Abstract:
543 Background: We have reported that carcinoembryonic antigen (CEA) response correlated with clinical outcomes of 1st-line cet-based therapy (Target Oncol 2017). Early tumor shrinkage (ETS) is considered to be an on-treatment biomarker for outcomes of chemotherapy; however, clinical biomarkers to predict outcomes earlier are warranted. Methods: This study included 69 pts who were assessable for CEA at baseline and 4 wks, and with observed survival time from 2 phase II trials of 1st-line therapy for KRAS exon2 wild-type mCRC; JACCRO CC-05 of cet plus FOLFOX (UMIN000004197) and CC-06 of cet plus SOX (UMIN000007022). We investigated the influence of baseline age, gender, PS, primary tumor sidedness (PTS), number of tumor sites, as well as the CEA decrease at 4 wks to the patient’s OS and PFS. Results: PTS and the CEA decrease at 4 wks were found to be important predictors to OS and PFS. Baseline CEA and CEA decrease at 4 wks were median of 31.0 (range, 1.0-20920.0) and median of 35% (range, -259%-97%), respectively. The STEP-analysis indicated that CEA response was most significantly associated with OS when a cut-off value of 50% for CEA-responder (HR 0.49, log-rank test p = 0.03). Median OS in responders (n = 25) and non-responders (n = 44) were 36.2 m and 21.5 m, respectively. When the same cut-off value was used, median PFS in responders and non-responders were 11.6 m and 6.5 m, respectively (HR 0.64, log-rank test p = 0.08). In addition, a multivariate Cox Proportional-Hazards Model with both PTS and CEA response as risk factors showed that PTS correlated with both OS and PFS. In pts with left-sided tumors, non-responders (n = 33) had shorter OS and PFS compared to responders (n = 23). In the above 2-covariate Cox proportional hazard model, adjusted HR for CEA 50% decrease is 0.55, p = 0.1; adjusted HR for PTS is 2.66, p = 0.008. Conclusions: Our analysis suggests 50% CEA decrease at 4 wks as an early on-treatment biomarker for 1st-line cet-based therapy in mCRC. It may potentially predict outcomes earlier compared to ETS. Also, CEA response at 4 wks may differentiate pts who receive more benefit from cet treatment in left-sided tumors. Clinical trial information: UMIN000004197 and UMIN000007022.

Abstract:
Cohesion Theory of Halliday and Hasan has been widely applied in different parts of language teaching. As many cohesive devices are used in language, it’s very necessary for language learners to understand and identify those devices. Now in China, a large number of college students have to take part in the CET-4 test every year. Thus, it is of great significance that some effective strategies can be applied in the test and help more students pass the test easily. Cohesion Theory and cohesive devices may be such strategies and they are important for the improvement of reading comprehension of CET-4. This article is aiming to use the CET-4 test papers in June 2019 as examples to illustrate that Cohesion Theory can be applied in reading comprehension of CET-4 with great effectiveness.

Abstract:
This paper presents the concept and algorithm of data mining and focuses on the linear regression algorithm. Based on the multiple linear regression algorithm, many factors affecting CET4 are analyzed. Ideas based on data mining, collecting history data and appropriate to transform, using statistical analysis techniques to the many factors influencing the CET-4 test were analyzed, and we have obtained the CET-4 test result and its influencing factors. It was found that the linear regression relationship between the degrees of fit was relatively high. We further improve the algorithm and establish a partition-weighted K-nearest neighbor algorithm. The K-weighted K nearest neighbor algorithm and the partition algorithm are used in the CET-4 test score classification prediction, and the statistical method is used to study the relevant factors that affect the CET-4 test score, and screen classification is performed to predict when the comparison verification will pass. The weight K of the input feature and the adjacent feature are weighted, although the allocation algorithm of the adjacent classification effect has not been significantly improved, but the stability classification is better than K-nearest neighbor algorithm, its classification efficiency is greatly improved, classification time is greatly reduced, and classification efficiency is increased by 119%. In order to detect potential risk graduating students earlier, this paper proposes an appropriate and timely early warning and preschool K-nearest neighbor algorithm classification model. Taking test scores or make-up exams and re-learning as input features, the classification model can effectively predict ordinary students who have not graduated.

Abstract:
We have identified a new member of the TGF-beta superfamily, CET-1, from Caenorhabditis elegans, which is expressed in the ventral nerve cord and other neurons. cet-1 null mutants have shortened bodies and male tail abnormal phenotype resembling sma mutants, suggesting cet-1, sma-2, sma-3 and sma-4 share a common pathway. Overexpression experiments demonstrated that cet-1 function requires wild-type sma genes. Interestingly, CET-1 appears to affect body length in a dose-dependent manner. Heterozygotes for cet-1 displayed body lengths ranging between null mutant and wild type, and overexpression of CET-1 in wild-type worms elongated body length close to lon mutants. In male sensory ray patterning, lack of cet-1 function results in ray fusions. Epistasis analysis revealed that mab-21 lies downstream and is negatively regulated by the cet-1/sma pathway in the male tail. Our results show that cet-1 controls diverse biological processes during C. elegans development probably through different target genes.

Abstract:
243 Background: The benefit of salvage chemotherapy such as weekly paclitaxel (TAX) is modest in metastatic (met) urothelial ca, with median TTP of ≤3 months and ORR of 10% (95% CI 4-18% [combined results, Vaughn 2002; Joly, 2009]). Cetuximab (CET) is a monoclonal antibody against the epidermal growth factor receptor (EGFR). High-grade urothelial cancer overexpress EGFR. We conducted a multicenter randomized, noncomparative phase II study to measure the efficacy of CET ± TAX in patients (pts) with previously treated met urothelial cancer. Methods: Pts with met urothelial cancer who received one line of chemotherapy in the adjuvant, neoadjvuant, or met setting were enrolled. Pts were randomized to CET 250mg/m2 (after 400 mg/m2 load) ± TAX 80 mg/m2 weekly. A cycle was 4 weeks (wks). Response (RECIST) was assessed by imaging every 8 wks. We used early progression to assess futility (Litwin Stats Med 2007). Either arm would close if 7 of the initial 15 pts in that arm progressed at the first disease evaluation. Either arm would be considered promising if ≥9/28 patients had PFS>16 wks (90.4% power to detect an improvement in Median [Med] PFS from 8 to 16 wks with a 7.1% type 1 error). Results: We enrolled 39 evaluable pts (30 male). Median age was 69 years (range 49-79). All pts received prior platinum-based chemotherapy. CET arm closed after 9 of the first 11 pts progressed by 8 wks (ASCO GU 2009). CET-TAX arm completed full accrual (28 pts), of which 11 had visceral disease and 13 received chemo for met disease. 10/28 pts had PFS>16 wks. Overall RR was 28.5%, (8/28 pts, 95% CI 13-49%) (2 CR, 6PR). 4 additional pts had unconfirmed PR. 2 pts have maintained PR 3 and 4 months after discontinuing CET-TAX. Med PFS for the CET-TAX was 115 days (16 weeks)(95% CI 58-174 days). Med PFS for pts with visceral disease was 84 days (95%CI 50-NR). Med PFS for pts who received prior chemo for met disease was 142 days (95% 58-NR). Med number of cycles for the CET-TAX was 3 (range 0-25). Grade 3 AEs occurring in more than 2 pts were rash (5), fatigue (4), anemia (4), low magnesium (3). Conclusions: EGFR inhibition with CET appears to augment the antitumor activity of TAX in pts with previously treated urothelial cancers. The CET-TAX combination merits further study to establish its role in treatment of urothelial cancers. [Table: see text]

Abstract:

Abstract:

Abstract:

Ceramic electrolytes that conduct either oxygen ions or protons at intermediate temperatures are important materials for use in e.g. solid oxide fuel cells, the prime candidate to produce electricity by electrochemical reactions. There are still challenges with respect to the cell performance which have to be solved before solid oxide fuel cells become a commercial success. One of the main topics in this field concerns the enhancement of ionic conductivity at suitable operation temperatures. During the last decade nanocrystalline materials have received considerable attention. Nanocrystalline ionic conductors may have higher ionic conductivity compared to traditional ceramics with grain sizes in the micrometer range.

The aim of this work has been to develop electrolyte ceramic materials with a designed microstructure. CeO2- and LaNbO4-based materials have been prepared through a complete route, from preparation of powders to densification of ceramics including characterization of selected properties. CeO2-based materials are oxygen ion conductors and show higher ionic conductivity compared to the more common yttria stabilized zirconia (YSZ), thus a lower operation temperature is possible. LaNbO4-based materials have recently been suggested as a promising proton conductor stable in CO2/H2O atmosphere.

In Paper I, powder synthesis of nanocrystalline CeO2-based powders (CeO2, Ce0.8Gd0.2O1.9 and Ce0.8Sm0.2O1.9) using combustion synthesis with glycine as fuel and nitrate as oxidizer is reported. The influence of glycine to nitrate (G/N) ratio on the pure CeO2-based powders was investigated. The influence of calcination temperature on crystallite size, surface area and carbonate species remaining from combustion reaction was studied, with special attention to powders prepared using a near-stoichiometric G/N-ratio. A G/N-ratio of 0.55 and calcination at 550°C in oxygen flow resulted in high quality powders with a crystallite size of ~10 nm with low degree of agglomeration due to the vigorous combustion. The G/N-ratio influenced the densification behavior of the powders. A G/N-ratio of 0.55 resulted in excellent sintering properties with an onset of sintering at ~600°C and fully dense materials were obtained at ~1300°C.

In Paper II, three different sintering techniques have been used to prepare dense (>95%) CeO2-based materials from the high-quality powders described in Paper I: Spark plasma sintering, hot pressing and conventional sintering. The three different sintering techniques resulted in different grain sizes, ranging from 160 nm by spark plasma sintering, to 50 μm by conventional sintering mainly due to difference in sintering temperature and the applied pressure. The materials were reduced after hot pressing and a minor reduction was observed after spark plasma sintering. The materials were easily reoxidized at temperatures above 200°C. The electrical conductivity, measured by van der Pauw method, revealed no clear dependence on grain size, but instead a dependence on the sintering method used. The substituted materials prepared by hot pressing had a lower electrical conductivity and higher activation energy compared to the materials prepared by both conventional and spark plasma sintering. Thus, it is proposed that the reduction of Ce observed during hot pressing might be detrimental for the ionic conductivity even after reoxidation. Hardness and fracture toughness, measured by Vickers indentation, were more influenced by chemical composition than the grain size of the materials. Higher fracture toughness and lower hardness were observed for pure CeO2 compared to the substituted materials.

A novel route to prepare large quantity of sub-micron LaNbO4-based powders by spray pyrolysis is presented in Paper III. An aqueous solution containing stable La-EDTA complex and Nb-malic acid complex was spray pyrolysed using an in-house spray pyrolysis unit. The pure, nonagglomerated powders had a particle size of ~0.1 μm, narrow particle size distribution and high purity after calcination at 800°C.

The sintering behavior, microstructure, phase content and electrical conductivity of La1-xAxNbO4 (x = 0, 0.005 and 0.02 and A = Ca, Sr and Ba) prepared by spray pyrolysis is presented in Paper IV. The powders had excellent sintering properties and achieved high density after conventional sintering at 1200°C or as low as 1050°C by hot pressing at 25 MPa. A grain size down to 0.4 μm was achieved by hot pressing. The acceptor doped materials had a more homogenous microstructure due to secondary phases inhibiting grain growth compared to pure LaNbO4. Liquid secondary phase was formed at elevated temperatures in acceptor doped LaNbO4, resulting in tremendous grain growth (~70 μm) and microcracking in La0.98Ba0.02NbO4. The solubility of Sr on La-site in LaNbO4 was determined to 1% at 1500°C, and similar low solubility of CaO and BaO in LaNbO4 was inferred. Protons were found to be the main charge carrier up to 1000°C in wet hydrogen. Higher grain boundary resistivity was observed compared to previous work, possibly due to lower sintering temperature resulting in secondary phases due to lower solubility of AO.

The thermal and mechanical properties of LaNbO4-based materials are presented in Paper V. The materials possessed a ferroelastic to paraelastic phase transition at ~500°C and the linear thermal expansion was significant lower for the paraelastic compared to the ferroelastic phase. The pure LaNbO4 had a significantly lower hardness compared to acceptor doped (Ca, Sr and Ba) LaNbO4 due to large grain size and microcracking. The fracture toughness of La0.98Sr0.02NbO4, measured by SEVNB method, was 1.7±0.2 MPa·m1/2. The ferroelastic properties were confirmed by non-linear stress-strain relationship and remnant strain. The remnant strain decreases with increasing temperature and increasing acceptor doping. The latter was possibly due to secondary phases pinning the ferroelastic domain boundaries.

Abstract:
The explosive 2,4,6-trinitrotoluene (TNT) is currently a main ingredient in munitions; however the compound has failed to meet the new sensitivity requirements. The replacement compound being tested is 2,4-dinitroanisole (DNAN). DNAN is less sensitive to shock, high temperatures, and has good detonation characteristics. However, DNAN is more soluble than TNT, which can influence transport and fate behavior and thus bio-availability and human exposure potential. The objective of this study was to investigate the environmental fate and transport of DNAN in soil, with specific focus on sorption processes. Batch and column experiments were conducted using soils collected from military installations located across the United States. The soils were characterized for pH, electrical conductivity, specific surface area, cation exchange capacity, and organic carbon content. In the batch rate studies, change in DNAN concentration with time was evaluated using the first order equation, while adsorption isotherms were fitted using linear and Freundlich equations. Solution mass-loss rate coefficients ranged between 0.0002 h(-1) and 0.0068 h(-1). DNAN was strongly adsorbed by soils with linear adsorption coefficients ranging between 0.6 and 6.3 L g(-1), and Freundlich coefficients between 1.3 and 34 mg(1-n) L-n kg(-1). Both linear and Freundlich adsorption coefficients were positively correlated with the amount of organic carbon and cation exchange capacity of the soil, indicating that similar to TNT, organic matter and clay minerals may influence adsorption of DNAN. The results of the miscible-displacement column experiments confirmed the impact of sorption on retardation of DNAN during transport. It was also shown that under flow conditions DNAN transforms readily with formation of amino transformation products, 2-ANAN and 4-ANAN. The magnitudes of retardation and transformation observed in this study result in significant attenuation potential for DNAN, which would be anticipated to contribute to a reduced risk for contamination of ground water from soil residues.

Abstract:
The aim of that work was to investigate the flavor impact of 4-acetyl gamma butyrolactone (solerone) and 4,5-dimethyl 3-hydroxy-2(5H) furanone (sotolon) on the typical flor-sherry wines aroma. The amounts of those two compounds were determined in red and white wines and in flor-sherry wines. The results showed that only sotolon was specific of the latters. By sensory analysis, solerone was proved to be not a key compound of the aroma of samples containing it. At the opposite, sotolon is in sufficient amounts in flor-sherries to he detected. Moreover, statistical analysis revealed its concentration to be correlated to the typical flavor intensity of that kind of wines. Finally, cultures of yeast (S. Bayanus) in different medium were performed. Amounts of sotolon similar to those of flor-sherry wines were detected in organic extracts of those medium, proving its biochemical origin. Further developpement of the results are discussed

The process of programmed cell death is important in the development and homeostasis of multicellular organisms. The conserved morphological events of this process have been termed apoptosis. The molecular mechanisms of apoptosis execution are also conserved. We have investigated the behavior of these shared components during programmed cell death in the nematode Caenorhabditis elegans. We have found that the CED-9 protein, an anti-apoptotic member of the Bcl-2 family of apoptotic regulators, is required for the sequestering of the CED-4 cell-death activator to mitochondria. In the absence of CED-9 in C. elegans embryos, we found that CED-4 protein translocates to the nuclear membrane. In addition, inducing excess programmed cell death by expression of the EGL-1 cell-death activator triggers the translocation of CED-4 from mitochondria to the nuclear membrane. We performed a genetic screen for mutations that trigger programmed cell death in ced-9 gain-of-function animals where cell death is blocked. We identified a mutation in cup-5, the C. elegans homolog of the human mucolipidosis type IV gene, which is mutated in a lysosomal storage disorder. We found that cup-5 is required for viability and that excess lysosomes accumulate in cup-5 mutants. In addition, cup--5 mutants contain excess programmed cell deaths, suggesting that apoptosis may play a role in the pathology of mucolipidosis type IV.

Mudge, Christopher, Bradley Sartain, Kurt Getsinger, and Michael Netherland. Efficacy of florpyrauxifen-benzyl on dioecious hydrilla and hybrid water milfoil - concentration and exposure time requirements. Engineer Research and Development Center (U.S.), September 2021. http://dx.doi.org/10.21079/11681/42062.

Full text
Abstract:
This study conducted small-scale trials under various concentration and exposure time (CET) scenarios to determine florpyrauxifen-benzyl activity on dioecious hydrilla and hybrid watermilfoil and determine impact on water stargrass and elodea. Hydrilla treated with 12, 24, or 36 μg active ingredient (a.i.) L⁻¹ florpyrauxifen-benzyl and exposed for 12, 24, or 48 hr under outdoor mesocosm conditions was reduced in biomass by 30-75% at 8 weeks after treatment (WAT). An additional hydrilla trial at the same herbicide concentrations, but under longer exposures (24, 72, or 168 hr), resulted in 33–85% plant control. Under indoor conditions, hybrid watermilfoil dry weight decreased 98–100% with subsurface applications of florpyrauxifen-benzyl under CET scenarios of 3–12 μg a.i. L⁻¹ at 3–24 hr exposure times in a growth chamber trial. Under shorter exposure periods (0.5–4 hr) in a follow-up trial, low doses (3–9 μg a.i. L⁻¹) achieved 50–100% control of hybrid watermilfoil. In the same trial, the nontarget species water stargrass and elodea proved relatively tolerant to the florpyrauxifen-benzyl at doses up to 6 μg a.i. L⁻¹ (4 hr exposure) and 9 μg a.i. L⁻¹ (1 hr exposure). These small-scale trials demonstrate florpyrauxifen-benzyl’s potential to selectively manage invasive species.

Full text
Abstract:

Nolan, Brian, Brenda Gannon, Richard Layte, Dorothy Watson, Christopher T. Whelan, and James Williams. Monitoring Poverty Trends in Ireland: Results from the 2000 Living in Ireland survey. ESRI, July 2002. http://dx.doi.org/10.26504/prs45.

Full text
Abstract:
This study is the latest in a series monitoring the evolution of poverty, based on data gathered by The ESRI in the Living in Ireland Surveys since 1994. These have allowed progress towards achieving the targets set out in the National Anti Poverty Strategy since 1997 to be assessed. The present study provides an updated picture using results from the 2000 round of the Living in Ireland survey. The numbers interviewed in the 2000 Living in Ireland survey were enhanced substantially, to compensate for attrition in the panel survey since it commenced in 1994. Individual interviews were conducted with 8,056 respondents. Relative income poverty lines do not on their own provide a satisfactory measure of exclusion due to lack of resources, but do nonetheless produce important key indicators of medium to long-term background trends. The numbers falling below relative income poverty lines were most often higher in 2000 than in 1997 or 1994. The income gap for those falling below these thresholds also increased. By contrast, the percentage of persons falling below income lines indexed only to prices (rather than average income) since 1994 or 1997 fell sharply, reflecting the pronounced real income growth throughout the distribution between then and 2000. This contrast points to the fundamental factors at work over this highly unusual period: unemployment fell very sharply and substantial real income growth was seen throughout the distribution, including social welfare payments, but these lagged behind income from work and property so social welfare recipients were more likely to fall below thresholds linked to average income. The study shows an increasing probability of falling below key relative income thresholds for single person households, those affected by illness or disability, and for those who are aged 65 or over - many of whom rely on social welfare support. Those in households where the reference person is unemployed still face a relatively high risk of falling below the income thresholds but continue to decline as a proportion of all those below the lines. Women face a higher risk of falling below those lines than men, but this gap was marked among the elderly. The study shows a marked decline in deprivation levels across different household types. As a result consistent poverty, that is the numbers both below relative income poverty lines and experiencing basic deprivation, also declined sharply. Those living in households comprising one adult with children continue to face a particularly high risk of consistent poverty, followed by those in families with two adults and four or more children. The percentage of adults in households below 70 per cent of median income and experiencing basic deprivation was seen to have fallen from 9 per cent in 1997 to about 4 per cent, while the percentage of children in such households fell from 15 per cent to 8 per cent. Women aged 65 or over faced a significantly higher risk of consistent poverty than men of that age. Up to 2000, the set of eight basic deprivation items included in the measure of consistent poverty were unchanged, so it was important to assess whether they were still capturing what would be widely seen as generalised deprivation. Factor analysis suggested that the structuring of deprivation items into the different dimensions has remained remarkably stable over time. Combining low income with the original set of basic deprivation indicators did still appear to identify a set of households experiencing generalised deprivation as a result of prolonged constraints in terms of command over resources, and distinguished from those experiencing other types of deprivation. However, on its own this does not tell the whole story - like purely relative income measures - nor does it necessarily remain the most appropriate set of indicators looking forward. Finally, it is argued that it would now be appropriate to expand the range of monitoring tools to include alternative poverty measures incorporating income and deprivation. Levels of deprivation for some of the items included in the original basic set were so low by 2000 that further progress will be difficult to capture empirically. This represents a remarkable achievement in a short space of time, but poverty is invariably reconstituted in terms of new and emerging social needs in a context of higher societal living standards and expectations. An alternative set of basic deprivation indicators and measure of consistent poverty is presented, which would be more likely to capture key trends over the next number of years. This has implications for the approach adopted in monitoring the National Anti-Poverty Strategy. Monitoring over the period to 2007 should take a broader focus than the consistent poverty measure as constructed to date, with attention also paid to both relative income and to consistent poverty with the amended set of indicators identified here.
