Abstract:
In dogs chronic degenerative valve disease (CDVD) is the most common cardiac disorder and the most frequent cause of cardiac failure seen in clinical practice. The aetiology of CDVD is unknown however recent investigations have focused on the molecular mechanisms that may be implicated in the development of the disease. This thesis investigates the impact of mechanical stimuli on the canine mitral valve at both the molecular and clinical level. At the molecular level canine valvular interstitial cells (VICs) were successfully extracted from valve tissue and shown to express proteins related to

SARM1 is an injury-induced nicotinamide adenine dinucleotide nucleosidase (NADase) that was previously shown to promote axonal degeneration in response to traumatic, toxic, and excitotoxic stressors. This raises the question of whether a SARM1-dependent program of axonal degeneration is central to a common pathway contributing to disease burden in neurological disorders. The degree to and mechanism by which SARM1 inactivation decreases the pathophysiology of such disorders is of interest to establish the rationale to pursue SARM1 as a therapeutic target. The chorda tympani nerve, also known as the taste nerve, runs uncovered through the middle ear cavity, a localization that exposes the nerve to pathological processes and surgical trauma in the middle ear. People operated on for otosclerosis tend to complain more about postoperative taste disturbances than those operated on for chronic otitis media. It has been suggested that this difference may be explained by gradual deterioration of chorda tympani nerve function caused by chronic otitis media infection and that further impairment caused by surgery is less noticeable in these patients.
