My thesis research began with a project in which we were trying to determine the function of embryonic stem cell (ESC)-specific miRNAs. Using luciferase constructs containing miRNA binding sites, luciferase expression was inhibited by endogenous miRNAs in ESCs, and by exogenous miRNAs in HeLa cells. Inhibition of luciferase expression by miRNAs was inhibited in HeLa cells using 2’O-methyl-oligonucleotides. Abstract Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) is now used as a routine technique for the fast and reliable identification of fungi at the species level and, currently, it represents an important phenotypic methodology based on proteomic profiles. The main limitations to MALDI-TOF MS for fungal identification are related to sample quality (e.g. quality of biological material such as rigidity or pigmentation of cell walls), sample preparation (e.g. the myriad of sample preparation methodologies that deliver different data sets to different MALDI-TOF MS databases) and the databases themselves (e.g. the 'black-box' commercial databases). This chapter presents an overview and discussion of the use of MALDI-TOF MS for fungal identification. The major known limitations of the technique for fungal taxonomy, and how to overcome these, are also discussed.

Fixation is vital for preserving tissue integrity for histological examination, aiming to maintain tissues in a lifelike state. Immediate fixation, post-surgery or autopsy, is crucial to prevent autolysis. Tissues are prepared for transformation into thin microscopic slices following fixation. One important method is to embed tissues in paraffin that matches their density. Using a microtome, paraffin-embedded tissues are cut into micrometer-thin slices, with paraffin serving as support. However, in order for water- and alcohol-soluble dyes to penetrate, paraffin must be eliminated during staining. The Hematoxylin and Eosin stain, commonly used in histology, gives distinct coloration to cellular components. Hematoxylin, which binds to nucleic acids, stains cell nuclei bluish-purple. Eosin, binding to cytoplasmic elements, imparts a pink hue to the background. Hematoxylin requires a mordant to enhance staining affinity. Mordants, high-molecular-weight metal salts, form strong bonds with tissues and dyes, ensuring effective staining.

Here, we developed two advanced surface engineering technologies, overcoming the above limitations, to prepare a black TiO2 photoanode that achieved the maximum photocurrent density reported to date. Moreover, we theoretically and experimentally revealed the formation mechanism of black TiO2 and its enhanced photoelectrochemical (PEC) performance. These surface engineering technologies are not only suitable for the preparation of efficient photoanode materials for PEC hydrogen production but also play a beneficial and promoting role in the research and development of new materials for hydrogen fuel cells and hydrogen storage.

Yeasts are ubiquitous and will be present in various types of patient specimens and patient populations. This chapter includes the most common yeasts that cause human disease and also yeast-like fungal organisms that may cause human disease. Examination of yeast preparations will show round to oval budding cells (blastoconidia). Some cells may be elongated, forming pseudohyphae, and a few species will show actual arthroconidia and true hyphae (Geotrichum and Trichosporon). Cryptococcus spp. or Rhodotorula spp. may produce capsules. Yeasts are best grown at 25 to 30ᴼC on Sabauraud's dextrose or Sabhi agar and often grow on primary bacteriological media without inhibitors. The addition of cycloheximide will inhibit many yeast strains. Yeast typically grows as cream-colored but may also be white, yellow, tan, orange, red, or dematiaceous (brown or black pigment). The two most medically common and significant yeasts are Candida albicans and Cryptococcus neoformans. Candida auris is now a top concern for hospitalized patients due to its excellent resistance to antifungal medications.

Herein, we summarized the latest progress of COFs as solid ion conductors in energy devices, especially lithium-based batteries and fuel cells. The design strategies and performance in terms of transporting lithium ions, protons, and hydroxide anions are systematically illustrated. Finally, the current challenges and future research directions on COFs in energy devices are proposed, laying the groundwork for greater achievements for this emerging material.

Protease nexin I (PN-1) is a cell-secreted protease inhibitor which modulates the activity of certain serine proteases in the extracellular environment. To probe PN-1 functions, we prepared a panel of monoclonal antibodies (mAbs) against it and studied their effects on a variety of PN-1 activities. Three highly potent inhibitors of PN-1 activity have been characterized. MAbs pl-6 and pl-9 stoichiometri-cally block PN-l-mediated anti-thrombin and anti-urokinase activity.
