The essay deals with the famous Florentine author and courtier Ottavio Rinuccini (1562–1621), more specifically with his librettos. Following an outline of Rinuccini’s career as a dramatist in the period 1579–1621, the prologues to his successful La rappresentazione di Dafne (1594/95) and his never performed Il Narciso (1608) are discussed. Abstract Background: We present the development of NECTIN4-CAR T cells targeting bladder cancer and evaluate the ability of thiazolidinedione, a class of anti-diabetic drugs, to specifically upregulate NECTIN4 expression and expand the therapeutic window of NECTIN4-CAR T cells. In addition, we assess efficacy of NECTIN4-CAR T cell therapy against a model of bladder cancer with acquired resistance to enfortumab vedotin (EV). Methods: NECTIN4-targeting chimeric antigen receptor (CAR) constructs were designed and cloned into a second-generation EQ-28ζ CAR backbone construct. Human primary T cells

Abstract Chimeric Antigen Receptor (CAR) T cell therapy represents a significant advance in the treatment of hematologic malignancies, as demonstrated by the high response rates and improved survival in diseases such as acute lymphoblastic leukemia (ALL), non-Hodgkin lymphoma (NHL), Diffuse large cell lymphoma (DLBCL) and chronic lymphocytic leukemia (CLL). The FDA has approved several CAR T cell therapies, including some approaches targeting CD19 (CART19) for adult and pediatric patients with B-cell malignancies. Response rates for CD19 CART therapy have been quite promising, reaching 80% ove

3-Aminobutanoic acid is an unnatural amino acid and by analogy with other amino acids, is represented in its zwitterionic form by R-CH(COO¯)NH3+ (where side group R = aryl, alkyl or any other group) in solid phase. Experimental IR and Raman modes near 3029, 2871, 1577, 1288 cm-1 measured for 3ABA are strongly in favor of the zwitterionic dimer structure formed of –N-H∙∙∙O–C bonding between –NH3+ and anion – CO2¯ groups. Zwitterionic monomer and dimer structures at B3LYP/6-311++G(d,p) level are computed. It has been found that the stable zwitterionic monomer and dimer structures are possible

A new type of multiphase nanoparticle-reinforced TiAl matrix composites ((Ti2AlC + Al2O3)p/TiAl composites) was successfully prepared by vacuum hot-pressing sintering using Ti powder and Al powder, which were ball-milled with different contents of stearic acid (CH3(CH2)16COOH). The component, microstructure, reaction mechanism, and mechanical properties were studied. The results indicated that the composites prepared by adding stearic acid as a process control agent during the ball-milling process not only contained γ-TiAl and α2-Ti3Al phases but also Ti2AlC and Al2O3 phases. Abstract Intravesical therapies have been the mainstay of non-muscle invasive bladder cancer (BC) management; however, their efficacy is limited by toxicities, recurrences, and supply shortages. Consequently, many patients are recommended cystectomy, which is fraught with high complication rates. Thus, bladder-sparing treatments present a major, unmet clinical need. Chimeric antigen receptor (CAR) T cell therapy, wherein T cells are engineered to express an artificial receptor to a target, is an immunotherapeutic approach with efficacy in hematologic malignancies. Given that metastatic CRPC has a tumor environment lacking immune infiltration and resisting immune checkpoint therapies, combination strategies that actively bring immune cells into the tumor are urgently needed (Choudhury et al., 2024). Abstract In the evolving landscape of glioblastoma (GBM) treatment, clinical trials exploring chimeric antigen receptor (CAR) T cell therapies have shown promise; however, their therapeutic efficacy remains constrained, particularly due to the immunosuppressive tumor microenvironment (TME). Emerging evidence highlights that the TME, especially myeloid-driven suppressive networks, plays a central role in resistance to CAR T cell therapy. Despite progress in elucidating these mechanisms, significant ambiguities endure, particularly regarding patient-specific TME heterogeneity and its impact on t
