Abstract:
Fatigue was studied in intact tibialis anterior muscle of anesthetized mice. The distal tendon was detached and connected to a force transducer while blood flow continued normally. The muscle was stimulated with electrodes applied directly to the muscle surface and fatigued by repeated (1 per 4 s), brief (0.4 s), maximal (100-Hz stimulation frequency) tetani. Force declined monotonically to 49 ± 5% of the initial value with a half time of 36 ± 5 s and recovered to 86 ± 4% after 4 min. Intracellular phosphate concentration ([Pi]) was measured by 31P-NMR on perchloric acid extracts of muscles. [

Current fluorescent monitors of free [Ca2+] in the sarcoplasmic reticulum (SR) of skeletal muscle cells are of limited quantitative value. They provide either a nonratio signal that is difficult to calibrate and is not specific or, in the case of Forster resonant energy transfer (FRET) biosensors, a signal of small dynamic range, which may be degraded further by imperfect targeting and interference from endogenous ligands of calsequestrin. We describe a novel tool that uses the cameleon D4cpv, which has a greater dynamic range and lower susceptibility to endogenous ligands than earlier cameleo

The colonial project of the French monarchy in the West Indies from the beginning understood an evangelizing will towards the indigenous populations. However, despite attempts to do so, the failure many missionaries is patent, as summarized with bitterness Caesar de Rochefort, attributing it to the fact that "the Indians of America are changing like Cameleons ". While they are gradually evacuated from the colonized islands, the missionary effort is devoted primarily to the servile population from Africa, to whom Catholicism is imposed. The conversion of slaves is more successful and allows to e

Abstract:
The first part of this thesis aims to develop new tools to explore mitochondrial Ca2+ dynamic in vivo by the improvement of a mitochondria targeting Cameleon probes and by creating a mitochondria targeted Channelrhodopsin. Genetically encoded calcium indicators (GECIs) allow quantitative Ca2+ measurements in different experimental models. Organelle-specific targeting signals are fused with the GECI’s sequence, achieving selective targeting to a specific organelle or cytoplasmic domain. Moreover, GECI’s coding sequences can be placed under the control of tissue specific or inducible promoters

Calcium (Ca2+) is one of the major intracellular messengers that impacts nearly every aspect of cell life. In particular, it plays essential roles in neuronal development, synaptic transmission and plasticity, as well as in the regulation of metabolic pathways and cell fate decisions. The first goal of my work was to study more in details the Golgi apparatus (GA), as an intracellular Ca2+ store. The Golgi complex may store up to 5% of the total cellular Ca2+ at higher concentrations and contains several different luminal Ca2+-binding proteins and Ca2+-release channels. The use of a specific

Research Doctorate - Doctor of Philosophy (PhD)<br>A divided response—between acceptance and rejection—currently exists among the Australian population toward people arriving in Australia from South Asia, particularly those seeking asylum. This rift has an analogue in the settler response to cameleers and other workers who arrived in Australia from across the Indian Ocean in the peri-Federation years (1890–1915). This research therefore examines the peri-Federation diaspora of South Asia to Australia to address the apparent persistence of a dissension for a period of more than one hundred year

The recognition of different microbe-associated molecular patterns triggers in the plant the activation of either an immune response or an accommodation program. In both types of responses, Ca2+ is a crucial intracellular messenger, mediating the early stages of the respective signalling pathways. In this work, we analyzed the cytosolic and nuclear Ca2+ changes activated by a set of chitin-related oligomers in different genetic backgrounds of Lotus japonicus roots by using specifically targeted aequorin-based Ca2+ reporters. By means of pharmacological and genetic approaches, we dissected the
