Abstract:
Café X is a coffee shop managed by 3 low-vision baristas. Visitors to this cafe are also dominated by visitors with visual impairments. Due to the limited visibility of the barista and some cafe visitors, several problems were found that could cause discomfort for workers and cafe visitors. Therefore, it is necessary to adjust the cafe's operating system to support performance and provide security and comfort to the baristas and visitors who come. The data collected are data on raw materials, machinery, and equipment for cafe operational activities, data on physical facilities, layout and actual physical environment of cafes, data on the application of 5S around the cafe area, as well as data on potential accidents that may arise while around the cafe area. Then the data will processed using the method of comparison with anthropometric data and design concepts in improvement for physical facilities and cafe layouts. Standardization comparisons to improve physical environment. Radar maps to determine the application of 5S culture. Fish bones and 5W+1H to describe the causes of potential accidents in the cafe area. The suggestions given are the adjustment of the machines and equipment used, redesign of the physical facilities and layout of the cafe, the proposed design of tools in the form of tactile stickers and the application of guiding blocks as directions, improvement of the physical environment, application of 5S culture to maintain the work area and provide suggestions prevention of accidents in café area.

Abstract:
<em>This research attempts to analyze the effect of religiosity and culture on purchasing decisions in buying cafe food in Bukittinggi partially or simultaneously. The design of this study is quantitative correlational. The population is Muslim teenagers in the city of Bukittinggi. The sample of this study was Muslim teenagers in the Bukit Tinggi town who bought cafe food. Furthermore, the determination of the sample to be used as the respondent was done by purposive sampling, by taking and selecting Muslim adolescents in Bukittinggi as respondents.Based on the results of the study it can be concluded that: partially Religiosity and Culture have a positive effect on purchasing decisions in buying cafe food, while Religiosity and Culture simultaneously influence purchasing decisions in buying cafe food where the contribution of the determination coefficient (R Square) is 38.8% and the remaining 61.2% is influenced by other variables which are not included in this similarity regression or variables not examined.</em>

Abstract:
Several factors affecting the cultural growth of the society are the correct function of social interaction, daily activities in the workplace, and increasing social action according to population growth rate need to be more evident. Review of past history showed the arrival of cafe culture in the Safavid era and Shah Abbas(I) and establishment of coffee houses in Qajar and Pahlavi era with social, economic and cultural function. In the first Pahlavi period, with the arrival of a new style of cafe from the west, the social approach and architectural style of the coffee house changed to a coffee shop. The young people of the society, as the audience, chose the coffee shop as a platform for social communication. In this article, we try to explain the course of physical and cultural developments of cafes and increase the benefit of young people from temporary urban social exchange centres as a third place (home, workplace and coffee shop) by interpreting the study examples of traditional and modern cafes in the period. We will pay attention to the Pahlavi, contemporary and modern West.

Abstract:
In the Indonesian context, we did not find any previous study on internet cafes that discussed the transformation from warnet to internet cafe. Based on our study in Yogyakarta, we found a new internet cafe style that we did not find in other cities. This study offers a new understanding of analyzing internet cafes that users not only consume a computer to access the internet, but they “consume” space as well. This study aims to explore that transformation and to reveal the expansion of consumption patterns. This study emphasizes the empirical investigation to gather the data. We conducted qualitative in-depth interviews and observations from January to June 2018. We also examined several previous studies about internet cafes to expose a gap that can be extended in this study. Based on findings, we found that the transformation from warnet was a way to survive amid mobile technology. This transformation gives an implication to the new means of consumption. However, the expansion of consumption patterns can be explored from the consumption of space and digital content. Overall, digital content is the new insight to understand that internet cafes are not only a place to copy contents, but this can be seen as a symbol of empowerment to local users. Therefore, internet cafes can be seen as media-industry bottom lines to promote a culture of resistance to digital content without going to the store or going to the cinema.

Abstract:
(1) Background: Critical gaps in the U.S. healthcare system perpetuate adverse reproductive health outcomes for Black people. Grounded in reproductive justice and trauma-informed care, Breastfeeding Awareness and Empowerment (BAE) has developed a program titled BAE Cafe to directly address these gaps by providing community-based lactation and perinatal mental health support. A literature review identified key programmatic gaps, namely, access to knowledge relevant to troubleshooting breastfeeding, peer support, community support and healthcare system support, and system-level factors that impede families and communities from accessing lactation support. (2) Methods: This paper describes BAE Cafe through a group process observation and participant survey. (3) Results: The observation of groups highlighted the core elements of the BAE Cafe model: knowledge, support and mental health support in a peer driven format. Participant survey feedback was overwhelmingly positive and highlighted the critical importance of lactation support for Black women by Black women and BAE’s role in participants’ decisions to continue breastfeeding. (4) Conclusions: BAE Cafe is a replicable, scalable, peer-driven and low-barrier intervention that has the potential to improve outcomes for Black families. Additional research and investment are now needed to assess large-scale implementation to reduce disparities and address health inequity across different contexts and settings.

This study intends to discuss the direction of building a cultural navigation system that can enjoy cultural tourism contents by mixing navigation systems within the basic framework of maps. In particular, the goal is to trace the location of Gyeongseong's coffee shops and cafes appearing in modern literature, apply them to their current location, examine them in terms of cultural heritage, and further develop them into the field of cultural tourism content using them. Coffee shops and cafes are one of the most important items in examining the culture of modern Gyeongseong, and have been studied in various ways in modern literature and cultural history. The roles of coffee shops and cafes have changed somewhat in modern times, but writers and artists of modern Gyeongseong were enthusiastic about coffee shops and cafes, the symbols of modern culture at the time. Numerous coffee shops and cafes have been opened, and the writers have described them in various media or novels. In this study, the location of coffee shops and cafes that appeared in modern Gyeongseong is substituted into the current location, and through cultural navigation, the location of the past and present, information on coffee shops and cafes, and how the coffee shops or cafes are expressed in literature. By providing a system that researches and introduces information on whether or not information has been made, it is intended to examine the cultural meaning of the contemporary coffee shop and cafe culture, and to provide its cultural value by restoring it in the form of a digital map.

This research is motivated by quite a number of failures experienced by businesses to family companies in Indonesia. Many of these failures can be a reflection of poor resource management in family companies. According to Schein, individual performance in organizations cannot be understood without taking into account organizational culture, because organizations determine the degree of organizational effectiveness. From this statement, understanding the organizational culture of family companies is very important, because there are many examples of how difficult it is for a family business or company in Indonesia to maintain its survival from generation to generation. Human resources are an important aspect of business, especially in a family business. The data in this study were obtained through in-depth interviews with several owners and visitors of the milk café Karangdoro Semarang. From this research, it can be seen that the milk cafe Karangdoro Semarang has a culture in which all employees and managers are members of their own family that has been running for up to 4 generations.

This study aims to analyze the influence of several marketing mix factors such as product, price, location, promotion, and community culture on purchasing decisions for the Iced Coffee Milk product variant at the Janji Jiwa Cafe. The population of this research is the visitors of the Janji Jiwa Cafe Volume 65 Surabaya. Samples were taken using random sampling technique. Samples were taken as many as 100 loyal buyers of Iced Coffee Milk at the Janji Jiwa Cafe Volume 65 Surabaya who were met during the distribution of the questionnaire. Data obtained from the distribution of questionnaires. The data analysis technique used multiple linear regression and hypothesis testing using F-test and t-test. Based on the F-test, it is known that simultaneously product, price, location, promotion, and culture have a significant effect on purchasing decisions because the significance value is less than 0.05, which is 0.000. Based on the t-test, it is known that the product has a significant effect on purchasing decisions because the significance value is smaller than 0.05, which is 0.002. Price has a significant effect on purchasing decisions because the significance value is smaller than 0.05, which is 0.007. Location has a significant effect on purchasing decisions because the significance value is smaller than 0.05, which is 0.005. Promotion has a significant effect on purchasing decisions because the significance value is smaller than 0.05, which is 0.004. Meanwhile, culture has a significant effect on purchasing decisions because the significance value is smaller than 0.05, which is 0.001.

The aim of this research is: (1) Identifying and analyzing internal and external environment of the company that influence the marketing strategy of Suoklat Cafe. (2) Formulating the alternative marketing strategy of Suoklat Cafe that is obtained from the analysis result. (3) Formulating and setting the strategy formula that is most suitable to improve Suoklat Cafe. The data collection is taken from April to August 2016 in Suoklat Cafe located in Surabaya.Internal Analysis (IFE) results the primary factor focusing on chocolate products with the score 0,172. Meanwhile, the main weakness from this business is the less appropriate management implementation process scoring 0, 117. When the two internal factors are combined, the average total score is 2, 635.External Analysis (EFE) results main opportunity that can be used by Suoklat Cafe in order to face the competition; hanging out culture in Cafe in Surabaya by 0,216. The main threat that can be obtained from the calculation shows through the strong factor of bargaining high supplier, scoring 0, 212. Based on the two external factors that is obtained, the average score is 2, 916. IE Analysis in Suoklat Cafe positions it in quadrant V, known as maintaining and preserving strategy. SWOT analysis results strategies that can be applied: (1) Maintaining the price, increasing the variation and the product quality in order to enchance the loyality and consumers' satisfactions, (2) Expanding the market share, (3) Maintaining and improving the service quality towards the consumers (4) improving the internal managemeny of the company (5) streghten the capital through cooperations with the media, the investors, and the governmeny agencies (6) analyzing the competitors to find out tje advantages and the disadvantages, and also streghten the consumer-supplier relationship to help increase the loyality (7) Improving the effeciency and effectiveness of the employee's work, expanding the location of the parking area or cooperating with local authorities, also increasing the technology. Riset ini memiliki tujuan untuk memahami bagaimana perilaku hedonisme di kalangan mahasiswa kurang mampu FIS UNP. Riset ini di kaji menggunakan teori masyarakat konsumsi dari Jean Baudrillard. Baudrillard berasumsi bahwa masyarakat mengkonsumsi suatu barang atau jasa bukan lagi dilihat pada pemenuhan kebutuhan (nneds), tetapi lebih kepada pemenuhan hasrat (desire). Metode yang dipakai dalam riset ini adalah pendekatan kualitatif tipe deskriptif, teknik pemilihan informan snowball sampling dan jumlah informan sebanyak 10 orang. Pengumpulan data dibuat dengan teknik observasi, wawancara, dan pengumpulan dokumen dan dikaji menggunakan teknik analsis data dari Miles dan Huberman. Hasil riset ini menyatakan Perilaku Gaya Hidup Hedonisme Di Kalangan Mahasisswa Kurang Mampu FIS UNP ditunjukkan melalui kegiatan-kegiatan sebagai berikut: (1) shopping; (2) Nongkrong di cafe; (3) nonton bioskop; (4) karaoke; (5) jalan-jalan/refreshing.

Abstract:
Goal. The purpose of the article is to substantiate the feasibility of introducing a craft cafe on the basis of a business hotel, in particular, introducing the local stage of raw raw materials preparation in the technology of preparing coffee beans. Indeed, according to the study, the provision of services for organizing the preparation of a coffee drink in hotels is not satisfactory. And the quality of raw materials leaves much to be desired. Methodology. The methodological basis of this work is the work of domestic and foreign scientists in the field of hospitality, scientific periodicals, Internet resources. The methodology includes generalizing and statistical data, which reflects the feasibility of introducing a preparatory operation for processing raw materials in the technology of making coffee precisely on the territory of a hospitality establishment. Results. As a result of the research, the international and domestic experience of introducing a craft cafe in a business hotel was analyzed. It was found that there is no such practice of implementation in domestic and foreign hospitality establishments. An algorithm was developed for the introduction of a craft cafe, ways of using secondary raw materials in additional services were proposed, and the positive effect of coffee on the human body was confirmed. The results of the implementation will help improve the culture of coffee consumption and expand its range, while providing the hospitality enterprise with the opportunity to develop and make significant profits. Scientific novelty. The necessity of introducing a craft cafe with its own roast on the territory of a hospitality enterprise has been proved. The world experience in the introduction of craft cafes in Ukraine has been systematized. The assortment of additional services that contribute to increasing the competitiveness of the institution, as well as which will enable business people during business trips to enjoy high-quality coffee drinks and maintain the beauty of their body, are presented. Practical significance. The materials presented in the work can be used to improve and expand the range of additional services provided in domestic hospitality enterprises: to expand the range of coffee beans of our own production with different degrees of roasting for the manufacture of various coffee drinks, to reduce costs when purchasing batches of finished beans at auctions, while increasing their competitiveness not only in Ukraine, but also far beyond its borders.

Pekalongan is one of the tourism destinations in Indonesia. Some cultures of Pekalongan can be developed and promoted especially in the culinary field. Pekalongan's special food and beverage offerings are one of the most potential products in attracting both local and foreign tourists. Not only because of its unique name but also because of the process of making and philosophy in a product that makes Pekalongan culinary specialties. But on its journey, Pekalongan cuisine has not been able to make the reason tourists come to visit Pekalongan. Akar alang-alang which is one of the special traditional drink ingredients of Pekalongan which is expected to be better known and available at Café Jlamprang, Krapyak Lor Village so that it can introduce Pekalongan's culinary culture more easily to the community and can embrace the young generation to get to know the Pekalongan culture. Because what is currently available are some drinks that use root-alang alang ingredients that can only be found in certain regions or in the form of instant drinks that can be found in souvenirs. In this study shows that Kalang has a unique taste and presentation. Then the production system can be done simply. Kalang has a high enough potential to be sold at Cafe Jlamprang, Krapyak Lor village so that if it is able to be developed and managed properly, efforts to preserve traditional drinks should be done not only by producers, but the government must also involve all elements of society.

Humanistic reflection on video games seems like a necessity nowadays, due to the enormous popularity of digital entertainment and the infl uence it exerts over modern culture. This need for a deeper refl ection also stems from the fact that, increasingly, video games give their players various pleasures – created by spectacular visual effects, exciting plot or an opportunity to solve logical puzzles. Electronic games are progressively becoming a tool of artistic pursuits and intellectual experiments that would be impossible in analogue media. An example of such phenomenon, and a game especially worthy of refl ection, is The Stanley Parable [Galactic Cafe, 2013]. The author of the article focuses on it.

Abstract The participatory dance performance STEHBLUES was created by the duo of Ninel Çam (an artist) and Chris Schaal (a filmmaker), who travel to different cities, mark a 2m x 2m area in a public space, and display an illustrated sign that lets passersby know that they can dance with the standing-and-waiting artist if they want. Chris Schaal documents the performances through still images and videos. Moving images are live-streamed to the World Cafe in Stuttgart. In STEHBLUES, the artist stands in the midst of the marked field in a public space, waiting until a stranger is willing to slow-dance with her. As soon as someone volunteers, a relationship is created in this sphere of dance and becomes an ephemeral part of the city, of the culture, of the world. STEHBLUES travels to different cities that have invited this project to be part of their culture or to cities with which the artists feel a connection.

Marakez Café & Resto is a restaurant that serves Middle Eastern specialties in Surakarta. Carrying the Café & Resto, Marakez has several spots and rooms, one of which is a VIP room provided for guests with facilities that can be considered exclusive compared to other rooms. The shape of this VIP room is actually very supportive, with a short sofa that allows customers to enjoy their food in a lesehan style, just like Middle Eastern culture, but unfortunately the interior elements in this room do not reflect the Moroccan feel as the name suggests. Based on the description that has been explained, it is necessary to design interior textile elements to build a Moroccan atmosphere because it has been supported by the shape of the room. This design uses the design method presented by Colin Clipson. The results of this design produce complementary interior styles that are based on ideas from the Moroccan interior, namely arch, zellij and mashrabiya. This design also combines two techniques in surface design, namely digital printing and embroidery, the product is an interior complement in the form of sofa upholstery, curtains and pillowcases.

The café for this community is a project to facilitate the activities of PKK women in Palmerah District, Jakarta. The majority of these communities are local residents who are also mostly Betawi ethnic, therefore, this design requires an affordable design, in the sense of a design that can represent local culture with the application of Betawi architecture that can meet limited funding. As is known, Betawi architecture is architecture that is rich in detail ornamentation and made of wood. The result of this design is a 'café' design that uses the most important Betawi ornament with a frame structure and iron construction.

The article is devoted to theoretical issues of success of confectionery business. It examines the factors affecting the process of economic activity of the confectionery enterprise. Sweets are a source of taste pleasure and positive emotions and for most consumers become a vital product. In the modern world a tea culture with sweets is widely developed, therefore confectionery business will always be quite promising. The business "with a sweet taste" has a lot of manifestations, but at the present time the tendency of development of such enterprises as cafe-confectioneries is obvious. Cafe-confectionery is a public catering enterprise that specializes in selling with consumption in its own accommodation a wide range of different types of confectionery products. The authors note, that in order to meet the client's expectations, the confectionery company should be able to navigate the market, know the wishes of consumers, effectively organize their production and serve the guests so that they return again. Modern confectionery business is riddled with service subtleties, special importance is the identification of the needs of its client. The need for sweets for modern people is becoming so natural that many people do not see their daily meals without confectionery. Each person satisfies the need for sweet, based on their preferences, as well as financial opportunities. There are many success factors; complex interaction of these factors among themselves will ensure a high demand for products, and hence the success of the café-confectionery. The article highlights groups of factors: organization of production, customer service, marketing and management. The authors analyze each group in detail, and gives an appropriate description of each success factor of the confectionery business.

This study aims to analyze the supporting and inhibiting factors for the development of Oesapa Beach attractions in Kupang City. This type of research is qualitative research using qualitative descriptive research methods. Data sources consist of primary data obtained through observation and interviews; and secondary data sources come from records, interview results and documents from relevant agencies in the study. The results of the study show that the factors that support the development of Oesapa Beach attractions are: (1) Tourist attraction, is anything that has a unique, easy, and value in the form of natural diversity, culture, and man-made results that are targeted or tourist visits; (2) Visitors, are someone who visits a tourist area with the aim of having fun and spending less time, energy and money in less than 24 hours. While the inhibiting factors for the development of Oesapa Beach attractions are: (1) Accessibility, is the degree of ease achieved by a person towards an object, service or environment; (2) Promotion is an effort to offer products or services with the aim of attracting consumers to buy or consume them; (3) Human Resources (HR), namely the workforce involved in tourism is still inadequate and professional; and (4) Funds, is one of the benchmarks for the progress of the development of sustainable tourism objects. Oesapa Beach has a tourist attraction in the form of beautiful and natural scenery, lopo-lopo lined on the beach, and lined with cafes with colorful tents and unique and different cafe designs. The article is devoted to the concept of public space and its functionality during the study of coffee shops in Novosibirsk. The necessity for public space per person is an urgent topic, since every person somehow finds himself inside this space and assumes a special role in society. Semiotic, theoretical and empirical research methods were used in the study of the topic. Important aspects in the formation of modern urban public spaces are their functionality and complexity, which determines the components of this environment. Public spaces fulfill the task of broadcasting urban culture from generation to generation and have the ability to adapt to the rapidly changing needs of residents. An analysis of the gender, age and psychological characteristics of coffee shop visitors makes it possible to understand the reasons for visiting coffee shops, as well as the relevance of this public space in general. The article analyzes the role of tables in the social practices of society and the age composition of visitors to the cafe of the city. As a result of the study, it was found that furniture inside public spaces affects not only the aesthetic component, but also the change in social relationships between people.

Under informed consent, subject with habit of tobacco and shisha smoker were included. Sociodemographic data (age, gender, duration, frequency of smoking), salivary flow rate and oral mucosa changes were documented. Eighteen subjects were recruited into this study. Most of shisha smoker was also tobacco smoker. Shisha was more practiced by male at age (15-24 years old). The oral mucosa changes such as keratosis, melanosis, leukoedema, coated tongue, gingivitis and xerostomia were found on subject with habit of tobacco smoking habit only or both shisha and tobacco smoking. Salah satu alasan masyarakat lebih memilih tempat tersebut dibanding perpustakaan adalah desain interior yang menarik dan aspek kenyamananya. Perpustakaan diharapkan mampu menyesuaikan diri dengan tren dan budaya masyarakat saat ini untuk menarik kembali minat pemustakanya. Penelitian ini bertujuan untuk menjelaskan konsep desain interior perpustakaan baik secara teoritis maupun praktis berdasarkan kajian literatur. Hasil penelitian menunjukkan bahwa desain interior merupakan bagian penting yang harus diperhatikan pengelola perpustakaan. Desain interior selain untuk menarik minat kunjung pemustaka juga dapat meningkatkan minat baca, membentuk citra positif perpustakaan, dan menumbuhkan kepuasan bagi pemustaka. On the other hand, currently in various shopping centers such as malls and tourist attractions have been established many public space with the concept of a mini library. One reason people prefer that place compared to the library is the attractive interior design and comfort aspects. Libraries are expected to adapt to the current trends and culture of society to reclaim the interests of its librarians. This study aims to explain the concept of library interior design both theoretically and practically based on literature review. The results showed that interior design is an important part that must be considered by library managers. Interior design in addition to attract visitors visit can also increase interest in reading, forming a positive image of the library, and foster satisfaction for pemustaka. In addition to considering the aesthetic aspect, the interior design of a library must still pay attention to its functional aspects. Post-Modern Space concept innovation with additional cafe and lounge can be applied in modern libraries while taking into account the layout, color, lighting, air circulation and sound system.

Introduction: the article deals with the essence of event-marketing as a popular tool for promotion in the market, the place of event-marketing in the complex of marketing communications is determined. A study on the development of the Park of culture and recreation “Olympic” based on the use of event-marketing tools is carried out. The results of the study of existing tools to promote the Park are given, the motives and preferences of potential guests of the Park on the basis of a survey are identified, recommendations for its development are given. Materials and methods: the article uses the publications of well-known scientists, methods of abstract-logical, systematic, economic and statistical, comparative analysis, graphic, methods of sociological and marketing research (in the survey of consumers of the Park of culture and recreation “Olympic”). The results of the study: the analysis of existing ways to promote the services of the Park of culture and recreation “Olympic” showed that the Park is not enough and not often held various event-events. In addition, the events are not diverse, which indicates the lack of use of event-marketing tools in the Park. In this regard, a variety of events was offered, within the framework of the implementation of such areas as sport events, exhibitions and art creativity, leisure activities for young people, romantic events, catering, opening and restoration of the existing Playground, modernization of the roller rink and art cafe, opening of the Park of extreme species, improvement of the Park and the opening of the school with clubs for the implementation of creative directions. Discussion and conclusion: Improvement program Park development using the tools of event marketing will contribute to the implementation of the Federal program “Formation of a comfortable urban environment for 2018–2022.g.”, to increase the customer traffic, to improve the visitor traffic, average check, increase the quality of life of the population. For organizations in the sphere of communication services, Event-marketing is the most effective direction of direct communication and is one of the most popular and progressively developing tools.

Nadine Fresco in her research on exiled Holocaust survivors uses the term diaspora des cendres (1981) to depict the status of Jewish migrants whose lives are forever marked by their tragic experience as well as a conviction that “the[ir] place of origins has gone up in ashes” (Hirsch 243). As a result, Jewish migrants and their children have frequently resorted to storytelling treated as a means of transferring their memories, postmemories and their condition of exile from the destroyed Eastern Europe into the New World. Since “[l]iterature of Australians of Polish-Jewish descent holds a special place in Australian culture” (Kwapisz Williams 125), the aim of this paper is to look at selected texts by one of the greatest Jewish-Australian storytellers of our time: Arnold Zable and analyse them according to the paradigm of an exiled flâneur whose life concentrates on wandering the world, sitting in a Melbourne café, invoking afterimages of the lost homeland as well as positioning one’s status on a map of contemporary Jewish migrants. The analyses of Zable’s Jewels and Ashes (1991) and Cafe Scheherazade (2001) would locate Zable as a memoirist as well as his fi ctional characters within the Australian community of migrants who are immersed in discussing their un/belonging and up/rootedness. The analysis also comprises discussions on mapping the past within the context of the new territory and the value of storytelling.

According to the new survey of Master Card (titled Consumer Purchasing Priorities) contained in Suara Pembaharuan 2016, 805 Indonesian’s people prefer to eat on the fast food outlet, in the second rank are food court with 615, and in the third rank are middle class restaurant or cafe with 225. Meanwhile, there is 1% Indonesian’s people who choose to come to restaurant for fine dining. Pizza Hut is the first fast food in Indonesia which serve pizza as a main dish. Consumer’s behaviour is the most to be considered remember all of that company did constituted by the market demands. Purpose of this research are to (1) know the characteristic of Pizza Hut consumer in West Jakarta; (2) know the influence of brand image to decision purchase of Pizza Hut in West Jakarta; (3) know the influence of product quality to decision purchase of Pizza Hut in West Jakarta; (4) know the influence of prices to decision purchase of Pizza Hut in West Jakarta; and (5) know the influence of product diferentiation to purhase decision of Pizza Hut in West Jakarta. Based on result in this research, characteristic of Pizza Hut consumer are consumer with the age range of the 20-30 years, women, with employment status is student or college student, know about pizza hut product from banner or commercial advertisement and buy pizza hut just one time in a month. Result of regression multiple linear analysis are brand image, product quality and product diferentiation are not significant influence to purhase decision of pizza hut in trust rate of 95%. Although of that, prices is significant influence to purchase decision of Pizza Hut product in West Jakarta.

The hybridity of Muslim youth with popular culture has had an influence on the da'wah packaging strategy carried out by many da'wah communities in Indonesia, including the Teras Da'wah Community in Yogyakarta, Indonesia. The packaging of da'wah with a more popular, fun, and modern cover has made Teras Da'wah as one of da'wah communities with the most wanted by many younger Muslims in Indonesia. This article aims to describe how the branding strategy is run by the Teras Da'wah Community using a qualitative research approach, namely through ethnographic and netnographic studies for six (6) months in the Teras Da'wah Community. The results of the study reveal that the appreciation of popular culture among young people today has a significant role in 'attracting' these millennials to recite the Koran in a relaxed but serious manner at Teras Da'wah. The form of this appreciation is packaged from the selection of an attractive logo, the interior of the room that resembles a cafe, the study content packaged in slang and the clustering of the study program according to the level of understanding of the members. The consequence of this strategy is the increasingly hybrid identity of Muslim youth in Indonesia due to the intersection between Islamic values and popular culture that has long been attached to them.Hibriditas anak-anak muda muslim dengan budaya populer telah membawa pengaruh pada strategi pengemasan dakwah yang dilakukan oleh banyak komunitas dakwah di Indonesia. Salah satu diantaranya yang telah menerapkan strategi tersebut adalah Komunitas Teras Dakwah di Yogyakarta, Indonesia. Pengemasan dakwah dengan cover yang lebih ngepop, fun, dan modern telah menjadikan Teras Dakwah sebagai komunitas dakwah yang diminati oleh banyak anak-anak muda muslim di Indonesia. Artikel ini bertujuan untuk mendeskripsikan bagaimana strategi branding yang dilakukan oleh Komunitas Teras Dakwah dengan menggunakan pendekatan penelitian kualitatif, yakni melalui studi etnografi dan netnografi selama enam (6) bulan di Komunitas Teras Dakwah. Hasil penelitian mengungkapkan bahwa apresiasi Teras Dakwah terhadap budaya populer di kalangan anak muda saat ini memiliki peran signifikan dalam ‘menarik’ milenial ini untuk mengaji dengan santai tapi serius di Teras Dakwah. Wujud apresiasi ini dikemas dari pemilihan logo yang menarik, interior ruangan yang menyerupai kafe, konten kajian yang dikemas dengan bahasa gaul dan klusterisasi program kajian sesuai dengan tingkat pemahaman anggota. Konsekuensi dari strategi ini adalah semakin hibridnya identitas anak-anak muslim di Indonesia akibat persinggungan antara nilai-nilai Islam dengan budaya populer yang sudah lama melekat pada diri mereka.

Topicality.The popularity of coffee industry is high. This business is actively developing in Ukraine. A coffee shop is an establishment where the high quality of the product, as well as the development of coffee consumption culture are always in the first place. An important component is the commercial activity on which the coffee industry is built. This is an indicator of the right culture: more often Ukrainians prefer coffee in some cafe or restaurant, or even takeaway, instead of home or office coffee. However, COVID 19 pandemic affected the decrease in visits by Ukrainians to coffee shops, cafes, restaurants and other catering establishments. The main share of coffee consumption is an imported product. Ukrainian production is formed by manufacturers who import coffee beans as a raw material, and in Ukraine they roast, receive, package, produce an article without caffeine, and sell it under their own brand. The fiercest competition is typical in low segments of coffee drinks and grains. The aim of the study and its methods. The purpose of this article is to analyse the current state of Ukrainian coffee industry. When conducting the research, general scientific methods have been applied: scientific and theoretical elaborations of blighty and foreign scientists, informational materials of statistical and reference publications, analysis, synthesis, generalisation, comparison. Results. The article analyses the current state of coffee industry in Ukraine. The greatest producers of coffee and the most popular brands of grain and ground coffee in Ukraine have been characterised. The structure of coffee consumption in Ukraine by types and in quantitative indicators has been analysed. It has been noted that Ukrainian coffee industry is represented by a large number of small operators who open their mobile coffee shops in places with high traffic. This is due to small initial investments, a short payback period, and the possibility to change the location. Since the tendencies of the world coffee market have a significant impact on coffee industry in Ukraine, the demand is also growing steadily. The amount of coffee import in Ukraine exceeded the volume of exports. In 2020, the scope of export reached 15,0 million dollars, USA, while in 2021 it increased to 15,9 million dollars, USA. The amount of import increased from 251,3 million dollars, USA in 2020 to 266,7 million dollars, USA in 2021. The import of coffee beans is aimed at meeting domestic consumers’ needs. Ukraine is completely dependent on import, due to the non-compliance with natural and climatic conditions, because growing coffee on the territory of Ukraine is impossible. Conclusions and discussion. So, in the course of this study, it has been established that coffee industry in Ukraine is developing more actively and even creating competition. A significant place in the restaurant industry is occupied by coffee shops, which increase competition and increase the quality of provided services. Modern coffee shops are restaurant establishments where coffee is made at a new level, with an individual approach to each guest. The current types of restaurant industry establishments are multi-format, so the creation of a national coffee shop is one of the advertising means of our country. Cafes with 3–4 seats are gaining popularity in Kyiv. On the other hand, recently, popular “to go” coffee shops are losing their market key positions. The market of coffee industry is going to grow, and become saturated after the end of the full-scale war. The infrastructure is going to be restored, and the restaurant industry has all the chances to be developed in the eastern region of Ukraine.

The list of topics includes: “Family”, “About Me”, “Study”, “Workday”, “Away”, “Rest”, “At the doctor”, “In a cafe”, and “Supermarkets, purchases”. The authors draw attention to controversial points in the educational material, requiring special commentary from the Russian language teacher. The article analyzes both positive and negative stereotypes, which are the basis for creating the primary image of a representative of Russian culture. The image of a Russian person represented in first-year textbooks is not ideal, both advantages and vices are noted. The authors also point out that some of the material in the Chinese textbooks no longer corresponds to today’s Russian reality and needs to be corrected. A comparative method is used in the analysis of the material. This turmoil mobilized everyone to overcome it, including Café Rumi worshipers. A spiritual café that offers spiritual needs that are being sought by the urban community. Through the application of routine zikir rituals, it can be a solution to this problem This study aims to find out whether the typical ritual remembrance activities at Café Rumi can have a significant impact and influence on the sobriety of the pilgrims who regularly attend zikir activities that are there. This study uses qualitative methods with descriptive analysis approach through direct observation, in-depth interviews and documentation process. After that, it is done with the data analysis process, compared, described later until the final conclusions are obtained. The target of being a researcher is the Cafe Rumi congregation, most of whom are city residents. This study yields conclusions in the form of the meaning of remembrance in Café Rumi is not merely a ritual but rather a zikir in an act of sharing with others. And from the reading side there is a modification of the addition and subtraction of the number of zikir. Zikir activities are carried out in three forms, namely spiritual meditation with zikir together led by kiyai, culture through sholawatan and Sufi dances accompanied by hadrah music, and social activities in the form of sharing food to people in need at a crossroads. The ritual of the typical Sufi ritual of remembrance is also able to increase the sobriety for worshipers at Café Rumi.

Abstract CD34+Thy-1+Lin− cells are enriched for primitive hematopoietic progenitor cells (PHP), as defined by the cobblestone area-forming cell (CAFC) assay, and for bone marrow (BM) repopulating hematopoietic stem cells (HSC), as defined by the in vivo SCID-hu bone assay. We evaluated the effects of different cytokine combinations on BM-derived PKH26-labeled CD34+Thy-1+Lin− cells in 6-day stroma-free cultures. Nearly all (>95%) of the CD34+Thy-1+Lin− cells divided by day 6 when cultured in thrombopoietin (TPO), c-kit ligand (KL), and flk2/flt3 ligand (FL). The resulting CD34hiPKHlo (postdivision) cell population retained a high CAFC frequency, a mean 3.2-fold increase of CAFC numbers, as well as a capacity for in vivo marrow repopulation similar to freshly isolated CD34+Thy-1+Lin− cells. Initial cell division of the majority of cells occurred between day 2 and day 4, with minimal loss of CD34 and Thy-1 expression. In contrast, cultures containing interleukin-3 (IL-3), IL-6, and leukemia inhibitory factor contained a mean of 75% of undivided cells at day 6. These CD34hi PKHhi cells retained a high frequency of CAFC, whereas the small population of CD34hiPKHlo postdivision cells contained a decreased frequency of CAFC. These data suggest that use of a combination of TPO, KL, and FL for short-term culture of CD34+Thy-1+Lin− cells increases the number of postdivision PHP, measured as CAFC, while preserving the capacity for in vivo engraftment.

Abstract We have previously shown that the sequential addition of a hypomethylating agent, 5-aza-2′-deoxyctidine (5azaD) and a histone deacetylase inhibitor, trichostatin A (TSA) is capable of changing the fate of adult bone marrow CD34+ cells (Milhem M et al Blood 2004). We have now studied whether these drugs could alter the behavior of dividing CD34+CD90+ cord blood (CB) cells. The 5azaD/TSA treated cultures yielded 10 times greater numbers of CD34+CD90+ cells as compared to the cultures containing cytokines alone after 9 days of culture. The 5azaD/TSA treated cultures contained 2 fold greater numbers of colony forming cells (CFC) and 14 fold greater numbers of long-term (5wk) cobblestone area forming cells (CAFC) in comparison to culture containing cytokines alone. Although the CFC/CAFC plating efficiency of cells in cultures exposed to cytokines alone declined during the time of incubation, the cloning efficiency of cells exposed to 5azaD/TSA was equivalent to that of primary CD34+ cells. In order to determine the effects of cell division on the behavior of CD34+CD90+ cells in the 5azaD/TSA treated cultures, we utilized the cytoplasmic dye, CFSE. All of the CD34+CD90+ cells within the 5azaD/TSA pre-treated cultures divided at least once after 9 days of culture. 60% of the 5azaD/TSA treated CD34+CD90+ cells divided 5 times or more while 40% divided 1–4 times. The CD34+CD90+ cells lacking 5azaD/TSA pre-treatment underwent more cell divisions (90%, 5 or more divisions). The CD34+CD90+ cells pre-treated with 5azaD/TSA which had undergone 1-2 cell divisions had 11 fold greater numbers of CFU-Mix and 9 fold greater number of CAFC as compared to CD34+CD90+ cell population cultured in presence of cytokines alone. Furthermore CD34+CD90+ cells having 5 and more divisions had 4 fold more CFU-mix and 6.5 fold more CAFC in comparison to cells lacking 5azaD/TSA exposure. The CD34+CD90+ cells experiencing 1–4 divisions had 60 fold greater number of CFU-mix and 54 fold more CAFC in contrast to culture treated with cytokines alone. The in vivo SCID repopulating potential of CD34+CD90+ cells generated in presence or absence of 5azaD/TSA was then evaluated. When 5x104 CD34+CD90+ cells having undergone 1-2 cell divisions were re-isolated from 5azaD/TSA pre-treated cultures, all mice contained human hematopoietic cells. In addition, 1 of 3 mice transplanted with CD34+CD90+ cells (5x104) having undergone 3 and more cell divisions isolated from cultures pre-treated with 5azaD/TSA also displayed human hematopoietic engraftment. Furthermore 1 of 3 mice transplanted with equal numbers of the 5azaD/TSA pre-treated CD34+CD90+ cells having undergone 5 and more cell divisions also had evidence of human multilineage hematopoietic engraftment. By contrast, an equivalent number of CD34+CD90+ cells which had undergone more than 3 or more than 5 cell divisions from the cultures containing cytokines alone were incapable of engrafting NOD/SCID mice. These data suggest that the sequential addition of 5azaD and TSA ex vivo is not only capable of expanding the numbers of CD34+CD90+ cells and assayable progenitor cells but also capable of preserving their SCID repopulating potential. We conclude that 5azaD/TSA treatment of CD34+CD90+ cells results in their retention of the cellular program required to maintain their marrow repopulating potential despite their undergoing multiple cell divisions.

The peculiarities of eating behavior of students of Maykop State Technological University and the Adygh State University of the Republic of Adygea have been investigated. Peculiarities of students’ perception of the quality of services of public catering enterprises, an assessment of the structure of expenses for the acquisition of various dishes at catering enterprises, an assessment of the impact of the social status of students on the structure of consumption of products of public catering enterprises, etc. have been considered. Particular attention is paid to identifying nutritional problems existing among students in the region. On the basis of the research the directions for improving healthy nutrition of students have been identified. A university system of healthy nutrition for students has been developed, based on the philosophy of youth taste and preferences: food behavior and reasonable quality: balance in basic substances, with specified beneficial properties, a recommended diet consisting only of healthy food products. The developed system of healthy nutrition will ensure the formation and development of food culture: aesthetic taste for food and its consumption in order to reorient young people from fast and unhealthy nutrition to a healthy diet, using «high cuisine» techniques (careful preparation and careful presentation of dishes / products designed to form a sustainable high motivation for healthy eating, which persists throughout the rest of life, the quality of which can be predicted with predominant indicators. Resultados: identificou-se a necessidade de ampliação do conhecimento sobre os cuidados com paciente em hemodiálise. Existe imprecisão na formulação de conceitos, características do tratamento e necessidade de ampliar conhecimento sobre a doença renal. Além da reduzida quantidade de profissionais que facilitem a aprendizagem e realizem orientações sobre o autocuidado no processo de adaptação com o tratamento hemodialítico. Conclusões: portanto, defende-se que os profissionais devam exercer a consciência crítica para o processo contínuo de formação para promover o cuidado apoiado.

Abstract Parathyroid hormone (PTH) induces activation and increases the number of osteoblasts. An increase the stem cells number (Lin-Sca1+cKit+) was observed in mice after 4 weeks of PTH treatment, that suggest osteoblasts participation in regulation of hematopoiesis (Calvi, et al., 2003, Nature, 425, 841). Long-term bone marrow culture (LTBMC) was used for the study of PTH influence on hematopoietic progenitors of different stages of maturation. Rat PTH (1–34) (final concentration, 10−7M) was supported during whole culture period. Cell number, colony-forming units in culture (CFU-GM, CFU-C-21 days) and cobblestone area forming cells (CAFC) were measured during 10 weeks. The number of mature cells in culture did not changed during PTH treatment. The number of studied progenitors did not changed significantly during 3 and 6 weeks of PTH treatment. The number of CFU-GM, CFU-C-21 days and CAFC 14 days, CAFC 21 days, CAFC 28 days increased 7 –10 fold in suspension fraction of LTBMC after 10 weeks of PTH treatment. Table 1. To evaluate the possibility of stem cells expansion by using of PTH treated adherent cell layers (ACL) of LTBMC, the PTH-treated for 4 and 8 weeks cultures, were irradiated with 40 Gy and seeded with 2 × 106 bone marrow cells depleted of adherent cells. Following 24 hours the number of survived CAFC 8 – 28 was analyzed. On PTH-treated ACL the number of CAFC 21 increased 2,7 ± 0,4 fold as compared with fresh bone marrow or culture with non-treated ACLs. Abstract:
Waste biomass coming from a local coffee company, which supplied burnt ground coffee after an incorrect roasting process, was employed as a starting material in the composting plant of the Experimental Station of the University of Naples Federico II at Castel Volturno (CE). The direct molecular characterization of compost using 13C-NMR spectra, which was acquired through cross-polarization magic-angle spinning, showed a hydrophobicity index of 2.7% and an alkyl/hydroxyalkyl index of 0.7%. Compost samples that were collected during the early “active thermophilic phase” (when the composting temperature was 63 °C) were analyzed for the prokaryotic community composition and activities. Two complementary approaches, i.e., genomic and predictive metabolic analysis of the 16S rRNA V3–V4 amplicon and culture-dependent analysis, were combined to identify the main microbial factors that characterized the composting process. The whole microbial community was dominated by Firmicutes. The predictive analysis of the metabolic functionality of the community highlighted the potential degradation of peptidoglycan and the ability of metal chelation, with both functions being extremely useful for the revitalization and fertilization of agricultural soils. Finally, three biotechnologically relevant Firmicutes members, i.e., Geobacillus thermodenitrificans subsp. calidus, Aeribacillus pallidus, and Ureibacillus terrenus (strains CAF1, CAF2, and CAF5, respectively) were isolated from the “active thermophilic phase” of the coffee composting. All strains were thermophiles growing at the optimal temperature of 60 °C. Our findings contribute to the current knowledge on thermophilic composting microbiology and valorize burnt ground coffee as waste material with biotechnological potentialities.

We have recently observed that the number and quality of mobilized peripheral blood stem cells (PBSC) was low in patients having received multiple rounds of chemotherapy. Moreover, grafts with low numbers of HSC and poor HSC quality had a high probability to cause graft failure upon their autologous infusion. Because ex vivo culture of stem cells has been suggested to present an attractive tool to improve hematological recovery or reduce graft size, we have studied the possibility that such propagation may affect stem cell quality. In order to do so, we have assessed the recovery of different stem cell subsets in CD34+ PBSC after a 7-day serum-free liquid culture using CAFC and LTC-CFC assays. A numerical expansion of stem cell subsets was observed in the presence of interleukin-3 (IL-3), stem cell factor, and IL-6, while stroma-contact, stromal soluble factors, or combined addition of FLT3-ligand and thrombopoietin improved this parameter. In contrast, ex vivo culture severely reduced the ability of the graft to produce progenitors in LTC while stromal soluble factors partly abrogated this quality loss. The best conservation of graft quality was observed when the PBSC were cultured in stroma-contact. These data suggest that ex vivo propagation of PBSC may allow numerical expansion of various stem cell subsets, however, at the expense of their quality. In addition, cytokine-driven PBSC cultures require stroma for optimal maintenance of graft quality.

In contrast, ex vivo culture severely reduced the ability of the graft to produce progenitors in LTC while stromal soluble factors partly abrogated this quality loss. The best conservation of graft quality was observed when the PBSC were cultured in stroma-contact. These data suggest that ex vivo propagation of PBSC may allow numerical expansion of various stem cell subsets, however, at the expense of their quality. In addition, cytokine-driven PBSC cultures require stroma for optimal maintenance of graft quality.

Abstract:
This study was carried out to determine the effect of β-mercaptoethanol (bME; at 50 μM) during a brief co-culture and the additional culture of inseminated oocytes on sperm penetration of porcine IVM oocytes. Cumulus-oocyte complexes (COCs) collected from 3–∼6-mm follicles were cultured in modified NCSU-37 supplemented with bME, eCG, hCG, and dibutyryl cAMP for 20 h and then in the same medium without eCG, hCG, and dibutyryl cAMP for additional 24 h. After IVM, the oocytes were denuded with 0.1% hyarulonidase, randomly divided into three groups (as described below) and fertilized in vitro. Treatment 1 (T1, control): Oocytes were co-cultured with fresh spermatozoa for 10 min in modified M199 medium (mM199) supplemented with 5 mM caffeine (+caff) in the absence of bME (bME-), then washed gently with caffeine-free mM199, and cultured for further 9 h in mM199; T2: co-culture for 10 min in mM199+caff bME− and additional culture in mM199 in the presence of bME (bME+); and T3: co-culture for 10 min in mM199+caff bME+ and additonal culture in mM199 bME+. At 9 h after insemination, oocytes were fixed and stained for the evaluation of sperm penetration. Results from four replicates were analyzed by ANOVA and Duncan's multiple range test. Sperm penetration rate was lower in T3 (79.8% in 92 oocytes) compared with T1 (95.3% in 84 oocytes) and T2 (93.3% in 93 oocytes). Percentage of eggs with male and female pronuclei in penetrated oocytes was higher in T1 (89.6%) than in T2 (56.4%) and T3 (57.2%). Monospermy rate was lower in T1 (48.9%) than in T2 (59.5%) and T3 (64.3%). These results demonstrated that, although the presence of β-mercaptoethanol during co-culture and the additional culture reduced the fertilization rate, the presence during the additional culture period affected the process of fertilization and increased the incidence of monospermy at 9 h after insemination. In conclusion, the progression of sperm penetration may be controlled by utilization of β-mercaptoethanol in porcine IVF system. This work was supported by JSPS-KAKEN (C16580230).

Abstract We here investigated whether receptor tyrosine kinase signaling (ckit, flt3) was sufficient to maintain hematopoietic stem cells (HSC) in culture, or whether, like in embryonic stem cells, signaling through gp130 is also required. Sorted CD34+ AC133+ (CD33/ CD38/ CD71)- cells from human umbilical cord blood (CB) were cultured in the presence of combinations of ckit-ligand stem cell factor (SCF), flt3-ligand (FL) and different stimulators of gp130 (IL-6, IL-11, LIF, OSM, CT1 and CNTF). Of these cytokines, SCF stimulated cell division by itself. Gp130-stimulating cytokines did not give any additional cell division activity. When SCF was included in the cytokine cocktail, progenitor levels (CFC, CAFCw2, CAFCw6) was very similar for all combinations of cytokines tested after this initital 6-day culture. To study more immature progenitors, cells were harvested after 6 days of serum-free culture and cultured for another 6 weeks on FBMD-1 stromal cells (LTC-CAFC). These experiments showed that in initial 6 day cultures with FL or gp130-stimulators alone, very little hematopoietic activity remained. SCF maintained CFC, and LTC-CAFCw2 and LTC-CAFCw6 at about half the original level. However, combinations of SCF with OSM consistently increased the number of LTC-CAFCw2 and w6 about two-fold. This finding suggests that very immature hematopoietic progenitors were at least maintained in serum-free cultures. To study the most immatore cells, we performed in transplantations into sublethally irradiated NOD/SCID mice. When CB cells were cultured with SCF alone, NOD/SCID-repopulating cell (CRU) frequency (1 in 900) dropped to about 20% in 6 days (1 in 5500). Similarly, cultures without growth factors or with FL or OSM showed a decrease to less than 10% of the original CRU number. In contrast, the combination of SCF and OSM maintained CRU at about half the original level (1 in 1900). This study shows that signals through ckit and gp130 synergize to maintain CRU level. However, to achieve expansion of human NOD/SCID-repopulating cell numbers, additional signaling pathways need to be stimulated.

This study reports the incidence of bacteraemia following 106 consecutive bedside percutaneous tracheostomies. Post-tracheostomy blood culture results were compared with other blood cultures from the same population. The incidence of positive post-tracheostomy blood cultures was 10.4% (11/106), compared with 6.6% (7/106) for other blood cultures (odds ratio 1.64, 95% confidence interval 0.61-4.40, P=0.46). Staphylococcus epidermidis was the most common organism cultured, 7/106 (6.6%) of post-tracheostomy cultures, compared with 3/106 (2.8%) for other cultures (odds ratio 2.43, 95% confidence interval 0.61-9.65, P=0.33). The other four post-tracheostomy cultures grew an organism cultured from that patient's tracheal secretions. Seventy-four patients were receiving antibiotics at the time of tracheostomy, of these 7 (9.5%) had positive blood cultures, a similar incidence (4 of 32, 12.5%) to those not receiving antibiotics (odds ratio 0.73, 95% confidence interval 0.20-2.70, P=0.90). We conclude bacteraemia is a common complication of percutaneous tracheostomy; the causative organisms come from the patients’ trachea or skin.

Abstract Long-term cytokine-supplemented or stromal cocultures of human CD34+ cells, particularly from cord blood (CB), show expansion of hematopoietic progenitors and stem cells. Ultimately, however cultures decline and terminally differentiate. Despite upregulation of telomerase activity in proliferating primitive hematopoietic cells, telomere shortening has generally been reported in long-term cultures of CB, bone marrow or G-CSF mobilized peripheral blood CD34+ cells. In earlier reports, we described a long-term culture of hematopoietic stem cells on a murine OP9 bone marrow stroma cell line transfected with an adenovector expressing thrombopoietin, which allowed an extensive proliferation and self-renewal of CB CD34+ cells for 4–5 months with sustained elevation of telomerase activity and without concomitant significant telomere shortening (Blood, 2004). Here, we evaluated adult healthy donor peripheral blood (PB) CD34+ cells in the same OP9/Tpo coculture system. To determine progenitor and stem cell production, standard CFC and 2ndry cobblestone area-forming cell assays (CAFC assayed at 5 weeks on MS5 stroma) were undertaken weekly with suspension cells. In addition telomere length was measured by telomere restriction fragment (TRF) assay, and telomerase activity by TRAP assay on input CD34+ cells, and weekly on culture suspension cells. Maximum total cell, CFC and CAFC production was seen in the first 4 weeks with up to 80-fold expansion in cell count, up to 4-fold expansion in CFC and up to 13-fold expansion in CAFC. Thereafter a continuous decrease in production of cells, CFC 2ndry CAFC was observed and cultures terminated at week 8. Mean telomere length of input PB CD34+ cells was 9,5 ± 0,5 kbp. After 4 weeks in culture, telomere length remained stable (9,6 ± 0,5 kbp). In 3/6 cultures terminated, cultures showed only a slightly decrease of telomere length compared to the input population (0,55 ± 0,1 kbp loss). However, in 2/6 cultures we could demonstrate an elevation of telomeres (+ 0,3 kbp) independent of a rapid loss of telomerase activity in all cultures during the culture period. Furthermore, the elevation of telomeres did not correlate with an enhanced stem/progenitor cell activity. These data confirm earlier results of granulocyte telomere change in myeloma patients following chemotherapy and tandem transplantation, where 154/193 patients showed an expected loss of telomeres during the treatment period, whereas 39/193 patients had an unexpected elevation of telomeres. We could now show in healthy donors that this phenomenon is independent of bone marrow stress due to chemotherapeutic treatment. In conclusion, we could show that the stromal coculture system with OP9/Tpo is highly effective in stem/progenitor cell expansion not only in CB but also in PB CD34+ cells. It is the first culture system, which allows expansion of hematpoietic cells without significant telomere erosion. We furthermore describe for the first time an age-independent healthy donor population which shows telomerase-independent, significant telomere elevation. Further studies have to demonstrate, if this phenomenon is potentially linked to a higher susceptibility for cancer disease.
