Il progetto di musealizzazione dell'area archeologica di Villa Clelia nasce dalla volontà di valorizzare il luogo dell'antica cattedrale di S. Cassiano, dedicata al santo patrono della città di Imola e sorta sulle sue spoglie. L'area si trova in una zona periferica e residenziale della città che si presenta come un vuoto urbano. Partendo da un accurato lavoro di rilievo e analisi storica del sito, il progetto propone una visita dello scavo archeologico lungo un percorso che scende alla quota dei reperti e risale attraverso tre livelli individuati. Un confronto tipologico con il sistema basilicale ravennate e milanese ha permesso di ipotizzare una ricostruzione planivolumetrica dell'antica cattedrale, di cui è stato evocato il fronte.

Progetto di valorizzazione e conservazione dell’area archeologica di Villa Clelia, Imola (BO). Il sito, che allo stato di fatto presenta dei manufatti che versano allo stato di rudere, gode di un importante riconoscimento nell’immaginario locale, in quanto presunto luogo di sepoltura delle antiche spoglie del santo martire Cassiano, patrono della città. Attorno ad esso si sarebbe presumibilmente eretta la “Basilica Beati Cassiani”, edificio per eccellenza dell’omonimo episcopio, citato dalle fonti storiche con il toponimo “Castrum Sancti Cassiani”. L’area presenta numerose criticità dovute alla sua conformazione: su tutte, la scarsa leggibilità nei confronti del tessuto urbano, a discapito della portata del valore storico e culturale dei beni presenti in situ. Obiettivo principale, nonché scopo di questa tesi, è quello di ridare identità ad un luogo attraverso un progetto di valorizzazione e conservazione attiva del palinsesto archeologico, affrontando tematiche legate al restauro, con altre a carattere urbanistico. Un’unità introduttiva, a supporto di un ‘testo’ che il visitatore dovrà comprendere successivamente, sarà la chiave di volta per la custodia della memoria del luogo.

Ce travail présente la réflexion sur la comparaison de la stratégie de l'ironie dans les oeuvres des deux auteures du début du XX siècle l'italienne Clelia Romano Pellicano et la française Colette. Nous avons comparé le cycle des Claudine de cette dernière avec les recueils de nouvelles Coppie et Novelle calabresi signées par Pellicano, retraçant aussi une biographie de cette femme écrivain méconnue. Pour cette étude, une notion sexuée d'ironie et d'humour au féminin a été créée, reconstituant le cadre des théories existantes que l'on a mis en relation aux études sur les dimensions interdiscursives et relationnelles propres à l'ironie littéraire. Notre discours a aussi été contextualisé par une reconstruction des aspects dominants de l'esprit de la Belle Époque. Nous avons identifié les termes du débat sur la surabondance et la qualité de la littérature féminine, qui était (dis)qualifiée de « muliebre » ; débat dans lequel Colette figurait comme élément principal de comparaison par rapport aux auteures italiennes. L'ironie de Pellicano et Colette s'est révélée être la stratégie discursive qui amorce un conflit (amoureux) avec l'Autre. Par l'ironie elles ont dénoncé les contradictions de leur temps et évoqué des sujets sensibles ou considérés scabreux (dans la relation entre les deux sexes, par exemple). Elles se sont aussi rapportées au canon littéraire réaliste de façon désacralisée ; aspect que nous avons étudié en analysant la relation de l'ironie au naturalisme-vérisme. Les auteures ont, en outre, instauré une relation de complicité avec leurs lecteurs notamment grâce à l'ironie et à l'auto-ironie dont elles font preuve quant à leur propre statut d'écrivaines
We have compared how irony was used by two women writers at the beginning of the 20th century: the French Colette and the Italian Clelia Romano Pellicano. We have examined the navels of Claudine's series by Colette, and the two collections of short-stories Coppie and Novelle calabresi by Pellicano. The missing biography of the Italian writer has also been reconstructed. For this analysis we have revised the notion of feminine irony (and humor) and of women's comedy, reconstrueting the panorama of the existing theories and reconsidering some studies about the interdiscursive and relational dimensions proper of literary irony. We have historically contextualised our analysis identifying the characteristics of Belle Epoque's esprit and the relationship between caricatures and literary charge. We have picked out the ternis of the debate of this period about the excess and the quality of that female literature disqualified by the adjective « muliebre ». In this debate Colette has appeared as term of comparison for the works of the Italian women witers. Pellicano and Colette's irony emerges as a discursive strategy through which it is possible to inaugurate a "caring" conflict with the Other, in order to uncover the contradictions of their time and to deal with delicated subjets or themes considered awkward (i. E. The authorities' corruption or the relationship between sexes). Both women writers were related to the literary realistic model of their time by an irreverent way. We have studied it also reflecting upon the relationship between irony and Naturalism. Through irony and even self-irony about their status as women writers, they have created a relation with their readers

Les neuf auteurs concernes, certaines tres connues (mme de lafayette, mlle de scudery), certaines moins connues (mlle de montpensier, mme de villedieu), certaines inconnues (mme de la calprenede, mlle de la rocheguilhen, mme de salvan. . . . ), ne firent pas un usage feministe direct de l'ecriture romanesque. C'est plus generalement la societe dans son ensemble sur laquelle elles posent un regard critique, ce qui leur permet d'envisager ensuite de nouvelles formes de sociabilite dont rend compte le code galant. Between 1654, the date of clelie, and 1678, year of la princesse de cleves, thirtyone novels written by women-writers were published. The nine authors, - some well-known (mme de lafayette, mlle de scudery), some less known (mme de villedieu, mlle de monpensier), some unknown (mlle de la roche-guilhen, mme de salvan,. . . ) - did not use the novel writing for a direct feminist intention : more generally, they criticized the whole society, what allowed them to propose new forms of sociability such as the galant code. Although they did not really invent a new type of novel-writing, their conditions of production lead them to look for new writing-strategies, based on a public success rather than on an institutional recognition. Setting the novel on a moral and historical level, they were the first to give a generic legitimacy to it

Abstract:
L’athérosclérose est une maladie inflammatoire chronique. L’une des caractéristiques des lésions d’athérosclérose est l’accumulation anormale de corps apoptotiques et nécrotiques, due à un défaut d’efferocytose, ceci entraînant la formation du cœur nécrotique. L’évolution de ce cœur nécrotique est également associée à une augmentation de l’inflammation et de la taille des plaques d’athérosclérose, mais aussi dans la survenue de complications telle que la rupture de plaque. Clec9a est un récepteur transmembranaire de type lectine C, majoritairement exprimé par une sous population de cellules dendritiques les DC-CD8α+. Il est capable de reconnaître un ligand spécifiquement exprimé par les corps nécrotiques, l’actine F. L’objectif de notre travail a été de savoir si Clec9a, qui est capable de reconnaître les corps nécrotiques, pouvait être impliqué dans la modulation de l’inflammation observée au cours du développement de l’athérosclérose. Au cours de cette étude, nous avons montré, in vivo partir de deux modèles murins (ApoE-/- et LDLr-/-), que la délétion de Clec9a entraîne une diminution significative de la taille des lésions dans un contexte d’hypercholestérolémie modérée. Cette athéro-protection observée en l’absence de Clec9a, est associée à une augmentation de l’expression de l’IL-10, qui est une interleukine anti-athérogène et anti-inflammatoire. Cet effet athéroprotecteur de l’absence de Clec9a n’est plus observé lorsque l’IL-10 est totalement invalidée. De plus, nous avons montré que l’invalidation de Clec9a spécifiquement dans les DC-CD8α+ entraîne, in vivo, une diminution de l’infiltration des macrophages et des lymphocytes T dans les lésions, ainsi qu’une augmentation de l’expression de l’IL-10, favorisant une diminution de la taille des lésions. La compréhension des mécanismes inflammatoires dans l’athérosclérose constitue un enjeu majeur pour prévenir les risques de complications comme la rupture de plaque ou la thrombose. Ainsi, ce travail met en évidence un nouveau rôle de Clec9a dans la régulation de l’inflammation dans l’athérosclérose et pourrait donc représenter une cible thérapeutique potentielle
Atherosclerosis is a chronic inflammatory disease. One of the characteristics of atherosclerotic lesions is the abnormal accumulation of apoptotic and necrotic cells, due to a deficiency of efferocytosis, which leads to the formation of the necrotic heart. The evolution of this necrotic core is also associated with an increase in inflammation and lesions of atherosclerosis, but also in the occurrence of complications such as plaque rupture. Clec9a is a C type lectin receptor, mainly expressed by a subpopulation of dendritic cells, which are the CD8α+ dendritic cells. This receptor is able to recognize a ligand expressed by necrotic cells, the actin F. The aim of our work was to find out if Clec9a, which can sense necrotic cells, could be involved in modulating the inflammation observed during the development of atherosclerosis. In this study, we have shown, in vivo with two mouse models (ApoE - / - and LDLr - / -), that the deletion of Clec9a leads to a significant decrease in the incidence of moderate hypercholesterolemia. This athero-protection observed in the absence of Clec9a, is associated with an increase in the expression of IL-10, which is an anti-atherogenic and anti-inflammatory cytokine. This athero-protective effect of the absence of Clec9a is abolished after total invalidation of IL-10. Furthermore, we report that specific knockdown of Clec9a in CD8α+-DC, in vivo, leads to a decrease in macrophage and lymphocyte infiltration in lesions, as well as an increase in IL-1 expression. 10, which promotes a decrease in lesions size. Understanding of inflammatory mechanisms in atherosclerosis is a major challenge to prevent the risk of complications such as plaque rupture or thrombosis. Thus, this work highlights a new role of Clec9a in the regulation of inflammation in atherosclerosis and could be therefore a potential therapeutic target

Abstract:
The development of dedicated cryptography systems for applications requiring low cost and consumption has been the current focus of research. This work addresses the design and performance analysis of cryptographic algorithms AES-128 (NIST standard), PRESENT-80 and CLEFIA-128 (ISO/IEC standard for Lightweight Cryptography), im-plemented in FPGA (Basys 3 Artix-7 - 28 nm technology) using VHDL. Performance metrics were analyzed and compared: occupied area in the FPGA, throughput (Mbps), efficiency (Mbps/slice), energy efficiency (Ws/bit) and current consumption. The metrics were obtained through the synthesis and implementation tool in FPGA, Vivado Design Suites (Xilinx), and by means of a current measurement prototype, which uses the Ada-fruit INA219 sensor board (Sensor from Texas Instruments) and microcontroller Arduino Uno (Atmega328 - Atmel). We also analyzed the graphical representation of current con-sumption through the mathematical model based on the Welch periodogram, applied on the current consumption variables during the data encryption process. The results show current curves that facilitate the identification and comparison of the algorithms. The data of area consumption, processing speed and efficiency in the FPGA obtained satisfactory performance in comparison with other implementations existing in the literature, besides providing relevant information to choose an algorithm of encryption.
O desenvolvimento de sistemas dedicados de criptografia, para aplicações que exigem baixo custo e consumo tem sido enfoque atual de pesquisas. Este trabalho aborda o projeto e análise de desempenho dos algoritmos de criptografia AES-128 (padrão NIST), PRESENT-80 e CLEFIA-128 (padrão ISO/IEC para Criptografia Leve), implementados em FPGA (Basys 3 Artix-7 – tecnologia de 28 nm), utilizando VHDL. Foram analisadas e comparadas as métricas de desempenho: área ocupada no FPGA, velocidade de proces-samento (Mbps), eficiência (Mbps/slice), eficiência energética (Ws/bit) e consumo de corrente. As métricas foram obtidas através da ferramenta de síntese e implementação em FPGA, Vivado Design Suites (Xilinx), e por meio de um protótipo de medição de corrente, que utiliza a placa sensor Adafruit INA219 (sensor da Texas Instruments) e microcontro-lador Arduino Uno (Atmega328 - Atmel). Foram analisadas também a representação grá-fica do consumo de corrente através do modelo matemático baseado no periodograma de Welch, aplicado sobre as variáveis de consumo de corrente durante o processo de encrip-tação de dados. Os resultados mostram curvas de corrente que facilitam a identificação e comparação dos algoritmos. Os dados de consumo de área, velocidade processamento e eficiência no FPGA obtiveram desempenho satisfatório, em comparação com outras im-plementações existentes na literatura, além de fornecer informação relevante para escolha de um algoritmo de criptografia.

Abstract:
My parallel reading of two seventeenth-century romances by two women, one English, one French, aims to illumine the early modern mapping of womanhood from a female perspective. Part one examines the discourse of virtuous women in the patriarchal societies of Urania and Clélie. Adopting an approach based on close stylistic analysis, I explore, on the one hand, the extent to which the marriage topos endows or does not endow these women with speech and power, and on the other the extent to which the marriage topos enables the utterance of a protofeminist discourse. While the marriage topos initially allows us to visualise these women as daughters, sisters, mothers and wives, it gradually unveils women not only as the apologists of true love, but also as androgynous heroines in the male-authored domain of politics. Having discussed the discourse (mostly oral) of Uranian and Scuderian heroines in the context of a society functioning on the basis of political alliances, I move on to analyse ‘The loci of the feminine’, i.e. the configuration of female spaces in two texts. This analysis is developed in parts two and three of this thesis. The first part begins by examining the cultural milieux of Mary Wroth and Madeleine de Scudéry, and explores some of the evidence regarding the possible existence of an early modern English equivalent of French ‘salons’. This chapter measures the extent to which Urania and Clélie might be constructed as illustrations of the highly intense activities of Wroth’s and Scudéry’s literary circles. The next chapter focusses on the Uranian and Scuderian fictionalisation of a predominantly female community. Part three assesses the ways in which these texts seem to inscribe themselves within a protofeminist project of re-evaluating female legacy and authorship in the realm of letters, and proceeds to explore more specifically the representation of literary creativity in Urania’s and Clélie’s female retreats. My final part examines the subject of the thesis in its literal sense, by analysing Wroth’s and Scudéry’s representations of the female body, and relates- where appropriate- images of women in Urania and Clélie to those found in the visual arts of the early modern period, such as emblems, engravings, paintings and masques.

Quelles relations, J. M. G. Le Clézio, engage dans une quête spirituelle, entretient-il avec le genre romanesque? Le roman est en question dans son œuvre sur plusieurs plans. Il rejette la linéarité du temps et le déterminisme social, psychologique, attribues au roman du XIXe siècle, dans une sorte de violence identificatoire. Pour rompre avec l'occident, il préconise un renoncement aux valeurs établies qui passe par l'ascèse. L'écriture se redéfinit comme recherche d'un langage affectif et de rythmes, proche de la poésie, rejoignant le silence. Dans ses romans, il utilise personnage, temps et récit mais en les déformant. Le personnage, indéterminé, se rattache au mythe et permet d'établir un pacte narratif ou se confondent protagoniste, auteur et lecteur dans une communauté des faiblesses et aspirations. Le temps perd sa linéarité causale pour se présenter dans la diversité d'expériences liées aux genres : conte, mythe, prophétie. Le récit refuse la mise en intrigue, se fracture sans cesse, surtout dans les premiers romans, et instaure une circularité, symbole de transcendance. Virtualité, évanescence et ambiguïté deviennent signes de l'être et fondent l'esthétique. Le roman parait contraire à l'exigence de vérité que suppose la quête. Cependant, par sa valeur d'image, il s'avère forme privilégiée pour éviter la clôture du sens, faire surgir l'ineffable, assurer la communication avec le lecteur. La réception des premières œuvres est problématique en raison d'une incessante redéfinition du code de lecture ; les derniers romans, plus cohérents, encourent un autre risque : celui de dissimuler la quête sous la dominante de la fiction. What are, in le Clezio's work, the interrelations between his quest to achieve essentiality and fictional form? The novel is put into question, in many ways. He refuses language as an instrument of domination, rejects linear time and both social and psychological determinism, regarded as characteristic of nineteenth-century novel. He wants to give up the inherited and established values of the west by renouncing the world as well as literature. The act of writing is defined as a search for an affective, rhythmic language, close to poetry, tending towards silence. In his novels, he uses characters, time and narrative but distorts them. The character, in his indeterminacy, is linked to myth and allows hero, writer and reader to meet and merge in a community of failures and aspirations. Time loses its causality to give rise to a diversity of experiences connected with literary genres: fairy tale, myth, prophecy. . . The narrative which rejects plot, is continually fragmented, especially in the first works. Beyond the breaking of the story, circularity appears as a symbol of transcendence. Virtuality, ambiguity and evanescence emerge as signs of being and the basis of aesthetics. The novel seems primarily opposed to the inescapable need to find truth. Nevertheless, with its value as an image, it emerges as a privileged form to keep meaning open, to give rise to the ineffable and ensure communication with the reader. The reader's reception of the first works is problematic owing to constant alterations in the reading code, while the last novels

Abstract:
Since his death in 1607, Abraham Fleming has never been completely forgotten about. This thesis covers all aspects of Fleming's life. It begins with his time at Cambridge and the relationships he forged there. It studies his varied and sometimes groundbreaking contributions to the books associated with him (with a focus on his English texts and translations). It also covers his ordination into the Church of England and subsequent career as a chaplain to Charles Howard, earl of Nottingham. It also elucidates his previously unknown life as a curate in the parish of St Nicholas, Deptford and as a deacon and priest St Pancras, Soper Lane, and finally his sermons at Paul's Cross in the grounds of St Paul's Cathedral. Fleming's legacy of at least 52 printed books, which includes original godly protestant treatises, English translations of Latin and Greek classical works, and books commemorating unusual occasions, have ensured that his name lived on in bibliographic catalogues. Since the 1950s a few scholars have considered Fleming's work on Holinshed's Chronicles as significant contributions to the text. However, the subsequent articles that have been written about him have been narrow in scope and at times unreliable. Recent studies of Fleming have considered him only as a minor writer, yet this thesis demonstrates that he was a literary figure of considerable significance. Fleming made an important contribution to the emerging public sphere, as foregrounded by Jurgen Habermas, that was lauded by his contemporaries but he has largely slipped from view. Before this doctoral research little was known about Fleming's career as a preacher in the Church of England, a career in which he proved just as diligent as when he was a “learned corrector” of books. The aim of this thesis has been to throw fresh light on the multi-faceted career of Abraham Fleming and establish him as a leading figure in late-Sixteenth century political and print culture.

Fundação de Amparo à Pesquisa do Estado do Rio de Janeiro
A presente pesquisa investiga a produção de subjetividade em seminários católicos. Por meio de diversos teóricos, girando em torno do eixo conceitual da Análise Institucional (em sua vertente socioanalítica e, principalmente, esquizoanalítica), pesquisou-se um importante seminário católico diocesano na região Sudeste, um dos primeiros fundados no Brasil. Um seminário é a instituição de internato em que muitos homens vivem, estudam e trabalham, durante oito anos em média, para se tornarem padres. Nossa pesquisa de campo utilizou-se da observação participante, oriunda da antropologia e sociologia, em diversas visitas ao estabelecimento, durante as quais fomos autorizados a participar de todas as atividades dos 110 seminaristas internos. Em seguida, alguns seminaristas, de todas as etapas da formação, e todos os 5 padres formadores foram entrevistados. Recorremos a entrevistas abertas de história de vida, segundo os procedimentos da História Oral italiana. Guiados pela distinção da filosofia de Spinoza de três gêneros de conhecimento, buscamos dramatizar a formação clerical em três níveis de compreensão: os Signos ou afectos, as Noções ou conceitos, as Essências ou perceptos. Por meio dessa trajetória, constatamos o predomínio da dimensão do instituído e de uma modelização homogeneizadora de tipo romana, resultando em que os clérigos ali formados se fechem em uma identidade sacerdotal claramente identificável e encapsulada na obediência aos centros de poder eclesiais: a Cúria romana, a Mitra diocesana e a Paróquia. Busca-se, na formação, reproduzir a subjetividade serializada segundo um Modelo sacerdotal institucionalizado, no qual as dimensões litúrgica e disciplinar são ressaltadas, em detrimento das dimensões místico-políticas sendo a perseguição à Teologia da Libertação um importante analisador dessa característica. A constante vigilância da pureza doutrinal, litúrgica, organizacional e teológica indicou-nos a pressão em reprimir a dimensão místico-profética, que range, querendo se expressar. Em vista disso, bem como de inúmeros outros analisadores, conceitos e personagens produzidos ao longo da pesquisa, pudemos constatar que o desejo clerical, modulado na formação seminarística, oscila entre dois pólos: um pólo sacerdotal-romano-paranóico e um pólo profético-libertador-esquizo. No primeiro, há redução à identidade hegemônica nascida nos centros de poder eclesiais, fechando-se à diferença, na busca de um projeto de imortalidade frente às intempéries da vida e transformações da História, produzindo práticas hierárquicas a partir de um pensamento de caráter transcendente, representativo. No segundo pólo, há busca de singularização, nascida do seguimento a Jesus, o conseqüente compromisso com os menores e excluídos dentro e fora da Igreja e o processo inerente a esta produção de sentido, criando-se, em conseqüência, uma radical imanentização da vida cristã e de seus pólos e transcendências: material/espiritual, fé/vida, Igreja/Mundo, mística/política. O seminário pesquisado, indicador das transformações micropolíticas da Igreja contemporânea, produz hegemonicamente um desejo sacerdotal-romano-paranóico, forjando funcionários do poder da Igreja, burocratas do aparelho de Estado romano, aplicadores de suas rubricas litúrgicas e normas doutrinais e morais, e não profetas do Reino de Deus, máquinas de guerra libertadoras quanto a tudo o que oprime a potência da vida e suas inauditas expressões singulares.
The present search investigates the production of subjectivity at catholic seminaries. Through several theorics, turning around the Institutional Analysis concept axis (in its socianalytical source and, mainly, schizoanalytical), a important diocesan catholic seminary at Southeastern region, one of the first founded in Brazil, was explored. A seminary is a boarding institution where many men live, study and work, during eight years on average, to become priests. Our field research used participant observation, from anthropology and sociology, in sundries visits to the establishment, during which we were allowed to participate in all seminarians activities. Afterwards, some seminarians, all training stages, and all trainers priests were interviewed. By the life history open interviews recourse, according to the italian Oral History procedures. Guided by the Spinozas philosophys distinction of three knowledge genders, we look for dramatize the clerical training in three understanding levels: the Signs or affects, the Notions or concepts, the Essences or percepts. Through this path, we caught the predominance of the instituted dimension and of the roman type homogenization standart, resulting that the clegyrmen who take degree there get close in to a clearly identifiable priesthood identity and encapsulated in obedience to the ecclesiastic power centers: the roman Curia, the diocesan Mitra and the Parish. The purpose, during the training, is to play the serialized subjectivity according to a established priesthood Model, in wich the liturgical and disciplinary dimensions are highlighted, over mystical-politics damaged dimensions the persecution to the Liberation Theology is an important analyser of this feature. The constant surveillance of doctrinal, liturgical, theological and organizational purity indicated us the pression on repress the mystical-prophetic dimension, and its creaks, wishing for express itself. In view of that, as well as numberless others analysers, concepts and characters produced along the survey, we could see that the clerical desire, modulated in the seminaristic training, oscillates between two poles: a priesthood-roman-paranoid pole and a prophetic-liberating-schizo pole. In the first, there is the reduction to the hegemonic identity borned in the ecclesiastics power centers, shutting off the difference, in the search way for a immortality project in face of the lifes storms and Historys changes, producing hierarchical practices from a thought of trancendent, representative kind. In the second pole, there is the search for singling, derived from the Jesus following, the concomitant commitment to the minors and excluded from within and outside the church and the inherent process in this sense production, creating, as consequence, a radical magnetization of Christian life and also of its poles and transcendencies: material/spiritual, faith/life, Church/World, mystical/ politics. The examineted seminary, a contemporary church micropolitical changes indicator, produces in a hegemony way a priesthood-roman-paranoid wish, forging Church power officials, roman state apparatus bureaucrats, its liturgical rubrics applicants and doctrinal and moral rules, and not Gods Kingdom prophets, wars machines setting life power and its unheard singular expression free from everything that oppress them.

Abstract:
Human infections with influenza A virus (IAV) exhibit mild to severe clinical outcomes as a result of differential virus-host interactions. C-type lectin receptors (CLRs) are pattern recognition receptors that may sense carbohydrates, proteins, or lipids derived from infected hosts or the invading microbes including bacteria, viruses, fungi, or parasites. CLR-viral interaction may lead to increased viral entry and spread; furthermore, their interactions have been reported to trigger downstream signaling that further modulates host’s innate immune responses through the induction of pro-inflammatory cytokines. To date, DC-SIGN and DC-SIGNR have been shown to mediate IAV entry; however, the potential interactions between other human transmembrane CLRs with IAV have not yet been systematically investigated. We utilized lentiviral-based pseudoparticles expressing influenza hemagglutinin (HA) to examine the binding potential between HA and a panel of human CLRs expressed in soluble form. CLEC5A was identified as a potential interacting target with the HA proteins derived from a highly pathogenic avian H5N1 virus A/VN/1203/04 (VN1203) or a human seasonal H1N1 virus A/HK/54/98 (HK5498), albeit at different binding intensity. Applying siRNA gene silencing, we confirmed that CLEC5A did not enhance influenza entry in human monocytic U937 cells that constitutively express CLEC5A or in the lentiviral-transduced stable CHO and CHO-Lec2 cells that overly expressed CLEC5A. To investigate downstream signaling upon engagement of CLEC5A to influenza virus, M-CSF or GM-CSF differentiated human macrophages with high expression levels of CLEC5A and DAP12, a known adaptor protein for CLEC5A upon phosphorylation to initiate signal transduction, was subjected to CLEC5A siRNA gene silencing followed by infection with recombinant A/PR/8/34 virus expressing HA and NA derived from either VN1203 (H5N1) or HK5498 (H1N1) viruses. RG-PR8xVN1203HA,NA (H5N1) exhibited a higher infectivity and induced higher levels of pro-inflammatory cytokines (TNF-( and IFN-α) and chemokines (IP-10, MCP-1, MIG and MIP-1α) secretion in M-CSF or GM-CSF differentiated macrophages while compared to that of the RG-PR8xHK5498HA,NA (H1N1) virus. Knocking-down CLEC5A in macrophages led to a universal reduction of cytokines and chemokines secretion after infection with either the RG-PR8xVN1203HA,NA, RG-PR8xHK5498HA,NA, RG-A/VN/1203/04 (H5N1) or A/Shanghai/2/2014 (H7N9) viruses, suggesting that CLEC5A plays a role as cytokine and chemokine amplifier after influenza infection. Since DAP12 phosphorylation is known to activate downstream signaling via Spleen tyrosine kinase (Syk), pre-incubation of M-CSF macrophages with a Syk inhibitor (Bay 61-3606) also lead to a significant reduction of TNF-α and IP-10 in infected macrophages. A higher mortality was observed in CLEC5A-/- mice while compared to the wild-type C57BL/6 mice after challenged with a lethal dose of RG-A/VN/1203/04 (H5N1) influenza virus suggesting that CLEC5A as a host innate response amplifier play a protective role upon influenza infection. In conclusion, we have identified CLEC5A as a novel host factor for influenza pathogenesis by modulating host innate inflammatory response.

En nous intéressant a deux écrivains contemporains, nous posons d'emblée la question du sens du roman après la remise en question de ce dernier au cours du vingtième siècle. L'expérience du temps dans l'œuvre de le Clézio et de Handke questionne l'espace littéraire sur sa capacité de reconfigurer le temps humain. Nous dégageons une structure d'une quête stratifiée du temps qui tente de transformer la question aporétique sur le temps en une poétique du récit par un réinvestissement du personnage et de l'intrigue. L'enjeu des romans s'affirme comme la nécessite d'une recollection temporelle capable de réinvestir le présent au-delà d'une dialectique de l'instant et de l'éternité. Des divergences narratives et signifiantes apparaissent. L'œuvre de le Clézio ne semble donner de sens à l'existence humaine qu'au regard d'un temps cosmologique qui été a l'homme tout pouvoir de changer le temps présent et qui renvoie à une expérience individualiste de la réalité. Le récit est incapable de dire le temps autrement que sous la forme rigide du discours. L'œuvre de Handke, elle, se constitue comme fable sur le temps. Le livre est réinvesti d'un sens universel en se transformant en horizon d'attente d'une humanité pacifiée. Nous découvrons ainsi, dans la première œuvre, une esthétique de l'expérience du temps et, dans la seconde, une poétique du récit ou l'expérience initiatique de la temporalité déplace la quête vers une éthique a redéfinir
Focussing on the work of two contemporary authors, the meaning of the novel, since the reassessment it has undergone during the twentiech century, is examined. The experience of time in le Clezio's and Handke's novels addresses the capacity of literature to reshaye human time. The analyses reveals a stratified quest for time which attemps to transform the aporetic question about time into a poetics of narrative thourgh the use of character and plot. The pivotal concern of the novels is seen to be the need for a temporal recollection capable of exploring the present beyond the dialectics of the instant and eternity. Points of divergence appear in the narrative and in the signifiers. Le clezio's work seems to assign meaning to human existence only with regard to a cosmological time wich deprives man of any power to change the present and wich reflects an individualist experience of reality. The narrative is incapable of expressing time other than in the rigid shape of discourse. Hank's wwriting, on the other hand, takes the form of a fable about time. The novel is endowed with universal significance opening perspectives of humanity pacified. Thus in the first work we discover an aesthetics of the experience of time, and in the second, a poetics of narrative where the initiating experience of temporality displaces the quest towards an ethics

We consider the myth of golden age as the main element of le clezio's texts ; indeed, in our point of view, his vision of the world is deeply influenced by the myth that gives to the le clezio's novels its real meaning. So we intend to study how the myth of golden age has "modeled" the fundamental parts of le clezio's books. That is to say his philosophical conception of space, of language, of game ; his position on original freedom and on modern slavery. We mostly used the theses of mircea eliade (the ontological aspect of the myth for example) because those theses allow to understand the various aspects of le clezio's thought. As an example, myth is defined by m. Eliade as "a story of a creation"; also the drift of le clezio consists in telling how modern man has himself lost golding age and has lived in a bad world. As well as the "fallingworld" is disqualified, as well as are exalted" the country of silence" and its people who could preserve oneself from the "fall". The texts of le clezio are built upon a series of oppositions assorted with a system of values that could be brought out by the structuralist method, even though we added other analyses including the social context, the author's personnality and the ontological aspect. Through the le clesio's novels, the myth of golden age finds again its sacred sense proper to the primitive people. Indeed le clezio's texts can help us to build a new world.

L'ecriture romanesque de le clezio aspire a une expression directe et sensorielle de la rencontre entre l'etre et le monde. Ses points de rupture avec les criteres du genre sont dictes par le desir de conferer au roman une nouvelle force originelle grace a un investissement poetique. Son refus d'une mise en intrigue est compense par une contemplation passionnee du monde et une poetisation d'anecdotes, le personnage psychologise est substitue par des figures "primitivistes". Une repartition volontairement simplifiee des caracteristiques spatiales et temporelles reflete une dialectique primitiviste opposant l'originel au non-originel. Car pour le clezio, une ecriture romanesque depouillee et minimaliste, ne reflete pas un vide metaphysique, mais suggere, plus qu'une solitude existentielle, la presence d'un cosmos mouvant. Une evocation sensorielle du monde contribue a souligner cette topique: le discours du corps supplante ainsi celui de l'intellect, les passions sont occultees par une nature omnipresente. Cette perspective modifiee, qui valorise la matiere au profit de l'homme, est perceptible dans une ecriture poetique fondee sur le sensoriel. Il en resulte un mimetisme typographique et rythmique destine a evoquer le lien magique entre mot et chose, un systeme metaphorique qui subordonne l'homme aux elements pour suggerer un cosmos qui se revele a l'homme a travers les sens dans une "extase materielle"; et un regard, mediateur de tous les sens, qui constitue un principe structurant des romans de le clezio. Ces techniques poetiques contribuent a la creation romanesque d'un lieu originel ou l'etre rencontre le monde
Le clezio's novelestic writing aspires to a direct and sensory expression of the meeting between the being and the world. The points in which he breaks with the criteria of the genre are dictated by the desire to contribute a new original force to the genre through investing the novel with poetic traits. He refuses to employ a proper plot, but this is compensated by a passionnate contemplation of the world and a poetisation of the anecdotes; the psychologized character is subsituted by "primitivistic" figures. The voluntarily simplified distribution of spatial and temporal characteristics reflects a primitivistic dialectic opposing the original and the nonoriginal. Le clezio's simplification of the novelestic writing does not reflect a metaphysical emptiness, but suggests, more than an existential solitude, the presence of a changing cosmos. Indeed, a sensory presentation of the world serves to underline this theme: the body's way of expressing itself supplants in this way that of the intellect; the passions are concealed by an omnipresent nature. This displacement of the perspective from man to substance is visible in a poetic style that concentrates on a sensory expression of the world. The result of this is a mimic typography and rhythm to evoke the magic bond between word and thing, a metaphoric system that subordinates man to the elements in order to suggest a cosmos that reveals itself to man through his senses in a "material ecstasy", and a look that is a mediator of all the senses and that forms a structural principle of le clezio's novels. These poetic techniques contribute to the novelestic creation of an original scene where the being meets the world

Cette these etudie la continuite et l'evolution de l'oeuvre de j. M. G. Le clezio, depuis le proces-verbal (1963) jusqu'a reve mexicain (1989). Le clezio offre a lire une seule histoire : celle d'une quete d'harmonie interieure, ses premieres oeuvres mettent en scene des apocalypses, des promethees, des icares ou des oedipes. Le protagoniste cherche alors son unite perdue : trois rencontres sont alors determinantes : celle des quatre elements. Celle d'autrui et celle d'une autre culture. Dans les oeuvres suivantes, au contraire, le heros renaitra donc tel hermes ou le phenix. Le present ouvrage distingue trois periodes dans l'oeuvre de le clezio : les annees de plumes et de revolte, les annees du feminin, les annees du mythe et du spirituel. La demonstration se compose de quatre parties: la quete d'harmonie des personnages, la quete d'un style leclezien. Les mythes et initiations dans l'oeuvre et enfin la relation entre mythe personnel et mythe collectif. Tous les chapitres insistent sur le parcours de l'oeuvre- du jeu d'ecriture a la question de l'unite retrouvee- mais aussi sur sa continuite, notamment par la recurrence de motifs symboliques ( visage, regard, silencen enfant, lumiere, musique) ainsi que par le fonctionnement de la reiteration. Grace a ses voyages, le clezio a decouvert la valeur du silence et a delaisse l'esprit cartesien. Chez lui, l'homme trouve son humanite lorsque s'etablit l'echange entre lui et les autres, entre lui et le passe. Entre lui et quelqu'au-dela. Sont alors eclairantes la rencontre et l'intuition de la presence, au coeur du reel, du sacre qui donne sens
Our work surveys both the continuity and the evolution from le proces-verbal (1963) to le reve mexicain (1989). Le clezio has written one and the same story : the basic quest of the being. As icare or prometheus, his protagonists are searching for the being. Through the meeting with the four elements, the protagonist makes headway towards a new harmony. Out of the chaos of the universe, the young hero is born again as a phoenix, a hermes. The present thesis deals with firstly, the protagonists' quests, secondly the writer's quest for a style, thirdly the mythe and fourthly the two-way interraction between the personnal myth and the collective ones. We delineate the writer's gradual shift from literature as a stylistic game to a more essential approach of man's successful life. Three stages have been found : first le clezio focuses on his writing, second he meets the woman and third he emphasises the spiritual and the mythic. We have also laid stress on patterns: face, vertigo, look, silence, child, light, music. . . Due to his staying among the mexican indians, le clezio has grown more responsive to mutism, the poetry of the myths ; hence his negative attitude towards occidental ideology which he regards as a misleading way to the true life. Today, le clezio merges autobiographical and universal patterns. Man will achieve his human status when he communicates with the other, the past, and the sacred, only then this intuition of the presence of the sacred makes sense

Resumo: O projeto explora o potencial catalítico de fosfolipases A2, isoladas de venenos de serpentes brasileiras para efetuar resolução enzimática de substratos com relevância científica, visto que nenhum trabalho anterior foi feito analisando-se sua enantiosseletividade. Foram feitos estudos sobre a resolução do Binol, do a-tetralol, do 1-feniletanol, do para-nitro-1-feniletanol, do ácido 3-(2-bromo-hexanoiloxi)-4-nitrobenzóico e ácido 3-(2-metil-hexanoiloxi)-4-nitrobenzóico.Devido a dificuldade de obtenção e purificação das fosfolipases, a enzima foi imobilizada utilizando a formação de um agregado com ligações cruzadas (Cross-Linked Enzyme Aggregate . CLEA). Os agregados foram produzidos com quatro tipos de precipitantes (solução 55 % de sulfato de amônio, polietilenoglicol 600 Da, dimetoxietano e acetona) e dois adicionantes (TRITON-X100 e polietilenodiimina). Com os testes, observou-se que o CLEA formado com sulfato de amônio, sem adicionante apresentou os melhores resultados, sendo utilizado nas reações de biocatálise. A resolução dos substratos foi feita com a esterificação dos álcoois, formando-se ésteres (acetatos, propanoatos e hexanoatos), e posterior hidrólise com a enzima não-imobilizada e CLEA da fosfolipase A2, para comparação. Alíquotas das reações foram e analisadas por GC/FID com fase estacionária quiral para estudo dos excessos enantioméricos. As reações foram feitas a temperatura ambiente e a 45 °C. Os resultados indicam atividade enzimática sendo possível obter o tetralol com 16% de e.e. utilizando-se o CLEA e o p-nitro-1-feniletanol com 19% de ee usando-se a PLA2 livre. Os outros álcoois foram obtidos com baixos ee. O ácido 3-(2-bromo-hexanoiloxi)-4-nitrobenzóico não pode ser analisado por sofrer hidrólise química completa no meio reacional, e com a hidrólise do ácido 3-(2-metil-hexanoiloxi)-4-nitrobenzóico foi possível a obtenção do ácido 2-metil-hexanóico com 9 % utilizando-se CLEA e 7 % com a enzima livre. A baixa enantiosseletividade foi interpretada como decorrente da fraca interação dos substratos com o sítio ativo da enzima
Abstract: This project explores the catalytic potential of fosfolipases A2, isolated from poisons of brazilian serpents to effect enzymatic substrate resolution with scientific relevance, since no previous work was made analyzing its enantioselectivity. Studies on the resolution of several compounds had been made, including Binol, a-tetralol, 1-phenylethanol, para-nitro-1- phenylethanol, 3-(2-bromohexanoiloxy)-4-nitrobenzoic acid and 3-(2-methylhexanoiloxy)-4-nitrobenzoic acid.Due to difficulty of attainment and purification of the phospholipase, the enzyme was immobilized using the formation of an aggregate with cross-links (Cross-Linked Enzyme Aggregate ¿ CLEA). These aggregates had been produced with four types of precipitation agents (ammonium sulphate solution 55%, polietileneglycol 600 Da, dimethoxyethane and acetone) and two additives (TRITON-X100 and poliethylenediimine). With the tests, it was observed that the CLEA formed with ammonium sulphate, without additives presented the best results, being used in the reactions of biocatalysis.The resolution of substrates was made with the alcohol¿s esterification, forming different (acetates, propanoates and hexanoates) followed by hydrolysis with the free enzyme and CLEA, for comparison. Aliquots of the reactions had been made and analyzed with GC/FID with quiral stationary phase for study of the enantiomerics excesses. The reactions had been made at ambient temperature and 45 °C.The results indicate enzymatic activity and was possible to get tetralol with 16% of ee using CLEA and p-nitro-1-phenylethanol with 19% of ee. The other alcohols had been gotten with low ee. The 3- (2-bromohexanoiloxy) - 4-nitrobenzoic acid cannot be analyzed by suffering complete chemical hydrolysis during the reaction, and with hydrolysis of acid the 3- (2-metilhexanoiloxi) - 4-nitrobenzoic the attainment of the acid 2-metilhexanoic with 9% was possible using CLEA and 7% with the free enzyme. The low enantioselectivity was explained due to the weak interaction of substrates with the active site of enzyme

Abstract:
Dendritic cells (DCs) play important roles at the interface between innate and adaptive immunity by priming and directing T cell responses. Much of our current knowledge of DC biology has come from mouse models in which DCs can be genetically manipulated, labelled or ablated. Here, novel models are presented using a strategy that targets DC precursors via genetic editing of the Clec9a locus. While validating a novel mouse model to inducibly deplete DCs using diphtheria toxin receptor (DTR) expression driven by Clec9a, it became clear that these Clec9a+/CreROSAiDTR mice suffer from unexpected lymph node (LN) hypocellularity and reduced frequencies of DCs in LNs, even in the absence of diphtheria toxin (DT) injection. This phenotype turned out to be a common feature of other mouse models in which DTR is expressed on DCs (e.g. CD11c-DTR and Langerin-DTR mice) and raises questions about the interpretation of results obtained with such animals. Therefore, in an alternative approach, mice were developed to constitutively lack DCs by expressing the diphtheria toxin alpha (DTA) subunit under control of the Clec9a locus. Unfortunately, these mice still harboured DCs and only showed partial reduction of one DC subset. Finally, seeding of tissues by DC precursors was examined. Clec9a+/CreROSA+/confetti mice were generated in which DC precursors stochastically express one of four fluorescent proteins, which is inherited by its daughter cells. 8- Colour microscopy of tissue sections and histo-cytometry analysis of the images was developed to analyse these mice. This approach will be used to determine how many daughter cells are produced when a single DC precursor seeds the small intestine (clonal burst size), whether these daughters are found among different DC subsets and whether seeding changes during inflammation. In summary, manipulation of the Clec9a locus proves to be an excellent way to study the DC lineage and DC precursor behaviour in the mouse.

Historians often describe how the ideas of national identity, race, religious affiliation, and political power greatly influenced the development of societies in colonial America. However, historians do not always make clear that these ideas did not exist independently of one another. Individuals in colonial Americans societies often conflated and incorporated one or more of these ideas with another. In other words, individuals did not always think of national identity and race and religious affiliation as independent entities. The specific case of the Reverend Morgan Godwyn illuminates just how connected these ideas were in the minds of some colonial Americans. As a minister in the Church of England, Godwyn spoke and wrote within an overtly religious context. His words, however, reveal that to him, religion and politics, national identity and race and ethnicity, could not be unpacked and viewed separately-each heavily influenced the others. Godwyn used his position as a cleric to challenge the authority of English colonial elites. He attempted to convince the English public of the necessity of reining in the growing powers of colonial elites in order to preserve the authority of the English monarch and the Church of England clergy. From studying Morgan Godwyn, one can see how complex and convoluted ideas-and simultaneously important-ideas of national identity, race, religion and political influence were in seventeenth-century colonial American societies.

Le clezio se caracterise par un discours critique sur la double tension de l'ecriture : elle est donnee a la fois comme une contingence et se presente comme un ensemble construit, issu d'un heritage culturel donne, auquel l'ecrivain ne peut echapper. La question recurrente de cette periode est la suivante : comment utiliser au mieux cet outil paradoxal? le recours a la dimension ludique du langage constitue une des reponses que choisit j. M. G. Le clezio, par la mise en oeuvre de situations romanesques, fondees sur les polarites agonistiques du jeu. Le jeu ontologique pose un rapport antagoniste entre la contingence de la vie humaine et la presence immuable de forces materielles et cosmiques. Un jeu de relations ambivalentes est expose, ou alternent fantasmes d'agressions deleteres et scenes d'extases fusionnelles avec la matiere. La fonction d'adaptation traditionnellement assignee au jeu, trouve une application paradoxale dans cet univers romanesque : les enjeux individuels engendrent une deconstruction du moi des personnages, qui invite le lecteur a se demander si le jeu ontologique ne consiste pas a apprendre a perdre pour mieux se perdre. Le jeu linguistique s'inscrit dans une strategie du pari litteraire : donner du jeu au langage. Le principe de tension entre la fonction cognitive et sensitive des experiences scripturales, constitue le parcours du "voyage du verbe" de j. M. G. Le clezio. Les nombreuses pratiques ludiques attestent de cette latitude temporairement donnee au langage, pour combler la circularite infinie de ses signes. La fonction sensitive du langage se fonde sur la presence de "la voix originelle" du monde, qui doit retrouver les forces elementaires de la vie. During the decade 1963-1973, le clezio's novels are characterized by a conflict within his writing he presents it as the result of contingency and a structured cultural background, which no writer can escape. How to find the best literary solution to this paradoxical problem, is the central preoccupation of this period. One solution is the adoption of playful language to dramatize adversarial events. The ontological play is founded on an antagonistic relation between the contingency of humain life and the immutable presence of material anhd cosmic forces. Le clezio depicts fantasies of frightful cosmic brutality, alternating with ecstatic union with the universe. Play, which usually acts as a means of integration, in the universe of his novels has the opposite function of alienation and disintegration of the heroes. This causes the reader wonder wether the ontological play does not consist in learning how to lose so in order to find the best way to lose one self. The linguistic play isin keeping with the great wager that is the core le clezio's writing "le voyage du verbe" is a tension between the cognitive and the sensory functions of experiments in writing. The author pratices elaborate abstract linguistic games, vacillating between then and concrete descriptions of sensory life. Ultimately le clezio chooses the latter, because it is in harmony with the original, authentic voice of the world. It achieves an ecstatic silence at the heart of the cosmos

"Cette thèse porte sur le roman autobiographique et l'autofiction à travers l'analyse d'un corpus comparatiste de textes de Paul Auster, J. M. G. Le Clézio et Elsa Morante. La question de recherche est la suivante : quels rapports ces textes entretiennent-ils avec l'autobiographie ? De quelle manière autobiographie et fiction s'entrecroisent chez Auster, Le Clézio et Morante ? L’approche se veut pluridisciplinaire : les outils théoriques de la linguistique de l'énonciation et de la narratologie, mais aussi la critique thématique, les études sur le roman autobiographique et sur la fiction sont ici mis à contribution. Dans la première partie sont analysés les "récits de mémoire" qui structurent ces textes. Si d’un côté le "Je" rétrospectif contribue à créer confusion entre le narrateur et l’auteur, de l'autre ces textes sont traversés par l'altérité : le "Je" est polyphonique et le récit est tissé par l'intertextualité. A travers le discours d'autrui se construit ce que Ricoeur appelle l'"identité narrative". Dans la deuxième partie, on focalise sur le "roman familial" (Marthe Robert) à l'œuvre chez ces écrivains mais aussi sur les images d'intimité (Bachelard) (dont le regressus ad uterum) qui se dégagent de leurs textes à partir des lieux de mémoire. Dans la troisième partie, on interroge d'abord la notion d'autofiction, que l'on adopte ici pour désigner les avatars contemporains du roman autobiographique. This is a PhD thesis about autobiographical novel and "autofiction" through the analysis of a comparative text corpus by Paul Auster, J. M. G. Le Clézio and Elsa Morante. The main research question is : which is the connection between these texts and autobiography ? Which is the intricate interrelationship between autobiography and fiction in Paul Auster, Le Clézio and Elsa Morante's works? Our approach is interdisciplinary : we used the tools of enunciation linguistics, narratological analysis, thematic critical perspective, studies about autobiographical novel and fiction. The first part of this thesis is based on the analyse of the "tales of memories" which characterize these texts. If on the one hand the first person retrospective narration contribute to confuse the narrator and the author, on the other hand alterity cross text corpus : the first person is polyphonic and a great number of intertextual references interweave these texts. This speech of others is the main topic of what Ricoeur calls "narrative identity". The second part, focuses on "family novel" (Marthe Robert) that characterize these texts structures and on images of inwardness (Bachelard) (as regressus ad uterum) that emerge from analysis of places of memory. The third part firstly questions the notion of "autofiction" we use here to describe the contemporary avatars of the autobiographical novel. Secondly, from de point of view of reception, we find onomastic, biographical, paratextual clues that contribute to tend towards an autobiographical reading of these texts. We finally analyse "intersubjective communication" (Couturier) that autofiction establishes between the author and the reader. In conclusion, we make the hypothesis that these writers wish both to show and hide themselves behind the veil of fiction like in hide and go seek game (Winnicott)

Abstract:
This thesis provides the first rigorous history of the concept of midlife crisis. It highlights the close connections between understandings of the life course and social change. It reverses accounts of popularization by showing how an idea moved from the public sphere into academia. Above all, it uncovers the feminist origins of the concept and places this in a historically little-studied tradition of writing about middle age that rejected the gendered "double standard of aging." Constructions of middle age and life-planning were not always oppressive, but often used for feminist purposes. The idea of midlife crisis became popular in the United States with journalist Gail Sheehy's Passages (1976), a critique of Erik Erikson's male-centered model of ego development and psychoanalytic constructions of gender and identity more generally. Drawing on mid-century notions of middle life as the time of a woman's entry into the public sphere, Sheehy's midlife crisis defined the onset of middle age, for men and women, as the end of traditional gender roles. As dual-earner families replaced the male breadwinner model, Passages circulated widely, read by women and men of different generations, including social scientists. Three psychoanalytic experts-Daniel Levinson, George Vaillant, and Roger Gould-rebutted Sheehy by putting forward a male-only concept of midlife as the end of a man's family obligations; they banned women from reimagining their lives. Though this became the dominant meaning of midlife crisis, it was not universally accepted. Feminist scholars, most famously the psychologist and ethicist Carol Gilligan, drew on women's experiences to challenge the midlife crisis, turning it into a sign of emotional instability, immaturity, and egotism. Resonating with widespread understandings of mental health and social responsibility, and confirmed by large-scale surveys in the late 1990s, this relegated the midlife crisis to a chauvinist cliché. It has remained a contested concept for negotiating the balances between work and life, production and reproduction into the present day.

Le clezio rend impossible le trace d'un geste stylistique unique, parce qu'une coupure radicale separe ses ecrits en deux versants inconciliables dont la ligne de crete est incarnee par l'experience biographique amerindienne bouleversante de l'auteur. Un premier bloc grouperait les recits du proces-verbal (1963) jusqu'aux geants (1973), et un second, initie par les voyages de l'autre cote (1975) se prolongerait jusqu'aux dernieres oeuvres parues, la quarantaine et poisson d'or (1997). Le trace de ces deux gestes d'ecriture heterogenes est interroge sous trois angles successifs : l'experience temporelle fictive et son rapport a l'autobiographique, l'ecriture de l'alterite, et l'ecriture du corps et de la voix. Or, par trois fois, une meme opposition structurale partage l'oeuvre en deux cotes. Abstract:
Selon un rapport de l’OMS, les maladies infectieuses figurent parmi les 3 causes les plus fréquentes de mortalité en début de vie. En effet, les nouveau-nés présentent une sensibilité particulièrement importante aux infections, de par leur système immunitaire toujours en développement et donc immature. Parmi les particularités de l’immunité néonatale, l’absence de cDCs CD11chigh CD8α+ durant la 1ère semaine de vie rend compte de l’incapacité du nouveau-né à développer des réponses de type Th1 et T CD8+ cytotoxiques, essentielles à la protection contre certains pathogènes comme Listeria monocytogenes (Lm). Mon travail de thèse a porté sur l’ontogénie des DCs conventionnelles CD11chigh CD8α+ et plus particulièrement sur la fonction de cette lignée de DCs en début de vie lors d’une infection par Lm. Au cours de cette étude, nous avons identifié, chez le nouveau-né, une population splénique de cDCs CD11chigh qui expriment les marqueurs CD24, CD205 et Clec9A mais pas le CD8α. Cette population, qui dépend du facteur de transcription Batf3, acquière le CD8α une fois transférée dans une souris adulte. Ces DCs néonatales, que nous nommerons DCs pre-CD8α Clec9A+, constituent donc les précurseurs des DCs CD8α+. L’étude fonctionnelle de ces DCs pre-CD8α Clec9A+ a montré qu’elles étaient capables de phagocyter Lm et de générer des réponses T CD8+ contre cette bactérie. De plus, ces cellules sécrètent de l’IL-12p40 et de manière unique de l’IL-10 en réponse à une stimulation par Lm. Par contre, contrairement aux cDCs CD8α+ adultes, nous n’avons observé aucune production d’IL-12p70 par les DCs pre-CD8α Clec9A+ en conditions physiologiques. Elles ne sécrètent pas non plus d’IL-23. Nous avons également montré que ces sécrétions d’IL-12p40 et d’IL-10 jouaient respectivement un rôle positif et négatif dans l’induction des réponses T CD8+ contre Lm. Ainsi, la génération des réponses T CD8+ contre Lm semble résulter, en début de vie, d’une balance entre la sécrétion de ces 2 cytokines aux propriétés antagonistes. Par ailleurs, nous avons démontré que ces cellules constituaient une cible privilégiée en vue d’améliorer les stratégies vaccinales en début de vie. En effet, l’administration à des nouveau-nés d’une construction anti-Clec9A/OVA, associée au poly(I:C), induit des réponses T CD8+ anti-Lm mémoires protectrices à l’âge adulte. Finalement, nous montrons que le TNF-α produit par les monocytes et les neutrophiles joue un rôle essentiel dans la génération des réponses T CD8+ en régulant notamment le statut et la fonction des DCs pre-CD8α Clec9A+ en début de vie. En effet, cette cytokine modifie, en faveur d’une production d’IL-12p40, la balance IL-12p40/IL-10 sécrétée par celles-ci. L’inhibition de la production d’IL-10 par le TNF-α pourrait s’expliquer au moins en partie par une inhibition de la β-caténine au sein des DCs pre-CD8α Clec9A+. En conclusion, nous avons caractérisé un précurseur des DCs CD8α+ biologiquement actif au sein de la rate du nouveau-né de 3 jours. Celui-ci représente une cible potentielle pour l’amélioration des stratégies vaccinales contre des bactéries intracellulaires ou des virus en début de vie, et ce pour autant que l’on puisse contrôler ses propriétés régulatrices.

Abstract:
L’étiologie des cancers cutanées est liée à des mutations génétiques résultant de l’exposition aux rayonnements ultraviolets (UV) émis par le soleil. La propagation des cellules cancéreuses dépend aussi des interactions avec les cellules présentes dans le microenvironnement circulant, notamment des fibroblastes associés au cancer (FAC) et des cellules immunitaires. Xeroderma pigmentosum (XP) est une maladie génétique qui comprend 7 groupes de complémentation génétique (XP-A à XP-G). Les patients XP présentent une déficience du mécanisme de réparation des lésions de l’ADN provoquées par les UV. Ces patients sont susceptibles au développement précoce de très nombreux cancers cutanées. XP-C est le groupe de complémentation le plus représenté en Europe. Chez ces patients, les carcinomes spino-cellularies (CSC) sont plus fréquents que les carcinomes baso-cellulaires (CBC) (taux 5 : 1). Les CSC ont un potentiel métastatique plus élevé que les CBC. Des travaux précédents ont suggéré que la réponse immunitaire chez les patients XP pouvait être altérée, incluant un déficit de l’activité cytolytique des cellules Natural Killer (NK) et une diminution du nombre des lymphocytes T circulants.L’objectif central de cette thèse était, d’identifier des facteurs du microenvironnement impliqués dans la progression des cancer cutanées agressifs, en prenant comme modèle de susceptibilité au cancer, des cellules de patients XP-C. Une analyse transcriptomique comparant les fibroblastes WT et des patients XP-C a permis d’identifier que CLEC2A, un ligand activateur du récepteur NKp65 des cellules NK, est exprimé par les fibroblastes WT mais pas par les fibroblastes XP-C. Nos travaux ont pu montrer une diminution du niveau d’expression de CLEC2A au cours de la sénescence réplicative ; une absence dans les FAC et dans les CSC et que, des facteurs solubles secrétés para les CSC diminuent l’expression de CLEC2A. Ces résultats suggèrent que la perte de CLEC2A peut induire un déficit d’activation des cellules NK au sein du microenvironnement tumoral et dans les dermes des patients XP-C. Par la suite, nous avons élaboré un modèle de culture de peau 3D, dans lequel nous avons introduit des cellules NK, en présence ou absence d’anticorps bloquants CLEC2A. Ce modèle nous a permis de montrer que l’interaction CLEC2A/NKp65 régule l’invasion des CSC via un dialogue entre fibroblastes et cellules NK. Skin cancer etiology is related to genetic mutations arising after ultraviolet (UV) sun exposure. The propagation of cancer cells is also dependent of a crosstalk with cells present in the surrounding microenvironment, mainly cancer associated fibroblasts (CAF) and immune cells. Xeroderma pigmentosum (XP) is a genetic disease that comprises seven groups of genetic complementation (XP-A to XP-G). XP patients present a default in the mechanism responsible for the repair of UV-induced DNA lesions. They are prone to develop skin cancers with high frequencies early in their life. XP-C is the most represented complementation group in Europe and in XP-C patients squamous cell carcinoma (SCC) are more frequent than basal cell carcinoma (BCC) (ratio 5:1). SCC have high metastatic potential compared to BCC. Previous studies suggested that the immune responses in XP patients could be altered with defects in their NK lytic activity and a decrease in the levels of circulating T lymphocytes. The main objective of this thesis was to identify microenvironment factors that could contribute to the progression of aggressive skin cancers using XP-C disease cells as a model of skin cancer susceptibility. Comparative transcriptomic analysis of WT and XP-C dermal patient’s fibroblasts revealed that CLEC2A, a ligand of the activating NK receptor NKp65 implicated in the activation of the innate immune system, is expressed in WT fibroblasts and absent in XP-C fibroblasts. Additional work showed that CLEC2A level is decreased in WT fibroblasts during replicative senescence, is absent in CAF and SCC, and is down regulated by soluble factors secreted by SCC cells. These results suggest that the loss of CLEC2A may induce a deficit of NK cell activation in the tumor microenvironment of SCC and in the dermis of XP-C patients. Elaboration of 3D skin culture models including NK cells and, in the presence or absence of blocking anti-CLEC2A antibody, allowed us to show that CLEC2A/NKp65 interaction regulates SCC cells invasion through a crosstalk between fibroblasts and NK cells. Our results suggest that the expression of CLEC2A in fibroblasts contributes to skin immune surveillance while, conversely, its absence under yet unidentified factors, favors the development of aggressive cancers in XP-C patients. CLEC2A could be a potential target in the fight against SCC progression

Dermatofitoses são infecções fúngicas de natureza crônica cujo principal agente etiológico é Trichophyton rubrum. Apesar de sua alta ocorrência mundial, pouco se sabe sobre os mecanismos imunológicos envolvidos nestas infecções. Neste trabalho investigamos a participação de duas classes de receptores de imunidade inata (NLRs e CLRs) na resposta a T.rubrum e avaliamos o perfil proteômico de macrófagos quando estimulados com o fungo. Observamos que T.rubrum foi capaz de induzir a produção de IL-1β dependente do inflamassomo NLRP3 e destacamos o papel da sinalização de IL-1 na modulação da resposta de IL-17. Determinamos os CLRs dectina-1 e dectina-2 como receptores essenciais na produção de citocinas inflamatórias e para o controle da infecção experimental. Curiosamente, a IL-17 e os linfócitos T e B foram dispensáveis para a eliminação do fungo. Também identificamos a proteína CLEC1A como uma novo receptor para fungos, envolvido no reconhecimento de glicolipídeos de T.rubrum. Por fim, a análise proteômica de macrofagos revelou a vimentina e a plastina-2 como duas proteínas potencialmente envolvidas na relação patógeno-hospedeiro.
Dermatophytosis are chronic fungal infections whose main causative agent is Trichophyton rubrum. Despite its high incidence worldwide, the immunological mechanisms underlying these infections remain largely unknown. Here we investigated the involvement of two classes of innate immune receptors (NLRs and CLRs) in the reponse to T.rubrum and performed a proteomic profiling of macrophages upon T.rubrum stimulation. We observed that T.rubrum was able to drive NLRP3 inflammasome-derived IL-1β production and highlighted IL-1 signaling as an important component in the shaping of the IL-17 response. We defined the CLRs dectin-1 and dectin-2 as key receptors for the induction of inflammatory cytokines and for the infection control in the in vivo settings. Curiously, IL-17 cytokines and T and B lymphocytes were dispensable for fungal clearance. In addition, we uncovered CLEC1A as a new receptor in fungal sensing, involved in the recognition of T.rubrum glycolipids. Finally, the proteomic analysis revealed Vimentin and Plastin-2 as two proteins potentially involved in the host-pathogen interaction.

La premiere partie est consacree a l'etude des civilisations meso-americaines et de leurs mythologies, source d'inspiration pour les deux auteurs consideres : il s'agit principalement d'etudier les cultures maya, azteque et lacandone. Ont ete privilegies les mythes fondateurs, les rites divinatoires et quelques themes fondamentaux de la tradition indienne comme le temps, le destin, les sacrifices et la magie des transformations, themes qui sont aussi developpes dans la deuxieme partie de ce travail en relation avec l'oeuvre des ecrivains contemporains qui nous interessent. Based on the principle that writing is not representative of sense, and thus the power of signification is not only property of the writer, but also of writings and their narrative construction, such texts offer to an "active" reading the plurality of their meanings. Engager la réflexion sur le thème du lieu dans un roman est très important, mais aussi très difficile, car il se situe souvent à tous les niveaux textuels. C’est pourquoi, lorsqu’on cherche à comprendre une fiction, il ne faut pas se contenter de décrire les lieux abordés ; il importe d’aller au-delà et saisir ce qui organise l’espace et lui confère parfois un rôle de personnage. Autrement dit, le lieu désigné n’est jamais un élément neutre ou insignifiant. Cette évidence apparaît avec force à la lecture de l’œuvre de Le Clézio. Parler des lieux chez l’auteur, c’est alors parler d’un espace complexe, aux dimensions multiples et variées (littéraire, sociologique, politique, philosophique, voire métaphysique) ; c’est, en fait, réfléchir sur les relations que le texte institue entre tous ses éléments et qui contribuent à donner du sens au récit. Sur ce plan, il se dégage de l’écriture leclézienne une récurrence, d’un livre à l’autre, de quelques lieux puisés dans le vécu de l’auteur, portant témoignage d’une époque et décrits d’une manière poétique, parfois avec fureur. Ces lieux peuvent être ramenés à deux espaces essentiels : la ville et l’espace naturel. Ce sont là deux thèmes obsessionnels du romancier-poète que l’on retrouve en parcourant sa production littéraire. Ces deux lieux porteraient en eux une dualité : la nature opposée à la civilisation. Le Clézio est en effet attiré par les vastes étendues, par les espaces immenses et illimités, ces lieux originels, où tout est pur, non usés par la durée du temps et non marqués par l’intervention de l’homme. À ces espaces mythiques s’oppose la civilisation occidentale, incarnée par la société urbaine, une société dépeinte négativement : espace de métal et du ciment, constamment caché derrière la brume, la société urbaine est un lieu de malaise et d’angoisse ; elle représente l’endroit où les hommes sont isolés, enfermés dans leur solitude et privés du contact avec la nature et avec les autres. Cette dichotomie entre deux mondes fait penser qu’il ne peut exister de compromis entre la nature et l’humanité. Or, rien n’est moins sûr. La pensée leclézienne comporte en effet plusieurs niveaux de lecture et soulève de nombreux questionnements : - Est-il vrai que Le Clézio essaye de nous introduire dans un univers scindé en deux : l’un bon et « sauvage », gardant ses liens avec le sacré, les origines, le mythe ; l’autre mauvais et menaçant, illustré par la ville labyrinthe et l’excès de la modernité ? - Assistons-nous plutôt à une approche poétique des lieux ? - Le but de Le Clézio n’est-il pas de peindre sa vision d’un monde, où le « Je » et le « Nous » se rencontrent et se découvrent mutuellement ? - Et si l’œuvre leclézienne ne fait que poser une question existentielle : ne sommes-nous pas qu’une infime partie de la vie régissant le cosmos ? C’est l’objet de ce travail d’apporter un début de réponse à ces interrogations
Engaging reflection on places or space, in a novel, is very important, but also very difficult, because it is at all textual levels. It is to approach a system of relations in which all the elements which forge it are interdependent and whose functioning creates meaning. This is why, when we try to understand a novel. We must not simply describe the places; we must go beyond and grasp what organizes space and gives it meaning. It must be recognized that the designated place is never a neutral or insignificant element. This evidence appears strongly in the reading of the art work. Talking about places at Le Clézio, it is to speak of a complex space, with multiple dimensions, it is, in fact, reflect on the relations that the text institutes between all its elements and which contribute to give meaning to the narrative. From this point of view, there emerges from the Leclozian work a recurrence from one book to another of a few places drawn from the experience of the writer. Bearing testimony of a time and described in a poetic way. These places can be divided into three spaces: the Desert, the City, the Sea. These are the obsessional themes of Le Clézio that can be found by browsing his literary production. These three subjects would carry in them a duality: nature opposed to civilization. The Clézio is attracted by the vast expanses, by the immense and unlimited spaces, these original places, where everything is pure, unused by the duration of time and not marked by human action: it is the desert, it is the sea.To these mythical spaces is opposed the western civilization incarnated by urban society, a society depicted negatively: space divided, divided into neighborhoods, urban society, that is to say the city,reflects social divisions and inequalities. It represents the place where men are confined, deprived of contact with nature and with others, and where the dislocation of the person who enters or feeds himself is organized this dichotomy between two worlds suggests that there can be no compromise between nature and humanity except through the abandonment of all social responsibilities and characteristics. But nothing is less certain. On the contrary, we believe that the Leclesian thought has several levels of reading and raises a dichotomy between two worlds, which suggests that there can be no compromise between nature and humanity except at the price of the abandonment of all responsibilities and social characteristics. L' objectif principal de ce travail est celui de démontrer les procédés utilisés par J.M.G. Le Clézio pour rendre le roman Le chercheur d'or (1985) un ensemble d'éléments mythiques et légendaires d'origines bien diversifées créant un récit qui ressemble à la poliphonie de Dostoiévski. L'écrivain utilise aussi le contexte historique des siècles XX et XXI pour accentuer les aspects associés à la realité de l'homme moderne et contemporain, démontrant comment le progrès technologique et les relations économiques peuvent nuire à l'être humain. Ainsi, les récits mythologiques et la réalité seront liés et les deux iront montrer à Alexis, le héros du roman, le chemin pour dévoiler le sens réel de son aventure en quête de l'or. Le chercheur d'or sera donc une oeuvre qui explore la realité réfléchie, en accord avec la théorie de Tânia Pellegrini (2009), car le roman met en valeur les événements historiques avec l'objectif de décrire ce qui est possible, mais pas nécessairement réel. En plus, l'interprétation mythologique, qui est été basée sur la théorie de Mircea Eliade (2011), ira soutenir l'idéologie interculturelle de Le Clézio et démontrer son habilité à mêler des genres du XIX siècle (le roman d'aventures) aux éléments du récit contemporain
O principal objetivo deste trabalho é mostrar como J.M.-G. Le Clézio consegue unir em Le chercheur d'or (1985) elementos do mundo mítico e lendário de diferentes origens, criando uma narrativa que se assemelha à polifonia dos romances de Dostoiévski. O autor utiliza-se também do contexto histórico dos séculos XX e XXI para evidenciar aspectos ligados à realidade do homem moderno e contemporâneo, mostrando como o progresso tecnológico e as relações econômicas afetam emocionalmente o ser humano. Desse modo, as narrativas mitológicas e a realidade estarão entrelaçadas e ambas irão mostrar a Alexis, herói do romance, o caminho para desvendar o verdadeiro significado de sua aventura na busca pelo ouro. Le chercheur d'or será, portanto, uma obra que explora a realidade refratada, de acordo com a teoria de Tânia Pellegrini (2009), pois destaca acontecimentos históricos com o objetivo de retratar algo possível, mas não necessariamente real. Além disso, a interpretação mitológica, baseada na teoria de Mircea Eliade (2011), servirá como suporte para que Le Clézio reafirme sua ideologia intercultural, ao mesmo tempo em que mostra sua habilidade em mesclar gêneros característicos do século XIX (romance de aventuras) a elementos da narrativa contemporânea

Abstract:
This capstone seeks to assess and support Homeboy Industries (HBI), a leader in wrap-around services for formerly gang-involved and incarcerated men and women, in their co-creation of a youth services committee and a comprehensive system of care for young people. In doing so, my strategic project consists of conducting stakeholder interviews, focus groups, and synthesizing those findings to present to the organization. The second part of the strategic project involves building and working with a team of individuals from various departments, including case management, mental health, education, job services within two separate agencies, Homeboy Industries and Learning Works Charter School Network, to create a youth services committee that can carry the work forward. In service of evaluating the progress of the strategic project, I will utilize the 4I Framework of Organizational Learning, developed by management professor, Mary Crossan, and her associates from the Ivey School of Business. The 4I Framework contains “four related (sub)processes-intuiting, interpreting, integrating, and institutionalizing-that occur over three levels: individual, group, and organization” (Crossan et al., 1999, p. 524). Ultimately, the goal is to help an already successful leader in wrap-around support services for adults, Homeboy Industries, create an “organized system of care for young people” (Torres, 2015). This goal can be achieved by maximizing its strengths, coupling them with best practices in youth development, and in creating a team that can place the needs of young people in its core mission. Creating an organized system of care, Homeboy Industries-style, can have national implications as the new secretary of education, John King, has made it a point to visit with the leaders of Homeboy Industries (August 2015, Appendix A) in search of models for empowering the youth in a non-traditional way. If clecha, or knowledge that is passed down in prison is the old way of empowering young people, as it often goes in one ear and out the other, then this capstone seeks to capture the experience of Homeboy Industries and Learning Works, the profound work of founder Father Greg Boyle and many amazing practitioners on site at HBI, and combine it with the wisdom of young people, to offer a new approach to empower youth in gangs, the ever-evolving, Homeboy Industries Way. See, the idea of clecha or street wisdom has been passed down for generations as the way that older homies “lace” (give) younger homies advice. In the research on best practices to reach gang involved youth, this clecha notion dates back to the curbside counselor of the 1930s from the seminal work of psychologist Clifford Shaw, but often times, that form of advice has not worked. This has created what Reed Larson, a pioneer in positive youth development, calls the Intentionality Paradox. According to Larson, the paradox lies in that adults want to be intentional with their advice-giving to young people because “it is easier to think about molding clay than about helping the clay mold itself.” (Larson, 2006, p. 682) Larson along with many other experts in the field of youth development are telling us, what young people have been saying for a long time, “stop telling me what to do.” They don’t care to know how much we know (about life or the struggle), they need to know (and feel) how much we actually care. Many adults care so much that they struggle to balance letting youth learn on their own, and sharing their own experiences or clecha. While we are trying to figure it out in the field of youth development and education, too many young people are dying. Every 26 seconds a young person drops out of school in the U.S. (American Graduate, 2016). Over 1 million youth per year are system-involved in “courts with juvenile jurisdiction handling delinquency cases.” (Hockenberry, 2015, p. 6) Thousands of those youth are ending up caged in juvenile halls and prison, and many are dying in our cities nationwide. We must search for new ways to engage and walk with youth in gangs. This is part of that search.

The desert is a unifying principle which brings together henri bosco, jean giono and j. M. G. Le clezio through three novels, le jardin d'hyacinthe, le chant du monde and desert. Stone, sky and water are the figures of the elemental which preside over the creation of the desert's forms and making of them a space charged with the memory of origins. But the desert represents above all a fertile figure because in the novel's organization its function is twofold : it is the privileged instrument of a positive and felicitous world vision, enabling the three authors to give voice to their principle of harmony and it is a needful subject matter which reveals the poetic writing moving on images of pathways. These images wrought from the motifs of the star, water, the bird and the flower, are the instruments of a poetic shaping of space. The desert, thus constructed is the hearth of the transfiguring experience ans reveals itself to be a vast expanse of harmonious well-being. It makes possible the crystallization of the dream of a space in which man regains the fundamental harmony of happiness in and oneness with the world.
