Abstract:
Biochemical analysis in horses is an important aid for determining correct clinical diagnosis of general, infectious, and some parasitic diseases. This work studied the biochemical and mineral indices in mares of two breeds: the Norik breed Muráň Plain type and the Hucul breed. A total of 34 mares of the Norik breed Muráň Plain type (aged 15.18 ± 5.99 years) and 28 Hucul mares (aged 9.03 ± 5.50 years) were used. Blood serum was analysed using the biochemical analyser Cobas c111 (Roche, Switzerland). Significant difference (P < 0.05) was found between the Norik breed Muráň Plain type and the Hucul mares in aspartate aminotransferase, alkaline phosphatase, lactate dehydrogenase and gamma-glutamyltransferase activity; significant difference (P < 0.01) was found in urea values; and highly significant difference (P < 0.001) was found in glucose values. The mineral profile elements showed a highly significant differences (P < 0.001) between the Norik breed Muráň Plain type and the Hucul mares in phosphorus, magnesium, iron, chloride, potassium, and sodium concentrations. The results confirmed that there are significant differences between horse breeds in some biochemical indices. Therefore, it is appropriate to determine reference values for other horse breeds, as well. To our knowledge, this is the first report that compares biochemical and mineral indices between the Norik breed Muráň Plain type and the Hucul breed.

Abstract:
Vitamin D deficiency and overweight have now become an important global problem in the field of health care as well as public health. A substantial hypovitaminosis D in children with obesity is often accompanied by metabolic disorders.
 The aim of the study was to determine the relationship between the levels of vitamin D and lipid metabolism markers in adolescents with overweight and obesity.
 Material and methods. 139 adolescents were examined (63 (45.4 %) girls and 76 (54.7 %) boys). Depending on the body mass index (BMI) 65 children were overweight and 74 were obese children. The mean age of teenagers was (15,4±2,3) years. To establish vitamin D status using the immune-enzyme method, blood serum levels of 25(OH)D were determined. Lipid metabolism markers were determined using the Roche Diagnostics Cholesterol reagent kit using and automatic Cobas c111 analyzer.
 Results. The study established a direct relationship between the level of vitamin D and high-density lipoprotein cholesterol and indirect with total cholesterol, triglycerides, low-density lipoprotein cholesterol, atherogenic index, body mass index and waist circumference. Using a simple linear regression analysis, it was determined that total cholesterol, low-density lipoprotein cholesterol and atherogenic index have the greatest correlation with vitamin D.
 Conclusions: Vitamin D deficiency in overweight and obese adolescents is associated with an increase in the body mass index, blood pressure, and atherogenic dyslipidemia.

Abstract:
Iron deficiency, with or without anemia, is common in pregnant women and more than half of the anemia’s in the world are due to the deficiency of iron in the serum. The aims of this study were to determine the percentage and level of serum iron among iron supplemented pregnant women in different trimesters and in different age groups among supplemented pregnant women in Sulaimani city. This study was carried out in the private clinic in the Sulaimani city-Kurdistan Region of Iraq. The pregnant women were participated and enrolled between the first of December 2018 and first of December 2019. Two hundred and seventy-five healthy and iron supplemented pregnant women were selected randomly and the questionnaire form, which contains information about age of mothers and their gestational age, was filled and serum iron level was measured by COBAS C111 analyzer. The results of this study found that the percentage of iron deficiency among participants was high 33%, 45%, and 52.6 % in the first trimester in different age groups <25, 25–35, and above 35 years old, respectively. However, in the third trimesters decreased to 12.5%, 7.1%, and 3.7% in <25, 25–35, and above 35 years old, respectively. Regarding serum iron levels, the present study found that there were significant differences between ages 25 and 35 with age <25 years. However, there was no significant difference between first, second, and third trimesters. This study concludes that the percentage of iron deficiency among supplemented pregnant women was high compared to other cities or other countries. Pregnant women who their ages <25 are at risk of serum iron deficiency. On the other hand, pregnancy trimesters had no effects on the serum iron level among supplemented pregnant women.

Abstract:
HbA1c is the test of choice in many countries to monitor and diagnose diabetes. There are continuing debates on the HBA1c usage. HBA1c has some limitations, for example, HBA1c test should not be used in pregnant women and children under the age of 13, vitamin B12 deficiency and Hypertriglycemia increases HBA1c levels, many other conditions such as iron deficiency anemia, stage 4 or 5 chronic kidney disease, hemoglobinopathy all interfere with HBA1c results. Moreover, sensitivity and specificity of the test vary among different ethnic groups. In this study, we aimed to investigate the knowledge of health professionals on HBA1c test limitations and to assess the frequency of HBA1c usage in the pediatric population. We also assessed the quality of the instruments used in Sulaimani laboratories (labs) to evaluate HBA1c. One hundred (100) diagnostic labs in Sulaimani city was surveyed to determine HBA1c instruments types used in labs. A one-page questionnaire on the practice and HBA1c limitations test was given to 100 health professionals. Most of the health professionals in this study were unaware of the limitations of the HBA1c test, 99% of the physicians were unaware of the inaccuracy of the point of cares testing (POCT) devices and the frequency of POCT uses in Sulaimani diagnostic labs. Surprisingly, Sulaimani Pediatric hospital used the HBA1c test to monitor and diagnose type 1 diabetes patients. Our survey also showed Cobas C111 analyzer was the most common (47.8%) type of instrument used in Sulaimani city. the advantage was used by 8% of the labs, Biohermes and auto accent was used by 4.3% of labs. Surprisingly, the Diabetes and Endocrine Center in Sulaimani used the POCT device SD A1cCare instrument. In conclusion, almost all health professionals were unaware of the HBA1c limitations. Continuing education programs to all health professional should be mandated in Sulaimani city hospitals to ensure high quality of the HBA1c test. Most importantly, health professionals need to be aware that POCT is widely used in Sulaimani labs and that POCT is considered an inaccurate method for HBA1c evaluation. Pediatric physicians need to be reminded that HBA1c is not recommended in children.

Abstract:
Achatina achatina snail specie are considered by many people in Nigeria, Ghana and other parts of West Africa to be the most prized snail for eating. In general, lectins bind to sugar moieties in cell walls or membrane, thereby change the physiology of the membrane to cause agglutination, mitosis or other biochemical changes in the cell. It has been deduced that lectins could be toxic and can as well be used as potent administrations that could be used or serve as substitutes for routine treatment or management of many disorders. Based on these, the toxicity of the Achatina achatina snail lectin in animals was investigated with a view to determining the nutritional value of the snail as food stuffs by carrying out tests to determine the blood values of Liver Function Tests (LFT) parameters in Albino Wistar Rats administered with the lectin. A total of 120 samples of the Nigeria Achatina achatina snail specie were collected, authenticated at the Zoology Department of the University of Nigeria, Nsukka and 80mls of pooled crude Lectin extract was obtained. Purifications were performed on 20mls of the crude extract in three steps viz, Ammonium sulphate precipitation and Dialysis (Partial purifications), Con A Sepharose 4B affinity Chromatography column (Complete purification). The affinity purified lectin was used for all the tests conducted in this research. The crude, partially and complete/affinity purified Lectin extracts were subjected to Haemagglutination tests. The Lectin was further assessed to determine its effects on Liver Function Tests (LFT) parameters viz, Total bilirubin (TB), Conjugate bilirubin (CB), Alkaline phosphatase (ALP), Aspartate transaminase (AST) and Alanine transaminase (ALT) as follows: A total of Thirty-five (35) male Albino Wistar Rats weighing 101-180g and aged 2-3 months obtained from the Animal house of University of Nigeria, Nsukka, were used in this research. The animals were Grouped into 5 (A-E) and allowed for 2 weeks acclimatization. Graded doses of 0.04ml, 0.05ml and 0.06ml of the Affinity purified Lectin were administered intra-peritoneally to each of the rats in Groups A-D (test groups) according to their body weights at intervals of 2 days for 1 week. Group E served as the control. Two (2) mls of blood was collected from each of the rats before and 24 hours after the last day of lectin administration for the following tests: TB, CB, ALP, AST and ALT (performed by means of Roche Cobas C111 automated chemistry analyser). The results of the research showed as follows: On complete/affinity purification, 15mls of pure sample containing only the high molecular weight lectin was obtained. The respective haemagglutination tests on the crude, partially and affinity purified Lectin showed on standardization preferential agglutinations with Blood group A type. Bar charts statistics show that there was Post lectin administration mean increase in TB, CB and AST when the Post administrations values were compared with the Pre values. The Bar charts statistics show that there was Post lectin administration mean decrease in ALP and ALT. However, the differences in the Pre and Post administration mean values of these parameters were further subjected to one way analysis of variance (ANOVA) test statistics aimed at determining whether the mean increases or decreases in these assessed parameters were statistically significant. The ANOVA statistics show that the effects of the lectin on all the assessed LFT parameters viz, TB, CB, ALP, AST and ALT were statistically insignificant (P > 0.05). The results obtained in this research has succeeded in demonstrating that the A. achatina snail lectin is non-toxic, non-carcinogenic and therefore point to its nutritive value as food stuff, hence supports the snail eating education.

Abstract:
Por su naturaleza, el laboratorio de urgencias ha de responder a la demanda de resultados en plazos muy cortos de tiempo.Hay ocasiones en que el o los analizadores de química se averían simultáneamente, y en ese momento un equipo de gases puede ser vital para suplir al analizador convencional. No obstante, para que el resultado sea fiable, el método o analizador debe ser validado. En nuestro servicio las determinaciones más solicitadas en el paciente crítico son, glucemia y lactato, es por esto que nuestro trabajo tiene como propósito validar como analizador de glucosa y lactato al equipo de gases COBASR b221. Se procesaron 210 muestras de sangre con el objetivo de estudiar la concordancia de resultados de estos analitos, en sangre total y en plasma. Para evaluar la concordancia entre las metodologías de análisis para los analitos considerados, se realizaron pruebas a través de comparación de medias apareadas. Se determinó el coeficiente de concordancia de Lin y su intervalo de confianza, además se construyó el gráfico de Bland-Altman. En el caso de la determinación de lactato, para determinar la importancia clínica de la diferencia entre los resultados obtenidos en ambos equipos, se calculó el porcentaje de concordancia entre ambos métodos utilizados, en base a tres rangos de concentración de lactato, clínicamente relevantes.El COBASR b221 tiende a sobreestimar ambos parámetros, esto nos lleva a concluir que los diferentes resultados aportados por este equipo tienen relevancia clínica y hay que ser cautos a la hora de valorar tanto glucosa como lactato.

Abstract:
Aim: This study evaluated D-dimer level in pregnant and non-pregnant women in Southwestern Nigeria in order to provide more information on the concentration and liable risks in this region.
 Study Design: This is a cross sectional study where convenience sampling method was applied in sample collection.
 Place and Duration of Study: Blood samples were collected from pregnant women attending the antenatal clinics of Federal Teaching Hospital Ido-Ekiti (FETHI), Ekiti; Federal Medical Centre (FMC), Owo; and LAUTECH Teaching Hospital (LTH), Osogbo in Southwestern Nigeria.
 Methodology: Exactly three hundred pregnant (300) and one hundred and fifty (150) apparently healthy non pregnant women were recruited for this study. The blood samples were analysed for haematocrit (HCT) and platelet count using Sysmex KX-2IN (Japan); prothrombin time (PT) and activated partial thromboplastin time (APTT) by Diagen reagents (Diagnostic Ltd., UK); the international normalized ratio (INR) was calculated from the PT results; and D-dimer quantitative assay using Tina Quant Gen 2 on Cobas C111 (Roche). Data analysis was performed using IBM-SPSS version 25.0; mean and standard deviation was used to summarize continuous variables and descriptive and Inferential statistical tests were employed with level of statistical significance was determined at p<0.05.
 Results: The mean D-dimer levels were significantly higher in the pregnant women (0.87 ± 1.00 ugFEU/ml) than in controls (0.31 ± 0.22 ugFEU/ml) with 42% of the pregnant population having elevated concentration while the mean PT, INR and HCT were significantly higher in controls than the subjects (p<0.05).Furthermore, the HCT, platelet, PT and INR were observed to be highest at first trimester; 36.04±5.09 (L/L), 182.72±35.11 (x109/L), 11.80±1.86 (seconds) and 0.35±0.15 respectively, decreasing across the second and the third trimester. On the other hand, the D-dimer and APTT increased exponentially from the first trimester; 0.42±0.18 (ugFEU/ml) and 30.80±3.30 (seconds), through the second and third trimesters respectively (p>0.05).
 Conclusion: This study shows a significant increase in D-dimer in the pregnant subjects when compared with the control and an exponential increase in the third trimester, also a significant reduction in some other baseline coagulation profile hence depicting D-dimer as a notable significant marker of coagulation and fibrinolysis. This therefore emphasizes the hypercoagulable state of pregnancy and a need for adequate monitoring.

Abstract:
Klinisk kemiska analyser har hög klinisk relevans. I serum/plasma kan olika parametrar kvantifieras. Dessa parametrar kan vara proteiner, enzymer, joner, metaller, lipider och kolhydrater. Med hjälp av referensintervall kan veterinärer ställa diagnos, följa behandling och sjukdomsförlopp. Parametrar detekteras med olika analysprinciper/metoder; kolorimetri, immunturbidimetri, enzymatisk metod och potentiometri. Djursjukhuset, AniCura, i Hässleholm mottagas både hundar och katter. Cirka 120 kemianalyser analyseras varje dag. AniCura har köpt in ett nytt våtkemiiinstrument, Indiko Plus, som ska ersätta Cobas C111. Med Indiko Plus tillkommer fler provpositioner, 8 nya analyser, ökad kapaciteten och underlättad användning.  Syftet med denna studie var att jämföra instrumenten och ta fram eget referensintervall som jämfördes med referensintervall framtaget av Thermo Fisher. Verifiering av det nya instrumentet genomfördes med precisionsstudie och linjäritetsstudie. Provtagning på hundar och katter utfördes av personal på AniCura. Jämförelsen gjordes med patientprover och referensintervall togs fram med hjälp av prover från friska hundar och katter. Jämförelsen visade att 9 av 13 analyser hade statistisk signifikant skillnad. Orsaken till det beror troligen på skillnaden av reagens, instrumentens ålder och tid mellan mätningar. Ett nytt referensintervall utarbetades och skiljde sig inte mycket från Thermo Fishers intervall. Vidare validering på grund av liten population rekommenderades. Precisionen för Indiko Plus blev godkänd. Linjäriteten blev icke linjär och berodde troligen på en dålig pipett och bör göras om.<br>Clinical chemical analyzes has high clinical relevance. In serum/plasma, different parameters can be quantified. Parameters can be proteins, enzymes, ions, metals, lipids, and carbohydrates. With reference intervals, veterinarians can set diagnosis, follow treatment and development of the disease. Parameters are detected with different analysis principles/methods; colorimetry, immunoturbidimetry, enzymatic method and potentiometry. The animal hospital, AniCura, in Hässleholm accept dogs and cats. About 120 chemical analyzes are analyzed every day. AniCura purchased a new instrument, Indiko Plus, which will replace Cobas C111. Indiko Plus provide, more sample positions, 8 new analyzes, increased capacity, and facilitated use. The purpose of this study was to compare the instruments and produce a new reference interval which was compared to the reference interval provided by Thermo Fisher. To verify Indiko Plus, a precision and linearity study were conducted. Blood sampling of dogs and cats was performed by staff at AniCura. The comparison was made with patient samples and the reference intervals were obtained using samples from healthy animals. The comparison showed 9 of 13 analyzes had a statistically significant difference. The reason for this is probably due to the difference in reagents, the age of the instruments and the time between measurements. A new reference interval was developed and did not differ much from the Thermo Fisher interval. Further validation due to low population was recommended. The precision for Indiko Plus was approved. The linearity study shows not linear trend but was likely due to a bad pipette and should be redone.
