Abstract:
Background CD19-targeted chimeric antigen receptor-engineered (CD19 CAR)-T cell immunotherapy has shown promising efficacy in patients with relapsed or refractory (R/R) B-cell malignancies. The potential benefits of repeat infusions of CD19 CAR-T cells are unknown, and the factors associated with response, CAR-T cell in vivo expansion, and progression-free survival (PFS) after repeat infusion of CD19 CAR-T cells have not been investigated. Methods We analyzed the outcomes of patients with R/R B-cell malignancies after a second infusion of CD19 CAR-T cells (CART2) on a phase 1/2 trial (NCT01865617) at our institution. Responses after CAR-T cell therapy were evaluated around day 28 after infusion and defined according to the 2018 NCCN guidelines for acute lymphoblastic leukemia (ALL), 2018 iwCLL for chronic lymphocytic leukemia (CLL), and the Lugano criteria for non-Hodgkin lymphoma (NHL). Logistic, Cox and linear regression were used for multivariable analyses of response, progression-free survival and peak CD8+ CAR-T in blood, respectively. Bayesian model averaging was performed for variable selection. Results Forty-four patients evaluable for response (ALL, n=14; CLL, n=11; NHL, n=19) were included in this study. The median age at the time of CART2 was 58 (range, 23-73). Patients were heavily pre-treated (median prior therapies, 6; range, 2-13), and 16 patients (36%) had bulky (≥ 5cm) nodal or extramedullary disease. The median time from the first CAR-T infusion (CART1) to CART2 was 70 days (range, 28-712). Twenty-eight patients (64%) had received a CART1 dose ≥ 2x106 CAR-T cells/kg. Fifteen patients (32%) had not responded to CART1, 22 (50%) relapsed or progressed after having initially responded (complete response [CR], n=15; partial response [PR], n=7) to CART1; 7 (16%) received CART2 in PR after CART1. All characteristics are shown in the Table. We observed responses in all disease types, including 3 of 14 ALL patients (21%; all CR/CRi), 4 of 11 CLL patients (36%; CR/CRi, n=3; partial response [PR], n=1), and 9 of 19 NHL patients (47%; CR, n=2; PR, n=7). After a median follow-up of 43 months (range, 16-66) in alive and responding patients, the estimated 4-year PFS probability in responders was 23% (95% CI, 9-59%). The 4-year overall survival probability in responders was 36% (95% CI 19-71%) compared to 24% (95% CI, 12-47) in non-responders. Multivariable logistic regression modeling identified predictors of response after CART2: CART1 lymphodepletion (high-intensity cyclophosphamide and fludarabine [CyFlu] vs no CyFlu, OR=12.19, 95% CI, 1.10-1689.85, p=0.04), and peak of in vivo CAR-T cell expansion after CART2 (OR=2.31 per log10 CD8+ CAR-T cell/µL increase, 95% CI, 1.17-5.29, p=0.01). In a multivariable Cox model, a higher peak of CD8+ CAR-T cells after CART2 (HR=0.47 per log10 CD8+ CAR-T cell/µL increase, 95%CI, 0.33-0.68, p<0.001); CART2 > CART1 cell dose was associated with longer PFS (HR=0.36, 95% CI, 0.16-0.86, p=0.02). This suggested that CD8+ CAR-T cell peak after CART2 and factors increasing CART2 peak (e.g. prevention of immune rejection or increase in the infused cell dose) are key elements associated with outcomes of CART2. Hence, we looked at factors associated with higher CD8+ CART2 peak. In multivariable linear regression, CART1 CyFlu predicted a higher peak of CD8+ CAR-T cells after CART2 (high-intensity CyFlu vs no CyFlu, p<0.001 ; low-intensity CyFlu versus no CyFlu, p=0.002) after adjusting for disease type (CLL vs ALL, p=0.02; NHL vs ALL, p=0.04) and the total CD19+ cell count in blood (p=0.02). CyFlu being the most commonly used lymphodepletion prior to CAR-T cell therapy, we evaluated the impact of CART1 CyFlu lymphodepletion intensity by comparing high-intensity to low-intensity CyFlu in our multivariable models. Logistic regression suggested higher probabilities of response to CART2 in patients who received high-intensity compared to low-intensity CyFlu prior to CART1 (OR=3.83, 95%CI, 0.85-21.83, p=0.08). In multivariable analysis, CART1 high-intensity CyFlu was associated with higher CD8+ CAR-T cell numbers at day 60 after CART2 compared to low-intensity CyFlu (p=0.01) after adjusting for disease type and total CD19+ cell count in the blood. Conclusion Our findings suggest outcomes after second infusions of CD19 CAR-T cells might be improved with high-intensity CyFlu lymphodepletion prior to CART1 and by increasing the CAR-T cell dose at the time of CART2. Purpose The paper aims to examine the influences of the competencies, social skills and continuous learning ability on career success and career satisfaction among workers aged 50 and over. Design/methodology/approach A mixed methods approach consisted of 920 surveys and 11 semi-structured interviews with working individuals aged 50 or above, registered at a job agency specialized in older employees in The Netherlands. Multiple regression and content analysis were used to analyze the findings. Findings The survey showed a positive relationship between the competencies and career success and career satisfaction. However, the interviews revealed that not all older workers perceived the need to continuously stay up-to-date as positive, pushing them into retirement. Originality/value The influence of competencies on career success and career satisfaction of older workers has received only little attention from researchers.

Abstract Background: Chimeric antigen receptor-modified T cells (CARTs) targeting CD19 has illustrated therapeutic efficacy in diffuse large B cell lymphoma (DLBCL). However, severe cytokine release syndrome (sCRS) reaction and CAR-T-cell-related encephalopathy syndrome (CRES) are the main adverse reactions of CART therapy. Because of the concern of severe CRES which is associated with potential mortality, patients with primary central nervous system Lymphoma(PCNSL) are excluded in most of CART clinical trials. CD70 is a promising therapeutic target due to the restricted expression pattern in normal tissues and overexpression in some lymphoma tissues, so CD70 specific CARTs maybe avoid CD19-negative relapse .Here we report a case with refractory and relapsed PCNSL who achieved long-term disease-free survival by combination therapy of CD19 and CD70 specific CARTs without inducing CRS or CRES. Methods: CD3+ cells were selected from the apheresis PBMC and activated before lentiviral 4SCAR infection. The cells were transduced with a caspase 9-inducible, safety-engineered lentivector CAR containing anti-CD19 or -CD70 scFv fused with multiple intracellular signaling domains: CD28/CD27/CD3z-iCasp9 (4SCAR19 or 4SCAR70). The quality of apheresis cells, gene transfer and T cell proliferation efficiencies, and effective CAR T infusion dose were quantitatively scored and documented. Tumor sections were immunohistochemically stained with different antibodies including anti-CD19 and anti-CD70 antibodies to confirm antigen expression. The patient received cyclophosphamide and fludarabine conditioning regimen before the dual CARTs cell infusions. Case presentation: A 67-year-old male who was diagnosed as PCNSL of DLBCL in 2011.The patient received 6 cycles of temozolomide and high does methotrexate, and achieved CR. In December 2016 he relapsed and was treated with right frontal lobe space-occupying resection and multi-course chemotherapy including 2 course of rituximab, high dose methotrexate and temozolomide ,and 6 courses of rituximab, high dose methotrexate and ibrutinib,and he achieved remission again.However, He relapsed again in August 2017, with the clinical symptoms of dizziness , fatigue, left corners of the mouth askew and occlusal difficulties.MRI presented a residual mass on the right side of the postoperative cavity of 26mm*35mm*30mm and stale hemorrhage in left basal ganglia. After confirmation for CD19 and CD70 expression in his tumor specimens, the pt enrolled in the clinicaltrail :"Combination CART cell therapy targeting hematological malignancies"(clinicaltrail.gov registry NCT03125577).He received 4SCAR19 and 4SCAR70 T cells targeting CD19 and CD70 following lymphodepletion chemotherapy with FC regimen conditioning (FLU 30mg/m2, d1-3; CTX 300mg/m2, d1-3) in October 2017. Brain MRI 1 month later showed complete remission and the pt had symptomatic improvement. To date, after more than 8 months of follow-up, the pt remains in CR (figure1) and CART cells still can be detected . There was no CRS or CRES during the treatment and in follow-up period. Conclusions: The study results demonstrate that CARTs are able to pass through the blood-brain barrier without inducing CRS or CRES, suggesting that CNS tumor is not an absolute contraindication to 4sCART therapy. In addition, the combination of CD19 and CD70 specific 4sCARTs could effectively target PCNSL and achieves long-term disease-free survival without sCRS or CRES. Abstract Background: CD19 targeting chimeric antigen receptor T cells(CART19) therapy have shown great therapeutic potential in relapsed and refractory B cell acute lymphoblastic leukemia (ALL), but associated with risk of life-threatening adverse effects as severe cytokine release syndrome (sCRS) and CAR-T-cell-related encephalopathy syndrome (CRES).It's been reported that high leukemia burden before CART therapy, and high does infused CART cells are associated with severity of CRS and CRES. To decrease the risk of severe adverse effect, we applied integrated therapeutic strategy of using fourth generation CART cells , reducing leukemia blasts burden in bone marrow, and decreasing the dose of infused CART cells (www.clinicaltrials.gov; #NCT03407859 and #NCT03125577 ). Methods: Between May 23, 2016 and July 2, 2018,the trial enrolled 20 patients (pts) who were exhausted with all available treatment options, life expectancy >2 months,CD19-positive and diagnosis of B lineage ALL.T cells were apheresis collected and transduced with an apoptosis-inducible, safety-engineered lentivector CAR with the following intracellular signaling domains: CD28/CD27/CD3ζ-iCasp9 (4SCAR). In vitro amplication of CART was not performed. In pts with bone marrow blasts exceeding 50.0%, VDCP or similar chemotherapy was given to reduce the tumor burden, and then received FC conditioning regimen (FLU 30mg/m2, d1-3; CTX 300mg/m2, d1-3), while FC regimen was directly carried out in pts with bone marrow blasts less than 50.0%.In this trial, Pts received single CD19-targeted CARTs or multi-CARTs targeting CD19 and an additional antigen of CD22 or CD123.The level of CAR-T cells and cytokines in peripheral blood, as well as tumor burden was measured after infusion. Results: 20 pts were enrolled and infused with CAR-T cells. The median age is 37.5 (16-67) years old. Of these pts, 5 had prior HSCT and 14 had adverse genetic abnormalities, including 4 pts (20%) who were Philadelphia chromosome-positive(Ph+). All pts were previously treated with 2-22 courses of cytotoxic chemotherapy regimens. The overall objective response rate was 85%(17/20), and the complete response(CR) rate was 80%(16/20). The complete remission rate of 12 pts receiving single CD19-targeted CART therapy was 83 %(10/12), while 33%(4/12) of them had disease relapse at 6 months after infusion. Of the 7 people who received multi-CARTs infusion, 71%(5/7) achieved complete remission, with relapse rate of 29%(2/7) at 6 months after infusion. 3 pts who relapsed post transplantation received combination therapy of anti-CD19 CART and anti-CD123 CART, and all achieved minimal residual disease-negative CR within 1 month after CART infusion, 2 of whom maintained disease-free survival for 7 months and 11 months to date, respectively. Among the 7 people who underwent HSCT after achieving CR, 6 of them maintained disease-free survival for 3 months to 9 months. At a median follow-up of 115.5 days (ranging from14 to 384), the median overall survival was 269 days and the median event-free survival was 232 days. During treatment and follow-up, the most common adverse events were grade 1-2 cytokine release syndrome (CRS), with an incidence of 55%.No grade 3 or higher CRS was observed.12 pts were infused with a dose equal to or exceeding 5*10^5/kg, 10 of whom had CRS response. While only 2 of the 8 pts who received the infusion dose of less than 5*10^5/kg had CRS reaction and both of them were grade 1 CRS, suggesting low CAR-T cell doses decrease the risk of CRS (P=0.009). Interestingly, the objective response rate did not differ significantly between the low dose and high dose group. Conclusions: Based on fourth generation CART system, a therapeutic strategy of low tumor burden and low CART infusion dose shows a safer profiling while remaining potent efficacy against leukemia. Accumulating evidence indicates that species interactions such as competition and predation can indirectly alter interactions with other community members, including parasites. For example, presence of predators can induce behavioural defences in the prey, resulting in a change in susceptibility to parasites. Such predator-induced phenotypic changes may be especially pervasive in prey with discrete larval and adult stages, for which exposure to predators during larval development can have strong carry-over effects on adult phenotypes. To the best of our knowledge, no study to date has examined possible carry-over effects of predator exposure on pathogen transmission. We addressed this question using a natural food web consisting of the human malaria parasite Plasmodium falciparum , the mosquito vector Anopheles coluzzii and a backswimmer, an aquatic predator of mosquito larvae. Although predator exposure did not significantly alter mosquito susceptibility to P. falciparum , it incurred strong fitness costs on other key mosquito life-history traits, including larval development, adult size, fecundity and longevity. Using an epidemiological model, we show that larval predator exposure should overall significantly decrease malaria transmission. These results highlight the importance of taking into account the effect of environmental stressors on disease ecology and epidemiology.

Abstract Introduction/Objective Omphaloceles are frequently associated with chromosomal abnormalities, including aneuploidy and segmental alterations. High resolution chromosomal microarray analysis (CMA) can detect segmental alterations < 5 Mb, which is not detectable by G-banding. However, the prognostic significance of the segmental alteration in infant with omphalocele is not elucidated. Methods To identify omphalocele cases with genetic studies, a CoPath database search (1/2000 - 7/2017) was performed with key words “omphalocele” and “genetic”. From 1/2000 to 12/2008, only G-banding was performed. From 1/2009 to 7/2017, omphalocele cases were screened with karyotyping. Cases with normal karyotype were reflexed to CMA. Copy number gains/losses and corresponding genes were analyzed by the Affymetrix Chromosome Analysis Suite. Results Follow-up data are available in 75% (67/89) cases. There is no significant difference of the overall survival rate of male and female patients (80.5% vs 76.9%; χ 2, p = 0.7645). There are 16.9% (15/89) omphaloceles with aneuploidy, 10.1% (9/89) cases with segmental alterations by CMA, and 73.0% (65/89) cases with normal CMA and/or normal karyotype. Although patients with segmental alterations have a significantly higher survival rate than those with aneuploidy (44.4% vs 0%, χ2, p = 0.0119), their overall survival rate is significantly lower than infant with normal CMA and/or normal karyotype (44.4% vs 82.8%; χ2, p < 0.0001). Infants with segmental alterations carry a significantly worse prognosis than infants with normal genetic study. Conclusion To date, this is the first study of the prognostic significance of segmental alteration in infants with omphalocele. Our data demonstrated that omphaloceles with segmental alterations carry a significantly worse prognosis than those with normal CMA and/or karyotype. It is crucial to convey the prognosis to the parents with a fetus carrying segmental alterations; so the family could make an informed decision and get ready for an infant with special needs.

The use of cowpea as a forage for Central Missouri has been limited. High mid summer temperatures and frequent drought suggest that an alternative forage for livestock may be advantageous during the hot summer months. The ability of cowpea to withstand drought, high temperature and maintain high foliage protein, justifies research of the forage potential of this crop. Forage yield potential of two cowpea cultivars was evaluated in field experiments in 1968 and 1989, For both years a split plot experimental design was utilized with cowpea cultivar the main plot and harvest date the subplot Dry matter yields for both cultivars were similar at the early harvest dates. However, both fresh and dry shoot yields of Vita 3 (V3) were significantly higher than that of California Blackeye #5 (CB #5) at the later harvest stages. CB #5 plants produced a significantly higher stem dry weight while V3 produced higher leaf dry matter. Foliage regrowth after clipping was substantial for both cowpea cultivars. However, V3 produced 30% higher growth than CB #5. In vitro dry matter digestibility (IVDDM) varied with sample date (plant age) and plant parts sampled. IVDDM was 77% for leaves and 60% for stems of CB#5, and 80% for leaves and 65% for stems of V3.

The epidemiological role of domestic animals in the spread and transmission of SARS-CoV-2 to humans has been investigated in recent reports, but some aspects need to be further clarified. To date, only in rare cases have dogs and cats living with COVID-19 patients been found to harbour SARS-CoV-2, with no evidence of pet-to-human transmission. The aim of the present study was to verify whether dogs and cats act as passive mechanical carriers of SARS-CoV-2 when they live in close contact with COVID-19 patients. Cutaneous and interdigital swabs collected from 48 dogs and 15 cats owned by COVID-19 patients were tested for SARS-CoV-2 by qRT-PCR. The time elapsed between owner swab positivity and sample collection from pets ranged from 1 to 72 days, with a median time of 23 days for dogs and 39 days for cats. All samples tested negative, suggesting that pets do not passively carry SARS-CoV-2 on their hair and pads, and thus they likely do not play an important role in the virus transmission to humans. This data may contribute to confirming that the direct contact with the hair and pads of pets does not represent a route for the transmission of SARS-CoV-2.

Undiluted tomato petiole sap from a variety by K rate experiment (48 treatment rep combinations) was used to measure K concentration via the battery operated portable Cardy meter and ICP laboratory instrumentation. Three sample 1998 dates, 16 July., 21 Aug., and 8 Sept., resulted in K sap readings by ICP of 3917, 2612, and 2297 ppm, respectively. At sap levels below 3000 ppm the linear Cardy:ICP correlation was r = 0.04, but above 3000 ppm only 0.53. From 3500 to 5000 ppm K the Cardy meter under estimated actual sap K by 200 to 900 ppm. For the years 1999 and 2001, tomato petiole sap at each sample date (4) was diluted 1:1 with deionized water. The linear regression equation describing the relationship between ICP and Cardy meter measurements was: Cardy K ppm = 0.733 * ICP + 685 (r = 0.92, n = 190). The Cardy meter error over the 2000 to 5000 ppm K range was 8 to 12%. Petiole sap K, measured by either Cardy or ICP, was highly correlated to whole leaf K concentration both years. But even though the slope of the regression lines was similar the intercepts were significantly different (P≤.01). The significant 0.32% K difference in whole leaf between years precluded the development of a common regression line to predict whole leaf K from Cardy petiole sap determinations. The Cardy meter can be reliably used for tomato petiole K determination provided the sap is diluted and the usual handling precautions are taken to prevent petiole moisture loss.

In this article, I discuss the potential of digital storytelling as a site where feminist scholars in academia can get together to reflect upon their concerns about neoliberal burdens in academia. I rely on the recent literature about the feminist interventions in higher education in addition to giving an account of the grass-roots digital storytelling movement around the world and its feminism oriented journey in Turkey. Focusing on the digital stories told at the pre-conference digital storytelling workshop in EMU (Eastern Mediterranean University) immediately before the 5th International Conference on Gender Studies in March 2015, the aim of that digital storytelling workshop was not only to facilitate the creation of digital stories told by feminist academics but also to carry these digital stories into the program of the conference in a special session that hosted both the screening of these digital stories and an open discussion about our neoliberal concerns in academia and everyday life. This article also provides an account of the main discussions and attempts to suggest the digital storytelling workshop as a tool for broadening the interactions between academic and activist feminisms.

Radiocarbon dating of 32 stratigraphic samples aided by Bayesian analysis has allowed the author to produce a high precision chronology for the construction and development of a continental Neolithic long barrow for the first time. She shows when and how quickly people living on the shore of the Baltic adopted pit graves, megalithic chambers and long barrows. Better than that, she provides a date for the famous cart tracks beneath the final barrow to 3420–3385 cal BC. Although other parts of the package — ploughing and pottery — are late arrivals, her analysis of the global evidence shows that Flintbek remains among the earliest sightings of the wheel in northern Europe.

The majority of people in Kuta Dame Village work as farmers. Several outreaches have been carried out to develop the agricultural sector in Kuta Dame village. However, some of the extension services rarely touched on effective technology to support agriculture in Kuta Dame village. One of the technologies that need to be developed by the Kuta Dame village farmer group in supporting agriculture is an automatic sprayer. This development is necessary because most farmers are still using the carrying type sprayer with a manual pump (diaphragm pump). The sprayer's diaphragm pump pressure produces a relatively low spray pressure, especially when the operator is tired of moving the pump lever. As a result, the spraying performance is not perfect, making the spray grains not smooth and not reaching the leaf surface evenly. A community service program was implemented to implement automatic sprayers for farmers in Kuta Dame village. This community service program aims to carry out the planning process and use of an electric sprayer appropriately to increase spraying fertilizers and pesticides on crops. Implementing this community service activity starts from planning activities, implementing academic counseling, implementing the practice, handing over an electric sprayer, monitoring, and satisfaction analysis.

Cocaine- and amphetamine-regulated transcript (CART) is a peptide suggested to play a role in gastrointestinal tract tissue reaction to pathology. Gastric ulceration is a common disorder affecting huge number of people, and additionally, it contributes to the loss of pig livestock production. Importantly, ulceration as a focal disruption affecting deeper layers of the stomach wall differs from other gastrointestinal pathologies and should be studied individually. The pig’s gastrointestinal tract, due to its many similarities to the human counterpart, provides a valuable experimental model for studying digestive system pathologies. To date, the role of CART in gastric ulceration and the expression of the gene encoding CART in porcine gastrointestinal tube are completely unknown. Therefore, we aimed to verify the changes in the CART expression by Q-PCR (gene encoding CART in the tissue) and double immunofluorescence staining combined with confocal microscopy (CART immunofluorescence in enteric nervous system) in the porcine stomach tissues adjacent to gastric ulcerations. Surprisingly, we found that gastric ulcer caused a significant decrease in the expression of CART-encoding gene and huge reduction in the percentage of CART-immunofluorescent myenteric perikarya and neuronal fibers located within the circular muscle layer. Our results indicate a unique CART-dependent gastric response to ulcer disease.

Prior to the advent of combined antiretroviral therapy (cART), human immunodeficiency virus-associated nephropathy (HIVAN) was inevitably associated with rapidly progressive renal failure and dialysis dependence. HIV-1 seropositive patients often met with untimely deaths due to complications of end-stage renal disease (ESRD), opportunistic infections, or other HIV-related end-organ failure. Although the association between cART and improved outcomes in HIVAN has been recognized for over 20 years, no randomized trials have specifically examined this effect to date. In terms of reversal of dialysis-dependent renal failure after cART initiation, only a handful of case reports exist. The authors report a case of a 44-year-old Latino male requiring thrice-weekly haemodialysis in the setting of biopsy-proven HIVAN who was able to stop dialysis in 7 months after being initiated on cART.

Long-distance migrants may be particularly vulnerable to climate change on both wintering and breeding grounds. However, the relative importance of climatic variables at different stages of the annual cycle is poorly understood, even in well-studied Palaearctic migrant species. Using a national dataset spanning 46 years, we investigate the impact of wintering ground precipitation and breeding ground temperature on breeding phenology and clutch size of 19 UK migrants. Although both spring temperature and arid zone precipitation were significantly correlated with laying date, the former accounted for 3.5 times more inter-annual variation. Neither climate variable strongly affected clutch size. Thus, although carry-over effects had some impact, they were weaker drivers of reproductive traits than conditions on the breeding grounds.

In the face of rapid and complex and unpredictable changes in life, educational innovation must be an important and critical priority because education is still the main support for improving the quality of human resources. The educational innovation model in the Indonesian Ministry of Education and Culture still tends to be more dominant in using a “top-down model” strategy, namely educational innovation developed from above by policy makers from the central level, to be implemented imperatively down to the lowest level of educational institutions. This will dull the creativity of the teaching staff and education staff. Thus, educational innovation should be balanced with a "bottom-up model" strategy, namely a model of educational innovation developed from below, which originates from the creativity of education personnel and educational staff in each educational institution implemented as an effort to improve the quality of education delivery. In order for educational innovation to run faster than other changes, it is necessary to look for more powerful models and strategies through various scientific approaches. The educational innovation model with a strategy of implementing the concept of "Dare to be different" is a concept used to motivate education providers to carry out educational innovations by not contradicting existing educational innovation models and strategies. This model focuses more on internalizing the enthusiasm for educational innovation, especially by educators. Dare to be different means daring to be different, so start with the word dare which stands for the words dream, attitude, relationship, and excellence.

Abstract Background: Multiple myeloma (MM) is an incurable plasma cell malignancies despite the advent of numerously new drugs. Survival was poor particularly for high risk patients such as R-ISS stage III. The preliminary data from our center showed that the median PFS after auto-HSCT was 24 months for patients with R-ISS III stage, while 17 months for patients that achieved PR or less after induction. Chimeric antigen receptor (CAR)-transduced T cells is a promising strategy for cancer immunotherapy. Our previous study showed good response for RRMM patients after CD19 and BCMA-specific CART therapy without severe CRS and other deadly side effects. To improve the survival of high risk patients, this study was designed to observe the safety and efficacy of combined infusion of CD19 and BCMA-specific CART cells after autologous transplantation (SZ-MM-CART02 study, NCT 03455972). Methods:18-65y NDMM in R-ISS stage III, or who only achieved PR or less after 4 cycles of PAD triplet induction were enrolled with serum creatinine (Cr) <2.0 mg/dL, and adequate hepatic, cardiac and pulmonary function. BCMA and CD19 expression on MM cells were analyzed by flow cytometry. Lymphocytes were collected from PBSCs and cultured with an anti-CD3 monoclonal antibody to activate T-cell proliferation. The cells were transduced with recombinant lentiviral verctors which respectively contained the anti-BCMA or anti-CD19 single chain variable fragment (scFv), the cytoplasmic portion of the OX40 and CD28 costimulatory moiety, and the CD3z T-cell activation domain. This is the new third generation CAR technique applied in clinic. BUCY were used as conditioning, followed by infusion of autologous stem cells. Median time to engraftment were 10 days for nutrophils. CART-19 (1×107/kg on d0) and CART-BCMA cells as split-dose (40% on d1 and 60% on d2) were infused derectly on d14 to d20 after autologous transplantation. Levels of CAR-transduced cells are measured by qPCR. The cytokine release syndrome (CRS) was graded according to the UPen cytokine release syndrome grading system. Neurotoxic side effects and other toxicities were assessed according to the CTCAE v 4.03. Plasma levels of IL-2, IL-4, IL-6, IL-10, TNF-alpha, IFN-gamma, and IL-17A proteins were determined with a cytokine kit. Imids alone were given as maintenance therapy. Responses were assessed by IMWG criteria. 10-color flow cytometry was used to monitor MRD regularly after CART treatment. The median of follow-up was 3 (2∼11) months. Results: To date, 9 patients have completed the CART cells infusion (cohort 1). All cases expressed BCMA >50% without CD19 expression on MM cells. CRS occurred in 9 patients (100%) grade1 or 2 associated with fever(n=9), fatigue (n=9), elevated IL-6 and CRP (n=9), elevated ALT (n=1, grade 1). Two patients needed to use low-dose vascular active drugs (pts 04 and 07). Other toxicities to date included coagulopathy (n=6, grade 1 for 4 pts and grade 2 for 2 pts), elevated troponin T (n=4, grade 1), and atrial flutter (n=1). There was no serious CRS or neurologic complications occurred in this group of patients. The ORR was 100% with all patients were monitored for a period of more than 2 months, which may be eventually further improved. There were 2 CR, 1 VGPR, 4 PR, 2 SD after induction; 3 CR, 2 VGPR, 4 PR after APBSCT; 3 CR, 6 VGPR after CART therapy. MRD negativity in BM increased from 37.5% after transplantation to 66.7% after CART therapy latest. Four patients (pts 02, 03, 06 and 07) obtained partial PR after transplantation, and got VGPR after CART cells infusion. We found dramatic in vivo CART expansion that median of peak value of CART copies was 1059.54 folds (ranged from 536.90 to 10997.93 folds) which was 100 folds to that with RRMM patients in our previous study. Conclusions: Tandom autologous transplantation and combined infusion of CART-19 and CART-BCMA cells could be another choice of consolidation treatment for high risk MM patients. Toxicities to date including CRS and organ function impairment seemed to be mild and reversable. It is worthy of further study to compare DFS, OS between single autologous transplantation and tandom transplantation with CART therapy. Immune environment in high risk patients with multiple myelomaI remodelled by auto-HSCT may contribute to more rapid expansion of CART cells than that in RRMM patients, suggesting that the extent of CART expansion depends more than tumor burden. In 1714 the Catalans, being in the camp of the losers in the War of the Spanish Succession, finally lost any hope to gain independence. 300 years later, nationalists, who are in power now in Catalonia, want to take advantage of that memorable date by organizing a regional referendum in order to choose their own path of development. In the aggravating conflict of interests between Madrid and Barcelona both parties use not only political and propaganda measures, but also financial leverages of influence. The ethnonational issue in the multiethnic Spain is far from being solved, because for centuries the process of forming a unified Spanish state was characterized by political union of several genetically related (except Basque) ethnic communities, although with significant socio-cultural and linguistic differences among them. It is not coincidental that regional identity in Spain is still extremely strong, while the interethnic consolidation of the Spanish nation can be characterized as incomplete, which can be seen, particularly, in the intensification in recent decades of radical nationalist and separatist sentiments in Catalonia, the Basque Country and some other autonomies. Among reasons which escalated confrontation between supporters and opponents of Catalonian independence, we should mention the global financial crisis that hit the regions of Spain as well as the overall national economy. The separatists have many barriers on their way, starting from the constitutional provisions proclaiming Spain a united and indivisible state which impede to carry out regional plebiscites, and ending by an ambiguous attitude towards the hypothetical independence of Catalonia by both the residents of the region and in other parts of the country. It is important to keep in mind that in today's world the possibility of breaking large multiethnic state into ethnically constituting elements is more possible than 40-50 years ago. Disintegration of Yugoslavia and the Soviet Union and the emergence on the world political map of Abkhazia, South Ossetia, and most recently of the Republic of Crimea - confirm this. The Catalonian nationalists in their arguments in favor of the sovereignty of Catalonia refer to the arbitrariness of the borders formed in the past between separate European countries and the regions. Modern state boundaries indeed are not the result of rational planning, democratic arrangements or consideration of the local population's opinion. In many ways, the current borders are the result of historical processes, often - of bloody wars and territorial annexations. In other words, they are the result of actions which are contrary to current international law and moral norms of behavior. Independent Catalonia is still a hypothesis, but under certain conditions and, more importantly, as a result of concerted efforts of regional nationalists it could become a reality.

The fate of pre-harvest-sprayed dicofol (DCF) on date fruits has been investigated. Date palm trees, variety Sukkari, were sprayed with DCF (18.5%, EC) at the rate of 200 ml/100 L. DCF residues in date fruits were determined at different time intervals using high performance liquid chromatography (HPLC) with UV detection at 220 nm. The method was based on extraction with ethyl acetate. The results showed that the HPLC response was linear (r2 > 0.98) for DCF in the range of 0.0 to 2 mμg. The limits of detection and quantification were 0.24 and 0.80 ppm, respectively. The method was developed by using spiked date fruits at levels of 0.25, 0.50 and 1.0 ppm. Recovery percentage was satisfactory with a range of 77.2 to 103.6% and an RSD ranging from 6.33 to 11.84%. A biphasic model was assumed in order to carry out the statistical study of the loss of DCF from date fruits. The results showed that the initial deposit of DCF on date fruits was 11.22 ppm, while the residue value decreased to 1.90 ppm seven days after spraying, which is below the tolerance limit for fruits and vegetables (2 ppm). Also, the data indicate that there was a faster rate of DCF loss in the first phase than the second one. This is clearly reflected in the t1/2 values, where the half-lives of DCF were 1.35 and 38.52 days, for the first and second phase model, respectively. The described method is rapid and sensitive, with satisfactory recoveries and reproducibility.

This study aims to design accounting applications for the preparation of financial statements at D'Haus Cake's business. The approach used in this research is a case study. The design process begins with studying the transactions and reports that D'Haus Cake’s business needs in the form of purchase and sale transaction forms, and general journals. The required reports are in the form of all transaction journal records, ledgers, trial balances, profit and loss reports, and statements of financial position. After studying the system requirements, then proceed to the application development stage using Microsoft Access 2013 software. The next stage is to test the application to obtain adequate confidence in transaction processing by comparing the results of processing with manual calculations carried out. The implementation process is carried out by converting the initial data on the conversion date and making input transactions up to the company's operating date. After implementing and converting, the final stage is to carry out training for users and carry out a process of improvement and adjustment to the application based on the feedback received from users. The process of designing and implementing the application has been considered successful after the user states that the application has met all user needs and operates well.

Over the past four years, Pat Thaddeus and I have been using the CfA 1.2 m telescope to carry out a new CO survey of the first Galactic quadrant with a sensitivity per solid angle 3–10 times higher than previous CO surveys of the plane, and 16 times higher than the only previous extensive wide-latitude survey (Dame et al. 1987). Since a preliminary purpose of this survey is the study of molecular gas out of the plane in the inner Galaxy, the observations have so far been confined mainly to several vertical strips across the plane covering a few degrees in Galactic longitude and 6°–8° in Galactic latitude; details of this on-going survey are given in Dame & Thaddeus (1995).

Consideration was given to basic variation principles and methods of calculating structural mechanics. Distinctive features of forming systems of equations of classical methods of structural mechanics and known numerical methods were demonstrated. Possible advantages of methods with respect to accuracy and rate of calculations were revealed. Consideration was given to possible improvements of existing mathematical models in elastoplastic domain. Possible directions of development in the field of engineering methods for calculating strength were proposed.

Abstract Climate change has advanced the phenology of many organisms. Migratory animals face particular problems because climate change in the breeding and the wintering range may be asynchronous, preventing rapid response to changing conditions. Advancement in timing of spring migration may have carry-over effects to other parts of the annual cycle, simply because advancement of one event in the annual cycle also advances subsequent events, gradually causing a general shift in the timing of the entire annual cycle. Such a phenotypic shift could generate accumulating effects over the years for individuals, but also across generations. Here we test this novel hypothesis of phenotypic response to climate change by using long-term data on the Arctic tern Sterna paradisaea. Mean breeding date advanced by almost three weeks during the last 70 years. Annual arrival date at the breeding grounds during a period of 47 years was predicted by environmental conditions in the winter quarters in the Southern Ocean near the Antarctic and by mean breeding date the previous year. Annual mean breeding date was only margiually determined by timing of arrival the current year, but to a larger extent by arrival date and breeding date the previous year. Learning affected arrival date as shown by a positive correlation between arrival date in year (i + 1) relative to breeding date in year (i) and the selective advantage of early breeding in year (i). This provides a mechanism for changes in arrival date being adjusted to changing environmental conditions. This study suggests that adaptation to changing climatic conditions can be achieved through learning from year to year.

In Bangladesh, the popularity of date and its consumption surge few-fold during Ramadan among Muslims owing to the profound emotion related to religious belief that breaking of dawn-to-dusk fasting with dates is fulfilling a Sunnah. The present study aimed to explore the microbiological quality of the five most popular fresh date samples (Nagal, Dhapas, Boroi, Morium, and Tunisia) purchased from different locations of Dhaka City, Bangladesh. Among 25 samples tested, 23 were observed to carry 3.30-5.65 Log CFU/gm aerobic bacteria and 3.30-5.36 Log CFU/gm yeasts and molds population. Coliform bacteria and Escherichia coli were not found in any of the samples analyzed in this study. However, except Nagal and Boroi from Mohammadpur and Mirpur, respectively, none of the samples were found safe to consume according to the microbiological grade recommended by Woolworths Quality Assurance Standard (WQAS), 2009, as fresh dates with microbial load can cause food poisoning or even foodborne intoxication. Date samples with less or no processing is responsible for this contamination which can be subsided and eliminated by appropriate handling and hygiene practices during postharvest processing.

<p><strong>Abstract.</strong> The Carte Rouge is one of the most notorious Hungarian ethnographical maps to date. The map was planned and executed by Count Pál Teleki (1879&amp;ndash;1941) in 1918. Teleki, who was the chief secretary of the Hungarian Geographical Society, later served as the prime minister of Hungary twice within his political career. The first edition of this particular map was published on the 21st of February in 1919. Teleki played a central role in arranging, ascribing and allocating Hungarian geographical works for the peace-negotiations following the First World War. The term “Carte Rouge”, derives from the French; critiquing the subjective colouring of the map. During this period of time, political correspondents and those in power were responsible for planning such maps in order to support their own political programs with regards to their respective national ties and territorial claims.</p><p>The red pockets on the map signify areas which designate nationality, ethnicity, and language claims, on the part of both the locals and/or the author of the map. Particularly of note, is that this map was viewed and used by the leading politicians of the Entente-powers.</p><p>On the 16th of January in 1920, the day on which the Hungarian delegations received the peace terms of the First World War, Albert Aponyi gave a speech of significant weight in front of the Entente representatives in Paris, after the peace terms were received. Aponyi (1846&amp;ndash;1933), who was the head of the Hungarian delegation, showed this particular map to those present at this meeting following his speech, as justification and proof of his points. Apponyi sent the map to Teleki and to the members of various delegations. Isaiah Bowman (1878&amp;ndash;1950), for example, also had a private copy of the map. Bowman's critique of Teleki's ethnographical map is most significant in this discussion:</p><p>„<i>This idea has occurred to me since examining a map by Count Teleki which gives altogether a wrong impression of the distribution of Magyars in Transylvania that we ought to keep a fairly good collection of propaganda maps, of which I have here several very striking examples, and sometime after I return I should like to write a little paper on the various types of lies and liars that I met in this form of cartography. It would be good popular education, because so many people regard what appears on a map as gospel truth, and almost none of the people, who print maps, understand map technique sufficiently well to distinguish between the various principles followed in the construction of maps. (…).</i>”</p><p>It is not certain as to whether Bowman was indeed writing about this particular map, however Teleki had at that point and time-edited no other ethnographical map. Others, for example, the Austrian Randolf Rungaldier (1892&amp;ndash;1981) had a very positive opinion about the Carte Rouge.</p><p>In total, and to the best of my knowledge, the Carte Rouge has 12 editions and 27 various versions.</p><p>In the following analysis, I will consider how the Carte Rouge can be compared to other, similar international examples of the region, based on archival research I conducted in Paris, London, Budapest and Milwaukee. For instance, Emmanuel de Martonne (1873&amp;ndash;1955), who also adopted a critical approach to the Carte Rouge, planned a comparable ethnographical map of Romania. This map used another method of cartographic representation; nonetheless, the various territorial concepts designated by the colour-selection, the borders and the represented (and not represented) territories are facets that have similar features.</p><p>Later Teleki &amp;ndash; as member of the Mossul-Commission &amp;ndash; designed an ethnographical map of the Turkish-Iraqi border-area, which afforded him international professional recognition. The method of cartographic representation was the same, as was utilized in the creation of the Carte Rouge.</p><p>Resultantly, the Carte Rouge inspired the production of other ethnographical maps in Hungary as well as in other countries. In Hungary, Ferenc Fodor (1887&amp;ndash;1962) &amp;ndash; as the member of the Hungarian Peace-Preparation Office (Béke-előkészítő Iroda) &amp;ndash; edited a manuscript map about Central-Europe using the same "representing-method". This map was published under the name of G. Bery in 1938. A part of this map was also reused as a propaganda poster. Later, during the Second World War Imre Jakabffy (1915&amp;ndash;2006) continued this form of mapping with a slightly modified version of this method of cartographic representation at the Institute of Political Science (Államtudományi Intézet) in Budapest. Subsequently, other ethnographical maps were edited using this method of cartographic representation by the French and by Romanians.</p><p>In sum, this map was a typical product of the period, as it supported a particular territorial concept. The opinions about the map &amp;ndash; not independent from the political context &amp;ndash; were very differential in form. Regardless, this method of cartographic representation had an extended international carrier in the fields of geography and cartography for years to come afterwards.</p></p>

PURPOSE Rare cancers are challenging for researchers, as clinicians and scientists have difficulty recruiting sufficient patient cases to power studies appropriately. Likewise, patients often are frustrated by a lack of specific information or evidence base for their cancer and, although eager to participate in research, have limited opportunities. We established CART-WHEEL.org, an online patient-entered database, to directly engage patients in the research process, collect rare cancer data, and facilitate their entry into additional research. PATIENTS AND METHODS Patients access CART-WHEEL.org directly online. Clinical information is collected from users via a streamlined questionnaire developed collaboratively with consumer groups to ensure accessibility and relevance. Data collected include the following: patient demographics, comorbidities, and risk factors and tumor diagnostic, biomarker, and treatment history. Patients can download a medical summary for personal use; consent for research use of data; and indicate willingness to be contacted about other research or clinical trials. We describe data collected to date and its validation, and we provide examples of how CART-WHEEL.org can facilitate rare cancer research. RESULTS From January 2010 to March 2018, 558 patients provided consent and entered their rare cancer data. One hundred distinct rare tumor types and patients from 22 countries were included. Validation of data entered by 21 patients with sarcoma against a hospital database demonstrated accuracy sufficient to facilitate future research in key fields, such as tumor site (95%) and histopathologic diagnosis (90%). Examples of CART-WHEEL–based disease-specific projects, subsequent recruitment to other rare cancer projects, and rare cancer patient cases of interest are described. CONCLUSIONS Online platforms like CART-WHEEL.org can engage consumers directly, facilitating collection of patient-entered rare cancer data for hypothesis generation, and connect patients with researchers to enable specific rare cancer research and clinical trials.

Risk assessment models for gray leaf spot of maize, caused by Cercospora zeae-maydis, were developed using preplanting site and maize genotype data as predictors. Disease severity at the dough/dent plant growth stage was categorized into classes and used as the response variable. Logistic regression and classification and regression tree (CART) modeling approaches were used to predict severity classes as a function of planting date (PD), amount of maize soil surface residue (SR), cropping sequence, genotype maturity and gray leaf spot resistance (GLSR) ratings, and longitude (LON). Models were development using 332 cases collected between 1998 and 2001. Thirty cases collected in 2002 were used to validate the models. Preplanting data showed a strong relationship with late-season gray leaf spot severity classes. The most important predictors were SR, PD, GLSR, and LON. Logistic regression models correctly classified 60 to 70% of the validation cases, whereas the CART models correctly classified 57 to 77% of these cases. Cases misclassified by the CART models were mostly due to overestimation, whereas the logistic regression models tended to misclassify cases by underestimation. Both the CART and logistic regression models have potential as management decision-making tools. Early quantitative assessment of gray leaf spot risk would allow for more sound management decisions being made when warranted.

The research investigated some factors influencing the rate of stabilisation of wastewater in the spring period in continental climate waste stabilisation ponds, and in particular the potential for bringing forward the discharge date by optimising storage capacity and dilution. Experiments using pilot and model-scale ponds were set up in Almaty, Kazakhstan. These simulated operating regimes for a facultative and storage/maturation pond system subject to ice cover from late November until late March. Two pilot-scale facultative ponds were operated at hydraulic retention times (HRT) of 20 and 30 days, with surface loading rates of 100 and 67 kg BOD ha−1 day−1. Effluent from the 20-day HRT facultative pond was then fed to two pilot-scale storage/maturation ponds which had been partially emptied and allowed to refill over the winter period with no removal of effluent. The paper discusses the results of the experiments with respect to selection of an operating regime to make treated wastewater available early in the spring. Preliminary results indicate that there may be potential for alternative operating protocols designed to maximise their performance and economic potential.

Sensitivity analysis can be used to carry out hedging strategies. The sensitivity value measures how much the price change of the option influenced by some parameters. The aim of this study is to determine the sensitivity analysis of the buying price of European option by using the Greek method on Black Scholes Formula. From this study we get the values of delta, gamma, theta, vega, and rho. The values of deltas, gamma, vega, and rho are positive, which means that the value of the option is more sensitive than the corresponding parameter. The most sensitive value of gamma is obtained when the stock price approaches the strike price and approaches the expiry date. The value of theta obtained is negative and hence the most sensitive theta value is when the value is getting smaller. While, the most sensitive value of vega is obtained when the stock price is close to the strike price and is far from the expiry date. The most sensitive value of rho is obtained when the stock price gets bigger and farther from the expiry date.

SummaryWheat is one of the modern world’s staple food sources. Its production requires good environmental conditions, which are not always available. However, agricultural practices may mitigate the effects of unfavorable weather or poor-quality soils. The influence of environmental and crop management variables on yield can be evaluated only based on representative long-term data collected on farms through well-prepared surveys.The authors of this work analyzed variation in winter wheat yield among 3868 fields in western and eastern Poland for 12 years, as dependent on both soil/weather and crop management factors, using the classification and regression tree (CART) method. The most important crop management deficiencies which may cause low wheat yields are insufficient use of fungicides, phosphorus deficiency, non-optimal date of sowing, poor quality of seeds, failure to apply herbicides, lack of crop rotation, and use of cultivars of unknown origin not suitable for the region. Environmental variables of great importance for the obtaining of high yields include large farm size (10 ha or larger) and good-quality soils with stable pH. This study makes it possible to propose strategies supporting more effective winter wheat production based on the identification of characteristics that are crucial for wheat cultivation in a given region.
