The shear failure mechanism induced by wave of seabed at the Chengdao Sea was analyzed, one model to calculate the shear failure was established, and the factor of safety of shear failure at the different locations was calculated, some conclusions were gotten: (1) The shear stress at the seabed induced by wave and gravity along the submarine slope are the main triggers to cause shear failure, and the shear stress at the seabed surface induced by wave and currents can be ignored; (2) At extreme sea conditions, the shear failure regions are mainly distributed within the north-west and south-east. The shear failure depth of north-west area is within 4m, and that at south-east is relatively large, beyond 4m in depth. (3) The shear failure of Chengdao Sea was mainly controlled by wave stress in seabed. The shear failure region and depth calculated in this paper can be referred in oil engineering facilities design and construction.

Abstract:
In recent years, investigating the trend of aquatic plant diversity in response to different disturbance events has received increasing interest. However, there is limited knowledge of the different stages of aquatic vegetation succession over a long period in eutrophic lakes. In this study, we analyzed aquatic plant species richness and its relation to the physical and chemical characteristics of water in Chenghai Lake for the period of 1980–2018. This study shows that the richness and distribution of aquatic vegetation in Chenghai Lake are related to chlorophyll-a concentration, dissolved nutrients, base cations, and micronutrients. The results show that the long-term succession of aquatic plants in this lake classified in different stages: (I) A peak in species richness occurred at an intermediate stage that lasted from 1980 to 1992, and this was caused by more aquatic species being able to coexist since the competition for resources was lower; (II) after 26 years of secondary succession (1992–2018), the diversity and distribution area of aquatic plants gradually declined because pioneer species or human activities may have altered habitat conditions to render habitats less beneficial to pioneer species and more suitable for new aquatic plant species. Thus, species diversity and growth performance of aquatic plants in their communities may be useful indicators of Chenghai Lake’s trophic status, especially during the transition period from a mesotrophic lake to a eutrophic one.

E-mail: wenzhou@csu.edu.cn; chengtao@ihcams.ac.cn Background: It is generally believed that multiple myeloma (MM) cells develop from either B cells or plasma cells in the bone marrow (BM). However, the nature of the earliest precursors of MM cells is still poorly defined and existing data are conflicting regarding the origin of MM cells. How these genes cooperate with other MM-related factors to induce MM oncogenic transformation in the hematopoietic hierarchy needs to be further defined. Materials and Methods: In this study, we employed single-cell gene expression analysis of HSC-PC lineage including HSC, MLP, B cells, Plasmablasts (PBs) and Plasma cells (PC) from one health donor (HD), one paired monozygotic twin(one is HD and another one suffered with MM), MM patients at diagnosis (AD) and relapse (RE) with FGFR3/IGH translocation. 672 individual cells from the 8 population were sorted for single cell real-time PCR reaction. Results: We focused on the HSC-PC lineage because of its implication in MM and identified several differentially expressed genes (DEGs) in the HSC compartment with continuous activation throughout HSC-PCs. Among those, BACH2FA and DDIT3 showed consistent up-regulation, while FOXO3 and IL1 were down-regulated continuously. 7 DEGs (SIPA1L2, RBM34, SPI1, MDM2, AKAP12, DKK1 and c-Myc) showed overlap among the three conditions (HD, AD and RE, respectively). Kaplan-Meier analysis revealed HSC-associated signatures (HAS) in overlapping and non-overlapping DEGs with poor survival outcome, p=0.000). To compare genetic alterations between sporadic and hereditary MM patients, we analyzed two groups of DEGs - one from sporadic MM patients (AD) and HD; the other from the hereditary MM patient (AD) and the healthy counterpart of its paired monozygotic twin- along the HSC differentiation hierarchy. We identified both overlapping and non-overlapping DEGs. For example, MDM2 and c-Myc were over-expressed at the HSC and the MLP stage, respectively, in both groups. However, ARUKA was not activated in the monozygotic twin group, presumably because of the lack of FGFR3 translocation. By using fluorescence in situ hybridization (FISH) analysis, we detected IGH and FGFR3 translocation in 50-60% and 11% MM patients, respectively. These chromosomal rearrangements have been used to identify tumor plasma cells. FGFR3-translocated was found in 18%, 23% and 44% for B cells, PBs and plasma cells, respectively. Our work extends previous discoveries of chromosome translocation in malignant plasma cells, suggesting existence of heterogeneity in cells with FGFR3 translocation. Furthermore, FGFR3 translocation was accompanied by alteration of ARUKA. While B cell differentiation associated genes begin to express in the plasmablast stage, expression of multi-drug resistance genes occurs only in plasma cells in relapsed patients. Together, these molecular alterations likely constitute key elements of a sophisticated genetic program underlying the occurrence of MM. Conclusion: Our findings reveal the existence of genetically distinct cellular origins of MM cells from the HSC differentiation hierarchy within each MM tumor mass. FGFR3 translocation was accompanied by alteration of ARUKA, indicating FGFR3 translocation might cause CIN gene activation. Finally, we find that a small percentage of HSCs from MM patients could harbor oncogenic activation, thereby pointing to the need of a more vigorous quality control in autologous HSC transplantation for MM patients. Abstract:
This study puts forward a new concept, "population urbanization level forecast - driving factor analysis - urban spatial planning analysis" for achieving efficient and intensive development of urbanization considering human-land harmony. We analyzed big data for national economic and social development, studied the development trends of population urbanization and its influencing factors using the grey system model in Chengmai county of Hainan province, China. In turn, we calculated the population of Chengmai coming years based on the forecasting urbanization rate and the corresponding amount of urban construction land, and evaluated the urban spatial planning with GIS spatial analysis method in the study area. The result shows that the proposed concept is feasible for evaluation of urban spatial planning, and is meaningful for guiding the rational distribution of urban space, controlling the scale of development, improving the quality of urbanization and thus promoting highly-efficient and intensive use of limited land resource.

Abstract:
Local thinning of submarine pipeline always exists due to the complexity and dynamic of marine environment loads. However, the location and extent of thinning areas are random. Mechanical research on the randomness of local thinning of submarine pipelines is significant for pipeline’s stress analysis and failure evaluation. In this paper, finite element model of locally thinned submarine pipeline is established and based on the Monte-Carlo method, the effect of input random variables on the submarine pipeline’s reliability is analyzed. Probability distribution ranges of equivalent stresses are got and further, relationship equation between confidence level and stress limit has been obtained. The result more accurately reveals the relationships of locally thinned pipeline and its elastic modulus with equivalent stress, which can provide foundation for safety analysis of locally thinned submarine pipeline with internal pressure.

Abstract:
It is well-known that land subsidence seriously affects the regional social and economic development and becomes a world-wide problem. Xixi-Chengnan district, Jiangsu province is one of the most important area affected by land subsidence in China. The damage of the hazard to physical, social, economical and environmental systems has been increasing during recent years, which mainly caused by the long period groundwater overexpolitation in the area. This research presents a vulnerability assessment model based on grey correlation analysis (GCA) for the regional land subsidence in Xixi-Chengnan district of Jiangsu province, by the assessment results, the rank of relative vulnerability of each township in the study area can be obtained without weighting and aggregating the vulnerbaility indicators and with avoiding the subjectivity of weighting in general vulnerability evaluation methods. The study can be expected to raise public awareness of land subsidence risk and vulnerability, lay the foundation for risk decision-making, and provide theories and technological supports for taking comprehensive and active measures to prevent and alleviate land subsidence vulnerability.

Abstract:
Taking the Western District of Chengchao Iron Mine as engineering background, based on the GPS monitoring data of nearly 8 years and the record of the ground surface movement, the law of ground strata subsidence and movement is studied. The studies have shown that the surface movement caused by underground mining has the characteristics of time delay and displacement angle development has the law of jumping. The strata movement caused by mining is divided into the stage of overlying strata collapse and the collapse stage of surrounding rock to goaf. In the first stage, the vertical stress plays a leading role and rock failure is shown as a regular barrel collapse. In the second stage, horizontal tectonic stress plays a leading role and the collapse mechanism of rock strata can be explained by the cantilever beam theory. According to the mechanism of strata movement analysis, strata movement of Chengchao Iron is divided into six regions, which respectively included vertical subsidence area, toppling sliding zone, dumping area, deformation zone, cumulative deformation area and undisturbed zone. The results of this paper can provide a theoretical basis for the surface movement prediction of caving mining in similar metal mine.

Abstract. Over the past decades, paleoenvironmental studies in the Indian summer monsoon region have mainly focused on precipitation change, with few published terrestrial temperature records from the region. We analysed the distribution of isoprenoid glycerol dialkyl glycerol tetraethers (isoGDGTs) in the sediments of Lake Chenghai in southwest China across the Pleistocene–Holocene transition, to extract both regional hydrological and temperature signals for this important transition period. The lake level was reconstructed from the relative abundance of crenarchaeol in isoGDGTs (%cren) and the crenarchaeol′/crenarchaeol ratio. The %cren-inferred lake level identified a single lowstand (15.4–14.4 ka cal BP), while the crenarchaeol′/crenarchaeol ratio suggests a relatively lower lake level between 15.4–14.4 and 12.5–11.7 ka cal BP, corresponding to periods of weakened ISM during the Heinrich 1 and Younger Dryas cold event. A filtered TetraEther indeX consisting of 86 carbon atoms (TEX86 index) revealed that lake surface temperature was similar to present-day values during the last deglacial period and suggests a substantial warming of ∼4 ∘C from the early Holocene to the mid-Holocene. Our paleotemperature record is generally consistent with other records in southwest China, suggesting that the distribution of isoGDGTs in Lake Chenghai sediments has potential for quantitative paleotemperature reconstruction.

According to some actual cases of pile insertion analysis, combined with specialty norms and principles of soil mechanics in this paper, a few common misconceptions in pile insertion analysis of offshore platforms in Chengdao Oilfield are summarized. Type of common misconceptions, causes and adverse effects are analyzed, and countermeasures are put forward to correct misunderstandings and improve the accuracy of pile insertion calculation and analysis. Cases of analysis errors can be easily found because of unclear soil layer classification, difficultly recognized geotechnical properties, inaccurately divided layer uniformity, inadequately considered structure and soil interaction responses, etc. in offshore platforms pile insertion analysis, thus many unnecessary losses appear in oilfield production. By improving survey quality, using a variety of means of investigation, paying more attention to soil layer uniformity in well field, properly evaluating strength of laminated soil, doing anti-slip platform evaluation when pile is inserted too shallow, etc. can reduce errors effectively.

Abstract:
The twenty first century has witnessed an emerging research interest in island urbanization, which will set further pressure on island ecological vulnerability (IEV), especially in those islands with a fixed link to the mainland. In this contribution, the IEV of eighteen towns and townships in Chongming Island is assessed based on an “exposure (E)-sensitivity (S)-adaptive capacity (A)” framework and by means of the entropy weight method for determination of the weight of fifteen indicators. The assessment results show that: (1) An index system consisting of 1 objective, 3 sub-objectives, 8 elements, and 15 indicators can be established and tested to reflect the IEV to island urbanization; (2) The overall ecological vulnerability of Chongming Island to urbanization is at a rather low level, with only three out of eighteen towns and townships at a moderate high and high IEV level, while the spatial distribution of IEV surrounds Chengqiao, the seat of the district government, and radiates in a fan-shaped manner; (3) Chengqiao inevitably leads among the towns and townships in its IEV value, and its westward urbanization has adversely affected the IEV of adjacent towns Xinhe and Jianshe. (4) Chenjia’s moderate low level of IEV comes as a surprise to the authors, due largely to its proximity to Shanghai. Our proposed E-S-A framework and assessment model could be rationally applied to similar islands with fixed links to the mainland nationally and internationally, which is the major contribution of our study.

Abstract:
With the application of ANSYS finite element analysis software and environmental data in ChengDao oil field, a nonlinear finite element model of suspended submarine pipeline coupled with sea-floor soil is developed. Discarding the traditional and imprecise pipeline model with double clamped or simply supported ends, the model is founded on the theory of elastic foundation beam, in which the nonlinear effect between pipeline and sea-floor soil is considered. Base on the model we developed, the influences of span length and shear strength of soil at the ends of span on the span’s natural frequency are discussed. And the effects of span length, shear strength of soil and current velocity on the dynamic response of span are also analyzed in this paper. The model we developed in this paper fits the actual structure better, and the analysis is instructive for preventing and controlling the suspending of submarine pipeline.

Abstract:
The Haikou Macun Port is the harbor located in the Chengmai Bay of the southern Qiongzhou Strait, China. The principle for the constructing harbor and waterway is the layout should be conformed to the current field. The layout planning port area is studied from the view of hydrodynamics with a wave-current coupling hydrodynamic model of the Mike21 software package. The results show that the large area flow field is substantially unchanged after project construction and the effect is restricted to a small region near around the Macun Port.
