Abstract:
The calcium-dependent antibiotic (CDA) is a non-ribosomally synthesised lipopeptide produced by Streptomyces coelicolor A3(2). The cda biosynthesis gene cluster occupies approximately 83 kb at the 10 o'clock region of the chromosome. A gene encoding a positive regulator of CDA production, designated cdaR, had been previously identified. Sequence similarly suggests that CdaR is a member of the Streptomyces antibiotic regulatory protein (SARP) family. CdaR is predicted to be 638 amino acid residues in size with a putative ATP/GTP-binding site motif A located in its C-terminal region. Mapping of the transcription start point of cdaR found that it possesses an unusually long untranslated leader sequence of 358 nt. cdaR transcripts were easily detectable at the earliest time point examined (24 h) and decreased over time. A 0.75 kb in-frame deletion in cdaR abolished CDA production. Nuclease protection experiments, using RNA isolated from a S. coelicolor cdaR deletion mutant, indicated that CdaR is required for transcription of some of the genes within the cda cluster, including itself. His6-tagged CdaR was produced in E. coli. Preliminary evidence from mobility shift assays suggested that CdaR binds directly to the 5' end of cdaR and also possibly to the promoter region of cdaPSI (peptide synthetase 1). Unexpectedly, introducing multiple copies of cdaR (approximately 50 copies/chromosome) into S. coelicolor 2377 and MT1110 did not detectably increase CDA production. Correspondingly cdaR transcripts only increased by ~ 30% in S. coelicolor MT1110 containing multiple copies of cdaR; this result suggests that transcription of cdaR is very tightly regulated.

Abstract:
In satellite applications, a circularly polarized satellite antenna is desirable with a pattern that results in constant received power while the distance between the transmitter and the receiver is changing. The Circular Disk - Annular Ring (CDAR) antenna satisfies these requirements along with other requirements for the satellite antenna. The CDAR antenna is a combination of a Circular Disk and an Annular Ring patch antennas. In this thesis, a circularly polarized CDAR antenna that is fed from a single point is designed at the center frequency of 8.2 GHz. This antenna is investigated and optimized to ease the fabrication process. The design parameters are defined on this report and optimized by using an Electromagnetic Simulation software program. In order to verify the theoretical results, Circular Disk - Annular Ring Antenna is produced as a prototype. Measurements of antenna parameters, electromagnetic field and circuit properties are interpreted to show compliance with theoretical and simulation results. The values of deviation between theoretical and experimental results are also discussed.

Abstract:
This thesis examines the legacy and consequences of Constructivism in art, from the early days of the Russian avant-garde to recent times and today. The Introduction explains how the concept of faktura, first theorised around 1912 by David Burliuk and others, came to designate the material qualities rather than the subject-matter of art. Chapter 1: Towards A Constructive Ideal, traces the progress of faktura in the reliefs of V.Tatlin from 1913. The ancestry of faktura in the Eastern icon tradition is emphasised, where a close relation between sight and touch already suggested a new type of encounter between the viewer and the object of art. The chapter further examines the importance of faktura to Suprematism, and examines A.Rodchenko’s appeal to line as a rational element of construction and as a weapon against ‘composition’ in art. Chapter 2: Time and the Viewer presents evidence of the importance to artists in the period 1940-70 of the real-time encounter between viewer and the art-object, first in American and British ‘Constructionism’, and then in the Minimal art of Judd, Morris and others. The chapter ends with a discussion of temporality in relation to abstract paintings of Rothko and de Kooning. Chapter 3: Irregular Curves: Science and ‘The Organic’ reprises the minority Constructivism of Mikhail Matyushin and Pyotr Miturich that claimed organic structures were superior to technicist ones. Evidence is presented that the rectilinear grid was always subject to challenge, initially in the art of Emma Kunz, Jean Arp and other pre-war modernists but latterly among those for whom ‘field’ and ‘curvature’ became relevant formats after 1945. Particularly with the development of computing from the 1970s, new geometries based on iteration and scale-invariancy assumed major relevance to constructed art. Chapter 4: Constructivism Now presents evidence of the application of Constructivist principles in recent art, initially in Dan Flavin’s ‘monuments’ to Tatlin and others and subsequently in so-called Neo-Geo and Op art of the 1970s and 1980s. From that period on, albeit often in a register of irony and ‘serious play’, faktura in a Constructivist sense continued, and continues today, to define the relation between viewer and object of art.

Abstract:
The purpose of this study was to better understand student learning in asynchronous computer-conference discourse (ASD) for non-native speakers of English in India through the Community of Inquiry (COI) framework. The study looked at ASD from an online course taught in the fall of 2015 to 25 students in a seminary in South India. All but one of the students were non-native speakers of English. The class consisted of 22 men and 3 women. Eight students spoke languages from the Dravidian family of languages (Malayalam, Tamil, Telegu and Kannada). Eight students were from the Northeastern states of Manipur, Nagaland and Tripura, where most languages are from the Sino-Tibetan family. Three students were native speakers of Indo-Aryan languages (Odiya and Assamese). Five students were from Myanmar representing several Sino-Tibetan languages. The COI is a framework used to understand learning in ASD, often used in online learning. To study the ASD of this group, critical discourse analysis (CDA) was used with the COI to capture the unique socio-cultural and linguistic conditions of this group. The study revealed that non-native speakers of English often reach the Exploration phase of learning but rarely show evidence of reaching the Resolution phase. This phenomenon was also observed in native English speakers as reported in the literature. Also, the structure of ASD showed that students took an examination approach to discussion shaped in part by their epistemology. This examination approach shaped how knowledge was constructed. CDA also showed that the discourse acquired an instructor-centered structure in which Resolution and Repair were initiated and finalized by the instructor. The study advances the COI framework by undergirding it with a theory of asynchronous discourse using critical discourse analysis and capturing cognitive, social and teaching presence phenomena for non-native speakers that were not observed through the traditional COI framework. These phenomena were driven by cultural, epistemological, and linguistic forces and require a rethinking of the COI for contexts outside of North America. The study also demonstrates that learning for non-native speakers in ASD is challenged by these very same forces. Therefore, design for online learning should account for these phenomena.

Novas medidas de controle das vazões excedentes surgiram nos últimos anos e buscam contornar as mudanças nos ciclos hidrológicos, imitando e mantendo as condições naturais existentes antes dos processos de urbanização. Uma dessas alternativas, conhecida genericamente pelo nome de reservatório de armazenamento, procura reter parte do escoamento superficial gerado durante as precipitações, para depois fazer sua devolução de forma lenta e gradual aos leitos dos córregos e rios, atenuando o pico dos hidrogramas e redistribuindo as vazões ao longo do tempo. Uma das variáveis desse reservatório de armazenamento, que permanece seco entre eventos chuvosos, é denominado de reservatório de detenção e constitui-se o objeto deste trabalho. Sua implantação e utilização são discutidas por meio das experiências da CDHU, através de estudos de casos reais, onde prova-se que a incorporação desse novo dispositivo de drenagem urbana é ainda um assunto polêmico, para o qual técnicos, governantes e população estão despreparados. É legítimo o anseio da sociedade por medidas sustentáveis e ambientalmente corretas, na qual os reservatórios de detenção se enquadram, porém sua implementação por força de medidas legais, muitas vezes, tem sido realizada de forma pouco conseqüente, sem planejamento e ponderação quanto às dificuldades e implicações dessa atitude. Denota-se também, a necessidade de avaliação da eficiência destes dispositivos e de normatização das formas de aplicação, dimensionamento, operação e manutenção. Além disso, é necessário a sensibilização, a conscientização e a educação em todos os níveis, pois existem paradigmas a serem quebrados e obstáculos a serem vencidos. Ao final do trabalho, sugere-se um pequeno roteiro com diretrizes gerais para incorporação dos reservatórios de detenção em novos empreendimentos, com o intuito de orientar e facilitar a utilização desses dispositivos, melhorando sua aceitação pela sociedade.
New stormwater control techniques have emerged during last years, trying to mimic and keep the natural conditions of hydrologic cycle. One of these measures, known as storage facility, detains part of the rainfall runoff to release it at controlled rates downstream, attenuating the hydrograph peak and redistributing the volume over a certain period of time. The focus of this report is the detention basin, a storage facility designed to empty out between runoff occurrences. Its insertion and use are discussed through CDHU experiences, based on real cases, where the results show that this new urban drainage control system is still a polemic issue, for what technicians, governments and population are unprepared. Society’s concerns about sustainable and environmentally correct measures, inside which detention pond is placed, are rightful, however its implementation in the course of legal procedures has been made carelessly, unplanned and without considerations of difficulties and impacts. In addition, evaluation of its performance along with regulation of using forms, dimensioning, operation and maintenance are also required. Moreover, sensitiveness, consciousness and education related to detention basin should be improved, since there are paradigms to be broken and obstacles to win. At the end of this report, it is suggested some guidelines to incorporate detention basins in new land development, not only to facilitate and instruct the use of this structure, but also to improve society’s acceptance.

Abstract:
This research aims at finding how BBC and AJE media represented the Israeli-Palestinian conflict. The study examines the news website, which reported the recent escalation of Jerusalem conflict in December 2017, where US President Trump have recognized Jerusalem as Israeli capital, and declared to move the US embassy from Tel-Aviv to Jerusalem. The study uses qualitative research, where it investigated the impact of ideology on media discourse by means of critical discourse analysis (DHA, Topoi, and Social actor’s representation). CDA was carried out for a sample of 8 news articles published on the websites of two networks: the British Broadcasting Corporation World news "BBCWN" and the Middle Eastern Qatari owned "Al Jazeera English". The articles were chosen within the month of December 2017. Articles were analyzed by means of the two-level analysis method, including the thematic and in-depth analysis. On the first, entry-level analysis, I focus on contents of texts and outline the discourse topics. While in the second phase I analyze the means of discursive strategies and the representation of social actors utilized throughout the text. The thematic analysis revealed that, both BBC and AJE have covered the incident similarly in general look. However, in-depth analysis showed that each network had portrayed the images of Israelis and Palestinians differently. On contrast of AJE, BBC tends to perceive Palestinians as a threat and the Israeli one as victims of the Palestinian violation. Moreover, the analysis revealed that each network had different ideologies and aims. The study concludes that AJE articles was biased pro-Palestinians, while BBC articles was biased pro-Israelis.

Abstract:
In this thesis we present the continuing work done examining a system in which an Andreev interferometer is used to probe the state of a flux qubit. In particular, we show that the back action of the interferometer on the qubit is low enough that an energy gap can still be observed in the qubit, and present the first experimental evidence of resonant excitation of a flux qubit detected using an Andreev interferometer. We begin by discussing the theory of flux qubits and Andreev interferometers individually. We then go on to examine what happens when with these two types of structures are combined, with particular attention being paid to the consequences for the coherence time of the qubit. We then discuss the practical elements of the experiment, notably the development of a tri-layer resists system that can be used to create high quality mesoscopic structures. We present the experimental results, which show the evidence for resonant excitation of a qubit detected using an Andreev interferometer. The quality of these resonances suggests that the system has a coherence time of less than 1ns. To conclude we examine some ways in which we believe the system can be improved in order to allow more detailed spectroscopic and time resolved measurements.

Abstract:
This research examines how companies adapt to more unpredictable environments by developing dynamic capabilities that help sustain innovation and change. The key explanatory construct is modular architecture - the intentional decomposition of systems (products or processes) into relatively independent sub-units with standard interfaces. The dissertation is structured as three semi-independent papers plus an introduction and a literature review. The literature review describes current theories of modular systems and discusses the similarities and differences to related concepts such as nearly decomposable systems, loose coupling, and vertical decomposition. The literature review identifies three gaps in the current literature. First, there has been a limited degree of theorising on the concept outside the field of technology management. Second, the current literature is based largely on anecdotal case studies and there is little quantitative evidence of the strategic value of modularity. Finally, there is a lack of understanding concerning the implementation of modular architectures. The first paper generalises from product to organisation design and derives five principles for modular organisational architectures. A hypothetical example is developed to illustrate how a large home appliances company might apply these principles to create a process platform consisting of reconfigurable building blocks. The chapter also proposes some extensions of current theory to more realistically adopt modularity concepts in organisation design. The second paper is based on a study that explores the current product and process architectures at three leading manufacturers of vacuum cleaners. It identifies some of the key barriers to increased flexibility among companies in mature industries. It also considers the relative value of modularity for established companies pursuing incremental innovation versus entrepreneurial companies introducing more radical innovations. The third paper presents the results from a survey questionnaire study of companies in the home appliances industry in the UK. and US. The relationship between market context, product and process architecture, strategic flexibility and firm performance is examined by means of structural equations modelling. The results show a positive relationship between modular product architectures and performance, with product model variety as a mediating variable. The thesis makes the following contributions to the existing literature: It complements extant theorising by generalising modular principles to organisation design. It contributes to methodology by developing a questionnaire for assessing modularity in product and process design. Finally, it provides one of the first empirical studies of the effects of modularity on strategic flexibility and firm performance.

Abstract:
This Thesis contains two parts. Part I details my work on KMOS, a new instrument for the VLT and Part II details my research on scaling relations for the massive galaxy cluster MS0451.6-0305. Part I: The K-Band Multi-Object Spectrograph (KMOS) is a new near infra-red (NIR) instru- ment to be installed at the Very Large Telescope (VLT). KMOS is capable of spatially resolved kinematics via 24 deployable integral field units (IFUs), each with a 2.8′′ field of view. I describe my contribution to the construction and optical characterisation of the spectrograph modules and instrument as a whole. Part II: GMOS-N spectroscopy has been used to obtain velocity dispersions (σ) and archival HST photometry has been used to determine the effective radii (Re) & average surface brightnesses within Re (⟨I⟩e) of 26 confirmed cluster members of MS0451.6-0305 at z = 0.55. The Kormendy Relation, Faber-Jackson Relation and Fundamental Plane have been produced for the cluster and have been compared to the results for a local reference sample in Coma. GMOS-N g′ and r′ band photometry has also been used to produce a Colour-Magnitude Relation for MS0451.6-0305. It is found that the KR and FJR disagree at the 2.1σ level with respect to luminosity evolution since z = 0.55. When correcting for size-evolution (SE), the magnitude offsets agree and are consistent with passive evolution of the galaxy stellar populations from a single burst of star formation 9.8+4.5-1.7 Gyrs ago or z = 1.7+∞-0.6. The median offset in the FP relation for the SE corrected data is also consistent with this formation epoch. Evidence is found for the evolution of the FP tilt in MS0451.6-0305, where it is shown to be steeper than that of the Coma FP, when expressed as a relation between galaxy mass and M/L.

Despite the growing stream of literature about English language education in China, there has been relatively little discussion of primary English language education. Past research has focused almost exclusively on secondary and tertiary English education. This research seeks to address this gap by examining primary English education policy. More specifically, this research provides insights into the extent to which each instance of language policy and planning can be seen as a product of its specific context. This thesis is based on an ethnographic study conducted in local education bureaus and three primary schools in Yulin, a Northwestern Chinese city with 3 million people. Drawing on critical discourse analysis (CDA) of political discourse(s), ethnographic field notes, questionnaires, interviews and classroom observation, this study investigates the influence of macro and micro levels of policy actors in disseminating and implementing the language policy. It also evaluates the apparent match and mismatch between macro policy and micro levels of practice within the local language policy and planning context. As such, I offer a lens for studying primary English education policy in China. The combination of critical discourse analysis (CDA) and ethnography represents a methodological contribution of this research and is used as a way of seeing if there are gaps between policy and practice; and, if so, what kind(s)? Thus, the main contribution of this study to the literature on language education policy is a framework for analyzing the interaction between policy and practice. This framework combines CDA and ethnography. CDA is used to try and identify underlying ideology and power relations in politically determined educational discourse, whereas the ethnographic part of the study is used to examine and interpret the implementation of policy among local education officials and teachers. Additionally, this research extends the current literature on English education policy by identifying that a lack of communication between macro and micro policy actors is a significant reason behind poor English language education quality. On the surface, local officials and teachers follow the instructions in the political discourse, due to a somewhat rigid hierarchical structure of Chinese society. In some aspects, though, local officials and teachers might veer away from, abandon or even change policy consciously or unconsciously in their daily routines and classes. Abstract:
The Congenital Dyserythropoietic Anemias (CDA) are rare hereditary hemolytic disorders with large bi- to multi-nucleated erythroblasts in the bone marrow. Hemolysis is negative in a direct antiglobulin test (DAT). Based on morphology and clinical picture, three major forms of CDAs, type I, II, and III have been defined. CDA III, dominantly inherited, constitutes the rarest type with a majority of cases belonging to a family in Västerbotten, Sweden. The genetic background of CDA I and CDA II has been linked to mutations in CDAN1 and SEC23B respectively. The mutation of CDA III has been linked to 15q22 in earlier studies. In this project we have defined the causative genetic lesion in two families with CDA III. The novel mutation KIF23 c.2747C>G (p.P916R) was shown to segregate with CDA III in the Swedish and American CDA III families and was absent in 356 healthy controls. KIF23 encodes mitotic kinesin-like protein 1 (MKLP1), which plays a central role in the last step of cytokinesis. RNAi-based knock-down and rescue experiments demonstrated that the p.P916R mutation causes cytokinesis failure in HeLa cells, resulting in increasing number of bi-nuclear cells, consistent with appearance of large multinucleated erythroblasts in CDA III patients. We conclude that CDA III is caused by a mutation in KIF23, encoding MKLP1, a conserved mitotic kinesin crucial for cytokinesis. Flow cytometry with eosin-5´-maleimide (EMA), anti-CD55 and anti-CD59 is commonly used when investigating non-autoimmune hemolytic anemias. Reduced fluorescence of EMA, typically detected in hereditary spherocytosis, is also seen in CDA II, while reduction of CD55 and CD59 characterizes paroxysmal nocturnal hemoglobinuria (PNH). We studied the flow cytometric profile of EMA, CD55, and CD59 on erythrocytes in CDA III. We found no abnormality of the erythrocyte membrane in CDA III and concluded that standard flow cytometry cannot be used to discriminate between CDA III and normal controls. In CDA I and CDA II a majority of patients, including those who are not transfusion dependent, suffer from iron overload, which, according to earlier studies, is not the case in CDA III. We found that individuals of the Västerbotten CDA III family carry mutations in the hemochromatosis (HFE) gene. Three CDA III patients with heterozygous or compound HFE mutations need treatment with phlebotomy due to iron overload. One of them carries heterozygous H63D mutation, which is not reported to lead to iron overload by itself in otherwise healthy individuals. We propose that molecular genetic testing of the HFE gene is indicated in all patients with CDA, including CDA III.

Calcium Dependent Antibiotic (CDA), is a nonribosomal lipopeptide antibiotic from Streptomyces coelicolor. It is an undecapeptide with an unusual N-terminal 2,3-epoxyhexanoyl fatty acid side chain. It contains many nonproteinogenic amino acids such as D-4-hydroxyphenylglycine, D-3-phosphohydroxyasparagien, L-3-methylglutamic acid and Z-2,3-dehydrotryptophan. CDA is very similar in structure to the antibiotic daptomycin, which under the trade name Cubicin has recently been approved for the treatment of skin infections. The work described in this thesis focuses on a number of different areas, with the overall aim of delineating the biosynthetic origin of CDA and investigating the possibility of producing modified cyclic lipopeptides through a combined chemical and biochemical approach. Strategies used in engineering the biosynthesis of CDA may then be applied to other related, nonribosomal peptides of therapeutic importance, in order to improve their biological activity.

De que forma burocracias e atores não-estatais influenciam o processo de implementação de políticas públicas? No contexto da política em estudo, ou seja, no âmbito do trabalho social em políticas de urbanização de favelas da Companhia de Desenvolvimento Habitacional e Urbano do Estado de São Paulo (CDHU), a pergunta de fundo é: como a agência situada de atores implementadores influenciou a criação de arenas participativas, num contexto organizacional-institucional adverso? Partindo desta pergunta, este trabalho analisa os diferentes perfis da burocracia implementadora, considerando suas trajetórias, valores, padrões relacionais e formas de atuação heterogêneas em três territórios urbanizados pela CDHU ao longo dos anos: Projeto Pantanal em São Paulo, Programa Serra do Mar em Cubatão e Projeto Pimentas em Guarulhos. Cada um destes três casos analisados conformou distintos cenários à implementação da política, com seus arranjos organizacionais-institucionais específicos. Os principais achados da pesquisa mostram que a construção de arenas participativas e a concretização de práticas do trabalho social se deram de forma gradual e experimental, a partir da atuação ativista dos diferentes agentes implementadores. Foram identificados quatro padrões de atuação engajada ou ativista da burocracia e oito distintos perfis ou papéis dos burocratas entrevistados. Os resultados também sugerem que a atuação criativa destes agentes é influenciada pelos variados cenários organizacionaisinstitucionais onde estão inseridos, pois conformam distintas estruturas de oportunidades ou constrangimentos à sua ação. Ainda assim, o que se percebe é que, a despeito do contexto conservador das administrações da CDHU, a burocracia implementadora, junto com atores não-estatais envolvidos na implementação da política, lançou mão de diversas estratégias de atuação no sentido de (i) criar e concretizar arenas participativas funcionais no âmbito da política e de (ii) influenciar a construção de um método de trabalho nas políticas de urbanização de favelas da Companhia, entre as diferentes áreas técnicas da CDHU.
How bureaucracies and non-state actors influence public policies implementation process? Concerning the studied case here, within social work in slum upgrading projects promoted by São Paulo State Housing and Urban Development Company (CDHU), the underlying question is: how the implementing actors\' agency could influence the creation of participatory arenas within an adverse institutional-organizational context? Starting from this question, this work analyzes the different implementing bureaucracy profiles, considering their trajectories, values, relational patterns and heterogeneous forms of action in three territories developed by CDHU over the years: Pantanal Project in São Paulo municipality, Serra do Mar Program in Cubatão and Pimentas Project in Guarulhos. These three analyzed cases conformed different scenarios to the policy implementation, with its specific organizational-institutional arrangements. The main research findings show that the development of participatory arenas and social work practices occurred in a gradual and experimental way, through the activist agency of implementing actors. Four patterns of activist bureaucratic action and eight different bureaucracy profiles (roles) were identified. The results also suggest that the creative performance of these agents is influenced by the various organizationalinstitutional scenarios in which they are located, conforming different opportunities and constraint structures for action. Nevertheless, despite the conservative context of CDHU administrations, the implementing bureaucracy, together with non-state actors involved in policy implementation, has adopted several acting strategies in order to (i) create functional participatory arenas within the policy and to (ii) influence the construction of a work methodology within CDHU slum upgrading projects, among the different technical sectors in the Company.

A dissertação utiliza a pesquisa como método experimental e investigativo para identificar os fatores condicionantes para a definição da infraestrutura de aquecimento de água para banho (gás, elétrico e solar térmico) nas edificações residenciais de interesse popular, construídas no município de São Paulo. A partir das informações de três projetos e do dimensionamento da infraestrutura de cada opção, determinam-se seus custos para um projeto típico, exclusivamente para o aquecimento de água para banho. Também são determinados os custos de aquisição, instalação, infraestrutura e operação de cada configuração. O método Life Cycle Cost Analysis (LCCA) é utilizado para comparar financeiramente diversas alternativas. Os resultados permitem avaliar o interesse na realização de infraestruturas combinadas de forma a possibilitar a qualquer momento a utilização de aparelhos instantâneos, elétrico ou a gás, com ou sem o apoio solar térmico. Essa infraestrutura combinada permite a liberdade de escolha ao usuário, de forma individual e a qualquer momento, do aquecimento de água para banho, tendo em vista o seu custo benefício, eventualmente associadas às oportunidades oferecidas pelas distribuidoras de energias e/ou pelos fabricantes de aparelhos e/ou políticas públicas. Os resultados alcançados apresentam importantes dados para subsidiar discussões e análises comparativas, estabelecendo os caminhos para orientar a escolha da infraestrutura.
The paper uses research as investigative and experimental method to identify the factors responsible for the definition of infrastructure for bathing water heating (gas, electric and solar thermal) in residential buildings of popular interest, built in São Paulo. Based on the information from three projects and the sizing of the infrastructure of each of them, the costs for a typical project exclusively for bathing water heating, are determined. Besides that, are also determined, the costs of acquisition, installation, infrastructure and operation of each configuration. The method - Life Cycle Cost Analysis (LCCA) is used to compare the various financial alternatives. The results allow to evaluate the interest in carrying out infrastructures combined to enable anytime instant use of appliance, electric or gas, with or without the solar thermal support. This combined infrastructure allows the user free choice, individually and at any time, for heating water for bathing, in view of its cost benefit, possibly combined with the opportunities offered by energy distributors and / or the device manufacturers and / or public policy. The results present relevant data to support comparative discussion and analysis, setting ways to guide the choice of infrastructure.

Rapping has long been used by people who are from the margin of society as a way to give a voice (Campbell, 2005; Ibrahim, 1999). As a member of the marginalized group and as the first and only Asian who claimed a seven-time victory on Freestyle Friday on Black Entertainment Television (BET), Jin Au-Yeung has received a noticeable amount of attention. At the same time, he has faced a lot of unfavourable experience as an Asian rapper in American society. This study employs Fairclough’s (1989) model of CDA approach to find out how Jin constructs his identity and establishes his ideology through his lyrics, and how his construction of identity and establishment of ideology reflect the social practice in American society. Fifteen songs written by Jin were chosen for the analysis according to the three interrelated stages in CDA: description, interpretation and explanation. Results show that Jin constructs his personal identities as a professional rapper and as a Chinese American and establishes his ideology of having one human nation despite the difference in races through his rap lyrics. These are achieved through the co-occurrence of “I” and “to be”, and promoted through the use of rhyming and code-switching. It was also interpreted that Jin’s personal identities and ideology are shaped through the social ideology on Asian Americans, which is probably reflected through the social practice in American society.

Due to projectedincrea ses in air traffic, there are several research efforts underway to evaluate ways to safely increase the capacity of the National Airspace System (NAS), improve operational efficiency, andre duce aircraft noise. At NASA Langley Research Center (LaRC) in Hampton, Virginia, two parallel research efforts have focusedon terminal area research: one is Airborne Precision Spacing (APS), and the other is the Quiet Aircraft Technologies (QAT) project. The APS objective is to increase terminal-area capacity without adversely affecting safety, whereas the QAT project objective is to develop noise- and fuel-efficient approach trajectories. The APS project developed a cockpit tool, called Airborne Merging and Spacing for Terminal Arrivals (AMSTAR), that issues speedco mmands to aircraft to maintain desired spacing between aircraft pairs. The APS studies showed an ability to increase runway capacity; however, capacity increases may negatively impact noise andemissio n levels in airport areas. The QAT project created efficient Continuous Descent Approaches (CDAs), which showedred uctions in aircraft ground noise and fuel consumption. Previous research has shown that CDA trajectories have adverse effects on runway capacity because aircraft must be spacedf urther apart at long distances from the runway to prevent separation losses at the runway threshold. To date, the APS and CDA concepts have been evaluated independently at LaRC.In this study, three different approaches to combining APS and CDA operations were evaluatedto determine the feasibility and benefits of combining these concepts. These methods combined AMSTAR with 3◦-flight-path-angle-CDA approach routes, 3◦-CDA routes with spoilers, and2 ◦-CDA routes without spoilers. Adding the use of spoilers allowedf aster responses to large speedr eductions issued by AMSTAR. This improvement was contrastedwith the effects of a shallower flight-path angle for greater deceleration capabilities. This research indicated that AMSTAR improved the performance of CDA operations, although full capacity improvements were not achieved. Whereas the 2◦-CDA routes were expectedto show the best results, the 3◦-CDA case with spoilers showed the least variability in thresholdspacing errors. All of the CDA routes were more noise, fuel, and time efficient than traditional step-descent routes that are commonly usedto day.

Denna C-uppsats granskar OmVärlden som behandlar bistånd, utveckling och globala frågor och som ställs till förfogande av Sida (Styrelsen för internationellt utvecklingssamarbete). Studien grundar sig i ett övergripande intresse för hur medierna påverkar samhällets uppfattning om verkligheten och för studien relevant, postkoloniala föreställningar om maktrelationer och strukturer i utvecklingsländer. Studien innefattar en förstudie i kombination med kritisk diskursanalytisk metod vilken appliceras på fem skilda reportage ur OmVärlden. Syftet är vidare att se hur journalistiska texters uppbyggnad bidrar till att konstituera samhällets föreställningar om rådande hegemoniska strukturer. För att definera studiens problemområde ställs frågor om reportagens dominerande teman och aktörer samt representationen av genus. Detta följs sedan av en utredning av Sidas eventuella röst och påverkan vad gäller OmVärldens redaktionella innehåll. För studien relevant har ett flertal teorier och begrepp behandlats, bland annat postkolonial teori (som berör kritisk granskning av hierarkiska maktrelationer) och intersektionalitet (som bidrar med att skapa förståelse för hur simultana och globala maktrelationer påverkar föreställningar om ojämnlikhet). Efter redovisning av samtliga anlyser och med stöd från behandlade teoretiska områden, gör vi tolkningen att reportagens tematiska och diskursiva sammansättning styr de postkoloniala maktförhållandenas produktion och reproduktion. Vidare konstaterar vi att Sidas röst framkommer genom, och därmed påverkar OmVärldens redaktionella innehåll. Vi drar följdaktligen slutsatser som grundar sig i analysens resultat om textsammansättningens utformning och betydelse. Vi tolkar resultaten som att OmVärlden explicit bidrar med en betydelsefull och nutida angelägen kunskap om utvecklingsländer och bistånd, vilket i enhet med Sida verkar för utveckling inom området. Tidningens implicita bidrag till en ideologisk omvärldsförändring tolkas dock som begränsad. Avslutningsvis behandlas ett flertal aspekter vad gäller fortsatt forskning inom ämnesområdet.

Nineteen cultivars and fourteen breeding lines were evaluated for partial resistance to crown rust Puccinia coronata race 264. Multivariate statistical methods such as principal component and cluster analyses were employed to identify significant resistance parameters and to group oat genotypes with similar rust resistance characteristics. This involved two separate investigations consisting of two experiments each, the first experiment conducted under field conditions and the second conducted under growth bench conditions. From both of the investigations a group of oat genotypes with high partial resistance to P. coronata race 264 was obtained. These are OA 712-17, OA 712-33, Glen, Woodstock, QO 220.13, and QO 574.21. These oat genotypes are currently being used as parents in crosses in the Macdonald Campus of McGill University oat breeding program.

O principal objetivo deste trabalho é avaliar a qualidade de um site de governo eletrônico. Para atingir essa meta, a pesquisa descreve a aplicação da ferramenta Quality Function Deployment, QFD, para melhoria da qualidade do site da Secretaria de Estado da Habitação de São Paulo, SH, e Companhia de Desenvolvimento Habitacional e Urbano, CDHU. CDHU é a empresa do governo do Estado de São Paulo responsável pela execução do plano de habitação de interesse social para o Estado. O QFD foi desenvolvido no Japão, na década de 60, para melhorar a qualidade de produtos industriais desde o projeto. Apesar disso, o foco deste estudo é sua aplicação para melhorar a qualidade do site, apresentando as adaptações necessárias ao modelo original e observando as possíveis limitações de sua aplicação para esse fim. Como ponto de partida para a pesquisa, foram realizadas entrevistas com os profissionais de informática da empresa, responsáveis pelo desenvolvimento e manutenção do site, com outros colaboradores que utilizam diariamente o site da companhia e com o ouvidor da CDHU. Essas entrevistas, juntamente com a revisão bibliográfica, formaram a base para a elaboração de um questionário para avaliar a percepção dos clientes da CDHU sobre o site. O questionário foi aplicado a mutuários da CDHU para captar a voz do cliente e construir a casa da qualidade, principal produto da aplicação do QFD. O QFD mostrou ser uma ferramenta útil para priorização das mudanças a serem feitas no site e para iniciar discussões sobre a necessidade de sistematizar rotinas de teste e de melhoria contínua do site, além da gestão do conteúdo. Este trabalho abrange os temas Qualidade da Informação e Governo Eletrônico e apresenta sugestões de melhoria para o site da SH/CDHU.

To achieve that goal, this paper describes the advantages of using the Quality Function Deployment (QFD) tool to improve the quality of Secretaria de Estado da Habitação de São Paulo, SH, and Companhia de Desenvolvimento Habitacional e Urbano, CDHU, website. CDHU is a governmental company of São Paulo state which is responsible for developing urban plans focused on low revenues families. QFD was developed in Japan, in the 60s, to improve the quality of industrial products from the project. Even tough the focus of this study is the use of such managerial tool as a website quality inducer, presenting the necessary adjustments to original QFD model and pointing out some possible limitations of such application. First of all, interviews were conducted with technology information professionals from CDHU, who are in charge for the website development and maintenance; with CDHU workers who access company website daily; and with CDHU ombudsman. These interviews along with bibliography review allowed the development of a questionnaire to evaluate CDHU customers perception on its site quality and performance. The answers captured the voice of the customer and were used to build the house of quality, main deliver of QFD. QFD demonstrated to be powerful tool to help prioritizing desirable changes in the website and also to start PDCA discussions about the site content management. This study covers the Information Quality and E-Government subjects and it also presents improvement suggestions to SH/CDHU website as an answer to clients willing and recommendations.

Energy efficiency has become an important feature in the design of process plants due to the rising cost of energy and the more stringent environmental regulations being implemented worldwide. In South Africa as in other African countries, most of the chemical plants were built during the era of cheap energy with little emphasis placed on energy efficiency due to the abundance of cheap utility sources such as coal and crude oil. In most of these plants, there exists significant potential for substantial process heat recovery by conceptual integration of the plant’s heat exchangers. Pinch Technology (PT) has been demonstrated to be a simple and very effective technique for heat integration and process optimization. This study applies the PT approach to retrofit the heat exchangers network of the Crude Distillation Unit (CDU), of a complex petroleum refinery with the aim to reduce utilities requirement and the associated gaseous pollutants emission. This objective is accomplished by firstly conducting an energy audit of the unit to scope for potential energy saving. The existing Heat Exchanger Network (HEN) was re-designed using the remaining problem analysis (RPA) to achieve improved process energy recovery while making maximum use of the existing exchangers. The aim is to maintain the existing plant topology as much as possible. This network was later relaxed trading heat recovery with number of heat transfer unit so as to optimize the capital cost. These were implemented in AspenPlus v7.2 environment. The cost implications of the retrofitted and evolved networks including the capital and operating costs were determined on a 5 years payback time basis. The Problem Table (PT) analysis revealed that the minimum utilities requirements are 75 MW and 55 MW for the hot and cold utilities respectively. Compared to the existing utilities requirements of 103 MW for hot utility and 83 MW for cold utility, this represent a potential savings of about 26 % and 33 % savings for the hot and cold utilities respectively. The target utilities usage in the re-designed network after applying Remaining Problem Analysis (RPA) was found to be 55 MW for the cold utility and 75 MW for hot utility. The relaxed HEN required a cold utility of 62.5 MW and hot utility of 81 MW. From the total cost estimation, it was found that, although an energy saving of 34% can be achieved by the re-designed network before relaxation, the capital cost, US$ 1670000 is significantly higher than for the existing network (about US$ 980000). The final relaxed network gave an energy saving of 34% and with total cost of US$ 1100000. It was recommended from the study after cost comparisons of the four different networks (the original network, the MER network, the relaxed network and a grass-root design) that the best network for the retrofit purpose was the relaxed HEN, because there is no major shift in deviation from the topology of the original network. From the analysis it was found that a 34% saving in energy cost could be achieved from this retrofit. The Total Annual Cost (TAC) for this network gives credence to the fact that this retrofit which applied the rules of pinch analysis can bring about real saving in energy usage.

With the new challenges of the digital world associated with disinformation, data breaches, and cybercrimes (Cadwalladr & Graham-Harrison, 2018; Connolly et al., 2016; Shipley & Bowker, 2013) many countries nowadays discuss approaches to Internet regulation. In Ukraine, which faced propaganda tactics employed by Russia as a part of the Ukrainian-Russian conflict (Yurkova, 2018), the need for information security in recent years became a major challenge. In 2017, authorities of Ukraine addressed the challenge by introducing sanctions to the most-used Russian web-platforms, including social networks Vkontakte and Odnoklassniki, search engine Yandex, and many other services. This study analyzed the discourse that was formed around the decision. The work incorporates the model of critical discourse analysis by Fairclough (1995a) and framing analysis by Pan & Kosicki (1993). This qualitative study analyzed materials from Ukrainian online media, TV stories, user comments, and political speeches from 16th-17th May 2017 – the dates when the decision on blocking of Russian web-sources was announced to the Ukrainian public. The research answers the questions about the dominant discourses in society regarding the approach of the authorities to Internet regulation, the main arguments and counter-arguments, and media framing. The results show that the discussion was locked in a trap of two dominant discourses of freedom of speech and national security, whereas alternative measures to restrictions were not represented on the public agenda.

There is growing interest in the impact of development controls on the development process. Many researches focus on whether development controls offer certainty and flexibility to developments. Nevertheless, not many researches explore the causes leading to delays in developments. Through examining the length of time spent in the stages of planning, lease and building controls of 57 completed Comprehensive Development Area (CDA) sites where the development processes were started and completed in the Metro Planning Committee Area in Hong Kong between 1990 and 2014, this paper explores if there are any factors accounting for the delay in the development process of the completed CDA sites. Besides, this research examines the relationships between the length of time involved in the development process and the characteristics of the completed CDA sites. Meanwhile, this research also examines 67 approved planning applications submitted for uncompleted CDA sites where the development processes have not been completed in order to explore the relationships between the stage of planning control of CDAs and particular variables. Through quantitative statistical analyses, this paper confirms that the stage of planning control does not cause a delay in the development process of the completed CDA sites. Approval rate of planning applications does not necessarily affect the development process of them. Instead, the relatively lengthy amount of time spent in the development process in the completed CDA sites can be stemmed from the change of mind of the planning applications’ applicants. The GFA of completed CDA sites affects the length of time spent in the development process. In addition, when the year of the first approved planning application of completed CDA sites is closer to year 2014, the length of time spent in the whole development process is less. For the 67 approved applications submitted for the uncompleted CDA sites, it is confirmed that the GFA of the proposed developments cause impacts on the length of time a planning application involved to get an approved decision from the planning authority. Based on the findings collected, it is concluded that the stage of planning control does not cause a delay in the completed CDA sites and, various factors play a part accounting for the length of time spent in the development process and in getting approved decisions from the planning authority.
