Abstract:
This tavern story about an 1832 Saturday night on the town in Brantford, Upper Canada, addresses the complexities of racialized relations in “public places” generally and the tavern’s bar room in particular. It juxtaposes tavern-goers who engaged in “heterogenous” sociability with the “‘high pressure’ prejudice” of a “‘Yankee’” barkeeper. It challenges us to understand what such moments of multiracial public life meant in a society permeated by racialized thought and practice. There was a strange contradiction between White settlers’ marginalization of Black and First Nations peoples and the sometimes easy accommodation afforded them in the public houses. Although accommodation to people of colour was also illegally, and sometimes violently, denied, tavern stories complicate historical interpretations focusing on conflict. Without questioning these analyses, or the evidence supporting them, the stories suggest that something more subtle was also going on. They invite serious attention to the colony’s many taverns as sites where people chose to relax racial boundaries as often as they chose to enforce them. Maybe it was just the whiskey and the wine; without comparable work on other public spaces, the typicality of a tavern-based history will remain an open question. But because “Indians” as well as the “blacks and whites” all went there, the taverns show how race, as one socially constructed category, shaped ordinary, everyday human interactions.

Abstract:
Abstract An important aspect of leukemia biology is the elucidation of the spectrum of chromosomal abnormalities and molecular mutations that cooperate in the pathways leading to leukemogenesis. To identify those chromosomal abnormalities that are associated with drug resistance and a poor outcome, we analyzed a cohort of 3,438 pts (primary MDS, 1155; AML de novo, 1832; and therapy-related MDS/t-AML, 451) ascertained between 1970–2007 at the University of Chicago. 866 (25%) had either -5/del(5q) (276, 8%), -7/del(7q) (313, 9%), or both (277, 8%). The incidence of loss of 5q, 7q, or both was 19% in primary MDS, 20% in AML de novo, and 61% in t-MDS/t-AML. Of note, 71% of the karyotypic abnormalities in these pts led to loss of chromosomal material. Balanced rearrangements accounted for 10% of the additional abnormalities, and the remaining 19% led to a gain of material. Abnormalities of chr 5 were associated with karyotypic instability. Pts with -5/del(5q) had an average of 7 karyotypic abnormalities, whereas pts with -7/del(7q) had an average of 3 abnormalities. Pts with both -5/del(5q) and -7/del(7q) had an average of 10 abnormalities. Only 13% of pts with an abnormality of chr 5 had -5/del(5q) as their sole karyotypic abnormality, whereas 41% of pts with an abnormality of chr 7 had -7/del(7q) as their sole karyotypic abnormality. In addition to increased complexity of the karyotype, abnormalities of chr 5 were associated with specific recurring abnormalities: -17/loss of 17p, 39%;-3/loss of 3p or 3q, 30%; -18/loss of 18q, 24%; -12/loss of 12p, 24%; +8, 23%; -13/del(13q), 18%; -20/del(20q), 18% (values refer to the percentage of each abnormality among pts with 5q loss). The relative frequency of these abnormalities in pts with -7/del(7q) ranged from 4–8%, and no specific abnormality was observed in more than 10% of cases with loss of 7q. These observations extend previous studies which demonstrated a high incidence of abnormalities of 17p, TP53 mutations, and complex karyotypes in t-MDS/t-AML characterized by abnormalities of chr 5 (Pedersen-Bjergaard J. NEJM152:1491, 2005). Our observations, together with those in the literature, suggest that different molecular pathways are involved in the pathogenesis of primary MDS/AML and t-MDS/t-AML associated with abnormalities of chr 5 or 7. The subgroup of pts with abnormalities of chr 7 without chr 5 abnormalities typically has mutations of the RAS pathway (KRAS1, NRAS, NF1, PTPN11), methylation of the promoter of CDKN2B/p15, and a poor prognosis. In contrast, pts with loss of 5q, with or without abnormalities of chr 7, are characterized by complex karyotypes, TP53 mutations and -3/loss of 3p or 3q, +8, -12/loss of 12p, -13/del(13q), -17/loss of 17p, -18/loss of 18q, and -20/del(20q), and a poor prognosis. Emerging technologies, such as high throughput genomic analysis, may facilitate further delineation of the genetic pathways leading to myeloid diseases. »When the Shadow comes nearest..« By Flemming Lundgreen-NielsenThis small contribution deals with two intricate lines from stanza 5 of Grundtvig’s 1824 poem .The Land of the Living.. They are usually taken to mean that when likeness in the shape of shadows comes nearest the real thing, the little ones observing them weep, because in human poetry and arts which strive to reflect eternity similarity is not and can never be identity. Chr. Thodberg has repeatedly since 1971 suggested a different understanding of the lines to the effect that »ligest« (i.e. .most similar to.) may be interpreted as »most vertical«, referring to the shrinkage or entire disappearance of shadows at noon, when the sun reaches zenith. Thodberg departs from an off-hand commentary by Grundtvig about this natural fact in a sermon delivered on 27 March 1823. The author tries to demonstrate that Grundtvig's varying usage of theword shadow (»Skygge«) both before and after 1824 makes it impossible to arrive at an unambiguous determination regarding the word in the said lines. Furthermore, the superlative degree »ligest« meaning »most vertical« neither seems to have been recorded in the language of Grundtvig and his contemporaries nor in older periods.Contrarily, »ligest« meaning »most similar to« can be found in ancient Danish proverbs which Grundtvig studied extensively from 1816 and until he edited and published a collection of them in 1845, and it can also be located in contemporary literature. Returning from there to the meaning of shadow, the author by means of quotes from Grundtvig’s brief but precise historical evaluations of Plato and his philosophy (in 1812 and 1833) is inclined to support the traditional interpretation that the shadows making little ones weep originate in the famous cave metaphor in the fifth book of Plato’s dialogue The Republic. Finally, two other details deriving from Grundtvig’s studies of Danish proverbs in the Peder Syv edition (1688) are mentioned to throw light on a couple of nebulous expressions in two other lyrical poems by him.

Abstract Charybdis lucifera (Fabricius, 1798), Demania reynaudii (H. Milne Edwards, 1834), Galene bispinosa (Herbst, 1783), and Neodorippe callida (Fabricius, 1798) are documented for the first time from the territorial waters of Bangladesh. Before this account, the families Dorippidae MacLeay, 1838 and Galenidae Alcock, 1898 were also unknown to the country. All specimens but Neodorippe callida were sampled from St. Martin’s Island, a sedimentary island on the continental shelf. We encountered N. callida from Dublar Char Island, a mangrove block extending into the Bay of Bengal. We collected D. reynaudii from a beach food stall, being displayed apparently unaware of the species’ toxicity, which seems not to be known in the region; the other species were sourced from artisanal seining activities. Here, taxonomic traits of these species are discussed respective to close congeners, and with necessary remarks on their locality.

Abstract:
In 1822, Robert Wilmot, the undersecretary of state for the colonies, introduced a bill to unite Upper and Lower Canada in the British House of Commons. In doing so, he was proposing not just an intercolonial union but a new constitution. He believed that because the Canadas’ constitution was granted by an Act of Parliament in 1791 it could be changed by Parliament without colonial consultation or consent. Whig parliamentarians and colonists contested this interpretation and raised questions about consent and the status of colonial constitutions. These debates in the 1820s reveal just how muddied thinking about colonial constitutions and consultation had become. Lower Canadian opposition to the bill has received significant attention from historians, yet this attempt at constitutional change also forced Upper Canadians to take unexpected positions, including interpreting the 1791 act as a written charter. Through these transatlantic debates, they were continuing to fashion a settler interpretation of British constitutionalism. This article traces these ideas by examining the movement of news and rumours through emerging, intertwined colonial and imperial public spheres, which illustrate the permeability of the line between public and private information. Inserting this failed constitution-writing effort into the longer history of Canadian constitutions sheds light on the limits to imperial intervention and encourages a broader rethinking of Canadian constitutional histories and the role of empire in a long nineteenth century.

By late May of 1835, unrest in Bytown had reached unprecedented proportions. All winter, the people of the town, the entrepôt of the Ottawa timber trade, had been bracing themselves, awaiting the annual visitation, the annual affliction, of the raftsmen who came each spring from high up the Valley to roister and riot in the streets of Bytown. Like the freshets in the streams, the raftsmen and social disorder arrived each April and May. But never before had their coming brought such organized violence as it did in 1835. For the Irish timberers, now had a leader, and a purpose. Peter Aylen, run-away sailor, timber king, ambitious schemer, had set himself at the head of the Irish masses, had moulded them into a powerful weapon. He had given them a purpose: to drive the French Canadians off the river and thus guarantee jobs and high wages in the timber camps to the Irish.

2509 Background: As a promising approach for some cancers, chimeric antigen receptor T cell therapy has limited success in solid tumors. Claudin18.2 (CLDN 18.2) is a stomach-specific isoform of Claudin-18, and highly expressed in gastric and pancreatic adenocarcinoma, the advanced form of both of which have urgent unmet medical needs. We previously developed and demonstrated ability of CLDN 18.2-specific CAR (CAR-CLDN18.2) T cells to eradicate CLDN 18.2-positive gastric cancer xenografts without obvious on-target off-tumor toxicity (Huang J. JNCI 2018). Methods: In this single-arm, open-label, first-in-human phase I pilot study (NCT03159819) to investigate the safety and explore the efficacy of the autologous CAR-CLDN18.2 T cells, patients with confirmed CLDN 18.2 positive advanced gastric or pancreatic adenocarcinoma aged 18 to 70 years received 1 or more cycles of CAR-CLDN18.2 T cell infusion(s) after lymphodepletion pretreatment (fludarabine and cyclophosphamide, with or without nab-paclitaxel) until disease progression or presence of intolerable toxicity. Adverse Event (AE) grade categorization is according to CTCAE 4.0, and tumor response was assessed per RECIST 1.1. Results: As of November 30th, 2018, 12 subjects with metastatic adenocarcinoma (7 gastric and 5 pancreatic) were treated with 1–5 cycles (total of 0.5 - 55 X 108) of CAR-positive T cells infusions. There were no serious adverse events, treatment-related death or severe neurotoxicity occurred in the study. No grade 4 AEs except for decreased lymphocytes, neutrophils and white blood cells. All cytokine release syndromes observed were grade 1 or 2. Among the 11 evaluable subjects, 1 achieved a complete response (gastric adenocarcinoma), 3 had partial responses (2 gastric adenocarcinomas and 1 pancreatic adenocarcinoma), 5 had stable disease and 2 had progression of disease. The total objective response rate was 33.3%, with median PFS of 130 days estimated using Kaplan-Meier method [95% CI (38, 230)]. Conclusions: This clinical study indicated that CAR-CLDN18.2 T cell therapy were safe and well tolerated and may have promising therapeutic efficacy in patients with advanced gastric and pancreatic adenocarcinoma. A concept of a car-following system that consists of a car-following equation together with a set of constraints is appropriate for micro-simulation models. Recognition was given to the asynchronous condition in the leader-follower relationship as it relates to the internal dynamics in simulation models. The Pitt car-following equation and criticisms of it were evaluated. Two modifications to the Pitt car-following equation were introduced that address the criticisms. The first modification consists of a change in the method by which the perception-reaction time is handled, eliminating the requirement that it must be much less than the simulation time step and removing a discrepancy between the application of car-following and emergency deceleration. The second modification consists of a multiplicative factor, K, that addresses issues related to overdamping and the dead band, which have been raised concerning the original formulation of the Pitt model. An Excel spreadsheet was used to implement the car-following system, and a series of scenarios were simulated. The results of the scenarios indicated that the addition of the factor K has a substantial effect on the behavior of vehicles in a platoon when the leader of the platoon is subjected to a deceleration. Issues of calibration and validation were also addressed.

Price and service elasticities of passenger car travel are estimated using stated preference and sample enumeration methodology. Moreover, the effects of hypothetical travel demand management policies are analyzed by changes on modal share using the elasticity estimates. The elasticity of passenger car travel with fuel price is estimated to be within the range of −0.078 to −0.171. The parameter estimate of the fare variable is estimated to be statistically insignificant in every subgroup of car users. This finding suggests that fare policies are relatively ineffective for increasing transit modal shares in Korea. Meanwhile, car users' responsiveness to changes in parking costs is estimated to be much higher than for fuel cost. This suggests that parking regulations or pricing policies may be effective in reducing travel by passenger car. The elasticity with in-vehicle time, which is a key attribute of public transport amenities, is estimated to be particularly high, implying that policy measures such as introducing express buses or express urban trains could be effective in reducing passenger car travel. The demand elasticity of service levels of mass transit represented by the degree-of-crowdedness proxy turns out to be very high. Reducing crowdedness in public transit can be very effective in attracting more passengers, or at least in retaining current patronage.

Po klęsce powstania listopadowego w przestrzeni wiejskiej Królestwa Polskiego pojawiły się donacje należące do rosyjskich wysokiej rangi oficerów i carskiej elity urzędniczej. Car Mikołaj I nadał im te dobra przede wszystkim za zasługi oddane podczas tłumienia polskiego powstania i wierną służbę. Jedną z największych donacji otrzymał generał piechoty Karol Wilhelm hrabia Toll, pochodzący z tzw. Bałtów (szlachty bałtyckiej). Był on jednym z najbardziej znaczących dowódców carskich w wojnie polsko-rosyjskiej w 1831 r. Otrzymał on w 1836 r. część dóbr skarbowych Uniejów o powierzchni ponad 5500 mórg. Donacja ta znajdowała się w posiadaniu rodziny von Tollów przez 80 lat. Po klęsce powstania styczniowego otrzymali oni jeszcze niewielką donację Grzybki. Ponadto byli właścicielami majątku ziemskiego Stemplew. Kolejne trzy pokolenia tej rodziny coraz bardziej wiązały się z regionem, a po utworzeniu niepodległej Rzeczypospolitej potomkowie pierwszego donatariusza pozostali w naszym kraju. Jednak nie byli już właścicielami donacji, którą przejęło państwo polskie w 1919 r. W pierwszej części niniejszego artykułu dokonano syntetycznej charakterystyki dóbr ziemskich będących w rękach hrabiów Tollów. Natomiast w drugiej części podjęto próbę odtworzenia genealogii uniejowskiej linii tego rodu oraz przybliżono nieraz skomplikowane losy potomków pierwszego donatariusza.

The queue discharge problem at a signalized intersection was analyzed with application of the modified Pitt car-following system. The Pitt car-following system was implemented on an Excel spreadsheet in the form of a prototype simulation model. The situation consisted of a single intersection in which discharging vehicles were unconstrained by downstream conditions. It was asserted that the car-following parameters used in the queue discharge problem are significantly different from those used in the uninterrupted-flow problem and that this is consistent with the two-fluid model. A number of scenarios were executed for each situation, using the spreadsheet implementation to examine a number of issues, including the effect of the following: free-flow speed, car-following parameter variations, vehicle length, heterogeneous traffic streams, and lane changing. It was shown that all of these issues have significant effects on discharge headways and that the Pitt car-following system has a far wider range of applicability than the simple equal headway model. The intuitiveness of the findings and calibration and validation issues were also addressed.

Abstract:
Crashes involving large trucks and passenger cars are important topics for research and countermeasure development since they represent more than 60% of all fatal truck crashes and because the passenger car occupant is much more likely to be killed. This study ( a) examined “fault” in total car–truck crashes using North Carolina Highway Safety Information System (HSIS) data for comparison with fault analyzed in previous studies of fatal crashes, ( b) used general estimates system (GES) crash data to verify unsafe driving acts (UDAs) identified by expert panels in past studies, and ( c) used North Carolina HSIS data to identify critical combinations of roadway facility type, roadway location, and crash type based on “total harm”—a measure combining both the frequency and severity of the crash. Fault in total North Carolina car–truck crashes was found to differ significantly from past fatal crash studies, with the truck driver being at fault more often than the car driver both overall and in certain crash types. Car drivers continue to be at fault much more often in head-on and angle crashes. While it was not possible to analyze all UDAs identified in prior studies, when possible, the current analyses revealed differences between the GES crash data results and the expert-based results, pointing to the need for better UDA methods if they are to be used to target treatments. Finally, using the total-harm analysis with North Carolina car–truck crashes indicated that undivided rural arterials and collectors should be primary targets for further investigation and for treatment.

Nine months into the introduction of car sharing in San Francisco, California, an estimated 7% of members' trips and more than 20% of vehicle miles traveled were by shared-use vehicles. Evidence suggests that access to shared cars is stimulating motorized travel. Most members do not own cars, and many appear to be leasing vehicles in lieu of walking and biking. Car-share vehicles are used more for personal business and social-recreational travel than for nondiscretionary, routine travel such as to work or school. Shared cars are generally not used during peak periods or to dense settings well served by transit, such as downtown. In this sense, car sharing appears to be stimulating a resourceful form of judicious automobility. Users are accruing substantial travel-time savings and willingly pay market prices for these benefits.

Abstract In every aspect of life, 19th century Spain was not peaceful. Despite tumultuous times, members of the Real Academia Española (Royal Spanish Academy) managed to publish a new edition of the Diccionario de la lengua castellana ( drae 1817). A notable achievement, they published one of the best editions of the academic dictionary ever. In this paper, I analyze the introduction of words and their meaning from the region of Andalusia. The diatopic labels corresponding to this Spanish region are used widely. I explain the existence of this dialectal lexicon in drae (1817) by connecting it to historical events around 1812. This was the same year when liberal politicians and intellectuals established the Spanish Constitution of 1812 (Constitución política de la monarquía española) in the Andalusian city of Cádiz.

Three car-following models were compared: the Greenshields single-regime model, the Pipes two-regime model, and a four-parameter single-regime model that amalgamates both the Greenshields and Pipes models. The four-parameter model proposed by Van Aerde and Rakha is less known but is currently implemented in the INTEGRATION 2.30 software. The Greenshields and Pipes models were considered because they represent state-of-the-practice models for several types of microscopic and macroscopic software. The Greenshields model is widely used in macroscopic transportation planning models. In addition, the Pipes model is implemented in a number of microscopic traffic simulation models including CORSIM and VISSIM. Steady-state car-following behavior is also related to macroscopic traffic stream models to develop calibration procedures that can be achieved using macroscopic loop detector data. The study concluded that the additional degree of freedom that results from including a fourth parameter (Van Aerde model) overcomes the shortcomings of the current state-of-the-practice traffic stream models by capturing both macroscopic and microscopic steady-state traffic behavior for a wide range of roadway facilities and traffic conditions. Also developed was a procedure for calibrating the Pipes car-following model using macroscopic field measurements that can be obtained from loop detectors. Although this calibration procedure does not overcome the inherent shortcomings of the Pipes model, it does provide an opportunity to calibrate the CORSIM and VISSIM car-following behavior to existing roadway conditions more efficiently and without the need to collect microscopic traffic data.
