Full textLaurentian Great Lakes ciscoes (Coregonus artedi, sensu lato) diversity arose via repeated local adaptive divergence. Much of this diversity has vanished or is threatened. For instance, the nigripinnis form is now extirpated from the Great Lakes but remains in L. Nipigon. Ciscoes with some characteristics of nigripinnis have recently been discovered with sympatric artedi in lakes historically covered by the Fossmill outlet, a proglacial drainage of L. Algonquin. This raises the evolutionary hypothesis that artedi and nigripinnis could have colonized the area as distinct pre-existing lineages. Hence, putative nigripinnis may represent a relict embodiment of the historical diversity of ciscoes from the Great Lakes. The objective of this research was to characterize the phenotypic diversity and the evolution of newly observed putative nigripinnis and artedi. To do so, fish were grouped by habitat of capture (benthic and pelagic) and by gill raker counts and contrasted morphologically and genetically. Differentiation within and across lakes was often observed and paralleled that of artedi and nigripinnis in L. Nipigon. Gill rakers, caudal peduncle and body depth were the most important traits differentiating cisco forms. These traits are associated with varying foraging and locomotion strategies. Genetic analyses using 6676 single nucleotide polymorphism (SNPs) markers show that nigripinnis populations found in and nearby Algonquin Provincial Park origin from an Atlantic glacial lineage and repeated in situ divergence. Pairs of sympatric ciscoes were on a continuum of ecological, morphological and genetic differentiation across lakes. The divergence of nigripinnis could be associated to different resource exploitation, such as alternative zooplankton prey. This thesis confirms, by a comparative and integrative approach, the occurrence of a nigripinnis form with an independent evolutionary origin relatively to reference nigripinnis in L. Nipigon. The newly discovered nigripinnis populations could be source for the reestablishment of the functional diversity of the Great Lakes.

Odontogenic cysts are classified according to their origin in: developmental cysts, which include the dentigerous cyst (CD), or in inflammatory cysts, which have the root cyst (CR) as the main representative. In lesional microenvironments with low oxygen concentrations, the Hypoxia Induced Factor 1α (HIF-1) influences mechanisms responsible for cell survival. Thus, the aim of this research was to analyze the immunoexpression of HIF-1α in root and dentigerous cysts. The sample consisted of 40 cases of odontogenic cysts, 20 of CD and 20 of CR. Verification of HIF-1α expression was performed by the immunohistochemistry technique using anti-HIF-1α primary antibody (Clone EP1215Y, Abcam®, 1: 500, overnight, citrate pH 6, Pascal). Quantitative analysis was performed by counting the number of epithelial cells with immunostaining in the cytoplasm or nucleus in five fields photographed at a magnification of 400x. A qualitative analysis was also performed, observing the intensity of cytoplasmic immunostaining. The collected clinical data, such as sex, age and location site, were expressed in absolute and percentage frequency form and compared by Pearson's Chi-square test or Fisher's Exact and by Binomial Logistic Regression. The number of positive epithelial cells was expressed as mean and standard deviation and analyzed using the Student's T-test using the Statistical Package for the Social Sciences program. For all the tests performed, a significance level of 5% (p <0.05) was established. Immunohistochemical analysis evidenced positive marking throughout the sample. For the total number of cells with cytoplasmic immunostaining, between CR (67.7 ± 12.6) and CD (64.5 ± 16.1), there was no significant difference between the lesions (p = 0,480) and the Same result was found for nuclear immunostaining (p = 0.354). When the intensity of strong cytoplasmic immunostaining was observed and the clinical characteristics, the CD group presented a lower number of cells immunized by HIF-1α (11.7 ± 13.6) for the age group up to 30 years when compared to the age group of Individuals older than 30 years (34.5 ± 20.0), showing statistical difference (p = 0.010). When the strong cytoplasmic immunostaining was observed between the CR and CD groups in the anterior region of the jaws, it was verified that the CR group presented a greater quantity of immunolabelled cells (28.5 ± 8.5) than the CD group (14 , 0 ± 11.4), with statistical difference (p = 0.014). Considering the results obtained in this research, it can be concluded that HIF-1α is present in the epithelial constituents of CR and CD. However, further studies are needed to better elucidate the role of HIF-1α in odontogenic cysts.
Os cistos odontogênicos são classificados conforme sua origem em: cistos de desenvolvimento, dos quais faz parte o cisto dentígero (CD), ou em cistos inflamatórios, que têm o cisto radicular (CR) como principal representante. Em microambientes lesionais com baixas concentrações de oxigênio, o Fator Induzido por Hipóxia 1α (HIF-1) influencia mecanismos responsáveis pela sobrevida celular. Assim, o objetivo desta pesquisa foi analisar a imunoexpressão do HIF-1α em cistos radiculares e dentígeros. A amostra foi constituída por 40 casos de cistos odontogênicos, sendo 20 de CD e 20 de CR. A verificação da expressão de HIF-1α foi realizada através da técnica de imunoistoquímica, utilizando anticorpo primário anti-HIF-1α (Clone EP1215Y,Abcam®, 1:500, overnight,citrato pH 6, Pascal). A análise quantitativa foi realizada por meio da contagem do número de células epiteliais com imunomarcação no citoplasma ou núcleo, em cinco campos fotografados no aumento de 400x. Realizou-se, também, uma análise qualitativa observando a intensidade de imunomarcação citoplasmática. Os dados clínicos coletados, tais como sexo, idade e sítio de localização, foram expressos em forma de frequência absoluta e percentual e comparados por meio do teste qui-quadrado de Pearson ou Exato de Fisher e por meio de Regressão Logística Binomial. O número de células epiteliais positivas foi expresso em forma de média e desvio-padrão e analisado por meio do teste de T de Student, utilizando-se o programa Statistical Package for the Social Sciences. Para todos os testes realizados, foi estabelecido o nível de significância de 5% (p <0,05). A análise imunoistoquímica evidenciou marcação positiva em toda a amostra. Para o total de células com imunomarcação citoplasmática entre os casos de CR (67,7±12,6) e CD (64,5±16,1), não houve diferença significativa entre as lesões (p=0,480), bem como o mesmo resultado foi encontrado para imunomarcação nuclear (p=0,354). Quando se observou a intensidade de imunomarcação citoplasmática forte e as características clínicas, o grupo de CD apresentou para faixa etária de até 30 anos menor número de células imunomarcadas por HIF-1α (11,7±13,6) quando comparada à faixa etária de indivíduos maiores de 30 anos (34,5±20,0), demonstrando diferença estatística (p= 0,010). Já quando observada a imunomarcação citoplasmática forte entre os grupos de CR e CD na região anterior dos maxilares, verificou-se que o grupo de CR apresentou maior quantidade de células imunomarcadas (28,5±8,5) que o grupo de CD (14,0±11,4), com diferença estatística (p= 0,014). Diante dos resultados obtidos nesta pesquisa, pode-se concluir que HIF-1α está presente nos constituintes epiteliais de CR e CD. No entanto, novos estudos são necessários para melhor elucidar o papel desenvolvido pelo HIF-1α nos cistos odontogênicos.
