Full textGranja Deportiva Chicureo será un Centro Deportivo y Recreativo, que buscará satisfacer las necesidades deportivas infantiles y de esparcimiento familiar de los habitantes de las comunas de Colina y Lampa (Chicureo y Batuco), que tengan hijos desde los 5 hasta los 12 años de edad, de los estratos socioeconómicos medio y alto, con la infraestructura y organización necesaria con una gran oferta de deportes orientada a los niños y enfocada a un gran segmento de la población del sector la cual es la clase media y alta. El plan de negocios está inserto dentro de la industria del deporte infantil donde la tendencia en la actualidad tanto a nivel país como a nivel mundial es aumentar la actividad física deportiva y disminuir el sedentarismo infantil. Existe un mercado potencial de acuerdo al estudio de campo definido en el plan de negocios, el principal aspecto distintivo es entregar un concepto original y único en la zona en donde hay alto grado de sustitutos clubes deportivos, colegios y escuelas deportivas pero no existe un club familiar en el sector orientado a los niños en su totalidad donde convive un restaurant temático infantil con menú para niños y adultos, con la mayor variedad de actividades deportivas del sector, en un ambiente natural al aire libre con las prestaciones de seguridad y con una excelente ubicación y conexión vial. Para ello se evalúa el proyecto a 5 años plazo, el primer año la meta es obtener el 4,1% del mercado objetivo a través de estrategias con un plan de marketing agresivo principalmente apoyado por la red de contactos de destacados deportistas y apoyados por sponsor deportivos, luego obteniendo año tras año un 0,3% adicional promedio para así llegar al quinto año al 5,4%, con estas proyecciones según lo evaluado se puede concluir que el proyecto es viable y rentable tanto en lo financiero como en la proyección del negocio, donde se obtiene un VAN positivo de $17.368.000, una TIR mayor a la tasa de descuento del 15% y se obtiene un Payback (plazo de recuperación de la inversión) de 5 años, financiado en un 52% por sus socios y diferencia apalancada por crédito bancario.

Full textLiver Trans-Arterial Chemo-Embolization (TACE) is recommended for non-resectable, intermediate Hepatocellular carcinoma (HCC). The use of drug delivery platforms during TACE allows slow release of chemotherapy into the tumors’ arterial supply and prolonged exposure of the tumor cells while minimizing systemic exposures. Lipiodol-emulsion is one of the most widely used drug delivery platforms for TACE of HCC. However, the release of chemotherapy occurs very rapidly because Lipiodol-emulsions have poor stability. First, we have demonstrated that high stability (using synthetic surfactant as emulsifier) improves the therapeutic properties of Lipiodol-emulsion in a VX2-tumor rabbit model. A literature review also indicates that Lipiodol-emulsion must be water-in-oil (w/o) for better tumor’ uptakes. Next, we analyzed technical parameters that formulate in-vitro w/o emulsions with better stability without any additional emulsifier. We found that progressive incorporation of chemotherapy in the Lipiodol results in much more predictable w/o type and higher stability and that higher ratio of Lipiodol/chemotherapy is also a critical parameter for stability. Then, we develop a new concept of emulsion for TACE that uses Polylactic-co-glycolic acid (PLGA) nanoparticles (Nps) to stabilize Lipiodol and chemotherapy (Pickering-emulsion). In-vitro analysis demonstrated that Pickering-emulsion is the only drug delivery system (versus Lipiodol-emulsion and drug eluding beads) that allows slow and complete release of various chemotherapies (doxorubicin, irinotecan, oxaliplatin) but also of antibodies (anti-CTLA4), making Pickering-emulsion an universal carrier, not only for TACE but also for trans-arterial immunotherapy. Finally, TACE with oxaliplatin were performed in a VX2-tumor rabbit model to compare in-vivo Pickering-emulsion and Lipiodol-emulsion. Thanks to its slow drug release, Pickering-emulsion significantly decreased the systemic exposure but significantly increased the tumor/liver ratio oxaliplatin exposure

Full textSince many years, special attention has been made to synthesize amphiphilic cyclodextrins (CDs) to obtain new supramolecular assemblies able to interact with preformed lipidic structures such as liposomes. The aim of these investigations is to combine the size specificity of cyclodextrins for guests and their capacity to vectorize them. The final objects could be of importance to transport or target without toxicity relevant molecules such as drugs through biological membranes. If there are many chemical synthesis of amphiphilic cyclodextrins in literature, few works describe the obtention of modified cyclodextrins by enzymatic route. Fisrt, β-CD derivatives, used as substrats in reaction catalysed by lipases, were synthesized in classical chemical ways from native β-CD. They were modified by adding spacer arm with different chain length, ending with carboxylic acid, alcohol groups or amine function. Then, the grafting of lipidic moiety was realised during transesterification and amidification reaction using various lipases and acyl donor in non-conventional media. Analysis by NMR has allowed to evaluate amphiphilic cyclodextrin’s properties in water. It was shown that the reaction between cyclodextrin, acetaldehyde and lipase leads to controlled polyconcondensation reaction whose mechanism has been studied in the second part of this work. Finally, different routes were explored with the aim to synthesize diacylglyceryl-CDs and glycerophospholipidyl-CDs derivatives

Full textThe polarization of light, despite being known since long time, is recently at the center of renewed interest. More and more high technology companies in the fields of safety and information transmission are starting to exploit this property. One bottleneck for their use comes from the limitation in the light transmission of current methods of polarization (typically up to 45%). In order to overpass this physical limitation, one possible approach would be to use fluorescent materials emitting polarized light. However, the synthesis and purification of such materials is complex and obtaining both enantiomers is not always possible. The current work focus on a new synthetic pathway, possibly simpler and more versatile, using chiral hybrid or inorganic nano-helices and organic fluorophores interacting together. The aggregation of chromophores around the template will form chiral fluorescent nano-objects. The first chapter explains how chirality is present in many fields and at every scale, from molecules to daily objects. We will discuss the way of inducting or transferring chirality. The second facet of this work, light-matter interactions, will also be explained, concerning both absorption and emission of light, but also on how molecular assembly can affect these properties. We will study into detail the very particular case of circular dichroism and circularly polarized luminescence. Finally, we will see the existing systems that are used to obtain these properties and the drawback of these materials. In this work, we chose to use two systems. The first, constituted of organic nano-helices in a silica shell, has the advantage of using the organic template confined in chiral nano-space to induce chirality to the organic chromophores in interaction with them molecularly, but also through aggregation due to the confinement. The disadvantage being that this system is not robust toward environmental changes. The alternative approach is to use the silica shell as an inorganic template for the covalent grafting of fluorophores onto its surface. In the second chapter, the method for the synthesis of nano-structures is described, along with an explanation on the choice and synthesis of the chromophores used in this study. Finally, the characterization processes used are detailed. The third chapter will focus on the results we obtained when integrating achiral chromophores into hybrid helices or by grafting then onto the silica surface. We will see the importance of the intermolecular assembly and of the interaction with a chiral environment to obtain circular dichroism and circularly polarized luminescence through chiral induction. Various fluorophores are presented and compared allowing the understanding of the key parameters for chirality induction of each type of structure.In the last chapter, more complex systems are studied using molecules presenting chiroptical properties in solution state or having the ability to form self-assemblies showing such properties. The objective will be to tune the chiroptical properties of these chromophores, by the use of hybrid helices to force a specific organization. The last part will focus on the synthesis of fluorescent carbon based quantum dots using hybrid structures. These quantum dots, can retain the shape of the original structure and show circular dichroism or circularly polarized luminescence without needing to form a complex with an external source of molecular chirality

Full textTherapeutic embolization is nowadays a first line treatment for haemorrhage, arteriovenous malformation or tumors. Among different embolization agents, non degradable microspheres(Embozene®, Bead BlockTM,…) are the most employed thanks to their well calibrated spherical shape which allows good occlusion. In some cases including treatment of uterine fibroids or chemo-sensitive tumors, it may be interesting to achieve a temporary embolization to avoid definitive destruction of the tissue. Temporary embolization would be possible using biodegradable microspheres. The aim of our work was to develop degradable microspheres having all requiredcharacteristics to be used as embolization material (well calibrated in size, biocompatible, rigide enough to resist blood pressure but elastic enough to remain intact during injection through catheter). To this purpose, we have developed hydrolysable hydrogel based microspheres by suspension polymerization. A wide range of microspheres was synthesized by varying the type of crosslinker and composition of the polymerization medium. In vitro test showed that the microspheres have suitable characteristics to pass through catheter. Degradation studies revealed a rapid diminution of G’ modulus and the pH of the supernatants, accompanied by an increase of swelling ratio due to the hydrolysis of the crosslinkings. Although microspheres were not totally degradable as expected (since a side reaction had created non degradable crosslinking during the polymerisation), characterisations showed promising results that the degradation did occur within a suitable time scale requirements for temporal embolization

Full textO objetivo principal deste estudo foi caracterizar e compreender de forma clara e direta a cefaléia associada à malformação de Chiari tipo 1 (MC-1). Entrevistamos 19 pacientes com MC-1, dentre os quais onze apresentavam a associação de algum tipo de cefaléia e MC-1, sendo a dor de cabeça avaliada por uma ficha clínica padronizada, na qual constavam quesitos relativos à localização, duração, fatores precipitantes, caráter e evolução da dor. Dentre os 11 pacientes estudados, cinco foram homens, com idades variando entre 30 e 75 anos. A cefaléia foi mais prevalente na região occipital, seja de forma isolada ou associada a demais regiões da cabeça. Os principais fatores precipitantes da cefaléia foram: tosse, atividade sexual e outras formas de esforço físico. A cefaléia durava até três horas em todos os pacientes, com exceção de um que apresentava dor contínua. A cefaléia apresentou caráter pulsátil na maioria dos pacientes. Observou-se neste estudo que mais da metade dos pacientes apresentaram cefaléia associada à atividade sexual, o que tem sido pouco descrito na literatura, devendo a MC-1 ser sempre lembrada entre as possíveis causas de cefaléia associada à atividade sexual

Chapter Iprovides a brief introduction to metal-metal bonds and extended metal atom chains. In Chapter IIspin crossover properties of series of tricobalt EMACs based on [Co3(dpa)4X2] motifs arepresented. The influence of the axial ligands and the symmetry of the molecule was examined andcorrelated with the SCO properties. Full textDuring the course of a unique evolutionary process, the sensory systems of cetaceans (whales, dolphins and porpoises) have secondarily adapted to life in an aquatic environment. While the extraordinary acoustic capacities of these animals have been widely studied, surprisingly little is known about their chemosensory abilities. The results of some sparse behavioural studies suggest that cetaceans can detect molecules secreted by, for example, a prey or congener. In contrast, anatomical and genetic investigations have concluded that cetacean chemosenses are greatly reduced or even absent. This poses the question; have cetaceans truly lost the use of the sensory channels that play a major role in the feeding, orientation and reproduction behaviours of other marine predators? The main objective of this thesis was, therefore, to establish whether these animals are capable of perceiving biologically-relevant chemical cues and exhibiting a behavioural response. For this project, I studied the reactions of both toothed (Odontoceti) and baleen (Mysticeti) whales to various soluble or volatile food-related stimuli. I first worked with captive bottlenose dolphins (Tursiops truncatus) and showed they could discriminate soluble extracts of their piscine prey. I then developed a novel protocol to measure the responses (surface behaviour and movements) of cetaceans to chemical cues in their natural habitat. Using this protocol, experiments performed in the western Mediterranean Sea revealed that both bottlenose dolphins and pilot whales (Globicephala melas) react to dimethyl sulfate (DMS), a volatile molecule found in areas of high primary productivity. Using the same protocol, I subsequently measured the reactions of a mysticete species, the humpback whale (Megaptera novaeangliae) to DMS and to krill extract (their natural prey) in three different locations: in their reproduction zone in the Indian Ocean, and in their feeding grounds in the North Atlantic and Antarctic Ocean. I found that the humpback whales reacted with an increased respiratory rate to both stimuli and that the krill extract also attracted them (chemotaxis). As the results of the behavioural studies suggested the presence of functional chemical senses, I initiated two further studies; (1) a neuroanatomical and immunohistochemical exploration of tissues in the oral and nasal pathways, in order to identify potential chemosensory receptor cells in four species of odontocete, and (2) a chemical analysis of odontocete secretions (urine or feces) in search for potential semiochemicals. Overall, this innovative and multidisciplinary research project revealed that chemical signals are potentially very important in the behavioral ecology of cetaceans. Moreover, these results contribute not only to our fundamental knowledge of the sensory biology of these animals, but they could also have important practical implementations in the conservation of these iconic and threatened species

Full textAlthough the biological importance of proteins and nucleic acids has long been known, it is only in recent years that scientists have realised the importance of sugars in many biological processes. Today, the major drawback, which is stopping the fast development of glycobiology, remains the lack of complex sugars perfectly pure and well characterised. To address this problem, we decided to combine the advantages of a supported synthesis with the use of enzymes. During such chemo-enzymatic syntheses, two methodologies may be used : either the oligosaccharide is made step by step on the support, using successive glycosyltransferases ; or the donor substrate is bound to the support and the acceptor, present in solution, is subsequently coupled to the support using a glycosyltransferase. Using the latter method, we synthesised a supported nucleotide sugar, GDP-fucose, a substrate of fucosyltransferases. To achieve this, after having proved the viability of the synthesis on model compounds, we functionalised and characterised a new soluble and dendrimeric support, a derivative of poly(ethylene glycol). This ultimately allowed us, after the binding of a synthon from L-galactose, to do the supported synthesis of GDP-fucose, which was obtained with a global yield of 37% after 8 chemical steps. Subsequently, our supported nucleotide sugar was used as a dollar substrate of fucosyltransferase 3 and gave us, after cleavage using nickel boride, a fluorescent derivative of the trisaccharide Lewis [a]. This work represents one of the first real chemo-enzymatic supported syntheses of an oligosaccharide and provides a future basis for many other oligosaccharide syntheses using our supported GDP-fucose

The purpose of the study deals with the synthesis of mono-glycosylcyclodextrines. Due to their high solubility, the latter are of great interest for chiral chromatography and for drug vectorisation. In order to reach this goal, three chemo-enzymatic strategies were developped : Aldolisation catalyzed by an aldolase. Transglycosylation catalyzed by glycosidases Amino-reduction in the presence of b-CD-NH2 and of 6-oxo-galactosides. In this work, we have described two new and efficient routes for the synthesis of O- and N-galactosyl-CD. A particular case of N-glycosylation, observed with 6-oxogalactose, showed the synthesis of b-cyclodextrinyl-azepane. The study of this reaction is completed by means of molecular modeling

Spiritual bypassing is the tendency to use spiritual beliefs, teachings, and practices to avoid dealing with one’s psychology, painful feelings, unresolved wounds, and developmental issues. Chiron, like his mentee Asclepius, embodies the Wounded Healer archetype. Chiron’s wounding and healing journey can serve as a roadmap for spiritual practitioners on how to navigate out of bypass and deepen their path toward self-realization and individuation. Developing an archetypal awareness of one’s wounds appears to reveal the aspects of one’s psychology defended against in spiritual bypass. This emerging roadmap and lessons from Chiron’s journey may help therapists, healers, and spiritual teachers accelerate their own path and assist clients.

Full textChiari malformation type I (CMI) is a congenital disease that has been traditionally defined as the chronic tonsillar herniation of at least 3 mm below the foramen magnum. It is characterized by an overcrowding of a normally developed hindbrain within a hypoplastic PCF, although both age at presentation and symptoms show high variability. The etiology of the disease remains unknown, but the most accepted hypothesis is that CMI is the result of an insufficient development of the paraxial mesoderm. Several evidences suggest that, at least in a subset of CMI patients, genetic factors may play an important role. This project focused on the identification of genetic risk factors for CMI, including genes responsible for the mendelian and also the complex forms of the disease. To achieve this goal, two different strategies were followed: 1) Identification of susceptibility genetic variants in CMI patients using a casecontrol association approach. 2) Genetic analyses of monogenic forms by a mutational screen of the candidate gene SUZ12 and by exome sequencing in two CMI patients ofthe same family. We recruited 530 CMI patients from 450 unrelated families. Given that tonsilar herniation can be caused by five distinct mechanisms, we developed a probability model to establish a diagnosis with 93% confidence in those patients with a hypoplastic PCF. This diagnostic tool allowed to define a clinical homogeneous group for further genetic studies. A cephalometric oropharynx morphometric study was also performed to identify the possible underlying anatomical anomalies leading to less known symptoms. The results of the genetics association study paint to a putative role for defective retinoic acid signalling and fetal vasculogenesis in causing hypoplasia of the basichondrocranium. However, when we studied the monogenic forms, we did not find any causal variant.

Full textConsidering the increasing appearance of pigmentation disorders caused by UV exposure, the development of new whitening agents is a major challenge for the cosmetics industry. Consumers are turning to ecoresponsible products, it is urgent to develop new methods of biocatalysis for the access to new depigmenting agents. Some glycosides, such as arbutin, have whitening properties which are still limited because of therapid hydrolysis of the O-glycosidic bond. The challenge of this project is synthesize analogues of this molecule in thioglycoside series. Endeed, this bond allows a greater stability against the hydrolysis. The mutation of anative glycosidase to dictyoglomus thermophilum gives access to a thioglycoligase. This makes possible the enzymatic synthesis of arbutine analogues. Six molécules were synthesized and tested as depigmenting agents. The synthesis methodology was then applied to the preparation of thioglycolipids. These moleculesare known for their hydrogellating properties allowing access to thermoreversible materials. Five molecules have been identified as hydrogellating agents. The rheological, thermal and structural properties have been characterized and showed significant differences depending of the compound structure. Lastly, the preliminary results on the formulation of a product that is both antipigmenting and texturizing are promising for the validation of an smart material concept for the cosmetic industry

Full textThe aim of fuis study is to understand coupled phenomena that occur in swelling porous materials like clays. Electro-chemo-hydro-mechanical contributions are taken into account to analyze transfers in such minerais. Ln a first fart, an general discussion is proposed to introduce mineralogical and physico- chemical considerations of clayey media. An important objective of this chapter is to show the crucial role of the microstructure. Ln a second part is presented an imbibition test in a MX80 bentonite powder. The hydraulic diffusivity versus water content curve's decrease is explained thanks to a double porosity model that shows the progressive collapse of mesopores due to swelling effects at the microscale. Thus a multi- scale analysis is necessary to weil describe clayey media behaviour. The third chapter exposes such a multi-scale modelling (periodic homogenization). It is based on the double-layer theory and introduces an innovative concept of virtual electrolyte solution. First numerical results are given in a simple geometry (parai lei platelets). Ln the next part are proposed numerical simulations of two kinds : response of the system to a chemical gradient and simulation of electro-osmosis. The end of this chapter puts into relief the necessity to integrate pH effects in the model. In the last part, chemical surface exchanges are incorporated in the modelling to understand pH and ionic force roles in electro-osmotic process

Full textThis thesis describes Chiron, visualization system which helps programmers detect memory system bottlenecks in their shared-memory parallel applications. Chiron is different from most other performance debugging tools in that it uses three-dimensional graphics techniques to display vast amounts of memory-performance data. Both code-and data-oriented information can be presented in several views. These views have been designed to help the user detect problems which cause coherence interference or replacement interference. Chiron’s interactive user-interface enables the user to manipulate the views and home in on features which indicate memory system bottlenecks. The visualized data can be augmented with more detailed numerical and correlations between the separate views can be displayed. The effectiveness of Chiron is illustrated in this thesis by means of three case studies.
