Numerous enzymatic reactions are controlled by the chemistry of metallic ions. This dissertation investigates the electronic properties of three transition metal (copper, chromium, and nickel) complexes and describes modeling studies performed on glutathione synthetase. The formation of DNA triple helices offers the possibility of selectively targeting specific genes to control their expression in vivo. This anti-gene strategy provides powerful tools for the development of therapeutics (anti-cancer drugs, drugs for viral infections) at the transcriptional level. DNA triplexes are formed when an oligonucleotide binds to the major groove of double helical DNA; the third strand can bind in either a parallel motif, or an anti-parallel motif. The requirement of low pH for the protonation of cytosine in the parallel binding motif makes the formation of triple helic

The avalanche of data that followed the sequencing of the human genome has revealed an overwhelming biological complexity. No simple molecular explanation exists for most of the diseases and, in consequence, simple therapies have low probability of success. The emerging field of systems pharmacology seeks drugs of broad impact on molecular networks. To achieve so, it is necessary to integrate heterogeneous data, at different levels of complexity, and find correlations between them. Cataloged from PDF version of thesis.
Includes bibliographical references.
Proteins perform myriad functions inside cells. In vitro studies of purified proteins yield important information about their function, but it is an important pursuit in cell biology to study proteins within the native context of intact cells, since their physiological functions might not be recapitulated in any other environment. The most basic question, the location of a protein within the cell, is difficult to answer,

Mathematical modeling has been instrumental to the development of natural sciences over the last half-century. Through iterated interactions between modeling and real-world exper- imentation, these models have furthered our understanding of the processes in biology and chemistry that they seek to represent. In certain application domains, such as the field of car- diac biology, communities of modelers with common interests have emerged, leading to the development of many models that attempt to explain the same or similar phenomena. As these communities have developed, however, reporting standa

On the LED side, in particular, Chapter 2 utilizes individual NCs to alter layered organic LED structures at nanometer scale, resulting in spectrally resolved electroluminescence from single colloidal CdSe/ZnS (core/shell

Abstract:
Type 2 phosphatidylinositol-5-phosphate 4-kinase (PI5P4K) converts phosphatidylinositol-5-phosphate to phosphatidylinositol-4,5-bisphosphate. Mammals have three genes, PIP4K2A, PIP4K2B and PIP4K2C that encode the enzymes PI5P4Kα, PI5P4Kβ and PI5P4Kγ respectively. Studies in mice showed that PI5P4Kβ is a negative regulator of insulin signaling (Lamia et al., 2004) and that co-deletion of Pip4k2b and Trp53 resulted in synthetic embryonic lethality (Emerling et al., 2013). Also, deletion of two alleles of Pip4k2a and one allele of Pip4k2b suppressed the appearance of tumors in Trp53-/- mice. Thes
