Full textENGLISH ABSTRACT: The terms business model and business model innovation have only gained prominence over the past two decades, whilst little of the research has been applied in a South African context. The main objective of this thesis was to ascertain the reasons for success of a local business through the research and application of the themes of business models, business model innovation and blue ocean strategy to a local organisation. The subject matter of the thesis was Van Loveren Family Wine Cellar, a family-owned South African wine farm that has seen significant success over the past 15 years in a highly competitive and fragmented local market. To complete this process, it was also necessary to gain an understanding of both the global and local wine industries, and how South Africa has grown within this environment. The format of the thesis is a case study, selected because of its relevance to gaining a rich understanding of the context of the research and the processes enacted, thereby facilitating the main objectives. The case study approach also provides a tool to aid in the education of business strategy students, allowing the principles to be applied to new scenarios. The findings of the study include that there has been a shift in both consumption and production of wine from the traditional Old World to the innovative New World. This shift is being driven by new consumers of wine who are less concerned with the mystique and legacy of Old World wine, instead demanding wines that fit their lifestyles through easy drinking taste profiles with an ease of selection. This phenomenon is present in South Africa, where growth in the market is seen at lower price points. New consumers who are selecting wines that compete with existing alcohol alternatives as gateway products are driving this demand. Van Loveren launched their Four Cousins Rosé in 2000, with exponential growth experienced between 2004 and 2008, driving the product to be one of South Africa’s best-selling brands. The findings of the study were that the Van Loveren Family Cellar had positioned itself, through business model innovation, to capture the growth in new wine drinkers. Van Loveren had built a business model that provided a framework within which strategy can take on competitors. Core to their success were the long-term relationships with key buyers and the collaborative partnerships that allowed Van Loveren to utilise external capabilities as part of their own business. The success was also based on the leap of faith to enter the market in a sweeter taste profile, through the development of a brand that resonated with consumers, and in packaging that provided an attractive price point. The combination of these decisions allowed Van Loveren to capture uncontested market and grow exponentially.

Full textENGLISH ABSTRACT: The wine industry significantly contributes to South Africa's water demand and subsequent pollution of the limited resource. Wastewater is produced throughout the year with an increase in volume and organic load during the vintage season. Anaerobic digestion (AD), specifically the upflow anaerobic sludge bed (UASB) technology has been shown to be feasible in the treatment of cellar wastewater. However, the legal standard for chemical oxygen demand (COD) for disposal in a natural water resource (75 rnq.L") is often not met. The aim of the study was to conduct a laboratory-scale investigation into the feasibility of combining pre- and post-ozonation processes with AD in order to achieve a final COD closer to the legal disposal limit. While acclimatising an UASB bioreactor containing mixed anaerobic granules to a cellar wastewater with a pH set at 8.0, stable-state conditions were not reached. Sucrose additions to the substrate, increased substrate loads, heattreatment of the substrate and an addition of isolated cellar effluent bacteria to facilitate degradation prior to AD, were all unsuccessful in maintaining stable-state in terms of COD removal efficiency. Once the substrate pH was re-set to 7.5, the reactor stabilised. The lowest efficient operational pH was found to be 5.73 resulting in a COD removal of 88% at a substrate COD < 5 000 rnq.L". At a substrate pH of 6.0, the lowest efficient operational hydraulic retention time (HRT) and corresponding organic loading rate (OLR) were 19.7 hand 9.75 kg COD.m-3d-1 , respectively, with the COD removal being maintained around 84%. The reactor effluent still had a final COD of 1280 rnq.L", which was well above the legal South African limit. Dominant bacteria were isolated from raw cellar wastewater and identified as Acinetobacter haemolyticus, Burkholderia cepacia and Cryseomonas luteola. In order to investigate the possibility that ozonation improved biodegradability, the growth of the isolates at 35°C was monitored over 24 h in sterile ozonated and non-ozonated substrates from the vintage and non-vintage seasons. All the isolates increased by at least 1.5 log cycles in the control substrates from both seasons. Ozonation of the wastewater batches for 10 min at a rate of 73 rnq.L" led to slightly increased growth of the inoculants in the substrate batch from the vintage season. For the substrates from the non-vintage season, ozonation had an inhibitory effect on the bacterial growth. A 5 min ozonation treatment at a concentration of 73 rnq.L" was found to be optimal for both a pre- and post-treatment to UASB-treatment of cellar wastewater. Both UASB treatment and ozonation were effective in reducing the COD by 85% and 20%, respectively. The COD reduction was improved to 88% when UASB treatment was combined with post-ozonation. The total reduction in total suspended solids (TSS) for the combined process was 97%, compared to 80% for UASB and 73% for an ozone treatment alone. The reduction for volatile suspended solids (VSS) was 98% compared to 81% for UASB and 73% for the ozone treatment alone. The total reduction when using a pre-ozonation UASB treatment combination was an average of 86% for COD. The TSS and VSS were both reduced by 95%. Biogas production increased from 1.4 L.d-1 to 3.8 L.d-1 when an ozonated wastewater was used as substrate. When the UASB treatment was combined with both a pre- and post-ozonation treatment process, the COD was reduced by 89% while TSS and VSS were both reduced by 99%. This study showed that pre- and post-ozonation treatment processes could successfully be utilised to improve UASB treatment of cellar wastewater. Although the legal limits for discarding into a natural resource were not met, significant progress was made in reducing COD levels. Cellar wastewaters do however, vary according to season and the wastewater composition could affect the efficiency of a pre-ozonation process.
AFRIKAANSE OPSOMMING: Die wynindustrie maak "n beduidende bydrae tot die eise wat aan Suid-Afrika se waterbronne gestel word en gevolglik die besoedeling van die beperkte hulpbron. Afloopwater, wat in volume en organiese lading gedurende die parstyd toeneem, word reg deur die jaar opgelewer. Anaërobiese vertering (AV), spesifiek die "Upflow anaerobic sludge blanket" (UASB) tegnologie, is alreeds suksesvol gebruik om kelderafloop te behandel. Die wetlike vereiste vir chemiese suurstof behoefte (CSB) vir storting in "n natuurlike hulpbron (75 rnq.L"), word egter dikwels nie bereik nie. Die doel van die studie was om in "n laboratorium-skaal ondersoek AV te kombineer met voor- en na-osoneringsprossesse, om sodoende te poog om "n CSB nader aan die wetlike standaard te verkry. Die vermindering in totale gesuspendeerde vastestowwe (TGV) vir die gekombineerde proses was 97%, in vergelyking met 80% vir UASB- en 73% vir osoonbehandeling alleen. Die vermindering in vlugtige gesuspendeerde vastestowwe (VGV) was 98% in vergelyking met 81% vir UASB- en 73% vir osoonbehandeling alleen. Full textThis study examines the validity of the mystery shopper research method, which was applied in the context of wine tourism. In the process of achieving this objective, the service quality of selected wine estates along the Stellenbosch Wine Route (SWR) was also established. Furthermore, the findings reveal whether this research method is beneficial to facilitate customer relationships. Wine tourism is considered a special interest tourism that has received heightened global attention in the past decade, both practically and academically. This growth is also mirrored domestically, as wine tourism is one of the major attractions in the Western Cape. Since 2006, the number of wine estates in the Stellenbosch region rose by 12.3%, resulting in more than 140 wine estates that are currently available. This ultimately means that there are more wine estates, which all compete for the same customer base, and this increases competitiveness. Moreover, being a relatively homogenous product offering, with the key differential point being the actual winescape, it is important that wine estates focus on delivering superior service quality in order to achieve competitive advantage. This also calls for a need to systematically test service quality and procedures to ensure that desired levels are maintained. The research methodology that was used is exploratory in nature, comprising of two in-depth interviews and sixteen mystery visits via judgmental sampling. The mystery visits were conducted at the Stellenbosch Hills and Bottelary Hills sub-routes. One popular and one less recognised wine estate per sub-route were observed. Findings showed that service quality at these estates are generally high, yet experiences at all estates were vastly different. The most important discovery is that the concept of mystery visitors is, in fact, an appropriate vehicle to assess service quality and to facilitate customer relationships. This method was successful in identifying the source of problems that led to sub-optimal performance, which allowed for corrective action to be taken. As a result, the service quality of the estates can be further increased to ensure a perfect experience on each occasion, which, over time, builds the reputati~n of the wine estate. Ultimately, this research has demonstrated that mystery visitors can be applied in the context of tourism on both an individual and collective scale. Full textENGLISH ABSTRACT: The alcoholic beverage industry needs objective analysis of grape juice and liqueur quality. Fourier transform infrared (FT-IR) spectroscopy with multivariate data analysis techniques is widely used in wine laboratories across South Africa for accurate, fast and high sample throughput analyses. In this study the potential of FT-IR spectroscopy is evaluated for the quantification of ammonia in freshly pressed grape juice. FT-IR spectroscopy is evaluated, using two different spectrometers, in respectively attenuated total reflection (ATR) - and transmission scanning modes for the quantification of alcohol, pH and invert sugar in spirit-based liqueurs. The ultimate aim was to implement the PLS regression algorithms developed at an industrial cellar and replace the complex and lengthy reference methods used at the time of this study. Principle component analysis (PCA) was performed prior to the calibration step to identify groupings and patterns within the spectra. The PLS calibration models were developed from samples collected at the cellar and using partial least square (PLS) regression. The models were evaluated using the performance criteria coefficient of determination (R2) and root mean squared error of cross validation (RMSECV) at calibration stage, and root mean square error of prediction (RMSEP) and residual predictive deviation ratio (RPD) at validation stage. The average RMSEP (1.88 mg/L) of the ammonia PLS calibration model was in agreement with the standard error of laboratory (SEL = 1.54 mg/L). The R2 (92.05) and average RPD (3.3) proposed a model with excellent precision for screening purposes that was ready to be transferred for use by the laboratory. Full textENGLISH ABSTRACT: The goal of this study was to investigate the feasibility of a small-scale biomass gasification system within the context of a cooperative wine cellar operation in the Western Cape of South Africa. Central to this goal was the questions whether the time for the implementation of such small-scale renewable energy technologies in South Africa has arrived, in light of the status quo which has changed drastically from the days of abundant and cheap coal-based electricity, and whether the new opportunities afforded by the CDM can help foster financial feasibility. There are various macro-drivers contributing to the current-day emphasis on renewable energy and cogeneration projects. The first and most pressing driver is the global climate change imperative, while the others include the increased aspiration of countries towards energy security, the realization of the importance of sustainable development and the subsequent renewable energy policies that falls within the ambit of sustainable development. Small-scale biomass gasification technology still poses some challenges, particularly when it comes to the gasification of agricultural residues, as with grape residues in the case of this project. The most important technical feasibility problem to overcome is the low ash agglomeration temperature of grape residues reported in the literature. Although the local equipment manufacturer foresees no problem in this regard with their system design - since they have conducted tests on sunflower seed residues, which have similar ash properties, without experiencing any agglomeration problems - the seriousness of this aspect will be confirmed during pilot trials. In order to be eligible for emissions reduction trading under the CDM, the project must adequately demonstrate that the emissions reductions are additional to the business-as-usual scenario, in both the environmental and financial sense. The project will satisfy the environmental additionality requirement, since in its absence the wine cellar will continue its full reliance on coal-based electricity from the grid. Financial additionality, in its strictest sense, requires for the project to be infeasible if it does not pursue CDM participation, which the financial feasibility modeling results indicated to be true in this case. A feasibility model was developed to - subject to the input parameter values assumed and basic assumptions made - be able to assess the financial viability of the project. The main assumptions were that the private feed-in into the national electricity grid was available to all IPPs, in spite of the fact that in its Medium Term Power Purchase Program me (MTPPP) of May 2008 Eskom had only requested expressions of interest from IPPs that had a generating capacity of 5MW and higher to install private base load capacity. This assumption is commensurate with the country's renewable energy targets and the official government aim of achieving a 30% contribution from IPPs to the national electricity mix. The second assumption was that the 65 to lOOclkWh offered by Eskom in its MTPPP will be applicable to sub-SMWe IPPs as well (Creamer, 2008), and that this lower bound electricity price of 65clkWh can be applied over the whole project lifetime. Inherent to this assumption is the further supposition that the progressive decline of electricity prices to an eventual level of 35clkWh by 2018 foreseen by Eskom will not materialize, due both to the sustained pressure an expanding South African economy will put on the considerable but time-consuming supply-side initiatives launched by Eskom, and the likelihood for price premiums to be introduced for clean electricity in order to meet the country's renewable energy targets. From the assumed input parameter values the initial capital and COM expenditures, operating revenues and costs over the project lifetime were determined, and then used to calculate the net cash flows, where after the NPV was computed to serve as the deciding criterion on financial feasibility. A discount rate of 18% was assumed, corresponding with the subjectively judged risks that the project posed as a small-scale renewable energy system within the wine cellar operations. In the case where the project excluded all CDM aspects, the NPV was negative at - ZAR342 573, but this improved to ZAR325 193 if the project participated in the CDM. Thus it was concluded that the project will only be financially feasible if it includes CDM participation, and that this positive contribution can be leveraged by pursuing a programmatic CDM approach. This entails the development of this project as part of a larger program in which similar projects are implemented as they arise, up to the official UN limit of 15MWe in total to still qualify as a small-scale program. The nature of the assumptions that form the backbone of this study indicate that the positive financial feasibility result in the case where the project includes COM participation will become practically relevant only over the short- to medium-term as these assumptions become reality in South Africa. Therefore, although the time for such small-scale renewable energy projects has clearly not arrived as yet, it would seem that it is indeed around the corner.
AFRIKAANSE OPSOMMING: Die mikpunt van die navorsing was om die lewensvatbaarheid van 'n klein-skaal biomassa vergassing sisteem binne die konteks van 'n kooperatiewe wynkelder in die Wes-Kaap van Suid-Afrika te ondersoek. 'n Sentrale aspek was die vrae of die tyd aangebreek het om klein-skaal volhouhare energie projekte in Suid-Afrika tot uitvoering te bring, gesien in die lig van 'n status quo wat drasties weg beweeg het van die tydperk toe elektrisiteit volop en goedkoop was, en of die geleentheid wat die CDM bied sulke projekte finansieel lewensvatbaar kan maak. Daar is verskeie eksterne makro-drywers wat bydrae tot die huidige fokus op sulke volhoubare energie projekte. Hieronder tel globale klimaatsverandering as die dringendste drywer, terwyl die res onder andere die hernuwe strewe van lande tot verbeterde energie-sekuriteit, die besef van die belangrikheid van volhoubare ontwikkeling en die daaropvolgende volhoubare energie beleid stappe insluit. Daar bestaan nog etlike tegniese uitdagings tot die toepassing van klein-skaal biomassa vergassing tegnologie, spesifiek met betrekking tot die vergassing van afval landbou byprodukte, soos wat die geval is met druiwe afval in hierdie projek. Die belangrikste tegniese uitvoerbaarheids-aspek wat aandag verg is die lae smeltpunt van druifafval-as wat in die literatuur rapporteer word. Alhoewel die plaaslike toerustings-vervaardiger nie enige probleme in hierdie verhand voorsien nie, aangesien toetse met sonneblom afval - met as-eienskappe rofweg identies aan die van druiwe-afval - geen smelting van die as getoon het nie, sal dit tog nogsteeds aan verdere toetse onderwerp word. Vir die projek om te kwalifiseer as geskik vir verhandeling in kweekhuisgas vermindering deur middel van die COM, moet dit voldoende bewys kan word dat alle uitlaatgas vermindering addisioneel is tot wat die geval sou wees in die gewone gang van besigheid. Hierdie addisionaliteit is relevant in beide 'n omgewings- en finansiele sin. Hierdie projek sal orngewings-addisionaliteit bevredig deurdat die wynkelder in sy afwesigheid volkome afhanklik van die steenkool-gebaseerde elektrisiteit vanaf die nasionale netwerk sou bly, terwyl finansiele addisionaliteit bewys is deur die finansiele lewensvatbaarheids-model wat getoon het dat die projek slegs ekonomies uitvoerbaar sal wees indien dit CDM deelname insluit. Die lewensvatbaarheids-model is ontwikkel om die ekonomiese uitvoerbaarheid van die projek te evalueer, onderworpe aan die aangenome inset parameter waardes en basiese aannames in die studie. Inherent tot hierdie aanname is die verdere veronderstelling dat die progressiewe daling in elektrisiteits-pryse tot 'n eventuele vlak van 35clkWh voorsien deur Eskom nie sal realiseer nie, beide as gevolg van die volgehoue druk wat die groeiende Suid-Afrikaanse ekonomie op die aansieniike, maar tydrowende, opwekkings-kapasiteit uitbreidings van Eskom behoort te plaas, en ook die hoe waarskynlikheid dat prys premiums vir skoon elektrisiteit ingestel word sodat Suid-Afrika sy volboubare energie telkens kan bereik. Die aanvanklike kapitaal en CDM uitgawes, en bedryfsuitgawes en - inkomstes oor die projekleeftyd is vanaf die aangenome inset parameter waardes afgelei, waarop die NPV van die projek uitgewerk is om te dien as die beslissende maatstaf van ekonomiese haalbaarheid. 'n Diskontokoers van 18% is gebruik, ooreenstemmend met die subjektief beraamde risikos wat die projek inhou as 'n klein-skaal volhoubare energie sisteem. Die resultate van die finansiele lewensvatbaarheids-model het getoon dat in die geval waar die projek geen CDM aspekte bevat nie, die NPV hoogs negatief sou wees met 'n waarde van -ZAR342 573, terwyl dit verbeter na ZAR325 193 as die projek CDM deelname insluit. Die gevolgtrekking is dus gemaak dat die spesifieke projek slegs ekonomies haalbaar sal wees indien dit wel CDM deelname insluit, en dat hierdie positiewe finansiele bydrae van die CDM geoptimaliseer kan word deur 'n programmatiese CDM benadering te volg. Dit behels die ontwikkeling van die projek as deel van 'n groter program waarin soortgelyke projekte mettertyd geimplementeer word soos hulle ontstaan, tot by die offisiele VN limiet van 15MWe om sodoende nog te kwalifiseer as 'n algehele klein-skaalse program. Die aard van die hoof-aannames in hierdie studie is indikatief dat die positiewe finansiele lewensvatbaarheid in die geval waar die projek CDM deelname insluit, eers oor die kort- tot medium-termyn prakties relevant sal word soos die aannames bevredig word. Dus, alhoewel die era van klein-skaalse volhoubare energie projekte nog nie aangebreek het in Suid-Afrika nie, lyk dit tog asof dit om die draai is.

Full textPhagocyte activation is critical role for host defense against several pathogens, so that studies developing alternatives for activating these cell are very important. We investigated the effects of BAY 41-2272 on phagocytes activation. For this purpose we evaluated several aspects indicating cellular activation, as phagocytosis, superoxide release and microbicidal activity. We used PBMC, neutrophils and THP-1 cell lineage cultured or not with BAY 41-2272 (1mM and 3mM) for 1h or 48h at 37°C. Superoxide release was tested by superoxide dismutase-inhibitable cytochrome c reduction assay; phagocytosis was evaluated through co-culture with zymosan particles and counting of ingested particles; and microbicidal activity was measured through co-incubation with enteropathogenic E. coli followed by counting of CFU from recovered bacteria from phagocytes. We analyzed the IL-12, IFN-g and TNF-a cytokine production and gp91phox gene expression. All cell types showed a response to treatments. PBMC, PMN and THP-1 treated with BAY 41-2272 produced significantly more superoxide (about 50% more), and presented significantly more phagocytic (about 54% more), microbicidal (about twice) activity than control group and production more TNF-a than control group. The expression of CYBB gene was more expressed on treated cells than control. BAY 41-2272, especially at 3mM, showed a great potential in activation of phagocytes in several aspects. This potential should be investigated at the light of new therapies seeking infection control, mainly in immunodeficiency.

A Crista Neural (CN) constitui uma população de células que emergem a partir das pregas dorsais do neuroepitélio durante a neurulação. Estas células migram ao longo de todo o eixo antero-posterior embrionário dos vertebrados até encontrarem seus locais definidos onde irão diferenciar em diversos derivados neurais e mesenquimais. As células da CN cefálica (CNC) e da CN Truncal (CNT) podem dar origem a neurônios e células gliais do sistema nervoso periférico, melanócitos e células endócrinas. Diferentemente do que ocorre com a CNC, a CNT possui uma limitada capacidade de originar elementos mesenquimais in vivo. Entretanto, sob determinadas condições in vitro, a CNT pode originar fenótipos mesenquimais. Até o momento, a maior parte dos estudos com a CNT foram realizados sobre substratos bidimensionais (2D), geralmente revestidos moléculas isoladas da matriz extracelular (colágeno, laminina, fibronectina) ou sobre monocamadas de fibroblastos embrionários (3T3). Atualmente, sabe-se que este tipo de ambiente não reflete a complexa fisiologia dos tecidos in vivo. No presente estudo nós realizamos cultivos de células isoladas da CNT sobre um extrato solúvel da membrana basal, comumente denominado de Matrigel. Elaboramos uma metodologia que permitiu criar dois microambientes dentro do mesmo poço de cultivo: um ambiente bidimensional (2D) e um ambiente tridimensional (3D). Verificamos que o Matrigel permite a diferenciação dos principais fenótipos da CNT (neurais: células gliais, células musculares lisas, neurônios, melanócitos e mesenquimais: condrócitos). Embora tenham sido observadas algumas variações na obtenção dos fenótipos descritos conforme o lote de Matrigel utilizado, o uso de uma mistura destes lotes permitiu normalizar a frequência de poços de cultivo contendo cada tipo celular. Além disso, 70% dos poços de cultivo apresentavam nódulos de cartilagem, os quais frequentemente se encontravam na região 3D. Interessantemente, esta frequência de condrócitos detectada utilizando um microambiente 3D de Matrigel, resultou ser muito maior quando comparado a trabalhos anteriores realizados sobre ambientes 2D convencionais. Estes dados sugerem que o Matrigel pode ser um ótimo substrato para estudar a diferenciação e multipotencialidade das células da CN.
Abstract : The Neural Crest (NC) is a cell population that detach from dorsal neural folds during neurulation. These cells migrate along the entire vertebrate embryonic axis until they find their final destination and differentiate into both neural and mesenchymal phenotypes. Cephalic NC (CNC) and Trunk NC cells (TNCCs) give rise to neurons and glia from the peripheral nervous system, melanocytes and endocrine cells. Differently from CNC, TNC have a limited capacity to give rise to mesenchymal derivatives in vivo. Nevertheless, under special culture conditions, TNC can give rise to mesenchymal phenotypes. Until now, most of studies of TNC were conducted on 2D substrates, usually covered with extracellular matrix molecules (e.g. collagen, laminin, fibronectin) or under fibroblast feeder-layers (3T3). Nowadays, it is worth knowing that this kind of environment does not mimic the complex tissue physiology in vivo. In the present study, we conducted cell cultures isolated from TNC on a soluble extract of basal membrane, commonly named Matrigel. We performed a new methodology that permitted to create two microenvironments in the same culture well: a two-dimensional (2D) and a three-dimensional (3D) environment. We observed that Matrigel permits the differentiation of the main TNC phenotypes (glial cells, melanocytes, smooth muscle cells, neurons and chondrocytes). Even though we observed some described variations on the phenotypes obtained according to the Matrigel lot used, the use of a mixture of lots allowed normalizing the frequency of culture wells containing each cell type. Moreover, 70% of culture wells contained cartilage nodules, which were frequently on the 3D zone. Interestingly, this frequency of chondrocytes detected using a 3D microenvironment of Matrigel resulted even higher than previous reports performed on traditional 2D cultures. Together, this data suggest that Matrigel could be an excellent substrate to study NC differentiation and multipotenciality.

Full textConstruction details available on the Internet and in books for various building components are today limited in their design. Terraces and cellar walls both have a challenging construction where high demands are made, as they are the house's protection against the external climate. The initiator WSP had experienced a problem that designers at the company designed differently when designing these types of details. The company therefore wanted to find a method where the designers could use the same type details as guidance. The purpose of this work was to facilitate the quality assurance of design details when designing terrace roofs and cellar walls by creating a basis for starting. The goal was then to produce clear construction details with an accompanying technical description. Through literature studies, project reviews, conversations with designers and experts on and outside the company, information has been collected to a sufficient extent as a basis for analysis and conclusion. Analysis and conclusion were conducted based on a longevity perspective focusing on the aspects; constructability, moisture safety, robustness and cost. The result clearly showed that the designers at the office designed these details differently and that they could not always justify their choices. The result also showed that the terrace roof as per the reverse concept is the best solution according to the longevity perspective, and for cellar walls is the important aspects to keep the wall dry and warm. For both cellar walls and terraces, there are different conditions that affect the production of construction details. This eventually led to two types construction details of terrace roofs and three for cellar walls.

O álcool atua no organismo podendo trazer várias doenças, entretanto sua ação no tecido ósseo ainda apresenta resultados controversos. O objetivo deste trabalho foi avaliar o efeito do consumo crônico de álcool a 20% em osteoblastos obtidos da calvária de ratos recém-nascidos. Foram utilizadas 18 ratas prenhas, tratadas durante a gestação e divididas em grupos conforme a dieta: álcool a 20%, grupo isocalórico, e controle. Aos três dias de vida, os recém-nascidos foram eutanasiados para remoção da calvária e isolamento das células por meio de digestão enzimática sequencial, sendo estas cultivadas por períodos de até 14 dias. Foram realizados testes para avaliar o efeito do álcool na adesão, proliferação e viabilidade celular, no conteúdo de proteína total, na atividade da fosfatase alcalina e nas formações nodulares de matriz mineralizada. Os resultados mostraram que em geral, a adesão celular não foi influenciada pelo consumo crônico de álcool, já que não foi demonstrada diferença estatística entre os grupos. Contudo, o grupo álcool apresentou aumento significativo na proliferação, exceto no período de 1 dia, e nas formações nodulares. Com relação a viabilidade celular, apenas no período de 3 dias houve aumento significativo de células no grupo álcool. Os valores representativos de proteína total variaram dependendo do período estudado, sendo maior no grupo controle com 7 dias, porém aos 14 dias houve maior média nos grupos isocalórico e álcool. Quanto à fosfatase alcalina observamos aumento de sua atividade nos grupos álcool e isocalórico em todos os períodos. Concluímos que nesta metodologia, o álcool não apresentou efeito deletério para os osteoblastos, talvez pelo curto tempo de administração
The alcohol affects the organism and may cause various diseases, even though its effects on bone metabolism are still controversial. The purpose of this paper was to evaluate the effects of alcohol 20% chronic consumption in osteoblasts obtained from the calvaria of newborn mice. The alcohol was administrated to pregnant mice throughout the entire pregnancy. For that purpose, 18 mice were used, divided in groups according to the diet: 6 receiving alcohol 20%, 6 belonging to the isocaloric group and 6 receiving water and ration at will. At three days of life, the newborns were euthanized so as to remove the calvaria and start the cell culture procedures. The osteoblastic lineage cells were isolated by sequential enzymatic digestion and the osteoblasts were cultivated for periods of 14 days or less. Tests were performed on the culture slides to evaluate the effect of alcohol based on adhesion, proliferation and cellular viability on total protein content, alkaline phosphatase activity and nodule formation of mineralized matrix. The results have shown that the alcohol group presented significant increase in proliferation, except for the period of one day, and in nodule formation. A significant increase in the alcohol group concerning cellular viability was only observed in the period of 3 days and there was no statistic difference in adhesion. The total protein content was higher in the control group in 7 days, and the average higher in the isocaloric and alcohol groups in 14 days, according to the periods of time studied. An increase in the activity of the alkaline phosphatase was observed in the isocaloric and alcohol groups in all periods of evaluation. By this methodology, we have concluded that the alcohol has not presented any deleterious effect on osteoblasts, possibly due to the short period of administration

Resumo: Transplantes autoplásticos de células-tronco periféricas (CTP) já vêm sendo realizados há algum tempo, tendo mostrado algumas vantagens em relação aos transplantes do mesmo tipo que utilizam medula óssea. Dentre estas vantagens, destaca-se a capacidade das CTP de produzir uma "pega" mais rápida do enxerto, encurtando o período de aplasia de medula óssea que se segue ao condicionamento do paciente. A coleta de CTP por aférese oferece maior conforto ao doador, que não precisa ser submetido à anestesia geral e às numerosas punções da crista ilíaca necessárias à obtenção do número de células suficientes para o transplante. Apesar das evidentes vantagens observadas no cenário autoplástico, a aplicação da mesma técnica aos transplantes alogênicos só se iniciou no começo dos anos 90. Este retardo se deveu ao temor de que os enxertos de CTP alogênicas, mais ricos em células T imunocompetentes, pudessem acarretar aumentos na incidência e gravidade da doença do enxerto-contra-o-hospedeiro (DECH). Outro empecilho à realização desses transplantes é a ecessidade de mobilização das CTP para a circulação sangüínea, que se fez possível nos transplantes autoplásticos, inicialmente, pelo uso de quimioterápicos citotóxicos; tal prática seria eticamente condenável em doadores sadios e, só mais recentemente, os fatores de crescimento hematopoiético humanos, obtidos por tecnologia de recombinação gênica (rhGCSF), ofereceram uma alternativa mais segura para conseguir essa mobilização. Neste estudo, não controlado analisamos os resultados de dezessete transplantes alogênios de CTP, comparando-os com aqueles obtidos em um grupo-controle que recebeu, contemporaneamente, transplantes alogênicos convencionais de medula óssea. O objetivo foi o de se estudar, comparativamente, o perfil de "pega" dos enxertos de CTP, assim como a morbidade associada ao procedimento. Para tanto, foram comparados vários parâmetros entre os dois grupos, que incluíram: tempo de "pega" do enxerto, tempo de permanência no hospital após o transplante, necessidade de transfusões, número de dias em uso de antimicrobianos ou nutrição parenteral, níveis séricos máximos de creatinina e bilirrubina, incidência e gravidade de DECH aguda, entre outros. Procuramos avaliar ainda os eventuais efeitos colaterais a curto prazo do rhG-CSF nos doadores. Os resultados mostraram que a incidência e gravidade da DECH aguda foram comparáveis entre os dois grupos, assim como a morbidade geral relacionada ao transplante. De fato, os transplantes com CTP apresentaram "pega" mais rápida e permitiram alta hospitalar mais precoce. A mobilização de CTP pelo uso de rhG-CSF e sua obtenção por sessões únicas de aférese mostraram-se, no período estudado, livres de efeitos colaterais ou complicações clínicas sérias para os doadores. O número de células foi suficiente para garantir a "pega" e estabilidade do enxerto em todos os pacientes, não se observando rejeições ou "falhas de pega". Aparentemente, as vantagens oferecidas pelas CTP em transplantes autoplástiéos também se aplicam aos transplantes alogênicos. Os resultados sugerem ser a técnica viável, porém devem ser interpretados com cautela devido às limitações metodológicas do estudo, que incluem o curto tempo de seguimento dos pacientes e a realização de análise de dados de grupos de pacientes distribuídos sem "randomização"
Abstract: Autologous blood stem celI (BSC) transplantation is already a welI established medical procedure, which has shown some advantages over the use of bone marrow for the same purpose. A faster engraftment rate is observed in these transplants, consequently shortening the period of marrow aplasia that folIows the conditioning phase. Besides, BSC colIection by apheresis offers more cornfort to the donors, avoiding the need for general anesthesia and marrow aspiration in order to obtain sufficient numbers of stem celIs for engraftment. Despite alI these advantages, the use of allogeneic BSC for transplantation did not start until the beginning of the Nineties. The application of the technique had always been hampered by the fear. that BSC grafts, which contain a larger amount of immunocompetent T -celIs, could result in intolerable increases of graft-versus-host disease (GVHD) incidence and severity. ColIection ofBSC by apheresis requires that these celIs first be mobilized to the circulation, a task acomplished initially by the use of antinoeplastic cytotoxic drugs. The administration of such agents to healthy donors would be a clearly unnacceptable practice, and, only Tecently, recombinant human hematopoietic growth factors (rhG-CSF) have offered a reasonable alternative for this purpose. In thi~ study, we retrospectively analyze the results of seventeen allogeneic BSC transplants, comparing them to those obtained in a control group of patients who received conventional allogeneic marrow transplants contemporarily fashion. The objective was to study the engraftment profile of the BSC grafts, as welI as the general transplant-associated morbidity. For this purpose, several parameters were assessed and compared between the two groups, including: time to engraftment, transfusion needs, number of days under antimicrobial treatment and parenteral nutrition, maximal serum levels of creatinine and bilirubin, incidence and severity of acute GVHD, among others. We also analyzed the eventual short-term deleterious effects of rhG-CSF upon donors. The results have pointed to a comparable incidence and severity of acute GVHD in the two populations, as well as similar transplant-associated morbidity profiles. Indeed, BSC grafts produced significantly faster engraftment and shorter hospital stays. Donor stem cell mobilization by rhG-CSF and their collection by single apheresis sessions have been devoid of significant side-effects or clinical complications in the study period. The numbers of cells collected have always proven sufficient to promote good engraftment, with no documentated of rejection or any other kind of graft failure. It seems apparent that the advantages offered by autologous BSC transplants can be shared by their allogeneic counterparts. The results suggest that allogeneic BSC transplantation is a feasible procedure, but one must not forget that this was not a randomized, prospective study, and that our follow-up time was not long enough to permit safe conclusions about the issue. More studies are necessary until we can replace alogeneic BSC transplantation for conventional bone marrow transplants

Resumo: Dada à necessidade de uma efetiva barreira tecidual precoce ao redor dos implantes osseointegrados, o presente estudo avaliou, in vitro, a influência das características físicas e químicas de materiais simulando superfícies de abutments de implantes em titânio (T) e zircônia (ZrO2) na adesão de células epiteliais gengivais OBA-9. Para este estudo foram utilizados espécimes em forma de discos (n=13) de titânio e zircônia, e como controles positivo e negativo, discos de esmalte bovino (EB) e lamínulas de vidro (LV), respectivamente. Foram avaliadas previamente a adesão celular a rugosidade superficial (Ra), energia livre de superfície (ELS) e os elementos químicos presentes na superfície identificados por meio de Espectrômetro de raios-X por Dispersão de Energia em Microscópio Eletrônico de Varredura (MEV). Após a adesão celular em 1 e 24 horas, foram avaliadas a viabilidade celular por meio do MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide); e a morfologia das células aderidas sobre as superfícies por meio de MEV. Os dados de viabilidade celular foram avaliados estatisticamente pelo teste de ANOVA a dois critérios fixos ("material" e "período de análise"), enquanto os dados de ELS foram submetidos aos testes de Kruskal-Wallis e Mann-Whitney, considerando um nível de significância de 5%. A superfície de T obteve o maior valor de ELS com 6,2N/m, seguida pela ZrO2 com 5,8N/m, LV com 4,9N/m, e com o menor valor o EB com 4,7N/m. Foi observado que os fatores material (p=0,336) e período de análise (p=0,400) não apresentaram efeito sobre os dados de viabilidade celular, assim como a interação entre eles (material*período de análise, p=0,559). A análise da morfologia celular mostrou que as células aderidas às superfícies de T e ZrO2 apresentaram espraiamento semelhante, sendo este maior quando comparado as superfícies de EB e LV. Abstract: Given the need for effective early mucosal barrier around dental implants, the present study evaluated, in vitro, the influence of physical and chemical characteristics of materials simulating abutments implants surfaces in titanium (T) and zirconium (ZrO2) on gingival epithelial cells OBA-9 adhesion. For this study, disks-shaped samples (n=13) of titanium and zirconium were tested, and as positive and negative control groups, bovine enamel disks (BE), and glass coverslips (GCS), respectively. Previously the cellular adhesion surface roughness (Ra), surface free energy (SFE) and the chemical composition of specimens by X-ray Spectrometer Dispersive Energy in Scanning Electron Microscope (SEM) were evaluated. After the periods of cellular adhesion, 1 and 24 hours, the cellular viability was evaluated by MTT Assay (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide) and the morphology of the epithelial cells adhered to the surfaces was analyzed using the SEM. The data of cell viability were statistically evaluated by two-way ANOVA ("Material" and "period of analysis"), while the SFE data were submitted to Kruskal-Wallis and Mann-Whitney tests, considering a significance level of 5%. The T specimens showed the highest values of SFE (6.2 N/m), followed by ZrO2 (5.8 N/m), GCS (4.9 N/m), and BE (4.7 N/m). It was observed that the factors material (p = 0.336) and periods of analysis (p = 0.400) showed no effect on cell viability data, as well as the interaction between them (material* period of analysis, p = 0.559). The analysis of cell morphology showed that cells adhered to the T and ZrO2 surfaces presented similar spreading, which was higher compared to the BE and GCS surfaces. It was concluded that there was no difference between the cellular adhesion to different materials for implant abutments, T and ZrO2, in evaluated periods. Full textThe interspecific fungal biodiversity (Illumina Mi-Seq) and the dynamics of Saccharomyces cerevisiae and Brettanomyces bruxellensis species were studied in a new winery and/or in 3 aging cellars, more specifically on the floor, the walls, the equipment and the outside of the barrels. In the new winery, an initial fungal consortium (yeasts and molds) is already present on all the winery environments before the arrival of the first harvest. This consortium consists of fungal genera such as Aureobasidium, Alternaria, Didymella and Filobasidium. These genera, that persist during two vintages, are not specific to the winery environment and seem to be adapted to natural or anthropic environments due to their ubiquitous character. The initial consortium is enriched by oenological genera (e.g. Hanseniaspora, Saccharomyces) that are introduced into the winery either by the harvest or by potential transfers between the different environments of the winery. However, these genera do not seem to persist or establish due to their low adaptation to the stressful conditions of the winery environment. The dynamics of wild S. cerevisiae in the new winery was also studied. No isolates belonging to this species were found before the arrival of the first harvest, confirming that this species is not specific to the winery environment and its presence is related to the alcoholic fermentation activity. However, the results obtained suggest a potential colonization of the new winery environment by certain S. cerevisiae strains. These so-called « colonizing » strains showed a higher capacity to form biofilms compared to non-implanted strains. This study highlights the importance of the winery environment that constitutes a true ecological niche for fungal populations capable of implantation during the winemaking process. In the environment of the 3 aging cellars, the equipment and the outside of the barrels (in direct contact with wine) are the environments that seem to be favorable for the development and installation of cultivable microbial populations (yeasts and lactic acid bacteria) and spoilage microorganisms (acetic acid bacteria and B. bruxellensis), unlike the floor and the walls where low microbial populations were found. Recurrent strains of B. bruxellensis have been found on both the equipment and the outside of the barrels and could be the cause of wine contamination during aging. These recurrent strains showed greater biofilm formation and resistance capacities which could explain the persistence of B. bruxellensis in aging cellars. These results highlight the importance of equipment cleaning and the regular monitoring of wines during aging in order to limit the contaminations

Full textTo be presented in dendritic cells MHC I, antigens are processed by immunoproteasome. The aim of the study was to examine the effect of pro-oxidants in dendritic cell-induced T CD8+ lymphocyte proliferation with focus on ubiquitin-proteasome system. The co-culture of DC incubated with Xanthine/Xanthine Oxidase system and T CD8+ isolated from mice immunized with DNA-HSP65 promoted T CD8+ proliferation. XaXO incubation was more efficient in promoting DC maturation compared to LPS. In addition, XaXO incubation decreased IP catalytic activity in relation to LPS, suggesting that increased T CD8+ proliferation is not directly related to IP activity. XaXO incubation did not alter the expression of 20S catalytic subunit, 19S regulatory unit or ⓹i subunit. XaXO incubation increased 11S regulatory unit content as well as that of ubiquitinated proteins. We suggest that the DC incubation with XaXO increased 11S content favoring its coupling to 20S in detriment of 19S. This would increase ubiquitinated protein levels and direct more peptide fragments for antigen presentation.

Resumo: Essa dissertação propôs-se a fazer uma análise do cinema contemporâneo a partir do universo dos filmes feitos com telefone celular, especificamente dos documentários. Para começar, procuramos entender em que contexto da história do cinema esses filmes se situam. Em seguida, mergulhamos nas características mais específicas do dispositivo desses filmes (conectividade, portabilidade e mobilidade), tendo como objeto de análise os filmes participantes do festival Pocket Films. Por fim, traçamos a relação dos filmes feitos com celular com a construção de processos de subjetividade no cinema

At first, we seek to understand the context at the history of cinema in which these movies take place. Resumo: Os cervídeos têm sofrido com a diminuição do seu habitat natural, segregação entre populações e diminuição da diversidade genética. Embora a criopreservação e estocagem de gametas e embriões seja usada como estratégia de preservação de populações selvagens, poucos estudos tem obtido sucesso devido a dificuldades na sua obtenção. Recentes estudos com células tronco de pluripotência induzida ("induced pluripotent stem cells" - iPSC) derivadas de células somáticas humanas e de camundongos conseguiram diferenciação em células germinativas, espermatozoides e oócitos. Deste modo, tem-se o objetivo a longo prazo de produzir gametas viáveis de cervídeos, a partir de células somáticas. Porém, como até a presente data células iPSC ainda não foram derivadas em cervídeos, o enfoque principal neste projeto foi estabelecer protocolos que propiciem a obtenção de linhagens iPSC estáveis. O processo de reprogramação de células somáticas em células iPSC é facilitado pelo uso de células-tronco adultas que possuam capacidade multipotente. Como pouco se conhece a respeito de células-tronco multipotentes em cervídeos, a primeira meta deste trabalho teve como objetivo obter biopsias de diversos tecidos com capacidade multipotente. Entre elas encontram-se as células-tronco do tecido adiposo, chifre e pele. Uma vez estabelecidas, as linhagens de cada tecido foram avaliadas quanto ao seu tempo de duplicação da população celular e foram induzidas à diferenciação em outros tipos de células (adipócitos, ... Abstract: Deer have been enduring pressures on their natural habitat, which causes segregation between populations and reduced genetic diversity. Although cryopreservation and storage of gamete and embryos can be used as a strategy for the preservation of wild populations, few studies have been successful due to difficulties to recover these materials. Recent studies in mice and humans have shown that induced pluripotent stem cells (iPSC) derived from somatic cells can be differentiated into germ cells, oocytes, and spermatozoa. Thus, we hypothesize that viable gametes could also be obtained from somatic cells in deer. To date, iPSC cells have not been derived in cervids and therefore our main goal is to establish protocols to obtain stable iPSC lines from deer somatic cells. The process of reprogramming somatic cells into iPSC cells is facilitated by the use of adult stem cells that still have the multipotent capacity. As little is known about multipotent stem cells in deer, the first objective of this study was to obtain biopsies from various tissues with multipotent differentiation capacity. Among these tissues are the stem cells from adipose tissue, antler, and skin. Once primary cultures were established, cells from each tissue were evaluated for doubling time and then induced in vitro to differentiate into adipocytes, osteocytes and chondrocytes lineages in order to assess their plasticity. Moreover, the expression of molecular markers was characterized by immunocytochemistry (OCT4, SOX2, Nanog, REX1) and RT-PCR (OCT4, SOX2, Nanog, LIN28, REX1). Full textCellular automata have been studied as computer models in many different areas. They have many properties, one of them being reversibility. Reversible cellular automata can be used, among other applications, for data compressing and encryption. Apparently, the reversible rules featured in the literature seem to have been derived through exhaustive searches in their corresponding spaces. However, it would be important the availability of an algorithm that would allow their direct and easy construction, different from what occurs in literature. This is the aim of this work. Along this line, we tried to come up with an algorithm to allow the identification of one-dimensional, reversible cellular automaton rules. This was based on reversible rules with 2 states and 2, 3, 4 and 5 cells per neighborhood, and on those with 3 states and 2 and 3 cells per neighborhood, all of them drawn out of exhaustive analysis and from the literature. By studying these rules it was possible to verify in each space that: all reversible rules are balanced; they are symmetrically distributed; a subset of them herein denoted primitive reversible rules, RPs have a simple formation law, defined by homogeneous blocks of states; and, if a rule is reversible, so are all its dynamically equivalent rules. In the attempt to obtain the targetted algorithm, an approach was explored in which the non-primitive reversible rules (the so-called derived rules, RDs) were supposed to be obtained from the primitives. Along this line, two ways to construct the RDs were tried out, one based upon using all RPs jointly as a group, and another, using them individually; however, neither of them led to a positive result. Additionally, relations between the properties of reversibility and conservativity of a rule have also been studied in the rule spaces considered.
Autômatos celulares têm sido estudados como modelos computacionais em diversas áreas, sendo que muitas são as suas propriedades, entre elas a reversibilidade. Autômatos celulares reversíveis podem ser usados, entre outras aplicações, para compactação ou encriptação de dados. Aparentemente, as regras reversíveis apresentadas na literatura parecem ter sido derivadas apenas através de buscas exaustivas em seus espaços correspondentes. No entanto, seria importante a existência de um algoritmo que permitisse construí-las fácil e diretamente, diferente do que acontece na literatura. Este é o objetivo deste trabalho. Neste sentido, buscou-se um algoritmo que permitesse identificar regras de autômatos celulares unidimensionais reversíveis. Para tanto, foram obtidas em análises exaustivas e na literatura todas as regras reversíveis de 2 estados e vizinhanças de 2, 3, 4 e 5 células, e de 3 estados e vizinhanças de 2 e 3 células. Com o estudo destas regras constatou-se em cada espaço que: todas as regras reversíveis são balanceadas; elas se distribuem simetricamente; um subconjunto delas aqui denominadas regras reversíveis primitivas, RPs possui lei de formação simples, definida por blocos homogêneos de estados; e, se uma regra é reversível, todas as suas equivalentes dinâmicas também o são. Na tentativa de se obter o algoritmo desejado explorou-se uma abordagem em que as regras reversíveis não primitivas (denominadas regras derivadas, RDs), seriam obtidas a partir das primitivas. Nesse sentido foram testados dois esquemas de construção das RDs, um baseado na utilização conjunta de todas as RPs, e outro, utilizando-as individualmente; entretanto, ambos não levaram a resultado positivo. Adicionalmente, estudou-se a relação entre as propriedades de reversibilidade e conservatividade de regras nos espaços considerados.

Full textThe aim of this dissertation is to apply and to compare the modeling system dynamics (MDS) and activity based costing (ABC) to measure the costs in cellular manufacture. Taking this into account, a simulation was accomplished using the concepts of MDS to obtain the costs of production of four products, in an environment of cellular manufacture, considering ABC method and the comparison of those values with the technique of traditional costing. As for the research methodology, this dissertation is of quantitative focus in which the research type is exploratory without establishment of hypotheses, however guided by their objectives. The obtained results demonstrate that the application of ABC together with MDS it is very satisfactory when compared with the traditional technique. In conclusion, it can be observed that the application of ABC with MDS can be used efficiently as an alternative method to costing in cellular manufacture.

Introdução: O câncer de mama possui etiologia variada, sendo influenciada tanto por fatores genéticos quanto por fatores ambientais. Dentro do contexto genético, o papel de genes que codificam moléculas do sistema imunológico tem sido alvo de interesse pela importância que esse sistema possui para o desenvolvimento do processo tumoral. A Interleucina-18 é uma importante citocina pró-inflamatória e que possui indicações como possível alvo-terapêutico. Possui polimorfismos de um único nucleotídeo localizados na região promotora do gene e que possuem atividade funcional. As células Natural Killer (NK) fazem parte da resposta imune inata, sendo a primeira linha de defesa do organismo contra vírus, bactérias e tumores. Estas células induzem a morte da célula-alvo quando não há o reconhecimento das moléculas de antígenos leucocitários humanos (HLA) de classe I, através de seus receptores, chamados Killer cell Immunoglobulin-like. Receptor (KIR) ou os receptores NKG2D. Vários estudos demonstram o envolvimento dos genes que codificam receptores de células NK na patogênese do câncer. Objetivo: Avaliação do papel de polimorfismos em genes envolvidos na regulação da atividade das células NK e o desenvolvimento do câncer de mama em pacientes do Estado de Santa Catarina. Resultados: A presença dos genótipos variantes dos polimorfismos IL18 -607 (C/A) [rs1946518] e IL18 -137 (G/C) [rs187238] no presente estudo apresentou um risco significativo para o desenvolvimento do câncer de mama, sendo que o modelo de herança mais significativo foi o codominante para o IL18 -607 (C/A) [rs1946518] (CC vs AA, P = 0,004, OR = 2,782 95%CI [1,385-5,589]). E o recessivo para IL18 -137 (G/C) [rs187238]. Em relação à análise do polimorfismo do gene NKG2D [rs2255336], não foi encontrada uma diferença significativa na presença do alelo variante entre pacientes e indivíduos do grupo controle. Para os genes KIR foi encontrado um aumento de risco de 5,50 (P = 0,046; [1,02-33,36]) para o desenvolvimento da doença na presença do gene KIR2DL3 e de 5,62 (P ? 0,0001; [2,81- 11,33]) na presença do gene KIR2DL2. A presença desses dois genes em conjunto apresentou um risco de 6,86 (P ?0001; [2,83-13,18]). A presença do gene KIR2DL5 mostrou-se como um fator de proteção em relação ao desenvolvimento do câncer de mama (P = 0,001; [0,06-0,53]). Conclusão: Estes resultados indicam um potencial papel dos polimorfismos dos genes envolvidos na regulação da atividade das células NK na patogênese do câncer de mama, indicando que esses genes do sistema imunológico são possíveis marcadores preditivos de risco para câncer de mama na população estudada.
Abstract : Introduction: Breast cancer has varied etiology, being influenced by both genetic factors and by environmental factors. Within the context of the genetic etiology, the genes that encode immune system molecules have been the target of interest because of the importance of this system in the development of tumor process. Interleukin-18 is an important pro-inflammatory cytokine and has possible indication as a therapeutic target. IL18 gene has some single nucleotide polymorphisms located in the promoter region of the gene that have functional activity on gene transcription. Natural Killer (NK) cells are part of the innate immune response, the first line of defense against viruses, bacteria and tumors. These cells induces death of the target cell when there is no recognition molecules in human leukocyte antigens (HLA) class I, through their receptors, called Killer cell immunoglobulin-like receptor (KIR) or NKG2D receptors. Several studies have shown the involvement of the genes encoding receptors of NK cells in the pathogenesis of cancer. Objective: Evaluation of the role of polymorphisms in genes involved in regulating the activity of NK cells and the development of breast cancer patients in the State of Santa Catarina. Results: The presence of genotype variants of IL18 polymorphisms -607 (C / A) [rs1946518] and IL18 -137 (G / C) [rs187238] in the present study showed a significant contribution to the development of breast cancer risk, and the most significant was the codominant inheritance model for the IL18 -607 (C / A) [rs1946518] (CC vs. AA, P = 0.004, OR = 2.782, 95% CI [1.385 to 5.589]). And the recessive model was for IL18 -137 (G / C) [rs187238]. Regarding the analysis of the NKG2D polymorphism (rs2255336) gene, a significant difference was not found in the presence of the variant allele between patients and control subjects. For KIRs genes we detected an increased risk of 5.50 (P = 0.046, 95% CI [1.02 to 33.36]) for the development of the disease in the presence of KIR2DL3 gene and 5.62 (P ? 0.0001; 95% CI [2.81-11.33]) for the presence of KIR2DL2 gene. The presence of both genes together increase the risk about 6.86 (P ?0001; 95% CI [2,83-13,18]). The presence of the KIR2DL5 gene introduced a protective factor in the development of breast cancer (P = 0.001, 95% CI [0.06 to 0.53]). Conclusion: These results indicate a potential role of polymorphisms in genes involved in regulating the activity of NK cells in the pathogenesis of breast cancer, indicating that these genes of the immune system, IL18, KIR2DL2 and KIR2DL3 are possible predictive risk markers for breast cancer in this population.

Resumo: A eficiência do tratamento do câncer sob vários aspectos, mesmo com os avanços ¿farmacotecnológicos¿, ainda permanece como desafio para a medicina. Diante desse fato, novos agentes que atuem de forma alvo-específico, apresentem poucos efeitos colaterais e possam impedir o ¿escape¿ das células tumorais à indução de morte, são extremamente desejáveis. No presente trabalho, foram abordados 3 aspectos da atividade antitumoral da riboflavina: indução da apoptose das células de câncer prostático, indução da diferenciação de células leucêmicas e aumento da biodisponibilidade intracelular do quimioterápico mitoxantrona. Sob esses 3 aspectos, através do estudo de vias de sinalização, identificamos mediadores moleculares responsáveis pela ação da riboflavina como antitumoral. A atuação da riboflavina irradiada em células de câncer de próstata foi dependente da inibição da PI3K. De forma interessante, observamos uma potencial ação inibitória da metástase, evidenciada pela inibição das metaloproteinases 2 e 9 e da angiogênese pela diminuição da expressão do VEGF. Em relação à diferenciação das células leucêmicas evidenciamos o envolvimento do receptor TNFR1, bem como das proteínas ciclina D, JNK, Src quinase e das proteínas tirosinas fosfatases SHP2 e PTP!. Portanto, proteínas com diferentes localizações celulares foram afetadas, culminando com a diminuição da proliferação, manutenção da sobrevivência celular, interrupção da progressão do ciclo celular e reorganização do citoesqueleto, efeitos metabólicos essenciais para a ocorrência da diferenciação. Esse trabalho de tese também demonstrou a aplicabilidade da técnica do Pepchip para a identificação de diferenças metabólicas entre duas linhagens das células da leucemia eritroblástica, K562 e Lucena. Outra abordagem interessante nesse trabalho foi o uso da riboflavina com a finalidade de aumentar a biodisponibilidade celular de quimioterápicos tradicionais e a utilização de inibidores de proteínas fosfatases como estratégia para reverter a resistência de células tumorais. De acordo com os resultados, esse trabalho aponta de forma inédita uma nova função da vitamina B2 como antitumoral
Abstract: Cancer therapy efficiency, under several aspects, even with the progress of ¿pharmacotechnology¿, remains as a challenge for the medicine. According to this factor, new agents that act on specific target, present low side-effects and prevent cancer cells ¿escaping¿ from death induction, are extremely desirable. In this work, 3 aspects of antitumoral property of riboflavin were evaluated: apoptosis induction of prostate cancer cells, leukaemic cells differentiation and increase of intracellular bioavailability of chemotherapic (mitoxantrone). Under these 3 aspects, and by signal transduction approach, we identified molecular mediators responsible for antitumoral activity of riboflavin. The action of irradiated riboflavin on prostate cancer cells was dependent on PI3K inhibition. Interestingly, we also observed a potential inhibitory action of metastasis, as demonstrated by the inhibition of metalloproteinases 2 and 9 and decreasing of angiogenesis by downregulation of VEGF. In relation to leukaemic cells differentiation we demonstrated the involvement of TNFR1, as well as cyclin D, JNK, Src kinase and protein tyrosine phosphatases SHP2 and PTP!. Therefore, proteins with different cellular localizations were affected culminating in decreasing of cell proliferation, maintaining cell survival, cell cycle arrest and cytoskeleton rearrangement, crucial metabolic effects for the occurrence of differentiation process. This work also demonstrated the applicability of Pepchip technique for identifying the differences between 2 erytroblastic leukaemia cell lines, K562 and Lucena. Other interesting approach, in this work, was the use of riboflavin for improving chemotherapic cellular bioavailability and the strategical use of protein phosphatase inhibitors for reverting tumor cells resistance. According to our findings, this work spotlights the novel function of the vitamin B2 as an antitumoral agent

Resumo: Riboflavina (Rb - Vitamina B2) é o precursor das flavocoenzimas essenciais flavina mononucleotídeo (FMN) e flavina adenina dinucleotídeo (FAD). Estas coenzimas participam de processos enzimáticos dependentes das reações de transferências de elétrons, que ocorrem nas vias de produção de energia, biossíntese, desintoxicação e sequestro de elétrons. O aumento dietético da riboflavina e piridoxina foi associado com maiores densidades minerais em mulheres e homens idosos. Fotoderivados da riboflavina demonstraram efeitos citotóxicos em células cancerosas de próstata e leucemias, entretanto, o efeito direto da Rb e seus fotoderivados em osteoblastos não foram examinados. Neste trabalho os efeitos biológicos da Rb e riboflavina irradiada (IRb) foram investigados na linhagem de pré-osteoblastos MC3T3-E1, um modelo bem aceito de osteogênese in vitro caracterizado pela indução de genes específicos associados com o fenótipo osteoblástico quando tratados com ácido ascórbico e ß-glicerofosfato. A viabilidade celular foi avaliada através da redução do MTT, da incorporação do corante vermelho neutro e do conteúdo de ácidos nucléicos. Marcadores de diferenciação osteoblástica foram analisados através do RT-PCR semi-quantitativo (osteopontina e osteocalcina) e através de análises colorimétricas de atividade da fosfatase alcalina (FAL) e síntese de colágeno pela coloração de picrosirius. As atividades das metaloproteinases (MMP) -9 e -2 foram avaliadas pela zimografia de gelatina. Microarranjos de peptídeos com subtratos específicos para quinases e imunoblotting foram usados para identificar os efeitos na sinalização celular. As atividades de enzimas antioxidantes (superóxido dismutase, catalase, glutationa peroxidase e glutationa S-transferase) foram determinadas em lisados celulares usando métodos espectrofotométricos. As atividades das caspases-8, -9 e -3 foram analisadas através de métodos colorimétricos. Na primeira análise Rb e IRb causaram a parada do ciclo celular na fase G0/G1 e também a inibição da quinase AKT, um mediador da proliferação. Flavinas causaram a diferenciação de pré-osteoblastos, evidenciada pelo aumento da expressão de osteocalcina, osteopontina e BMP-2. Atividades mais elevadas de MMP-9 e MMP-2 também foram observadas. A capacidade das flavinas em engatilhar a diferenciação de osteoblastos foi reforçada pelo aumento da conexina 43, diminuição da caveolina-1 e repressão da sinalização Notch. Na segunda análise, nós encontramos que as interações entre Rb, em sua forma irradiada e não-irradiada, e indutores osteogênicos (ácido ascórbico e ß-glicerofosfato) afetaram significativamente a proliferação de osteoblastos, a atividade de FAL, biossíntese de colágeno, expressão de osteocalcina e osteopontina, a atividade das MMP-2 e MMP-9 e a expressão de fatores osteoclastogênicos (RANKL e osteoprotegerina). Nós também encontramos que os efeitos das flavinas em osteoblastos nesta segunda etapa foram independentes das suas propriedades antioxidantes. A atividade biológica da combinação de indutores osteogênicas com Rb e seus fotoprodutos foi associada com a ativação de diferentes vias de sinalização (AKT, FAK, CaMKII), caspases -8, -9 e -3 e aumento da expressão e/ou estabilização de fatores de transcrição osteoblásticos (Runx2 e ß-catenin). Este estudo nos trouxe fortes evidências que altas concentrações de Rb e IRb geraram um microambiente osteogênico através da modulação de diferentes vias de sinalização, além de promover um efeitos aditivo durante a diferenciação das células pré-osteoblasticas MC3T3 induzida por ácido ascórbico e ß- glicerofosfato. Em resumo, este estudo aponta para uma potencial aplicação da Rb e seus fotoprodutos no desenvolvimento do fenótipo osteoblástico e, consequentemente, uma alternativa terapêutica coadjuvante para osteoporose.
Abstract: Riboflavin (Rb-Vitamin B2) is the precursor of essential flavocoenzymes, flavin mononucleotide (FMN) and flavin adenine dinucleotide (FAD). These coenzymes participate in numerous enzymatic processes dependent on electron transfer reactions that occur in energyproducing, biosynthetic, and detoxifying and electron-scavenging pathways. Increase dietary riboflavin and pyridoxine intake has been associated with higher bone mineral density in elderly men and women. Photoderivatives of riboflavin have been shown strong activity in haematological malignancy and prostate cancer cells, however, the direct effect of Rb and its photoderivatives on osteoblast has not been examined. In this work, the biologic effects of Rb and irradiated riboflavin (IRb) were investigated in the MC3T3-E1 pre-osteoblastic cell line, a well-accepted model of osteogenesis in vitro characterized for the induction of specific genes associated with the osteoblastic phenotype when treated with ascorbic acid and ß-glycerophosphate. Cell viability was assessed by MTT reduction, neutral red uptake and nucleic acids content. Osteoblastic differentiation markers were analyzed by semiquantitative RT-PCR (osteopontin and osteocalcin), alkaline phosphatase (ALP) activity measured colorimetrically and collagen synthesis by Sirius red staining. Metalloproteinases (MMP) -9 and -2 activities were assayed by gelatin zymography. Peptide microarray of substrate specificity to kinases and immunoblotting were used to identify the effects on signal transduction pathways. Antioxidant enzyme activities (superoxide dismutase, catalase, glutathione peroxidase and glutathione Stransferase) were determined in cellular lysate using spectrophotometric methods. Caspase-8, -9 and -3 activation were measured by a colorimetric assay. In the first analysis Rb and IRb caused cell cycle arrest at G0/G1 phase and accordingly inhibited AKT kinase, a proliferation mediator. Flavins caused differentiation of preosteoblast cells as evidenced by increase of osteocalcin, osteopontin and BMP2 expressions. In addition, higher MMP-9 and -2 activities were observed. Importantly, the capacity of flavins to trigger osteoblasts differentiation was also reinforced by upregulation of connexin 43, down regulation of caveolin-1 and negative modulation of Notch cascade. In the second analysis, we found that the interaction between Rb and IRb and osteogenic inductors (ascorbic acid and ß-glycerophosphate) significantly affected the osteoblast proliferation, alkaline phosphatase activity, collagen biosynthesis, osteopontin and osteocalcin mRNA expression, MMP-2 and MMP-9 activities and the expression of osteoclastogenesis factors (RANKL and OPG). We also showed that the effects of flavins in osteoblasts cells were independent on flavins antioxidant property. The biological activity of the combination of osteogenic medium with riboflavin and its photoderivatives was associated with the activation of different signaling pathways (AKT, FAK, CaMKII), caspases -8, -9 and -3, and up-regulation and/or stabilization of osteoblastic transcription factors (Runx2 and ß-catenin). This study brought out strong evidences that high concentration of Rb and IRb generates an osteogenic microenvironment through modulating different mediators of signaling pathways, besides of the additive effect of riboflavin and its photoproducts during the ascorbate and ß-glycerophosphateinduced osteoblast differentiation of MC3T3-E1 cells. In summary, this study pointed out the potential application of Rb and its photoproducts in osteoblasts phenotype development and, consequently, it is possible use as an alternative therapeutic adjuvant of osteoporosis.

Leprosy is an infectious disease characterized by different clinical forms that are associated with the type of immune response against Mycobacterium leprae. Polymorphisms present in the first intron of IFN- may have an important role in the regulation of the immune response, which could have functional consequences for gene transcription. Later studies identified the present of five polymorphisms in the IL12RB2 promoter region, this polymorphisms will may transcriptional factor suppress IL12R2 expression affect the development of cellular immune response. In the present study we investigated a possible association of the +874 T/A SNP and IFN-, microsatellite and polymorphisms present in the IL12RB2 promoter region associated with susceptibility to leprosy or with development of clinical forms in patients from brazilian Legal Amazon states. Nucleotide sequencing was performed with samples from 118 leprosy patients and 114 controls subjects, as well as immunophenotyping of CD4 +, CD8 + and CD69 + T cells by flow cytometry. Between leprosy patients and controls subjects, as well as between PB and MB patients were no observed significant differences among the studied genotypes in the polymorphisms of IL12RB2 studies. The polymorphisms -1035, -1023 and -464 present an absolute correlation (p<0.0001) having a combination of a same genotype in this positions. The microsatellite encoding 16 CA repeats was significantly associated with PB compared to MB patients (p = 0.019), being individuals homozygous for the +874 A allele present IFN- and the mean level of CD4 + and CD69 + T cells were higher. Our data suggest that polymorphisms present in the first intron of IFN- and of the IL12RB2 promoter region are not associated with susceptibility to leprosy, nevertheless, the +874 A/A genotype and microsatellite encoding 16 CA repeats may be related to a higher cellular immune response in patients and are consistently more frequently detected in PB patients.
A hanseníase é uma doença infecciosa caracterizada por apresentar diferentes formas clínicas que estão associadas com o tipo de resposta imune ao Mycobacterium leprae. Polimorfismos genéticos presentes no primeiro íntron de IFN- podem apresentar importante papel na regulação da resposta imune, podendo resultar em consequências funcionais na transcrição do gene. Estudos anteriores identificaram a presença de cinco polimorfismos na região promotora de IL12RB2, sendo que estes polimorfismos poderiam suprimir fatores de transcrição na expressão de IL12R2, afetando o desenvolvimento da resposta imune celular. No presente estudo investigamos uma possível associação do SNP +874, do microssatélite de IFN- e de polimorfismos presentes na região promotora de IL12RB2 com a susceptibilidade à hanseníase ou com desenvolvimento das formas clínicas em pacientes dos Estados da Amazônia Legal brasileira. O sequenciamento nucleotídico foi realizado em 118 pacientes com hanseníase e 114 indivíduos controles, assim como a imunofenotipagem de células T CD4 +, CD8 + e CD69 + pela citometria de fluxo. Entre pacientes com hanseníase e indivíduos controles, assim como entre pacientes PB e MB não foram observadas diferenças significativas entre os genótipos estudados nos polimorfismos de IL12RB2. Os polimorfismos - 1035, -1023 e -464 apresentam uma correlação absoluta (p<0,0001) havendo combinação de um mesmo genótipo nas respectivas posições. O microssatélite com 16 repetições foi significativamente associado com pacientes PB quando comparado aos pacientes MB (p = 0,019). Indivíduos homozigóticos ao alelo +874 A apresentaram maiores níveis de IFN- e maior média de células T CD4 + e CD69 +. Nossos dados sugerem que polimorfismos presentes no primeiro íntron de IFN- e da região promotora de IL12RB2 não estão associados com a susceptibilidade à hanseníase. No entanto, o genótipo +874 A/A e 16 repetições de CA de IFN- podem estar relacionados à alta resposta imune celular observada nestes pacientes, sendo também mais frequentemente encontrados em pacientes PB.
