von DgcE beteiligt. Zudem wurden neu identifizierte Curli-regulierende Gene (rbsK, rbsR, ydcI, yieP, puuR) untersucht, wobei keines über das PdeR/DgcM/MlrA-Modul in die c-di-GMP-vermittelte CsgD-Expression eingreift. Flagellare Verknotungen in der unteren Schicht von E. coli Makrokolonien tragen zur Morphogenese dieser Makrokolonien bei. Diese Arbeit zeigt, dass Flagellenverknotungen zu einer verminderten Expression der Master-PDE PdeH beitragen, wodurch vermutlich die zelluläre c-di-GMP-Konzentration steigt und somit die Biofilmbildung begünstigt wird.
Biofilm formation of E. coli K12 requires the expression of the biofilm master regulator CsgD. This process depends on the signaling molecule c-di-GMP, which is synthesized by 12 diguanylate cyclases (DGCs with GGDEF domains) and degraded by 13 phosphodiesterases (PDEs with EAL domains). DgcE is the most complex DGC with a MASE1-containing transmembrane region (TM), three PAS, a GGDEF and a degenerate EAL domain, and it is essential for biofilm formation. This work shows that the regulation of the DgcE activity is highly complex. It was possible to assign activating and inhibitory roles to single domains of DgcE with regard to the expression of biofilm matrix components. C-di-GMP produced by DgcE is necessary for biofilm matrix production. The dimerization of DgcE is mainly mediated by the PAS region, whereas the EAL domain counteracts an active form of DgcE. DgcE-mediated matrix synthesis requires the activating signal input of the GTPase YjdA and its partner protein YjcZ. DgcE, YjdA and YjcZ form a protein complex in which YjdA directly interacts with YjcZ and the TM of DgcE. The interaction between DgcE and YjdA as well as the matrix expression depend on the GTPase activity of YjdA. Thus, it is proposed that GTP serves as an intracellular signal regulating the activation of DgcE by YjdA/YjcZ. The MASE1-containing TM proved to be a central hub for signal integration. It is both required for DgcE activity and for a massive degradation of DgcE. Furthermore, newly discovered curli-regulating genes (rbsK, rbsR, ydcI, yieP, puuR) have been analyzed. None of those gene products act on CsgD expression via the PdeR/DgcM/MlrA module. Flagellar entangling within the bottom layer of E. coli macrocolonies determines morphogenesis of macrocolonies. The data presented here suggest that the master PDE PdeH is somehow down-regulated by flagellar entangling, which probably results in a higher cellular c-di-GMP concentration, thereby promoting biofilm formation.

Kutane T-Zell-Lymphome (CTCL) stellen eine heterogene Gruppe von non-Hodgkin-Lymphomen dar. Die häufigsten Entitäten sind die Mycosis fungoides, das Sézary-Syndrom sowie CD30+ lymphoproliferativen Erkrankungen (cALCL). Todesliganden-vermittelte Apoptose ist auch für die Lymphozyten-Homöostase von essentieller Bedeutung. Zunächst wurden die CTCL-Zelllinien mit systemischen T-Zell-Lymphomzellen hinsichtlich ihrer Apoptosesensitivität verglichen. Hierbei zeigte sich eine ausgesprochene Resistenz aller kutanen Zelllinien gegenüber TRAIL- und TNF-α-induzierter Apoptose. Anhand der Aktivierung der Caspasensignalkaskade war eine frühe Blockierung der Apoptose nachweisbar. Überdies konnte der Verlust von CD95, TNF-R1, Caspase-10 sowie von Bid in verschiedenen CTCL-Zelllinien nachgewiesen werden. Die Apoptoseresistenz konnte hingegen mit einer generellen und starken Expression des anti-apoptotischen Proteins c-FLIP korreliert werden. Des Weiteren wurde der Einfluss von CD30-Stimulation auf die durch CD95-induzierte Apoptose in CD30+-Zelllinien untersucht. Hierbei zeigte sich, dass CD30 Stimulation zur Aktivierung von NF-kB und zu erhöhter c-FLIP Expression führt, was mit Apoptoseresistenz korrelierte. Die Bedeutung der in vitro-Ergebnisse zeigte sich auch durch eine weitgehende Parallelität dieser zu untersuchten Biopsieproben von CTCL-Patienten. Schließlich wurden verschiedene NSAIDs auf ihre Fähigkeit hin untersucht, die Expression von c-FLIP zu vermindern und Apoptose zu induzieren. Dies wurde sowohl in CTCL-Zelllinien als auch in Tumor-T-Zellen von CTCL-Patienten untersucht. NSAIDs aktivierten beide Apoptosesignalwege und Diclofenac sensitiviert überdies für TRAIL-vermittelte Apoptose. In Ihrer Gesamtheit vermittelt die vorliegende Arbeit einen Einblick in die komplexen Apoptoseregulationsmechanismen in kutanen T-Zell-Lymphomen und identifiziert c-FLIP als einen zentralen Resistenzfaktor sowie NSAIDs als mögliche CTCL-Therapieoption.
Cutaneous T cell lymphomas (CTCL) form a heterogeneous group of non-Hodgkin lymphomas. Its most frequent forms are Mycosis fungoides, Sézary syndrome and CD30-positive cutaneous anaplastic large-cell lymphoma. Death ligands critically contribute to lymphocyte homeostasis by induction of apoptosis and may further represent safeguard mechanisms to prevent lymphoma development and tumor growth. First, we analyzed induction of apoptosis by death ligands in CTCL cell lines and compared it to that in systemic T-cell lymphoma cells. This revealed for CTCL cells a pronounced resistance to death ligands. In particular, there was no caspase activation in non-responsive CTCL cells, indicating an early blockade of the apoptosis signal. Furthermore a loss of CD95, TNF-R1, caspase-10, as well as of Bid was found in several cell lines. Changes at the receptor expression level were largely ruled out, whereas, consistent and strong expression of c-FLIP was correlated with resistance. In addition, we investigated the CD30/CD95 crosstalk in CD95-sensitive cALCL cell lines. Experiments showed that CD30 ligation leads to NF-κB-mediated c-FLIP upregulation in cALCL cells, which in turn conferred resistance to CD95-induced apoptosis. Parallels with regard to expression of apoptosis regulators were seen in peripheral blood mononuclear cells and biopsies of CTCL patients. Finally, we evaluated non-steroidal anti-inflammatory drugs (NSAIDs) for their capacity to downregulate c-FLIP expression and apoptosis induction in CTCL cells as well as in tumor T cells from CTCL patients. NSAID treatment resulted in an activation of both apoptosis signaling pathways and, furthermore, Diclofenac pre-treatment sensitized for TRAIL-induced apoptosis. In conclusion, this study elucidates defects in apoptosis regulation, proved the significance of c-FLIP for the survival of CTCL cells and provides a rationale for the use of NSAIDs as a potentially new therapeutic option for CTCL patients.

Der sekundäre Botenstoff c-di-GMP kommt in vielen Bakterienspezies vor und fördert dort die Bildung von Biofilmen. Für Abbau und Synthese von c-di-GMP in der Zelle sorgen Diguanylatzyklasen (DGCs) und Phosphodiesterasen (PDEs), an deren N-Terminus häufig Sensordomänen die enzymatische Aktivität steuern. In dieser Arbeit wurde die Regulation und Wirkungsweise einer Gruppe von PDEs mit einer neuartigen Sensordomäne, der CSS-Domäne, genauer charakterisiert. Die CSS-Domäne ist im Periplasma lokalisiert und besitzt zwei hochkonservierte Cysteine, von denen eines in dem namensgebenden CSS-Motiv arrangiert ist. Die Integration in die innere Membran erfolgt durch zwei Transmembrandomänen (TM1 und TM2), wobei TM2 die Verbindung zu der C-terminal gelagerten EAL-Domäne mit PDE-Aktivität herstellt. Die Analyse von PdeC als eine von fünf CSS-PDE in Escherichia coli K-12 zeigte, dass die Bildung einer Disulfidbrücke zwischen den konservierten Cysteinen vom oxidierende DsbA/DsbB-System katalysiert wird und zu einer Verminderung der enzymatischen Aktivität der EAL-Domäne führt. Im Gegensatz dazu führt die reduzierte freie Thiol-Form der CSS-Domäne zu einer stark erhöhten PDE-Aktivität verbunden mit der Dimerisierung über die TM2. Die Reduktion der CSS-Domäne führt außerdem zur Prozessierung durch die HtrA-Proteasen DegP und DegQ in ein membranständiges Fragment aus TM2+EAL-Domäne mit hoher enzymatischer Aktivität. Der Abbau durch DegP und DegQ im Periplasma ist sehr effizient. Auf der cytoplasmatischen Seite hingegen erfolgt die weitere Degradierung durch noch unbekannte Proteasen eher langsam, wodurch es unter bestimmten Bedingungen zur Akkumulierung der hochaktiven TM2+EAL-Form kommt. Durch das Zusammenspiel von redox-abhängiger Aktivitätskontrolle und Proteolyse der c-di-GMP-spezifischen PDE PdeC wird die Produktion der amyloiden Curli-Fasern und Cellulose reguliert, welche Hauptbestandteil der extrazellulären Matrix von Biofilmen sind.
The second messenger c-di-GMP promotes biofilm formation in many bacterial species. Phosphodiesterases (PDEs) and diguanylatcyclases (DGCs) - often controlled by various N-terminal sensor domains - degrade and synthesize c-di-GMP. The aim of this study was to characterize the mode of activation and physiological function of a new class of sensor domains, the CSS domain, which is coupled to an EAL domain with PDE activity and therefore potentially controlling c-di-GMP degradation. The CSS sensor domain contains two highly conserved cysteins in the periplasmic loop, of which one is arranged in the characteristic CSS motif. Integration into the inner membrane is ensured by two flanking transmembrane domains (TM1 and TM2), with TM2 providing a connection to the C-terminal EAL domain. Analysis of PdeC as one of 5 CSS-PDEs in Escherichia coli K-12 revealed a close linkage to the disulfide bond (DSB) system as well as important periplasmic proteases. Thus, formation of a DSB between the two conserved cysteins is promoted by the oxidizing DsbA/DsbB-system and reduces enzymatic activitiy of the EAL domain. In contrast, the free thiol form increases PDE activity and dimerizes via the TM2 domain. Moreover, reduction of the CSS domain results in degradation by the periplasmic HtrA proteases DegP and DegQ. These redundantly process PdeC to a shorter fragment containing the TM2 and EAL domain only, which shows dimerization as well and has high PDE activity. Degradation in the periplasm mediated by DegP and DegQ is very efficient. In contrast, further proteolysis in the cytoplasm by yet unidentified proteases is rather slow, allowing the accumulation of the highly active TM2+EAL form. Finally, the interplay of redox dependent activity control and proteolysis of the c-di-GMP specific PDE PdeC regulates production of curli and cellulose as major matrix components of the bacterial biofilm.

Lebewesen nutzen nukleotid-basierte sekundäre Botenstoffe um extra- und intrazelluläre Signale zur Induktion einer entsprechenden Zellantwort weiterzuleiten. Das zyklische Dinukleotid c-di-AMP steuert verschiedene physiologische Prozesse und ist für viele Bakterien unter bestimmten Bedingungen essentiell. Dieses Signalmolekül muss präzise reguliert werden, da seine Akkumulation oft toxisch ist. Diadenylatzyklasen mit einer DAC-Domäne synthetisieren c-di-AMP, welches von Phosphodiesterasen (PDE) mit DHH/DHHA1- oder HD-Domänen abgebaut wird. Streptomyceten sind im Boden lebende, Gram-positive Actinobakterien mit einem komplexen Lebenszyklus, während welchem sie vielzählige sekundäre Metabolite, inklusive Antibiotika, produzieren. Die Regulierung des zellulären c-di-AMP und seine Bedeutung in der Streptomyceten-Biologie waren zu Beginn dieser Studie weitgehend unbekannt. Zur c-di-AMP-Synthese nutzen Streptomyces die DAC DisA, besitzen aber keine der typischen PDEs sowie die meisten der bekannten c-di-AMP-bindenden Effektoren. Diese Arbeit zeigt, dass DisA die wichtigste c-di-AMP-Synthetase in Streptomyces venezuelae ist. AtaC wurde als eine PDE identifiziert, welche eine neue Klasse von c-di-AMP PDEs begründet. Während eine ataC-Deletion zu Störungen in Differenzierung und Wachstum in S. Venezuelae führt, führt die Inaktivierung von disA zur Sensitivität gegenüber erhöhten Konzentrationen von monovalenten Kationen im Medium. CpeA und CpeD wurden als erste c-di-AMP-bindende Proteine im Streptomyces Signalnetzwerk charakterisiert. Die entsprechenden Gene sind in Operons mit putativen Kation/Proton-Antiportern cpeB bzw. cpeE kodiert und die jeweiligen Genprodukte interagieren c-di-AMP-abhängig in vivo. Obwohl Deletion von cpe und Überexpression von cpeABC in S. venezuelae keine Phänotypen zeigten, verbesserte die CpeABC-Expression in Escherichia coli das Wachstum in Kalium-supplementierten Medien, was auf eine Funktion von cpe in der Regulation von Kalium hindeutet.
Nucleotide second messengers are used by all forms of life to transduce extra and intracellular signals and translate them into a physiological cell response. The cyclic dinucleotide c-di-AMP is a signaling molecule involved in diverse functions in bacterial physiology and is essential for many bacteria under certain growth conditions. However, this second messenger has to be tightly regulated since increased levels of c-di-AMP can be toxic. In many bacteria diadenylate cyclases with a conserved DAC domain synthesize c-di-AMP and phosphodiesterases (PDEs) with a DHH/DHHA1 or HD domain degrade it. Streptomyces spp. are soil-inhabiting gram-positive Actinobacteria characterized by a sophisticated developmental life cycle during which they produce various secondary metabolites, including antibiotics. The regulation and role of c-di-AMP is not well understood in Streptomyces biology. For c-di-AMP synthesis, streptomycetes utilize the DAC DisA but do not encode any canonical PDE and most of the known effector proteins for c-di-AMP signal transduction are absent. In this work, I demonstrated that DisA is the primary c-di-AMP synthetase in Streptomyces venezuelae and characterized AtaC as the founding member of a novel class of c-di-AMP-specific PDEs. In S. venezuelae, deletion of ataC interferes with development and growth, whereas disA inactivation affects bacterial survival under high ion osmotic stress conditions. Further, I identified CpeA and CpeD as the first c-di-AMP-binding proteins in Streptomyces. The respective genes are encoded in operons with the predicted cation/proton antiporters cpeB and cpeE, respectively, and the gene products interact in vivo in a c-di-AMP-dependent manner. Although neither cpe deletion nor overexpression of cpeABC produced a phenotype in S. venezuelae, expression of cpeABC in Escherichia coli improved growth in liquid media supplemented with potassium, suggesting that Cpe transporters are involved in potassium homeostasis.

Die fünf CSS-Domänen Phosphodiesterasen aus Escherichia coli K12 (E. coli) gehören zu den weit verbreiteten c-di-GMP-PDEs. Ein Vertreter, PdeC, wurde bereits von Herbst et al. (2018) charakterisiert. Durch DsbA/DsbB geförderte Disulfidbrückenbindung (DSB) in der CSS-Domäne von PdeC wird die PDE-Aktivität des Proteins gehemmt. Gegenteilig ist die freie Thiolform, in Abhängigkeit von der TM2 als Dimerisierungs-Domäne, enzymatisch aktiver. Diese Form wird von den periplasmatischen Proteasen DegP und DegQ prozessiert. Ein irreversibel aktiviertes TM2+EAL-Fragment wird generiert, dass durch weitere Proteolyse langsam entfernt wird. Die Reduktion der CSS-Domäne von PdeC zur der freien Thiolform stimuliert die PDE-Aktivität der EAL-Domäne in vitro. Zusammen mit den Daten von Herbst et al. (2018) wird die CSS-Domäne in dieser Arbeit als eine neue sensorische Domäne charakterisiert, dessen Aktivität durch einen DSB/Thiol-Schaltmechanismus reguliert wird. Ein weiterer Vertreter de CSS-Domänen PDEs, PdeN, scheint nicht über die Ausbildung einer DSB in der CSS-Domäne reguliert und aktiviert zu werden. Nach erfolgter Proteinbiosynthese wird die Proteinkonzentration vielmehr über den N-Terminus reguliert, wobei saure Wachstumsbedingungen das Protein maßgeblich induzieren und die Aktivität erhöhen. Wird das Protein erfolgreich in die Membran eingelagert, kann es bedingt durch die strukturelle DSB seine PDE-Aktivität entfalten und die Biofilmmatrixproduktion maßgeblich beeinflussen.
The five CSS domain phosphodiesterases from Escherichia coli K12 (E. coli) belong to the widespread group of c-di-GMP PDEs. One representative, PdeC, has already been characterized by Herbst et al. (2018). DsbA/DsbB promoted disulfide bond (DSB) formation in the CSS domain of PdeC inhibits the PDE activity of the protein. On the contrary, the free thiol form is more enzymatically active, depending on the TM2 as the dimerization domain. This form is processed by the periplasmic proteases DegP and DegQ. An irreversibly activated TM2 + EAL fragment is generated that is slowly removed by further proteolysis. The reduction of the CSS domain of PdeC to the free thiol form stimulates the PDE activity of the EAL domain in vitro. Together with the data from Herbst et al. (2018) the CSS domain is characterized as a new sensory domain whose activity is regulated by a DSB / thiol switch mechanism. All five E. coli K12 CSS domain PDEs share a similar domain architecture, but redox biochemistry, proteolysis, and PDE activity differ within the protein family. On the basis of the PDE activity of non-DSB variants, PdeB, PdeC and PdeG were characterized as activatable (reduction increases PDE activity) and PdeD and PdeN as non-activatable (reduction inactivated PDE activity). Another representative of the CSS domain PDEs, PdeN, does not seem to be regulated and activated by forming a DSB in the CSS domain. After protein biosynthesis the protein concentration is rather regulated via the N-terminus, with acidic growth conditions significantly inducing the protein and increasing its activity. If the protein is successfully inserted in the membrane, it can develop its PDE activity due to the structural DSB and influence the biofilm matrix production significantly.

CRM Treasury Systems AB is one of the Nordic Countries’ leading companies within treasury management. Their main product CRM Finance is a software program for financial administrators with roots back to the early 90s. Computer usage and software has changed considerably since then and the company is currently considering choosing a new technology to base future products upon. In this thesis Oracle ADF, Microsoft Lightswitch beta 1 and Silverlight with WCF RIA Services, are compared in order to select a suitable technology to base future applications on. The frameworks are used to implement a simple application with a small number of screens displaying database data in order to get to know the frameworks and capture the time required to get started. After the basic applications have been implemented in all the three frameworks and evaluated it is decided to continue by implementing a number of more advanced authorization and internationalization scenarios. It is decided to continue with only two of the selected frameworks due to lack of time once the simple application has been implemented successfully. Lightswitch beta 1 is chosen for discontinuation due to an uncertain future for the application as it is the first public available beta of the product. Based on the experience from the implementations performed, the time required for them as well as how the platforms fit the company’s requirements the .NET platform with Silverlight + WCF RIA Services is recommended as the base for future applications.

Streptomyceten weisen einen komplexen Lebenszyklus auf, dessen Verlauf durch den sekundären Botenstoff Bis-(3´- 5´)-zyklisches dimeres Guanosinmonophosphat (c-di-GMP) und die c-di-GMP-Effektorproteine BldD und RsiG reguliert wird. Der Auf- bzw. Abbau von c-di-GMP wird von Diguanylatzyklasen (DGC) mit GGDEF-Domänen bzw. Phosphodiesterasen (PDE) mit EAL- oder HD-GYP-Domänen katalysiert. In S. venezuelae, einem Modellorganismus der Streptomyceten, konnten zehn potenziell c-di-GMP metabolisierende Enzyme identifiziert werden, von welchen mit RmdA und RmdB zwei GGDEF-EAL-Tandem-Proteine im Fokus dieser Arbeit stehen. Die chromosomale Deletion der für RmdA und RmdB kodierenden Gene führt zu einer ausgeprägten Verzögerung der Entwicklung in S. venezuelae. Mit Hilfe chromosomaler Mutationen konnten die EAL-Motive der EAL-Domänen als essenziell für die in vivo Funktion beider Proteine identifiziert werden. Beide Proteine zeigen in vitro PDE-Aktivität und RmdA konnte als bifunktionales Enzym charakterisiert werden, da es in vitro auch DGC-Aktivität aufweist. Mittels Nukleotidextraktionen konnte RmdB als Haupt-PDE in S. venezuelae identifiziert werden, welche über den gesamten Entwicklungsverlauf für den Abbau von c-di-GMP verantwortlich ist. Aber auch RmdA hat während des Übergangs von der vegetativen zur reproduktiven Wachstumsphase Einfluss auf die globale zelluläre c-di-GMP Konzentration. Durchgeführte Transkriptomanalysen und qRT-PCR-Experimente ergaben, dass in den Deletionsmutanten die Expression einiger wichtiger entwicklungsspezifischer Gene im Vergleich zum Wildtyp herunterreguliert ist. Dies ist vermutlich auf die erhöhten c-di-GMP Konzentrationen in den Deletionsmutanten zurückzuführen, wodurch die Aktivität der c-di-GMP-Effektorproteine BldD und RsiG beeinflusst wird und die verzögerte Entwicklung der Deletionsmutanten erklärt werden kann. Weiterhin konnte gezeigt werden, dass RmdB mit dem Sigmafaktor der Sporulation, WhiG, interagieren kann.
Streptomycetes show a complex life cycle. The transition between the different developmental stages is regulated by the secondary messenger bis- (3´- 5´) -cyclic dimeric guanosine monophosphate (c-di-GMP) and the c-di-GMP effector proteins BldD and RsiG. c-di-GMP is synthesized by diguanylate cyclases (DGCs) with GGDEF domains, and its degradation is catalyzed by phosphodiesterases (PDE) with EAL or HD-GYP domains. In S. venezuelae, the Streptomyces strain which was used as a model organism in this work, there are ten potentially c-di-GMP metabolizing enzymes, of which two GGDEF-EAL tandem proteins, RmdA and RmdB, are the focus of this work. The deletion of the genes coding for RmdA and RmdB leads to a pronounced developmental delay in S. venezuelae. With the help of chromosomal mutations, the EAL motif was identified as essential for the in vivo function of RmdA and RmdB. Furthermore, both proteins were characterized in vitro as active PDEs and RmdA as a bifunctional enzyme, which also showed DGC activity. RmdB was identified as the master PDE in S. venezuelae by means of nucleotide extraction and is responsible for the hydrolysis of c-di-GMP over the course of development investigated. Also RmdA has an influence on the global cellular c-di-GMP concentration during the transition from the vegetative to the reproductive growth phase. A transcriptome analysis, qRT-PCR experiments and related follow-up experiments showed that the deletion of rmdA and rmdB leads to a differential expression of genes which code for important development-specific factors and regulators. This is presumably due to the increased c-di-GMP concentrations in the deletion mutants, with the c-di-GMP effector proteins BldD and RsiG delaying the transition to the next growth phase. Furthermore, it could be shown that RmdB can interact with the sigma factor of sporulation, WhiG.

In dieser Arbeit wurde die Affinitätsreifung des murinen anti-c-myc-Peptid-Antikörpers 9E10 analysiert. Hierfür wurden Fab-Fragmente mit Keimbahnrückmutationen gentechnisch hergestellt und in ihrem Bindungsverhalten zum humanen c-myc-Peptid charakterisiert. Das von 9E10 erkannte Epitop besitzt die Aminosäuresequenz EQKLISEEDLLRKR mit den darin sehr selektiv erkannten Schlüsselpositionen LISEXXL.Der 3300-fache Affinitätsgewinn während der 9E10-Reifung kommt sowohl durch eine Zunahme der Assoziations- als auch durch eine Abnahme der Dissoziationsgeschwindigkeit des Komplexes zustande. Der Affinitätsgewinn resultiert weniger aus zusätzlichen Kontakten des Antikörpers zum Peptid, sondern vor allem aus der Beeinflussung der Konformation und/oder der Flexibilität der an der Bindung beteiligten CDRs. Die außergewöhnlich lange CDR-H3 liefert einen wesentlichen Beitrag zur Affinitätsreifung. Die variable leichte Domäne dient dabei mit der langen CDR-L1 und -L3 als eine Bindungsplattform für die flexible CDR-H3. Änderungen in der Spezifität von 9E10 sind vorrangig auf die Reifung der variablen schweren Domäne zurückzuführen. Dabei ist die selektive Erkennung der Schlüsselpositionen im Peptid im Anfangsstadium der Affinitätsreifung von 9E10 stark ausgeprägt.
In this work the affinity maturation of the murine anti c-myc-peptide antibody 9E10 was analysed. Therefore Fab fragments with reversed mutations directed towards germline genes were genetically produced and characterised for their binding to the human c-myc peptide. The epitope recognized by 9E10 consists of the amino acid sequence EQKLISEEDLLRKR of which the key positions LISEXXL are very selectively recognized. The maturation of 9E10 leads to a 3300-fold higher affinity, which is achieved by a faster association as well as by a slower dissociation of the complex. For the gain in affinity formation of additional contacts to the peptide is less important than conformational and/or flexibility changes of the CDRs which are involved in binding. The exceptionally long CDR-H3 contributes essentially to the affinity maturation. The variable light domain serves thereby with its long CDR-L1 and -L3 as a binding platform for the flexible CDR-H3. Changes in specificity of 9E10 are primarily due to maturation of the variable heavy domain. Selective recognition of the key positions in the peptide is already highly pronounced in the initial stage of affinity maturation of 9E10.

Abstract:
Das Lipopolysaccharid Bindende Protein (LBP) ist ein überwiegend in der Leber synthetisiertes Akutphaseprotein. Es bindet den Zellwandbestandteil Lipopolysaccharid (LPS) Gram-negativer Bakterien und transportiert es zu zellulären Rezeptoren, wodurch das angeborene Immunsystem aktiviert wird. In dieser Arbeit wird die Regulation der LBP-Expression in Interleukin (IL)-1, IL-6 und Dexamethason (Dex) stimulierten humanen Hepatomzelllinien HuH-7 und HepG2 untersucht. Der wichtigste Stimulator ist dabei IL-6, dessen Wirkung über die Transkriptionsfaktoren (TF) Stat-3, C/EBP-beta und AP-1 vermittelt wird. Für alle 3 TF konnten aktive Bindungsstellen auf dem LBP-Promotor nachgewiesen werden. Für IL-1-Effekte die u. a. über den TF NF-kappaB vermittelt werden, konnten ebenfalls aktive Bindungsstellen nachgewiesen werden. Die Wirkung von Dex wird über Glucocorticoid Responsive Elements (GREs) vermittelt. Auf dem LBP-Promotor befinden, sich wie gezeigt werden konnte, mehrere aktive GREs, wobei einige verstärkend und einige hemmend wirken. Eine zu beobachtende Synergiewirkung von Dex und IL-6 wird durch die Aufregulation des IL-6-Rezeptors durch Dex verursacht. Die LBP-Expression kann durch TGF (Transforming Growth Factor)-beta gehemmt werden. Der TGF-beta-Signalweg über Smads ist in den Hepatomzellen aktiv, vermittelt aber nicht den TGF-beta-Hemmeffekt, sondern eine geringe stimulierende Wirkung, die bei alleiniger TGF-beta-Inkubation auftritt. Die inhibierende Wirkung von TGF-beta wird durch Gfi-1- und AP-1-Bindungsstellen vermittelt. Die Gfi-1-Bindungsstelle nimmt dabei, wie hier erstmals gezeigt werden konnte, eine herausragende Stellung ein. Die Aufklärung der LBP-Regulation und dabei besonders die Hemmung der LBP-Expression kann mittelfristig dazu beitragen, den klinischen Verlauf von inflammatorischen und infektiösen Erkrankungen zu beeinflussen und bietet daher Potenzial für neue Therapieansätze.
Lipopolysaccharide (LPS) binding protein (LBP) is an acute phase protein with the ability to bind and transfer LPS of Gram-negative bacteria. This soluble pattern recognition molecule represents an important defense principle of the host. Regulation of the hepatic acute phase response and its termination are important mechanisms for limiting systemic inflammatory activity of the host. Here were analyze the cooperation of Interleukin (IL)-1, IL-6, and Dexamethasone (Dex) at LBP expression in the hepatoma cell lines HuH-7 and Hep G2. The major inducer of LBP expression is IL-6. Within the LBP promoter numerously highly consensus binding sites such as AP-1, C/EBP-beta? and STAT3 are present, that confer transcriptional activity as shown by truncation and mutation experiments. Additionally, activate NF-kappaB sites activated by IL-1 were detected at the LBP promoter. By mutation experiments of the promoter furthermore were found differentially active glucocorticoid response elements (GREs). The promoter contains GREs enhancing the activity as well as inhibitory ones. The enhancing effect towards LBP expression by Dex was mediated by IL-6. Dex stimulated the expression of the IL-6 receptor and therefore upregulated the IL-6 pathway. Transforming Growth Factor (TGF)-beta is able to inhibit LBP expression in stimulated cells. An AP-1 binding site was identified mediating inhibitory TGF-beta effects towards LBP promoter activity. Furthermore it was shown that a growth factor independence (Gfi)-1 binding site localized near the AP-1 site is essential for mediating the TGF-beta inhibitory effect. The relevancy of the Gfi-1 site fore mediating TGF-beta effects indicates a novel mechanism for understanding inhibitory TGF-beta effects at the transcriptional level. In summary the complex regulation of LBP were elucidate which may help to eventually develop novel intervention strategies for acute phase, sepsis, and septic shock.

In der vorliegenden Arbeit wurden die Einflüsse der landwirtschaftlichen Nutzung auf die Struktur und Biomasse von Nematoden entlang eines Tiefengradientens und innerhalb zweier Vegetationsperioden an einem Ackerstandort untersucht. Die Freilandanalyse der Nematodengemeinschaft wies auf ein mit Nährstoffen angereichertes und gestörtes Ökosystem, mit einer geringen Diversität, hin. Entlang des Tiefengradienten bildeten die Nematoden Metacommunities, welche Umweltgegebenheiten wie Nahrungsquellen und abiotischen Faktoren widerspiegeln. Signifikant höhere Biomassen wurden unter Weizen im Vergleich zu Mais als Ackerfrucht beobachtet. Die Streuapplikation induzierte einen „bottom-up“ Effekt mit größeren Biomassen in den niedrigen als in den höheren trophischen Stufen. Die Nematoden Biomassen sowie die faunistischen Indizes (Channel Index, Enrichment Index) zeigten, dass der Kohlenstofffluss im Bakterienkanal des Bodens dominierte. Allerdings deckte das 13C Pulse-Labelling Experiment im Feld auf, dass der Kohlenstofffluss durch die Pilzgemeinschaft sowie die pilzfressenden Nematoden wesentlich höhere Umsatzraten aufweist. In einem 14C-Laborexperiment wurde ein vollständiges Budget des Flusses von wurzelbürtigem Kohlenstoff in das Nahrungsnetz der Nematoden bestimmt. Hierbei wiesen die pflanzenparasitären Nematoden die höchsten 14C-Gehalte innerhalb weniger Tage auf, da sie direkt an den Wurzeln fressen und über die Herbivorenkette den Nährstoffzyklus im Boden eröffnen.
The present research considered the effects of agricultural management practice on the nematode community structure and biomass in three different depths and two successive vegetation periods at an arable field site. The resource quality was manipulated by crop plant and organic amendment, to investigate the major soil carbon pathways based on roots, bacteria and fungi. The nematode community analysis pointed to a highly enriched and disturbed ecosystem with low biodiversity. Along the depth gradient the nematodes formed distinct metacommunities, reflecting resource availability and abiotic environmental factors. Wheat supported significantly greater nematode biomass than maize. The litter amendment induced bottom-up effects, with greater biomass allocation at lower than at higher trophic levels. The biomass of nematode families as well as faunal indices (Channel Index, Enrichment Index) revealed a predominance of the bacterial carbon channel in the arable soil. A 13C pulse-labelling experiment investigating the flux of root- derived C into the soil food web revealed high turnover rates in the fungal carbon pathway. This was evident for soil fungi as well as for fungal-feeding nematodes and contradicts general assumptions of a more active bacterial pathway in arable soils. A laboratory experiment with 14C isotope was used to compile a complete budget for the root-derived carbon in the nematode food web. Here plant-feeding nematodes, which feed on living plant roots, thereby opening the root C cycling into the food web, showed highest amounts of 14C allocation within a few days.
