Full textThis thesis presents three years of work with the CMS experiment, in the context of the first LHC collisions. Electron objects were studied in particular, as major tools for multi-lepton analyses, in particular the H → ZZ(∗) → 4ℓ analysis. During the first months of collisions, we took part in the validation of data registered by the electromagnetic calorimeter. We also measured the efficiency of the level-1 electron and photon trigger during the whole 2010 year. The plateau efficiency is of 99. 6 % (resp. 98. 5 %) on electrons in the barrel part (resp. In the endcap part) of the calorimeter. In order to optimize the discovery potential, we built a new electron charge measurement algorithm. In CMS, this measurement is affected by the large amount of material present in the inner tracker. The performance of this algorithm was measured on 2010 data, for electrons from Z boson decay passing a standard selection. The probability of charge mis-identification is of 1. 06 % (0. 19 % with a specific selection), in agreement with the simulation. The physics analysis that was built during this PhD searches doubly charged Higgs bosons decaying into lepton pairs. For the amount of data registered in 2010, one background event is expected to pass the selection, while the amount of signal events depends on the mass hypothesis and on the model. One event was found on data, in agreement with the background expectation, hence the signal was excluded on larger mass ranges than previous experiments: a mass limit was set between 122 GeV and 176 GeV, depending on the model

Full textIn the last 20 years the importance of "high-risk" human papilloma viruses (HPVs) in the development of cancer has been well documented, particularly in the development of cervical cancer. The two viral oncoproteins E6 and E7 participate in a number of cellular processes leading to the immortalization of infected cells. In particular, the process of p53 degradation mediated by E6/E6-AP has yet to be fully elucidated. The understanding of the molecular interaction of "high-risk" HPV E6 with its cellular partners will provide important information for the understanding of cellular immortalization and for the development of specific inhibitors. Furthermore, the conception of targeting strategies will be beneficial for both the diagnosis and treatment of tumours. The first axe of my work concerned the selection, by phage display, of peptides specifically targeting cells derived from cervical tumours. These peptides were used to redirect the diphtheria toxin to SiHa cells, a cell line derived from a human cervical carcinoma. The second axis concerned the definition by phage display of several groups of peptide ligands of the HPV 16 E6 oncoprotein. One of these groups of peptides presented significant homology with a region of p53. The study in vitro as well as on cultured cells of the p53 protein mutated in this region revealed its importance in the degradation of p53 by E6

Full textHuman papilloma viruses infect distinct epithelial cells and cause lesions, which lead simple warts towards cancer. Two viral oncoproteins, E6 and E7, take part in a synergistical way in several cellular parthways, which can lead to immortalisation or transformation. The most cited functions are the degradation of the anti-oncogene p53 by E6 and of the protein pRb by E7. E6 is mainly implicated in the late stages of malign progression towards cancer. This correlates with its ability to degrade PDZ domain containing proteins. These proteins are located at cell-cell junctions and act as scaffolding molecules for building junction and signalisation complexes. Degradation of these proteins often induces invasive cell phenotypes. During my Ph. D. I studied the interaction between PDZ domain containing protein MAGI-1 and the C-terminal domain of E6. I produced the PDZ1 domain of MAGI-1 and the C-terminal domain of E6. Then I set up two methods for measuring protein-protein interactions by Biacore and by comparative chromatographic retention, which allowed to determine the affinity of the interaction. Finally I studied the complex by NMR. The structure calculations of the MAGI-1 PDZ1 bound to a C-terminal E6 peptide is near to completion. The detailed analysis will allow to understand the binding mechanism and the rational design of inhibitors. The interaction measurement techniques will allow to screen inhibitors directed against E6 or PDZ domains at a medium or high throughput for the aim of identifying new therapeutical molecules

Full textFracture of the femoral neck may be a life-threatening condition in the elderly and ay have a poor functional outcome in young adults with the risk of avascular necrosis of the femoral head. Perfusion-tomodenstometry might be useful in studying the remaining vascularization after recent femoral neck fracture showing hypoperfusion thus giving an estimation for the risk of necrosis. Perfusion-tomodensitometry was performed in 20 patients. Images were analysed after delineation of an area of interest. Topographic adjustment was performed and results were analysed after modeling according to the pattern of diffusion. A single pattern was not found. Inter-individual variability was greater than intr-individulal variability. Difference in pattern between patients were important. No difference could be shown between the groups of "unaffected hips", "fractured hips", "fracture of the femoral neck", "extracapsular fracture of the proximal femur" although there was a trend against "fracture of the femoral neck". The only correlation found was between both sides of the same patient, although a tendency towards a difference between both sides of the "neck group" could appear. Analysis of our results does not confirm our work hypothesis. The only correlation found is most likely related to a tomodensitometric evaluation of the bone mineral density. Perfusion tomodensitometry clearly is not suitable to assess residual perfusion of the femoral head nor estimating the risk of avascular necrosis. However, the trends observed might indicate that the difference observed coulde become statistically significant by an increased number of cases

Full textBeing sensitive to the polarization of the light backscattered by the object under study rather than its intensity, polarimetric imaging may reveal some properties of the object to which ordinary intensity is blind. Polarimetric imaging thus features a strong potential for the development of new optical diagnostic for medical purposes. Polarimetric imaging is particularly sensitive to any modifications of the spatial structure of fibrillar collagen in connective tissues, which in turn are known to occur in many diseases, among which cervical dysplasias and genital prolapse. This work was mainly devoted to studying, by means of polarimetric microscopy, the collagen structure in histological plates taken from conisations (front part of uterine cervices extracted because of suspicion of dysplasias) and vaginal tissue. The objective was to eventually correlate the observations with the presence or absence of the considered disease. After a brief introduction, the first chapter describes the current management of cervical cancer, its strengths and weaknesses and the need to develop a new optical diagnostic technique. The second chapter is devoted to the prolapse, with particular emphasis on the usefulness of early detection of persons at risk. The third chapter summarizes the properties of the light polarization needed to understand the following. The results are reported in the last two chapters. In healthy vaginal wall tissue, the collagen is differently oriented in each of the three main layers (adventice, muscularis and mucosa) while this structure disappears in presence of prolapse. About uterine cervices, we observed a well organized collagen layer, just beneath the malpighian epithelium, with much larger retardation in healthy regions with respect to dysplastic ones. This so far unknown layer seems to be primariy responsible for the main trends observed in thick samples in backscattering geometry, namely much stronger retardation and depolarization in healthy versus dysplastic regions of malpighian epithelium

Full textMeasurements of W bosons produced in pPb collisions at nucleon-nucleon centre-of-mass energy $\sqrt{s\rm{_{NN}}}=5.02$ TeV are presented in the muon plus neutrino decay channel. The data sample of 34.6 nb-1 integrated luminosity was collected by the CMS detector. The W boson differential cross sections, lepton-charge asymmetry and forward/backward asymmetry are computed as a function of the lepton pseudorapidity, for muons of transverse momentum higher than 25 GeV/$c$. These observables are compared to two sets of parton distributions (PDF). One of two assumes nuclear modifications (EPS09) while the other is simply a superposition of free proton PDF CT10). Some of the observables deviate from expectations based on unmodified and currently available nuclear PDF. One in particular slightly deviates from both predictions which may indicates dependence of nuclear PDF as a function of the valence quark flavor

Full textThe proto-oncogene c-cbl is cytoplasmic protein expressed in heamatopoetic tissues and testis, implicated in cellular signalling either as a scaffolding protein and/or as a negative modulator of cellular signalling. Although it has been studied extensively, not much attention was granted to examine it's role in the testis, one of the principal organs of it's expression. We report here that c-Cbl expression in rat and mouse testis is limited to developing germ cells with strongest expression in pachytene spermatocytes and round spermatids and its expression is upregulated by testosterone that can be inhibited by the anti-androgen flutamide. Androgen-regulated apoptosis occurring at the end of meiosis I is markedly lower in the testis of c-Cbl KO mice, while KOc-Cbl Mouse Embryo Fibroblasts (MEFs) show an increased sensitivity to apoptosis provoked by oxidative stress. We also cloned three other isoforms of p120c-Cbl from pachytene spermatocytes, named p115c-Cbl, p90c-Cbl and p85c-Cbl after their expected size, harbouring either one (D1 – p115; D2 - p90) or two (D1 & D2 - p85) deletions. The expression of p120c-Cbl and p115c-Cbl can be detected in testis, lung, spleen, thymus and brain while p90c-Cbl and p85c-Cbl expression seems to be restricted to germ cells. The deletion D1 corresponds to exon 10 (amino acids 459-502), deletion D2 originates from alternative splicing, stretching over exons 11 to 15 (amino acids 514-768). Both deletions contain PR regions with a Grb2 binding site but only p85c-Cbl failed to bind to Grb2, showing that the presence of any of these two sites is sufficient for Grb2 – c-Cbl binding. P90c-Cbl transfected cells had shown increased proliferation compared to the other isoforms. The above results suggest that c-Cbl has a finely regulated role in the regulation of spermatogenesis and some of this regulation is realised through a balance between alternative isoforms

Full textOur aim is to lead a formal survey in geometric modeling and computational geometry in order to improve the programming techniques and ensure the algorithms correctness. The originality of the means we use to achieve our purpose rely on one hand on the fact that the specifications and the formai proofs of programs are expressed in the formalism of the Calculus of Inductive Constructions implemented in the Coq system, and on the other hand on the fact that we work in a topology-based geometric modeling framework where the planar subdivisions are described by combinatorial oriented maps. We present a formai case study in computational geometry on a classical problem which involves elementary geometric objects: computing the incremental convex hull ofa finite collection ofplanar points. Our work consists in designing two functional convex hull algorithms, automatically extracting a program in OCaml augmented with input handling and a graphical display of the result, and formally proving their total correctness. Proofs consist in checking the termination of the algorithms as well as highlighting several useful topologic and geometric properties. Finally, we derive a C++ implementation of an optimized program from our specification and integrate it into the library developped in our team

Full textThe use of concepts borrowed from Business Process Management (BPM), quality standards and interoperability allows to be interested by the organization and intergration of enterprise business processes , bringing us to propose a step of implementing a multi-views quality reference. The goal of our work is to present how to integrate several processes using a common reference frame offering various viewpoints. This generalizable step is applied to the integration of two quality standards, ISO 9001v2000 and Capability Maturity Model Integration (CMMI), in order to generate a multi-views quality reference frame allowing a certification relatively to the two standards. This reference frame takes into account chapters imposed by ISO and the recommendations of CMMI. During the implementation of our reference frame, we were interested to the definition of a cost and effort estimation methodology in order to rationalize this critical process for the enterprise. The implementation of the quality reference frame was accompanied by the definition of a step ensuring the interoperability of defined processes with those of customers and/or supplier. An audit project methodology, a documentary reference frame and a competences reference frame were also developed in order to ensure the implementation and the integration of the quality reference frame

Catalytic Membrane Reactors (CMR) are economical attractive processes for natural gas reforming to syngas. The CMR studied consisted of a mixed ionic and electronic conductor dense layer (La(1-x)SrxFe(1-y)GayO3-d) which can separate oxygen from air and transport it, a porous support to ensure the mechanical strength of the reactor and a catalytic layer improving methane reforming. Firstly, a material with a good dimensional stability was chosen and shaped by tape casting. Secondly, the influence of the microstructure of the membrane and the architecture of multilayer reactors on semipermeability properties was studied. Finally, a multilayer dense membrane was elaborated in order to improve performances of the CMR in industrial working conditions

Full textCerebral cortical development is a finely regulated process, depending on diverse progenitor cells. Abnormal behavior of the latter can give rise to cortical malformations. Mutations in Eml1/EML1 were identified in the HeCo mouse, as well as in three families presenting severe subcortical heterotopia (SH). SH is characterized by the presence of mislocalized neurons in the white matter. At early stages of corticogenesis, abnormally positioned apical radial glia progenitors (aRG) were found cycling outside the proliferative ventricular zone (VZ) in the HeCo cortical wall. I focused my research on characterizing aRG in the VZ to assess why some cells leave this region and thus to further understand SH mechanisms. Combining confocal and electron microscopy (EM), I uncovered abnormalities of centrosomes and primary cilia in Eml1-mutant aRGs: primary cilia are shorter, and often remain basally oriented within vesicles. Searching for Eml1-interacting partners using mass spectrometry (MS), combined with exome sequencing of SH patient DNAs, allowed us to identify a ciliary Eml1-interacting partner, RPGRIP1L, showing mutations in a SH patient. Gene ontology analyses of MS data pointed to Golgi apparatus and protein transport as enriched categories. Indeed, Golgi abnormalities were identified in HeCo aRGs. Altogether, these data indicate that the Golgi-to-primary cilium axis is perturbed in Eml1mutant conditions, pointing to new intracellular pathways involved in severe neurodevelopmental disorders
