In this study, the effects of certain chromatographic conditions on various modified and unmodified oligonucleotides were investigated. At the forefront of this study was the investigation of a new Ion-pair Reversed-phase liquid chromatography (IP-RPLC) method, that had the potential to replace a previously established triethylammonium acetate (TEAA) IP-RPLC method developed for oligonucleotide separations. This thesis incorporates studies about the structural characterization of proteins in solution mainly by means of small angle X-ray scattering (SAXS). The major part of the thesis has as main focus the study of the effect of stress conditions on the conformation of Human Serum Albumin (HSA), the most abundant plasma protein.
