This thesis describes studies into the pathogenesis of Cholesterol—related early vascular injury. In addition, this thesis examines the potential reversibility of this injury with 2 agents: a) simvastatin, an HMGCoA reductase inhibitor, in a model independent of any reduction in lipids and 2) high dose aspirin. Hypercholesterolaemia led to a decrease in NO bioavailability, in association with a decrease in the enzyme, endothelial nitric oxide synthase and increased oxidative stress. Statins are widely used to treat hypercholesterolemia. Statins inhibit cholesterol biosynthesis, thereby activating genes involved in cholesterol homeostasis, which are under the control of the Sterol Regulatory Element (SRE). Statins also have cholesterol-independent beneficial cardiovascular effects mediated through the phosphoinositide 3-kinase (PI3-K) / Akt signaling pathway and by inhibition of protein prenylation. Because statins inhibit the synthesis of isoprenoids, they can act by inhibiting the small signaling GTPases Ras and Rho, which require post-translational prenylation to become

There is worldwide concern over the possible estrogenic effects of organic chemicals on a variety of wildlife and indeed on humankind. In the U.K., estrogenic compounds in sewage treatment works (STW) effluents have been implicated in causing the increases in egg yolk protein production observed in caged male trout and other fish species. At the initiation of the present study, few of the estrogenic compounds in STW effiuents had been recognised, although circumstantial evidence suggested that steroidal hormones were primary candidates.
