Abstract:
Nature employs complex networks of protein-tailoring enzymes to effect the post-translational modification of proteins in vivo. By comparison, modern chemical methods rely upon either nonspecific labeling techniques or upon the genetic incorporation of bioorthogonal handles. To develop truly robust bioconjugates it is necessary to develop methods which possess the exquisite activity and specificity observed in biological catalysts. One attractive strategy to achieve this is the engineering of protein-tailoring enzymes possessing user-defined specificity and high catalytic efficiency.<br>Che

Estrogens enhance transcription of target genes by stimulating the association of the estrogen receptor (ER) and related coactivators to estrogen response elements or to transcription complexes formed at activator protein (AP)-1 or specificity protein (Sp)-binding sites. Protein arginine methyltransferases (PRMTs) are enzymes that catalyze the methylation of protein arginine residues, resulting in the formation of monomethylarginine, and/or asymmetric or symmetric dimethylarginines. Recently, our laboratory identified two novel SVMPs, atragin and k-like, with similar primary sequence (~60%) and 3D structure. Atragin binds to αvβ3 integrin, inhibits NIH3T3 cell migration and possesses heparin binding ability. However, the binding model of atragin/αvβ3 integrin and the role of heparin binding on

Thus we need a real time and high-throughput method to analyze glycosyltransferase activity. In this research, we focus on the UDP-glycosyltransferase from Bacillus cereus (BcGT-1) catalytic reaction. BcGT-1 was successfully over-expressed in E. coli BL21 (DE3) and purified by DEAE column. In this research, we use a quick colorimetric activity assay for glycosyltransferase BcGT-1. The assay principle is based on the glycosylation activity of B
