Full textYodA of E. Coli is a protein of stress, obtained in significant quantity during the over-expression in fermentor of E. Coli strain containing a plasmid carling for the transacylase CyaC. The similarity of behaviour of YodA and CyaC in the biochemical characterization was strong and only the mass spectrometry revealed without ambiguity that the isolated protein was YodA, an endogenous protein of stress. The determination of its three-dimensional structure by diffraction of x-rays was carried out on three crystal forms obtained in absence and in the presence of cations: zinc and cadmium. These structures were refined with resolutions close to 2. 0 A, sufficient to interpret all the structural information's. YodA is made of two structural domains but does not present any relevant similarity with known structures. One can consider it as a model of a new folding, representative protein of a subfamily of sequences similar to YodA (YrpE of B subtilis and pXO1-130 of B anthracis). The number and the position of cations vary from a structure to another: zinc addition leads to three sites while cadmium complexes four sites. It is remarkable that the protein known as native also fixes a nickel, which undoubtedly is released from the affinity column used during the purification. This event illustrates the tendency of the protein to complex cations in a major site, always occupied in the structure. This site implies three histidine residues and the hypothesis is thus made that the biological functions of YodA would be the complexation and transport of cations. The validity of this hypothesis is corroborated as YodA is always associated with the stress induced by cadmium in the intracellular medium YodA also appears in the presence of hydrogen peroxide, another known agent of stress. This study may be the base for future research on YodA in the process of stress responses

Full textWork presented here is about Arabidopsis thaliana P1B-ATPases HMA2, HMA3 et HMA4, cation transporters found in different plant membranes. Cation content study of mutant plants hma2 and hma4 precised important role of HMA4 in upward translocation of Zn and Cd, and its high affinity for Cd. Cation content measure of seeds from different genotypes showed a complex effect of modulations of related genes expression levels, HMA4 overexpression leading to a seed Zn content similar the loss-of-function mutant one. These results confirm the importance of HMA4 and show the needs to adapt construction to biotechnological aims. To precise the role of residus conserved among the P1B-ATPases family, I studied the effect of the expression of a HMA4 variant for the highly conserved domain CPC. Obtained in planta results suggest an interaction with the native transporter leading to a loss of HMA4 activité. To perform a structure/function study on these transporters, L.lactis has been shown as the best candidate to produce HMA3. Due to HMA3 expression, a gain of Cd tolerance has been observed and allowed to validate three HMA3 variants, mutated in the pore or the ATP hydrolysis site, as affected in the protein activity. L.lactis membranes enriched with HMA3 or variants allowed an in vitro reconstitution in proteoliposomes and the measurement of a Cd transport activity competing with Zn and inhibited by vanadate

Full textThis thesis aimed to improve the comprehension of cadmium (Cd) absorption by maize and alpine pennycress plants in order to model better its transfer into edible and hyperaccumulating plants. Since the model used is sensitive to the characteristics of root absorption, the first objective was to develop a rigorous method for measuring those parameters. Two protocols were finalized to describe precisely Cd net influx in both root compartments (apoplast and symplast) according to Cd concentration in solution. The results highlighted, for the first time, the existence of a low-affinity transport system (LATS) that works at the same time with the high-affinity transport system (HATS). However, the kinetics parameters measured through these experimentations did not succeed in improving significantly the model: Cd uptake by maize is overestimated by 100%, and Cd uptake by alpine pennycress is under-estimated by 66%. Several factors were investigated in order to understand the reasons of this difference. We showed that the experimental conditions used to measure the kinetics parameters present characteristics that are able to modify their values significantly: the ionic composition of the solution of exposition to Cd, Cd concentration during growth, and the root structure that forms in hydroponics. This thesis suggests to choose aeroponics as controlled culture condition, and to measure the kinetics parameters in an ionic composition that is representative of the close vicinity of the root cell walls and membranes in soil. This composition remains to be investigated

Full textThe DNA double helix is a stable structure that ensures both the protection and transmission of genetic information. To access this information, a large family of multifunctional enzymes called helicases performs the separation of complementary bases of DNA. Some of these helicases in humans are associated with cancer predisposition syndromes. This is the case of Bloom syndrome (BS), a recessive genetic disease that results in an increase in genetic instability but where no phenomenon of haploinsufficiency or dominant negative is found in carriers heterozygotes. Yet we recognize that the Bloom syndrome protein (BLM) adopts a multimeric structure in vitro, but the expression among some heterozygotes of an inactive enzyme is not considered as a risk factor. To explain this paradox, we studied the structure of the BLM and find that it works as a monomer, a new result which justifies why most inactive forms does not influence the degree of cancer predisposition. On the other hand, cadmium toxicity is potentially linked to the inactivation of the BLM helicase as cells exposed to cadmium present analogies with those of patients with Bloom syndrome. Indeed, we observed in vitro, that low concentrations of cadmium reduce helicase activity by promoting its oligomerization. These studies provide new information on the molecular mechanism of the BLM helicase and emphasize its importance in maintaining genome integrity

Full textCadmium chalcogenide nanoplatelets present unique optical properties resulting from their atomically precise thickness control. They have emerged as a new promising class of nanomaterial for optoelectronic applications. The goal of this thesis is to promote new optical properties through the design and surface chemistry of nanoplatelets. Firstly, CdSe/CdSe1-xTex core/crown heterostructures were synthesized and showed bicolor emission at the scale of a single nanoplatelet. This bicolor emission results from a competition between the localization of the electron in the CdSe core driven by the conduction band offset and the direct exciton recombination in the crown favored by the high exciton binding energy. Silver doped nanoplatelets were synthesized in the second part. This doping, performed through partial cation exchange, creates a silver-related emissive state in the band gap. The amount of silver incorporated in the nanoplatelet allows a progressive control of the emission color from green to red thanks to the coexistence of the band edge and the silver related emission. The silver state is strongly bound and is located 340 meV above the bulk valence band. Finally, the modification of the surface chemistry with bromide and oleylamine reduces the surface stresses and improves the surface passivation. This leads to a shift, beyond the discrete confinement, of the emission wavelength and increases the quantum yield up to 75 %

Full textMercury cadmium telluride (HgCdTe or MCT) is a time-honored material for condensed matter physics, whose history nowadays more than fifty years long may serve as an excellent example of remarkable progress made in research on semiconductors and semimetals. The ternary compound HgCdTe implies two important aspects, which largely contributed to its undoubted success in solid-states physics.The present PhD thesis primarily aims at filling some of existing gaps in our understanding of the electronic band structure in 2D and quasi-2D heterostructures based on HgTe/HgCdTe and InAs/InSb materials, which both may be tuned into topologically insulating phase using particular structural parameter. To explore their properties, the primal experimental technique, infrared and THz magneto-spectroscopy operating in a broad of magnetic fields, is combined with complementary magneto-transport measurements. This combination of experimental methods allows us to get valuable insights into electronic states not only at the Fermi energy, but also in relatively broad vicinity.The observed magneto-optical response - due to intraband (cyclotron resonance) and interband inter-Landau level excitations - may be interpreted in the context of previous studies performed on bulk samples , quantum wells and superlattices, but also compared with theoretical expectations. Here we aim at achieving the quantitative explanation of the collected experimental data, but also further developing a reliable theoretical model. The latter includes the fine-tuning of the band structure parameters present in the established Kane model, but even more importantly, identifying additional relevant (high-order) terms and finding their particular strengths, needed to achieve quantitative agreement with our experiments. One may expect that corrections due to these additional terms will more affect the valence subbands, which are in general characterized by relatively large effective masses. Consequently, valence subbands have larger density of states compared to conduction band or, when the magnetic field is applied, rather narrow spacing (and possibly large mixing) of Landau levels

Full textThis work is devoted to the crystallographic study and the electrochemical and optical properties of cadmium phosphates and fluorophosphates. The investigation of the NaF - CdO - NaPO3 system revealed a new fluorophosphate of formula Na2CdPO4F and the polymorphism of the phosphate NaCdPO4. The crystal structures of these two phases were solved using XRD powder diffraction data, from materials obtained by a ceramic route. The characterization of the fluorophosphate Na2CdPO4F was also carried out by infrared spectroscopy, Raman scattering and Nuclear Magnetic Resonance. The electrochemical study, carried out on solid solutions of the Na2(Cd/Mn)PO4F type, revealed that the simultaneous presence of two divalent cations was beneficial for the improvement of the sodation/desodation rates. The luminescence properties of the red emitters NaCdPO4: Eu3+ and Na2CdPO4F: Eu3+ are closely correlated with the structural properties. Some formulations of NaCdPO4: Eu3+ type are of interest for application as red components in light emitting devices. The study of the LiF - CdO - NaPO3 system revealed the existence of an original fluorophosphate of formula Li4Na41Cd30P30O117F20. Its crystal structure, determined on monocrystal, is characterized by the presence of a novel entity PO3F2, where the phosphorus admits a five-fold coordination

Full textSemiconductor colloidal nanocrystals are quite attractive, because of their physical properties, such as discrete energy levels. However, devices prepared from semiconductor nanocrystals are still facing limitations due to a high environmental sensitivity of their organic shell. In order to increase their optical properties, core-shell nanocrystals have thus been synthesized. Scanning tunneling microscopy is the appropriate tool to image and probe the electronic properties of individual nanostructures and. This system can be compared to a double barrier tunnel junction, where the transport properties are governed by the transmission probability across both potential barriers (chapter 1). In order to investigate the Coulomb effect in those quantum objects by tunneling spectroscopy (this technique being described in chapter 2), the thesis has first focussed on a prototypical model: an isolated silicon dangling bond, where its charge state has been changed in a controlled manner (chapter 3). Then, PbSe/CdSe core-shell nanocrystals have been studied and a general method is described to correctly identify the electrical nature of the charge carriers in the tunneling spectra (chapter 4). In contrast to the core nanocrystals the transport through core-shell structures reveals, for a majority of nanocrystals, that the same type of charge carrier tunnel on both sides of the apparent gap. Charging peaks are also observed and allow the measurements of the charging energy in these systems. A similar transport regime is obtained for CdSe/CdS dot in rod nanocrystals (chapter 5), demonstrating the reproducibility of the characterized transport phenomena of nanoheterostructure

Full textThe development of emerging applications of CdSe nanocrystals requires a detailed understanding of the band-edge exciton fine structure and relaxation pathways. This thesis is focused on cryogenic spectroscopy of single nanocrystal with a remarkable photostability. Photoluminescence spectra as a function of temperature and under external magnetic fields provide a spectral fingerprint of the low energy sub-levels, revealing the morphology and the crystal structure of individual nanocrystals. In order to probe the entire band-edge exciton fine structure, a high resolution luminescence excitation technique has been developed. Zeeman and anisotropy-induced splittings are used to reveal the entire 8-state band-edge fine structure, enabling complete comparison with band-edge exciton models. State selective excitation allows the preparation of single quantum states. It is also used to map the hole spin relaxation pathways between the fine structure sub-levels.Charged quantum dots provide an important platform for a range of emerging quantum technologies. Double shell CdSe nanocrystals are engineered to efficiently ionize at cryogenic temperatures, resulting in trion emission with a single sharp zero-phonon line and a near-unity quantum yield. Zeeman splitting of this line enables direct determination of electron and hole g-factors. Spin relaxation is observed in high fields, enabling identification of the trion charge. Importantly, we show that spin flips are completely inhibited for Zeeman splittings below the low-energy bound for confined acoustic phonons. This charac- teristic unique to colloidal quantum dots has potential applications in single spin coherent manipulation
