Full textCounter-current chromatography (CCC) is a preparative purification technique that works with the twoliquid phases of a biphasic liquid system. One phase is used as the mobile phase when the other phase isused as the stationary phase. There is no solid support: centrifugal fields are used to obtain a support-freeliquid stationary phase. This work contains an exhaustive bibliographic study of what can be found in theliterature concerning continuous chromatographic processes. The multi-dual-mode (MDM) process was foundto be the best one able to purify large amount of crude mixtures. The MDM method starts with a classicalseparation of the mixture followed by a switch of both the liquid phase nature and the flowing direction. Themobile phase flowing e.g. in a descending direction becomes the stationary phase. The previous stationaryphase becomes the mobile phase flowing in the ascending direction (or vice versa). The purified compoundsof the introduced mixture are eluted at one side of the column or the other according to their polarity. TheMDM method was used to purify a crude sample of Coomassie Blue: the polar part of the dye was eluted atthe column top (or head) and the apolar part at the column bottom (or tail) while the essential part of the dyewas trapped inside the CCC column. The work also presents a new small volume (30 mL) hydrostatic CCCcolumn. It is shown that this column could be used to test quickly the potential of a given biphasic liquidsystem

Full textPreparative two-dimensional chromatography is gaining interest in the elucidation of complex samples as it allows the collection of a large number of molecules with high recovered purity and quantity. While the second dimension is often selected to be liquid chromatography (LC), centrifugal partition chromatography (CPC) is a technique with multiple advantages representing a suitable first dimension. In order to purify several molecules of interest in plant matrices, the comprehensive CPCxLC represents a technique with high potential. After explaining its interest and the issues related to the preparative separation in comprehensive mode, the development of such a separation is studied according to three axes. Firstly, a purification of two targeted molecules in Edelweiss plant is carried out at industrial scale thanks to the realization of 2D-contour plot. This application allows to expose the interest of the separation and to highlight the locks related to the conditions of total transfer of the fractions in second dimension. In a second part, the comprehensive CPCxLC separation is developed with the total transfer of the sample in second dimension applied to the purification of five target compounds from Edelweiss plant. The key points of the CPCxLC separation, namely the sampling time and the second dimension transfer, are studied with regard to the LCxLC separation in order to ensure a separation quality allowing the total recovery of the compounds. Finally, the third part consists in the implementation of a CPCxLC system selection methodology based on the quantitative evaluation of the potential of two-dimensional systems to generate distance between peaks. This selection procedure is developed on the sample Cyclopia genistoides with the objective of isolating eight target compounds

Full textGreen analytical chemistry development represents one of the main issues of the 21th century. Many investigators in analytical chemistry are actually involved in the development of well-established analytical methods that prevent irreversible damage to humans and environment. In the domain of lipid analysis, structural diversity as well as difference in solubility of these compounds is leading to work with a very large polarity range to separate lipids by classes. The normal-phase liquid chromatography (NPLC) allows realising the elution of compounds in order of increasing polarity. The solubilisation of lipid classes requires the use of a mixture of several solvents, among them chloroform, n-heptane, dichloromethane. Moreover, organic solvents traditionally used in NPLC, although well performing are raising different problems due to their original source, i.e. fossil hydrocarbons, volatility and toxicity for humans and environment. One of the ways to avoid such solvents is the substitution with alternative solvents, as proposed by various players in green chemistry. The interest raised by these solvents, is promising in terms of reduction of solvent use with a significant environmental impact. The properties of supercritical fluid chromatography using CO₂ are similar to NPLC and also offer a green alternative to this method. This work has highlighted that separation methods could be developed with alternative solvents to n-heptane, methanol and chloroform. Their use is compatible with liquid and supercritical chromatography and offer better selectivity in terms of separation of lipid classes. The compatibility of alternative solvents with ELSD and mass spectrometry was also evaluated, which showed that the unavailability of sufficient purity could be an issue. However, this problem was also observed with the use of commercial solvents, which presented impurities such as fatty acids, polymers and antioxidants

Full textSFC is an old analytical technique that took time to establish itself in the morals of chromatographers, remaining for many years in the shadow of HPLC and GC. Small and non-polar molecules are easily analysed using this technique because of the low polarity of carbon dioxide. What about polar compounds? Several methods can be used to analyse them. An intermediate technique between SFC and HPLC was used, unified chromatography.This research deals with methods for analysing molecules of higher polarity, such as amino acids. Stationary phases were studied using the LSER model to understand the behaviour of such molecules in a supercritical environment. A method development was performed for amino acid analysis. This method was then used for various applications (food supplements, plant of pharmaceutical interest)

Full textMembrane chromatography (MC) is an alternative to traditional resin packed columns chromatography. The solute mass transport in the membrane occurs in convective through-pores rather than in stagnant fluid inside the pores of the resins particles, which is limited by the slow diffusive transport. MC offers the main advantage of reducing diffusion phenomena, shorter residence time and lowered pressures drops, and thus, facilitates rapid purification of large quantities of molecules. A wide range of chromatographic membranes involving different molecules retention mechanisms (ion exchange, affinity, etc...) is now commercialized. Despite their success, the influence of the geometry of the membrane chromatography devices remains relatively unexplored from a theoretical point of view. This doctoral thesis is aimed to clarify some ambiguous points related to this technique

Full textThis thesis aimed at developing monolithic phases, on the one hand by studying the different parameters influencing the formation of the monolithic structure and on the other hand by developing a cladding procedure in order to apply these monoliths in chromatography. A new procedure based on the use of the F68 triblock copolymer was compared to the classical PEG, in the formation of silica monoliths, and was demonstrated to be promising since it leads to co-continuous structures presenting both smaller throughpores, in a wider composition range, and mesporous structures without any alkaline treatment. Moreover, an original cladding method was developed and allowed use of such monoliths in chromatography. The obtained separation performances were improved seven times if compared to those reported in the literature. Nevertheless, peak broadening due to phase heterogeneity and cladding still occurs. The high permeability of the columns was used by coupling several columns to achieve the efficiency required for the separation of phospholipids

Full textNowadays, the development of chiral molecule is a strategic issue for the pharmaceutical industry. During the development of a new molecular entity, supercritical fluid chromatography using enantioselective stationary phases is essential for the analysis or the purification of racemates. Despite the advantages of SFC, the analysis of some compounds like basic compounds may still be a challenge. Their analysis most of the time, requires an additive in the mobile phase. In many cases additives concentration is chosen arbitrarily, based on the columns supplier guidelines and work already published, regardless of the impact of the additive concentration on the resolution. This work has shown the impact of the additives concentration for the analysis of enantiomers by SFC, in particular a significative impact on the enantioselectivity of chiral stationary phases, with for some of them a reversal elution order depending on the additive concentration. The second objective is to improve our knowledge on the mechanism of action of the additive both on the stationary phase and the mobile phase. The last part is dedicated to the presentation of a few examples highlighting the positive impact of the variation of the additives concentration for enantiomer separation

Full textThe goal of this thesis is the development of technological and numerical tools for the monitoring of indoor air quality. That comprises the design of miniaturized sensors capable of detecting a large number of pollutants, and the development of inverse models allowing the reconstruction of pollutant sources from concentration measurements. In this work we present the realization of a micro-gas chromatograph, miniaturized chemical-analysis system fabricated with silicon-based MEMS technology. The separation column can separate multiple volatile organic compounds in less than a minute. The integrated thermal conductivity detector, based on a 4-points measurement technique coupled with an original temperature control circuit, can detect compounds separated in the column. In order to improve the performances of such micro-chromatographs, stochastic injection techniques have been developped. We have shown that these techniques easily allow to reduce the detection noise by an order of magnitude so that low-concentrated pollutants can be detected. We also observed that it is possible to follow in real time the mean evolution of the concentration with such techniques. We finally developped an original multiplexing technique that allows to monitor multiple sample streams simultaneoulsy with a single sensor. Finally, this work is completed by the development of 2D inverse models that allow, from data measured by micro-chromatographs placed in a room, to reconstruct pollutant sources, concentration fields or airflow patterns. Those models are based on an optimal control approach and use the resolution of Navier-Stokes and diffusion-advection equations with the finite element method. POD orthonormal basis are used for the regularization of the problem

Full textThe introduction describes the principles of countercurrent chromatography and especially its centrifugal partition chromatography version, as long as new or to-come industrial apparatuses. Usual strategies for the solvent system choice in case of an elution development are compared. Principles and theory of the displacement mode are particularly developed in their solid-liquid and support-free versions. Personal work bears on several applications and betterments of both elution and displacement modes. In elution development, a saponin-containing extract of Bacopa monnieri, an Indian Ayurvedic plant, has been fractionated, and isolation of an hemisynthetic alkaloid is described. In displacement development, a new solvent system range is described for the pH-zone refining mode and applied to the purification at an actual preparative scale of galanthamine from Leucojum aestivum. A new type of ion-exchanger adapted to support-free chromatography is developed. Benzalkonium chloride is used as a strong cationic exchanger. A numerical model allows graphical simulations, and extensive experimental work is done on optimization of the method. Several applications of this development mode are described, one of them being purification of rosmarinic acid from Lavandula vera cell culture extract

Full textA chromatographic technique has been developed in order to characterize high temperature only soluble polymers. The chromatographic device, built from commercial units, is now reliable. It is an automated and easy to modulate device. The operating conditions for polyolefin fractionation have been determined on the basis of theoretical considerations (self-focusing process) as well as on practical considerations (experience is higt temperature chromatography and in room temperature LAC). These conditions could be easily adapted to other higt temperature soluble polymers (polyamides). Different types of polyolefins have been characterized. The fractionation of non polar polyolefins (polyethylenes, polypropylenes) is delicate and should be improved, but offers yet very interesting perspectives (study of copolymers based on propylene, of polyolefin blends) In the case of polar copolymers based on ethylene, higt temperature LAC allows the determination of the chemical composition distribution within a very short period of time (about 1 hour). This distribution cannot be obtained with other analytical techniques. Higt temperature LAC analysis can also differentiate a copolymer from a polymer blend. LAC - SEC cross fractionation at higt temperature allows a very fine analysis of polymers that can be used for the study of the synthesis - process - structure - properties relationship

Full textThe question of marine litter and their environmental impact gains importance. It was shown that 80% of plastics results from terrestrial sources, while 20% come from coastlines and seas. Many authors point to a serious environmental problem involved to the absorption of POPs (persistent organic pollutants) by plastic detritus, thus initiating a potential source of contamination of the food chain. All authors attribute the fragmentation to "microplastic" (size < 5mm) to phenomena related to degradation, under UV without ever having demonstrated or analysed it. The rivers constituting then a privileged vector of transport of the terrestrial waste towards the seas and oceans, we decided to highlight (and quantify) the presence of microplastics in a river known as "wild" as the Allier. At the laboratory, we develop simulation protocols for the fragmentation of plastics under UV irradiation of various plastic materials in fresh water. We analyse both the chemical molecules resulting from the degradation of plastics and transferred to the aqueous medium as well as the molecular changes that can undergo the material

Full textAntisense oligonucleotides (ASO) have the ability to inhibit or modulate the expression of a target gene by various mechanisms. Bioconjugation of ASO with a lipid is a very promising approach which has shown an improvement in the delivery of the antisense sequence and therefore, in the therapeutic efficacy of the oligonucleotide. These new therapeutic agents, lipid oligonucleotides (LON), are amphiphilic molecules and are able to self-assemble to form supramolecular objects. Their pharmaceutical development requires suitable analytical methods to study the purity of the LONs synthesized and to be able to quantify them in the formulations but also to characterize the supramolecular assemblies formed.In this work, different methods were investigated in ion-pairing reversed-phase HPLC and hydrophilic interaction chromatography, capillary electrophoresis (CE) and size exclusion chromatography (SEC) for LON analysis with various chemical structures. Despite the different parameters studied, the asymmetry of the peaks obtained by HPLC limits its use for assays. On the other hand, a quantitative method has been developed and validated in CE in the presence of cyclodextrins (CD). The complexation constant between free LON and CDs as well as the electrophoretic mobility of the complex were determined. Finally, the potential of SEC and CE for the characterization of supramolecular objects of LON was assessed

The first part of this thesis was devoted to the design and fabrication of mesoporous silicon chromatographic columns using different clean room techniques (RIE, Cathode sputtering, photolithography. . . ). 1 carried out the various fabrication stages starting from a silicon substrate doped with phosphorus. The colurons characterization was performed using SEM imaging and optical microscopy. A chromatographic separation of two molecules was managed by using silica as stationnary phase. The second part of this work dealt with a Cellular study on Biology and proteomic. The effects of Nafenopine (a peroxisomes proliferator) on the microsomiales proteins of the hepatics cells ofrodents were studied in collaboration with the U. F. R. Des Sciences Médicales et Pharmaceutiques. After the lysis of the hepatic cells of the subjects and the recovery of their microsomiales proteins, the sample was injected into a two-dimensional liquid chromatography device conceived for proteomic, the ProteomeLab PF2D. This apparatus separates proteins according to two types of liquid chromatography: their isoelectric point, by chromatofocusing, as weIl as their hydrophobicity, by RP-HPLC. After differential study of the proteins map, a bio-marker was discovered and identified by Westem Blotting. This is the Cytochrome P450 4Al
