The adenovirus codes for a protease which is essential for virion infectivity. This protease requires the presence of a peptide cofactor in order to develop optimal activity. This peptide, GVQSLBCRRRCF, originates from the C-terminal of a viral protein, pVI, and some evidence regarding its specificity came from observations showing that neither of the peptides GVQSLKRRRAF or KRRRCF was able to activate the protease, indicating that both the cysteine and the N-terminal were important in the activation process. The adenovirus type 2 origin of DNA replication is located within the terminal 51 bp of the viral genome and contains three recognisable domains: the minimal origin or 'core' and binding sites for the cellular transcription factors Nuclear Factor I and Nuclear Factor III. RNA splicing is a process where introns are removed and exons are joined together. Human adenovirus type 2 pre-mRNAs undergoes intensive alternative splicing and produce more than 40 differently spliced mRNAs.  This thesis work is focused on the identification of new splice sites in adenovirus. By virtue of Illumina mRNA sequencing technology we have identified 255 splice sites. Splice site analysis of the introns revealed the presence of three types of splice sites GT-AG (61.2%), GC-AG (25.9%) and AT-AC (12.9%). Among 255 splice sites, 224 were new. Canine parvovirus type 2 (CPV-2), the aetiological agent of haemorrhagic enteritis in dogs emerged in 1978 worldwide. In the mid 1980’s, the original CPV-2 had evolved and was completely replaced by 2 variants, CPV 2a and 2b. In 2000, a new variant of CPV (CPV-2c) was detected in Italy and now circulates in other countries. Haemorrhagic enteritis in dogs is a major disease in South Africa and Nigeria. Both infection rates with CPV-2 and case fatality rates in young dogs are high. CPV-2 is a small, negative-sense, single-stranded DNA virus of 5.2kb long and a member of the Parvoviridae family,

The effects of productive adenovirus infection on host gene expression were studied using a line of methotrexate-resistant HeLa cells containing amplified amounts of the gene coding for dihydrofolate reductase (DHFR). These cells contain several hundred copies of the DHFR gene and overproduce DHFR RNA and protein.
