Gale, Zoe. "Glial cell line-derived neurotrophic factor effects on dental pulp cells and osteoblast-like cells." Thesis, University of Birmingham, 2012. http://etheses.bham.ac.uk//id/eprint/3268/.

Abstract:
Glial cell line-derived neurotrophic factor (GDNF) is a growth factor promoting survival, proliferation and differentiation of neural crest cells. Neural crest cells play an important role within mesenchymal tissues during dental pulp and calvarial bone development. GDNF also has a role within non-neuronal tissues and is expressed during dental development. GDNF null mutations prevent the formation of the mineralised hard tissues of the tooth. The hypothesis for this study was that GDNF affects mesenchymal dental pulp cells (DPC), promoting the regenerative responses of mineralised tissues. Th

Abstract:
The basic helix-loop-helix protein, activated B-cell factor-l (ABF-l), is a transcription factor involved in cellular proliferation and differentiation. ABF-1 shows patterns of expression that are stimulated in activated B cells, which may suggest ABF-1 is involved in the regulation of B-cell differentiation. To investigate the functional role of ABF-1, we have generated a human plasmacytoma cell line with inducible expression of ABF-1. We were able to induce expression of ABF-1 by using the tetracycline repressor system (teton/off). With addition of tetracycline we were able to stimulate the

UV radiation induces two of the most abundant mutagenic and cytotoxic DNA lesions such as cyclobutane–pyrimidine dimmers (CPDs) and 6-4 photoproducts (6-4PPs) and their Dewar valence isomers. However, cells have developed a number of repair mechanisms to counteract the DNA damage caused by UV. Although skin cancer is a common in Taiwan, the pathogenesis is still incompletely studied. In this proposal, we would like t

Beta-secretase (BACE1) that cleaves amyloid precursor protein (APP) to produce the amyloid 42 peptide fragment, ultimately resulting in the formation of amyloid plaques, plays a key role in the pathogenesis of AD. Thus, BACE1 is an attractive target for the development of inhibitor drugs to treat AD. Nowadays numerous in vitro BACE1 activity assay are commercially available. Our current efforts focused on the establishment of a FRET-based scr

A mouse L cell mutant termed gro29 has been isolated (Tufaro et al. 1987) and shown to be defective in the growth of two enveloped viruses, vesicular stom- atitis virus (VSV) and herpes simplex virus type 1 (HSV-1). In this thesis it is demonstrated that the rate of transport and processing of HSV-1 glycoproteins was impeded in infected gro29 cells. In addition, HSV-1 virions failed to exit the cell and accumulated in cytoplasmic vacuoles. The phenotype of the uninfected gro29 cell was examined. It was determined that gro29 cells were resistant to the lectins ricin and modeccin, and sho

The cause of SCA8 was originally proposed associated with CTG trinucleotide repeat at 3’UTR (untranslated region) of ATXN8OS gene, lying on chromosome 13q21. However, recent studies suggest that bidirectional transcription of ATXN8OS occurs, with its anti-strand, ataxin8 (ATXN8), which encodes a polyQ protein in the CAG orientation, and ATXN8OS transcribed into potentially pathogenic CUG transcripts. SCA8 may thus has both RNA and protein gain of function mecha

碩士
臺灣大學
微生物學研究所
98
Oral squamous-cell carcinoma (OSCC) is common worldwide with low survival rate after therapy. Previously, we found elevated Treg, Th17 and a subset of IL-17+ Foxp3+ tumor infiltrating lymphocytes (TILs) in OSCC by immunohistochemistry and flow cytometry. However, the origin of these subsets was unclear. In this study, a mixed CD4 T cells-tumor cells coculture system was established to delineate the induction and/or expansion of the IL-17+ Foxp3+ T cells. The percentage of either Foxp3+ or IL-17+Foxp3+ T cells was increased after co-cultured in the presence of an
